Abstract
Un método para generar una línea celular que es capaz de complementar adenovirus recombinantes con E1 suprimida, que comprende proporcionar una célula humana con una molécula de ácido nucleico que comprende secuencias E1 adenovirales que codifican los productos génicos E1A y E1B funcionales y carecen de secuencias pIX, para integrar la molécula de ácido nucleico en el genoma de dicha célula humana y para expresar E1A, E1B 55 kD y E1B 21 kD en dicha célula.
Description
Sistemas de empaquetado para adenovirus recombinante humano destinados a terapia génica.
La invención se refiere al campo de la tecnología del ADN recombinante, más concretamente al campo de la terapia génica. En particular la invención se refiere a la terapia génica en la que se utilizan materiales derivados de adenovirus, en particular adenovirus recombinante humano. Especialmente se refiere a vectores derivados de virus novedosos y líneas celulares de empaquetamiento novedosas para vectores basados en adenovirus.
La terapia génica es un concepto desarrollado recientemente para el cual se puede y se han ideado una amplia gama de aplicaciones.
En la terapia génica se introduce una molécula que porta información genética en algunas o todas las células de un huésped, como resultado de lo cual se añade información genética al huésped en un formato funcional.
La información genética añadida puede ser un gen o un derivado de un gen, tal como un ADNc, que codifica una proteína. En este caso el formato funcional significa que la proteína puede ser expresada por la maquinaria de la célula huésped.
La información genética también puede ser una secuencia de nucleótidos complementaria a una secuencia de nucleótidos (ya sea ADN o ARN) presente en la célula huésped. El formato funcional en este caso consiste en que la molécula de ADN (ácido nucleico) añadida o las copias realizadas de la misma in situ sean capaces de emparejar las bases con la secuencia complementaria presente en la célula huésped.
Entre las aplicaciones se incluyen el tratamiento de las alteraciones genéticas suplementando una proteína u otra sustancia que, debido a dicha alteración genética, no se encuentra presente o al menos está presente en cantidades insuficientes en el huésped, el tratamiento de tumores y (otras) enfermedades adquiridas tales como las enfermedades (auto)inmunes o infecciones, etc.
Como puede resultar evidente a partir de lo anterior, existen básicamente tres enfoques diferentes en terapia génica, uno dirigido a compensar una deficiencia presente en un huésped (mamífero); el segundo dirigido a la separación o eliminación de sustancias no deseadas (organismos o células) y el tercero a la aplicación de una vacuna recombinante (tumores o microorganismos foráneos).
Para el propósito de la terapia génica, se han propuesto como vehículos adecuados adenovirus que portan deleciones.
Los adenovirus son virus con ADN sin envuelta. Los vectores de transferencia génica derivados de adenovirus (denominados vectores virales) tienen numerosos rasgos que los hacen particularmente útiles para la transferencia génica con tales fines. Por ejemplo, la biología de los adenovirus está caracterizada con detalle, el adenovirus no está asociado a patologías humanas graves, el virus es extremadamente eficaz al introducir su ADN en la célula huésped, el virus puede infectar una amplia variedad de células y tiene una amplia gama de huéspedes, el virus puede ser producido en grandes cantidades con relativa facilidad, y el virus se puede volver de replicación deficiente mediante deleciones en la región temprana 1 (E1) de genoma viral.
El genoma de los adenovirus es una molécula de ADN de doble hebra lineal de aproximadamente 36.000 pares de bases con la proteína terminal de 55 kDa unida covalentemente al extremo 5' de cada hebra. El ADN de Ad contiene Repeticiones Terminales Invertidas (ITR) idénticas de aproximadamente 100 pares de bases dependiendo de la longitud exacta del serotipo. Los orígenes de replicación virales están localizados en las ITR exactamente en los extremos del genoma. La síntesis de ADN se produce en dos fases. Primero, la replicación se desarrolla mediante desplazamiento de la hebra, generando una molécula dúplex hija y una hebra desplazada parental. La hebra desplazada es de hebra sencilla y puede formar un intermedio denominado "panhandle", que permite el inicio de la replicación y la generación de una molécula dúplex hija. Alternativamente, la replicación puede desarrollarse desde ambos extremos del genoma simultáneamente, obviando el requerimiento de formar la estructura "panhandle". La replicación se resume en la Figura 14 adaptada de (Lechner and Kelly, 1977).
Durante el ciclo de infección productivo, los genes virales son expresados en dos fases: la fase temprana, que es el período hasta la replicación del ADN viral, y la fase tardía, que coincide con el inicio de la replicación del ADN viral. Durante la fase temprana sólo se expresan los productos génicos tempranos, codificados por las regiones E1, E2, E3 y E4, que portan numerosas funciones que preparan la célula para la síntesis de las proteínas estructurales virales (Berk, 1986). Durante la fase tardía se expresan los productos génicos virales tardíos además de los productos génicos tempranos y se disparan la síntesis de ADN y de proteínas de la célula huésped. Por consiguiente, la célula se dedica a la producción de ADN viral y de proteínas estructurales virales (Tooze, 1981).
La región E1 del adenovirus es la primera región del adenovirus expresada después de la infección de la célula diana. Esta región consta de dos unidades transcripcionales, los genes E1A y E1B, ambos requeridos para la transformación oncogénica de cultivos de roedor primarios (embriónarios). Las principales funciones de los productos génicos E1A son:
i) inducir a las células quiescentes a entrar en el ciclo celular y reanudar la síntesis de ADN celular, y
ii) activar transcripcionalmente el gen E1B y las otras regiones tempranas (E2, E3, E4). La transfección de las células primarias con el gen E1A solo puede inducir una proliferación ilimitada (inmortalización), pero no da como resultado una transformación completa. Sin embargo, la expresión de E1A en la mayoría de los casos da como resultado la inducción de la muerte celular programada (apoptosis), y sólo ocasionalmente se obtiene la inmortalización (Jochemsen y col., 1987). La co-expresión del gen E1B es requerida para evitar la inducción de la apoptosis y para hacer que se produzca la transformación morfológica completa. En líneas celulares inmortales establecidas, un elevado nivel de expresión de E1A puede causar la transformación completa en ausencia de E1B (Roberts y col., 1985).
Las proteínas codificadas por E1B ayudan a E1A a redirigir las funciones celulares para permitir la replicación viral.
Las proteínas E1B de 55 kD y E4 de 33 kD, que forman un complejo que se localiza esencialmente en el núcleo, funcionan inhibiendo la síntesis de las proteínas del huésped y facilitando la expresión de los genes virales. Su principal influencia consiste en establecer un transporte selectivo de ARNm virales desde el núcleo hasta el citoplasma, simultáneamente al comienzo de la fase tardía de la infección. La proteína E1B de 21 kD es importante para corregir el control temporal del ciclo de infección productivo, evitando de ese modo la muerte prematura de la célula huésped antes de que el ciclo vital del virus se haya completado. Los virus mutantes incapaces de expresar el producto génico E1B de 21 kD manifiesta un ciclo de infección acortado que está acompañado de una degradación excesiva del ADN cromosómico de la célula huésped (fenotipo deg) y un efecto citopático intensificado (fenotipo cyt) (Telling y col., 1994). Los fenotipos deg y cyt son suprimidos cuando además el gen E1A es mutado, indicando que estos fenotipos son una función de E1A (White y col., 1988). Además, la proteína E1B de 21 kDa disminuye la velocidad mediante la cual E1A pone en marcha los otros genes virales. No se sabe todavía a través de qué mecanismo E1B de 21 kD sofoca estas funciones dependientes de E1A.
Los vectores derivados de adenovirus humanos, en los que al menos la región E1 ha sido suprimida y remplazada por un gen de interés, han sido extensamente utilizados para experimentos de terapia génica en fase pre-clínica y clínica.
Como se ha establecido antes, todos los vectores de adenovirus utilizados en la actualidad en terapia génica tienen una deleción en la región E1, donde se puede introducir información genética novedosa. La deleción de E1 vuelve al virus recombinante de replicación deficiente (Stratford-Perricaudet and Perricaudet, 1991). Los autores de la presente invención han demostrado que los adenovirus recombinantes son capaces transferir eficazmente genes recombinantes al hígado de rata y al epitelio de las vías respiratorias de monos rhesus (Bout y col., 1994b; Bout y col., 1994a). Además, los autores de la presente invención (Vincent y col., 1996a; Vincent y col., 1996b) y otros (ver, v.g., Haddada y col., 1993) han observado una transferencia génica mediada por adenovirus in vivo muy eficaz a una variedad de células tumorales in vitro y a tumores sólidos en modelos animales (tumores de pulmón, glioma) y xenoinjertos humanos en ratones inmunodeficientes (pulmón) in vivo (revisado por Blaese y col., 1995).
En contraste por ejemplo con los retrovirus, los adenovirus a) no se integran en el genoma de la célula huésped; b) son capaces de infectar células que no están en división y c) son capaces de transferir eficazmente genes recombinantes in vivo (Brody and Crystal, 1994). Esos rasgos hacen de los adenovirus atractivos candidatos para la transferencia génica in vivo, por ejemplo, de genes suicidas o citoquinas a células tumorales.
No obstante, un problema asociado con la tecnología de adenovirus recombinante actual es la posibilidad de generación no deseada de adenovirus de replicación competente (ARC) durante la producción de adenovirus recombinante (Lochmüller y col., 1994; Imler y col., 1996). Esta está causada por la recombinación homóloga entre secuencias solapantes del vector recombinante y los constructos del vector presentes en la línea celular complementadora, tales como las células 293 (Graham y col., 1977). El ARC en los lotes que se van a utilizar en pruebas clínicas no es deseable debido a que el ARC i) replicará de una manera descontrolada; ii) puede complementar los adenovirus recombinantes de replicación defiente, causando una multiplicación descontrolada de los adenovirus recombinantes y iii) los lotes que contienen ARC inducen una lesión tisular significativa y por tanto fuertes efectos secundarios patológicos (Lochmüller y col., 1994). Por lo tanto, se debe probar primero que los lotes que van a ser utilizados en pruebas clínicas están libres de ARC (Ostrove, 1994). En un aspecto de la invención se resuelve este problema de la producción de virus ya que los autores de la presente invención han desarrollado células de empaquetamiento que no tienen secuencias solapantes con un nuevo vector básico y por tanto son adecuadas para la producción segura a gran escala de adenovirus recombinantes.
Uno de los problemas adicionales asociados con el uso de vectores de adenovirus recombinantes es la reacción de defensa del huésped frente al tratamiento con adenovirus. En resumen, los adenovirus recombinantes son sometidos a la deleción de la región E1 (ver arriba). Los productos de E1 de adenovirus desencadenan la transcripción de los otros genes tempranos (E2, E3, E4), que por consiguiente activan la expresión de los genes tardíos del virus. Por lo tanto, generalmente se pensaba que los vectores con E1 suprimida no expresarían ningún otro gen del adenovirus. No obstante, recientemente se ha demostrado que algunos tipos de células son capaces de expresar genes de adenovirus en ausencia de las secuencias de E1. Esto indica, que algunos tipos de células poseen la maquinaria para conducir la transcripción de los genes de adenovirus. En particular, se demostró que tales células sintetizan E2A y las proteínas tardías del adenovirus.
En el marco de la terapia génica, esto significa que la transferencia del gen recombinante terapéutico a células somáticas no sólo produce la expresión de la proteína terapéutica sino que también produce la síntesis de proteínas virales. Las células que expresan las proteínas adenovirales son reconocidas y eliminadas por los Linfocitos T Citotóxicos, que de este modo, a) erradican las células transducidas y b) ocasionan inflamaciones (Bout y col., 1994a; Engelhardt y col., 1993; Simon y col. 1993). Como esta reacción adversa está impidiendo la terapia génica, se han sugerido numerosas soluciones a este problema, tales como a) utilización de agentes inmunosupresores después del tratamiento; b) conservación de la región E3 del adenovirus en el vector recombinante (ver la solicitud de patente EP 0.707.071) y c) utilización de mutantes ts de adenovirus humanos, que tienen un punto de mutación en la región E2A (patente WO 94/28938).
No obstante, estas estrategias para salvar la respuesta inmune tienen sus limitaciones.
El uso de los adenovirus recombinantes mutantes ts disminuye la respuesta inmune en cierto grado, pero era menos eficaz al prevenir las respuestas patológicas en los pulmones (Engelhardt y col., 1994a).
La proteína E2A puede inducir una respuesta inmune por sí misma y juega un papel fundamental en la puesta en marcha de la síntesis de proteínas de adenovirus tardías. Por lo tanto, es atractivo elaborar adenovirus recombinantes que tienen mutaciones en la región E2, haciéndolos sensibles a la temperatura (ts), como se ha reivindicado en la solicitud de patente WO 94/28938.
La principal desventaja de este sistema es el hecho de que, aunque la proteína E2 es inestable a la temperatura no permisiva, la proteína inmunogénica todavía está siendo sintetizada. Además, es de esperar que la proteína inestable active la expresión del gen tardío, aunque en un grado bajo. Los adenovirus recombinantes mutantes ts125 han sido sometidos a ensayo, y se ha informado de una expresión génica recombinante prolongada (Yang y col., 1994b; Engelhardt y col., 1994a; Engelhardt y col., 1994b; Yang y col., 1995). Sin embargo, la patología en los pulmones de ratas Cotton todavía era elevada (Engelhardt y col., 1994a), indicando que el uso de los mutantes ts sólo produce una mejora parcial de la tecnología de los adenovirus recombinantes. Otros (Fang y col., 1996) no observaron una expresión génica prolongada en ratones ni en perros utilizando adenovirus recombinantes ts125. Una dificultad adicional asociada con el uso de adenovirus mutantes es que se observa una elevada frecuencia de reversión. Estos revertientes son o bien revertientes reales o bien el resultado de segundas mutaciones en el sitio (Kruijer y col., 1983; Nicolas y col., 1981). Ambos tipos de revertientes tienen una proteína E2A que funciona a la temperatura normal y por lo tanto tienen una toxicidad similar a la del virus de tipo salvaje.
En otro aspecto de la presente invención los autores de la presente invención suprimen por lo tanto las secuencias codificadoras de E2A del genoma del adenovirus recombinante y transfectan estas secuencias E2A a las líneas celulares (de empaquetamiento) que contienen las secuencias E1 para complementar los vectores de adenovirus recombinantes.
Los principales obstáculos de este enfoque son a) que E2A debe ser expresada a niveles muy elevados y b) que la proteína E2A es muy tóxica para las células.
La presente invención describe por lo tanto en otro aspecto el uso del gen E2A mutante ts125, que produce una proteína que no es susceptible de unirse a secuencias de ADN a la temperatura no permisible. Se pueden mantener elevados niveles de esta proteína en las células (debido a que no es tóxica a esta temperatura) hasta que se realiza la puesta en marcha a la temperatura permisiva. Esto se puede combinar colocando el gen E2A mutante bajo la dirección de un promotor inducible, tal como por ejemplo el promotor inducible por esteroides tet, el de la metalotioneína, el del receptor 1 de ácido retinoico u otros sistemas inducibles. Sin embargo en otro aspecto más de la invención, se describe el uso de un promotor inducible para controlar el momento de producción de E2A salvaje tóxica.
Dos notables ventajas adicionales del adenovirus recombinante con E2A suprimido son el incremento de la capacidad para albergar secuencias heterólogas y la selección permanente de células que expresan la E2A mutante. Esta segunda ventaja se refiere a la elevada frecuencia de reversión de la mutación ts125: cuando se produce la reversión en una línea celular que alberga E2A ts125, ésta será letal para la célula. Por lo tanto, existe una selección permanente para aquellas células que expresan la proteína E2A mutante ts125. Además, como los autores de la presente invención en un aspecto de la presente invención generan adenovirus recombinantes con E2A suprimido, no tendrán el problema de la reversión en sus adenovirus.
Los autores de la presente invención describen la construcción de combinaciones novedosas y mejoradas de líneas celulares de empaquetamiento (novedosas y mejoradas) y vectores de adenovirus recombinantes (novedosos y mejorados). Los autores de la presente invención proporcionan:
4 Los constructos de empaquetamiento que se van a utilizar para la generación de líneas celulares de empaquetamiento complementadoras de célulasdiploides (no exclusivamente de origen humano) sin necesidad de selección con genes marcadores. Estas células son inmortalizadas mediante la expresión de E1A. Sin embargo, en este caso concreto es esencial la expresión de E1B para evitar la apoptosis inducida por las proteínas E1A. La selección de las células que expresan E1 se logra mediante la selección en cuanto a la formación de focos (inmortalización), como se describe para las células 293 (Graham y col., 1977) y las células 911 (Fallaux y col., 1996), que células de riñón embriónico humano (HEK) y retinoblastos embriónicos humanos (HER) transformados con E1, respectivamente.

- Tras la transfección de las células HER con el constructo pIG.E1A.E1B (Fig. 4), se pudieron establecer siete líneas celulares independientes. Estas líneas celulares fueron denominadas PER.C1, PER.C3, PER.C4, PER.C5, PER.C6, PER.C8 y PER.C9. PER indica PGK-E1-Retinoblastos. Estas líneas celulares expresan las proteínas E1A y E1B, son estables (v.g. PER.C6 durante más de 57 pases) y complementan los vectores de adenovirus carentes de E1. Los rendimientos de adenovirus recombinante obtenidos en las células PER son algo superiores a los obtenidos en las células 293. Una de estas líneas celulares (PER.C6) ha sido consignada en la ECACC con el número 96022940.

- Nuevos vectores de adenovirus con deleciones de E1 ampliadas (deleción nt. 459 – 3.510). Esos vectores virales carecen de secuencias homólogas a las secuencias de E1 en dichas líneas celulares de empaquetamiento. Estos vectores adenovirales contienen secuencias promotoras pIX y el gen pIX, ya que pIX (de sus secuencias promotoras naturales) sólo puede ser expresado a partir del vector y no por las células de empaquetamiento (Matsui y col., 1986, Hoeben and Fallaux, pers. Comm: Imler y col., 1996).

- Las líneas celulares de empaquetamiento que expresan E2A basadas preferiblemente en líneas celulares establecidas que expresan E1A o células diploides que expresan E1A + E1B (ver en 2 -4). La expresión de E2A o bien está bajo el control de un promotor inducible o bien el mutante ts125 E2A es dirigido por un promotor inducible
o constitutivo.
8. Vectores de adenovirus recombinantes como se ha descrito antes (ver 6) pero que portan una deleción adicional de las secuencias E2A.
Los vectores de adenovirus según la presente invención conservan al menos una porción del genoma viral que es requerida para la encapsidación del genoma en las partículas del virus (la señal de encapsidación), así como al menos una copia de al menos una parte funcional o un derivado de la Repetición Terminal Invertida (ITR), esto es secuencias de ADN derivadas de los extremos del genoma del adenovirus lineal. Los vectores según la presente invención también contendrán un transgen conectado a una secuencia promotora para dirigir la expresión del transgen.
Los principales problemas asociados con los vectores derivados de adenovirus actuales son:
A) La fuerte inmunogenicidad de la partícula de virus
B) La expresión de los genes del adenovirus que reside en los vectores adenovirales, dando como resultado una respuesta de las células T citotóxicas frente a las células transducidas.
C) La baja cantidad de secuencias heterólogas que pueden ser acomodadas en los vectores actuales (Hasta aproximadamente un máximo de 8.000 pb de ADN heterólogo).
Información sobre A). La fuerte inmunogenicidad de las partículas de adenovirus produce una respuesta inmunológica del huésped, incluso después de la administración del vector adenoviral. Como resultado del desarrollo de los anticuerpos neutralizantes, una posterior administración del virus será menos eficaz o incluso completamente ineficaz. Sin embargo, una expresión prolongada o persistente de los genes transferidos reducirá el número de administraciones requeridas y puede evitar el problema.
Información sobre B). Los experimentos realizados por Wilson y colaboradores han demostrado que después de la transferencia génica mediada por adenovirus a animales inmunocompetentes, la expresión del transgen disminuye gradualmente y desaparece aproximadamente 2 -4 semanas después de la infección (Yang y col., 1994a; Yang y col., 1994b). Esto está causado por el desarrollo de una respuesta de las Células T Citotóxicas (CTL) frente a las células transducidas. Las CTL fueron dirigidas contra proteínas de adenovirus expresadas por los vectores virales. En la síntesis por las células transducidas de la proteína de unión al ADN de adenovirus (el producto del gen E2A), se pudieron establecer proteínas pentónicas y de la fibra (productos génicos tardíos). Estas proteínas de adenovirus, codificadas por el vector viral, fueron expresadas a pesar de la deleción de la región E1. Esto demuestra que la deleción de la región E1 no es suficiente para evitar completamente la expresión de los genes virales (Engelhardt y col., 1994a).
Información sobre C). Los estudios de Graham y colaboradores han demostrado que los adenovirus son capaces de encapsidar ADN de un tamaño de hasta un 105% el del genoma normal (Bett y col., 1993). Los genomas más grandes tienden a ser inestables dando como resultado la pérdida de secuencias de ADN durante la propagación del virus. Combinando deleciones en las regiones E1 y E3 del genoma viral aumenta el tamaño máximo del foráneo que puede ser encapsidado hasta aproximadamente 8,3 kb. Además, algunas secuencias de la región E4 parecen ser innecesarias para el crecimiento del virus (añadiendo otras 1,8 kb a la capacidad máxima de encapsidación). Asimismo la región E2A puede ser suprimida del vector, cuando el producto del gen E2A es proporcionado en trans en la línea celular de encapsidación, añadiendo otras 1,6 kb. No obstante, es improbable que la capacidad máxima del ADN foráneo puede ser incrementada significativamente más allá de 12 kb.
Las aplicaciones de las invenciones descritas son esbozadas más abajo y se ilustrarán en la parte experimental, que sólo está destinada a este fin, y no se debe utilizar para reducir el alcance de la presente invención como comprenderá una persona experta en la técnica.
Los constructos, en particular pIG.E1A.E1B, se utilizarán para transfectar células humanas diploides, tales como Retinoblastos Embriónicos Humanos (HER), células de Riñón Embriónico Humano (HEK), y células de Pulmón Embriónico Humano (HEL). Las células transfectadas serán seleccionadas por el fenotipo transformado (formación de foco) y sometidas a ensayo en cuanto a su capacidad para soportar la propagación de adenovirus recombinantes con E1 suprimida, tal como IG.Ad.MLPI.TK. Tales líneas celulares se utilizarán para la generación y producción (a gran escala) de adenovirus recombinantes con E1 suprimida. Tales células, infectadas con adenovirus recombinante también están destinadas a ser utilizadas in vivo en forma de un productor local de adenovirus recombinante, v.g. para el tratamiento de tumores sólidos.
Se utilizan células 911 para la titulación, generación y producción de vectores de adenovirus recombinantes (Fallaux y col., 1996).
Las células HER transfectadas con pIG.E1A.E1B han dado como resultado 7 clones independientes (células denominadas PER). Estos clones se utilizan para la producción de vectores de adenovirus con E1 suprimida (incluyendo vectores de adenovirus no solapantes) o con E1 defectuosa y proporcionan la base para la introducción
v.g. de constructos E2B o E2A (v.g. ts125E2A, ver más abajo), E4, etc., que permitirán la propagación de vectores de adenovirus que tengan mutaciones v.g. E2A o E4.
Todas las líneas celulares diploides transformadas pueden ser utilizadas como base para la generación de líneas de empaquetamiento de "la siguiente generación", que soporten la propagación de los adenovirus recombinantes carentes de E1, que también portan deleciones en otros genes, tales como E2A y E4. Por otra parte, proporcionarán la base para la generación de vectores adenovirales mínimos como se describe aquí.
Se utilizan y se utilizarán células de empaquetamiento que expresen secuencias E2A para la generación y producción (a gran escala) de adenovirus recombinante con E2A suprimida.
Las líneas celulares de empaquetamiento de adenovirus humano recién generadas (E2A o ts125E2A; E1A + E2A; E1A + E1B + E2A; E1A + E2A/TS125; E1A + E1B + E2A/TS125) son y serán utilizadas para la generación y producción (a gran escala) de vectores adenovirus recombinante con E2A suprimida. Además, serán aplicadas in vivo para la producción local del virus recombinante, como se describe para las células diploides (ver arriba).
Los vectores de adenovirus recién desarrollados que albergan una deleción de E1 de los nt. 459-3.510 serán utilizados para el propósito de la transferencia génica. Estos vectores también son la base para el desarrollo de vectores de adenovirus sometidos a deleción adicionalmente que son mutados v.g. para E2A, E2B o E4.
SECCION EXPERIMENTAL
1. Línea celular 911
Los autores de la presente invención generaron una línea celular que alberga las secuencias E1 del adenovirus de tipo 5, susceptible de complementar en trans un adenovirus recombinante con E1 suprimida (Fallaux y col., 1996).
Esta línea celular fue obtenida mediante transfección de retinoblastos embriónicos humanos diploides (HER) con pAd5XhoIC, que contiene los nt. 80 – 5.788 de Ad5; uno de los transformantes resultantes fue denominado 911. Se ha demostrado que esta línea celular es muy útil en la propagación de adenovirus recombinantes carentes de E1. Se encontró que era superior a las células 293. A diferencia de las células 293, las células 911 carecen de un fenotipo totalmente transformado, que muy probablemente es la causa del mejor funcionamiento como línea de empaquetamiento de adenovirus:
se pueden realizar más rápido los análisis en placa (4 -5 días en lugar de 8 -14 días en 293),
las monocapas de células 911 sobreviven mejor bajo una cubierta de agar de lo requerido para los análisis en
placa, superior amplificación de los vectores con E1 suprimida.
Además, a diferencia de las células 293 que fueron transfectadas con ADN adenoviral sometido a cizalla, las células 911 fueron transfectadas utilizando un constructo definido. Las eficacias de transfección de las células 911 son comparables a las de 293.
Nuevos constructos de empaquetamiento.
Las secuencias de adenovirus derivan o bien de pAd5.SalB, conteniendo los nt. 80 – 9.460 del adenovirus de tipo 5 humano (Bernards y col., 1983) o bien de ADN de Ad5 de tipo salvaje.
PAd5.SalB fue digerido con SalI y XhoI y el fragmento grande fue religado y este nuevo clon fue denominado pAd5.X/S.
El constructo pTN (construido por el Dr. R. Vogels, IntroGene, Holanda) fue utilizado como fuente de promotor PGK humano y de gen NEO.
Promotor PGK Humano y gen NEOR.
La transcripción de las secuencias de E1A en los nuevos constructos de empaquetamiento es dirigida por el promotor PGK humano (Michelson y col., 1983; Singer-Sam y col., 1984), derivado del plásmido pTN (donación de
R. Vogels), que utiliza pUC119 (Vieira y Messing, 1987) como esqueleto. Este plásmido también fue utilizado como una fuente de gen NEO fusionado a la señal de poli-adenilación del Virus de la Hepatitis B (HBV).
Con el fin de remplazar las secuencias de E1 de Ad5 (ITR, origen de replicación y señal de empaquetamiento) por secuencias heterólogas los autores de la presente invención amplificaron las secuencias E1 (nt. 459 a nt. 960) de Ad5 mediante PCR, utilizando los cebadores Ea1 y Ea2 (ver la Tabla I). El producto de la PCR resultante fue digerido con ClaI y ligado en Bluescript (Stratagene), predigerido con ClaI y EcoRV, dando como resultado el constructo pBS.PCRI.
El vector pTN fue digerido con las enzimas de restricción EcoRI (parcialmente) y ScaI, y el fragmento de ADN que contenía las secuencias promotoras de PGK fue ligado en PBS.PCRI digerido con ScaI y EcoRI. El constructo resultante PBS.PGK.PCRI contiene el promotor PGK humano conectado operativamente a las secuencias E1 de Ad5 desde el nt. 459 al nt. 916.
pIG.E1A.E1B.X fue elaborado remplazando el fragmento ScaI-BspEI de pAT-X/S por el correspondiente fragmento de PBS.PGK.PCRI (conteniendo el promotor PGK conectado a las secuencias de E1A).
pIG.E1A.E1B.X contiene las secuencias codificadoras de E1A y E1B bajo la dirección del promotor PGK.
Como las secuencias Ad5 desde el nt. 459 al nt. 5.788 de Ad5 están presentes en este constructo, también la
proteína pIX del adenovirus está codificada por este plásmido.
Con el fin de introducir el promotor de E1B completo y fusionar este promotor de tal manera que el codón AUG de E1B de 21 kd funcione exactamente como el codón AUG de NEOR, los autores de la presente invención amplificaron el promotor E1B utilizando los cebadores Ea3 y Ep2, donde el cebador Ep2 introduce un sitio NcoI en el fragmento de la PCR. El fragmento de la PCR resultante, denominado PCRII, fue digerido con HpaI y NcoI y ligado en pAT-X/S, que había sido predigerido con HpaI y con NcoI. El plásmido resultante fue denominado pAT-X/S-PCR2. El fragmento NcoI-StuI de pTN, que contenía el gen NEO y parte de la señal de poli-adenilación del Virus de la Hepatitis B (HBV), fue clonado en pAT-X/S-PCR2 (digerido con NcoI y NruI). El constructo resultante: pAT-PCR2-NEO. La señal de poliadenilación fue completada remplazando el fragmento ScaI-SalI de pAT-PCR2-NEO por el correspondiente fragmento de pTN (dando como resultado pAT.PCR2-NEO.p(A)). El ScaI-XbaI de pAT.PCR2.NEO.p
(A) fue remplazado por el correspondiente fragmento de pIG.E1A.E1B-X, conteniendo el promotor PGK conectado a los genes E1A.
El constructo resultante fue denominado pIG.E1A.NEO, y contiene por lo tanto las secuencias E1 de Ad5 (nt. 459 a nt. 1.713) bajo el control del promotor PGK humano.
PIG.E1A.E1B fue elaborado amplificando las secuencias que codificaban los aminoácidos N-terminales de E1B de 55 kd utilizando los cebadores Eb1 y Eb2 (introduce un sitio XhoI). El fragmento de la PCR resultante fue digerido con BglII y clonado en BglII/NruI de pAT-X/S, obteniéndose de ese modo pAT-PCR3.
PIG.E1A.E1B fue construido introduciendo las secuencias poli(A) de HBV de pIG.E1A.NEO aguas abajo de las secuencias de E1B de pAT-PCR3 mediante intercambio del fragmento XbaI -SalI de pIG.E1A.NEO y el fragmento XbaI XhoI de pAT.PCR3.
PIG.E1A.E1B contiene los nt. 459 a nt. 3.510 de Ad5, que codifican las proteínas E1A y E1B. Las secuencias de E1B están terminadas en el aceptor de empalme en el nt. 3.511. No se encuentran secuencias pIX en este constructo.
PIG.NEO fue generado clonando el fragmento HpaI -ScaI de pIG.E1A.NEO, conteniendo el gen NEO bajo el control del promotor E1B de Ad5, en pBS digerido con EcoRV y ScaI.
Este constructo se utiliza cuando se transfectan células establecidas con constructos E1A.E1B y se requiere selección NEO. Debido a que la expresión de NEO está dirigida por el promotor E1B, se espera que las células resistentes a NEO expresen simultáneamente E1A, lo que también es ventajoso para mantener elevados niveles de expresión de E1A durante el cultivo a largo plazo de las células.
La integridad de los constructos pIG.E1A.NEO, pIG.E1A.E1B.X y pIG.E1A.E1B fue evaluada mediante mapeo con enzimas de restricción; además, partes de los constructos que habían sido obtenidos mediante análisis PCR fueron confirmados mediante análisis de la secuencia. No se encontraron cambios en la secuencia de nucleótidos.
Los constructos fueron transfectados en células BRK (Baby Rat Kidney) primarias y sometidos a ensayo en cuanto a su capacidad para inmortalizar (pIG.E1A.NEO) o transformar completamente (pAd5.XhoIC, pIG.E1A.E1B.X y pIG.E1A.E1B) estas células.
Los riñones de ratas WAG-Rij de 6 días de edad fueron aislados, homogeneizados y tratados con tripsina. Se transfectaron placas subconfluentes (5 cm de diámetro) de los cultivos de células BRK con 1 o 5 !g de pIG.NEO, pIG.E1A.NEO, pIG.E1A.E1B, pIG.E1A.E1B.X, pAd5XhoIC, o con pIG.E1A.NEO junto con PDC26 (Van der Elsen y col., 1983), que portaban el gen Ad5.E1B bajo el control del promotor temprano de SV40. Tres semanas después de la transfección, cuando los focos eran visibles, las placas fueron fijadas, teñidas con Giemsa y sometidas a recuento de los focos.
Una visión de conjunto de los constructos de empaquetamiento de adenovirus generados, y de su capacidad para tranformar BRK, se presenta en la Fig. 6. Los resultados indican que los constructos pIG.E1A.E1B y pIG.E1A.E1B.X son capaces de transformar células BRK de una manera dependiente de la dosis. La eficacia de la transformación es similar para ambos constructos y es comparable a la encontrada con el constructo que se utilizaba para elaborar las células 911, esto es pAd5.XhoIC.
Como se esperaba, pIG.E1A.NEO apenas era capaz de inmortalizar BRK. No obstante, la co-transfección de un constructo de expresión de E1B (PDC26) daba como resultado un incremento significativo del número de transformantes (18 versus 1), indicando que la E1A codificada por pIG.E1A.NEO es funcional.
Los autores de la presente invención concluyen por lo tanto, que los constructos de empaquetamiento recién generados son adecuados para la generación de nuevas líneas de empaquetamiento de adenovirus.
Generación de líneas celulares con nuevos constructos de empaquetamiento. Líneas celulares y cultivo celular
Se hicieron crecer células de carcinoma bronquial A549 humano (Shapiro y col., 1978), retinoblastos embriónicos humanos (HER), células de riñón embriónico humano (HEK) transformadas con Ad5-E1 (293; Graham y col., 1977) y células HER transformadas con Ad5 (911, Fallaux y col, 1996)) y células PER en Medio de Eagle Modificado de Dulbecco (DMEM) suplementado con Suero de Ternera Fetal al 10% (FCS) y antibióticos en una atmósfera con el 5% de CO2a 37°C. El medio de cultivo, los reactivos y los sueros fueron adquiridos de Gibco Laboratories (Grand Island, NY). Los plásticos para el cultivo fueron adquiridos de Greiner (Nürtingen, Alemania) y Corning (Corning, NY).
La construcción de los vectores adenovirales IG.Ad.MLP.nls.lacZ, IG.Ad.MLP.luc, IG.Ad.MLP.TK e IG.Ad.CMV.TK se describe con detalle en la solicitud de patente EP 0707071.
El vector adenoviral IG.Ad.MLP.nls.lacZ contiene el gen lacZ de E. Coli, que codifica la 1-galactosidasa, bajo el control del promotor tardío principal (MLP) de Ad2. IG.Ad.MLP.luc contiene el gen de la luciferasa de luciérnaga dirigido por el MLP de Ad2. Los vectores adenovirales IG.Ad.MLP.TK e IG.Ad.CMV.TK contienen el gen de la timidina quinasa (TK) del Virus Herpes Simplex bajo el control del MLP de Ad2 y el intensificador/promotor de Citomegalovirus (CMV), respectivamente.
Todas las transfecciones se realizaron mediante ADN precipitado con fosfato de calcio (Graham y Van Der Eb, 1973) con el GIBCO Calcium Phosphate Transfection System (GIBCO BRL Life Technologies Inc., Gaithersburg, USA), según el protocolo de los fabricantes.
Se lavaron con PBS cultivos subconfluentes de células 293, 911 y A549 y PER transformadas con Ad5-E1 que crecían exponencialmente y se rasparon en tampón Fos-RIPA (Tris 10 mM (pH 7,5), NaCl 150 mM, NP40 al 1%, dodecilsulfato de sodio (SDS) al 0,1%, NA-DOC al 1%, fluoruro de fenilmetilsulfonilo (PMSF) 0,5 M, inhibidor de tripsina 0,5 mM, NaF 50 mM y vanadato de sodio 1 mM). Al cabo de 10 minutos a la temperatura ambiente, los productos lisados fueron aclarados por centrifugación. Las concentraciones de proteína fueron medidas con el estuche de análisis de proteínas de Biorad, y se cargaron 25 !g de proteína celular total sobre un gel de SDS-PAA al 12,5%. Tras la electroforesis, las proteínas fueron transferidas a nitrocelulosa (1h a 300 mA). Los patrones teñidos previamente (Sigma, USA) se desarrollaron en paralelo. Los filtros fueron bloqueados con seralbúmina bovina (BSA) al 1% en TBST (Tris 10 mM, pH 8, NaCl 15 mM, y Tween-20 al 0,05%) durante una hora. Los primeros anticuerpos fueron el anticuerpo A1C6 anti-Ad5-E1B de 55 kDA monoclonal de ratón (Zantema y col., no publicado), el anticuerpo C1G11 anti-Ad5-E1B de 221 kDA monoclonal de rata (Zantema y col., 1985). El segundo anticuerpo era un anticuerpo anti-ratón de cabra marcado con peroxidasa de rábano picante (Promega). Las señales fueron visualizadas mediante aumento de la quimioluminiscencia (Amersham Corp. UK).
Se aisló ADN de elevado peso molecular y se digirieron 10 !g por completo y se fraccionaron sobre un gel de agarosa al 0,7%. Se realizó la transferencia Southern a Hybond N+ (Amersham, UK) con una solución de transferencia de NaOH 0,4 M, NaCl 0,6 M (Church and Gilbert, 1984). La hibridación se realizó con un fragmento SspI-HindIII de 2.463 nt. De pAd5.SalB (Bernards y col., 1983). Este fragmento consta de los pb. 342-2.805 de pAd5. El fragmento fue radiomarcado con dCTP a-P32 con el uso de cebadores hexanucleotídicos al azar y ADN polimerasa de Klenow. Las transferencias Southern fueron expuestas a una película Kodak XAR-5 a -80°C y a una pantalla Phospho-Imager que fue analizada mediante el soporte lógico B & L Systems Molecular Dynamics.
Se generaron líneas celulares de carcinoma bronquial humano A549 transformadas con Ad5-E1 mediante transfección con pIG.E1A.NEO y selección en cuanto a la resistencia a G418. Se establecieron treinta y un clones resistentes a G418. La co-transfección de pIG.E1A.E1B con pIG.NEO rindió siete líneas celulares resistentes a G418.
PER
Se generaron células de retina embriónica humana (HER) transformadas con Ad5-E1 mediante transfección de células HER primarias con el plásmido pIG.E1A.E1B. Las líneas celulares transformadas fueron establecidas a partir de focos bien separados. Los autores de la presente invención fueron capaces de establecer siete líneas celulares clonales, que denominaron PER.C1, PER.C3, PER.C4, PER.C5, PER.C6, PER.C8 y PER.C9.
Uno de los clones PER, a saber PER.C6, ha sido consignado en la ECACC con el número 96022940.
La expresión de las proteínas E1A y E1B de 55 kDa y 21 kDa de Ad5 en las células A549 y PER establecidas fue estudiada por medio de transferencia Western, utilizando anticuerpos monoclonales (mAb). El mAb M73 reconoce los productos de E1A, mientras los mAb AIC6 y ClGll están dirigidos contra las proteínas E1B de 55 kDa y de 21 kDa, respectivamente.
Los anticuerpos no reconocían las proteínas en los extractos de las células A549 parentales o de las células HER primarias (datos no mostrados). Ninguno de los clones de A549 que se generaban por co-transfección de pIG.NEO y pIG.E1A.E1B expresaba niveles detectables de las proteínas E1A o E1B (no mostrados). Algunos de los clones de A549 que se generaban por transfección con pIG.E1A.NEO expresaban las proteínas E1A de Ad5 (Fig. 7), pero los niveles eran muy inferiores a los detectados en los productos lisados de proteína de las células 293. Los niveles de E1A en estado estacionario detectados en los extractos de proteína de las células PER eran muy superiores a los detectados en los extractos de células derivadas de A549. Todas las líneas celulares PER expresaban niveles similares de proteínas E1A (Fig. 7). La expresión de las proteínas E1B, concretamente en el caso de E1B de 55 kDa, era más variable. En comparación con 911 y 293, la mayoría de los clones de PER expresan elevados niveles de E1B de 55 kDa y de 21 kDa. El nivel en estado estacionario de E1B de 55 kDa en PER-C3 era el más alto. Ninguno de los clones de PER perdía expresión de los genes E1 de Ad5 tras el paso seriado de las células (no mostrado). Los autores de la presente invención encontraron que el nivel de expresión de E1 en las células PER permanecía estable durante al menos 100 duplicaciones de la población. Los autores de la presente invención decidieron caracterizar los clones de PER con más detalle.
Para estudiar la disposición de las secuencias que codifican E1 de Ad5 en los clones de PER se realizaron análisis Southern. El ADN celular fue extraído de todos los clones de PER, y de las células 293 y 911. El ADN fue digerido con HindIII, que corta una vez en la región E1 de Ad5. La hibridación de Southern del ADN digerido con HindIII, utilizando una sonda específica para E1 de Ad5 radiomarcada reveló la presencia de numerosas copias integradas de pIG.E1A.E1B en el genoma de los clones de PER. La Figura 8 muestra el patrón de distribución de las secuencias de E1 en el ADN de elevado peso molecular de las diferentes líneas celulares PER. Las copias se concentran en una única banda, lo que sugiere que están integradas como repeticiones en tándem. En el caso de PER.C3, C5, C6 y C9 los autores de la presente invención encontraron bandas de hibridación adicionales de bajo peso molecular que indican la presencia de copias truncadas de pIG.E1A.E1B. El número de copias fue determinado utilizando un Phospho-Imager. Los autores de la presente invención estimaron que PER.C1, C3, C4, C5, C6, C8 y C9 contienen 2, 88, 5,4, 5, 5 y 3 copias de la región codificadora de E1 de Ad5, respectivamente, y que las células 911 y 293 contienen 1 y 4 copias de las secuencias E1 de Ad5, respectivamente.
Se generan adenovectores recombinantes mediante co-transfección de plásmidos adaptadores y del fragmento ClaI grande de Ad5 en células 293 (ver la solicitud de patente 0707071). El ADN del virus recombinante se forma mediante recombinación homóloga entre las secuencias virales homólogas que están presentes en el plásmido y el ADN de adenovirus. La eficacia de este método, así como la de las estrategias alternativas, depende enormemente de la transfectabilidad de las células coadyuvantes. Por lo tanto, los autores de la presente invención compararon las eficacias de transfección de algunos de los clones de PER con células 911, utilizando el gen lacZ que codifica a 1galactosidasa de E. Coli como informador (Fig. 9).
Los rendimientos de adenovirus recombinantes obtenidos tras la inoculación de 293, 911, PER.C3, PER.C5 y PER.C6 con diferentes vectores de adenovirus se presentan en la Tabla II.
Los resultados indican que los rendimientos obtenidos en las células PER son al menos tan elevados como los obtenidos en líneas celulares existentes.
Además, los rendimientos del vector de adenovirus novedoso IG.Ad.MLPI.TK son similares o superiores a los rendimientos obtenidos para otros vectores virales en todas las líneas celulares sometidas a ensayo.
Generación de nuevos vectores de adenovirus (Fig. 10)
Los vectores de adenovirus recombinantes utilizados (ver la solicitud de patente 0707071) tienen suprimidas las secuencias E1 de 459 al nt. 3.328.
Como el constructo pIG.E1A.E1B contiene las secuencia de los nt. 459 a 3.510 de Ad5 existe un solapamiento de la secuencia de 183 nt. Entre las secuencias E1B del constructo de empaquetamiento pIG.E1A.E1B y los adenovirus recombinantes, tales como v.g. IG.Ad.MLP.TK. Las secuencias solapantes son suprimidas de los nuevos vectores de adenovirus. Además, las secuencias no codificadoras derivadas de lacZ, que están presentes en los constructos originales, también fueron suprimidas. Esto se logró (ver la Fig. 19) mediante amplificación por PCR de las secuencias poli(A) de SV40 de pMLP.TK utilizando los cebadores SV40-1 (introduce un sitio BamHI) y SV40-2 (introduce un sitio BglII). Además, las secuencias de Ad5 presentes en este constructo fueron amplificadas desde el nt. 2.496 (Ad4, introduce un sitio BglII) a nt. 2.779 (Ad5-2). Ambos fragmentos de PCR fueron digeridos con BglII y ligados. El producto de ligadura fue amplificado mediante PCR utilizando los cebadores SV40-1 y Ad5-2. El producto de la PCR obtenido fue cortado con BamHI y AflII y fue ligado en pMLP.TK digerido previamente con las mismas enzimas. El constructo resultante, denominado pMLPI.TK, contiene una deleción en las secuencias E1 del adenovirus desde el nt. 459 al nt. 3.510.
La combinación del nuevo constructo de empaquetamiento pIG.E1A.E1B y el adenovirus recombinante pMLPI.TK, que no tienen ningún solapamiento de secuencia, se presenta en la Figura 11. En esta figura, también se presenta la situación original, donde se indica el solapamiento de la secuencia.
La ausencia de secuencias solapantes entre pIG.E1A.E1B y pMLPI.TK (Fig. 11a) excluye la posibilidad de recombinación homóloga entre el constructo de empaquetamiento y el virus recombinante, y por lo tanto es una mejora significativa para la producción de adenovirus recombinante en comparación con la situación original.
En la Fig. 11b se representa la situación para pIG.E1A.NEO e IG.Ad.MLPI.TK. Se espera que cuando pIG.E1A.NEO es transfectado a células establecidas, sea suficiente para soportar la propagación del adenovirus recombinante con E1 suprimida. Esta combinación no tiene ningún solapamiento de secuencia, evitando la generación de ARC mediante recombinación homóloga. Además, este sistema de empaquetamiento conveniente permite la propagación de adenovirus recombinantes que tienen suprimidas sólo las secuencias E1A y no las secuencias E1B. Los adenovirus recombinantes que expresan E1B en ausencia de E1A son atractivos, ya que la proteína E1B, en particular E1B de 19 kD, es capaz de evitar la lisis de las células humanas infectadas mediante el Factor de Necrosis Tumoral (TNF) (Gooding y col., 1991).
Generación del adenovirus recombinante derivado de pMLPI.TK.
Se generó un adenovirus recombinante mediante co-transfección de células 293 con ADN de pMLPI.TK linealizado con SalI y ADN wt de Ad5 linealizado con ClaI. El procedimiento se representa esquemáticamente en la Fig. 12.

Claims (6)
- REIVINDICACIONES1. Un método para producir un adenovirus recombinante en una célula, que comprende: a) proporcionar una célula depositada con el núm. 96022940 en el ECACC, o uno de sus derivados; b) introducir en dicha célula una molécula de ácido nucleico recombinante basada en o derivada de un adenovirus, teniendo dicha molécula de ácido nucleico recombinante una señal de encapsidación funcional y al menos una Repetición Terminal Invertida funcional o uno de sus fragmentos o derivados funcionales, donde dicha molécula de ácido nucleico recombinante y dicha célula comprenden entre las dos todos los elementos que son necesarios para generar un adenovirus recombinante que comprende dicha molécula de ácido nucleico recombinante, y donde dicha molécula de ácido nucleico recombinante no tiene secuencias solapantes con las secuencias de ácido nucleico presentes en dicha célula que permite la recombinación homóloga conduciendo a virus de replicación competente en dicha célula; y c) producir el adenovirus recombinante en dicha célula.

- Un método de acuerdo con la reivindicación 1, donde dicha molécula de ácido nucleico recombinante está en forma lineal y comprende una Repetición Terminal Invertida en o cerca en ambos extremos.

- Un método de acuerdo con la reivindicación 1, donde dicha molécula de ácido nucleico recombinante se forma en el interior de la célula de empaquetamiento mediante recombinación entre las moléculas de ácido nucleico que comprenden diferentes partes del genoma del adenovirus.

- Un método de acuerdo con la reivindicación 1, donde dicha molécula de ácido nucleico recombinante se introduce en dicha célula de empaquetamiento en forma de un adenovirus recombinante que comprende dicha molécula de ácido nucleico recombinante.

- Un método de acuerdo con una cualquiera de las reivindicaciones 1-4, donde dicha molécula de ácido nucleico recombinante tiene una deleción de al menos los nucleótidos 459-3510 de la región E1.

- 6.
- El uso de una célula de empaquetamiento depositada con el núm. 96022940 en el ECACC, o uno de sus derivados para la producción de un adenovirus recombinante.
