Opfindelsen angår en fremgangsmåde til isolering af insulinderivater med basisk karakter fra proteinblandinger.
Ved fremstillingen af human insulin ved gentekniske metoder sker biosyntesen af insulinet via et precursor-5 eller forløbermolekyle, proinsulinet. I dette er B-kæden forbundet med A-kæden via C-peptidet (= "Connecting Peptide") . Ved den gentekniske produktion ved hjælp af Escherichia coli får man undertiden først et fusionsprotein, i hvilket der foran proinsulinet desuden er koblet et fremmed 10 protein, eksempelvis β-galactosidase.
Dette fremmede protein skal før den videre oparbejdning først fraspaltes, eksempelvis ved behandling med cyan-halogenid (jf. DE offentliggørelsesskrift nr. 3.440.988).
Efter cyanhalogenidfraspaltningen omdannes cysteingrupper 15 ved sulfitolyse (behandling med natriumsulfit og natrium-tetrathionat) til deres S-sulfonatform. Molekylet kan ud fra den form ved hjælp af reduktiv tilbagefoldning (dvs. ved behandling med mercaptoethanol i basisk opløsning) ved korrekt dannelse af dets disulfidbroer overføres til dets 20 naturlige rumstruktur. Proinsulinet eller præproinsulinet (- derivat af proinsulinet, forstavelsen "præ" refererer til en eller flere yderligere aminosyrer ved proinsulinets N-terminal) omdannes ved enzymatisk spaltning (eksempelvis med trypsin) til en blanding af spaltningsprodukter, som 25 indeholder en insulinforløber, insulin-Arg-B31-B32 og C-peptidet. Foruden disse dannes der desuden nogle biprodukter, såsom insulin-Des-Thr-B30, insulin-Arg-B31 samt ufuldstændigt spaltede mellemprodukter.
De nævnte insulinderivater og forbindelser, som dannes 3 0 i forstadier ved den kemiske behandling af udgangsmaterialet, skal separeres. I den forbindelse er det særligt vanskeligt at fraskille insulinderivater med basisk karakter, som udviser derivatiseringer i nærværelse af aminosyrer, som efter færdigdannelse af den tertiære struktur ligger i det indre 35 af molekylet.

Dette viser sig i et sammenligningsforsøg (jf. del B) :
Den ionbytterfremgangsmåde, som er beskrevet i DE patentskrift nr. 2.629.568, og som er udviklet til rensning 5 af insulin, har den særegenhed, at der til undgåelse af proteinaggregationer til elueringsvæsken sættes et ikke--ionisk tensid. Denne fremgangsmåde, som er velegnet til rensning af insuliner, fører ved forsøg på separation og isolering af basiske proteiner, som eksempelvis fås ved 10 tryptisk spaltning af proinsulin af genteknisk oprindelse, til en fuldstændig utilstrækkelig separation. Dette tydeliggøres ved den tilsvarende elueringsprofil (jf. del B) .
Man erkender, at toppene for de enkelte peptider flyder sammen.
15 Man kender ligeledes allerede fremgangsmåder, hvorved man skal kunne opnå separation af de nævnte proinsulin-spaltningsprodukter .
I Journ. of Biol. Chem. 24£, 1365-1374 (1971) beretter Steiner et al. om en fremgangsmåde til isolering af C-peptid 20 ud fra en proinsulinspaltningsblanding. Ved denne fremgangsmåde (der er ikke angivet udbytter) chromatograferer man ved hjælp af en svagt sur carboxymethylgruppeholdig (CM) kationbytter på cellulosebasis. Til belastnings-og eluerings-opløsningen sættes der i den forbindelse 7 mol urinstof pr.
25 liter for at undgå en aggregation af proteinerne. Tilstedeværelsen af på den måde høje urinstofkoncentrationer er uheldig, fordi den fører til derivatisering af proteiner, frem for alt til carbamoylering af frie aminogrupper. I Protein Engineering, bd. 1, nr. 3, 205-213 (1987) beskriver 30 Markussen et al. en ionbytterchromatografifremgangsmåde ved hjælp af diethyl-2-hydroxypropylaminoethyl-holdige (QAE) anionbyttere på en matrix af tredimensionelt tværbundet polysaccharid-netværk.
Ved denne fremgangsmåde - for anionbytterchromato-35 grafien angives der ingen udbytter - arbejder man til in-hibering af proteinaggregationer i en 60%'s ethanolopløsning.

En ulempe ved sådanne koncentrerede ethanolopløsninger består i de nødvendige sikkerhedsforanstaltninger, som især er nødvendige ved arbejde med koncentrerede organiske opløsningsmidler i teknisk målestok. På den anden side forekommer 5 der, som efterarbejder har vist det, denatureringer af proteinerne ved de nævnte alkoholkoncentrationer.
I bestræbelserne på at tilvejebringe en bedre separations- og isoleringsfremgangsmåde for insulinderivater med basisk karakter ud fra proteinblandinger - indeholdende 10 sådanne insulinderivater har det overraskende vist sig, at dette formål kan opnås ved chromatografi af proteinblandingerne på en stærkt sur kat ionbyt ter og eluering ved hjælp af en saltgradient med kun en relativt ubetydelig mængde alkanol. Det er især overraskende, at man derved i forhold 15 til teknikkens stade såvel kan forbedre separationseffekten samt i vid udstrækning undgå de yderligere derivatiseringer af proteinerne under separationen. Det er bemærkelsesværdigt, at man endog kan fraskille derivater, der er dannede ved forbehandlingen af proinsulinet og har derivatiseringer i det 20 indre af de i tertiærstrukturen foreliggende proteiner.
Der iagttages ikke aggregationer af de proteiner, der skal separeres. Genanvendeligheden af ionbytterne bereder ingen vanskeligheder.
Opfindelsen angår følgelig en fremgangsmåde til isole-25 ring af insulinderivater med basisk karakter fra proteinblandinger, som indeholder sådanne insulinderivater, og som er tilvejebragt ved enzymatisk spaltning af proinsulin og/eller dets derivater af naturlig, halvsyntetisk eller genteknisk oprindelse, ved ladning af en ionbytter med pro-30 teinblandingen og eluering, og denne fremgangsmåde er ejendommelig ved, at man som ionbytter anvender en stærkt sur kationbytter, idet ladningen deraf gennemføres ved en pH-værdi mellem ca. 2,5 og ca. 5,5, fortrinsvis mellem ca. 3,5 og ca. 4,0, og at man gennemfører elueringen ved hjælp af 35 en blanding af vand og -alkanol, som indeholder fra ca.
10 til ca. 50%, fortrinsvis fra ca. 20 til ca. 40%, især 4 DK 173502 B1 ca. 30% alkanol, idet man indstiller elueringsopløsningens pH-værdi på fra ca. 3,5 til ca. 4,0 og eluerer med en ammonium- eller alkalimetalsaltgradient, der ligger mellem ca. 0 og ca. 1 mol pr. liter, især mellem ca. 0,15 og ca.
5 0,35 mol pr. liter.
Ved hjælp af den her omhandlede fremgangsmåde kan man ud fra spaltningsblandinger af vilkårlige proinsuliner, eller af derivater deraf, såsom af abepræproinsulin, isolere insulinderivaterne med basisk karakter i høje udbytter. Som 10 ionbyttere kommer i princippet alle stærkt sure kat ionbyttere på tale. Man foretrækker stærkt sure kationbyttere, hvis funktionelle gruppe er sulfogruppen, især sulfopropylgruppen -CH2-CH2-CH2-SO3H, eksempelvis på en matrix af hydrofile hydroxygruppeholdige vinylpolymere - eksempelvis "Fractogel 15 TSK", producent Fa. Merck. Darmstadt), acryl-copolymere (såsom "SP Trisacryl M", producent Réactifs IBF, Villeneuve-la-Garenne, Frankrig) eller geler med et indhold af tværbundet agarose (såsom "S-Sepharose'r, producent Fa. Pharmacia, Upsala, Sverige). Især foretrækkes "S-Sepharose".
20 Forud for ladningen af den stærkt sure kationbytter med blandingen af spalteprodukter skal denne nyækvilibreres med en pufferopløsning. Denne pufferopløsning består af en blanding af vand og Ci_4-alkanol med et alkanolindhold på fortrinsvis fra ca. 10 til ca. 50 volumenprocent, især for-25 trinsvis fra ca. 20 til ca. 40 volumenprocent, især på ca.
30 volumenprocent. Foretrukne alkanoler er ethanol og iso-propanol, især isopropanol. Yderligere tilsætningsstoffer, der kan sættes til pufferopløsningen, er eksempelvis salt, fortrinsvis fysiologisk acceptabelt mineralsalt, en eller 30 flere vilkårlige organiske syrer, fortrinsvis mælkesyre, en base, fortrinsvis NaOH, og/eller konserveringsmidler. pH-værdien for pufferopløsningen ligger mellem ca. 2,5 og ca.
5,5, især fortrinsvis mellem ca. 3,5 og ca. 4,0.
Ladningen af den stærkt sure kationbytter kan ske 35 ved, at man opløser blandingen af spaltningsprodukter i en pufferopløsning - med den ovenfor beskrevne sammensætning 5 DK 173502 B1 og den ovenfor beskrevne pH-værdi og bringer den således tilvejebragte opløsning i kontakt med den stærkt sure kat-ionbytter.
Elueringsopløsningen, som i princippet kan have en 5 lignende sammensætning som den ovenfor beskrevne pufferopløsning, har en pH-værdi fra 3,5 til 4,0. Især egnet er en elueringsfremgangsmåde, ved hvilken elueringsopløsningen har en tidsmæssig saltkoncentrationsgradient med fortrinsvis lineært forløb. Denne koncentrationsgradient kan eksempelvis 10 pålægges ved, at der i begyndelsen af elueringen foreligger en ubetydelig saltkoncentration (i grænsetilfældet gående mod 0) i elueringsopløsningen og ved, at saltkoncentrationen øges under elueringsforløbet. På denne måde kan man opnå en særlig virkningsfuld separation af proteinblandingen. En 15 foretrukken salt-koncentrationsgradient varierer fra tæt ved 0 mol salt/liter (i begyndelsen af elueringen) til ca.
1 mol salt/liter (ved slutningen af elueringen), især fortrinsvis fra ca. 0,15 (ved begyndelsen af elueringen) til ca. 0,35 mol/liter (i slutningen af elueringen). Til salt-20 tilsætningen kommer mange organiske og uorganiske salte på tale. Man foretrækker fysiologisk acceptable salte, såsom ammonium- og alkalimetalsalte, især fortrinsvis natriumsalte, især natriumchlorid.
Den her omhandlede separationsfremgangsmåde kan gen-25 nemføres på forskellig måde. Gennemførelsen ved søjlefremgangsmåden eller ved batch-fremgangsmåden er at foretrække. Temperaturen, som ved ionbytterchromatografien fortrinsvis skal holdes konstant, kan varieres indenfor et bredt område.
Man foretrækker et temperaturinterval fra ca. -10 til ca.
30 50°C, især fra ca. 15 til ca. 25°C.
Ved hjælp af det følgende udførelseseksempel (A) skal opfindelsen belyses nærmere. Ved hjælp af den dertil hørende sammenligning (B) skal man vise overlegenheden af den her omhandlede fremgangsmåde i forhold til en fremgangs-35 måde til rensning af insulin ifølge teknikkens stade (DE patentskrift nr. 2.629.568).
6 DK 173502 B1 A) Udføringseksempel (ifølge opfindelsen) 6 liter "S-Sepharose" opslæmmes en en vandig pufferopløsning, som indeholder 50 mmol mælkesyre pr. liter, 30% isopropanol og 1 mol natriumchlorid pr.
5 liter, og hvis pH-værdi ligger på ca. 3,5, og fyldes i en søjle med en diameter på 10 cm op til en højde på ca. 80 cm. Søjlen ækvilibreres med 10 liter vandig startpufferopløsning (50 mmol mælkesyre pr. liter, 30% isopropanol, 0,15 mol natriumchlorid pr. liter 10 pH-værdi lig med ca. 3,5). 15 g krystalliseret spalt ningsblanding, som indeholder 6,2 g insulin-Arg-B31--32, 3 g insulin-Arg-B31 og insulin-Des-Thr-B30 samt ukendte spaltningsmellemprodukter samt derivater, dannet ved tryptisk spaltning af præprohumaninsulin, 15 opløses i 3 liter startpuffer og sættes til søjlen.
Derpå elueres der med en vandig opløsning indeholdende 50 mmol mælkesyre pr. liter og 30% isopropanol ved en pH-værdi på 3,5 (indstillet med NaOH) og en gradient fra 0,15 mol natriumchlorid pr. liter til 0,35 20 natriumchlorid pr. liter. Elueringsopløsningen har et rumfang på 2 x 20 liter. Gennemstrømningshastigheden er 1,5 liter pr. time. Fraktioner, hvis insulin--Arg-B31-32-indhold ifølge HPLC (høj tryks væske chroma-tografi) er over 90%, opsamles, fortyndes med vand i 25 forholdet 1:1 og fældes ved tilsætning af 10 ml 10%'s
ZnCl2-opløsning pr. liter opløsning og indstilling til pH-værdi lig med 6,8. Udbyttet er 5 g insulin--Arg-B31-32, hvilket svarer til et udbytte pr. trin, beregnet på insulin-Arg-B31-32, på 80%.
30 B) Sammenligning
Sammenligningseksempel 1 gennemføres ifølge DE-patent-skrift nr. 2.629.568 og eksempel II ifølge opfindelsen. I tabel I er fremgangsmådeparametrene og de 35 opnåede resultater stillet overfor hinanden. I fig.
1 er vist elueringsprof il en, optaget under elueringen, 7 DK 173502 B1 for sammenligningseksempel l, fig. 2 den samme for eksempel II ifølge opfindelsen. Man overfører absorptionen A i UV-området ved en bølgelængde på 278 nm (A27ø) mod elueringstiden eller fraktionsnumrene.
5 Elueringsprofilerne viser, at den her omhandlede fremgangsmåde fører til en bedre separation af de basiske proteiner end den ifølge DE-patentskrift nr. 2.629.568 gennemførte fremgangsmåde. Tilsvarende opnås der ved den her omhandlede fremgangsmåde lige-10 ledes et højere produktudbytte og en højere produkt renhed (jf. tabel I) .

Claims (4)
1. Fremgangsmåde til isolering af insulinderivater med basisk karakter fra proteinblandinger, som indeholder sådanne insulinderivater, og som er tilvejebragt ved en- 5 zymatisk spaltning af proinsulin og/eller dets derivater af naturlig, halvsyntetisk eller genteknisk oprindelse, ved ladning af en ionbytter med proteinblandingen og eluering, kendetegnet ved, at man som ionbytter anvender en stærkt sur kationbytter, idet ladningen deraf gennemføres 10 ved en pH-værdi mellem ca. 2,5 og ca. 5,5, fortrinsvis mellem ca. 3,5 og ca. 4,0, og at man gennemfører elueringen ved hjælp af en blanding af vand og Ci_4-alkanol, som indeholder fra ca. 10 til ca. 50%, fortrinsvis fra ca. 20 til ca. 40%, især ca. 30% alkanol, idet man indstiller elueringsopløsnin-15 gens pH-værdi på fra ca. 3,5 til ca. 4,0 og eluerer med en ammonium- eller alkalimetalsaltgradient, der ligger mellem ca. 0 og ca. 1 mol pr. liter, især mellem ca. 0,15 og ca. 0,35 mol pr. liter.
2. Fremgangsmåde ifølge krav 1, kendete g-20 net ved, at man som stærkt sur kationbytter anvender en kationbytter med sulfogrupper, især med sulfopropylgrupper.
3. Fremgangsmåde ifølge krav 1 eller 2, kendetegnet ved, at den til eluering anvendte blanding af vand og C1-4-alkanol som C^_4-alkanol indeholder ethanol 25 eller isopropanol, især isopropanol.
4. Fremgangsmåde ifølge et eller flere af kravene 1--3, kendetegnet ved, at ladnings- og eluerings-opløsningen indeholder en puffersubstans, fortrinsvis på basis af en organisk syre, især mælkesyre. 30
