Description
- Die Erfindung betrifft eine pharmazeutische Formulierung, insbesondere eine Tablette, die ein neues Polymorph eines Makrolidantibiotikums enthält.
- Dirithromycin ist ein Makrolidantibiotikum, das sich von Erythromycin ableitet. Das Antibiotikum ist in US-A 4 048 306 beschrieben und hat den chemischen Namen Erythromycin, 9-Desoxy-11-desoxy-9,11-[imino[2- (2-methoxyethoxy)ethyliden]-oxy-, [9S, 16R]. Das durch das in US-A 4 048 306 beschriebene Verfahren hergestellte Dirithromycin wird in Form eines Polymorphs gebildet, das hierin als "Form I" bezeichnet wird. Es hat das folgende Röntgendiffraktionsmuster am Pulver, worin D für den Interplanarabstand und I/I&sub0; für die relative Intensität stehen:
- Es wurde nun festgestellt, daß die Form I metastabil ist und daher nicht gut zur Verwendung in pharmazeutischen Formulierungen, wie Tabletten, geeignet ist.
- Jedoch wurde nun überraschenderweise und gemäß der vorliegenden Erfindung erkannt, daß ein zweites Polymorph von Dirithromycin, hierin als "Form II" bezeichnet, stabil ist und daher zur Verwendung in pharmazeutischen Formulierungen, wie Tabletten, gut geeignet ist.
- Daher wird bei einer Ausführungsform der Erfindung eine Tablette bereitgestellt, die Dirithromycin Form II als Wirkstoff enthält. Die Tablette enthält natürlich auch ein oder mehrere pharmazeutisch annehmbare Hilfsstoffe, wie Bindemittel, Gleitmittel, Desintegratoren, Füllstoffe usw..
- Dieses neue Polymorph hat im wesentlichen die folgenden Röntgendiffraktionsmuster am Pulver, worin D für den Interplanarabstand und I/I&sub0; für die relative Intensität stehen:
- Die obigen Röntgendiffraktionsmuster am Pulver wurden mit einem Nicolet 12V Pulverdiffraktometer aufgenommen, das mit einem Graphitmonochrometer mit einer Kupferbestrahlung von λ = 1,5818/Angström ausgestattet ist. Der Fachmann erkennt, daß die Intensitätswerte von Faktoren beeinflußt werden, wie der Probenvorbereitung und den Unterschieden zwischen den Instrumenten. Die D-Abstände stellen daher das Wesentliche der Diffraktionsmuster dar.
- Da der Wirkstoff der Tablette (Form II) als Pharmazeutikum verwendet werden soll, sollte er in einem pharmazeutischen Reinheitsgrad vorliegen. Für Stabilitätszwecke sollte die Form II keine wesentlichen Mengen an Form I enthalten. Vorzugsweise sollten mit Form II weniger als 10 % Form I, am besten weniger als 5 % vorkommen.
- Dirithromycin ist ein sehr wirksames Antibiotikum und weist gegenüber anderen Makrolidantibiotika beträchtliche Vorteile auf, da es nur einmal am Tag in einer Gesamtdosis von bis zu 500 mg verabreicht werden muß. Jedoch bedarf es einiger Sorgfalt bei der Herstellung oral annehmbarer Formulierungen, da die Verbindung gegenüber sauren Medien, wie sie im Magen vorkommen empfindlich ist. In diesem Zusammenhang wird auf die US-A 4 755 385 verwiesen, und es ist bevorzugt, daß die erfindungsgemäße Tablette von einer Schicht ummantelt wird, die gegenüber Magensäften resistent ist, das heißt einer "enteralen Ummantelung". Die Art solcher Ummantelungen ist dem Fachmann gut bekannt. Häufig wird ein Lack zur Bildung der Ummantelung verwendet.
- Die folgenden Beispiele 1 bis 4 erläutern die Herstellung der Form II.
- Eine Menge von 20 g nicht-solvatisiertem Dirithromycin wird zu einem Lösemittelgemisch gegeben, das aus 81 ml Aceton und 9 ml Wasser besteht. Die Lösung wird auf etwa 60ºC aufgeheizt. Die Lösung wird auf 60ºC gehalten bis das Reaktionsvolumen 35 ml beträgt. Es wird eine weitere Menge Wasser von 100 ml langsam während einer Stunde zugegeben. Das Gemisch kann auf Raumtemperatur abkühlen. Das abgekühlte Gemisch wird in ein Eisbad gestellt und für 1 Stunde gerührt. Der Feststoff wird durch Filtration isoliert und mit 25 ml gekühltem Lösemittel gewaschen, das sich aus 67 % Wasser und 33 % Aceton zusammensetzt. Der Feststoff wird mit einer Menge von 40 ml Wasser bei Umgebungstemperatur gewaschen. Der nasse Kuchen des Acetonsolvats von Dirithromycin wird bei Umgebungsbedingungen über Nacht stehengelassen.
- Eine Menge von 180 ml Wasser wird zum nassen Kuchen des Acetonsolvats gegeben. Das Gemisch wird auf 70ºC aufgeheizt und wird bei 70ºC unter einer Stickstoffspülung für 4 Stunden gerührt. Der Feststoff wird unmittelbar durch Filtration isoliert und mit einer Menge von 30 ml Wasser gewaschen, das auf 70ºC erwärmt wurde. Der isolierte Feststoff wird im Vakuum bei 40ºC über Nacht getrocknet. Der Feststoff wird als Form II Dirithromycin durch Röntgendiffraktometrie am Pulver identifiziert.
- Gesamtausbeute: 90,8 %.
- Eine 10,0 g Probe des Acetonsolvats von Dirithromycin wird unter Rühren zu 100 ml Wasser unter Stickstoffspülung gegeben. Die Temperatur des Reaktionsgemisches wird auf 74ºC angehoben und es wird bei etwa 72-75ºC für 4 Stunden gerührt. Das warme Gemisch wird vakuumfiltriert und mit etwa 35 ml 60ºC warmen Wasser gewaschen. Der Feststoff wird bei 50ºC über Nacht im Vakuum getrocknet. Der Feststoff wird als Form II Dirithromycin durch Röntgendiffraktometrie am Pulver identifiziert. Die Gesamtausbeute der Reaktion beträgt 8,74 g (87,4 %).
- Eine Menge von 15 g Form I Dirithromycin wird zu 150 ml Wasser gegeben. Das Reaktionsgemisch wird auf 74ºC aufgeheizt und für 4 Stunden bei 74ºC gerührt. Der Feststoff wird durch Filtration isoliert und mit zwei Waschschritten mit jeweils 40 ml 70ºC warmen Wasser gewaschen. Die Probe wird in einem Vakuumofen bei 25ºC für etwa 68 Stunden getrocknet. Das Produkt wird als Form II Dirithromycin durch Röntgendiffraktometrie am Pulver identifiziert.
- Ausbeute: 97,5 %
- Eine Menge von 3,01 g des Form I Dirithromycins wird zu 15 ml Ethylacetat gegeben. Das Reaktionsgemisch wird auf etwa 76ºC aufgeheizt und man läßt das Gemisch kochen bis das Reaktionsvolumen etwa 10 ml beträgt. Eine Menge von 20 ml n-Octan wird zugegeben. Das Gemisch kann auf Raumtemperatur abkühlen. Der Feststoff wird durch Filtration isoliert. Die Probe wird bei Raumtemperatur getrocknet. Das Produkt wird durch Röntgendiffraktometrie am Pulver als Form II Dirithromycin identifiziert.
- Gesamtausbeute: 95 %
- Allgemeine Verfahren zur Herstellung pharmazeutischer Formulierungen, die Dirithromycin enthalten sind in US-A 4 755 385 beschrieben. Im wesentlichen beinhaltet das Herstellungsverfahren das Mischen der Form II mit den notwendigen pharmazeutisch annehmbaren Trägern gefolgt von einem Pressen zu Tabletten und wahlweise das Ummanteln mit einer Schicht, die gegen Magensäfte resistent ist. Das folgende spezifische Beispiel erläutert jedoch die erfindungsgenmäße Herstellung einer ummantelten Tablette.
- Die folgenden Daten betreffen eine Tablette, die 250 mg Dirithromycin Form II als Wirkstoff enthält.
- Die Chargen können als einzelne Einheit oder als Teil hiervon hergestellt und alle Bestandteile als trokkene Mischung vor dem Pressen gemischt werden. Alle Bestandteile werden zur Sicherheit gesiebt. Dirithromycin (Form II), Magnesiumcarbonat, mikrokristalline Cellulose, Natriumstärkeglycolat, Hydroxypropylcellulose und Magnesiumstearat werden sorgfältig in einem Bandmischer oder einem anderen geeigneten Mischertyp, wie einem V-Mischer, einem konischen Mischer usw. gründlich gemischt. Das entstehende Pulvergemisch wird mittels eines Rollverdichters oder einer äquivalenten Trockengranulierausrüstung verdichtet. Die Granulierung wird erforderlichenfalls mittels eines oszillierenden Granulators, einer Fitz-Mühle oder einer anderen geeigneten Mahlausrüstung auf eine bestimmte Größe eingestellt. Die größeneingestellte Granulierung wird mit zusätzlicher mikrokristalliner Cellulose, Croscarmellose-Natrium und Magnesiumstearat in einem Bandmischer oder einem anderen geeigneten Mischertyp, wie einem V-Mischer, einem konischen Mischer usw. gemischt. Nach dem Mischen wird das Gewicht mit dem theoretischen Wert verglichen. Die Granulierung wird in einer herkömmlichen Preßmaschine wie einer Manesty Betapress usw. gepreßt. Die Gewichtsüberprüfungen der Tablettenkerne werden während dem Preßverfahren routinemäßig durchgeführt und die Proben werden zum Testen vereinigt.
- Die Untergrundummantelung wird durch Lösen der Hydroxypropylmethylcellulose 2910 (E-5), Polyethylenglykol, Propylenglykol und Benzylalkohol in gereinigtem Wasser hergestellt. Diese Ummantelungslösung kann mittels eines Groen Kessels, Topfes oder einem Äquivalent hierzu und einem Mittel zur Bereitstellung einer Durchmischung, wie einem Lightening Mischer oder einem Äquivalent hierzu hergestellt werden.
- Diese Ummantelungslösung wird auf die obigen Tablettenkerne in einer Accela Cota Ummantelungspfanne oder einer äquivalenten Filmummantelungsausrüstung aufgetragen. Geeignete Sprühsysteme, wie das Accela- Spray II, die Nordson Sprüheinheit, die Graco Sprüheinheit oder eine äquivalente Sprühausrüstung werden verwendet. Abwechselnde Sprüh- und Trockerzyklen werden erforderlichenfalls verwendet, bis die gewünschte Menge an Ummantelungslösung aufgetragen wurde.
- Die enterale Ummantelungssuspension wird durch Mischen der wäßrigen Methacrylsäurecopolymerdispersion (L30D), der Farbmischung Weiß T3166-WE und Triethylcitrat mit gereinigtem Wasser hergestellt. Diese Ummantelungssuspension kann mittels eines Groen Kessels, Topfes oder einem Äquivalent hierzu und einem Mittel zur Bereitstellung einer Durchmischung, wie einem Lightening Mischer oder einem Äquivalent hierzu hergestellt werden.
- Diese Ummantelungssuspension wird auf die obigen untergrundbeschichteten Tabletten in einer Accela Cota Ummantelungspfanne oder einer äquivalenten Filmummantelungsausrüstung aufgetragen. Geeignete Sprühsysteme, wie das Accela-Spray II, die Nordson Sprüheinheit, die Graco Sprüheinheit oder eine äquivalente Sprühausrüstung werden verwendet. Abwechselnde Sprüh- und Trockenzyklen werden erforderlichenfalls verwendet bis die gewünschte Menge an Ummantelungssuspension aufgetragen wurde. Die Tabletten können durch leichtes Bestäuben mit Talkum geschönt werden (um den Glanz der ummantelten Tabletten zu steigern). Es haften nur Spurenmengen dieses Poliermittels an den geschönten Tabletten.
Claims (3)
1. Tablette, die als Wirkstoff Dirithromycin Form II zusammen mit einem oder mehreren pharmazeutisch
annehmbaren Hilfsstoffen enthält, wobei dieses Dirithromycin Form II im wesentlichen das folgende
Röntgendiffraktionsmuster am Pulver aufweist, worin D für den Interplanarabstand und I/I&sub0; für die relative Intensität
stehen:
2. Tablette nach Anspruch 1, die mit einer oder mehreren Schichten ummantelt ist, welche gegenüber Magensäften
resistent sind.
3. Pharmazeutische Formulierung, die als Wirkstoff Dirithromycin Form II nach Anspruch 1 zusammen mit einem
oder mehreren pharmazeutisch annehmbaren Hilfsstoffen enthält.
