Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Zwischenverbindungen fuer die Herstellung von Verbindungen der allgemeinen Formel (I). Das erfindungsgemaesse Verfahren zeichnet sich dadurch aus, dass 2-(2-pyrrolidinyl)-ethanol mit 1-Brommethylzyklohexen zur Umsetzung gebracht wird, dann das so erhaltene 1-Zyklohexenylmethyl-2-hydroxyethylpyrrolidin mit Thionylchlorid erwaermt wird, um zu dem 1-Zyklohexenylmethyl-2-chlorethylpyrolidin zu gelangen. Verfahren zur Herstellung neuer substituierter heterozyklischer Phenoxyamine
Die vorliegende Erfindung betrifft ein Verfahren zur Herstellung neuer substituierter heterozyklischer Phenoxyamine mit wertvollen pharmakologischen Eigenschaften.
Die 'erfindungsgemäß hergestellten Verbindungen stellen wertvolle Lokalanästhetika dar.
Als Lokalanästhetika wurden bisher verwendet Xylokain, Prokain und Kokain.
Die erfindungsgemäß substituierten heterozyklischen Phenoxyamine sind neu. Angaben über Verfahren zu ihrer Herstellung sind nicht bekannt.
Ziel der Erfindung ist die Bereitstellung von neuen Verbindungen mit verbesserter Wirkung auf das Zentralnervensystem für die Anwendung als Lokalanästhetika.
Der Erfindung liegt die Aufgabe zugrunde, Verfahren zur Herstellung neuer Lokalanästhetika aufzuzeigen. Gegenstand der Erfindung ist ein Verfahren zur Herstellung von Zwischen-

das man nach, dem erfindungsgemäßen Verfahren erhält, wird für die Herstellung von Verbindungen der Formel I gemäi3 dem in der Patentanmeldung AP C 07 D / 222 455 beschriebenen Verfahren verwendet. -
Das Verfahren der vorliegenden Erfindung wird durch das folgende Beispiel veranschaulicht,
1. 2-»/i-(1-Zyklohexenylmethyl)~2~pyrrolidinyl7ethanol
16,4 g 2-(2-Pyrrolidinyl)ethanol (0,143 Mol) und 84,3 ml 1,78 N alkoholisches Kaliumhydroxid (0,150 Mol) werden in einen 500-ml-Kurzhals-Rundkolben eingegeben, ausgestattet mit einem Rührwerk, einem Thermometer, einem Rückflußkühler und einem Tropftrichter. Im Anschluß daran werden 28 g 1-Brommethylzyklohexen (Reinheit 90 %) tropfenweise zugesetzt.
Die Temperatur steigt von 20 0C 'auf 55 0C an, und es bildet sich ein Niederschlag. Wach Ablauf einer einstündigen Reaktion werden die Salze abfiltriert und das Filtrat zur Trockne eingedampft.

Das Rückstandsöl wird mit 150 ml Wasser und sodann mit Salzsäure behandelt,, bis der pH-Wert = 1 ist, Nach 2 Extraktionen mit 100 ml Ethylether auf einmal wird die wäßrige Phase mit Soda alkalisch gemacht» Hieran anschließend erfolgt eine dreimalige Extraktion mil; 100 ml Ether. Die jeweiligen Extrakte werden über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft. Das Rückstandsöl wird unter Vakuumbedingungen destilliert. Dabei werden 20,7 g der Verbindung erhalten (Siedepunkt unter einem Druck von 2 mm Hg 100 bis 107 °C; n^° = 1,506).
2. 2-/1-(1-Zyklohexenylmethyl)-2-pyrrolidinyl7ch.loyQthan
17,8 g 2-/i-(1-Zyklohexenylmethyl)-2-pyrrolidinyl7ethanol (0,085 Mol) und 50 ml Chloroform werden in einen 250-ml-Kurzhals-Rundkolben eingegeben, der mit einem Rührwerk, einem Thermometer, einem Rückflußkühler und einem Tropftrichter ausgestattet ist. Anschließend werden 15,3 ml Thionylchlorid tropfenweise zugegeben. Die Temperatur wird dabei durch Abkühlung zwischen etwa 20 0G und 25 0C gehalten.
Das Gemisch wird sodann am Rückflußkühler 3 Stunden lang gekocht. Die Löschung wird zur Trockne eingedampft, dann der Rückstand mit 20 ml Toluol behandelt. Nach Eindampfen zur Trockne unter Vakuumbedingungen wird der Rückstand in 100 ml Ethylazetat suspendiert. Die Kristalle werden filtriert und in einem Trockenofen bei 50 0C getrocknet.

Die neuen Verbindungen werden hergestellt, in-dem ein Phenol entsprechend der allgemeinen Formel (V):

Lage ist, eliminiert zu werden, kondensiert wird. Die Gruppe Y wird beispielsweise für ein Halogenatom stehen, im besonderen für Chlor, Brom oder Jod, um zu einer Verbindung der allgemeinen Formel (I) zu gelangen:

wobei die Definition von m, n, A, X und E ungeändert bleibt. .
Diese Verbindungen können in ein Salz überführt werden. Sie können durch Umsetzung mit einer optisch aktiven Säure in ihre pharmaKologisch aktiven Isomere getrennt werden.
Die Phenole der Formel (V) werden in der Form der Phenolate der Alkalimetalle verwendet, im besonderen als Natriumphenolat. Die betreffenden Phenolate werden dabei zum Beispiel durch die Umsetzung der Phenole mit den Alkoholaten der Alkalimetalle gewonnen.
Die Reaktion zwischen den Verbindungen der Formel (V) und den Verbindungen der Formel (VI) wird in einem inerten

organischen Lösungsmittel, wie etwa unter Verwendung von Toluol, Xylol, ..., ausgeführt, wobei die Umsetzung bei der Rückflußtemperatur des Reaktionsgemisches erfolgt.
Die Verbindungen werden hergestellt und anschließend nach normalen Verfahren getrennt, isoliert und gereinigt, wozu beispielsweise die Extraktion, die Bildung von Salzen, das Umkristallisieren, die Chromatographie usw. zu rechnen s ind.
Die Salzbildung der Verbindungen der Formel (I) wird vorzugsweise durch die Zugabe einer Mineralsäure, wie etwa unter Einsatz von Salzsäure, Bromwasserstoffsäure, Orthophosphorsäure, oder einer organischen Säure, wie etwa unter Verwendung von Fumarsäure, Zitronensäure oder Oxalsäure, erreicht.
Die Verbindungen der allgemeinen Formel (I) können ebenfalls beispielsweise mit Alkylsulfaten oder -halogeniden umgesetzt werden, um zu quaternären Ammoniumsalzen zu gelangen.
Die Verbindungen der Formel (I) können in einer an und für sich bekannten Art und Weise beispielsweise durch Wasserstoffperoxid und Mangan(IV)-oxid oxydiert werden, um zu den entsprechenden N-Oxiden zu gelangen.
Die Trennung der Verbindungen der allgemeinen Formel (I) wird mit einer optisch aktiven Säure durchgeführt.

entspricht, können aus den Phenolen durch Azetylierung, Chlorierung, Entazetylierung und anschließende Reinigung gewonnen werden.
Die Ausgangsνerb indungen der Formel (V), wobei X dem Brom entspricht, können aus dem o-Nitrophenol durch Bromierung, Alkylierung der Phenolfunktion, Reduktion der Nitrogxnappe, Diazotierung und Aufspaltung gewonnen werden.
Die Ausgangs verb indungen der Formel (VI) können in Übereinstimmung mit dem 4·, Verfahren gemäß der Beschreibung von YAO-HUA-WU und J. R. CORRIGAN in der Zeitschrift J. Org. Chem. (1961), Seite 1531, hergestellt werden.
Pharmakologisch besonders wertvoll sind die folgenden, nach dem erfindungsgemäßeη Verfahren hergestellten Verbindungen:

Die Erfindung wird nachstehend an einigen Beispielen näher erläutert.
Dabei sollen diese Verkörperungen in bezug auf die Art und Weise ihrer Leistungsfähigkeit und die Möglichkeiten ihres Einsatzes nicht beschränkt sein*

1. 2-/T-» (1 -zyklohexenylmethyl )-2-»pyrrolidlnyl7äthanol
16.4 g 2-(2-Pyrrolidinyl)äthanol (0,143 Mol) und 84,3 ml 1,78 N alkoholisches Kaliumhydroxid (0,150 Mol) werden in einen 500-ml-Kurzhals-Rundkolben eingegeben, ausgestattet mit einem Rührwerk, einem Thermometer, einem Rückflußkühler und einem Tropftrichter» Im Anschluß daran werden 28 g 1-Brommethylzyklohexen (Reinheit 90%) tropfenweise zugesetzt.
Die Temperatur steigt von 20 0C auf 55 0C an, und es bildet sich ein Niederschlag. Nach Ablauf einer einstündigen Reaktion werden die Salze abfiltriert und das Filtrat zur Trockne eingedampft·
Das Rückstandsöl wird mit 150 ml Wasser und sodann mit Salzsäure behandelt, bis der pH-Wert =» 1 ist. Nach 2 Extraktionen mit 100 ml Äthyläther auf einmal wird die wäßrige Phase mit Soda alkalisch gemacht· Hieran anschließend erfolgt eine dreimalige Extraktion mit 100 ml Äther, Die Jeweiligen Extrakte werden über Magnesiumsulfat getrocknet, filtriert und zur Trockne eingedampft· Das Rückstandsöl wird unter Vakuumbedingungen destilliert· Dabei

werden 20,7 g der Verbindung erhalten (Siedepunkt unter einem Druck von 2 mm Hg 100 bis 107 0C; ώ|° = 1,506).
2. 2-/ff-(i-Zyklohexenylmethyl)~2-pyrrolidinyl7chloräthan
17,8 g 2-2i-(i-Zyklohexenylmethyl)-2-pyrrolidinyl7äthanol (0,085 Mol) und 50 ml Chloroform werden in einen 250-ml-Kurzhals-Rundkolben eingegeben, der mit einem Rührwerk, einem Thermometer, einem Rückflußkühler und einem Tropftrichter ausgestattet ist. Anschließend werden 15>3 cal Thionylchlorid tropfenweise zugegeben. Die Temperatur wird dabei durch Abkühlung zwischen etwa 20 0C und 25 °C gehalten.
Das Gemisch wird sodann am Rückflußkühler 3 Stunden lang gekocht. Die Lösung wird zur Trockne eingedampft, dann der Rückstand mit 20 ml Toluol behandelt. Nach Eindampfen zur Trockne unter Vakuumbedingungen wird der Rückstand in 100 ml Äthylazetat suspendiert. Die Kristalle werden filtriert und in einem Trockenofen bei 50 0C getrocknet.

O 59KrfindungsanspruchVerfahren zur Herstellung von Zwischenverbindungen für die Herstellung von Verbindungen der Formel I(CH0)(DO - (GH2)m - CH - (CH2)n -N-R.worin m = O oder 2,η = O oder 2, unter der Voraussetzung, daß in + 2=2und worinA = Wasserstoff, ' .Alkoxygruppe mit 1 bis 4 Kohlenstoffatomen in>der Alkylgruppe, wie etwa Methoxy, Ethoxy, linearem oder verzweigtem Propoxy, Butoxy, Alkenyloxygruppe mit 2 bis 6 Kohlenstoffatomen in der Alkenylgruppe, wie etwa Vinyloxy, Propenyloxy (Allyl), Butenyloxy, Pentenyloxy, Hexenyloxy;X = Halogen, wie etwa Fluor, Chlor, Brom; R = Zyklohexenylmethyl,gekennzeichnet dadurch, daß 2~(2-Pyrrolidin3?-l-eth.anol mit 1-Brommethylzyklohexen zur Umsetzung gebracht wird, dann das so erhaltene i-Zyklohexenylmethyl-2-hydroxyethylpyrrolidin mit Thionylchlorid erwärmt wird, um zu dem 1-Zyklohexenylmet.hyl-2-chlorethylpyrrolidin zu gelangen,
