Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer 3,3'-Triphenylmethandicarbonsäurederivate, welche eine antiinflammatorische Aktivität beim töpischen Aufbringen auf einen Entzündungsbereich aufweisen.
5 Gewisse 3,3'-Triphenylmethandicarbonsäurederivate, welche den Verbindungen der nachfolgend gegebenen Formel I strukturell verwandt sind, waren als Zwischenprodukte für die Herstellung von Farbstoffen bereits bekannt. I.N. Ivanova und Mitarbeiter haben die Herstellung einiger von diesen Verbin-lo düngen, wie z.B. 4"-Nitro-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenylme-
thandicarbonsäure,
im Zusammenhang mit physikalischen Bestimmungen an ihren oxidierten Derivaten beschrieben [Uch. Uap. Saratovsk. Gos. 15 Univ. 71 (1959), 195-200, referiert in Chem. Abstr. 57,7158c; Zhur. Anal. Khim. 26 (1971), 1253-1258, referiert in Chem. Abstr. 75,126024c]. Es wurde nun gefunden, und darauf beruht die vorliegende Erfindung, dass die neuen 3,3'-Triphe-nylmethandicarbonsäurederivate der unten definierten Formel 20 I unerwarteterweise bei lokaler Anwendung auf eine entzündete Fläche antiphlogistische Eigenschaften entfalten.
Die erfindungsgemäss erhältlichen Verbindungen entsprechen der folgenden Formel:
1 bis 6 Kohlenstoffatomen steht. Die Herstellung der pharmazeutisch annehmbaren Salze dieser Verbindungen bildet ebenfalls einen Gegenstand der Erfindung.
40 Das Verfahren besteht darin, dass man ein Oxim der Formel:
in welcher R obige Bedeutung hat, mit einem wasserabspal-55 tenden Mittel bei einer Temperatur im Bereich von 50 bis 200 °C behandelt und die erhaltene Verbindung gegebenenfalls mit einer ein pharmazeutisch annehmbares Kation liefernden Base umsetzt.
Ein besonders gut geeigneter Wert für R, wenn dieses für 60 ein C1_6-Alkylradikal steht, ist z.B. ein Methyl-, Äthyl-, Pro-pyl- oder Butylradikal.
Ein besonders gut geeignetes Dehydratisierungsmittel ist z.B. Thionylchlorid, ein Gemisch aus N,N-Dimethylformamid 65 und Natriumformiat, ein Gemisch aus Natriumacetat und Essigsäure, oder Essigsäureanhydrid. Bei Verwendung von Essigsäureanhydrid als Dehydratisierungsmittel ist das erste isolierte Produkt eine Verbindung der Formel:

Diese Verbindung kann jedoch ohne weiteres hydrolysiert werden, indem man sie mit einer wässrigen Säure, z.B. Schwefelsäure oder Salzsäure, oder einer wässrigen Base, z. B. Natronlauge oder Kalilauge, bei einer Temperatur von 20 bis 100 °C hydrolysiert.
Die Wasserabspaltung wird bei erhöhter Temperatur, nämlich bei 50 bis 200 °C, und insbesondere bei 100 bis 150 °C, durchgeführt, wobei gegebenenfalls ein inertes Lösungsmittel oder Verdünnungsmittel, z.B. 1,1,2,2-Tetrachloräthan, vorhanden sein kann.
Die Ausgangsprodukte der Formel II können durch Reaktion einer Formylverbindung der Formel:

in welcher R obige Bedeutung hat, mit Hydroxylamin unter für die Bildung von Oximen von Aldehyden herkömmlichen Bedingungen hergestellt werden.
Ihrerseits können die Formylverbindungen der Formel IV durch Reaktion eines Dialdehydes der Formel:
ganischen Base, die ein pharmazeutisch annehmbares Kation liefert, wie ein Triäthanolaminsalz.
Die hierin angewendete Nomenklatur ist auf folgendem System aufgebaut, von dem Formel Ia als Beispiel gegeben wird:

Eine besonders bevorzugte neue Verbindung der Formel I ist
20 4"-Cyano-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-
methandicarbonsäure.
Die Verbindungen der Formel I haben antiinflammatori-sche Eigenschaften, wenn sie topisch auf den Bereich einer Entzündung aufgebracht werden. Sie sind besonders geeignet 25 zur Behandlung von Entzündungserkrankungen oder von Ent-zündungszuständen der Haut bei Warmblütern.
Zusätzlich zu den antiinflammatorischen Eigenschaften haben bestimmte der Verbindungen der Formel I antibakterielle Eigenschaften. Diese antibakteriellen Eigenschaften stellen le-30 diglich einen verwertbaren Zusatz zu den antiinflammatorischen Eigenschaften dar und sie sind für sich nicht ausreichend, um die Verwendung einer Verbindung, die diese Eigenschaften besitzt, lediglich als antibakterielles Mittel zu rechtfertigen.
35 Die antiinflammatorischen Eigenschaften einer Verbindung der Formel I können in einem Standardtest, der die Hemmung der durch Crotonöl erzeugten Entzündung des Mausohrs verwendet, gezeigt werden. Die Aktivität der jeweiligen Verbindung bei diesem Test hängt von ihrer jeweiligen chemischen 40 Struktur ab. Spezifische Verbindungen der Formel I, wie sie hierin beschrieben werden, ergeben jedoch eine signifikante Hemmung der Entzündung bei einer topisch aufgebrachten Dosis von 100 /ug bis 2000 /ig pro Ohr. Bei der aktiven Dosis wurden bei diesem Test keine offenkundigen toxischen Effekte 45 festgestellt.
Die antibakteriellen Eigenschaften der Wirkstoffe können anhand eines Standardtests gezeigt werden, welcher auf der Hemmung des Wachstums von grampositiven Bakterien, beispielsweise von Strep. faecalis und Staph. aureus, die in be-50 kannter Weise gezüchtet werden, beruht. Die antibakterielle Aktivität der einzelnen Verbindungen hängt zwar von ihrer jeweiligen chemischen Struktur ab, doch erweisen sich im allgemeinen spezielle Verbindungen der Formel I, wie sie hierin beschrieben werden, bei Konzentrationen von 10 bis 100 ppm 55 aktiv.
Die Erfindung wird durch die Beispiele näher erläutert.
in Gegenwart von konzentrierter Schwefelsäure, vorzugsweise bei einer Temperatur von 10 bis 30 °C, hergestellt werden.
Wird ein pharmazeutisch annehmbares Salz gewünscht, dann wird die Verbindung der Formel I mit einer Base umgesetzt, die ein pharmazeutisch annehmbares Kation liefert, z.B. mit Natriumhydroxid, Calciumhydroxid, Ammoniak oder Tri-äthanolamin.
Ein besonders gut geeignetes Salz eines Wirkstoffes der Formel I ist beispielsweise ein Alkalimetallsalz, wie ein Natriumsalz, ein Erdalkalimetallsalz, wie ein Calciumsalz, ein Ammonium- oder ein Aluminiumsalz oder das Salz einer or-
Beispiel 1
Essigsäureanhydrid (2,1 ml) wurde zu einer Suspension des 60 Oxims von
4"-Formyl-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-
methandicarbonsäure (4,35 g) gegeben und das Gemisch wurde 24 Stunden lang am Rückfluss erhitzt. Nach dem Abkühlen auf 20 bis 25 °C wurde 65 das Gemisch heftig mit 2N-wässriger Natriumhydroxidlösung (ungefähr 40 ml) gerührt, so dass das Gemisch nahe bei einem pH-Wert von 11 gehalten wurde. Die wässrige Phase wurde sodann abgetrennt und mit Salzsäure auf einen pH-Wert von

3 angesäuert. Der ausgefällte Feststoff wurde abfiltriert, mit Wasser gewaschen und aus einem Gemisch aus Methanol und Acetonitril umkristallisiert, wodurch 4"-Cyano-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-
methandicarbonsäure,
Fp. 287 bis 289 °C (Zersetzung), erhalten wurde.
Das Ausgangsprodukt kann wie folgt erhalten werden: Therephthalaldehyd (25 g) wurde protionenweise zu einem gerührten Gemisch aus konzentrierter Schwefelsäure (98 % Gewicht/Gewicht, 300 ml) und Wasser (23 ml) gegeben, und das Gemisch wurde auf 20 bis 25 °C abgekühlt. o-Cresotsäure (2-Hydroxy-3-methylbenzoesäure) (58 g) wurde sodann rasch zugegeben, und das Gemisch wurde 16 Stunden bei 20 bis 25 °C weitergerührt. Das Reaktionsgemisch wurde hierauf in Eiswasser (1,5 Liter) gegossen und der ausgefällte Feststoff wurde abfiltriert, mit Wasser gewaschen und aus 50%igem wässrigem Aceton (Volumen/Volumen) umkristallisiert, wodurch
4"-Formyl-4,4'- dihydroxy-5,5'-dimethyl-3,3'-triphe-
nylmethandicarbonsäure,
Fp. 274 bis 277 °C (Zersetzung), erhalten wurde.
Hydroxylaminhydrochlorid (16,0 g) wurde bei 20 bis 25 °C zu einer Lösung von
4"-Formyl-4,4'-dihydroxy-5,5 ' -dimethyl-3,3 ' -triphenyl-methandicarbonsäure
(42,0 g) in einem Gemisch aus Wasser (300 ml) und wässriger Ammoniaklösung (Dichte 0,88) (25 ml) gegeben. Das Gemisch wurde bei der gleichen Temperatur 1 Stunde lang gerührt. Sodann wurde der pH-Wert durch Zugabe von Eisessig (ungefähr 20 ml) auf 4 eingestellt. Der ausgefällte Feststoff wurde abfiltriert, mit Wasser gewaschen und mit Essigsäure aus der Lösung in einem Gemisch aus wässrigem Ammoniak und Aceton wieder ausgefällt. Auf diese Weise wurde das Oxim der
5 4"-Formyl-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-methandicarbonsäure,
Fp. 275 bis 277 °C (Zersetzung), erhalten.
Beispiel 2
io Das Oxim der
4"-Formyl-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-
methandicarbonsäure (750 g) wird einem Gemisch von 5 Litern 98—100%iger Ameisensäure und 10 Litern 1,4-Dioxan zugegeben, und hierauf 15 wird mit 60 g Hydroxylaminhydrochlorid und 60 g Natriumformiat versetzt. Das Gemisch wird während 10 Stunden unter Rückfluss zum Sieden erhitzt und anschliessend auf etwa 5 °C abgekühlt. Der dabei ausgeschiedene feste Stoff wird durch Abfiltrieren abgetrennt, mit 3 Litern Wasser gewaschen und 20 sofort aus 2,5 Litern 2-Methoxyäthanol umkristallisiert. Man erhält
4"-Cyano-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenyl-
methandicarbonsäure vom Smp. 267 bis 270 °C (Zersetzung) in 90%iger Ausbeute. 25 Eine nochmalige Umkristallisation aus einem Gemisch von 2-Methoxyäthanol und Acetonitril (1:2, Volumen/Volumen) erhöht den Smp. der Verbindung auf 286 bis 289 °C (Zersetzung).
Die Zugabe von Hydroxylaminhydrochlorid kann ausgelas-30 sen werden, es werden aber in diesem Fall etwas geringere Ausbeuten an der 4"-Cyanoverbindung erhalten.

- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet, dass die Wasserabspaltung durch Behandlung mit Natriumformiat bei einer Temperatur von 100 bis 150 °C durchgeführt wird.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer 3,3'-Triphenylmethan-dicarbonsäurederivate der Formel:in welcher R für Wasserstoff oder ein C^-Alkylradikal steht, und ihrer pharmazeutisch annehmbaren Salze, dadurch gekennzeichnet, dass man ein Oxim der Formel:in welcher R obige Bedeutung hat, mit einem wasserabspaltenden Mittel bei einer Temperatur im Bereich von 50 bis 200 °C behandelt und die erhaltene Verbindung gegebenenfalls mit einer ein pharmazeutisch annehmbares Kation liefernden Base umsetzt.
- 3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass als Ausgangsprodukt das Oxim der 4"-Formyl-4,4'-dihydroxy-5,5 '-dimethyl-3,3 '-triphenyl-methandicarbonsäure verwendet wird.
- 4. Verfahren zur Herstellung neuer 3,3'-Triphenylmethan-dicarbonsäurederivate der in Anspruch 1 gegebenen Formel I und ihrer pharmazeutisch annehmbaren Salze, dadurch gekennzeichnet, dass man ein Oxim der in Anspruch 1 gegebenen Formel II mit Essigsäureanhydrid bei einer Temperatur von 100 bis 150 °C behandelt und das durch die Wasserabspaltung entstandene Zwischenprodukt der Formel:durch Behandlung mit einer wässrigen Säure oder Base bei einer Temperatur im Bereich von 20 bis 100 °C hydrolysiert.
