Description
Die Erfindung betrifft ein Verfahren zur Herstellung neuer 3,3'-Diarylmethandicarbonsäure-derivate, welche eine antiinflammatorische Aktivität beim topischen Aufbringen auf einen Entzündungsbereich aufweisen.
Gewisse 3,3'-Triphenylmethandicarbonsäure-derivate der nachfolgend gegebenen Formel I, in welcher R für ein Methylradikal und B für ein Phenyl, 2-Chlorphenyl-, 4-Chlorphenyl-, 2,6-Dichlorphenyl-, 2,5-Dichlorphenyl- oder 2,3,6-Trichlor-phenylradikal stehen oder R für Wasserstoff und B für ein
2,4-Dinitrophenylradikal stehen, waren bereits bekannt, vor allem als Zwischenprodukte für die Herstellung von Farbstoffen. I.N. Ivanova und Mitarbeiter haben die Herstellung einiger von diesen Verbindungen im Zusammenhang mit physikalischen Bestimmungen an ihren oxidierten Derivaten beschrieben [Uch. Zap. Saratovsk. Gos. Univ. 71 (1959), 195-200, referiert in Chem. Abstr. 57, 7158c; Zhur. Anal. Khim. 26 (1971), 1253—1258, referiert in Chem Abstr. 75, 126024 c] Es wurde nun gefunden, und darauf beruht die vorliegende Erfindung, dass die neuen 3,3'-Diarylmethandicarbonsäure-derivate der unten definierten Formel I unterwarteterweise bei lokaler Anwendung auf eine entzündete Fläche antiphlogistische Eigenschaften entfalten.
Die erfindungsgemäss erhältlichen Verbindungen entsprechen der folgenden Formel:

Das Verfahren besteht darin, dass man eine Salicylsäure der Formel:

in welcher R die obige Bedeutung hat, mit einem Aldehyd der Formel:

in welcher B die obige Bedeutung hat, in Gegenwart einer starken Säure umsetzt und die erhaltene Verbindung gegebenenfalls mit einer ein pharmazeutisch annehmbares Kation liefernden Base umsetzt.
Eine besonders gut geeignete starke anorganische Säure ist z. B. konzentrierte Schwefelsäure, Salzsäure oder Fluorwasserstoffsäure. Eine besonders gut geeignete starke organische Säure ist Trifluoressigsäure. Das Verfahren wird geeigneterweise beispielsweise bei 0 bis 50° C und vorzugsweise bei 10 bis 30° C durchgeführt. Die Dauer des Verfahrens beträgt 30 min bis 4 Tage, je nach der Reaktivität der Reaktionsteilnehmer. Die starke anorganische oder organische Säure wird geeigneterweise im Überschuss verwendet, doch kann auch ein herkömmliches inertes Verdünnungsmittel zugesetzt werden. Die Salicylsäure der Formel II wird geeigneterweise im Überschuss, beispielsweise in einem Überschuss von 1 Moläquivalent gegenüber dem Aldehyd der Formel III verwendet.
Wird ein pharmazeutisch annehmbares Salz gewünscht, dann wird die Verbindung der Formel I mit einer Base umgesetzt die ein pharmazeutisch annehmbares Kation liefert, z. B. mit Natriumhydroxid, Calciumhydroxid, Ammoniak oder Tri-äthanolamin.
Ein besonders gut geeignetes Salz eines Wirkstoffes der Formel I ist beispielsweise ein Alkalimetallsalz, wie ein Natriumsalz, ein Erdalkalimetallsalz, wie ein Calciumsalz, ein Ammonium- oder ein Aluminiumsalz oder das Salz einer or5

ganischen Base, die ein pharmazeutisch annehmbares Kation liefert, wie ein Triäthanolaminsalz.
Ein besonders gut geeigneter Wert für R, wenn dieses für ein C^-AIkylradikal steht, ist z. B. ein Methyl-, Äthyl-, Pro-pyl- oder Butylradikal. Wenn dieses für ein Halogenatom steht, dann handelt es sich z. B. um ein Fluor-, Chlor- oder Bromatom.
Ein besonders gut geeigneter Wert für ein Halogenatom, wenn dieses für den Fall als Substituent vorhanden ist, dass B für ein gegebenenfalls substituiertes Phenylradikal steht, ist z. B. ein Fluor-, Chlor- oder Bromatom.
Ein besonders gut geeigneter Wert für B, wenn dieses für ein gegebenenfalls substituiertes Phenylradikal steht, ist z. B. ein Phenyl- oder ein 3-Nitro-, 4-Nitro-, 4-Fluoro-, 2-Chloro-, 4-Chloro-, 4-Bromo-, 4-Cyano-, 4-Carboxy-, 4-Carbamoyl-, 2,6-Difluoro-, 2,4-Dichloro-, 2,6-Dichloro-, 2,4,6-Trichloro-, 2-Fluoro-6-chloro-, 2-Chloro-4-cyano-, 2-Chloro-4-nitro-, 2-Chloro-5-nitro-, 2-Bromo-6-chloro- oder 4-Formyl-phenyl-radikal.

Die hierin angewendete Nomenklatur ist auf folgendem System aufgebaut, von dem Formel la als Beispiel gegeben wird:
15 Eine besonders bevorzugte neue Verbindung der Formel 1 ist 4"-Cyano-4,4'-dihydroxy-5,5'-dimethyI-3,3'-triphenyl-methandicarbonsäure.
Die Verbindungen der Formel I haben antiinflammatorische Eigenschaften, wenn sie topisch auf den Bereich einer 20 Entzündung aufgebracht werden. Sie sind besonders geeignet zur Behandlung von Entzündungserkrankungen oder von Ent-zündungszuständen der Haut bei Warmblütern geeignet.
Zusätzlich zu den antiinflammatorischen Eigenschaften haben bestimmte der Verbindung der Formel I antibakterielle 25 Eigenschaften. Diese antibakteriellen Eigenschaften stellen lediglich einen verwertbaren Zusatz zu den antiinflammatorischen Eigenschaften dar und sie sind für sich nicht ausreichend, um die Verwendung einer Verbindung, die diese Eigen

schatten besitzt, lediglich als antibakterielles Mittel zu rechtfertigen.
Eine besondere Gruppe von neuen Verbindungen der Formel I, die sowohl antiinflammatorische als auch antibakterielle Eigenschaften haben, enthält die Verbindungen 2", 4"-Dichloro-4,4'-dihydroxy-3,3'-triphenylmethandicarbon-säure und 4.'-Cyano-4,4'-dihydroxy-5,5'-dimethyl-3,3'-tri-phenylmethandicarbonsäure.
Die antiinflammatorischen Eigenschaften einer Verbindung der Formel I können in einem Standardtest, der die Hemmung der durch Crotonöl erzeugten Entzündung des Mausohrs verwendet, gezeigt werden. Die Aktivität der jeweiligen Verbindung bei diesem Test hängt von ihrer jeweiligen chemischen Struktur ab. Spezifische Verbindungen der Formel I, wie sie hierin beschrieben werden, ergeben jedoch eine signifikante Hemmung der Entzündung bei einer topisch aufgebrachten Dosis von 100 bis 2000(ig pro Ohr. Bei der aktiven Dosis wurden bei diesem Test keine offenkundigen toxischen Effekte festgestellt.
Die antibakteriellen Eigenschaften der Wirkstoffe können anhand eines Standardtests gezeigt werden, welcher auf der Hemmung des Wachstums von grampositiven Bakterien, beispielsweise von Strep.faecalis und Staph.aureeus, die in bekannter Weise gezüchtet werden, beruht. Die antibakterielle Aktivität der einzelnen Verbindungen hängt zwar von ihrer jeweiligen chemischen Struktur ab, doch erweisen sich im allgemeinen spezielle Verbindungen der Formel I, wie sie hierin beschrieben werden, bei Konzentrationen von 10 bis 1000 ppm aktiv.
Die Erfindung wird durch die Beispiele näher erläutert.
Beispiel 1
Die vorstehenden Verbindungen wurden alle in der folgenden allgemeinen Weise erhalten:
Der substituierte Benzaldehyd (0,15 Mol) wurde portio-s nenweise zu gerührter gekühlter (10°C) konzentrierter Schwefelsäure (93% G/V, 153 ml) gegeben. Das Salicylsäurederivat (0,335 Mol) wurde sodann rasch zugesetzt und das Gemisch wurde 40 min bis 4 Tage lang je nach den jeweils verwendeten Ausgangsstoffen bei 10 bis 25° C gerührt. Hierauf wurde das io Reaktionsgemisch in Eiswasser gegossen und der ausgefallene Farbstoff wurde abfiltriert und gründlich mit Wasser gewaschen. Der Feststoff wurde sodann unter Verwendung des angegebenen Lösungsmittels bzw. der angegebenen Lösungsmittel kristallisiert.

Beispiel 2
Terephthalaldehyd (25 g) wurde portionsweise zu einem 20 gerührten Gemisch aus konzentrierter Schwefelsäure (98% Gewicht/Gewicht, 300 ml) und Wasser (23 ml) gegeben und das Gemisch wurde auf 20 bis 25° C abgekühlt. o-Cresotsäure (2-Hydroxy-3-methylbenzoesäure) (58 g) wurde sodann rasch zugegeben und das Gemisch wurde 16 Stunden lang bei 20 bis 25 25° C weitergerührt. Das Reaktionsgemisch wurde hierauf in Eiswasser (1,51) gegossen und der ausgefällte Feststoff wurde abfiltriert, mit Wasser gewaschen und aus 5()%igem wässrigem Aceton (Volumen/Volumen) umkristallisiert, wodurch 4"-Formyl-4,4'-dihydroxy-5,5'-dimethyl-3,3'-triphenylmethandi-30 carbonsäure, Fp. 274 bis 277° C (Zersetzung), erhalten wurde.

- 2. Verfahren nach Anspruch I, dadurch gekennzeichnet, dass die Salicylsäure der Formel II in einem Überschuss von 1 Moläquivalent gegenüber dem Aldehyd der Formel III eingesetzt wird.2PATENTANSPRÜCHE 1. Verfahren zur Herstellung neuer 3,3'-Diarylmethandi-carbonsäurederivate der. Formel:HOCHHOOCCOOHin welcher R für Wasserstoff, ein C^-Alkylradikal oder ein Halogenatom steht und B ein 4-Pyridylradikal oder ein Phe-nylradikal, das gegebenenfalls 1 bis 3 Substituenten aus der Gruppe Halogenatome, Nitro-, Cyano-, Carbamoyl-, Carboxy-und Formylradikale trägt, darstellt, sowie ihrer pharmazeutisch annehmbaren Salze, jedoch ausgenommen solche Verbindungen der Formel I, bei welchen R für ein Methylradikal und B für ein Phenyl-, 2-Chlorphenyl-, 4-Chlorphenyl-, 2,6-Dichlor-phenyl-, 2,5-Dichlorphenyl- oder ein 2,3,6-Trichlorphenylra-dikal stehen, sowie solche Verbindungen, bei welchen R für Wasserstoff und B für ein 2,4-DinitrophenylradikaI stehen, dadurch gekennzeichnet, dass man eine Salicylsäure der Formel:RHOHOOCin welcher R die obige Bedeutung hat, mit einem Aldehyd der Formel:B-CHO (III)in welcher B die obige Bedeutung hat, in Gegenwart einer starken Säure umsetzt und die erhaltene Verbindung gegebenenfalls mit einer ein pharmazeutisch annehmbares Kation liefernden Base umsetzt.
- 3. Verfahren nach Anspruch 1 oder 2, dadurch gekennzeichnet, dass die Umsetzung in Gegenwart von Schwefelsäure bei einer Temperatur von 0 bis 50° C durchgeführt wird.
- 4. Verfahren nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass in den Ausgangsprodukten der Formeln II und III R für ein Methylradikal und B für ein 4-Cyano-phenylradikal stehen.
