ダービリア抽出物は、ダービリアの抽出物である。ダービリアは、ダービリア科(Durvilleaceae)ダービリア属(Durvillea)に属する海藻であり、Durvillea antarctica、Durvillea potatorum、Durvillea willanaなどはBull kelpと呼ばれる。ダービリア抽出物は、ダービリアの植物体の全体及び一部のいずれからの抽出物であってもよい。抽出物の調製条件は特に限定されず、溶媒としては、アルコールを含む溶媒が好ましく、アルコール/水が0/100~70/30(V/V:体積比)の溶媒であることがより好ましく、アルコール/水が0/100~40/60の溶媒であることが更に好ましい。抽出温度は、特に制限はないが、通常5~80℃、好ましくは5~50℃である。抽出時間は、通常1~24時間である。抽出は撹拌しながら行なうのが好ましい。抽出pHは、極端な酸性、アルカリ性に傾かなければ特に制限はない。
ラズベリーケトンは、4-(4-ヒドロキシフェニル)-2-ブタノン及び/又はp-ヒドロキシベンジルアセトンである。ラズベリーケトンは、ラズベリーの香り成分の一つである。ラズベリーケトンは、バラ科キイチゴ属に属する植物(例:ラズベリー)の植物体(例:低木、果実)等の天然物に由来していてもよいし、化学合成等による人工的な製造物であってもよい。
アーティチョーク葉抽出物は、アーティチョーク(学名Cynara scolymus L.(Compositae))の葉の抽出物である。アーティチョーク葉抽出物は、葉の一部及び全部のいずれからの抽出物であってもよい。抽出物の調製条件は特に限定されず、溶媒としては、アルコールが好ましく、エタノールがより好ましい。
ローズマリー抽出物は、ローズマリーの抽出物である。ローズマリーは、マンネンロウともいい、学名をRosmarinus officinalis L.といい、シソ科に属する常緑性低木である。ローズマリー抽出物は、ローズマリーの植物体の全体及び一部のいずれからの抽出物であってもよい。抽出物の調製条件は特に限定されず、溶媒としては、水、アルコール又はこれらの組み合わせが好ましく、水とエタノールの組み合わせがより好ましい。
イソフラボンは、3-フェニルクロモン(3-フェニル-4H-1-ベンゾピラン-4-オン)である。イソフラボンは、グリコシドの形態から糖を分離して得られるアグリコン(イソフラボンアグリコン)であることが好ましい。イソフラボンは、天然物に由来していてもよいし、化学合成等による人工的な製造物であってもよい。
ブドウ種子抽出物は、ブドウ(ブドウ科(Vitaceae)のつる性低木)の種子の抽出物である。抽出物の抽出対象は、種子である。ブドウ種子抽出物の調製条件は特に限定されず、溶媒としては、水、アルコールが好ましい。溶媒として水を用いる場合には、ブドウの種子を水にて70℃以上(好ましくは80~120℃、より好ましくは80~100℃)の抽出温度範囲で抽出することが好ましい。ブドウ種子抽出物には、プロアントシアニジンが含まれ、その含有量は、通常約38質量%である。
松樹皮抽出物は、松(マツ属(Pinus)の植物)の樹皮の抽出物である。松としては、パイナス・ラジアタ(Pinus Radiata)が例示される。抽出物の調製条件は特に限定されないが、溶媒としては水が好ましい。松樹皮抽出物には、プロアントシアニジン、カテキン、フラボノイド等が含まれる。
コレウス・フォルスコリ抽出物は、コレウス・フォルスコリの植物体の抽出物である。コレウス・フォルスコリは、シソ科コレウス属の多年草で、学名はColeus Forskohlii,Coleus barbatus,Plectranthus Barbatusである。コレウス・フォルスコリ抽出物は、コレウス・フォルスコリの植物体の全体及び一部のいずれからの抽出物であってもよい。抽出物の調製条件は特に限定されない。コレウス・フォルスコリ抽出物には、ジテルペンの一種であるフォルスコリンが含まれる。
カカオ抽出物は、カカオ(アオギリ科カカオ、学名をTheobroma cacao)の抽出物である。カカオ抽出物は、カカオの植物体の全体及び一部のいずれからの抽出物であってもよいが、抽出部位としては例えば、果実(カカオポッド)、種子が挙げられ、種子が好ましく、外殻皮(カカオハスク)であることがより好ましい。外殻皮とは、果実の中の白いパルプ質の果肉に包まれたカカオ豆(種子)の周りの皮を意味する。カカオ抽出物の調製条件は特に限定されないが、溶媒としてはアルコールが好ましく、含水エタノール(例:エタノール50~80容量%の含水エタノール)がより好ましい。
結晶セルロース、アルギン酸ナトリウム、キサンタンガム等の多糖類;
α化デンプン、ヒドロキシプロピルスターチ、コーンスターチ、ポテトスターチ等のスターチ及びその誘導体;
ショ糖、グルコース、キシリトール、エリスリトール、ソルビトール、ラクチトール、トレハロース、パラチノース、パラチニット(還元パラチノース)、マンニトール、マルチトール、ラクチトール、乳糖、果糖、粉末還元麦芽糖水飴等の糖類及び糖アルコール類;
粉末セルロース、部分α化デンプン、エチルセルロース等のセルロース及びその誘導体;
軽質無水ケイ酸、酸化チタン、水酸化アルミニウムゲル、合成ケイ酸アルミニウム、三ケイ酸アルミニウム、二酸化ケイ素、カオリン、カカオ脂、クエン酸又はその塩、ステアリン酸又はその塩、リン酸水素カルシウム、リン酸水素ナトリウム。
・ラット腸間膜脂肪細胞の調製
SDラット(オス、10週齢、日本エスエルシー(株))を2匹用いた。エーテル麻酔下でラットを安楽死させて解剖し、腸間膜脂肪組織を摘出した。摘出組織を滅菌シャーレに入れ、1%の抗生物質(Antibiotic-Antimycotic Stabilized、シグマ社製、A5955)を添加した氷冷生理食塩水(大塚生食注、大塚製薬(株))に浸し、血液及び毛を洗浄(毛が見えなくなるまで液を変えて数回洗浄)し、リンパの除去を行った。眼科鋏にて組織の切断(組織1gに対して25回切断)を行い、1mg/mLのコラゲナーゼ(放線菌由来Collagenase S-1、新田ゼラチン(株))を添加したPBS溶液で、37℃40分間、数分おきに攪拌しながら組織を分散した。目安として1匹分の組織を20mLコラゲナーゼ溶液で処理した。
各実験例及び比較実験例で用いる試料を調製した。試料の成分としては、ラクトフェリン(DMV製)、ラズベリーケトン(高砂香料工業株式会社製)、アーティチョーク葉抽出物(一丸ファルコス株式会社製、商品名バイオベネフィティF)、カカオ抽出物(オリザ油化製、カカオエキス-P)、ブドウ種子抽出物(キッコーマン株式会社製、商品名グラヴィノール)、松樹皮抽出物(Enzo Nutraceuticals Limited製、エンゾジノール)、コレウス・フォルスコリ抽出物(Natural Remedie社製)、ダービリア抽出物、ローズマリー抽出物(ライオン株式会社製、商品名レオミール)、大豆イソフラボンアグリコン(キッコーマン株式会社製)、ショウガエキス(日本粉末薬品株式会社製)、エピガロカテキンガレート(和光純薬工業株式会社製)、クロロゲン酸(和光純薬工業株式会社製)を用いた。アーティチョーク葉抽出物として用いたバイオベネフィティFは、賦形剤としてデキストリンを70質量%含むが、表1に示すアーティチョーク葉抽出物の配合量は、該賦形剤を除いて換算した数値である。
(単独評価用の試料)

(複合評価用の試料(+ラクトフェリン30ppm))

・ラクトフェリン以外の各成分10ppm+ラクトフェリン10ppm
グリセロールの定量は、F-キット グリセロール(ロシュ社製、製品番号:14820)を用いて行った。吸光度の測定は、Novaspec Plus(Amersham Bioscience社製)で行った。蒸留水11mLに溶解した溶液I 1.0mL、蒸留水2.0mL、上述した各試料0.1mL、溶液II 0.01mLを加えた。よく混和し、5分間室温で反応させ、AB340mmの吸光度(E1)を測定した。そこに溶液III 0.01mLを加え、よく混和した。7分間室温で反応させ、吸光度(E2)を測定した。Standardは、キット付属の標準液を用いた。

表1に示すように、実験例1~9における複合評価の脂肪分解促進率は、各成分の濃度に相当する単独評価における脂肪分解促進率を足し合わせた数値よりも上回っていた。
試料として表2に示す組み合わせを用いたほかは、実験例1と同様に脂肪分解促進率を評価した。結果を表2に示す。
Claims (3)
- (A)ラクトフェリンと、
(B)ダービリア抽出物、ラズベリーケトン、アーティチョーク葉抽出物、ローズマリー抽出物、イソフラボン、ブドウ種子抽出物、松樹皮抽出物、コレウス・フォルスコリ抽出物、及びカカオ抽出物からなる群より選ばれる1種以上の成分と
を含有する脂肪分解促進組成物。 - 食品、飼料又は医薬品である、請求項1に記載の脂肪分解促進組成物。
- (A)ラクトフェリンと、
(B)ダービリア抽出物、ラズベリーケトン、アーティチョーク葉抽出物、ローズマリー抽出物、イソフラボンアグリコン、ブドウ種子抽出物、松樹皮抽出物、コレウス・フォルスコリ抽出物、及びカカオ抽出物からなる群より選ばれる1種以上の成分と
を食品又は飼料に添加して、食品又は飼料に脂肪分解促進効果を付与する方法。
