Abstract
Специфические полинуклеотидные последовательности в составе гетерологичной кассеты экспрессии вводят в геном кукурузы в область, фланкированную 5'- и 3'-концами гена. С помощью пары полинуклеотидных праймеров производят ампликон для выявления варианта кукурузы, обозначенного как MIR604. Полученное растение кукурузы обладает устойчивостью к блошке длинноусой. 9 н. и 13 з.п. ф-лы, 2 ил., 4 табл.
Description
Claims (22)
1. Выделенная молекула нуклеиновой кислоты, предназначенная для определения варианта кукурузы MIR604, которая содержит нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4 и их комплементы.
2. Пара полинуклеотидных праймеров, включающая первый полинуклеотидный праймер и второй полинуклеотидный праймер, которые функционируют вместе в присутствии в образце ДНК-матрицы из варианта кукурузы MIR604 с образованием ампликона, который предназначен для определения варианта кукурузы MIR604, где первая праймерная последовательность представляет собой или является комплементарной геномной последовательности растения кукурузы, фланкирующей инсерционный сайт гетерологичной последовательности ДНК, которая встроена в геном растения кукурузы варианта кукурузы MIR604, а вторая полинуклеотидная праймерная последовательность представляет собой или является комплементарной гетерологичной последовательности ДНК, которая встроена в геном растения кукурузы варианта кукурузы MIR604.
3. Пара полинуклеотидных праймеров по п.2, в которой первый полинуклеотидный праймер содержит по меньшей мере 10 последовательно расположенных нуклеотидов от положения 1 до положения 801, представленных в SEQ ID NO: 3, или их комплементы.
4. Пара полинуклеотидных праймеров по п.3, в которой первый полинуклеотидный праймер содержит нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 7-SEQ ID NO: 15 и их комплементы.
5. Пара полинуклеотидных праймеров по п.2, в которой первый полинуклеотидный праймер содержит по меньшей мере 10 последовательно расположенных нуклеотидов от положения 507 до положения 1570, представленных в SEQ ID NO: 4, или их комплементы.
6. Пара полинуклеотидных праймеров по п.5, в которой первый полинуклеотидный праймер содержит нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 39-SEQ ID NO: 46 и их комплементы.
7. Пара полинуклеотидных праймеров по п.2, в которой второй полинуклеотидный праймер содержит по меньшей мере 10 последовательно расположенных нуклеотидов, представленных SEQ ID NO: 58, SEQ ID NO: 59, SEQ ID NO: 60, SEQ ID NO: 61, SEQ ID NO: 62 или SEQ ID NO: 63, или их комплементы.
8. Пара полинуклеотидных праймеров по п.7, в которой второй полинуклеотидный праймер содержит нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 16-SEQ ID NO: 38 и их комплементы.
9. Пара полинуклеотидных праймеров по п.2, в которой первый полинуклеотидный праймер имеет последовательность, представленную в SEQ ID NO: 15, а второй полинуклеотидный праймер имеет последовательность, представленную в SEQ ID NO: 28.
10. Пара полинуклеотидных праймеров по п.2, в которой первый полинуклеотидный праймер имеет последовательность, представленную в SEQ ID NO: 45, а второй полинуклеотидный праймер имеет последовательность, представленную в SEQ ID NO: 27.
11. Набор для выявления ДНК, который содержит по меньшей мере одну молекулу ДНК, несущую нуклеотидную последовательность достаточной длины, которая представляет собой или является комплементарной SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3 или SEQ ID NO: 4, которая функционирует в качестве ДНК праймера или зонда, специфического для варианта кукурузы MIR604.
12. Растение кукурузы MIR604, устойчивое к блошке длинноусой, которое содержит нуклеотидную последовательность, представленную в SEQ ID NO: 1, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4, или их комплементы.
13. Способ выявления присутствия в биологическом образце ДНК варианта кукурузы MIR604, заключающийся в том, что:
(а) приводят в контакт образец с первым полинуклеотидным праймером и вторым полинуклеотидным праймером, которые функционируют вместе в реакции амплификации нуклеиновой кислоты в присутствии ДНК-матрицы из варианта кукурузы MIR604 с образованием ампликона, который предназначен для определения варианта кукурузы MIR604;
(б) осуществляют реакцию амплификации нуклеиновой кислоты, получая тем самым ампликон; и
(в) выявляют ампликон.
(а) приводят в контакт образец с первым полинуклеотидным праймером и вторым полинуклеотидным праймером, которые функционируют вместе в реакции амплификации нуклеиновой кислоты в присутствии ДНК-матрицы из варианта кукурузы MIR604 с образованием ампликона, который предназначен для определения варианта кукурузы MIR604;
(б) осуществляют реакцию амплификации нуклеиновой кислоты, получая тем самым ампликон; и
(в) выявляют ампликон.
14. Способ по п.13, в котором ампликон содержит нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: l, SEQ ID NO: 2, SEQ ID NO: 3, SEQ ID NO: 4 и их комплементы.
15. Способ по п.13, в котором ампликон содержит SEQ ID NO: 1, где первую полинуклеотидную праймерную последовательность выбирают из группы, включающей SEQ ID NO: 7-SEQ ID NO: 15 и их комплементы, где вторую полинуклеотидную праймерную последовательность выбирают из группы, включающей SEQ ID NO: 16-SEQ ID NO: 38 и их комплементы.
16. Способ по п.13, в котором ампликон содержит SEQ ID NO: 2, где первую полинуклеотидную праймерную последовательность выбирают из группы, включающей SEQ ID NO: 39-SEQ ID NO: 46 и их комплементы, где вторую полинуклеотидную праймерную последовательность выбирают из группы, включающей SEQ ID NO: 16-SEQ ID NO: 38 и их комплементы.
17. Способ выявления в биологическом образце ДНК варианта кукурузы MIR604, заключающийся в том, что:
(а) приводят в контакт образец, содержащий ДНК, с зондом, который гибридизуется в строгих условиях гибридизации и отмывки с ДНК MIR604 и не гибридизуется в строгих условиях гибридизации и отмывки с ДНК растения кукурузы, отличного от варианта MIR604;
(б) подвергают образец и зонд гибридизации и отмывке в строгих условиях; и
(в) выявляют гибридизацию зонда с ДНК варианта MIR604.
(а) приводят в контакт образец, содержащий ДНК, с зондом, который гибридизуется в строгих условиях гибридизации и отмывки с ДНК MIR604 и не гибридизуется в строгих условиях гибридизации и отмывки с ДНК растения кукурузы, отличного от варианта MIR604;
(б) подвергают образец и зонд гибридизации и отмывке в строгих условиях; и
(в) выявляют гибридизацию зонда с ДНК варианта MIR604.
18. Биологический образец, предназначенный для определения растения кукурузы MIR604, ткани или семян, где образец содержит нуклеотидную последовательность, которая представляет собой или является комплементарной нуклеотидной последовательностью, выбранной из группы, включающей SEQ ID NO: 1 и SEQ ID NO: 2, и где последовательность можно выявлять в образце с помощью метода амплификации нуклеиновой кислоты или гибридизации нуклеиновой кислоты.
19. Биологический образец по п.18, который выбирают из группы, включающей кукурузную муку, кукурузную мучку, кукурузный сироп, кукурузное масло, кукурузный крахмал и зерновые продукты, которые полностью или частично состоят из полученных из кукурузы побочных продуктов.
20. Экстракт для определения растения кукурузы MIR604, ткани или семян, который содержит нуклеотидную последовательность, которая представляет собой или является комплементарной нуклеотидной последовательностью, выбранной из группы, включающей SEQ ID NO: 1 и SEQ ID NO: 2, где последовательность можно выявлять с помощью метода амплификации нуклеиновой кислоты или гибридизации нуклеиновой кислоты.
21. Экстракт по п.20, где образец выбирают из группы, включающей кукурузную муку, кукурузную мучку, кукурузный сироп, кукурузное масло, кукурузный крахмал и зерновые продукты, которые полностью или частично состоят из полученных из кукурузы побочных продуктов.
22. Способ получения растения кукурузы, устойчивого по меньшей мере к блошке длинноусой, заключающийся в том, что:
(а) осуществляют половое скрещивание первого родительского растения кукурузы со вторым родительским растением кукурузы, где первое или второе родительское растение кукурузы содержит ДНК варианта кукурузы MIR604, получая тем самым множество растений поколения первой генерации;
(б) отбирают растение поколения первой генерации, обладающее устойчивостью к заражению по меньшей мере блошкой длинноусой;
(в) осуществляют самоопыление растения поколения первой генерации, получая тем самым множество растений поколения второй генерации;
(г) отбирают из растений поколения второй генерации растение, которое обладает устойчивостью к заражению по меньшей мере блошкой длинноусой, где растения поколения второй генерации содержат нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 1 и SEQ ID NO: 2.
(а) осуществляют половое скрещивание первого родительского растения кукурузы со вторым родительским растением кукурузы, где первое или второе родительское растение кукурузы содержит ДНК варианта кукурузы MIR604, получая тем самым множество растений поколения первой генерации;
(б) отбирают растение поколения первой генерации, обладающее устойчивостью к заражению по меньшей мере блошкой длинноусой;
(в) осуществляют самоопыление растения поколения первой генерации, получая тем самым множество растений поколения второй генерации;
(г) отбирают из растений поколения второй генерации растение, которое обладает устойчивостью к заражению по меньшей мере блошкой длинноусой, где растения поколения второй генерации содержат нуклеотидную последовательность, выбранную из группы, включающей SEQ ID NO: 1 и SEQ ID NO: 2.
