Abstract
치환된 2-(2,6-다이옥소피페리딘-3-일)프탈이미드와 치환된 1-옥소-2-(2,6-다이옥소피페리딘-3-일)아이소인돌린은 포유동물에서 TNFα의 농도를 감소시킨다. 대표적인 화합물은 1-옥소-2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4,5,6,7-테트라플루오로아이소인돌린이다.

종양괴사인자α또는 TNFα는 다수의 면역자극제들에 대한 반응에서 단핵식세포(mononuclear phagocyte)에 의하여 최초로 방출되는 사이토카인(cytokine)이다. TNFα는 동물이나 사람에게 투여될 때, 염증, 열, 심장혈관의 변화, 출혈, 응고 및 급성감염과 쇼크상태에서 나타나는 것과 유사한 급성반응들을 일으킨다. 그러므로 과도하거나 조절되지 않는 TNFα 생성은 다수의 질병상태와 연관되어 왔다. 이러한 질병들에는 내독혈증(endotoxemia) 및/또는 중독성 쇼크증후군{Tracey et al., Nature 330, 662-664(1987) 및 Hinshaw et al., Circ. Shock 30, 279-292(1990)}; 악태증(cachexia){Dezube et al., Lancet, 335 (8690), 662 (1990)} 및 12,000pg/mL을 초과하는 TNFα농도가 ARDS환자들의 폐흡기에서 검출되는 성인호흡곤란증후군이 포함된다{Millar et al., Lancet 2(8665), 712-714 (1989)}. 재조합 TNFα의 체계적 주입도 ARDS에서 전형적으로 나타나는 변화들을 일으킨다{Ferrai-Baliviera et al., Arch. TNFα는 관절염을 포함하는 골흡수성 질환들과 관련되는 것으로 보인다. 백혈구는 활성화되었을 때 골흡수를 일으킬 것이며, 이는 TNFα가 관여한다는 데이터를 제시하는 활성이다{Bertolini et al., Nature 319, 516-518 (1986) 및 Johnson et al., Endocrinology 124(3), 1424-1427(1989)}. 또한 TNFα는 용골세포(osteoblast)의 기능저해와 관련된 활성화와 용골세포형성을 자극하여 실험실내 및 생체내 골형성을 저해하고 골흡수를 자극하는 것으로 보인다. TNFα는 많은 골흡수성 질환들과 관련될 수 있지만, 그중 질병과 가장 밀접한 관련은 종양이나 숙주조직에 의한 TNFα의 생산과 과칼슘혈증(hypercalcemia)과 관련된 악성종양간의 관계이다{Calci. Tissue Int. 뇌말라리아는 TNFα의 고혈중 농도와 연관된 치명적인 초급성 신경성 증후군이고 말라리아환자들에게서 발생하는 가장 심각한 후유증상이다. 혈청내 TNFα의 농도는 질병의 심각성과 급성 말리리아에 감염된 환자의 예후와 직접적으로 관련되어 있다{Grau et al., N. Engl. J. Med. 320(24), 1586-1591(1989)}.
대식세포(macrophage)에 의해 유도되는 혈관형성은 TNFα에 의하여 매개되는 것으로 알려져 있다. 라이보비크(Liebovich)등{Nature, 329, 630-632(1987)}은 매우 낮은 양의 TNFα를 투여할 때 쥐의 각막(cornea)과 분화중인 닭의 융모요막(chorioallantoic membrane)에서 생체내 모세혈관형성이 유도됨을 밝혔고, 이는 TNFα가 염증, 상처재생 및 종양증식에서 혈관생성을 유도하는 것과 관련된 물질 중 하나임을 나타낸다. 또한 TNFα의 생성은 암의 조건들, 특히 유도된 종양들과 관련되어 있다{Ching et al., Brit. J. Cancer, (1955) 72, 339-343 및 Koch, Progress in Medicinal Chemistry, 22, 166-242(1985)}.
또한 TNFα는 만성 폐렴성 질환들과 관련되어 있다. 실리카입자들이 축적되면 섬유성 반응에 의하여 유도된 침투성 호흡불능질환인 규분증(silicosis)이 유도된다. TNFα에 대한 항체는 쥐에서 실리카에 의해 유도되는 폐섬유증(lung fibrosis)을 완전히 차단하였다{Pignet et al., Nature, 344:245-247 (1990)}. (혈청과 분리된 대식세포들에서) 고농도의 TNFα의 생성은 실리카와 석면에 의하여 유도된 섬유증의 동물모델들에서 제시되어 왔다{Bissonnette et al., Inflammation 13(3), 329-339 (1989)}. 폐유육종증(lung sarcoidosis)환자들의 폐포에 있는 대식세포는 정상적인 공여자의 대식세포와 비교할 때 과량의 TNFα를 자발적으로 배출하는 것으로 밝혀졌다{Baughman et al., J. Lab. Clin. Med. 115(1), 36-42 (1990)}.
또한 TNFα는 재주입 손상이라 불리는 재주입으로 인한 염증반응과 관련되어 있고 혈액흐름이 감소된 후 발생하는 조직손상의 주된 원인이다{Vedder et al., PNAS 87, 2643-2646 (1990)}. 또한 TNFα는 내피세포의 특성을 변화시키고 트롬보모듈린(thrombomodulin)의 발현을 감소조절할 뿐만 아니라 항응고성 단백질 C 경로를 억제시키고 조직인자의 응고전구체 활성을 증가시키는 것과 같은 다양한 응고전구체 활성을 가진다{Sherry et al., J. Cell Biol. 107, 1269-1277 (1988)}. TNFα는 (염증반응의 초기단계 동안) 초기 생성과 함께 심근경색, 졸중 및 순환성 쇼크를 포함하되 이에 한정되지 않는 여러 중요한 질병들에서 조직손상의 유사매개체로 만드는 염증활성의 전활성을 가진다. 세포간 유착분자(ICAM, intercellular adhesion molecule) 또는 내피세포의 내피백혈구유착분자(ELAM, endothelial leukocyte adhesion molecule)와 같은 TNFα에 의해 유도된 유착분자의 발현도 중요할 수 있다{Munro et al., Am. J. Path. 135(1), 121-132 (1989)}.
단일클론 항-TNFα 항체들로 TNFα를 차단하는 것이 류마티스성 관절염{Elliot et al., Int. J. Pharmac. 1995 17(2), 141-145}과 크론병{von Dullemen et al., Gastroenterology, 1995 109(1), 129-135}에 도움이 되는 것으로 나타났다.
더욱이 지금은 TNFα가 HIV-1의 활성화를 포함하는 레트로바이러스 복제의 강력한 활성인자임이 알려져 있다{Duh et al., Proc. Nat. Acad. Sci. 86, 5974-5978 (1989);Poll et al., Proc. Nat. Acad. Sci. 87, 782-785 (1990);Monto et al., Blood 79, 2670(1990);Clouse et al., J. Immunol. 142, 431-438 (1989);Poll et al., AIDS Res. Hum. Retrovirus, 191-197 (1992)}. AIDS는 사람의 면역결핍바이러스(human immunodeficiency virus, HIV)가 T 림프구에 감염된 결과이다. HIV-1, HIV-2와 HIV-3의 적어도 3종류 또는 3개 변종의 HIV가 확인되었다. HIV감염의 결과로서 T세포를 매개로 한 면역성이 손상되고 감염된 사람들은 심각한 기회감염 및/또는 특이적 종양(neoplasm)을 나타낸다. T 림프구에 HIV가 감염되기 위해서는 T 림프구의 활성화가 필요하다. T세포가 활성화된 후 T림프구를 감염시키는 HIV-1, HIV-2와 같은 다른 바이러스들과 그러한 바이러스성 단백질의 발현 및/또는 복제는 그러한 T세포 활성화에 의하여 매개되거나 유지된다. 일단 활성화된 T 림프구에 HIV가 감염되면, T 림프구는 HIV 유전자발현 및/또는 HIV 복제를 허용하기 위하여 활성화된 상태를 계속 유지해야만 한다. 사이토카인, 특히 TNFα는 T 림프구 활성화를 유지하는 역할을 함으로써 활성화된 T세포에 의하여 매개되는 HIV 단백질의 발현 및/또는 바이러스복제와 관련되어 있다. 따라서 HIV로 감염된 환자에서 사이토카인의 생성, 특히 TNFα의 생성의 억제 또는 저해와 같은 사이토카인 활성의 방해는 HIV 감염에 의하여 일어나는 T 림프구의 유지를 제한하는 것을 돕는다.
단핵구, 대식세포 및 쿠퍼 세포(kupffer cell)와 글리얼 세포(glial cell)와 같은 관련세포들도 HIV감염의 유지에 관련되어 있다. T세포와 마찬가지로 이 세포들은 바이러스 복제의 표적이고 바이러스 복제의 정도는 그 세포들의 활성화상태에 의존한다{Rosenberg et al., The Immunopathogenesis of HIV Infection, Advances in Immunology, 57 (1989)}. TNFα와 같은 사이토카인은 단핵구 및/또는 대식세포에서 HIV 복제를 활성화시키는 것으로 나타났고{Poli et al., Proc. Natl. Acad. Sci., 87, 782-784 (1990)}, 따라서 사이토카인의 생산 또는 활성의 방지 또는 저해는 T세포들에 대한 HIV 진행을 제한하는 것을 돕는다. 추가연구의 결과, TNFα가 시험관내 HIV의 활성화에서 공통인자임이 확인되었고 세포의 세포질에서 발견되는 핵내 조절단백질을 통한 명확한 활성메카니즘이 제시되었다(Osborn, et al., PNAS 86 2336-2340). 이러한 증거는 TNFα합성의 감소가 HIV 감염에서 전사와 그에 따른 바이러스 생산을 감소시킴으로써 항바이러스효과를 가질 수 있음을 제시한다.
T세포와 대식세포주의 잠복성 HIV의 AIDS 바이러스 복제는 TNFα에 의하여 유도될 수 있다{Folks et al., PNAS 86, 2365-2368 (1989)}. 세포의 세포질에서 발견되고 조절유전자서열(LTR)에 결합하여 HIV 복제를 촉진하는 유전자조절단백질(NFκB)를 활성화하는 TNFα의 능력에 의하여 활성을 유도하는 바이러스에 대한 분자메카니즘이 제시된다{Osborn et al., PNAS 86, 2336-2340 (1989)}. 환자의 말초혈액 단핵구에서 증가된 혈청 TNFα와 고농도의 자발적 TNFα생산에 의하여 악태증과 관련된 AIDS에서의 TNFα가 제시되었다{Wright et al., J. Immunol. 141(1), 99-104 (1988)}. TNFα는 거세포 바이러스(CMV), 인플루엔자 바이러스, 아데노바이러스 및 헤르페스족(herpes family)과 같은 다른 바이러스 감염에 대해 기재된 바와 비슷한 이유로 다양한 작용에 관련되어 왔다.
핵내 인자 κB(NFκB)는 다상유전의 전사활성인자이다{Lenardo, et al., Cell 1989, 58, 227-29). NFκB는 다양한 질병과 염증상태에서 전사활성인자로서 관련되어 왔고 TNFα에 대한 제한뿐 아니라 HIV전사의 활성인자로 작용하는 것을 포함하여 사이토카인의 농도를 조절하는 것으로 생각된다{(Dbaibo, et al., J. Biol. Chem. 1993, 17762-66, Duh et al., Proc. Natl. Acad. Sci. 1989, 86, 5974-78; Bachelerie et al., Nature 1991, 350, 709-12; Boswas et al., J. Acquired Immune Deficiency Syndrome 1993, 6, 778-786; Suzuki et al., Biochem. 및 Biophys. Res. Comm. 1993, 193, 277-83; Suzuki et al., Biochem 및 Biophys. Res. Comm. 1992, 189, 1709-15; Suzuki et al., Biochem. Mol. Bio. Int. 1993, 31(4), 693-700; Shakhov et al., Proc. Natl. Acad. Sci. USA 1990, 171, 35-47; 및 Staal et al., Proc. Natl. Acad. Sci. TNFα와 NFκB의 농도는 상호의 루프에 대해 영향받는다. 이상에서 기재된 바와 같이 본 발명의 화합물은 TNFα와 NFκB의 농도에 영향을 미친다.
많은 세포내 기능들은 아데노신 3',5'-사이클릭 모노포스페이트(cAMP)의 농도에 의하여 매개된다. 그러한 세포내 기능들은 염증성 상태와 천식, 염증 및 다른 상태를 포함하는 질병의 원인이 된다{Lowe 및 Cheng, Drugs of the Future, 17(9) 799-807, 1992}. 염증성 백혈구에서 cAMP의 증가는 그들의 활성과 그에 따른 TNFα와 NFκB를 포함하는 염증성 매개체들의 배출을 저해한다. 또한 cAMP의 농도증가는 평활근의 이완을 유도한다.
그러므로 감소한 TNFα 농도 및/또는 증가한 cAMP 농도는 많은 염증성, 감염성, 면역학적 및 악성질환들의 치료에 대한 가치있는 치료전략을 구성한다. 이러한 질환에는 패혈증 쇼크, 패혈증, 내독소 쇼크, 혈액투석쇼크 및 패혈증증후군, 후간헐성 재주입손상, 말라리아, 미코박테리아 감염, 뇌막염, 건선, 출혈성 심장마비, 섬유성질환, 악태증, 이식거부반응, 암, 자기면역질환, AIDS의 기회감염, 류마티스성 관절염, 류마티스성 척추염, 골관절염, 다른 관절질환, 크론병, 궤양성 대장염, 다발성 경화증, 전신홍반성 낭창, 나병의 ENL, 방사능증 및 과산소에 의한 폐포손상 등이 포함된다. TNFα의 효과를 억제하기 위하여 이전에는 덱사메타손 (dexamethasone)과 프레드니솔론(prednisolone)과 같은 스테로이드를 이용하는 것에서부터 다클론 및 단일클론 항체를 사용하는 것에까지 노력을 기울여 왔다{Beutler et al., Science 234, 470-474 (1985); WO92/11383}.
본 발명은 치환된 2-(2,6-다이옥소피페리딘-3-일)-프탈이미드와 치환된 2-(2,6-다이옥소피페리딘-3-일)-1-옥소아이소인돌린, 그들의 투여를 통하여 포유동물에서의 종양괴사인자α(tumor necrosis factor α)의 농도를 감소시키며 염증성 질병 및 자가면역성 질병을 치료하는 방법 및 그러한 유도체들의 약학적 조성물에 관한 것이다.
본 발명은 여기에서 보다 충분히 설명되는 어떤 종류의 비폴리펩타이드화합물들이 TNFα의 농도를 감소시킨다는 발견에 근거한다.
특히 본 발명은 화학식 Ⅰ에서:
X와 Y중 하나는 C=O이고 X와 Y중 다른 하나는 C=O 또는 CH2;
(i) 다른 것들과 독립적으로 각각의 R1, R2, R3및 R4는 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시 또는 (ii) R1, R2, R3및 R4중 하나는 -NHR5이고 R1, R2, R3및 R4의 나머지는 수소이고;
R5는 수소 또는 1-8개 탄소원자의 알킬이고;
R6은 수소, 1-8개 탄소원자의 알킬, 벤조, 클로로 또는 플루오로이고;
R7은 m-페닐렌, p-페닐렌 또는 n이 0-4의 값을 가지는 -(CnH2n)-이고;
R8과 R9는 각각 독립적으로 수소 또는 1-8개 탄소원자의 알킬이거나 또는 R8과 R9는 함께 테트라메틸렌, 펜타메틸렌, 헥사메틸렌 또는 X가 -O-, -S-또는 -NH-인 -CH2CH2XCH2CH2-이고;
R10은 수소, 1-8개 탄소원자의 알킬 또는 페닐이며;
(b) 양자화될 수 있는 질소원자를 가진 상기 화학식 Ⅰ의 화합물들의 산추가염에 관한 것이다.
첫 번째 바람직한 그룹의 화합물들은 R1, R2, R3, R4및 R6중 적어도 하나는 수소 이외의 다른 것을 가지는 상기 화학식 I의 화합물이다. 이 중, 바람직한 그룹의 화합물들은 R1, R2, R3및 R4이 서로 독립적으로 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시이고; R6가 수소, 메틸, 에틸 또는 프로필이고; R8과 R9는 각각 독립적으로 수소 또는 메틸이고; R10은 수소인 화합물이다. 이러한 화합물들 중에서 첫 번째로 바람직한 서브그룹은 R7이 m-페닐렌 또는 p-페닐렌인 상기 화합물들이며, 두 번째로 바람직한 서브그룹은 R7이 0-4의 n값을 가지는 -(CnH2n)-인 상기 화합물들이다.
다음으로 바람직한 그룹의 화합물들은 R1, R2, R3및 R4중 하나가 -NH2이고 그 나머지는 수소이며; R6이 수소, 메틸, 에틸 또는 프로필이고; R8과 R9는 각각 독립적으로 수소 또는 메틸이고; R10은 수소인 상기 화학식 I의 화합물이다. 이러한 화합물들 중에서 첫 번째로 바람직한 서브그룹은 R7이 m-페닐렌 또는 p-페닐렌인 상기 화합물들이며, 두 번째로 바람직한 서브그룹은 R7이 0-4의 n값을 가지는 -(CnH2n)-인 상기 화합물들이다.
알킬은 1-8개의 탄소원자를 포함하는 1가의 포화된 가지가 있거나 똑바른 탄화수소사슬을 나타낸다. 그러한 알킬그룹으로서 대표적인 것은 메틸, 에틸, 프로필, 아이소프로필, 뷰틸, 아이소뷰틸, sec-뷰틸 및 tert-뷰틸이다. 알콕시는 에테르성 산소원자를 통하여 분자의 잔기에 결합된 알킬그룹을 말한다. 그러한 알콕시그룹의 대표적인 것은 메톡시, 에톡시, 프로폭시, 아이소프로폭시, 뷰톡시, 아이소뷰톡시, sec-뷰톡시 및 tert-뷰톡시이다. 바람직하게는 R1, R2, R3및 R4는 클로로, 플루오로, 메틸 또는 메톡시이다.
화학식 I의 화합물들은 자격이 있는 전문가의 감독하에서, TNFα의 바람직하지 못한 효과들을 저해하기 위하여 사용된다. 그 화합물들은 단독으로 또는 항생제, 스테로이드 등을 포함하는 다른 치료제와 함께 경구, 직장, 또는 비경구적으로 치료가 필요한 포유동물에 투여될 수 있다.
본 발명의 화합물들은 각각 헤르페스 바이러스에 의한 질병, 바이러스성 결막염, 건선, 아토피성 피부염 등의 바이러스 감염들과 같이 과량의 TNFα생성에 의하여 매개되거나 악화되는 풍토병의 치료 또는 예방에 국부적으로 사용될 수 있다.
또한 그 화합물들은 TNFα생성의 방지 또는 저해를 필요로 하는 사람 이외의 포유동물들의 수의학적 치료에도 사용될 수 있다. 동물에서 치료적 또는 예방적으로 치료를 요하는 TNFα에 의하여 매개된 질병에는 특정 바이러스 감염들만이 아니라 상기에 기재된 질병들과 같은 질병상태들도 포함된다. 예를 들면 다른 렌티바이러스(lentivirus)들 뿐만 아니라 고양이과 동물의 면역결핍바이러스, 말의 감염성 빈혈증 바이러스, 염소의 관절염 바이러스, 비스나 바이러스(visna virus) 및 매디 바이러스(maedi virus)가 포함된다.
그 화합물들은 폼알데하이드를 화학식 ⅡA의 중간물질과 개시반응시켜 제조할 수 있다.
상기 화학식 ⅡA에서 X와 Y는 상기에 정의된 바와 같고;
다른 것들과 독립적으로 각각의 R1, R2, R3및 R4는 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시 또는 (ii) R1, R2, R3및 R4중 하나는 나이트로 또는 보호된 아미노이고 R1, R2, R3및 R4의 나머지는 수소이며;
R6은 수소, 1-8개 탄소원자의 알킬, 벤조, 클로로 또는 플루오로이다.
그 결과물인 식 Ⅲ의 N-하이드록시메틸 중간물질과 식 Ⅳ의 카복실산 유도체를 일반적으로 알려진 방법을 사용하여 결합한다.
상기 반응식 1에서 Hal은 클로로, 브로모 또는 아이오도와 같은 반응적인 할로겐이다.
여기서 사용되는 보호그룹들은 최종 치료화합물들에서는 일반적으로 발견되지 않고 보호되지 않으면 화학적 조작과정에서 변경될 수 있는 그룹들을 보호하기 위하여 합성의 어떤 단계에서 계획적으로 도입되는 그룹들을 의미한다. 그러한 보호그룹들은 합성의 이후 단계에서 제거되고 따라서 그러한 보호그룹을 가진 화합물들은 (일부 유도체들이 생물학적 활성을 나타내지 않더라도) 처음에는 화학적 중간물질들로 중요하다. 따라서 그 보호그룹의 정확한 구조는 중요하지 않다. 그러한 보호그룹들을 형성하고 제거하기 위한 많은 반응들은 본 명세서에 인용에 의하여 포함된 간행물들, 예를 들어 "유기화학에서의 보호그룹(Protective Groups in Organic Chemistry)", Plenum Press, 런던 및 뉴욕, 1973; Greene, Th. 아미노그룹은 마일드한 조건에서 선택적으로 제거할 수 있는 아실그룹, 특히 벤질옥시카본일, 폼일을 이용하거나, 카본일그룹의 1- 또는 α위치에 가지가 있는 저급 알카노일그룹, 특히 예를 들면 트라이플루오로아세틸(trifluoroacetyl)과 같은 카본일그룹의 α위치가 치환된 저급 알카노일 그룹인 피발로일(pivaloyl)과 같은 3차 알카노일(alkanoyl)을 이용한 아미드로 보호될 수 있다.
결합물질은 다이사이클로헥실카보드이미드와 N,N'-카본일다이이미다졸과 같은 시약을 포함한다.
결합반응 후 식 Ⅴ의 화합물을 예를 들어 소듐 아이오다이드의 존재하에서 아민과 반응시키는 것과 같이 종래의 방법으로 아민화할 수 있다.
선택적으로, 식 Ⅲ의 화합물은 식 ⅣA의 보호된 아미노카복실산과 반응할 수 있다.
상기 반응식 2에서 Z는 보호된 아미노 그룹이다.
이러한 결합반응 후, 아미노 보호그룹 Z는 제거된다.

화학식 ⅡA의 중간물질 중 일부는 본 명세서에 인용에 의하여 포함된 설명서, 출원 일련번호 08/790,258과 08/701,494에 기재되어 있다. 더욱이 R1, R2, R3및 R4치환분으로 적절히 치환된 알킬 o-브로모메틸벤조에이트는 트라이에틸 아민과 같은 산수용체의 존재하에서 α-R6-치환된 α-아미노글루타이미드염과 반응하여 X와 Y중 하나는 C=O이고 다른 하나는 CH2인 화합물을 만들 수 있다.
또한 X와 Y가 모두 C=O인 화학식 ⅡA의 화합물은 아세트산과 소듐 아세테이트의 존재하에서 R1, R2, R3및 R4로 적절히 치환된 프탈릭 안하이드라이드가 α-R6-치환된 α-아미노글루타이미드염과 반응하여 제조될 수 있다.
상기 반응에서 사용된 α-R6-치환된 α-아미노글루타이미드염은 아미노 그룹이 보호된 α-R6-치환된 글루타민을 고리화하여 제조할 수 있다. 고리화반응은 예를 들어 다이메틸아미노피리딘과 같은 산수용체의 존재하에서 N,N'-카본일다이이미다졸로 수행할 수 있다. 반응이 종료되면 보호그룹은 적당한 방식으로 제거할 수 있다. 예를 들어 보호그룹이 N-벤질옥시카본일 그룹이라면 이는 촉매적인 수소첨가에 의해 제거할 수 있다.
α-R6-치환된 글루타민은 아미노 그룹이 보호된 α-R6-치환된 글루탐산 안하이드라이드를 암모니아로 처리하여 제조할 수 있다. 마지막으로, α-R6-치환된 글루탐산 안하이드라이드는 아세틱 안하이드라이드와 상응하는 α-R6-치환된 글루탐산에서 얻을 수 있다.
화학식 Ⅰ및 ⅡB의 화합물들은 카이랄리티(chirality)의 중심을 가지고 광학적 이성체로서 존재할 수 있다. 두 개의 카이랄 중심을 가진 다이아스테레오 이성체(diastereomer)뿐 아니라 이성체들의 라세메이트(racemate)들과 개별적인 이성체들 자체도 본 발명의 범위에 속한다. 라세메이트들은 그대로 사용되거나 카이랄 흡착제를 사용한 크로마토그래피에 의하여 기계적으로 개별적인 이성체들로 분리될 수 있다. 선택적으로 개별적인 이성체들은 실질적으로 서로 독립되기 위하여, 즉 광학적 순도가 95% 이상인 형태로 되기 위해서, 카이랄 형태로 제조되거나 10-장뇌설폰산, 장뇌산, α-브로모장뇌산, 메톡시 아세트산, 탈타산(tartaric acid), 다이아세틸탈타산, 사과산, 피롤리드온-5-카복시산 등의 개별적인 이형질체 (enantiomer)와 같이 카이랄산을 가진 염을 형성한 후 용해된 염기들의 하나 또는 둘 다를 제거하고 그 과정을 선택적으로 반복함으로써 혼합체로부터 화학적으로 분리될 수 있다.
또한 본 발명은 화학식 I 및 ⅡB의 화합물들의 생리학적으로 허용가능한 비독성산 추가염을 포함한다. 그러한 염에는 과염소산, 과브롬산, 인산, 황산, 메탄설폰산, 아세트산, 탈타산, 락트산, 숙신산, 시트르산, 사과산, 말레산, 솔브산, 아코니트산, 살리실산, 프탈산, 엠본산, 에난트산 등과 같은 유기 및 무기산들로부터 유도된 염들이 포함된다.
경구투약형태는 단위투약당 1-100㎎의 약을 포함하는 정제, 캡슐, 당의정 및 유사한 형태의 압축된 약학적 형태들을 포함한다. 20-100㎎/㎖를 포함하는 등장식염수는 근육내, 포막내(intrathecal), 정맥내 및 동맥내 투여경로를 포함하는 비경구투여에 사용될 수 있다. 직장투여는 코코아버터와 같은 종래의 담체로부터 조성된 좌약의 사용을 통하여 효과를 낼 수 있다.

다음의 실시예들은 본 발명의 특징을 더 예시하기 위한 것으로 본 발명의 범위를 한정하는 것으로 해석될 수 없고, 그 범위는 첨부된 특허청구범위에 의해서만 한정된다.
(실시예 1)
N-벤질옥시카본일-α-메틸-글루탐산
2N 수산화나트륨(62mL)에 용해된 α-메틸-D,L-글루탐산(10g, 62mmol)의 교반용액에 벤질 클로로폼에이트(12.7g, 74.4mmol)를 30분 이상 첨가하였다. 첨가가 완료된 후 반응혼합물을 실온에서 3시간동안 교반하였다. 교반하는 동안 2N의 수산화나트륨(33mL)을 첨가하여 pH를 11로 유지하였다. 그 후 반응혼합물을 에테르(60mL)로 추출하였다. 수용성층을 얼음조에서 냉각시키고 4N 염산(34mL)으로 pH =1까지 산성화시켰다. 그 결과 얻어지는 혼합물을 에틸 아세테이트(3x100mL)로 추출하였다. 화합된 에틸 아세테이트 추출물을 소금물(60mL)로 세척하고 건조시켰다(MgSO4). 진공상태에서 용매를 제거하여 오일형태의 N-벤질옥시카본일-α-메틸클루탐산 15.2g(83%)을 얻었다:1H NMR(CDCl3) δ 8.73(m, 5H), 5.77(b,1H), 5.09(s, 2H), 2.45-2.27(m,4H), 2.0(s, 3H).
유사한 방식으로 α-에틸-D,L-글루탐산과 α-프로필-D,L-글루탐산으로부터 각각 N-벤질옥시카본일-α-에틸글루탐산과 N-벤질옥시카본일-α-프로필글루탐산이 얻어진다.
(실시예 2)
N-벤질옥시카본일-α-메틸-글루타믹 안하이드라이드
N-벤질옥시카본일-α-메틸-글루탐산(15g, 51mmol)과 아세틱 안하이드라이드 (65mL)의 교반혼합물을 질소하에서 30분동안 환류시키면서 가열하였다. 반응혼합물을 실온에서 냉각시킨 후 진공에서 농축하여 더 이상 정제하지 않고도 다음 반응에 사용할 수 있는 오일형태의 N-벤질카본일-α-메틸글루타믹 안하이드라이드(15.7g)를 얻었다:1H NMR(CDCl3) δ 7.44-7.26(m, 5H), 5.32-5.30(m,2H), 5.11(s, 1H), 2.69-2.61(m,2H), 2.40-2.30(m, 2H), 1.68(s, 3H).
유사한 방식으로 N-벤질옥시카본일-α-에틸클루탐산과 N-벤질옥시카본일-α-프로필글루탐산으로부터 N-벤질카본일-α-에틸클루타믹 안하이드라이드와 N-벤질카본일-α-프로필클루타믹 안하이드라이드가 각각 얻어진다.
(실시예 3)
N-벤질옥시카본일-α-메틸아이소글루타민
메틸렌 클로라이드(100mL)에 용해시킨 N-벤질카본일-α-메틸글루타믹 안하이드라이드(14.2g, 51.5mmol)의 교반용액을 얼음조에서 냉각시켰다. 냉각시킨 용액에 가스상태의 암모니아를 기포형태로 2시간동안 주입하였다. 반응혼합물을 실온에서 17시간동안 교반하고 물(2 x 50mL)로 추출하였다. 화합된 수용성 추출물을 얼음조에서 냉각시킨 후 4N 염산(32mL)로 pH 1까지 산성화시켰다. 그 결과로 생긴 혼합물을 에틸 아세테이트(3 x 80mL)로 추출하였다. 화합된 에틸 아세테이트 추출물을 소금물(60mL)로 세척한 후 건조시켰다(MgSO4). 진공상태에서 용매를 제거하여 11.5g의 N-벤질옥시카본일-α-아미노-α-메틸아이소글루타민을 얻었다:1H NMR(CDCl3/ DMSO) δ 7.35(m, 5H), 7.01(s,1H), 6.87(s, 1H), 6.29(s,1H), 5.04(s, 2H), 2.24-1.88(m, 4H), 1.53(s, 3H).
유사한 방식으로 N-벤질카본일-α-에틸글루타믹 안하이드라이드 및 N-벤질카본일-α-프로필글루타믹 안하이드라이드로부터 각각 N-벤질옥시카본일-α-아미노-α-에틸아이소글루타민 및 N-벤질옥시카본일-α-아미노-α-프로필아이소글루타민이 얻어진다.
(실시예 4)
N-벤질옥시카본일-α-아미노-α-메틸글루타이미드
테트라하이드로퓨란(50mL)에 용해된 N-벤질옥시카본일-α-메틸아이소글루타민(4.60g, 15.6mmol), 1,1'-카본일다이이미다졸(2.80g, 17.1mmol) 및 4-다이메틸아미노피리딘(0.05g)의 교반혼합물을 질소하에서 환류시키면서 17시간동안 가열하였다. 반응혼합물을 진공에서 농축시켜 오일로 만들었다. 오일을 물(50mL)에 넣어 1시간동안 슬러리로 만들었다. 그 결과 생성된 현탁액을 여과한 후 고체를 물로 세척하고 공기로 건조시켜 흰색 고체상태의 미가공산물 3.8g을 얻었다. 미가공 산물을 플래시 크로마토그래피(메틸렌 클로라이드: 에틸 아세테이트 8:2)로 정제하여 흰색 고체형태의 N-벤질옥시카본일-α-아미노-α메틸글루타이미드 2.3g(50%)을 얻었다: 융점 150.5-152.5℃;1H NMR(CDCl3) δ 8.21(s, 1H), 7.34(s, 5H), 5.59(s, 1H), 5.08(s, 2H), 2.74-2.57(m, 3H), 2.28-2.25(m, 1H), 1.54(s, 3H);13C NMR(CDCl3) δ 174.06, 171.56, 154.68, 135.88, 128.06, 127.69, 127.65, 66.15, 54.79, 29.14, 28.70, 21.98; HPLC: 워터스 노바-팩 C18 칼럼, 4미크론, 3.9x150mm, 1mL/min, 240nm, 20/80 CH3CN/0.1% H3PO4(aq), 7.56min(100%); C14H16N2O4에 대하여 계산된 분석: C, 60.86; H, 5.84; N, 10.14. 실제 분석: C, 60.88; H, 5.72; N, 10.07.

(실시예 5)
α-아미노-α-메틸글루타이미드 하이드로클로라이드
N-벤질옥시카본일-α-아미노-α-메틸글루타이미드(2.3g, 8.3mmol)를 약하게 가열하면서 에탄올(200mL)에 용해시키고 그 결과 얻어진 용액을 실온까지 냉각시켰다. 이 용액에 4N 염산(3mL)을 첨가한 후 10% Pd/C(0.4g)를 첨가하였다. 그 혼합물을 파르장치(Parr apparatus)내 50psi의 수소하에서 3시간동안 수소와 화합시켰다. 그 혼합물에 물(50mL)을 첨가하여 생산물을 용해시켰다. 이 혼합물을 물(50mL)로 세척된 셀라이트 패드(Celite Pad)로 여과하였다. 그 여과물을 진공에서 농축시켜 고체잔류물을 얻었다. 그 고체를 에탄올(20mL)에 넣어 30분동안 슬러리로 만들었다. 그 슬러리를 여과하여 흰색 고체형태의 α-아미노-α-메틸글루타이미드 하이드로클로라이드 1.38g(93%)을 얻었다:1H NMR(DMSO-d6) δ 11.25(s, 1H), 8.92(s, 3H), 2.84-2.51(m, 2H), 2.35-2.09(m, 2H), 1.53(s, 3H); HPLC: 워터스 노바-팩 C18칼럼, 4미크론, 1mL/min, 240nm, 20/80 CH3CN/0.1% H3PO4(aq), 1.03min(94.6%).

3-(3-나이트로프탈이미도)-3-메틸피페리딘-2,6-다이온

3-(3-아미노프탈이미도)-3-메틸피페리딘-2,6-다이온

메틸 2-브로모메틸-3-나이트로벤조에이트

α-아미노-α-메틸글루타이미드 하이드로클로라이드를 동량의 α-아미노-α-에틸글루타이미드 하이드로클로라이드 및 α-아미노-α-프로필글루타이미드 하이드로클로라이드로 치환함으로써 3-(1-옥소-4-나이트로아이소인돌린-1-일)-3-에틸피페리딘-2,6-다이온 및 3-(1-옥소-4-나이트로아이소인돌린-1-일)-3-프로필피페리딘-2,6-다이온이 얻어진다.
(실시예 10)
3-(1-옥소-4-아미노아이소인돌린-1-일)-3-메틸피페리딘-2,6-다이온
3-(1-옥소-4-나이트로아이소인돌린-1-일)-3-메틸피페리딘-2,6-다이온(1.0g, 3.3mmol)을 약하게 가열하면서 메탄올(500mL)에 용해시킨 후 실온까지 냉각시켰다. 질소하에서 이 용액에 10% Pd/C(0.3g)을 첨가하였다. 혼합물을 파르장치내 50psi의 수소하에서 4시간동안 수소와 화합시켰다. 혼합물을 셀라이트를 통과시켜 여과하고 셀라이트를 메탄올(50mL)로 세척하였다. 여과물을 진공에서 농축시켜 회색을 띠는 흰색 고체를 얻었다. 그 고체를 메틸렌 클로라이드(20mL)에 넣어 30분동안 슬러리로 만들었다. 그 후 그 슬러리를 여과하고 그 고체를 건조시켜(60℃, <1mm) 흰색 고체상태의 3-(1-옥소-4-아미노아이소인돌린-1-일)-3-메틸피페리딘-2,6-다이온 0.54g(60%)을 얻었다: 융점 268-270℃;1H NMR(DMSO-d6) δ 10.85(s, 1H), 7.19-7.13(t, J=7.63Hz, 1H), 6.83-6.76(m, 2H), 5.44(s, 2H), 4.41(s, 2H), 2.71-2.49(m, 3H), 1.9-1.8(m, 1H), 1.67(s, 3H);13C NMR(DMSO-d6) δ 173.7, 172.49, 168.0, 143.5, 132.88, 128.78, 125.62, 116.12, 109.92, 56.98, 46.22, 29.04, 27.77, 20.82; HPLC, 워터스 노바-팩/C18칼럼, 4미크론, 1mL/min, 240nm, 20/80 CH3CN/0.1% H3PO4(aq), 1.5min(99.6%); C14H15N3O3에 대하여 계산된 분석: C, 61.53; H, 5.53; N, 15.38. 실제 분석: C, 58.99; H, 5.48; N, 14.29.
3-(1-옥소-4-나이트로아이소인돌린-1-일)-3-에틸피페리딘-2,6-다이온과 3(1-옥소-4-나이트로아이소인돌린-1-일)-3-프로필피페리딘-2,6-다이온으로부터 3-(1-옥소-4-아미노아이소인돌린-1-일)-3-에틸피페리딘-2,6-다이온과 3-(1-옥소-4-아미노아이소인돌린-1-일)-3-프로필피페리딘-2,6-다이온이 각각 유사하게 얻어진다.

각각 50mg의 1-옥소-2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4,5,6,7-테트라플루오로아이소인돌린을 포함하는 정제는 다음의 방법으로 제조될 수 있다:

모든 고체성분들은 먼저 0.25mm 메쉬폭을 가진 체로 거른다. 만니톨과 락토오즈를 혼합하고, 젤라틴 용액을 첨가하여 알갱이로 만들고 2mm 메쉬폭을 가진 체로 걸러, 50℃에서 건조시키고 다시 1.7mm 메쉬폭을 가진 체로 거른다. 2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4-아미노프탈이미드, 글리신과 사카린을 조심스럽게 혼합하고, 만니톨, 락토오즈알갱이, 스테알산 및 활성을 첨가하여 전체를 완전히 혼합한 후 압착하여 양면이 오목하고 윗면에 단선 그루브를 가진 약 10mm 직경의 정제로 만든다.

각각 10mg의 2-(2,6-다이옥소에틸피페리딘-3-일)-4-아미노프탈이미드를 포함하는 정제는 다음의 방법으로 제조될 수 있다:

각각 100mg의 1-옥소-2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4,5,6,7-테트라플루오로아이소인돌린을 포함하는 젤라틴 건조캡슐은 다음의 방법으로 제조될 수 있다:

미세결정형 셀룰로오즈.............................30.0g
라우릴 황산나트륨..................................2.0g

라우릴 황산나트륨을 0.2mm 메쉬폭을 가진 체로 걸러 1-옥소-2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4,5,6,7-테트라플루오로아이소인돌린과 10분 동안 혼합한다. 미세결정형 셀룰로오즈를 0.9mm 메쉬폭을 가진 체로 걸러 첨가하고, 전체를 다시 10분동안 잘 혼합한다. 마지막으로 마그네슘 스테아레이트를 0.8mm 폭을 가진 체로 걸러서 추가하여 3분간 혼합한 후 혼합물을 크기가 0(늘어난)인 젤라틴 건조캡슐에 각각 140mg씩 넣는다.

0.2% 주사액이나 주입액은 예를 들어 다음의 방법으로 제조될 수 있다:

1-옥소-2-(2,6-다이옥소-3-메틸피페리딘-3-일)-4,5,6,7-테트라플루오로아이소인돌린을 1000mL의 물에 용해시켜 미세필터로 여과한다. 버퍼용액을 첨가하고 물을 넣어 전체가 2500mL가 되도록 만든다. 1회투약 단위형태를 만들기 위하여 각각 1.0 또는 2.5mL를 (각각 2.0 또는 5.0mg의 이미드를 포함하는) 유리 앰플에 넣는다.
Claims (12)
- (a)다음의 식의 화합물들:상기 식에서:X와 Y중 하나는 C=O이고 X와 Y중 다른 하나는 C=O 또는 CH2;(i) 다른 것들과 독립적으로 각각의 R1, R2, R3및 R4는 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시 또는 (ii) R1, R2, R3및 R4중 하나는 -NHR5이고 R1, R2, R3및 R4의 나머지는 수소이고;R5는 수소, 1-8개 탄소원자의 알킬 또는 CO-R7-CH(R10)NR8R9이고;R6은 수소, 1-8개 탄소원자의 알킬, 벤조, 클로로 또는 플루오로이고;R7은 m-페닐렌, p-페닐렌 또는 n이 0-4의 값을 가지는 -(CnH2n)-이고;R8과 R9는 각각 독립적으로 수소 또는 1-8개 탄소원자의 알킬이거나 또는 R8과 R9는 함께 테트라메틸렌, 펜타메틸렌, 헥사메틸렌 또는 X가 -O-, -S-또는 -NH-인 -CH2CH2XCH2CH2-이고;R10은 수소, 1-8개 탄소원자의 알킬 또는 페닐이며;(b) 양자화될 수 있는 질소원자를 가진 상기 화합물들의 산추가염을 포함하는 그룹으로부터 선택된 2,6-다이옥소피페리딘.
- 제1항에 있어서, 각각 R1, R2, R3및 R4가 서로 독립적으로 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시이고; R6가 수소, 메틸, 에틸 또는 프로필이고; R7은 m-페닐렌 또는 p-페닐렌이고; 각각 R8과 R9는 서로 독립적으로 수소 또는 메틸이며; R10은 수소인 것을 특징으로 하는 화합물.
- 제1항에 있어서, 각각 R1, R2, R3및 R4는 서로 독립적으로 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시이고; R6가 수소, 메틸, 에틸 또는 프로필이고; R7은 n이 0-4의 값을 가지는 -(CnH2n)-이고; 각각 R8과 R9는 서로 독립적으로 수소 또는 메틸이며; R10은 수소인 것을 특징으로 하는 화합물.

- 제6항에 있어서, 각각 R1, R2, R3및 R4는 서로 독립적으로 할로, 1-4개 탄소원자의 알킬 또는 1-4개 탄소원자의 알콕시이고, R6은 메틸, 에틸 또는 프로필인 것을 특징으로 하는 화합물.
- 제6항에 있어서, 각각 R1, R2, R3및 R4중 하나는 -NH2이고 R1, R2, R3및 R4의 나머지는 수소이며, R6은 메틸, 에틸 또는 프로필인 것을 특징으로 하는 화합물.
- 청구항 1에 따른 화합물의 유효량을 포유동물에게 투여하는 것을 포함하는 포유동물에서 TNFα의 바람직하지 못한 농도를 감소시키는 방법.
- 청구항 6에 따른 화합물의 유효량을 포유동물에게 투여하는 것을 포함하는 포유동물에서 TNFα의 바람직하지 못한 농도를 감소시키는 방법.
- 담체와 결합하여 포유동물에서의 TNFα의 농도를 감소시키기 위하여 단일 또는 수회투약량으로 투여하기에 충분한 양의 청구항 1에 따른 화합물을 포함하는 약학적 조성물.
- 담체와 결합하여 포유동물에서의 TNFα의 농도를 감소시키기 위하여 단일 또는 수회투약량으로 투여하기에 충분한 양의 청구항 6에 따른 화합물을 포함하는 약학적 조성물.
