Abstract
Substituert 2-(2,6-dioxopiperidin-3 -yl)-tfalimider. og l-oxo-2-(2,6-dioxopiperdin-3-yl) isoindoliner reduserer nivåene av TNFa hos pattedyr. En typisk utførelse er l-oxo-2-(2,6- dioxo-3-metylpiperidin-3-yl)-4,5,6,7-tetrafluorisoindolin.
Description
Den foreliggende oppfinnelse vedrører 2,6-dioksopiperidiner. Oppfinnelsen angår også farmasøytiske preparater som inneholder disse.
Tumor nekrose faktor a, eller TNfa, er en cytokin som blir frigjort primært av enkjernete fagocytter som svar på et antall immunostimulatorer. Når administrert til dyr eller mennesker, forårsaker den inflammasjon, feber, kardiovaskulaere effekter, blødning, koagulering og akutt faserespons lignende slike som blir observert under akutte infeksjoner og sjokk-tilstander. Usedvanlig stor eller ukontrollert TNfa-produksjon er således innblandet i et antall av sykdomstilstander. Disse innbefatter endotoksemia og/eller toksisk syndrom {Tra-cey et al., Nature 330, 662-664 (1987) og Hinshaw et al., Circ. Shock 30. 279-292 (1990)}; cachexia {Dezube et al., Lancet, 335 (8690), 662 (1990)} og "Adult Respiratory Dis-tress Syndrome" når TNfa konsentrasjon er i overskudd av 12,000 pg/ml hår blitt påvist i pulmonart oppsuget materiale fra ARDS pasienter {Millar et al., Lancet 2(8665), 712-714 (1 989)}. Cerebral malaria er et dødelig hyperakutt neurologisk syndrom forbundet med høye blodnivåer av TNfa og er den mest alvorlig komplikasjon som opptrer hos malaria pasienter. Nivåer av serum TNfa samsvarte direkte med sykdomsgraden og prognosen hos pasienter med akutt malaria angrep { Grau et al, N. Engl. J.
Med 320(24), 1586-1591 (1989)}.
Makrofagindusert angiogenesis er kjent å bli formidlet av TNfa. Leibovich et al. (Nature, 329, 630-632 (1987)} viste at TNfa forårsaker in vivo kapillar blodkardannelse i rottekor-nea og utviklingen av kylling-chorioallantoiske membraner ved meget små doser og antyder at TNfa forårsaker angiogenese ved inflammasjon, sårhelbredelse og tumor vekst. TNfa produksjon har også blitt forbundet med kreftlignende tilstander, spesielt induserte tumorer {Ching et al, Brit J. Cancer, (1955) 72, 339-343, og Koch, Progress in Medicinal Chemistry, 22. 166-242 (1985)}.
TNfa spiller også en rolle i området for kroniske pulmonare
inflammatoriske sykdommer. Avsetningen av silikapartikler leder til silikose, en sykdom med gradvis tiltagende åndedrett-svikt forårsaket av en fibrotisk reaksjon. Antikropp til TNfa blokkerte fullstendig silikaforårsaket lungefibrose hos mus
{Pignet et al, Nature, 344:245-247 (1990)}. Høye nivåer av TNfa produksjon (i serumet og isolerte makrofager) har blitt vist i dyreforsøk av silika- og asbestpåvirket fibrose {Bis-sonnette et al. Inflammasjon 13(3), 329-339 (1989)}. Alveola-re makrofager fra pulmonare sarkomatøse pasienter har også blitt funnet spontant å frigi store mengder av TNfa sammen-lignet med makrofager fra normale donatorer {Baughman et al, J. Lab. Clin. Med. 115(1), 36-42 (1990)}.
TNfa er også implisert i betennelsesreaksjonen som følger re-perfusjon, kallet reperfusjonsskade, og er en hovedårsak til vevskader etter blodtap {Vedder et al, PNAS 87, 2643-2646
(1990)}. TNfa endrer også verdiene av endotelceller og har forskjellige prokoagulerende aktiviteter, slik som å produsere en økning i vevsfaktor pro-koagulerende aktivitet og un-dertrykkelse av det antikoagulerende protein C-virkning så vel som nedregulering av ekspresjon av trombomodulin (Sherry et al, J. Cell Biol. 107, 1269-1277 (1988)}. TNfa har proinflammatoriske aktiviteter som sammen med tidligere produksjon (under de begynnende stadier av et inflammatorisk forløp) gjør den til en sannsynlig frembringer av vevsskader ved flere alvorlige sykdommer inkludert, men ikke begrenset til myokardiell infarkt, slag og sirkulasjonssjokk. TNfa-blokkering med monoklonale anti-TNfa antikropper har vist seg å være fordelaktig ved reumatisk arteritt (Elliot et al., Int. J. Pharmac. 1995 17(2), 141-145} og Crohn's sykdom {von Dullemen et al., Gastroenterology, 1995 109(1), 129-135} .
Dessuten har det vært kjent at TNfa er en potent aktivator for retrovirusreplikasjon innbefattet aktivering av HIV-1.

(1992)}. AIDS forårsakes av infeksjon av T-lymfocytter med "Human Immunodeficiency Virus" (HIV). I det minste har tre typer eller stammer av HIV blitt identifisert, dvs, HIV-1, HIV-2 og HIV-3 . Som en konsekvens av HIV-infeksjon, blir T-celleformidlet immunitet svekket, og infiserte individer ma-nifesterer alvorlige opportunistiske infeksjoner og/eller uvanlige neoplasmer. HIV- inntreden i lymfocyttene krever T-lymfocyttaktivering. Andre viruser, slik som HIV-1, HIV-2 in-fiserer T-lymfocytter etter T-celleaktivisering og slik virus protein ekspresjon og/eller replikasjon blir formidlet eller opprettholdt av slik T-celleaktivering. Er en aktivert T-lymfocytt blir infisert med HIV, må T-lymfocytten kontinuer-lig bli opprettholdt i en aktivert tilstand for å tillate HIV-genekspresjon og/eller HIV-replikasjon. Cytokiner, spesielt TNfa, er implisert i aktivert T-celleformidlet HIV-proteinekspresjon og/eller virusreplikasjon ved å spille en rolle ved opprettholdelse av T-lymfocytt aktivering. Derfor, interferens med cytokin aktivitet, slik som ved forhindring eller hemming av cytokin produksjon, spesielt TNfa, hos et HIV-infisert individ, hjelper, understøtter derfor i å be-grense opprettholdelse av T-lymfocytt forårsaket av HIV-infeksjon.
Monocytter, makrofager og beslektede celler, slik som kupf-fer- og glialceller, har også vært implisert i opprettholdelse av HIV-infeksjonen. Disse celler, som T-celler, er mål for virusreplikasjon, og nivået for virusreplikasjon er av-hengig av aktiveringstilstanden til cellene. Cytokiner, slike som TNfa, har vist seg å aktivere HIV-replikasjon i monocytter og/eller makrofager {Poli et al., Proe. Nati. Acad. Sei., 87, 782-784
(1990)},derfor, forhindring eller hemming av cytokinproduk-sjon eller aktivitet hjelper ved begrensning av HIV-progresjon for T-celler.
Ytterligere studier har identifisert TNfa som en vanlig faktor ved aktivering av HIV in vitro og har fremskaffet en klar mekanisme for aktivering via et kjerneregulerende protein funnet i cytoplasma i celler (Osborn, et al., PNAS 86 2336-2340). Denne indikasjon tyder på at en reduksjon av TNfa-syntesen kan ha en antiviruseffekt ved HIV-infeksjoner, ved redusering av transkripsjon og således virusproduksjon.
AIDS virusreplikasjon av latent HIV i T-celle og makrofagre linjer kan påvirkes av TNfa (Folks et al, PNAS 86, 2365-2368
(1989)}. En molekylær mekanisme for viruspåvirkende aktivitet blir formodet av TNfa's evne til å aktivere et genregulerende protein (NFkB) funnet i cytoplasmaet til celler som aktiverer HIV-replikasjon ved binding til virusregulerende gensekvenser (LTR) {Osborn et al, PNAS 86, 2336-2340 (1989)). TNfa i AIDS-assosiert cachexia blir framkalt av forhøyet serum TNfa og
høye nivåer av spontan TNfa-produksjon i periferisk blodmono-cytter fra pasienter (Wright et al, J. Immunol. 141(1), 99-104 (1988)). TNfa er blitt innblandet ved forskjellige roller med andre virusinfeksjoner, slik som cytomegaliaviruset (CMV), influensaviruset, adenoidviruset og herpesvirusene av lignende grunner som de som er anført.
Kjernefaktoren kB (NFkB) er en pleiotropisk transkripsjonal aktivator (Lenardo, et al, Cell 1989, 58. 227-29). NFkB har blitt innblandet som en transkripsjonal aktivator ved en rekke sykdommer og inflammatoriske tilstander og er antatt å regulere cytokine nivåer omfattende, men ikke begrenset til TNfa og kan også være en aktivator for HIV-transkripsjon (Dbaibo, et al, J Biol. Chem. 1993, 17762-66; Duh et al, Proe. Nati. Acad. Sei. 1989, 86, 5974-78; Bachelerie et al, Nature 1991, 350, 709-12; Boswas et al, J. Acquired Immune Deficiency Syndrome 1993, 6, 778-786; Suzuki et. al, Biochem. And Biophys. Res. Comm. 1993, 193, 277-83; Suzuki et al., Biochem. And Biophys. Res Comm. 1992, 189, 1709-15; Suzuki et al, Biochem. Mol. Bio. Int. 1993, 31(4), 693-700; Shakhov et al., Proe. Nati. Acad. Sei. USA 1990, 171, 35-47; og Staal et al, Proe. Nati. Acad. Sei. USA 1990, 87, 9943-47). Inhibering av NFkB- binding kan således regulere transkripsjon av cytokin (e) gen(er) og gjennom denne modulasjon og andre meka-nismer være nyttig ved inhibering av mange sykelige tilstander. Forbindelsene som beskrevet her, kan hemme virkningen av NFkB i kjernen og er således nyttig ved behandlingen av en rekke forskjellige sykdommer omfattende, men ikke begrenset til reumatisk arteritt, reumatisk spondylitt, osteo-arteritt, andre arteritiske tilstander, septisk sjokk, septisk, endotoksisk sjokk, transplantert vertssykdom, avvisning ("wasting"), Crohn's sykdom, inflammatorisk tarmsykdom, ulce-røs colitt, multippel sklerose, systemisk lupus erytematøs, ENL i spedalskhet, HIV, AIDS og opportunistiske infeksjoner i AIDS. TNfa og NFkB nivåer blir influert av en innbyrdes til-bakekobling. Som anført ovenfor, innvirker forbindelsene i henhold til foreliggende oppfinnelse på nivåene av både TNfa og NFkB.
Mange cellefunksjoner blir formidlet av nivåer av adenosin 3<1>,5<1->syklisk monofosfat (cAMP). Slike.cellefunksjoner kan bidra til inflammatoriske tilstander og sykdommer inkludert astma, inflammasjon og andre tilstander (Lowe and Cheng, Drugs of the Future, 17( 9), 799-807, 1992). Det har blitt vist at økningen av cAMP i inflammatoriske leukocytter hemmer deres aktivering og den etterfølgende frigjøring av inflammatoriske formidlere, inkludert TNfa og NFkB. Økede nivåer av cAMP leder også til avslapping av glatt luftveismuskulatur.
Minskning av TNfa nivåer og/eller økning av cAMP nivåer ut-gjør således en verdifull terapeutisk strategi for behandling av mange inflammatoriske, infeksiøse, immunologiske eller on-dartete sykdommer. Disse omfatter, men er ikke begrenset til septisk sjokk, sepsis, endotoksisk sjokk, hemodynamisk sjokk og sepsis syndrom, postischemisk reperfusjonsskade, malaria, mycobakteriell infeksjon, meningitt, psoriasis, kongestiv hjertesvikt, fibrotisk sykdom, cachexia, transplantasjonsav-visning, cancer, autoimmun sykdom, opportunistiske infeksjoner av AIDS, reumatisk arteritt, reumatisk spondylitt, osteoartrititt, andre artrittiske tilstander, Crohn's sykdom, ulcerativ colitt, multippel sklerose, systemisk lupus erytro-matosis, ENL i spedalskhet, bestrålningsskade og hyperoksisk alveolar skade. Tidligere bestrebelser ble ledet mot under-trykkelsen av virkningene av TNfa og har strukket seg fra an-vendelsen av steroider slik som dexamethason og prednisolon, til anvendelse av både polyklonale og monoklonale antikropper {Beutler et al., Science 234, 470-474 (1985); WO 92/1 1383}.
Den foreliggende oppfinnelse er basert på oppdagelsen av at bestemte klasser av ikke-polypeptidforbindelser som mer fullstendig er beskrevet nedenfor, minsker nivåene av TNfa.
Spesielt vedrører oppfinnelsen 2,6-dioksopiperidiner som ka-rakteriseres ved formelen

Betegnelsen alkyl betyr en monovalent mettet forgrenet eller rettkjedet hydrokarbonkjede inneholdende fra 1 til 8 karbonatomer. Representative for slike alkylgrupper er metyl, etyl, propyl, isopropyl, butyl, isobutyl og sek-butyl.
I en foretrukket utførelse angår oppfinnelsen en forbindelse som er karakterisert ved at den er 3(3-aminoftalimid)-3-metyl-piperidin-2,6-dion i racemisk form eller R- eller S-isomerer derav eller syreaddisjonssalt derav.
I en ytterligere utførelse angår oppfinnelsen en forbindelse som er karakterisert ved formelen
i racemisk form eller R- eller S-isomerer derav.
I nok en utførelse angår oppfinnelsen en forbindelse karakterisert ved formelen

Oppfinnelsen angår også et farmasøytisk preparat karakterisert ved at det omfatter en forbindelse ifølge krav 1. Forbindelsene i henhold til formel I blir anvendt, under overoppsyn av kvalifisert personell, til å hemme de uønskede virkninger av TNfa. Forbindelsene kan administreres oralt, rektalt eller parenteralt, alene eller i kombinasjon med andre terapeutiske midler omfattende antibiotika, steroider, etc, til et pattedyr i behov av behandling.
Forbindelsene i henhold til foreliggende oppfinnelse kan også anvendes topikalsk ved behandling eller profylakse av topiske sykdomstilstander formidlet eller forverret av overdreven TNfa-produksjon, henholdsvis, slik som virusinfeksjoner, slike som de som er forårsaket av herpes-viruser, eller virus conjuktivitt, psoriasis, atopisk dermatitt, etc.
Forbindelsene kan også anvendes ved veterinær behandling av pattedyr foruten mennesker ved behov av forebygging eller hemming av TNfa-produksjon. TNfa-formidlende sykdommer for behandling, terapeutisk eller profylaktisk, hos pattedyr ink-luderer sykdomstilstander av den art som beskrevet ovenfor, men spesielt virusinfeksjoner. Eksempler omfatter katteimmun-sviktvirus, hesteinfeksiøs anaemiavirus, caprin arterittvi-rus, visnavirus, og maedi-virus, så vel som andre lentiviru-ser.
I denne forbindelse skal det henvises til en søknad avdelt fra herværende søknad, idet nevnte avdelte søknad er rettet mot 2,6-dioksopiperidin eller R- og S-isomerer derav, karakterisert ved at det er valgt fra gruppen bestående av

Forbindelsene som har et chiralt sentrum kan forefinnes som optiske isomerer. Både racematene av disse isomerer og de individuelle isomerer så vel som diastereomerene, når det forefinnes to chirale sentra, er innen rammen for foreliggende oppfinnelse. Racematene kan anvendes som slike eller kan separeres mekanisk i deres individuelle isomerer slik som ved hjelp av kromatografi ved anvendelse av en chiral absorbent. Alternativt kan de enkelte isomerer fremstilles i chiral form eller separeres kjemisk fra en blanding ved dannelse av salter med en chiral syre, slik som de individuelle enantiomerer av 10-kamfersulfonsyre, kamfersyre, a-bromkamfersyre, metok-syeddiksyre, vinsyre, diacetylvinsyre, maleinsyre (eplesyre), pyrrolidon-5-karboksylsyre og lignende og så frigjøring av en eller begge av de oppløste baser, eventuelt gjenta prosessen, for å oppnå enten én eller begge vesentlig fri fra den andre, det vil si med en optisk renhet på >95 %.
Den foreliggende oppfinnelse vedrører også de fysiologiske akseptable ikke-toksiske syreaddisjonssalter av forbindelsene med formler I og IIB. Slike salter omfatter slike som er av-ledet fra organiske og uorganiske syrer slik som, uten begrensning, saltsyre, hydrobromsyre, fosforsyre, svovelsyre, metaneulfonsyre, eddiksyre, vinsyre, melkesyre, ravsyre, sit-ronsyre, eplesyre, maleinsyre, sorbinsyre, aconitinsyre, sa-licylsyre, ftalsyre, embonsyre, ønantsyre og lignende.
Orale doseringsformer omfatter tabletter, kapsler, dragéer, og lignende formet, sammentrykte farmasøytiske utformninger inneholdende fra 1 til 100 mg av medikamentet per enhetsdose. Isotoniske saltoppløsninger inneholdende fra 20 til 100 mg/ml kan anvendes for parenteral administrering som omfatter intramuskulær, intratekal, intravenøs og intra-arteriell ru-tiner ved administrering. Rektal administrering kan utføres ved bruk av suppositorer formulert fra konvensjonelle bærere slik som kakaosmør.
Farmasøytiske sammensetninger omfatter således én eller flere av forbindelsene med formel I og/eller IIB blandet med i det minste én farmasøytisk akseptabel bærer, fortynningsmiddel eller eksipient. Ved fremstilling av slike sammensetninger blir de aktive ingredienser vanligvis blandet med eller for-tynnet med en eksipient eller innelukket i en slik bærer som kan være i en form av en kapsel eller luktepose. Når eksi-pienten tjener som et fortynningsmiddel, kan det være et faststoff, semifaststoff, eller et materiale i væskeform som virker som en oppløsning, bærer, eller medium for den aktive ingrediens. Sammensetningen kan således være i form av tabletter, piller, pulver, eliksirer, suspensjoner, emulsjo-ner, oppløsninger, siruper, myke og harde gelatinpiller, stikkpiller, sterile injiserbare oppløsninger og sterile pak-kede pulver. Eksempler på egnede eksipienter omfatter laktose, dekstrose, sakkarose, sorbitol, mannitol, stivelse, gum acacia, kalsiumsilikat, mikrokrystallin cellulose, polyvinyl-pyrrolidinon, cellulose, vann, sirup og metylcellulose. Sammensetningene kan dessuten inneholde smøremidler slik som talkum, magnesiumstearat og mineralolje, fuktemidler, emul-gerings- og suspensjonsmidler, konserveringsmidler slik som metyl- og propylhydroksybenzoater, søtningsmidler eller smaksstoffer.
Sammensetningene blir fortrinnsvis formulert i enhetsdose-ringsform, hvilket betyr fysisk diskrete enheter egnet som en enhetsdosering, eller en forutbestemt del av en enhetsdose som kan administreres i en enkel eller multippel doseringskur til humane subjekter og andre pattedyr. Hver enhet inneholder en forutbestemt mengde av aktivt materiale beregnet til å gi den ønskede terapeutiske effekt i en blanding med en egnet farmasøytisk eksipient. Sammensetningene kan formuleres slik at de fremskaffer en øyeblikkelig, vedvarende eller forsinket frigjøring av aktiv ingrediens etter administrering til pasi-enten ved anvendelse av prosedyrer som er velkjente for fag-mannen .
De følgende eksempler vil tjene til ytterligere å illustrere oppfinnelsen, men skal ikke oppfattes som en begrensning av oppfinnelsens ramme, hvilken ramme er definert av de etter-følgende patentkrav.
Eksempel 1
N- benzyloksykarbonyl- a- metyl- glutaminsyre
Til en omrørt oppløsning av a-metyl-D,L-glutaminsyre (10 g, 62 mmol) i 2 N natriumhydroksid (62 ml) ved 0-5 °C ble tilsatt benzyl klorformat (12,7 g, 74,4 mmol) under 30 min. Etter at tilsetningen var fullført, ble reaksjonsblandingen om-rørt ved værelsestemperatur i 3 timer. Under denne tiden ble pH-verdien holdt ved 11 ved tilsetning av 2 N natriumhydroksid (33 ml). Reaksjonsblandingen ble deretter ekstrahert med eter (60 ml). Det vandige sjikt ble kjølt i et isbad og deretter gjort surt med 4 N saltsyre (34 ml) til pH=l. Den resulterende blandingen ble ekstrahert med etylacetat (3 x 100 ml). De forente etylacetatekstraktene ble vasket med salt-oppløsning (60 ml) og tørket (MgS04) . Oppløsningsmidlet ble fjernet i vakuum og ga 15,2 g (83%) av N-benzyloksykarbonyl-a-metylglutaminsyre som en olje: <X>H NMR (CDC13) d 8,73(m, 5H) . 5,77(b, 1H), 5,09(s, 2H), 2,45-2,27(m, 4H), 2,0(s, 3H).
På samme måte ble det av a-etyl-D,L-glutaminsyre og a-propyl-D,L-glutaminsyre oppnådd henholdsvis N-benzyloksykarbonyl-a-etylglutaminsyre og N-benzyloksykarbonyl-a-propylglutaminsyre.
Eksempel 2
N- benzyloksykarbonyl- g- metyl- glutaminanhydrid
En omrørt blanding av N-benzyloksykarbonyl-a-metyl-glutaminsyre (15 g, 51 mmol) og eddiksyreanhydrid (65 ml) ble oppvarmet under tilbakeløp under nitrogen i en tid av 30 min. Reaksjonsblandingen ble kjølt til værelsestemperatur og deretter konsentrert i vakuum for å gi N-benzylkarbonyl-a-metyl-glutaminanhydrid som en olje (15,7 g) som kan anvendes i nes-te reaksjonstrinn uten ytterligere rensning: ■""H NMR (CDCl3)d 7,44-7,26 (m, 5H) , 5,32-5,30 (m, 2H) , 5,11

På en lignende måte ble det av N-benzyloksykarbonyl-a-etyl-glutaminsyre og N-benzyloksykarbonyl-a-propylglutaminsyre oppnådd henholdsvis N-benzylkarbonyl-a-etylglutaminanhydrid og N-benzylkarbonyl-a-propylglutaminanhydrid.
Eksempel 3
N- Benzyloksykarbonyl- g- metylisoglutamin
En omrørt oppløsning av N-benzylkarbonyl-a-metylglutamin-anhydrid (14,2 g, 51,5 mmol) i metylenklorid (100 ml) ble kjølt i et isbad. Gassformet ammoniakk ble boblet inn i den kjølte oppløsningen under 2 timer. Reaksjonsblandingen ble omrørt ved værelsestemperatur i 17 timer og deretter ekstrahert med vann (2 x 50ml). De kombinerte vandige ekstrakter ble kjølt i et isbad og gjort surt med 4 N saltsyre (32 ml) til pH 1. Den resulterende blanding ble ekstrahert med etylacetat (3 x 80 ml). De kombinerte etylacetatekstraktene ble vasket med saltoppløsning (60 ml) og deretter tørket (MgS04) . Oppløsningsmidlet ble fjernet i vakuum og ga 11,5 g N-benzyl-
oksykarbonyl-a-amin-a-metylisoglutamin:

På en lignende måte ble det fra N-benzylkarbonyl-a-etyl-glutamanhydrid og N-benzylkarbonyl-a-propylglutaminanhydrid oppnådd henholdsvis N-benzyloksykarbonyl-a-amino-a-etyl-isoglutamin og N-benzyloksykarbonyl-a-amin-a-propyliso-glutamin.
Eksempel 4
N- Benzyloksykarbonyl- a- amin- ot- metylglutarimid
En omrørt blanding av N-benzyloksykarbonyl-a-metylisoglutamin (4,60 g, 15,6 mmol), 1,1' karbonyldiimidazol (2,80 g, 17,1 mmol), og 4-dimethylaminopyridin (0,05 g) i tetrahydrofuran (50 ml) ble oppvarmet under tilbakeløp under nitrogen i 17 timer. Reaksjonsblandingen ble deretter konsentrert i vakuum til en olje. Oljen ble oppslemmet i vann (50 ml) il time. Den resulterende suspensjon ble filtrert, og de faste stoffer ble vasket med vann og lufttørket og ga 3,8 g av råprodukt som et hvitt fast stoff. På en lignende måte ble det fra N-benzyloksykarbonyl-a-amino-a-etylisoglutamin og N-benzyloksykarbonyl-a-amin-a-propyl-isoglutamin oppnådd henholdsvis N-benzyloksykarbonyl-a-amin-a-etylglutarimid og N-benzyloksykarbonyl-a-amin-a-propylglutarimid.
Eksempel 5
N- benzyloksykarbonyl- a- amino- a- metylglutarimid N-benzyloksykarbonyl-a-amin-a-metylglutarimid (2,3 g, 8,3 mmol) ble oppløst i etanol (200 ml) ved mild oppvarming og den resulterende oppløsning tillatt å kjøle til værelsestemperatur. Til denne oppløsning ble tilsatt 4 N saltsyre (3 ml) fulgt av 10 % Pd/C (0.4 g). Blandingen ble hydrert i et Parr-apparat under 50 psi hydrogen i 3 timer. Til blandingen ble tilsatt vann (50 ml) for å oppløse produktet. Denne blandingen ble filtrert gjennom et "Celite"-filter som ble vasket med vann (50 ml). Filtratet ble konsentrert i vakuum for å gi en fast rest. Det faste stoff ble suspendert i etanol (20 ml) i 30 min. Det ble på lignende måte fra N-benzyloksykarbonyl-a-amin-a-etylglutarimid og N-benzyloksykarbonyl-a-amin-a-propylglutarimid oppnådd henholdsvis a-amin-a-ethylglutarimid-hydroklorid og a-amin-a-propylglutarimidhydroklorid.
Eksempel 6
3-( 3- nitroptalimid)- 3- metylpiperidin- 2, 6- dion En omrørt blanding av a-amino-a-metylglutarimidhydroklorid (1,2 g, 6,7 mmol), 3-nitroptalin anhydrid (1,3 g, 6,7 mmol) og natrium acetat (0,6 g, 7,4 mmol) i eddiksyre (30 ml) ble oppvarmet til tilbakeløp under nitrogen i 6 timer. Blandingen ble deretter kjølt og konsentrert i vakuum. Det resulterende faste stoff ble suspendert i vann (30 ml) og metylenklorid (30 ml) i 30 min. Det ble på lignende måte fra a-amin-a-ethylglutarimidhydro-klorid og a-amin-a-propylglutarimidhydroklorid oppnådd henholdsvis 3-(3-nitroptalimid)-3-etylpiperidin-2,6-dion og 3-(3-nitrophthalimid)-3-propylpiperidin-2,6-dion.
Eksempel 7
3-( 3- Aminoftalimid)- 3- metyl- piperidin- 2, 6- dion
3-(3-nitrofthalimid)-3-methylpiperidin-2,6-dion (0,5 g, 1,57 mmol) ble oppløst i aceton (250 ml) med mild oppvarming og deretter tillatt å kjøle til værelsestemperatur. Til denne oppløsning ble tilsatt 10 % Pd/C (0,1 g) under nitrogen. Blandingen ble hydrert i et Parr-apparat ved 50 psi med hydrogen i 4 timer. Blandingen ble deretter filtrert gjennom "Celite", og filteret ble vasket med aceton (50 ml). Filtratet ble konsentrert i vakuum og ga et gult fast stoff. Det faste stoffet ble suspendert i etylacetat (10 ml) i 30 minutter. Eksempel 8
Mety1- 2- brommety1- 3- ni trobenzoat
En omrørt blanding av metyl 2-brommetyl-3-nitrobenzoat ( 17,6 g, 87,1 mmol) og n-bromravinimid (18,9 g, 105 mmol) i karbon-tetraklorid (243 ml) ble oppvarmet under mildt tilbakeløp med en 100 W lyskolbe plassert i en avstand på 2 cm og strålende på reaksjonsblandingen over natten. Etter 18 timer ble reaksjonsblandingen kjølt til værelsestemperatur og filtrert. Filtratet ble vasket med vann (2 x 12 0 ml), saltoppløsning (120 ml) og tørket (MgSC>4) . Oppløsningsmidlet ble fjernet i vakuum for å gi et gult fast stoff. 3-( l- okso- 4- nitroisoindolin- l- yl)- 3- metylpiperidin- 2, 6- dion Til en omrørt blanding av a-amin-a-metylglutarimidhydroklorid (2,5 g. 14,0 mmol) og metyl-2-brommetyl-3-nitrobenzoat (3,87 g, 14,0 mmol i dimetylformamid (40 ml) ble tilsatt trietyl-amin (3,14g, 30,8 mmol). Ved substituering av ekvivalente mengder av a-amino-a-etyl-glutarimid hydroklorid og a-amino-a-propylglutarimid hydroklorid for a-amino-a-metylglutarimid hydroklorid, blir det oppnådd henholdsvis 3-(l-okso-4-nitroisoindolin-l-yl)-3-etyl-piperidin-2,6-dion og 3-(l-okso-4-nitroisoindolin-l-yl)-3-propylpiperidin-2,6-dion.

3-( l- Okso- 4- aminoisoindolin- 1- yl)- 3- metylpiperidin- 2, 6- dion
3(l-okso-4-nitroisoindolin- 1-yl)-3-metylpiperidin-2,6-dion (l,0g, 3,3 mmol) ble oppløst i metanol (500 ml) med mild oppvarming og tillatt å kjøle til værelsestemperatur. Til denne oppløsning ble tilsatt 10 % Pd/C (0,3 g) under nitrogen. Blandingen ble hydrert i et Parr-apparat ved 50 psi hydrogen i 4 timer. Blandingen ble filtrert gjennom celitt, og celit-

Tabletter, hver inneholdende 50 mg l-okso-2-(2,6-diokso-3-metylpiperidin-3-yl)-4,5,6,7-tetrafluorisoindolin, kan bli fremstilt på følgende måte:
Sammensetning ( for 1000 tabletter)
De faste ingredienser blir først tvunget gjennom en sikt med 0,6 mm maskevidde. Den aktive ingrediens, laktose, talkum, magnesiumstearat og halvdelen av stivelsen blir deretter blandet. Den andre halvdelen av stivelsen blir suspendert i 40 ml vann, og denne suspensjonen blir tilsatt til en kokende oppløsning polyetylenglykol i 100 ml vann. Den resulterende pasta blir tilsatt til de pulverformede substanser, og blandingen blir granulert, dersom nødvendig ved tilsetning av vann. Alle faste ingredienser blir først tvunget gjennom en sikt med 0,25 mm maskevidde. Mannitolen og laktosen blir blandet, granulert med tilsetning av gelatinoppløsning, tvunget gjennom en sikt med 2 mm maskevidde, tørket ved 50 °C og igjen tvunget gjennom en sikt med 1,7 mm maskevidde. 2-(2,6-diokso-3-metylpiperidin-3-yl)-4-aminftalimid, glysinet og sakkarinet blir omhyggelig blandet, mannitolen, laktosegranulatet, stea-rinsyren og talkumet blir tilsatt, og det hele blir blandet grundig og presset til tabletter med diameter på omtrent 10 mm, hvilke tabletter er konkave på begge sider og har et brekkespor på den øvre side.

Tabletter, hver inneholdende 10 mg 2-(2,6-dioksoetylpiper-idin-3-yl)-4-aminoftalimid, kan fremstilles på følgende måte:
Sammensetning ( for 1000 tabletter)
De faste ingrediensene blir først tvunget gjennom en sikt med 0,6 mm maskevidde. Deretter blir den aktive imidingrediensen, laktosen, talkumet, magnesiumstearatet og halvdelen av stivelsen straks blandet. Den andre halvdelen av stivelsen blir suspendert i 65 ml vann, og denne suspensjon blir tilsatt til en kokende oppløsning av polyetylenglykolen i 260 ml vann. Den resulterende pasta blir tilsatt til de pulverformede substansene, og det hele blir blandet og granulert, hvis nød-vendig med tilsetning av vann. Granulatet blir tørket over natten ved 35 °C, tvunget gjennom en sikt med 1,2 mm maskevidde og presset til tabletter med diameter på omtrent 10 mm, hvilke tabletter er konkave på begge sider og har en knekke-spalte på den øvre siden.

Gelatin-tørrfylte kapsler, hver inneholdende 100 mg av 1-okso-2-(2,6-diokso-3-metylpiperidin-3-yl)-4,5,6,7-tetra-fluorisoindolin kan fremstilles på følgende måte:
Sammensetning ( for 1000 kapsler)
Natriumlaurylsulfatet blir siktet inn i l-okso-2-(2,6-diokso-3-metylpiperidin-3-yl)-4,5,6,7-tetrafluorisoindolin gj ennom en sikt med 0,2 mm maskevidde, og de to komponentene blir straks blandet i 10 minutter. Den mikrokrystalline cellulosen blir deretter tilsatt gjennom en sikt med 0,9 mm maskevidde, og det hele blir igjen straks blandet intensivt i 10 minutter. Til slutt blir magnesiumstearatet tilsatt gjennom en sikt med 0,8 mm maskevidde, og etter blanding i ytterligere 3 minutter, blir blandingen oppdelt i n porsjoner på 140 mg hver til størrelse 0 (forlenget) gelatin-tørrfylte kapsler.

En 0,2 % injeksjons- eller infusjonsoppløsning kan fremstilles, for eksempel på følgende måte: l-okso-2-(2,6-diokso-3-metylpiperidin-3-yl)-4,5,6,7-tetra-fluorisoindolin blir oppløst i 1000 ml vann og filtrert gjennom et mikrofilter. Bufferoppløsningen blir tilsatt, og det hele blir fylt opp med vann til 2500 ml. For å fremstille doser ingsenhetsf ormer , blir porsjoner på 1,0 eller 2,5 ml hver ført inn i glassampuller (hver inneholdende henholdsvis 2,0 eller 5,0 mg imid).
