Claims (1)
- 【特許請求の範囲】 1. 複数のアッセイ成分を用いる、サンプル中の核酸分析物を検出するための 核酸ハイブリダイゼーションアッセイであって、該複数のアッセイ成分の各々は 、少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメントを含有し 、 少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメント中に、A- TおよびG-C塩基対が形成される条件下でアデノシン(A)、チミジン(T)、シチジン (C)、グアノシン(G)、またはウリジン(U)と効果的には塩基対を形成しない第1 のヌクレオチド単位を取り込ませる工程を包含する改良がされている、核酸ハイ ブリダイゼーションアッセイ。 2. 前記第1のヌクレオチド単位が、第2の相補的なヌクレオチド単位と塩基 対を形成し得る、請求項1に記載の方法。 3. 前記第1および第2のヌクレオチド単位が、以下からなる相補的塩基対の 群から交換可能に選択される請求項2に記載の方法: および ここでRは、ポリヌクレオチドに取り込まれたときに、該第1および第2のヌク レオチド単位に、相補的なヌクレオチド単位との塩基対を形成させ得るね格であ り、そしてR'は水素、メチル、a-またはb-プロピニル、臭素、フッ素、あるい はヨウ素である。 4. 複数のアッセイ成分を用いる、サンプル中の核酸分析物を検出するための 核酸ハイブリダイゼーションアッセイであって、該複数のアッセイ成分の各々は 少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメントを含有し、 アッセイストリンジェンシーが、Tm1ハイブリッド複合体を選択的に不安定化 するように制御され得る、Tm1ハイブリッド複合体およびTm2ハイブリッド複合体 を取り込ませる工程を包含する改良がされている、核酸ハイブリダイゼーション アッセイ。 5. 複数のアッセイ成分を用いる、サンプル中の核酸分析物を検出するための 液相サンドイッチハイブリダイゼーションアッセイであって、該複数のアッセイ 成分の各々は少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメン トを含有し、該アッセイは、(a)該分析物を固体支持体に直接的または間接的に 結合させる工程、(b)該分析物を標識する工程、および(c)分析物会合標識の存在 を検出する工程を包含し、 少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメント中に、A- TおよびG-C塩基対が形成される条件下でアデノシン(A)、チミジン(T)、シチジン (C)、グアノシン(G)、またはウリジン(U)と効果的には塩基対形成しない第1の ヌクレオチド単位を取り込ませる工程を包含する改良がされている、液相サンド イッチハイブリダイゼーションアッセイ。 6. 複数のアッセイ成分を用いる、サンプル中の核酸分析物を検出するための 液相サンドイッチハイブリダイゼーションアッセイであって、該複数のアッセイ 成分の各々は少なくとも1つのハイブリダイズするオリゴヌクレオチドセグメン トを含有し、該アッセイは、(a)該分析物を固体支持体に直接的または間接的に 結合させる工程、(b)該分析物を標識する工程、および(c)分析物に会合した標識 の存在を検出する工程を包含し、 アッセイストリンジェンシーが、Tm1ハイブリッド複合体を選択的に不安定化 するように制御され得る、Tm1ハイブリッド複合体およびTm2ハイブリッド複合体 の取り込みを包含する改良がされている、液相サンドイッチハイブリダイゼーシ ョンアッセイ。 7. 以下の構造式を有する化合物を合成する方法であって、 ここで、R1は、水素、ヒドロキシル、スルフヒドリル、ハロゲノ、アミノ、アル キル、アリル、および-OR2(ここでR2はアルキル、アリル、シリル、またはリン 酸塩である)からなる群から選択され: a) 以下の構造式 を有する化合物を、3'および5'ヒドロキシル基の両方を保護するのに適切 な試薬と反応させる工程; b) 工程(a)の産物を、O6-オキシ部分を求核置換を受けやすい官能基に変換す るのに適切な試薬と反応させ、それにより官能基化O6部分を産生する工程; c) 工程(b)の産物の2-アミノ基を酸化する工程; d) 工程(c)の産物を、該官能基化したO6部分を置換する求核試薬と反応させ る工程; e) 工程(d)の産物を、該保護された3'および5'ヒドロキシル基を脱保護する のに適切な試薬と反応させる工程 を包含する方法。 8. 以下を包含する2'-デオキシ-イソグアノシンを合成する方法: a) 2'-デオキシグアノシンをt-ブチルジメチルシリル(TBDMS)クロライドと反 応させることにより、2'-デオキシグアノシンを3',5'-O-(t-ブチルジメチルシリ ル)2-2'-デオキシグアノシンに変換する工程; b) 3',5'-O-TBDMS2-2'-デオキシグアノシンを4-トルエンスルホニルクロラ イドと反応させることにより、3',5'-O-TBDMS2-2'-デオキシグアノシンをO6-(4- トルエンスルホニル)-3',5'-O-TBDMS2-2'-デオキシグアノシンに変換する工程; c) O6-(4-トルエンスルホニル)-3',5'-O-TBDMS2-2'-デオキシグアノシンを フェノールで処理することにより、該O6-(4-トルエンスルホニル)基を置換して 、O6-(4-(メチルチオ)フェニル)-3',5'-O-TBDMS2-2'-デオキシグアノシンを得る 工程; d) 中性条件下でO6-(4-(メチルチオ)フェニル)-3',5'-O-TBDMS2-2'-デオキシ グアノシンを亜硝酸t-ブチルで処理することにより、該O6-(4-(メチルチオ)フェ ニル)-3',5'-O-TBDMS2-2'-デオキシグアノシンの2-アミノ基をオキシ官能基に酸 化して、O6-(4-(メチルチオ)フェニル)-3',5'-O-TBDMS2-2'-デオキシキサントシ ンを得る工程;および e) 高温で該O6-(4-(メチルチオ)フェニル)-3',5'-O-TBDMS2-2'-デオキシキサ ントシンのO2-(4-(メチルチオ)フェニル)基を水酸化アンモニウムで置換し、3', 5'-O-TBDMS2-2'-デオキシ-イソグアノシンを得る工程。 9. サンプル中の核酸分析物を検出するためのキットであって、少なくとも1 つのハイブリダイズするオリゴヌクレオチドプローブ、該分析物とハイブリッド 複合体を形成し得るプローブのセグメント、および該ハイブリッド複合体を検出 するための手段を含み、ここで該少なくとも1つのハイブリダイズするオリゴヌ クレオチドプローブは、A-TおよびG-C塩基対が形成される条件下で、アデノシン (A)、チミジン(T)、シチジン(C)、グアノシン(G)、またはウリジン(U)と効果的 には塩基対を形成しない第1のヌクレオチド単位を含有する、キット。 10.以下を含有する請求項9に記載のキット: (a) 捕獲プローブのセットであって、ここで該捕獲プローブは、A-TおよびG- C塩基対が形成される条件下で、A、T、C、G、またはUと効果的には塩基対を形成 しない第1のヌクレオチド単位を含有する、捕獲プローブのセット; (b) 第1および第2のハイブリダイズするオリゴヌクレオチドセグメントを 含有する捕獲エクステンダー分子のセットであって、ここで該第1のハイブリダ イズするオリゴヌクレオチドセグメントは該捕獲プローブとハイブリッド複合体 を形成し得、そして該第2のハイブリダイズするオリゴヌクレオチドセグメント は該核酸分析物の予め決定されたセグメントとハイブリッド複合体を形成し得る 、捕獲エクステンダー分子のセット; (c) 第3および第4のハイブリダイズするオリゴヌクレオチドセグメントを 含有する標識エクステンダー分子であって、ここで該第3のハイブリダイズする オリゴヌクレオチドセグメントは、該捕獲エクステンダー分子のセットが結合す るもの以外の核酸分析物のセグメントとハイブリッド複合体を形成し得る、捕獲 エクステンダー分子; (d) 第5および第6のハイブリダイズするオリゴヌクレオチドセグメントを 含有する随意のプレアンプリファイヤー分子であって、ここで該ハイブリダイズ するオリゴヌクレオチドセグメントは、A-TおよびG-C塩基対が形成される条件下 で、A、T、C、G、またはUと効果的には塩基対を形成しない第1のヌクレオチド 単位を含有し、そして該プレアンプリファイヤー分子は、該標識エクステンダー 分子および複数の増幅マルチマーとハイブリッド複合体を形成し得る、プレアン プリファイヤー分子; (e) 第7および第8のハイブリダイズするオリゴヌクレオチドセグメントを 含有する増幅マルチマーであって、ここで該ハイブリダイズするオリゴヌクレオ チドセグメントは、A-TおよびG-C塩基対が形成される条件下で、A、T、C、G、ま たはUと効果的には塩基対を形成しない第1のヌクレオチド単位を含有し、そし て該増幅マルチマーは、該標識エクステンダー分子またはプレアンプリファイヤ ー分子、および複数の同一のオリゴヌクレオチドサブユニットとハイブリッド複 合体を形成し得る、増幅マルチマー;および (f) 標識を含有する標識プローブであって、該同一のオリゴヌクレオチドサ ブユニットとハイブリッド複合体を形成するように設計され、そして直接的また は間接的に検出可能なシグナルを提供する、標識プローブ。 11. アプタマーとして有用なオリゴヌクレオチドであって、複数の相補的塩 基対を含有する分子内オリゴヌクレオチドハイブリッド複合体を含有し、該塩基 対の少なくとも1つが、A-TおよびG-C塩基対を通常形成する条件下で、アデノシ ン(A)、チミジン(T)、シチジン(C)、グアノシン(G)、またはウリジン(U)と効果 的には塩基対を形成しない相補的な非天然ヌクレオチド単位を含有し、ここで該 非天然ヌクレオチド単位は、該塩基対の特異性が該アプタマーの二次構造の維持 には必要とされないオリゴヌクレオチドセグメント内に含有される、オリゴヌク レオチド。 12.以下の工程を包含するアプタマーを調製するための方法: (a) 標的分子を提供する工程; (b) 該標的分子とオリゴヌクレオチドのランダマープールとを、該オリゴヌ クレオチドが該標的分子に結合するのに好都合な条件下で接触させる工程; (c) 該標的分子に結合し、そしてオリゴヌクレオチド-標的複合体を形成する 該オリゴヌクレオチドを、該標的分子に結合しない該オリゴヌクレオチドから分 離する工程: (d) 該オリゴヌクレオチド-標的複合体から該オリゴヌクレオチドを解離させ る工程; (e) ポリメラーゼ連鎖反応を用いて該オリゴヌクレオチドを増幅する工程; (f) 少なくとも1回(b)から(e)の工程を繰り返し、最終的なアプタマー構築 物を形成する工程;および (g) 該最終アプタマー構築物中の1つ以上のヌクレオチド単位を、A-Tおよび G-C塩基対を通常形成する条件下で、アデノシン(A)、チミジン(T)、シチジン(C) 、グアノシン(G)、またはウリジン(U)と効果的には塩基対を形成しない非天然ヌ クレオチド単位で置換する工程。 13. 第1および第2のハイブリダイズするセグメントを含有するアンチセン ス分子であって、ここで該第1のハイブリダイズするセグメントは標的オリゴヌ クレオチドとハイブリッド複合体を形成し得、そして該第2のセグメントは、A- TおよびG-C塩基対を形成する条件下で、アデノシン(A)、チミジン(T)、シチジン (C)、グアノシン(G)、またはウリジン(U)と効果的には塩基対を形成しない少な くとも1つのヌクレオチド単位を含有し、そして第2のアンチセンス分子とハイ ブリッド複合体を形成し得る、アンチセンス分子。
