Description
の化合物の調製方法であって、
式(II):
の化合物をビルスマイヤー試薬と反応させることを含む、
式(I)の化合物の調製方法に関する。
本明細書で用いられるとき、「ビルスマイヤー試薬」なる用語は、式(III)
の化合物を意味する。
のホルムアミドと活性化剤との反応によって好適に行われる。
好ましくは、化合物(II)とビルスマイヤー試薬との反応は、好適な溶媒中で起こる。別法では、反応は、溶媒の非存在下で行われ得る。
本発明の一実施態様において、反応シーケンスにはハロゲン交換ステップが含まれる。
本発明の好適な実施態様では、R1は水素である。
a1)式(XII)(1,1−ジフルオロアセトン):
a2)式(XIII)の化合物をジメチルホルムアミド及び活性化剤と反応させて式(IX)(3−ジフルオロメチル−1−メチル−1H−ピラゾール−4−カルバルデヒド):
a3)塩基の存在下で酸化剤を用いて、式(IX)の化合物を、式(XI)の化合物に酸化すること、
を含む、式(XI)の化合物の生成方法に関する。
上述の方法ステップa1)は好ましくは0℃〜50℃、さらに好ましくは10℃〜25℃の温度範囲にて行われる。
好適な活性化剤は、例えば、オキシ塩化リン、ホスゲン、または塩化チオニルであり、好ましくはオキシ塩化リンである。
方法ステップa3)用の好適な酸化剤は、例えば、過酸化水素である。酸化剤の好適な量は、例えば、少なくとも1当量であり、好ましくは10〜20当量である。
a1)不活性溶媒の存在下にて、式(VII)の化合物をメチルヒドラジンと反応させて式(II)の化合物を生成すること;並びに
a2)オキシ塩化リン及びジメチルホルムアミドを用いて、その化合物を式(I)の化合物に変化させること、
を含む、式(I)の化合物の生成方法である。
前記態様において、ステップa1)及びa2)は、先に述べたように行われる。

Claims (13)
- Hal及びHal’がFである、請求項1に記載の方法。
- Hal及びHal’がClである、請求項1に記載の方法。
- 酸化剤が過酸化水素である、請求項4に記載の方法。
- 該酸化が、塩基の存在下で行われる、請求項4または5に記載の方法。
- Hal及びHal’を、ClからFへ変換するハロゲン交換のステップを含む、請求項3または4に記載の方法。
- 該ビルスマイヤー試薬が、POCl3及びジメチルホルムアミドからその場で生成される、請求項1〜7のいずれか一項に記載の方法。
