Abstract
Description
本出願は、35 U.S.C. § 119(e)の下で、2013年7月25日に出願された米国仮出願第61/858,558号、名称「SPHERICAL NUCLEIC ACID-BASED CONSTRUCTS AS IMMUNOSTIMULATORY AGENTS FOR PROPHYLACTIC AND THERAPEUTIC USE(予防的および治療的使用のための免疫刺激剤としての球状の核酸ベース構築物)」の優先権を主張し、この仮出願はその全体が参照により本明細書に組み込まれる。
本発明は、TLRのアゴニストなどの核酸相互作用複合体のアゴニストのナノスケール構築物、ならびにそれらの方法および組成物に関する。
免疫系は、病原体および老化または悪性の宿主細胞を含む、異物、有害物および不要物質を取り除くための、高度に進化した極めて精密な内因性のメカニズムである。治療的または予防的目的のための免疫系の調節は、特定の免疫細胞の活性を調節する化合物の導入により可能であることが知られている。主な例はワクチンであり、これは病原体およびがん細胞に対する防御を誘導する能力を示す。最初の近代的なワクチン製剤は、生きている/弱毒化または不活化病原体を含有したが、これらは多くの場合、毒性が強すぎるとみなされるか、または防御免疫を提供しなかった。精製したタンパク質誘導体および他の抗原性サブユニットワクチン戦略が追求されたが、これらは一般に、軽度に予防的または非効率的な応答をもたらす。現在有効な免疫はほとんどの場合において、免疫刺激性化合物の使用が必要であることが知られていることが理解されており、該免疫刺激性化合物は特に、細胞媒介性免疫および免疫学的記憶を含む、より堅牢で特異的で長く続く応答を誘導するために必要な信号を提供する。これらの応答の性質は、導入される免疫刺激性化合物(複数可)の種類によって調節することができる。実際、適切な抗原刺激の存在下で一緒に投与される免疫刺激性化合物を使用して、感染症およびがんを含む様々な病気を処置または予防する潜在能力により、多様な免疫応答の誘発が可能であると考えられている。これらはまた、子供や老人などの免疫不全の集団のワクチン接種にも、使用できる可能性がある1。
他の側面において、本発明は、核酸相互作用複合体のアゴニストのコロナ、および該コロナに組み込まれた抗原を含む、ナノスケール構築物である。いくつかの態様において、抗原の表面密度は、少なくとも0.3pmol/cm2である。他の態様において、抗原は、少なくとも2つの異なる種類の抗原を含む。
さらに他の側面において、本発明は、核酸相互作用複合体の少なくとも2種のアゴニストが組み込まれたコロナを含み、ここで前記アゴニストが、TLR3、7/8、および/または9アゴニストからなる群から選択される、ナノスケール構築物である。
いくつかの態様において、核酸相互作用複合体のアゴニストは、スペーサーを含む。
ナノスケール構築物は、いくつかの態様において、任意に金属である、コロナの中心におけるナノ粒子コアを含む。金属コアは、金、銀、白金、アルミニウム、パラジウム、銅、コバルト、インジウム、ニッケル、およびそれらの混合物からなる群から選択することができる。いくつかの態様において、ナノ粒子コアは、金を含む。他の態様において、ナノスケール構築物は分解性である。
本発明の態様において、コロナは球状コロナである。
本明細書に記載のナノスケール構築物および担体から構成されるワクチンが、本発明の他の側面により提供される。
本発明のナノスケール構築物を細胞に送達することによる、治療剤を細胞に送達する方法が、他の側面において提供される。
標的分子の発現を調節するための方法が、本発明の他の側面において提供される。方法は、本発明のナノスケール構築物を細胞に送達することを含む。いくつかの態様において、標的分子は、TLR3、7、8、および9からなる群から選択されるTLRである。
他の側面によれば、本発明は、対象を処置する方法であって、該対象に対して、本明細書に記載のナノスケール構築物を、免疫応答を刺激する有効量で投与することを含む、前記方法である。いくつかの態様において、対象は、感染症、がん、自己免疫疾患、アレルギー、または喘息などのアレルギー性疾患を有する。

本明細書中で使用される核酸相互作用複合体は、核酸分子と相互作用し、その相互作用に応答して免疫応答を生成するように刺激される、分子または分子複合体を指す。分子または分子複合体は、例えば、受容体であってもよい。いくつかの態様において、核酸相互作用複合体は、パターン認識受容体(PRR)複合体である。PRRは、先天性免疫系の細胞によって発現されて病原体関連分子パターン(PAMP)を識別するタンパク質からなる、免疫系の原始的な部分であり、これは、微生物病原体または細胞ストレス、並びに、細胞損傷の間に放出される細胞成分に関連する、ダメージ関連分子パターン(DAMP)に関連付けられている。PRRとしては、限定はされないが、膜結合PRR、例えば受容体キナーゼ、トール様受容体(TLR)、およびC型レクチン受容体(CLR)(マンノース受容体およびアシアロ糖タンパク質受容体)など;細胞質PRR、例えばRIG−I様受容体(RLR)、RNAヘリカーゼ、植物PRR、およびNonRDキナーゼ;および分泌PRRを含む。

TLR7媒介性免疫応答は、TLR7シグナル伝達に関連する応答である。TLR7媒介性免疫応答は、一般に、IFN−αおよび、IP−10およびI−TACなどのIFN誘導性サイトカインの誘導によって特徴付けられる。TLR7媒介性免疫応答において誘導されるサイトカインIL−1α/β、IL−6、IL−8、MIP−1α/βおよびMIP−3α/βのレベルは、TLR8媒介性免疫応答において誘導されるものよりも低い。
TLR8媒介性免疫応答は、TLR8シグナル伝達に関連する応答である。この応答はさらに、例えば、IFN−γ、IL−12p40/70、TNF−α、IL−1α/β、IL−6、IL−8、MIP−1α/βおよびMIP−3α/βなどの炎症誘発性サイトカインの誘導により、さらに特徴付けられる。
本明細書で使用される場合、「TLR7/8アゴニスト」は全体として、TLR7および/またはTLR8シグナル伝達を増加させることができる、任意の核酸を意味する(すなわち、TLR7および/またはTLR8のアゴニスト)。いくつかのTLR7/8リガンドは、TLR7シグナル伝達のみを誘導し(例えば、TLR7特異的アゴニスト)、いくつかはTLR8シグナル伝達のみを誘導し(例えば、TLR8特異的アゴニスト)、他は、TLR7およびTLR8シグナル伝達の両方を誘導する。
TLR7またはTLR8シグナル伝達のレベルは、シグナル伝達の既存のレベルに対して強化することができるか、またはシグナル伝達のバックグラウンドレベルを超えて誘導することができる。TLR7リガンドとしては、限定されないが、C8置換グアノシンなどのグアノシン類似体、本質的にGおよびUからなるリボヌクレオシドの混合物、グアノシンリボヌクレオチドおよびRNAまたはRNA様分子(PCT/US03/10406)、およびアデノシンベースの化合物(例えば6−アミノ−9−ベンジル−2−(3−ヒドロキシ−プロポキシ)−9H−プリン−8−オール、およびSumitomoにより作製された類似化合物(例えば、CL-029))が挙げられる。

本明細書で使用する用語「TLR9アゴニスト」は、TLR9シグナル伝達を増加させることができる任意の薬剤を意味する(すなわち、TLR9のアゴニスト)。TLR9アゴニストには、具体的には、限定することなく、免疫刺激性核酸、および特にCpG免疫刺激性核酸が含まれる。
いくつかの態様において、核酸相互作用複合体のアゴニストは、免疫刺激性オリゴヌクレオチドである。本明細書で使用する「免疫刺激性オリゴヌクレオチド」とは、免疫応答を誘導することができる免疫刺激性モチーフまたは骨格を含有する任意の核酸(DNAまたはRNA)である。免疫応答の誘導とは、免疫細胞の数または活性の任意の増加、またはサイトカインなどの免疫因子の発現または絶対レベルの増加を意味する。免疫細胞としては、限定はされないが、NK細胞、CD4+Tリンパ球、CD8+Tリンパ球、B細胞、樹状細胞、マクロファージおよび他の抗原提示細胞を含む。サイトカインとしては、限定はされないが、インターロイキン、TNF−α、IFN−α、βおよびγ、Fltリガンド、および共刺激分子を含む。免疫刺激性モチーフとしては、限定はされないが、CpGモチーフおよびTリッチモチーフを含む。
二本鎖ポリヌクレオチドは、その全長にわたって二本鎖であることができ、それがオーバーハングする一本鎖配列を有さず、したがって平滑末端であることを意味する。他の態様において、二本鎖ポリヌクレオチドの二本の鎖は、1または2以上の一本鎖オーバーハングを生成する、異なる長さを有することができる。本発明の二本鎖ポリヌクレオチドは、ミスマッチおよび/またはループまたは膨隆(bulge)を含むことができる。いくつかの態様において、オリゴヌクレオチドの長さの少なくとも約70%、80%、90%、95%、96%、97%、98%、または99%が、二本鎖である。いくつかの態様において、本発明の二本鎖ポリヌクレオチドは、少なくとも、または1、2、3、4、5、6、7、8、9、10、11、12、13、14、もしくは15個までの、ミスマッチを含有する。
糖部分の修飾は、2’−O−メチルヌクレオチドを含むことができ、これは「メチル化」と呼ばれる。いくつかの例において、本発明に関連するポリヌクレオチドは、修飾または非修飾の糖部分のみを含んでもよく、一方他の例において、ポリヌクレオチドは、修飾されたいくつかの糖部分および修飾されていないいくつかの糖部分を含む。
いくつかの側面において、2’−O−メチル修飾は、二本鎖核酸に対するインターフェロン応答などの、望ましくない細胞ストレス応答を減少させるために有益であり得る。修飾された糖としては、D−リボース、2’−O−アルキル(2’−O−メチルおよび2’−O−エチルを含む)、すなわち、2’−アルコキシ、2’−アミノ、2’−S−アルキル、2’−ハロ(2’−フルオロを含む)、2’−メトキシエトキシ、2’−アリルオキシ(−OCH2CH=CH2)、2’−プロパルギル、2’−プロピル、エチニル、エテニル、プロペニル、およびシアノ基等が挙げられる。糖部分はまた、ヘキソースであることができる。
用語「疎水性修飾」とは、塩基の修飾であって、全体の疎水性が増加し、塩基がそれでも通常のワトソン−クリック相互作用に近いものを形成することができる、前記修飾を意味する。塩基修飾の非限定的な例としては、5位のウリジンおよびシチジンの修飾を含み、例えばフェニル、4−ピリジル、2−ピリジル、インドリル、およびイソブチル、フェニル(C6H5OH);トリプトファニル(C8H6N)CH2CH(NH2)CO)、イソブチル、ブチル、アミノベンジル;フェニル;ナフチル、などである。

いくつかの態様において、ポリヌクレオチドはDNAとRNAの両方を含むことができる。
CpG配列は、ヒトDNAにおいては比較的稀であるが、細菌などの感染性生物のDNAでは通常に見出される。ヒト免疫系は明らかに、CpG配列を感染の早期警告サインとして認識し、他の免疫刺激剤で頻繁に見られる副作用を引き起こすことなく、侵入する病原体に対する即時かつ強力な免疫応答を開始するように、進化してきた。したがって、CpG含有核酸は、この先天性免疫防御機構に依存して、免疫療法のためのユニークで天然の経路を利用することができる。免疫調節に対するCpG核酸の効果は、以下の文献に広範囲に記載されている:米国特許第6,194,388号、および公開された特許出願、例えばPCT US95/01570、PCT/US97/19791、PCT/US98/03678;PCT/US98/10408;PCT/US98/04703;PCT/US99/07335;およびPCT/US99/09863。
ナノスケール構築物の免疫刺激性オリゴヌクレオチドは、好ましくは長さが6から100塩基の範囲である。しかしながら、6ヌクレオチドより大きい任意のサイズ(複数kb長であってもよい)の核酸は、充分な免疫刺激モチーフが存在する場合、本発明による免疫応答を誘導することができる。好ましくは、免疫刺激性核酸は、ヌクレオチドのサイズが8〜100の範囲、いくつかの態様においては8〜50または8〜30である。
いくつかの態様において、免疫刺激性オリゴヌクレオチドは、ホスホロチオエート(PS)骨格などの修飾された骨格を有する。他の態様において、免疫刺激性オリゴヌクレオチドは、ホスホジエステル(PO)骨格を有する。さらに他の態様において、免疫刺激性オリゴヌクレオチドは、混合されたPOおよびPS骨格を有する。
核酸相互作用複合体および抗原のアゴニストを含む、本発明に関連するモダリティは、当技術分野で知られている任意の手段によってナノ粒子コアに付着することができる。オリゴヌクレオチドをナノ粒子に付着させるための方法は、米国特許出願公開第2010/0129808号に詳細に記載され、これは参照により組み込まれている。
ナノ粒子は、ポリヌクレオチドを付着するために官能化することができる。代替的にまたは付加的に、ポリヌクレオチドを官能化することができる。官能化のための1つのメカニズムはアルカンチオール法であり、これにより、オリゴヌクレオチドは、それらの3’または5’末端でアルカンチオールにより官能化され、その後、金ナノ粒子または他の金属、半導体、もしくは磁気材料を含むナノ粒子に付着される。かかる方法は、例えば、Whitesides, Proceedings of the Robert A. Welch Foundation 39th Conference On Chemical Research Nanophase Chemistry, Houston, Tex., pages 109-121 (1995)、およびMucic et al. Chem. Commun. 555-557 (1996)に記載されている。オリゴヌクレオチドはまた、例えば、米国特許第5,472,881号に記載され参照により組み込まれているように、ホスホロチオエート基などの他の官能基を用いて、または、Burwell, Chemical Technology, 4, 370-377 (1974)およびMatteucci and Caruthers, J. Am. Chem. Soc., 103, 3185-3191 (1981)に記載され参照により組み込まれているように、置換アルキルシロキサンを用いて、ナノ粒子に付着することができる。いくつかの例において、ポリヌクレオチドは、ポリヌクレオチドを5’または3’チオヌクレオシドで終端させることにより、ナノ粒子に付着することができる。他の例において、熟成プロセスを使用して、ポリヌクレオチドをナノ粒子に付着することができ、これは以下に記載されており、参照により組み込まれる:米国特許第6,361,944号、第6,506, 569号、第6,767,702号および第6,750,016号、およびPCT公開第WO 1998/004740号、第WO 2001/000876号、第WO 2001/051665号および第WO 2001/073123号。
ナノスケール構築物は、複数の化学物質を用いて設計することができる。例えば、DTPA(ジチオールホスホロアミダイト)結合を使用することができる。DTPAは、チオールによるフレアの細胞内放出に抵抗し、信号対雑音比を高めるのに役立ち得る。
本明細書に記載の方法によって生産されたコンジュゲートは、他の方法によって生産されたものよりもかなり安定である。この増大した安定性は、ナノ粒子コアの表面上の、またはコロナの表面を形成する、オリゴヌクレオチドの高密度化に起因する。界面活性剤の存在下で、例えば約0.01%ドデシル硫酸ナトリウム(SDS)、Tween、またはポリエチレングリコール(PEG)の存在下で、塩の添加を行うことにより、塩熟成処理は約1時間で行うことができる。
本発明の側面は、ナノスケール構築物の、治療および/または診断的使用のための対象への送達に関する。粒子は、in vivoでの投与のために、それのみで投与してもよく、または任意の適切な医薬担体中、例えば生理食塩水などの液体、もしくは粉末中で投与してもよい。それらはまた、より大きな担体粒子と共に、または投与デバイス内で送達することもできる。粒子を処方してもよい。本発明の製剤は、薬学的に許容し得る溶液中で投与することができ、該溶液は、薬学的に許容し得る濃度の、塩、緩衝剤、保存剤、適合性担体、アジュバント、および任意に他の治療成分を日常的に含有することができる。いくつかの態様において、本発明に関連するナノスケール構築物は、ローションなどの物質(例えば、アクアホル(aquaphor))と混合し、対象の皮膚に投与されて、これにより、ナノスケール構築物が対象の皮膚を介して送達される。当該技術分野で知られているナノ粒子送達の任意の方法は、本発明の側面と適合性であり得ることが理解されるべきである。
したがって、一側面において本発明には、核酸相互作用複合体のアゴニストが、免疫刺激効果を媒介するのに非常に有効であるという発見が関与する。核酸相互作用複合体のこれらのアゴニストは、治療的および予防的に免疫系を刺激して、がん、感染症、アレルギー、喘息、自己免疫疾患、および他の障害を処置し、がん化学療法後の日和見感染から保護するために有用である。例えば、TLRアゴニスト刺激から生じる、強力だがバランスのとれた細胞性および体液性免疫応答は、侵入する病原体およびがん細胞に対する身体自身の自然防御システムを反映する。
アレルギーを有する対象とは、アレルゲンに応答してアレルギー反応を有するか、これを発症するリスクのある対象である。アレルギーは、物質(アレルゲン)に対する後天性の過敏性を指す。アレルギー状態としては、限定はされないが、湿疹、アレルギー性鼻炎または鼻感冒、枯草熱、結膜炎、気管支喘息、蕁麻疹(じんましん)、および食物アレルギー、ならびに他のアトピー状態が挙げられる。
対象は、ヒトまたは脊椎動物を意味し、限定はされないが、イヌ、ネコ、ウマ、ウシ、ブタ、ヒツジ、ヤギ、シチメンチョウ、ニワトリ、霊長類、例えばサル、および魚(水産養殖種)、例えばサケを含む。したがって本発明はまた、非ヒト対象におけるがんおよび腫瘍、感染症、およびアレルギー/喘息を処置するために使用することができる。
本明細書で使用される抗原は、免疫応答を誘発することができる分子である。抗原としては、限定はされないが、細胞、細胞抽出物、タンパク質、ポリペプチド、ペプチド、多糖類、多糖複合体、多糖類および他の分子のペプチドおよび非ペプチド模倣物、小分子、脂質、糖脂質、炭水化物、ウイルスおよびウイルス抽出物ならびに、寄生虫およびアレルゲンなどの多細胞生物が挙げられる。抗原という用語は、広義に、宿主免疫系によって外来性であるとして認識される任意の種類の分子を含む。抗原としては、限定はされないが、がん抗原、微生物抗原、およびアレルゲンを含む。
真菌の例としては、Cryptococcus neoformans、Histoplasma capsulatum、Coccidioides immitis、Blastomyces dermatitidis、Chlamydia trachomatis、Candida albicansを含む。
他の医学的に関連する微生物は文献に広く記載されており、例えば、C.G.A Thomas, Medical Microbiology, Bailliere Tindall, Great Britain 1983を参照されたい;この内容全体は、参照により本明細書に組み込まれる。
抗ウイルス剤は、ウイルスによる細胞の感染または細胞内でのウイルスの複製を防止する化合物である。抗ウイルス薬の種類は抗細菌薬よりも大幅に少なく、その理由は、ウイルス複製のプロセスが宿主細胞内のDNA複製に非常に密接に関連しており、非特異的な抗ウイルス剤は多くの場合、宿主に対して毒性であるからである。抗ウイルス剤によって遮断または阻害することができる、ウイルス感染のプロセス内のいくつかの段階がある。これらの段階は、ウイルスの宿主細胞への付着(免疫グロブリンまたは結合ペプチド)、ウイルスの脱殻(例えば、アマンタジン)、ウイルスmRNAの合成または翻訳(例えばインターフェロン)、ウイルスRNAまたはDNAの複製(例えば、ヌクレオチド類似体)、新しいウイルスタンパク質の成熟(例えば、プロテアーゼ阻害剤)、およびウイルスの出芽および放出を含む。
本明細書で使用する「自己抗原」は、正常な宿主組織の抗原を指す。正常な宿主組織は、がん細胞を含まない。したがって、自己抗原に対して開始された免疫応答は、自己免疫疾患の文脈において望ましくない免疫応答であり、正常組織の破壊および損傷に寄与し、一方がん抗原に対して開始される免疫応答は、望ましい免疫応答であり、腫瘍またはがんの破壊に寄与する。したがって、自己免疫疾患の処置を目的とした本発明のいくつかの側面において、CpG免疫刺激性核酸を自己抗原、特に、自己免疫疾患の標的である自己抗原と共に投与することは推奨されない。
本発明のキットは、いくつかの場合において、本発明の組成物に関連して提供される任意の形態の指示書であって、当業者が、指示書が本発明の組成物と関連することを認識するような様式の、前記指示書を含むことができる。例えば、指示書は、キットに関連する組成物および/または他の組成物の、使用、修飾、混合、希釈、保存、投与、組立、格納、包装、および/または調製のための指示を含むことができる。いくつかのケースにおいて、指示書は、例えば、試料に対する等の特定の使用のための、組成物の使用についての指示書を含むことができる。指示書は、当業者によってかかる指示書を含む適切な媒体として認識できる任意の形態で提供されてよく、例えば、記載されたまたは公開された、口頭、可聴(例えば、電話)、デジタル、光学的、視覚的(例えば、ビデオテープ、DVDなど)、または電子通信(インターネットまたはウェブベースの通信を含む)であって、任意の様式で提供されるものである。
本発明は、以下の例によりさらに説明されるが、さらに限定するものと解釈されるべきでは決してない。本出願を通して引用されている全ての参考文献の内容全体(参考文献、発行済み特許、公開特許出願、および同時係属中の特許出願を含む)は、参照により本明細書に明示的に組み込まれる。
例1:
材料および方法
免疫刺激性SNAの合成
免疫刺激性SNA(isSNA)の合成を、以下の本質的な変更を加え、他の場所で説明したようにして7−12実施する。簡潔に述べると、20mLの13nmの金コロイドを、10%Tween 20およびスルフヒドリル修飾核酸(TLR3、7/8、9)のTLRアゴニスト配列と、5μMなどの適切な濃度で混合し、一晩反応させる。抗原の添加は、他の場所で説明したようにして実施することができる10。SNAの精製は、繰り返しの超遠心分離を75,000xgで30min行って実施することができる。
RAW 264.7細胞株はATCCから入手した。RAW-BlueマクロファージはInVivoGenから入手した。Ramos-BlueおよびTHP1-XBlue細胞は、InVivoGenから入手した。これらはすべて、販売業者の推奨に従って培養した。
結果と考察
本発明のナノスケール構築物は、溶液中の核酸相互作用複合体の未処方アゴニスト(CpGオリゴヌクレオチド)と比べて、マクロファージにおいて効力を大幅に増強することが見出された(図2)。RAW-Blueマクロファージは、ウェル当たり65kの細胞でプレーティングし、一晩接着させた。実験の日に、細胞をAST−008−psまたはCpG 1826−psでオリゴの指示された濃度で、30min(上パネル)、4h(中央パネル)、または一晩(下パネル)処置した。30minおよび4hの時点については、全上清を吸引し、細胞を洗浄し、試験薬剤なしの完全増殖培地を投与した。一晩の時点で、細胞の活性化状態を、QuantiBlueアッセイキットを用いて決定した。結果は、特に短い時点において、AST−008−psが、CpG 1826−psの17800nMよりも有意に低い、188nMのEC50を実証していることを示す(9〜1000倍の減少、平均から1標準偏差)。4hの時点で、AST−008−psは、CpG 1826−psよりも低いEC50(32nM対57nm)およびより強い活性化状態を実証する。この違いは、一晩のインキュベーションの後に小さくなり、この時点でEC50は互いに統計的に差がなかった。これは、標的細胞と薬剤の滞留時間が制限された条件下、特に30分以下で、AST−008−ps製剤が、より迅速かつ強い免疫活性化をもたらすことを示唆する。
なお、本発明のナノスケール構築物は、いくつかの異なるCpGオリゴ配列に対して、複数倍の効力の増加があったことが判明した(図6)。Ramos-Blue細胞を播種し、製造業者の推奨プロトコルに従って、示された化合物および対照を使用して活性化した。オリゴ1826(上パネル)および1668(下パネル)を試験した。特に、SNA化合物は、対照と比較して化学的性質とは無関係に、遊離オリゴよりも有意に低いEC50値を示す。これは、これらの配列に対して、オリゴのSNA製剤が数倍強力であることを示唆している。
本発明のナノスケール構築物は、CpGオリゴよりも迅速かつ持続的な活性を有することが観察された(図8)。細胞を記載のようにプレーティングし、活性化をQuantiBlueを用いて測定した。結果は、6nMのオリゴにおいて、PS SNAが、遊離の1668 PSオリゴよりも著しい活性化を実証することを示す。
ホスホロチオエート修飾の、配列依存的な様式でアゴニスト活性を調節する能力を試験した(図9)。Raw Blue細胞をプレーティングし、示したアゴニストで処置した。オリゴ1826(上パネル)および1668(下パネル)を試験した。その結果、内部ホスホロチオエート修飾(C*G)および2つの5’ホスホロチオエート結合(5’PS2)が、配列依存的に見える様式で、免疫刺激性SNASの活性に効果を及ぼすことを示す。
CpG PO/POのナノスケール構築物の活性化の時間経過を試験した(図11)。試験された構築物は、インキュベーションの>4hまで活性化されない。Raw Blue細胞をプレーティングし、示したアゴニストで処置した。結果は、POおよびPO SNAが、インキュベーションが4時間を超えるまで、接着したRaw Blue細胞を強力に活性化しないことを示す。
5’CholのCpG POナノスケール構築物は、低いnM範囲内で活性化を示し、一方5’C18は、活性を抑止した(図12)。5’コレステロール修飾(5’Chol)は、特に低濃度で、オリゴ用量依存的にアゴニストの効力を増大することができる。C18分子による5’末端の修飾(5’C18)は、全ての活性を排除するように見える。
マクロファージによる、低レベルのIFN−γ分泌が実証された(図14)。Raw Blueマクロファージを、ウェル当たり65kの細胞でプレーティングし、一晩接着させた。実験の日に、示された化合物を、細胞で一晩インキュベートした。サイトカイン分泌の程度(処置後、24時間−左パネル、または48時間−右パネル)を、上清を回収し、示されたサイトカインの濃度をELISAで測定することにより、決定した。結果は、IFN−γが、これらの化合物で刺激されたRaw Blueマクロファージによって、感知できる程度に産生されないことを示す。
免疫治療用SNA(すなわち、AST−008)は、新規で汎用的な技術プラットフォームを提供する。これらによる多価の免疫調節剤の送達は応答を最適化し、リンパ腫モデルにおける大幅な腫瘍の減少が観察されている。さらにこれらは、遊離オリゴヌクレオチドまたはミョウバンよりも高い、in vivoでの強力かつバランスのとれたT細胞応答を誘発する。SNAは、単一のナノ粒子上で治療ワクチン抗原およびアジュバントと共存でき、遊離の免疫刺激性CpGオリゴデオキシヌクレオチドよりも、増強された活性および速い動態を有することができる。さらに、SNAは、複数の免疫刺激性受容体(例えば、TLR3、4、7/8、9)を同時に標的とする可能性を有する。それらは、例えば、がん免疫療法およびワクチンにおいて使用可能である(予防的または治療的)。
免疫治療用SNA(AST−008)の模式図を図15に示す。SNAは、単一のナノ粒子上に治療ワクチン抗原およびアジュバントと共存でき、複数の免疫刺激性受容体(例えば、TLR3、4、7/8、9)を同時に標的とし得る。
AST−008は、in vitroで、対応するCpGオリゴデオキシヌクレオチド(オリゴ)よりも高い炎症誘発応答を誘導する。この知見を実証するアッセイを行った。データは、図17Aおよび17Bに一組のグラフとして示されている。図17Aは、CTLオリゴ、CTL SNA、CpG 1826およびAST−008によって誘導されるTNF、IL−12、およびIL−6の発現レベルを示す。図17Bは、示された薬剤に起因するNF−KBの活性化を示す。
AST−008はまた、単回皮下用量の投与後に流入領域リンパ節を標的とする。AST−008は、金コアの光散乱を増強するために銀染色され、その後エオシンで対比染色された。4×の明視野倍率を使用した。データを図18に示す。
当業者は、日常的な実験のみを用いて、本明細書に記載の本発明の特定の態様に対する多くの均等物を認識し、または確認することができるであろう。かかる均等物は、以下の特許請求の範囲に含まれることが意図される。

- 核酸相互作用複合体のアゴニストのコロナ、および該コロナに組み込まれた抗原を含み、ここで前記抗原の表面密度が少なくとも0.3pmol/cm2である、ナノスケール構築物。
- 抗原が、少なくとも2つの異なる種類の抗原を含む、請求項2に記載のナノスケール構築物。
- 核酸相互作用複合体の少なくとも2種のアゴニストが組み込まれたコロナを含み、ここで前記アゴニストが、TLR3、7/8、および/または9アゴニストからなる群から選択される、ナノスケール構築物。
