Abstract
Description
(抗HA効果による抗ウイルス性評価)
(実施例1〜27)
表1に示す市販のヨウ素化合物および一価の銅化合物の粉末を、それぞれMEM(Minimum Essential Medium Eagle、MPバイオメディカル社製)100μlに懸濁液濃度5、0.5質量%となるように調整し、抗ウイルス性を評価した。なお、本明細書において、懸濁液濃度とは、懸濁液を構成するヨウ化物や一価の銅化合物と溶媒等の全成分の質量を100%とし、その中の特定成分(例えばヨウ化物や一価の銅化合物)の質量%を意味する。
(評価方法)
実施例1から27について、赤血球凝集(HA)の力価(HA価)を定法により、目視にて完全凝集を判定した。対象ウイルスとして、MDCK細胞を用いて培養したインフルエンザウイルス(influenza A/北九州/159/93(H3N2))を用いた。
(インフルエンザウイルスおよびネコカリシウイルスに対する不活化効果による抗ウイルス性評価)
ウイルスは、前述したように、脂質を含むエンベローブと呼ばれている膜で包まれているウイルスと、エンベロープを持たないウイルスに分類できる。そこで、エンペロープの有無による不活化効果を評価した。エンペロープを持つウイルスとしてインフルエンザ(influenza A/北九州/159/93(H3N2))を、エンペロープを持たないウイルスとしてノロウイルスの代替ウイルスとして一般的に用いられるネコカリシウイルス(F9株)を使用した。
(実施例28〜31)
市販のヨウ化銅(I)粉末(和光純薬工業株式会社製 和光一級)をMEM希釈液100μlに懸濁液濃度5、1、0.2、0.1質量%になるように懸濁し、それぞれの懸濁液を実施例28、29、30、31とし、ネコカリシウイルスとインフルエンザウイルスに対する抗ウイルス性を評価した。
市販の塩化銅(I)粉末(和光純薬工業株式会社製 試薬特級)をMEM希釈液100μlに懸濁液濃度2、1、0.5、0.25質量%になるように懸濁し、それぞれの懸濁液を実施例32、33、34、35とし、ネコカリシウイルスとインフルエンザウイルスに対する抗ウイルス性を評価した。
実施例28〜35についての抗ウイルス性の評価は、ウイルスを高精度で検出可能なプラーク法での感染価測定で行った。具体的には、各試験サンプル100μl及びブランクとしてのMEM希釈液とに、それぞれウイルス液を100μlづつ加え、インキュベータにて25℃、200rpmにて振蘯して反応させた。所定時間振蘯後、ウイルスと各サンプル中の化合物との反応を停止させるために20mg/mlのブイヨン蛋白を1800μl加えた。さらに、各反応サンプルが10-2〜10-5になるまでMEM希釈液にて希釈を行い(10段階希釈)、ネコカリシウイルス反応サンプルはコンフルエントCrFK細胞に、インフルエンザウイルス反応サンプルはMDCK細胞に100μl、反応後のサンプル液を接種した。90分間のウイルス吸着後、0.7%寒天培地を重層し、ネコカリシウイルスは48時間、インフルエンザウイルスは64時間、34℃、5%CO2インキュベータにて培養後、ホルマリン固定、メチレンブルー染色を行い形成されたプラーク数をカウントして、ウイルスの感染価(PFU/0.1ml,Log10);(PFU:plaque-forming units)を算出することで、抗ウイルス性を評価し、表3及び表4に結果を示した。
Claims (6)
- 少なくとも1種の一価の銅化合物の粒子を有効成分として含むことを特徴とする抗ウイルス剤。
- 前記一価の銅化合物が、塩化物、酢酸物、硫化物、ヨウ化物、臭化物、過酸化物、酸化物、またはチオシアン化物であることを特徴とする請求項1に記載の抗ウイルス剤。
- 前記一価の銅化合物がCuCl、CuOOCCH3、CuBr、CuI、CuSCN、Cu2SおよびCu2Oからなる群から少なくとも1つ選択されることを特徴とする請求項2に記載の抗ウイルス剤。
- 請求項1から3のいずれか1つに記載の抗ウイルス剤が含有されている、または外面に固定されていることを特徴とする繊維構造体。
- 請求項1から3のいずれか1つに記載の抗ウイルス剤が含有されている、または外面に固定されていることを特徴とする成型体。
- 請求項1から3のいずれか1つに記載の抗ウイルス剤が含有されている、または外面に固定されていることを特徴とするフィルムまたはシート。
