Claims (1)
- 【特許請求の範囲】 1.ハムスターEF−1α転写調節DNAを含む、精製及び単離されたDNA 。 2.前記調節DNAが配列番号:1に示される核酸配列を含む請求の範囲第1 項記載のDNA。 3.前記調節DNAが、当該調節DNAに作動可能に連結された遺伝子の転写 を促進することができる配列番号:1に示される核酸配列の一部分を含む請求の 範囲第1項記載のDNA。 4.前記調節DNAが、配列番号:1に示されるヌクレオチド中、約2114 位のヌクレオチドから約3656位のヌクレオチドまでを含む請求の範囲第3項 記載のDNA。 5.請求の範囲第1乃至4項のいずれかに記載のDNAを含む宿主細胞であっ て、該DNAがEF−1αをコードする配列に作動可能に連結されていない宿主 細胞。 6.請求の範囲第1乃至4項のいずれかに記載のDNAを含むベクターであっ て、該DNAがEF−1αをコードする配列に作動可能に連結されていないベク ター。 7.請求の範囲第6項記載のベクターで形質転換またはトランスフェクトされ た宿主細胞。 8.タンパク質をコードするDNA配列に作動可能に連結されたハムスターE F−1α調節DNAを含み、該DNA配列はハムスターEF−1α以外のタンパ ク質をコードしているキメラポリヌクレオチド。 9.前記調節DNAが、配列番号:1に示される核酸配列を含む請求の範囲第 8項記載のキメラポリヌクレオチド。 10.前記調節DNAが、配列番号:1に示される核酸配列の一部分を含み、該 一部分が当該調節DNAに作動可能に連結された遺伝子の転写を促進することが できる請求の範囲第8項記載のキメラポリヌクレオチド。 11.前記調節DNAが、配列番号:1に示されるヌクレオチド中、約2114 位のヌクレオチドから約3656位のヌクレオチドまでを含む請求の範囲第10 項記載のキメラポリヌクレオチド。 12.前記タンパク質をコードするDNA配列が、EF−1α以外のハムスター 細胞の内在性タンパク質をコードする請求の範囲第8項記載のキメラポリヌクレ オチド。 13.前記DNA配列が、ハムスター細胞に異種性のタンパク質をコードする請 求の範囲第8項記載のキメラポリヌクレオチド。 14.請求の範囲第8乃至13項のいずれかに記載のキメラポリヌクレオチドで 形質転換またはトランスフェクトされた宿主 細胞。 15.請求の範囲第8乃至13項のいずれかに記載のキメラポリヌクレオチドを 含む発現プラスミド。 16.配列番号:28に示される核酸配列をさらに含む請求の範囲第15項記載 の発現プラスミド。 17.配列番号:28に示される前記核酸配列が、キメラポリヌクレオチドに対 して3’側に位置する請求の範囲第16項記載の発現プラスミド。 18.請求の範囲第15項記載の発現プラスミドで形質転換またはトランスフェ クトされた宿主細胞。 19.請求の範囲第16または17項のいずれかに記載の発現プラスミドで形質 転換またはトランスフェクトされた宿主細胞。 20.プラスミドpDEF2。 21.プラスミドpDEF14。 22.プラスミドpDEF1./ICM3H.2。 23.プラスミドpNEF1/ICM3L.3。 24.プラスミドpDEF1/F2GH.1。 25.プラスミドpNEF1/F2GL.1。 26.プラスミドpDEF1/CTN.1。 27.プラスミドpDEF2/HPH.4。 28.プラスミドpDEF10/MDC.1。 29.請求の範囲第20乃至28項のいずれかに記載の発現プラスミドで形質転 換またはトランスフェクトされた宿主細胞。 30.宿主細胞において目的の遺伝子の転写を増大させるための方法であって、 目的の遺伝子に作動可能に連結された位置にて、該宿主細胞のゲノム配列内にハ ムスターEF−1α調節DNAを含むDNAを統合させる工程を含む方法。 31.統合されるべき前記DNAがさらに、標的配列を含む請求の範囲第30項 記載の方法。 32.前記宿主細胞がチャイニーズハムスター卵巣細胞である請求の範囲第30 項記載の方法。 33.前記目的の遺伝子が、チャイニーズハムスター卵巣細胞の内在性遺伝子で ある請求の範囲第32項記載の方法。 34.前記所望の遺伝子がチャイニーズハムスター卵巣細胞に異種性であり、チ ャイニーズハムスター卵巣細胞のゲノムDN Aに安定に統合される請求の範囲第30項記載の方法。
