本発明は、免疫障害及び炎症性障害、特にPD-1/PD-L1及び/又はPD-L2系により制御される免疫障害及び炎症性障害のための処置についての必要性に対処する。この必要性に対処するために、本発明は抗PD-1抗体を提供し、これはPD-1とPD-L1の間での相互作用を遮断しない。一態様において、本発明は抗体を提供し、それはPD-1とPD-L1の間での相互作用を増強する。本発明の一態様では、本発明の抗体は、PD-1シグナル伝達経路を活性化する。一態様では、本発明の抗体は抗PD-1アゴニスト抗体である。PD-1アゴニズムによって、PD-1が発現されているが、しかし、そのリガンドにより関与しないであろう、又は、場合により、関与しうるヒト疾患において自己反応性T細胞の増殖及びエフェクター機能を阻害することにより、免疫バランスが回復される。一態様では、本発明の抗体は、免疫障害及び炎症性障害ならびに移植片拒絶を処置する際に有用である。例えば、本発明の抗体は、PD-1とPD-L1の間の相互作用を調節することにより、特にPD-1経路を活性化することにより軽減することができる疾患又は障害の処置及び/又は予防のために有用である。一態様では、本発明の抗体は、全身性硬化症(SSc)、全身性エリテマトーデス、多発性筋炎、巨細胞性動脈炎、乾癬、乾癬性関節炎、強直性脊椎炎、又は炎症性腸疾患を処置及び/又は予防する際に有用である。

MHC型A、C、D、E、H、G系統のマウスを、組換え単量体ヒトPD-1又はヒトPD-1-ヒトFc-Hisタンパク質で免疫化した。この組換えタンパク質についての遺伝子記号はPDCD1であり、GeneIDは5133である。血清学を次に、結合のためにヒトPD-1抗原を発現するCHOヒトPD-1細胞を使用したフローサイトメトリーにより評価した。選択された血清学的に陽性のマウスに、B細胞単離前に最終追加免疫を与えた。全ての選択されたマウスが、血清中で陽性の抗体価を示した。陽性の血清学で、脾細胞を、抗原特異的B細胞の回収のために回収した。全ての手順を、IACUC(Institutional Animal Careand Use Committee)により承認されたプロトコルに従って行った。
マウスリード抗体723C2を、723C2のマウス可変ドメイン及びヒト定常IgG1WT、IgG1KO、又はIgG4Proドメインからなるキメラ抗体に変換した。マウス抗体723C2軽鎖可変領域(Vκ)及び重鎖可変領域(VH)の配列を、本明細書中の上の表1及び2において示す。IgG4Proは、Fab アーム交換を防止する1つの置換変異(Ser228Pro)を有する。IgG1KOは、エフェクター機能(ADCC)を低下させるために、ヒンジ領域中に2つの変異、Leu234Ala及びLeu235Alaを有する。キメラ抗体を生成し、抗体の機能を確認し、正しい配列が得られていることを保証する。ヒトIgG1WT、IgG1KO、及びIgG4Proフォーマット中のキメラ723C2の配列を表8中に示す。ヒトIgG1WT及びIgG4Pro中のキメラ723C2は、H-CDR3における変異、DCからDYを含む。しかし、ヒトIgG1KO中のキメラ723C2は変異を有さない。この部位における変異を表8中で強調している。抗体の可変領域を次に、設計及びスクリーニング過程を通じてヒト化させる。ライブラリーを作成し、ここでヒト残基及びマウス残基を、任意の所与の位置においてヒト残基又はマウス残基のいずれかがありうるように変動させた。そのようなライブラリーを、ヒト生殖系列とマウス抗体の間で異なるアミノ酸について作成した。親マウス抗体の機能を保持しているクローンだけを選択した。抗体723C2についての代表的なヒト化可変領域を表5及び6中に示す。
この様式において、抗体A、抗体B、抗体C、抗体D、及び抗体Eは、マウス抗体723C2(ヒトIgG4Pro/カッパ骨格中にクローニングされた)から由来するヒト化抗体であった。抗体A、B、C、D、及びEを表7中に示す。
A)組換えヒトPD-1に結合するヒトIgG4Pro骨格におけるキメラ抗PD-1抗体の動態及び親和性を下に示す(表9)。動態及び結合親和性を、ProteOn XPR36(Biorad、カリフォルニア州ハーキュリーズ)を使用し、単一カラム精製後の一過性トランスフェクションから生成された材料を使用して測定した。
B)親和性を、マウス抗体723C2から由来するヒト化抗PD-1抗体について測定した。動態結合データが、ProteOn XPR36(Biorad、カリフォルニア州ハーキュリーズ)を使用して測定され、1:1結合モデルに包括的に適合され、組換えヒトPD-1との相互作用が 1nM~10nMの範囲中であることを実証した(表10)。抗体PD1AB-6-4P(Celgeneに対するWO2017/058859において開示されているIgG4Pro骨格中の抗体)もテストした。
C)カニクイザルPD-1に結合する抗PD-1抗体の親和性及び動態データをProteOn XPR36で測定し、1:1結合モデルに包括的に適合させた(表11)。抗体、PD1AB-6-4Pもテストした。
D)ヒトPD-1への分子選択性
細胞ベースのアッセイにおけるヒトPD-1タンパク質に対する抗PD-1抗体の選択性を、フローサイトメトリーにより評価した。ヒトPD-1タンパク質を発現しない親 Jurkat細胞又はヒトPD-1タンパク質を発現するJurkat細胞を、下に示す濃度のAlexaFluor 647で標識した抗PD-1抗体とインキュベートした。コントロールとして、親細胞及びPD-1発現 Jurkat細胞を、抗TNPアイソタイプコントロール抗体とインキュベートした。インキュベーション後、細胞を洗浄して非結合抗体を除去し、PFA中で固定し、次に染色緩衝液中で洗浄した。Jurkat細胞への抗体の結合を、フローサイトメトリーにより評価された。非染色細胞をまた、ネガティブコントロールとしてフローサイトメトリーにより評価した。抗PD-1抗体は、少なくとも1マイクロモルまでヒトPD-1に選択的に結合し、ヒトPD-1タンパク質を発現する Jurkat細胞への用量依存的な抗体結合及びPD-1発現を欠く親 Jurkat細胞へのAlexaFluor 647標識抗PD-1抗体の結合の欠如により示される通りである。抗体C(Ab C)を使用した代表的な実験の結果を図1中に示す。
ヒトPD-L1-Fcは、BioRad ProteOn XPR36機器でのGLMチップのチャンネル1~3に 60μg/mLの濃度で結合したアミンであった;3つのテスト抗体、抗体C、MK-3475(ペンブロリズマブ)、及びPD1AB-6-4Pは、30μg/mLでそれぞれチャネル 4、5、及び6上に結合したアミンであった。ヒトPD1-Fcをチップ表面上のチャネル1~ 6にわたり25nMの濃度で注入した。センサーグラムは、PD-L1とPD-1受容体の間での特異的結合を示す(図2A)。500nM 抗体C、MK-3475、及びPD1AB-6-4Pを25nM PD1-Fcと事前に混合し、チップ上の全てのチャネルにわたり分析物として注入し、個々の抗体がPD-1へのPD-L1の結合を阻害するか否かを評価した。抗体C及びPD1AB-6-4Pの両方が、センサーグラムにより実証されるように、PD-1抗原への結合についてPD-L1と非競合的である。MK-3475及びPD-L1は、競合アッセイにおいて観察された非結合センサーグラムに基づくPD-1との互いの潜在的な結合ブロッカーである(図2B)。
PD-1/PD-L1相互作用を、H-CDR3中でのDCからDYへの変異を伴わないヒトIgG4Pro骨格中でのPD-1アゴニスト抗体723C2の存在において調べた。複数のアッセイを利用して、抗体723C2によってPD-L1のPD-1への結合が増強されることを実証した。生化学的ELISAベースのアッセイを利用して、プレート結合PD-L1へのPD-1の結合を評価した(BPS Bioscience)。白色96ウェルマイクロプレートを、PBS溶液中の2μg/mlでの50μlのPD-L1で、一晩4℃でコーティングした。上清を除去し、プレートを、製造元であるBPS Bioscienceにより提供された1×イムノ緩衝液(カタログ番号72005)で3回洗浄し、その後にブロッキング緩衝液を用いた室温(RT)で1時間にわたるブロッキングが続いた。抗体を、関連コントロールと共に加え、その後に室温で2時間にわたる0.5ng/ml(10ng)のPD-1ビオチンの添加が続いた。プレートをブロッキング緩衝液で10分間にわたり遮断した。ストレプトアビジンホースラディッシュペルオキシダーゼ二次抗体を、洗浄したプレートに1時間にわたり加えて、その後にPD-1アッセイ緩衝液での洗浄が続いた。プレートを10分間にわたり遮断した。化学発光基質混合物を読み取り直前にプレートに加えた。化学発光シグナルは、ルミノメーター(Envision)で、又は化学発光を読み取ることが可能なマイクロタイタープレートで読み取った。
PD-1とPD-L1との増強した相互作用が、抗体723C2の存在において観察されたが、アイソタイプコントロールで処理されたサンプルと比較し、増加した化学発光シグナルにより示される(図3A)。抗体723C2は、図3A中で723C2-4Pとして命名されている。これは、MK3475(公知の抗PD-1アンタゴニスト抗体)とは対照的であり、それは、PD-L1とPD-1との相互作用を遮断した。抗体PD1AB-6-4Pは、このアッセイにおいて PD-1-PD-L1の限定的な増強を実証した(図3A)。
細胞ベースのアッセイを利用して、抗体723C2の存在においてPD-1/PD-L1相互作用の増強を実証するELISAベースの結果を確認した。ここで、PD-1/PD-L1の相互作用を、DELFIA(解離増強ランタニド蛍光イムノアッセイ)受容体-リガンド結合アッセイ(Perkin Elmer)を用いて、PD-1を過剰発現するCHO細胞への可溶性PD-1の結合を測定することにより評価した。
10,000個の細胞を播種し、37℃+5% CO2インキュベーター(加湿インキュベーター)で一晩インキュベートした。ビオチン標識PD-L1EC 10(130nM)及び10μlのPD-1抗体を各々のウェルに加えて、RTで1時間にわたりインキュベートした。プレートを50μlの1×TRF 洗浄緩衝液で2回洗浄した。20μLのEu-ストレプトアビジン試薬をアッセイプレートに加えて、室温で1時間にわたりインキュベートした。Enhancement Solutionを加えて、RTで30分間にわたりインキュベートした。プレートを蛍光プレートリーダーで読み取った(励起:320又は340nm、発光:615nm)。このアッセイでは、ELISAアッセイの確認では、この細胞ベースのアッセイにおける抗体723C2の存在によって、PD-1/PD-L1相互作用が強化された(図3B)。抗体723C2は、図3B中で723C2-4Pとして命名されている。
PD-1がCHO細胞で発現された第2の細胞ベースのアッセイをまた、PD-1/PD-L1相互作用を評価するために利用した。このアッセイでは、PD-1を発現するCHO細胞へのPD-L1多量体結合をフローサイトメトリーにより測定した。50μlの2×106細胞/mlを各々のウェルに加えた(100,000個細胞/ウェル)。細胞を遠心分離し、50μlの示した濃度の抗体中に再懸濁し、氷上で60分間にわたりインキュベートした。PDL1-ビオチン及びストレプトアビジン-APCを染色緩衝液中で組み合わせた(1μg/ml PDL1-ビオチン+0.25μg/mlストレプトアビジン-APC)。50μlの2×PDL1-ビオチン/ストレプトアビジン-APC 混合液を細胞に加え、氷上で60分間にわたりインキュベートする。細胞を洗浄し、180μlの染色緩衝液+20μPFA中に再懸濁し、データをBD LSR IIで取得した。図3C中に示すように、この細胞ベースのアッセイはまた、抗体723C2の存在における PD-1へのPD-L1の増強した結合を実証した。抗体PD1AB-6-4Pは、PD-L1結合に対する効果を有さなかった一方で、MK3475アンタゴニスト抗体は、PD-1へのPD-L1の結合を阻害した(図3C)。抗体723C2は、図3C中で723C2-4Pとして命名されている。
上に記載するCHO PD-1-PD-L1Delphia-Eu TRFアッセイをまた、抗体C、抗体PD1AB-6-4P、抗体1-4Pro、抗体PD1B1090-4Pro、抗体PD1B1094-4Pro、及び抗体ANB-030-4Proを使用して実施した。抗体1-4Proは、Eli LillyへのWO2019/168745において記載されている抗体1の重鎖及び軽鎖可変領域をIgG4-Pro骨格に含む。抗体PD1B1090-4Pro及び抗体PD1B1094-4Proは、Janssen BiotechへのWO2018/226580において記載されている、それぞれPD1B1090及びPD1B1094の重鎖及び軽鎖の可変領域をIgG4-Pro骨格中に含む。抗体ANB-030-4Proは、CAS番号 CAS2412764-40-8の下に記載されている抗体ANB-030の重鎖及び軽鎖可変領域をIgG4-Pro骨格中に含む(また、AnaptysbioへのWO2020/247648において記載されているAPE12537の重鎖及び軽鎖可変領域に対応する)。IgG4-Pro骨格中の抗TNP抗体も含まれた。
抗体Cは、濃度依存的にPD-1\PDL-1結合の一貫した(N=3) 増強を示した(図3D)。全ての他の抗PD-1アゴニストは、PD-1\PDL-1結合の増強を一貫して示さなかった(図3D)。
THP-1/Jurkat PD1NFAT 共培養アッセイを、複数のキャンペーンから生成された抗PD1抗体のアゴニスト活性を評価するために開発した。THP-1細胞株をATCCから得た。Jurkatレポーター細胞株を内部で生成した。Jurkatレポーター細胞は、細胞表面上でヒトPD-1(hPD-1)を過剰発現し、NFAT駆動型ルシフェラーゼレポーターも発現し、刺激への応答における細胞の活性化状態が測定される。Jurkat PD1NFAT細胞は、THP-1細胞の存在においてCD3xCD33 BiTEで活性化される。BiTEの抗CD33アームは、THP-1細胞上に発現されるCD33に結合する一方で、抗CD3アームはJurkat細胞上のCD3分子に結合する。BiTEは、THP-1とJurkat細胞を会合させる役割を果たし、Jurkat細胞を活性化しながら、2つの細胞間に免疫シナプスの形成をもたらす。Jurkat PD-1NFAT細胞の活性化を、NFAT 駆動型ルシフェラーゼレポーターにより測定する。このアッセイを、アゴニスト抗体を同定するために、抗PD1抗体の存在において実行した。活性化における20%又はそれ以上の低下を示した分子を、ルシフェラーゼシグナルの喪失により示されるように、アゴニスト抗体として分類した(表12)。表12中の抗PD-1抗体306E6から820C3は、マウスIgG1骨格上にあった。これらの抗体のいくつかを、ヒトIgG4Pro骨格(表12中でキメラ抗体として示されている)での追加のプロファイリングのために選択した。
上位の抗PD1抗体を選択するために使用された初代細胞アッセイは、hPD1ノックインマウス脾細胞アッセイであった。脾臓を、マウスPD1の代わりにヒトPD1を発現するC57BL/6マウスから収集した。脾細胞を脾臓から単離し、0.1μg/mlの濃度の抗CD3(クローン2C11)で活性化した。T細胞の活性化を、MSD分析(Meso Scale Discovery)によりmIFNγレベルを定量化することにより、48時間後に測定した。このアッセイは、THP-1/Jurkat PD1NFAT スクリーニングアッセイより選択された抗PD1抗体の存在において実行された。このアッセイから同定された上位の分子を、mIFNγの阻害%(50%又はそれ以上)及び配列クレードに基づいて選択した。阻害値及びIC50値を表13中に示す。
抗PD-1アゴニスト抗体を、ヒト初代細胞アッセイにおいて、IFNγ産生により測定されるように、T細胞の機能活性を調節するそれらの能力についてさらに特徴付けた。PBMCをヒト全血から単離して、1.5pMの抗CD3(クローンOKT3、BioLegend)で活性化させた。T細胞の活性化及び機能を、MSD分析によりhIFNγレベルを定量化することにより、72時間後に評価した。同定された抗PD-1アゴニスト抗体は、アイソタイプコントロール処理細胞と比較して、IFNγ分泌を低下させることができた(表14A)。
抗体C、抗体1-4Pro、抗体PD1B1090-4Pro、抗体PD1B1094-4Pro、抗体ANB-030-4Pro、及びアバタセプトをまた、このアッセイにおいてテストした。結果を表14B中に示す。抗体C、IgG1野生型骨格中の及びIgG1KO骨格中の抗体1-4Pro、抗体PD1B1090-4Pro、抗体PD1B1094-4Pro、及び抗体ANB-030-4Proの可変領域、ならびにアバタセプトをまた、このアッセイにおいてテストした。結果を、下に要約する表14C中に示す。各々の実験では、5人のドナーをテストした。
IgG1KO骨格中の抗体1-4Pro、抗体PD1B1090-4Pro、抗体PD1B1094-4Pro、及び抗体ANB-030-4Proの可変領域についてのIFNγの阻害は、30nMを上回るIC50値で40%未満であった。
抗PD-1アゴニスト抗体を、Th17分化T細胞によるIL-17分泌の機能的阻害についてテストした。初代細胞共培養アッセイを、PD-1によるIL-17の調節を評価するために開発した。PBMC から単離されたヒト初代T細胞は、以下の傾斜条件下でTh17分化した:CD4 T細胞は、Th17傾斜培地(X-VIVO15培地+IL-1β(10ng/mL)、IL-23(10ng/mL)、IL-6(10ng/mL)、IL-2(2ng/mL)、TGFβ(0.5ng/mL)、5μg/mL 抗IL4、5μg/mL 抗IFNγ)中で4日間にわたり0.5μg/mlプレート結合抗CD3(クローンUCHT1)で刺激した。4日後、細胞を抗CD3コーティングプレートから取り出し、Th17傾斜培地を含むフラスコに移した。分化後、Th17細胞を少なくとも3日間にわたり休止させ、次に自己単球と共培養し、PD-1抗体の存在において40fM 抗CD3(クローンOKT3)で再刺激した。共培養系は、抗PD-1抗体がアゴニスト活性を示すために必要なFc要件に起因して要求された。IL-17の阻害が、このアッセイにおいて、抗PD-1アゴニスト抗体の存在において観察された。抗体のIC50及びIL-17応答の最大阻害を下の表中に示す(表15)。最大阻害をアイソタイプコントロール抗体に対して比較した。
濾胞性ヘルパー T(Tfh)細胞活性をインビトロで阻害する抗PD-1アゴニスト抗体の能力を評価するためのアッセイを開発した。CD4T細胞及び自己単球をALLCELLSから得た。T細胞を、IL-23(25ng/ml)及びTGFβ(5ng/ml)の存在においてDynabeads Human T-Activator CD3/CD28(Gibco)で細胞を5日間にわたり活性化することによりTfh系統に傾斜させ、次に、細胞を洗浄し、活性化ビーズを除去した後、4.5pM 抗CD3(クローンOKT3、BioLegend)及び抗PD-1アゴニスト抗体の存在において自己単球と組み合わせた。24時間後、上清を収集し、IL-21の存在についてアッセイした(Meso Scale Discovery、MSD V-Plex Human IL-21Kit)。再刺激されたTfh分化細胞のIL-21産生は、抗PD-1アゴニスト抗体により阻害した。代表的なIC50及びEmax阻害値を表16中に示す。
アゴニスト抗体の機能活性に対するFc-Fcg受容体相互作用の役割を、異なる骨格フォーマット(IgG1野生型、IgG1KO、又はIgG4 Pro)又は二価抗体フラグメント(F(ab’)2フラグメント)での候補PD-1アゴニスト抗体を利用することにより特徴付けた。抗体バリアントの機能活性を、上に記載するヒトPBMCアッセイにおいて活性化 T細胞からのIFNγ産生を調節する能力により評価した。機能的アゴニスト活性は、IFNg産生における低下により測定され、親の723C2及び820C3抗体の二価F(ab’)2フラグメントでは失われる(図4A及び4B)。対照的に、ヒトIgG4Pro骨格における全長抗体は、用量依存的な様式においてIFNγ産生を阻害した(図4A及び4B、それぞれ723C2-4P及び820C3-4Pとして命名する)。これらのアッセイでは、ヒトPBMCを全血から単離し、抗PD-1抗体又は示された抗PD-1抗体のF(ab’)2フラグメントの存在において1.5pMの抗CD3クローンOKT3で活性化した。72時間後、上清中のヒトIFNガンマサイトカインレベルをMSD分析により測定した。
これは、Fc相互作用が抗PD-1抗体の機能的アゴニスト活性のために要求されることを示唆しているため、723C2抗体をIgG1WT、IgG1KO、及びIgG4Pro骨格で生成して、これらの相互作用をさらに特徴付けた(それぞれ723-IgG1WT、723-IgG1KO、及び723-IgG4Pro、下の表17中)。IgG1WT及びIgG4 Proの両方が、異なる程度でヒトFc受容体に結合する一方で、IgG1KO骨格はFc受容体への大きく低下した結合を有する。IgG4 Pro上の抗PD-1アゴニスト抗体が、ヒトPBMCアッセイにおける最も高い程度のIFNγ阻害を示した一方で、IgG1KO上の抗体は、大きく低下した結合を示した(表17)。まとめると、これらのデータは、抗PD-1抗体の機能的アゴニズムがFc相互作用に依存的であることを示す。
インビボ異種CD4+T細胞GvHDマウスモデルを使用して、PD-1アゴニスト抗体の有効性をテストした。群当たり8匹のNSG マウス(NOD.Cg-PrkdcscidIl2rgtm1Wjl/SzJ、Jackson Laboratory)に、健常ドナーleukopaksからの5×106個CD4+T細胞(ネガティブセレクションにより精製)を静脈注射した。マウスに、以下に従って0.625mg/kg IPを週2回投与した:群1:723(IgG4-Pro)、群2:PD1AB-6-4P(IgG4-Pro)、群3:抗TNPアイソタイプ(IgG4-Pro)、群4:アベルマブ(hIgG1-LALAPG)、群5:抗TNPアイソタイプ(hIgG1-LALAPG)、群6:CTLA4-Ig(hIgG1-LALA)。TNPはトリニトロフェノールである。LALAは、抗体のエフェクター機能を破壊するために一般に使用されるLeu234Ala/Leu235Ala変異を表する。PGはPro329Gly変異を表し、それは、Fcガンマ受容体への結合を防止することによりエフェクター機能を排除する。
3回の実験的繰り返しを実行し、各々が固有のドナーを伴った。第4週までに、ヒト細胞蓄積の有意な阻害が、テストされた全てのドナーについてのそれらのアイソタイプが一致されたコントロールと比較して、群1、2、及び6において認められた(表18)。4週目での炎症性サイトカインの定量化は、全てのドナーにおいてヒトIFNγ、TNFα、及びIL-10のレベルにおける有意な低下を示した(表19)。ヒトIL-1β、IL-2、IL-4、IL-6、IL-8、IL-12p70、及びIL-13をまたテストしたが、しかし、全てが、アッセイについての検出の限度を下回った。
抗体Cの薬物動態(PK)を、0.1、0.3、及び1.5mg/kgの単回静脈内(IV)ボーラス投与又は1.5mg/kgの皮下(SC)投与後に、中国起源の雄カニクイザルにおいて評価した(n=3/群)。抗体Cの血清中濃度を、2つの異なる MSDイムノアッセイフォーマットを使用して決定された:(1)「総」薬物の一般的な抗ヒト捕捉及び検出アッセイならびに(2)抗原(PD1-ECD)捕捉及び抗ヒト検出を伴う「遊離」薬物アッセイ。両方のアッセイのPK プロファイルを重ね合わせることができ、内因性sPD-1が抗体Cの測定に干渉せず、TMDDに対するほとんど又はまったく効果を有さなかったことを示唆している。抗体Cは、全体的なクリアランスへの標的媒介性薬物動態(TMDD)の寄与を示唆する、0.1~0.3mg/kgの間の用量依存的なCLを示した(遊離アッセイと「総」アッセイの両方を使用)。それぞれの用量の各々についてのNCA薬物動態パラメータの要約を、下の表20中に示す。
CHO細胞におけるトランスフェクション及び産生:
CHO-E細胞を~2x10E6個細胞/mLで、Irvine BalanCD Transfectory CHO + 4mM L-グルタミン(又はGlutamax)中にトランスフェクトする。1Lのトランスフェクションのために要求される量は、0.15mgのHC DNAプラス0.3mgのLC DNA及び1.05mgのフィラーDNA(ニシンの精子)及び0.15mgのXBP1DNAである。DNAを100mLのOptiPro SFM中に希釈し、0.2μmフィルターを通じて滅菌ろ過する。0.75mLのMirus TransIT Proトランスフェクション試薬を、希釈したDNA 混合物に加え、DNA複合体を、調製したCHO-E細胞に直ぐに加え、振盪フラスコを、37℃、5% CO2、140rpmのシェーカーに戻す。トランスフェクション後の24時間に、温度を32℃に変えて、2mLのGibco Anti-Clumping Agent及び100mLのIrvine Transfectory Supplementを、トランスフェクトされた細胞に加える。トランスフェクション後の5日に、シェーカーの温度を30℃に変える。200mLのIrvine Transfectory Supplementを、グルコースレベルがいつ2g/L~1g/Lの間に下落するかに依存して、5日目又は7日目の間に加える。トランスフェクトされた培養物を10日間にわたり維持する。回収は、細胞をスピンダウンし、その後の0.2μm PESフィルター(Thermo Scientific)を通じた滅菌ろ過により行う。
回収後、清澄化された細胞培養上清を、以下のように、プロテインAバイオセンサーを伴うForteBio/Pall Octet Red 96機器により、力価についてサンプリングした。
抗体A、抗体C、及び抗体Eについての力価は18~38mg/Lの間であり、タンパク質精製からの約80%回収、及びSEC精製後の98%を上回る単量体を伴った。タンパク質を、10mMヒスチジン-HCl、pH6.0を含む最終緩衝液中で緩衝液交換して、4℃で少なくとも4ヶ月間にわたり安定であり、この緩衝液中での180mg/mlまでの溶解度を伴う。
AUC:0.5~1mg/mlの濃度での沈降速度方法により測定される分析用超遠心分離;SEC:サイズ排除クロマトグラフィー。%M:単量体のパーセント。
材料及び方法
マウス抗体及び試薬。抗hPD1(EH12.2H7)(Biolegend、329912);抗hCD48(Bio-gems、10511-25-500);IgG1(カタログ番号16-4714-85)、抗hCD3(OKT3)(16-0037-85)、抗hCD3(UCHT1)(16-0038-85)、及び抗CD11a(140011982)(eBiosciencesから);抗hCD71(Southern Biotech、9670-14)。aCD3/aCD28ヒトT細胞アクチベーターDynabeads(Gibco、11131D)。
Imagestream。Jurkat PD-1細胞を、AF-488コレラ毒素(Life Technologies、V-34403)及び架橋抗体(Jackson ImmunoResearch)及びAPCaCD3(Biolegend、317318)、PV786aPD-1(Biolegend、329930)、又はAPCaCD48(Sigma、SAB4700193)のいずれかを用いて、XVIVO 15培地(Lonza)中で、氷上で10分間にわたりインキュベートした。細胞を、予熱したX-VIVO 15への移入により活性化し、追加の12分間にわたりインキュベートした。細胞の活性化を、冷PBS-2%PFA(約1:10の比率、細胞:PFA)の添加により停止させ、細胞を、氷上で20分間にわたり固定溶液中でインキュベートした。細胞を洗浄し、XVIVO中に再懸濁し、Imagestreamソフトウェアを使用してキャップ形成及び周囲長閾値について分析した。
フローサイトメトリー。1×105個のJurkat、Jurkat-PD-1、又はaCD3/aCD28で刺激された初代ヒトT細胞を、1mg/mlの初代 MAbと4℃で1時間にわたりインキュベートした、又は二重特異性分子がテストされた場合、8点結合曲線を6.25mg/mlの開始濃度から生成し、1:4で段階希釈した。6.25mg/mlの開始濃度から1:4に段階希釈する。細胞を洗浄し、それぞれ1:100希釈のPE-抗マウスIg(Life Technologies、P852)、又は1:800希釈のPE-ヤギ抗ヒトF(ab’)2(Invitrogen AHI1707)を用いて4℃で1時間にわたり染色した。サンプルを洗浄し、1×固定/溶解緩衝液(eBioscience、00-5333-57)中で固定し、LSR2(BD)で分析した。
PD-1相補性アッセイ。全長 PD-1-PK及び細胞内全長 SHP1-EA 融合タンパク質を過剰発現する2×104個のJurkat T細胞をDiscoverX(DRX-BI-080515A)から購入し、製造元の説明に従って培養した。細胞を細胞播種培地(DiscoverX、93-0563R4B)中に再懸濁し、一次マウス又はヒト抗体と4℃で30分間にわたりプレインキュベートした。実験に依存して、細胞を、10mMのpan-Srcキナーゼ阻害剤PP2(Abcam、ab120308)又は不活性類似体PP3(Abcam、ab120617)と追加でプレインキュベートした。細胞を洗浄し、架橋二次ヤギ抗マウスIgG(Thermo Scientific、31170)を伴って、又は伴わずに処理した。細胞を、384-white Opti-Plates(PerkinElmer)に移し、Flash Detection試薬(DiscoverX、93-0247)を受けて、EnVision Plate Reader(PerkinElmer)において読み取った。
初代huT細胞活性化。初代ヒトPan-T細胞(AllCells、PB009-1F)を500nMのCell Trace Violet(Life technologies、カタログ番号c34557)で標識した。Epoxy-dynabeads M450(Invitrogen、14011)を、製造元の指示に従って、2.5mgのマウスAbs/107ビーズで共有結合的にコーティングした。細胞を未刺激で放置するか、又はAbコーティングされたエポキシ ビーズの存在において、プレートに結合した抗CD3(UCHT1)(250及び500ng/mL)で刺激した。細胞を96時間後に回収し、BV510抗CD4(BD、562970)Abs及びPeCy7 抗CD8(BD、335787)Absで染色し、細胞増殖を、LSR2(BD)中でのCell Trace Violet希釈により分析した。初代記憶CD4+/CD45RO+ T細胞(AllCells、PB009-7F)を、それぞれ1mg/ウェルのプレート結合アイソタイプコントロール(ISO)又はBsAbの存在において1mg/ウェルのプレート結合抗CD3(UCHT1)で刺激した。培養上清を72時間で回収し、IL-2及びIL-10分泌(MSD)について分析した。
BsAbsの生成及びコンストラクトの設計。二重特異性抗体(BsAb)を、公開された抗CD48(US2012/0076790)及び抗PD1(WO2011/110621Al)配列から生成し、それらをビルディングブロックとして使用した。二重特異性コンストラクトを、2つの異なる標的可変領域(IgG1-KO)のヘテロ二量体化を促進するノブインツーホール技術を用いて設計し、抗PD-1及び抗CD48(PD-1/CD48)、又はコントロールとして抗PD-1及び抗TNP(PD-1/ISO)を含むBsAbsを生成した(図7A)。それぞれの標的について得られた可変領域配列を、ヒト定常領域を含むpTT-5(カナダ国立研究評議会からライセンス供与)発現ベクター中にクローニングした。簡単には、可変領域のアミノ酸配列を、哺乳動物での発現のためにコドン最適化した。標的V遺伝子の軽鎖及び重鎖を、結合リンカーセグメントを含む同じ発現ベクター中にクローニングした。ベクターを、EcoRI及びNheI認識部位を使用した制限酵素消化により直線化した。可変領域についてのDNA配列を、Integrated DNA Technologies(IDTDNA)からGブロック(dsDNA)として注文し、ベクター及び隣接するリンカーセグメントへの重複した相同末端を伴う。Gブロックを次に、製造元のプロトコルに従って、Gibsonアセンブリ方法(NEBuilder HiFiキット、New England Biolabs、カタログ番号E5510S)を介して連結させた。従来のクローニングを次に、アセンブリ混合物をコンピテント細胞(NEB 5-alpha C2987、New England Biolabs)中に形質転換することにより完了し、100μg/mlカルベニシリン(Teknova)を伴うLB寒天プレート上で、37℃で一晩増殖させた。個々のコロニーを採取し、カルベニシリンを伴うLB 培地中で、37℃で一晩増殖させた。インサートについての陽性クローンを、Lasergeneソフトウェア パッケージ(DNAstar)を使用した配列分析により確認した。配列が確認されたプラスミド DNAを0.5L培養中でスケールアップし、次に、製造元のプロトコルに従って、Plasmid Plus megaprepキット(Qiagen、カタログ番号12981)を介して精製した。
CHO-E一過性トランスフェクション。CHO-E細胞を、2mMグルタミンを添加したFS-CHO中で2e6個細胞/mLでトランスフェクトする。1L mAbトランスフェクション容量について、1mgの軽鎖(LC)プラスミドDNA及び0.5mgの重鎖(HC)プラスミド DNAを100mLのOptiPro SFM(Gibco)中で希釈し、0.2μmフィルター(Millipore)を通じて滅菌ろ過する。1.5mLのTransIT Pro(Mirus Bio LLC)トランスフェクション試薬を加えて、室温で15~30分間にわたりインキュベートする。複合体を次に、調製したCHO-E細胞に加えて、振盪フラスコをシェーカーに戻す。トランスフェクション後の24時間に、10mLのAnti-Clumping Agent及び150mLのCHO CD Efficient Feed B(両方ともGibcoから)をトランスフェクトした細胞に加えて、温度を32℃に変える。トランスフェクトされた培養物を6~12日間にわたり維持し、細胞増殖、生存率、及び栄養消費について培養を通して定期的にモニターする。培養物の回収を、4℃で4700rpmでの遠心分離により完了し、その後に滅菌ろ過が続く。
BsAbs精製。収集した培養上清を、1.0ml/分で、緩衝液A(DPBS、pH7.2)で事前に平衡化したGE(カタログ番号11003493)からの1ml HiTrap MabSelect SuReカラムにロードする。カラムを緩衝液A、緩衝液B(DPBSプラス1.0M NaCl)の各々の10mlで、1ml/分の緩衝液 Aで再び洗浄する。次に、結合したタンパク質を30mM酢酸ナトリウム、pH3.5で溶出する。5ml画分を1%体積対体積の3M酢酸ナトリウム、pH~9で中和する。最終緩衝液は、プロテインA 溶出後に60mM NaOAc、pH~5である。単量体のパーセンテージは、aSECにより、PD1/ISOについて71%&PD1/CD48について63%であった。
MabSelect Sureで精製された材料をさらに研磨して、陽イオン交換により凝集物を除去した。Thermo FisherからのPoros GoPure HS Pre-packedカラム(カタログ番号 4481316)をイオン交換のために使用した。緩衝液A(60mM NaOAc. pH 5.0)で事前に平衡化した1mlのPoros HSカラム上にプロテインA サンプルをロードし、10カラム体積の緩衝液Aでカラムを洗浄する。次に、結合したタンパク質を、20カラム体積中の緩衝液B(60mM NaOA、1M NaCl、pH 5.0)中の0%から40%の勾配で、0.5ml/分で溶出する。ピーク付近の画分をプールし、塩濃度を100mM NaClに調整する。ろ過ユニットでサンプルを無菌的にろ過する。タンパク質濃度を測定し、エンドトキシンレベルを測定し、SDS-PAGEならびにaSECを実行する。
NFATルシフェラーゼアッセイ。Jurkat PD-1NFATレポーター細胞株を社内で生成した。GeneCopoeiaからのヒトPD-1(EX-B0169-M02)をベクター中にクローニングし、それを、エレクトロポレーションを介してJurkat細胞(ATCC)中にトランスフェクトした。NFATルシフェラーゼレポーター(Promega E8481)を次に、エレクトロポレーションを介してPD-1発現クローン中にトランスフェクトした。THP-1細胞株をから購入しATCC(TIB-202)、製造元の指示に従って培養した。
Jurkat PD-1及びNFATレポーター細胞をアッセイ培地(RPMI、2% HI-FBS)中に再懸濁し、3×104個細胞/条件を、BsAbsの用量(100nMの開始濃度及び1:3希釈)を用いて、384の平底Opti-Plates中で15分間にわたりプレインキュベートした。THP-1細胞(3×104個細胞/条件)を加えて、細胞を、CD3×CD33アクチベーターの10nM溶液を用いて、37℃で6時間にわたり刺激した。NFATレポーターを、Steady-Glo(登録商標)Luciferase Assay reagent form 15 min(Promega、E2520)の添加により分析し、EnVisionプレートリーダーにおいて読み取った。
CD48及びPD-1の架橋によって、PD-1リン酸化が促進される。CD48は、マウス及びヒトのリンパ球における確立された脂質ラフト及びIS常在タンパク質である(Elishmereni and Levi-Schaffer,2011)。PD-1及びCD3のものと比べて、脂質ラフトにおけるCD48の存在及び存在量をより良く評価するために、本発明者らはImageStream実験を行って、PD-1を過剰発現するJurkat細胞におけるコレラ毒素(CT)誘導性脂質ラフト合体(キャッピング)とこれらの受容体の共局在を単一細胞レベルで定量化した。CTにより誘導された脂質ラフトキャップ内のCD48、CD3、及びPD-1の共局在の分析を、フルオロフォア標識MAbを使用して行った。この分析によって、PD-1とは異なり、CD3及びCD48がCTにより誘導されたキャッピング内で容易に観察されることが示された。周囲長の定量化では、より小さな周囲長の値がキャッピングと相関し、活性化後にCD48のキャッピングが明らかであるが、しかし、CD3よりもわずかに少ないことが明らかになった。対照的に、PD-1は通常、CTと共局在しなかったが、PD-1が、脂質ラフトに富むISに動員されるために、活性な過程(例、PDL-1との相互作用)を要求するという仮説と一致している(Yokosuka et al.,2012)。
脂質ラフト中へのCD48の存在及びSrcキナーゼ(T細胞のLck)との構成的会合によって、CD3と同様に、CD48とPD-1の近似が PD-1活性化/リン酸化を誘導するという仮説を本発明者らは立てることが可能であった。なぜなら、PD-1活性化のための基準機構が、PD-1の細胞内ITSM及びITIMドメインのLck媒介性リン酸化を要求するからである(Chemnitz et al.,2004;Parry et al.,2005;Sheppard et al.,2004)。この仮説をテストするために、カスタマイズされたJurkat細胞株を、bガラクトシダーゼ(PK)の1つの半分を伴うヒトPD-1融合タンパク質(PK)、及びbガラクトシダーゼの相補的な半分を伴うサイトゾル全長SHP1融合タンパク質(EA)を発現するように生成した。PD-1活性化を、このように、リン酸化PD1へのSHP1の動員に起因する PK/EA相補性の機能として測定し、それによって機能的なbガラクトシダーゼが産生される。これらの細胞におけるPD-1、CD48、及びCD3の発現を確認した後、実験を設定し、架橋時にPD-1活性化を誘導する、CD48に対する MAbの潜在能を評価した(図5)。PD-1活性化は、PD-1MAb又はFc特異的二次F(ab’)2抗体の非存在において誘導されなかった。二次抗体との架橋によって、PD-1活性化が約3倍だけ誘導された;しかし、CD48又はCD3 Mabの存在において、PD-1活性化が約9倍だけ増強され、CD48又はCD3とPD-1との密接な会合が、PD-1活性化を増強させることができることを示している。自己架橋により誘導される低レベルのPD-1活性化は驚くべきことではなかった。なぜなら、試験によって、Lckの小さな分画が、T細胞におけるPD-1と構成的に会合していることが実証されているためである(Sheppard et al.,2004)。
