Claims (12)
1. Medij za kulturu stanica, naznačen time, da služi za uzgoj epitelnih matičnih stanica, koji sadrži bazalni medij za kulturu i nadalje sadrži
a. BMP inhibitor,
b. Wnt agonist odabran iz R spondin porodice, i
c. između 5 i 500 ng/ml mitogenih faktora rasta odabranih od epidermalnog faktora rasta (EGF), transformirajućeg faktora rasta alfa, fibroblastnog faktora rasta (FGF), neurotrofnog moždanog faktora i keratinocitnog faktora rasta (KGF).
2. Medij za kulturu stanica prema patentnom zahtjevu 1, naznačen time, da BMP inhibitor je noggin.
3. Medij za kulturu stanica prema patentnom zahtjevu 2, naznačen time, da se noggin dodaje u bazalni medij za kulturu u koncentraciji od najmanje 10 ng/ml, najmanje 20 ng/ml, najmanje 50 ng/ml, ili najmanje 100 ng/ml, pri čemu koncentracija noggina poželjno iznosi oko 100 ng/ml.
4. Medij za kulturu stanica prema bilo kojem od prethodnih zahtjeva, naznačen time, da je Wnt agonist koji se bira iz R spondin porodice, bilo koji od R spondina 1, R spondina 2, R spondina 3, ili R spondina 4.
5. Medij za kulturu stanica prema patentnom zahtjevu 4, naznačen time, da se kod Wnt agonista radi o R spondinu 1, koji se dodaje u bazalni medij za kulturu u koncentraciji koja poželjno iznosi najmanje 50 ng/ml, najmanje 100 ng/ml, najmanje 200 ng/ml, najmanje 300 ng/ml, najmanje 500 ng/ml, pri čemu koncentracija poželjno iznosi oko 500 ng/ml.

7. Medij za kulturu stanica prema bilo kojem od prethodnih patentnih zahtjeva, naznačen time, da mitogeni faktor rasta sadrži EGF.
8. Medij za kulturu stanica prema patentnom zahtjevu 7, naznačen time, da se EGF dodaje u bazalni medij za kulturu u koncentraciji od oko 50 ng/ml.
9. Medij za kulturu stanica prema bilo kojem od prethodnih patentnih zahtjeva, naznačen time, da medij za kulturu stanica sadrži noggin kao BMP inhibitor, EGF kao mitogeni faktor rasta, i R spondin 1 kao Wnt agonist, pri čemu medij za kulturu stanica opcionalno nadalje sadrži Wnt3a kao Wnt agonist.
10. Medij za kulturu stanica prema bilo kojem od prethodnih patentnih zahtjeva, naznačen time, da medij za kulturu stanica nadalje sadrži:
a. Rock inhibitor; ili
b. Notch agonist, opcionalno DSL peptid.
11. Medij za kulturu stanica prema bilo kojem od prethodnih patentnih zahtjeva, naznačen time, da bazalni medij za kulturu sadrži Advanced DMEM/F12 (poboljšani DMEM/F12) ili Advanced RPMI (poboljšani RPMI), opcionalno dopunjen s jednim ili više B27, N-acetilcisteina i/ili N2.
12. Medij za kulturu stanica prema bilo kojem od prethodnih patentnih zahtjeva, naznačen time, da medij za kulturu stanica ne sadrži serum.
