Claims (21)
1. Transgeni murinski sisavac, naznačen time, da posjeduje integriranu molekulu nukleinske kiseline u svojem genomu, koja kodira prerazmještaj varijabilnog područja humanog imunoglobulina slabog lanca, pri čemu se taj slabi lanac može spariti s najmanje dva različita jaka lanca koje kodiraju murinski sisavci, tako da se različitost u specifičnosti protutijela zadržava kroz prerazmještaje i hipermutacije u jakim lancima, gdje spomenuti slabi lanac nadalje sadrži stalno područje murinskog slabog lanca.
2. Transgeni murinski sisavac prema zahtjevu 1, naznačen time, da je to miš.
3. Transgeni miš prema zahtjevu 2, naznačen time, da je integracija na mjestu koje je otporno na prigušenje.
4. Transgeni miš prema zahtjevu 3, naznačen time, da je integracija na Rosa-mjestu.
5. Transgeni murinski sisavac prema bilo kojem od zahtjeva 1 do 4, naznačen time, da se pribavlja molekula nukleinske kiseline koja kodira slabi lanac, zajedno sa sredstvom koje dopušta ekspresiju spomenute molekule nukleinske kiseline, bitno ograničenu na stanice roda B-stanica.
6. Transgeni murinski sisavac prema zahtjevu 5, naznačen time, da se pribavlja molekula nukleinske kiseline koja kodira slabi lanac, zajedno sa sredstvom koje dopušta ekspresiju molekule nukleinske kiseline koja kodira slabi lanac, većinom za vrijeme određene faze u razvoju B-stanica.
7. Transgeni murinski sisavac prema zahtjevu 6, naznačen time, da spomenuto sredstvo sadrži promotor odabran iz skupine od sljedećih: CD19, CD20, μHC, VpreB1, VpreB2, VpreB3, λ5, Igα, Igβ, κLC, λLC i BSAP (Pax5).
8. Transgeni murinski sisavac prema zahtjevu 6 ili 7, naznačen time, da spomenuto sredstvo sadrži cre/lox-sustav.
9. Transgeni miš prema bilo kojem od zahtjeva 1 do 8, naznačen time, da se spomenuti slabi lanac može sparivati s najmanje dva murinska jaka lanca.
10. Transgeni murinski sisavac prema bilo kojem od zahtjeva 1 do 9, naznačen time, da se spomenuti slabi lanac može sparivati se s najmanje dva humana jaka lanca.
11. Transgeni murinski sisavac prema bilo kojem od zahtjeva 1 do 10, naznačen time, da se funkcionalno prigušuje barem jedno od endogenih mjesta koja kodiraju endogeni slabi lanac.
12. Transgeni murinski sisavac prema bilo kojem od zahtjeva 1 do 11, naznačen time, da se funkcionalno prigušuje endogeno K-mjesto slabog lanca.
13. Transgeni murinski sisavac prema bilo kojem od zahtjeva 1 do 12, naznačen time, da je humana VK-sekvenca, ona sekvenca slabog lanca, koju kodira molekula nukleinske kiseline.
14. Transgeni murinski sisavac prema zahtjevu 13, naznačen time, da je sekvenca zametka, ona sekvenca slabog lanca, koju kodira molekula nukleinske kiseline.
15. Transgeni murinski sisavac prema zahtjevu 14, naznačen time, da se sekvenca zametka bazira na 012.
16. Transgeni murinski sisavac prema zahtjevu 15, naznačen time, da sekvenca zametka je IGKV1-39*01 / IGKJ1*01.
17. Transgeni murinski sisavac prema zahtjevu 1, naznačen time, da molekula nukleinske kiseline koja kodira slabi lanac, u smjeru 5'-3' sadrži sljedeće: VK-promotor, humani predvodnik, humani V-gen i stalno područje štakora (K).
18. Transgeni murinski sisavac, naznačen time, da se on pribavlja s kazetom ekspresije koja u smjeru 5'-3' sadrži sljedeće: VK-promotor, predvodnik, prerazmješteni humani V-gen koji se može spariti s najmanje dva različita jaka lanca koja kodira murinski sisavac, te stalno područje štakora (K).
19. Postupak za proizvodnju željenog protutijela, naznačen time, da obuhvaća izlaganje murinskog sisavca prema bilo kojem od zahtjeva 1 do 18, antigenu, tako da se reakcija protutijela inducira, te izoliranje protutijela specifičnih za taj antigen.
20. Postupak za proizvodnju željenog protutijela, naznačen time, da obuhvaća izlaganje murinskog sisavca prema bilo kojem od zahtjeva 1 do 18, antigenu, tako da se reakcija protutijela inducira i izoliraju se stanice koje proizvode takva protutijela, te kultiviranje i uzgoj spomenutih protutijela.
21. Postupak za proizvodnju željenog protutijela, naznačen time, da obuhvaća izlaganje murinskog sisavca prema bilo kojem od zahtjeva 1 do 18, antigenu, tako da se reakcija protutijela inducira i izoliraju se molekule nukleinske kiseline koje kodiraju barem dio takvog protutijela, zatim umetanje spomenute molekule nukleinske kiseline ili njezine kopije ili njezinog derivata u kazetu ekspresije, te ekspresiju spomenutog protutijela u stanici domaćina.
