Claims (15)
1. Plasminogenaktivator des im menschlichen Gewebe vorkommenden Typs, der im wesentlichen frei von anderen Proteinen menschlichen Ursprungs
ist. .
2. Plasminogenaktivator des im menschlichen Gewebe vorkommenden Typs, unbegleitet von assoziierter, natürlicher Glykosylierung.
3. Plasminogenaktivator des im menschlichen Gewebe vorkommenden
Typs, gebildet durch rekombinante Wirtszellen.
4. Plasminogenaktivator des im menschlichen Gewebe vorkommenden
Typs in im wesentlichen reiner Form.
5. Plasminogenaktivator des im menschlichen Gewebe vorkommenden Typs nach Anspruch 1-4, enthaltend eine Sequenz eines Polypeptides, die
sich vom N-Terminus der normalerweise ersten Aminosäure des genannten, im menschlichen Gewebe vorkommenden Plasminogenaktivators aus erstreckt.
6. Eine DNA-Sequenz,enthaltend eine Sequenz, die für einen Plasmingenaktivator
des im menschlichen Gewebe vorkommenden Typs kodiert.
7. Ein replizierbarer Expressionsvektor, der fähig ist, in
einem transformanten Mikroorganismus oder einer transformanten Zellkultur eine DNA-Sequenz nach Anspruch 6 zu exprimieren.

8. Das Plasmid paRIPA' oder pt-PAtrp12.
9. Ein Mikroorganismus oder eine Zellkultur, der (die) mit dem
Vektor nach Anspruch 7 oder 8 transformiert ist.
10. Ein Mikroorganismus nach Anspruch 9, erhalten durch Transformierung
eines E.coli-Stammes.

11. Eine Zusammensetzung,umfassend eine therapeutisch wirksame
Menge eines Plasminogenaktivators des im menschlichen Gewebe vorkommenden
Typs nach Anspruch 1 - 5,vermischt mit einem pharmazeutisch verträglichen
Trägermittel.
12. Die Zusammensetzung nach Anspruch 11, geeignet für parenterale
Verabreichung.
■13. Die Verwendung von Plasminogenaktivator des im menschlichen
Gewebe vorkommenden Typs nach Anspruch 1 - 5 für die Behandlung von Gefäßerkrankungen
oder -zustände oder für die Herstellung von pharmazeutischen
Zusammensetzungen, die für solche Behandlungen nützlich sind.
14. Ein Verfahren, das die Expression von DNA-kodierendem
Plasminogenaktivator des im menschlichen Gewebe vorkommenden Typs in
einer rekombinanten Wirtszelle umfaßt.
15. Ein Verfahren zur Herstellung von menschlichem t-PA, welches Verfahren umfaßt:
a) Herstellung eines replizierbaren Expressionsvektors, der fähig ist,
die menschliches t-PA kodierende DNA-Sequenz in einer geeigneten
Wirtszelle zu exprimieren;
b) Transformierung einer Wirtszellenkultur, um eine rekombinante Wirtszelle zu erhalten;
c) Kultivieren der genannten rekombinanten Wirtszellen unter Bedingungen,
die die Expression der das genannte t-PA kodierenden DNA-Sequenz zur
Erzeugung von menschlichem t-PA ermöglichen; und die
d) Gewinnung des genannten menschlichen t-PA.
