Description
- Diese Erfindung betrifft eine Serie von Pyrazolo[4,3-d]pyrimidin-7-onen, die wirkungsstarke und selektive Inhibitoren von cyclischer Guanosin-3',5'-monophosphatphosphodiesterase (cGMP PDE) sind, und die in verschiedensten therapeutischen Bereichen, einschließlich der Behandlung verschiedener kardiovaskulärer Störungen, wie Angina, Hypertension, Herzversagen und Arteriosklerose, verwendet werden können.
- Die Verbindungen der Erfindung zeigen eher eine Selektivität für die Inhibierung von cGMP PDEs als cyclische Adenosin- 3',5'-monophosphatphosphodiesterasen (cAMP PDEs), und als Folge dieser selektiven PDE-Inhibierung werden die cGMP-Konzentrationen erhöht, was seinerseits zu einer vorteilhaften Blutplättchen-anti-aggregierenden, anti-vasospastischen und vasodilatatorischen Wirksamkeit sowie zur Potenzierung der Wirkungen des vom Endothelium stammenden Relaxationsfaktors (EDRF) und von Nitrovasodilatatoren führen kann. Daher sind die Verbindungen bei der Behandlung zahlreicher Störungen verwendbar, einschließlich stabiler, instabiler und varianter (Prinzmetal-) Angina, Hypertension, kongestivem Herzversagen, Arteriosklerose, Zuständen reduzierter Durchgängigkeit der Blutgefäße, z.B. postperkutanter transluminaler Koronarangioplastie (post-PTCA), peripheren Gefäßerkrankungen, Schlaganfall, Bronchitis, chronischem Asthma, allergischem Asthma, Heuschnupfen, Glaukom und durch Störungen der Darmmotilität charakterisierten Erkrankungen, z.B. Reizdarmsyndrom (IBS).
- Die EP-A-0 201 188 offenbart bestimmte Pyrazolo[4,3-d]pyrimidin-7-one als Adenosin-Rezeptor-Antagonisten und PDE- Inhibitoren, die bei der Behandlung kardiovaskulärer Störungen, wie Herzversagen oder Herzinsuffizienz, verwendet werden können. Diese Verbindungen sind jedoch weder besonders wirkungsstarke PDE-Inhibitoren, noch wird beansprucht, daß sie selektive cGMP PDE-Inhibitoren sind.
- Die Verbindungen der vorliegenden Erfindung sind Iene der Formel (I):
- worin R¹ H, C&sub1;-C&sub3;-Alkyl, C&sub3;-C&sub5;-Cycloalkyl oder C&sub1;-C&sub3;-Perfluoralkyl bedeutet; R² H, C&sub1;-C&sub6;-Alkyl, gegebenenfalls substituiert durch OH, C&sub1;-C&sub3;-Alkoxy oder C&sub3;-C&sub6;-Cycloalkyl oder C&sub1;-C&sub3;-Perfluoralkyl darstellt; R³ C&sub3;-C&sub6;-Alkyl, C&sub3;-C&sub6;-Alkenyl, C&sub3;-C&sub6;-Alkinyl, C&sub3;-C&sub7;-Cycloalkyl, C&sub1;-C&sub6;-Perfluoralkyl oder (C&sub3;-C&sub6;-Cycloalkyl)-C&sub1;-C&sub6;-alkyl ist; R&sup4; zusammen mit dem Stickstoffatom, an das es gebunden ist, eine Pyrrolidinyl-, Piperidino-, Morpholino- oder 4-N-(R&sup6;)-Piperazinyl-Gruppe bildet; R&sup5; H, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub3;-Alkoxy, NR&sup7;R&sup8; oder CONR&sup7;R&sup8; bedeutet; R&sup6; H, C&sub1;-C&sub6;-Alkyl, (C&sub1;-C&sub3;-Alkoxy)-C&sub2;-C&sub6;-alkyl, Hydroxy- C&sub2;-C&sub6;-alkyl, (R&sup7;R&sup8;N)-C&sub2;-C&sub6;-Alkyl, (R&sup7;R&sup8;NCO)-C&sub1;-C&sub6;-Alkyl, CONR&sup7;R&sup8;, CSNR&sup7;R&sup8; oder C(NH)NR&sup7;R&sup8; darstellt; und R&sup7; und R&sup8; jeweils unabhängig H, C&sub1;-C&sub4;-Alkyl, (C&sub1;-C&sub3;-Alkoxy)-C&sub2;-C&sub4;-alkyl oder Hydroxy-C&sub2;-C&sub4;-alkyl sind; und pharmazeutisch annehmbare Salze hievon.
- Wenn nichts anderes angegeben ist, können in der obigen Definition Alkyl- oder Perfluoralkyl-Gruppen mit drei oder mehr Kohlenstoffatomen gerad- oder verzweigtkettig sein. Außerdem können Alkenyl- oder Alkinyl-Gruppen mit vier oder mehr Kohlenstoffatomen oder Alkoxy-Gruppen mit drei Kohlenstoffatomen gerad- oder verzeigtkettig sein.
- Die Verbindungen der Formel (I) können ein oder mehrere asymmetrische Zentren enthalten, und daher können sie als Enantiomere oder Diastereoisomere vorliegen. Die Erfindung schließt sowohl Mischungen als auch getrennte einzelne Isomere ein.
- Die Verbindungen der Formel (I) können auch in tautomeren Formen vorliegen, und die Erfindung schließt sowohl Mischungen als auch getrennte einzelne Tautomere ein.
- Ebenfalls in der Erfindung eingeschlossen sind radioaktiv markierte Derviate von Verbindungen der Formel (I), die für biologische Untersuchungen geeignet sind.
- Die pharmazeutisch annehmbaren Salze der Verbindungen der Formel (I), die ein basisches Zentrum enthalten, sind Säureadditionssalze, welche mit pharmazeutisch annehmbaren Säuren gebildet werden. Beispiele schließen ein: Hydrochlorid-, Hydrobromid-, Sulfat- oder Bisulfat-, Phosphat- oder Hydrogenphosphat-, Acetat-, Citrat-, Fumarat-, Gluconat-, Lactat-, Maleat-, Succinat- und Tartratsalze. Verbindungen der Formel (I) können auch pharmazeutisch annehmbare Metallsalze, insbesondere Alkalimetallsalze, mit Basen bilden. Beispiele schließen die Natrium-und Kaliumsalze ein.
- Eine bevorzugte Gruppe von Verbindungen der Formel (I) ist jene, worin R¹ H, Methyl oder Ethyl bedeutet; R² C&sub1;-C&sub3;-Alkyl, gegebenenfalls substituiert durch OH oder Methoxy, darstellt; R³ C&sub2;-C&sub3;-Alkyl oder Allyl ist; R&sup4; zusammen mit dem Stickstoffatom, an das es gebunden ist, eine Piperidino- oder 4-N-(R&sup6;)-Piperazinyl-Gruppe bildet; R&sup5; H, NR&sup7;R&sup8; oder CONR&sup7;R&sup8; bedeutet; R&sup6; H, C&sub1;-C&sub3;-Alkyl, Hydroxy-C&sub2;-C&sub3;-alkyl, CONR&sup7;R&sup8;, CSNR&sup7;R&sup8; oder C(NH)NR&sup7;R&sup8; darstellt; und R&sup7; und R&sup8; jeweils unabhängig H oder Methyl sind.
- Eine besonders bevorzugte Gruppe von Verbindungen der Formel (I) ist jene, worin R¹ Methyl bedeutet; R² n-Propyl darstellt; R³ Ethyl, n-Propyl oder Allyl ist; R&sup4; zusammen mit dem Stickstoffatom, an das es gebunden ist, eine 4-N-(R&sup6;)-Piperazinyl-Gruppe bildet; R&sup5; H bedeutet; und R&sup6; H, C&sub1;-C&sub3;-Alkyl oder 2-Hydroxyethyl darstellt.
- Besonders bevorzugte einzelne Verbindungen der Erfindung schließen ein:
- 5-[2-Allyloxy-5-(4-methylpiperazinylsulfonyl)-phenyl]-1- methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
- 5-[2-Ethoxy-5-(piperazinylsulfonyl)-phenyl]-1-methyl-3- n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
- 5-[2-Ethoxy-5-(4-methylpiperazinylsulfonyl)-phenyl]-1- methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
- 5-[2-Ethoxy-5-[4-(2-propyl)-piperazinylsulfonyl]-phenyl]-1- methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;

- Die Verbindungen der allgemeinen Formel (I) können hergestellt werden durch das Umsetzen einer Verbindung der allgemeinen Formel (II):
- (worin R¹, R² und R³ wie vorstehend definiert sind, und Y ein Halogenatom, vorzugsweise ein Chloratom, bedeutet) mit einer Verbindung der allgemeinen Formel (III):
- worin R&sup4; und R&sup5; wie vorstehend definiert sind.
- Die Reaktion wird allgemein bei Raumtemperatur, vorzugsweise in Anwesenheit eines Lösungsmittels, beispielsweise eines Alkanols mit 1 bis 3 Kohlenstoffatomen, unter Verwendung eines Überschußes von (III) durchgeführt, um das Säure-Nebenprodukt (HY) auszuspülen.
- Verbindungen der allgemeinen Formel (II) können hergestellt werden aus Verbindungen der allgemeinen Formel (IV):
- (worin R¹, R² und R³ wie vorstehend definiert sind) unter Verwendung bekannter Verfahren zur Einführung einer SO&sub2;Y-Gruppe (worin Y wie vorstehend definiert ist) in einen aromatischen Ring, wenn Y beispielsweise ein Chloratom darstellt, durch Wirkung von Chlorsulfonsäure bei. oder nahe bei 0ºC.
- Wenn R³ eine Gruppe darstellt, die unter den Chlorsulfonylierungsbedingungen abspaltbar ist, z.B. Allyl, kann die genannte Gruppe im Endstadium der Synthese eingeführt werden. Daher wird das Phenol der allgemeinen Formel (IV), worin R³ die Bedeutung H hat, und R¹ und R² wie vorstehend definiert sind, die durch Pd-vermittelte Entschützung des O-Allyl-Analogons erhalten werden kann, wie in Beispiel 25 veranschaulicht, chlorsulfonyliert, wobei eine Verbindung der allgemeinen Formel (II), worin Y die Bedeutung Cl hat, R³ die Bedeutung H hat, und R¹ und R² wie vorstehend definiert sind, erhalten wird. Die letztere wird dann mit dem geeigneten Amin (III) umgesetzt, wobei eine Verbindung der allgemeinen Formel (I), worin R³ die Bedeutung H hat, und R¹, R², R&sup4; und R&sup5; wie vorstehend definiert sind, erhalten wird, die schließlich O-alkyliert wird, wobei eine Verbindung der allgemeinen Formel (I), worin R¹, R², R³, R&sup4; und R&sup5; wie für die Formel (I) definiert sind, erhalten wird. Die Alkylierung kann unter Standard-Bedingungen unter Verwendung des geeigneten Alkylhalogenids, z.B. Allylbromid, in Anwesenheit einer Base, wie Kaliumcarbonat, in einem geeigneten Lösungsmittel, z.B. 2-Butanon, bei Rückflußtemperatur der Reaktionsmischung bewirkt werden. Alternativ dazu kann die Alkylierung unter herkömmlichen Mitsunobu-Reaktionsbedingungen erzielt werden.
- Im Fall von anderen Verbindungen der Formel (IV), die mit den Chlorsulfonylierungs-Reaktionsbedingungen inkompatibel sein können, z.B. jenen, worin R² Hydroxy-C&sub1;-C&sub6;-alkyl bedeutet, kann die Hydroxy-Gruppe mit einer Acyl-Gruppe, wie Acetyl oder Benzoyl, geschützt werden. Die genannte Schutzgruppe wird anschließend im Endstadium der Synthese unter Standard-Basenhydrolysebedingungen entfernt, wobei Verbindungen der allgemeinen Formel (I), worin R² Hydroxy-C&sub1;-C&sub6;-alkyl bedeutet, und R¹, R³, R&sup4; und R&sup5; wie für die Formel (I) definiert sind, erhalten werden. Diese letzteren Verbindungen könne auch zufällig als Nebenprodukte durch Chlorsulfonylierung der entsprechenden Alkoxy- Analoga erhalten werden, d.h. Verbindungen der allgemeinen Formel (IV), worin R² (C&sub1;-C&sub3;-Alkoxy)-C&sub1;-C&sub5;-alkyl bedeutet, gefolgt vom Umsetzen des Rohprodukts mit dem erforderlichen Amin (III), wie in Beispiel 48 veranschaulicht.
- Verbindungen der allgemeinen Formel (IV) können hergestellt werden aus Verbindungen der allgemeinen Formel (V):
- (worin R¹, R² und R³ wie vorstehend definiert sind) unter Verwendung bekannter Cyclisierungsverfahren zur Bildung eines Pyrimidinon-Rings. Daher kann die Cyclisierung beispielsweise bewirkt werden durch die Behandlung von (V) mit einer Base, wie Natriumhydroxid oder Kaliumcarbonat, gegebenenfalls in Anwesenheit von Wasserstoffperoxyd, in einem Ethanol-Wasser-Medium bei Rückflußtemperatur während 2 bis 40 Stunden. Unter diesen Bedingungen kann auch das verwandte Nitril der allgemeinen Formel (VI), worin R¹, R² und R³ wie vorstehend definiert sind, als Vorläufer für (IV) verwendet werden.
- In einem alternativen Cyclisierungsverfahren können Verbindungen der allgemeinen Formel (IV) durch die Behandlung von (V) mit Polyphosphorsäure bei oder nahe bei 140ºC während 6 bis 18 Stunden erhalten werden.
- Verbindungen der allgemeinen Formeln (V) und (VI) können hergestellt werden aus Verbindungen der allgemeinen Formeln (VII) bzw. (VIII)
- (worin R¹ und R² wie vorstehend definiert sind) durch das Umsetzen mit einer Verbindung der allgemeinen Formel (IX):
- (worin R³ und Y wie vorstehend definiert sind).
- Die Reaktion wird allgemein unter Verwendung eines Überschusses von (IX) in Anwesenheit eines Überschusses eines aliphatischen tertiären Amins, wie Triethylamin, durchgeführt, das als Spülmittel für das Säure-Nebenprodukt (HY) wirkt, gegebenenfalls in Anwesenheit eines Katalysators, wie 4-Dimethylaminopyridin, in einem inerten Lösungsmittel, wie Dichlormethan, bei 0ºC bis 25ºC während 2 bis 6 Stunden.
- Die Amine der Formel (III), die Aminopyrazole der Formeln (VII) und (VIII) und die Acyihalogenide der Formel (IX) können, wenn sie nicht im Handel erhältlich sind, durch herkömmliche, in der Literatur beschriebene Syntheseverfahren aus leicht erhältlichen Ausgangsmaterialien unter Verwendung von Standard-Reagentien und -Reaktionsbedingungen erhalten werden.
- Bestimmte Verbindungen der allgemeinen Formel (I), worin R&sup4; zusammen mit dem Stickstoffatom, an das es gebunden ist, eine 4-N-(R&sup6;)-Piperazinyl-Gruppe bildet, und R&sup6; wie vorstehend definiert, jedoch nicht Wasserstoff ist, können direkt aus dem entsprechenden 4-N-unsubstituierten Piperazin-Analogon hergestellt werden, d.h. einer Verbindung der allgemeinen Formel (I), worin R&sup6; Wasserstoff bedeutet, unter Verwendung geeigneter Standard- Syntheseverfahren.
- Alle obigen Reaktionen sind zur Gänze herkömmlich, und geeignete Reagentien und Bedingungen für ihre Wirksamkeit können leicht durch Bezugnahme auf Standard-Nachschlagewerke und auf die nachstehend vorgesehenen Beispiele ermittelt werden. Ebenso sind Alternativen und Variationen zur Herstellung von allen durch die Formel (I) definierten Verbindungen für Fachleute ersichtlich.
- Die biologischen Wirksamkeiten der Verbindungen der vorliegenden Erfindung wurden durch die folgenden Testverfahren bestimmt.
- Die Affinitäten der Verbindungen für cGMP und cAMP PDEs werden durch die Bestimmung ihrer IC&sub5;&sub0;-Werte untersucht (der Inhibitor-Konzentration, die für eine 50 % Inhibierung der Enzymaktivität erforderlich ist). Die PDE-Enzyme werden aus Kaninchen-Blutplättchen und Ratten-Nieren im wesentlichen durch das Verfahren von W.J. Thompson et al. (Biochem., 1971, 10, 311) isoliert. Die Calcium/Calmodulin (Ca/CAM)-unabhängigen cGMP PDE- und die cGMP-inhibierten cAMP PDE-Enzyme werden aus Kaninchen- Blutplättchen erhalten, wohingegen aus den vier Haupt-PDE-Enzymen der Ratten-Niere die Ca/CAM-abhängige cGMP PDE (Fraktion I) isoliert wird. Die Tests werden unter Verwendung einer Modifikation des "Chargen"-Verfahrens von W.J. Thompson und M.M. Appleman (Biochem., 1979, 18, 5228) durchgeführt. Die Ergebnisse dieser Tests zeigen, daß die Verbindungen der vorliegenden Erfindung wirkungsstarke und selektive Inhibitoren beider cGMP PDEs sind.
- Diese wird durch die Bestimmung der Fähigkeit einer Verbindung ermittelt, die durch den Blutplättchenaktivierungsfaktor (PAF) induzierte Blutplättchen-Aggregation in vitro zu inhibieren, und die Blutplättchen-anti-Aggregationswirkung in vitro von Aktivatoren von Guanylatcyclase, wie Nitroprussid und EDRF, zu potenzieren. Gewaschene Blutplättchen werden im wesentlichen durch das Verfahren von J.F. Mustard et al. (Methods in Enzymol., 1989, 169, 3) hergestellt und die Aggregation wird unter Verwendung von Standard-Turbidimetrie-Techniken bestimmt, wie von G.V.R. Born, J. Physiol. (Lond), 1962, 162, 67P, beschrieben.
- Diese wird nach der intravenösen oder oralen Verabreichung einer Verbindung an spontan hypertensive Ratten ermittelt. Der Blutdruck wird über eine Kanüle aufgezeichnet, die in die Karotisarterie von Tieren, die entweder bei Bewußtsein oder narkotisiert sind, eingeführt wurde.
- Zur Verabreichung an Menschen bei der heilenden oder prophylaktischen Behandlung von Angina, Hypertension oder kongestivem Herzversagen liegen orale Dosierungen der Verbindungen allgemein im Bereich von 4 bis 800 mg pro Tag für einen durchschnittlichen erwachsenen Patienten (70 kg). Daher enthalten für einen typischen erwachsenen Patienten einzelne Tabletten oder Kapseln 2 bis 400 mg aktive Verbindung in einem geeigneten pharmazeutisch annehmbaren Vehikel oder Träger zur Verabreichung als einzelne oder mehrfache Dosen einmal oder mehrere Male pro Tag. Dosierungen für intravenöse, bukkale oder sublinguale Verabreichung liegen typischerweise im Bereich von 1 bis 400 mg pro Einzeldosis, je nach Bedarf. In der Praxis bestimmt der Arzt das tatsächliche Dosierungsschema, das für einen einzelnen Patienten am geeignetsten ist, und dieses variiert mit dem Alter, dem Gewicht und der Reaktion des betreffenden Patienten. Die obigen Dosierungen sind Beispiele des durchschnittlichen Falls, es kann jedoch Einzelfälle geben, in denen höhere oder niedrigere Dosierungsbereiche angezeigt sind, und diese sind im Umfang dieser Erfindung eingeschlossen.
- Zur Verwendung bei Menschen können die Verbindungen der Formel (I) allein verabreicht werden, sie werden jedoch allgemein in Mischung mit einem pharmazeutischen Träger verabreicht, der in Abhängigkeit von beabsichtigten Verabreichungsweg und der pharmazeutischen Standard-Praxis ausgewählt wird. Beispielsweise können sie oral, bukkal oder sublingual in Form von Tabletten, die Exzipienten, wie Stärke oder Lactose, enthalten, oder in Kapseln oder Ovula entweder allein oder in Mischung mit Exzipienten oder in Form von Elixieren oder Suspensionen, die Geschmacks- oder Farbstoffe enthalten, verabreicht werden. Die Verbindungen können auch parenteral, beispielsweise intravenös, intramuskulär, subkutan oder intrakoronar, injiziert werden. Zur parenteralen Verabreichung werden sie am besten in Form einer sterilen wässerigen Lösung verwendet, die andere Substanzen enthalten kann, beispielsweise ausreichend Salze oder Glucose, um die Lösung isotonisch mit Blut zu machen.
- Daher sieht die Erfindung eine pharmazeutische Zusammensetzung vor, welche eine Verbindung der Formel (I), oder ein pharmazeutisch annehmbares Salz hievon, zusammen mit einem pharmazeutisch annehmbaren Verbindungsmittel oder Träger umfaßt.
- Die Erfindung sieht auch eine Verbindung der Formel (I) vor, oder ein pharmazeutisch annehmbares Salz hievon, zur Verwendung in der Medizin, insbesondere zur Behandlung von Angina, Hypertension, Herzversagen oder Arteriosklerose.
- Weiters sieht die Erfindung die Verwendung einer Verbindung der Formel (I) vor, oder eines pharmazeutisch annehmbaren Salzes hievon, bei der Herstellung eines Medikaments zur Behandlung von stabiler, instabiler und varianter (Prinzmetal-) Angina, Hypertension, kongestivem Herzversagen, Arteriosklerose, Schlaganfall, peripheren Gefäßerkrankungen, Zuständen reduzierter Durchgängigkeit der Blutgefäße, z.B. post-PTCA, chronischem Asthma, Bronchitis, allergischem Asthma, Heuschnupfen, Glaukom oder durch Störungen der Darmmotilität charakterisierten Erkrankungen, z.B. IBS.
- Die Erfindung schließt auch neue Zwischenverbindungen ein, die hier geoffenbart sind, wie jene der Formeln (II) und (IV).
- Die Herstellung der Verbindungen der Erfindung wird nun detallierter unter Bezugnahme auf die folgenden Versuchsbeispiele veranschaulicht. Die Reinheit der Verbindungen wurde routinemäßig durch Dünnschichtchromatographie (DC) unter Verwendung von Merck Kieselgel 60 F&sub2;&sub5;&sub4;-Platten überwacht. ¹H-Nuklearmagnetresonanzspektren wurden unter Verwendung eines Nicolet QE-300-Spektrometers aufgezeichnet und stimmen in allen Fällen mit den vorgeschlagenen Strukturen überein.
- Einer Mischung von 24,1 g (0,132 mol) 3-n-Propylpyrazol-5- carbonsäureethylester (hergestellt durch das Verfahren von Chem. Pharm. Bull., 1984, 32, 1568) und 16,8 g (0,133 mol) Dimethylsulfat wurde 2,5 h lang auf 90ºC erhitzt. Die Mischung wurde in Dichlormethan gelöst und die Lösung mit Natriumcarbonat-Lösung gewaschen. Die organische Phase wurde abgetrennt, getrocknet (MgSO&sub4;) und im Vakuum eingedampft, wobei ein Feststoft erhalten wurde. Chromatographie auf 300 g Silicagel, wobei mit Dichlormethan eluiert wurde, ergab 20,4 g (79 %) des Produkts als farbloses Öl. Rt 0,8 (Kieselerde; Dichlormethan, Methanol, Essigsäure 80:20:1).
- 20,2 g (0,10 mol) 1-Methyl-3-n-propylpyrazol-5-carbonsäureethylester wurden in 50 ml (0,30 mol) wässeriger 6n Natriumhydroxid-Lösung suspendiert. Die Mischung wurde 2 h lang auf 80ºC erhitzt, mit 50 ml Wasser verdünnt und mit 25 ml konzentrierter Salzsäure angesäuert. Durch Filtration wurden 12,3 g (71 %) Carbonsäure als blaßbraune Kristalle erhalten, Fp. 150- 154ºC.
- Berechnet für C&sub8;H&sub1;&sub2;N&sub2;O&sub2;: C, 57,13; H, 7,19; N, 16,66 %
- Gefunden: C, 56,99; H, 7,25; N, 16,90 %.
- 12,1 g (0,072 mol) 1-Methyl-3-n-propylpyrazol-5-carbonsäure wurden portionsweise einer Mischung von 13 ml Oleum und 11 ml rauchender Salpetersäure zugesetzt, wobei die Temperatur unter 60ºC gehalten wurde. Nach dem Zusatz wurde die Mischung über Nacht auf 60ºC erhitzt und dann auf Raumtemperatur abgekühlt, bevor sie auf Eis gegossen wurde. Durch Filtration des Niederschlags wurden 11,5 g (75 %) Nitropyrazol als weißer Feststoff erhalten, Fp. 124-127ºC.
- Berechnet für C&sub8;H&sub1;&sub1;N&sub3;O&sub4;: C, 45,57; H, 5,20; N, 19,71 %
- Gefunden: C, 45,43; H, 5,22; N, 19,42 %.
- 11,3 g (0,053 mol) 1-Methyl-4-nitro-3-n-propylpyrazol-5- carbonsäure wurden zu 50 ml Thionylchlorid zugesetzt, und die erhaltene Mischung wurde 3 h lang am Rückfluß erhitzt. Dann wurde die Reaktionsmischung abgekühlt und überschüssiges Thionylchlorid im Vakuum abgedampft. Der ölige Rückstand wurde in 50 ml Aceton gelöst und die Lösung vorsichtig zu einer Mischung von 50 g Eis und 50 ml konzentrierter wässeriger Ammoniumhydroxid-Lösung zugesetzt. Der Niederschlag wurde durch Filtration gesammelt, wobei 8,77 g (78 %) Pyrazolcarboxamid als blaßgelber Feststoft erhalten wurden, Fp. 141-143ºC.
- Berechnet für C&sub8;H&sub1;&sub2;N&sub4;O&sub3;: C, 45,28; H, 5,70; N, 26,40 %
- Gefunden: C, 45,22; H, 5,71; N, 26,12 %.
- 3,45 g (16,2 mmol) 1-Methyl-4-nitro-3-n-propylpyrazol-5- carboxamid und 18,4 g (81 mmol) Zinndichloriddihydrat wurden in Ethanol suspendiert, und die Mischung wurde 2 h lang am Rückfluß erhitzt. Die erhaltene Lösung wurde auf Raumtemperatur abgekühlt, durch den Zusatz von wässeriger 2n Natriumhydroxid-Lösung auf pH 9 basisch gemacht und 3 x mit je 150 ml Dichlormethan extrahiert. Die organischen Extrakte wurde kombiniert, getrocknet (MgSO&sub4;) und im Vakuum eingedamptt. Durch Zerreiben des Rückstands mit Ether wurden 2,77 g (94 %) Aminopyrazol als cremetarbener Feststoff erhalten, Fp. 98-101ºC.
- Berechnet für C&sub8;H&sub1;&sub4;N&sub4;O: C, 52,73; H, 7,74; N, 30,75 %
- Gefunden: C, 52,84; H, 7,81; N, 30,38 %.
- Eine Lösung von 6,1 g (33,0 mmol) 2-Ethoxybenzoylchlorid in 50 ml Dichlormethan wurde zu einer gerührten Lösung von 3,0 g (16,4 mmol) 4-Amino-1-methyl-3-n-propylpyrazol-5-carboxamid, 0,02 g (0,164 mmol) 4-Dimethylaminopyridin und 3,34 g (33,0 mmol) Triethylamin in 50 ml Dichlormethan bei 0ºC zugesetzt. Die erhaltene Mischung wurde auf Raumtemperatur erwärmen gelassen und weitere 2 h lang gerührt. Das Lösungsmittel wurde im Vakuum abgedampft, der Rückstand in 250 ml einer 19:1 Mischung von Dichlormethan und Methanol gelöst und dann die Lösung mit 100 ml 1n Salzsäure gewaschen, getrocknet (MgSO&sub4;) und im Vakuum eingedamptt. Das Rohmaterial wurde auf 200 g Silicagel chromatographiert, wobei mit einer 97:3 Mischung von Dichlormethan und Methanol eluiert wurde, um einen rosa Feststoft zu ergeben; durch Kristallisation aus Ethylacetat-Hexan wurden 2,2 g (40 %) des Pyrazol-5-carboxamids als blaßrosa Feststoff erhalten, Fp. 153-155ºC.
- Berechnet für C&sub1;&sub7;H&sub2;&sub2;N&sub4;O&sub3;: C, 61,80; H, 6,71; N, 16,96 %
- Gefunden: C, 61,66; H, 6,77; N, 16,95 %.
- 223 g (0,676 mol) 4-(2-Ethoxybenzamido)-1-methyl-3- n-propylpyrazol-5-carboxamid wurden portionsweise zu einer Lösung von 54 g (1,35 mol) Natriumhydroxid und 224 ml 30 % Wasserstoffperoxid-Lösung in 2000 ml Wasser zugesetzt. 700 ml Ethanol wurden zugesetzt, und die erhaltene Mischung wurde 2,5 h lang am Rückfluß erhitzt, abgekühlt, dann im Vakuum eingedampft. Der erhaltene Feststoff wurde mit 380 ml 2n Salzsäure behandelt, unter externer Kühlung, und die Mischung wurde 1 x mit 700 ml und 3 x mit je 200 ml Dichlormethan extrahiert. Die kombinierten organischen Extrakte wurden aufeinanderfolgend 3 x mit je 400 ml gesättigter wässeriger Natriumcarbonat-Lösung und 300 ml Kochsalz lösung gewaschen, dann getrocknet (Na&sub2;SO&sub4;) und im Vakuum eingedampft.

- 10,0 g (32,1 mmol) 5-(2-Ethoxyphenyl)-1-methyl-3-n-propyl- 1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on wurden zu 20 ml Chlorsulfonsäure bei 0ºC unter einer Stickstoffatmosphäre portionsweise zugesetzt. Nach Rühren über Nacht wurde die Reaktionslösung vorsichtig zu 150 ml Eis-Wasser zugesetzt und die wässerige Mischung 4 x mit je 100 ml einer 9:1 Mischung von Dichlormethan und Methanol extrahiert. Die kombinierten Extrakte wurden getrocknet (Na&sub2;SO&sub4;) und im Vakuum eingedampft, wobei 12,8 g (97 %) des gewünschten Sulfonylchlorids als weißer Feststoff erhalten wurden, Fp. 179-181ºC.

- 703 mg (5,50 mmol) 4-Carbamoylpiperidin wurden zu einer gerührten Suspension von 750 mg (1,80 mmol) 5-(5-Chlorsulfonyl- 2-ethoxyphenyl)-1-methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on in 50 ml Ethanol bei Raumtemperatur zugesetzt. Die erhaltene Mischung wurde 4 Tage lang gerührt, bevor das Lösungsmittel im Vakuum abgedampft wurde. Der Rückstand wurde in 100 ml einer 9:1 Mischung von Dichlormethan und Methanol gelöst und die Lösung mit 100 ml gesättigter wässeriger Natriumcarbonat-Lösung gewaschen. Die wässerige Phase wurde 3 x mit je 100 ml Dichlormethan-Methanol-Mischungen weiter extrahiert, und alle organischen Fraktionen wurden kombiniert, getrocknet (MgSO&sub4;) und im Vakuum eingedampft, wobei ein Feststoff erhalten wurde. Kristallisation aus einer Mischung von Methanol- Dimethylformamid ergab 446 mg (49 %) des Titel-Sulfonamids als cremefarbenen Feststoff, Fp. 274-276ºC.

- Eine Mischung von 0,78 g (1,5 mmol) 5-[2-Ethoxy-5-(4- thiocarbamoylpiperazinylsulfonyl)-phenyl]-1-methyl-3-n-propyl- 1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on, 426 mg (3,0 mmol) Methyljodid und 20 ml Methanol wurde 2 h lang am Rückfluß gerührt, dann abkühlen gelassen. Der erhaltene weiße Feststoff wurde abfiltriert und aus Ethylacetat-Methanol kristallisiert, wobei 0,70 g (71 %) der Titelverbindung als farblose Kristalle erhalten wurden, Fp. 227-228ºC.

- 0,5 g (0,75 mmol) 5-{2-Ethoxy-5-[4-methylthioimidoyl)piperazinylsulfonyl]-phenyl}-1-methyl-3-n-propyl-1,6-dihydro-7H- pyrazolo[4,3-d]pyrimidin-7-onhydrojodid wurden zu einer 33 % Lösung von Methylamin in 20 ml Ethanol zugesetzt, und die Mischung wurde 18 h lang bei Raumtemperatur gerührt. Die Lösung wurde im Vakuum eingedampft und der Rückstand mit Ether zerrieben. Durch Chromatographie des erhaltenen Feststoffs auf 10 g Silicagel, wobei ein Methanol in Dichlormethan Elutionsgradient (0 bis 4 %) verwendet wurde, gefolgt vom Zerreiben des Rohprodukts mit Ether, wurde ein hellbraunes Pulver erhalten. Kristallisation aus Ethylacetat-Methanol ergab 112 mg (23 %) der Titelverbindung als farblose Kristalle, Fp. 253-255ºC.

- 0,34 g (0,99 mmol) 1-Methyl-4-(2-n-propoxybenzamido)-3-n- propylpyrazol-5-carboxamid wurden zu einer gerührten Mischung von 1,0 ml 30 % Wasserstoffperoxid-Lösung, 0,54 g (3,92 mmol) Kaliumcarbonat, 10 ml Wasser und 5 ml Ethanol zugesetzt. Die Mischung wurde 38 h lang am Rückfluß erhitzt und dann im Vakuum eingedampft. Der Rückstand wurde in 20 ml Wasser suspendiert, dann die Mischung mit 2n Salzsäure angesäuert und 3 x mit je 20 ml Dichlormethan extrahiert. Die Extrakte wurde kombiniert, getrocknet (Na&sub2;SO&sub4;) und im Vakuum eingedampft. Der erhaltene Rückstand wurde auf 6 g Silicagel chromatographiert, wobei ein Methanol in Dichlormethan Elutionsgradient (0,0 bis 1,0 %) verwendet wurde, um ein Öl zu ergeben, wobei durch dessen aufeinanderfolgendes Zerreiben mit Ether 0,19 g (59 %) des erforderlichen Produkts als weißer Feststoff erhalten wurden, Fp.111- 114ºC.

- Dieses Sulfonylchlorid wurde aus 5-(2-n-Propoxyphenyl)-1- methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 8 hergestellt und als weißer Feststoff (92 %) erhalten.

- Dieses Sulfonamid wurde aus N-(2-Hydroxyethyl)-piperazin und 5-(5-Chlorsulfonyl-2-n-propoxyphenyl)-1-methyl-3-n-propyl- 1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 9 hergestellt und als farblose Nadeln (66 %) erhalten, Fp. 158-159ºC.

- Eine Lösung von 3,93 g (0,02 mol) 2-Allyloxybenzoylchlorid in 20 ml Dichlormethan wurde einer gerührten partiellen Lösung von 3,64 g (0,02 mol) 4-Amino-1-Methyl-3-n-propylpyrazol-5- carboxamid in 50 ml Pyridin tropfenweise zugesetzt, und die erhaltene Mischung wurde bei Raumtemperatur über Nacht in einer trockenen Atmosphäre gerührt. Das Lösungsmittel wurde im Vakuum abgedampft und der Rückstand zwischen 50 ml Dichlormethan und 50 ml gesättigter wässeriger Natriumcarbonat-Lösung verteilt. Die organische Schicht wurde abgetrennt und die wässerige Schicht erschöpfend mit weiterem Dichlormethan extrahiert. Die kombinierten organischen Lösungen wurden 3 x mit je 30 ml 2m HCl, dann 1 x mit 30 ml Kochsalzlösung gewaschen und getrocknet (Na&sub2;SO&sub4;). Nach Filtration und Eindampfen des Filtrats im Vakuum wurde das Rohprodukt aus Ethylacetat kristalliert, wobei 4,525 g (66 %) der Titelverbindung erhalten wurden, Fp. 132-134ºC.

- Eine Mischung von 1,2 g (0,0035 mol) 4-(2-Allyloxybenzamido)-1-methyl-3-n-propylpyrazol-5-carboxamid, 0,70 g (0,018 mol) Natriumhydroxid, 34 ml Wasser und 8 ml Ethanol wurde 5 h lang am Rückfluß gehalten. Nach dem Abkühlen wurde die Lösung erschöpfend mit Ethylacetat extrahiert. Die kombinierten Extrakte wurden mit 30 ml Kochsalzlösung gewaschen, getrocknet (Na&sub2;SO&sub4;), filtriert, und das Lösungsmittel wurde im Vakuum abgedampft, wobei ein Rohprodukt erhalten wurde, das aus Ethylacetat/Hexan kristallisiert wurde, um 0,476 g (37 %) der Titelverbindung zu ergeben, Fp. 116-119ºC.

- Eine Mischung von 0,25 g (0,0008 mol) 5-(2-Allyloxyphenyl)- 1-methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7- on, 0,145 g (0,0015 mol) Phenol, 0,131 g (0,0015 mol) Piperidin und 0,046 g (0,00004 mol) Tetrakis-(triphenylphosphin)palladium(0) in 5 ml absolutem Ethanol wurde über Nacht unter Stickstoff am Rückfluß gehalten. Die Mischung wurde Abkühlen gelassen, das Lösungsmittel im Vakuum abgedampft und der Rückstand in 40 ml Ethylacetat gelöst. Diese Lösung wurde 3 x mit je 10 ml Wasser, 3 x mit je 10 ml 1 M HC1 und 1 x mit 10 ml Kochsalzlösung gewaschen. Nach Trocknen (Na&sub2;SO&sub4;) und Filtration wurde das Filtrat im Vakuum eingedampft, wobei das Rohprodukt erhalten wurde. 0,021 g (10 %) des Titel-Phenols wurden nach Zerreiben mit Diethylether und Kristallisation aus Ethylacetat/Pentan erhalten, Fp. 233-238ºC.

- 0,239 g (0,00084 mol) 5-(2-Hydroxyphenyl)-1-methyl-3- n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on wurden zu 3 ml gerührter Chlorsulfonsäure, gekühlt auf 0ºC, unter einer Stickstoffatmosphäre portionsweise zugesetzt, und die erhaltene tiefrote Lösung wurde 18 h lang bei Raumtemperatur gerührt. Dann wurde die Reaktionmischung vorsichtig zu gerührtem Eis/Wasser tropfenweise zugesetzt, wobei ein brauner Feststoff erhalten wurde. Die letztere Mischung wurde 3 x mit je 30 ml Dichlormethan extrahiert, die kombinierten Extrakte wurden getrocknet (Na&sub2;SO&sub4;) und filtriert, und das Filtrat wurde im Vakuum eingedampft, wobei 0,24 g (75 %) eines braunes Feststoffs erhalten wurden, der im nächsten Schritt ohne weitere Reinigung verwendet wurde; Rt 0,3 (Kieselerde; Dichlorinethan; Methanol; 95:5).
- Eine Lösung von 0,235 g (0,0006 mol) 5-(5-Chlorsulfonyl-2- hydroxyphenyl)-1-methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on und 0,5 ml (0,0045 mol) N-Methylpiperazin in 40 ml Ethanol wurde 18 h lang bei Raumtemperatur gerührt. Die Lösung wurde im Vakuum eingedampft und der Rückstand zwischen 40 ml Ethylacetat und 40 ml Wasser verteilt. Der feine Niederschlag wurde abfiltriert, mit Wasser, dann Ethylacetat gewaschen und aus Ethylacetat/DMF kristallisiert, wobei 0,260 g (49 %) der Titelverbindung als cremetarbenes Pulver erhalten wurden, Fp. 283-284ºC.
- Berechnet für C&sub2;&sub0;H&sub2;&sub6;N&sub6;O&sub4;S: C, 53,80; H, 5,87; N, 18,82 %
- Gefunden: C, 53,53; H, 5,89; N, 18,40 %.
- 0,02 ml (0,00023 mol) Allylbromid wurden zu einer gerührten Suspension von 0,103 g (0,00023 mol) 5-[2-Hydroxy-5-(4-methylpiperazinylsulfonyl)-phenyl]-1-methyl-3-n-propyl-1,6-dihydro-7H- pyrazolo[4,3-d]pyrimidin-7-on und 0,032 g (0,00023 mol) Kaliumcarbonat in 10 ml 2-Butanon zugesetzt, und die Mischung wurde 8 h lang am Rückfluß erhitzt. Nach dem Abkühlen wurde die Reaktionsmischung im Vakuum eingedampft und der Rückstand in 20 ml Wasser suspendiert. Die wässerige Suspension wurde 3 x mit je 20 ml Ethylacetat extrahiert, die kombinierten Extrakte wurden getrocknet (Na&sub2;SO&sub4;) und nach Filtration im Vakuum eingedampft, wobei ein Öl erhalten wurde. Säulenchromatographie auf 2 g Silicagel, wobei ein Methanol in Dichlormethan Elutionsgradient (0 bis 3 %) verwendet wurde, gefolgt vom Eindampfen der geeigneten Fraktionen im Vakuum, ergab einen Halbfeststoff, der in Aceton gelöst wurde; durch Eindampfen der Lösung in Vakuum wurden 0,011 g (10 %) der Titelverbindung erhalten, Fp. 151- 153ºC, Rt 0,5 (Kieselerde; Dichlormethan, Methanol; 95:5), m/e 487 (M&spplus;+1).
- Dieses Amid wurde aus 4-Amino-1,3-dimethylpyrazol-5- carboxamid (erzeugt durch die Methode von J. Med. Chem.,1987, 30, 91), gemäß dem Verfahren von Beispiel 6 hergestellt und als weißer Feststoff (81 %) erhalten, Fp. 178-181ºC.
- Berechnet für C&sub1;&sub5;H&sub1;&sub8;N&sub4;O&sub3;: C, 59,59; H, 6,00; N, 18,53 %
- Gefunden: C, 59,89; H, 6,05; N, 18,44 %.
- 1,6 g (5,29 mmol) 4-(2-Ethoxybenzamido)-1,3-dimethylpyrazol-5-carboxamid wurden zu 50 g Polyphosphorsäure zugesetzt, und die Mischung wurde 6 h lang auf 140ºC erhitzt. Die Lösung wurde abgekühlt, in 100 ml Eis-Wasser gegossen, dann die Mischung mit 10 % wässeriger Natriumhydroxid-Lösung basisch gemacht und 3 x mit je 100 ml Dichlormethan extrahiert. Die organischen Extrakte wurden kombiniert, getrocknet (MgSO&sub4;) und im Vakuum eingedampft. Der Rückstand wurde auf Silicagel chromatographiert, wobei mit einer 97:3 Mischung von Dichlormethan und Methanol eluiert wurde. Durch Kristallisation des Rohprodukts aus wässerigem Ethanol wurde die Titelverbindung als farbloser Feststoff erhalten, Fp. 201-204ºC.

- Dieses Sulfonylchlorid wurde aus 5-(2-Ethoxyphenyl)-1,3- dimethyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 8 hergestellt und in einer quantitativen Ausbeute als weißer Feststoff erhalten. Rf 0,3 (Kieselerde: ther). Es wurde ohne weitere Reinigung verwendet.
- Die folgenden Verbindungen wurden aus 5-(5-Chlorsulfonyl-2- ethoxyphenyl)-1,3-dimethyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on und dem geeigneten Amin gemäß dem Verfahren von Beispiel 9 hergestellt. % Ausbeute Analyse % (Theorie in
- 3-n-Propylpyrazol-5-carbonsäure (hergestellt durch die Methode von Chem. Pharm. Bull. 1984, 32, 1568) wurde gemäß dem Verfahren von Beispiel 3 nitriert, wobei die Titelverbindung (75 %) als farbloser Feststoff erhalten wurde, Fp. 169-173ºC.
- Berechnet für C&sub7;H&sub9;N&sub3;O&sub4;: C, 42,21; H, 4,55; N, 21,10 %
- Gefunden: C, 42,35; H, 4,56; N, 21,07 %.
- Eine Mischung von 7,8 g (39,2 mmol) 4-Nitro-3-n-propylpyrazol-5-carbonsäure und 35 ml Thionylchlorid wurde 3 h lang am Rückfluß erhitzt. Das Lösungsmittel wurde im Vakuum abgedampft und der feste Rückstand zu 40 ml wässeriger Ammoniumhydroxid- Lösung bei 0ºC portionsweise zugesetzt. Dann wurde die Mischung mit 60 ml Wasser verdünnt und 3 x mit je 100 ml einer 9:1 Mischung von Dichlormethan und Methanol extrahiert. Die organischen Fraktionen wurden kombiniert, getrocknet (MgSO&sub4;) und im Vakuum eingedampft, und der Rückstand wurde aus Ethanol kristallisiert, wobei 1,0 g (13 %) des Carboxamids als farbloser Feststoff erhalten wurden, Fp. 202-206ºC.
- Berechnet für C&sub7;H&sub1;&sub0;N&sub4;O&sub3;: C, 42,42; H, 5,09; N, 28,27 %
- Gefunden: C, 42,35; H, 5,01; N, 28,38 %.
- Eine Lösung von 198 mg (1,0 mmol) 4-Nitro-3-n-propylpyrazol-5-carboxamid in 5 ml Methanol wurde tropfenweise zu einer Mischung von 113 mg (2,97 mmol) Natriumborhydrid, 5 mg 10 % Palladium-auf-Kohle und 3 ml Wasser zugesetzt. Die Mischung wurde 3 h lang bei Raumtemperatur gerührt, filtriert und das Lösungsmittel im Vakuum abgedampft. Durch Kristallisation des Rückstands aus Ethylacetat-Methanol wurden 61 mg (36 %) der Titelverbindung als cremefarbener Feststoff erhalten, Fp. 196- 201ºC. Rt 0,4 (Kieselerde; Dichlormethan, Methanol, Ammoniumhydroxid; 90:10:1).

- Das Titel-Sulfonylchlorid wurde aus 5-(2-Ethoxyphenyl)-3- n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 8 hergestellt und als weißer Feststoff (58 %) erhalten. Rt 0,25 (Kieselerde; Ether).
- Es wurde ohne weitere Reinigung verwendet.
- Das Titel-Sulfonamid wurde aus 5-(5-Chlorsulfonyl-2- ethoxyphenyl)-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 9 hergestellt und als weißer Feststoff (70 %) erhalten, Fp. 236-239ºC.

- 10,7 g (60,0 mmol) N-Bromsuccinimid wurden zu einer Lösung von 8,78 g (50,0 mmol) 5-Chlor-1,3-dimethyl-4-nitropyrazol in 100 ml Tetrachlorkohlenstoff zugesetzt, und die Lösung wurde am Rückfluß erhitzt, wobei sie mit sichtbarem Licht (150 W Wolframlampe) 3 Tage lang bestrahlt wurde. Während der gesamten Reaktion wurden in Intervallen 6 x Mengen von je 50 mg Benzoylperoxid zugesetzt. Das Lösungsmittel wurde im Vakuum abgedampft und der Rückstand auf Silicagel chromatographiert, wobei mit einer 1:1 Mischung von Dichlormethan und Hexan eluiert wurde, um 8,0 g (63 %) des Bromids als cremefarbenen Feststoff zu ergeben, Fp. 80-82ºC.

- Gefunden: C, 23,95; H 2,05; N, 16,31 %.
- Eine Lösung von 5,0 g (19,6 mmol) 3-Brommethyl-5-chlor-1- methyl-4-nitropyrazol in 50 ml Methanol wurde mit 5,75 g (33,8 mmol) Silbernitrat behandelt und die Mischung 2 h lang am Rückfluß erhitzt. Die abgekühlte Reaktionsmischung wurde filtriert und das Filtrat im Vakuum eingedampft. Der Rückstand wurde zwischen 100 ml Ethylacetat und 50 ml Wasser verteilt, und die wässerige Phase wurde mit einer weiteren Menge von 50 ml Ethylacetat extrahiert. Die organischen Extrakte wurden kombiniert, getrocknet (MgSO&sub4;) und im Vakuum eingedampft. Durch Chromatographie auf Silicagel, wobei mit einer 97:3 Mischung von Dichlormethan und Methanol eluiert wurde, wurden 1,6 g (40 %) des Titel-Pyrazols als weißer Feststoff erhalten, Fp. 59-63ºC.
- Berechnet für C&sub6;H&sub8;ClN&sub3;O&sub3;: C, 35,05; H, 3,92; N, 20,44 %
- Gefunden: C, 34,65; H, 3,83; N, 20,05 %.
- Eine Lösung von 205 mg (1,0 mmol) 5-Chlor-3-methoxymethyl- 1-methyl-4-nitropyrazol, 130 mg (2,0 mmol) Kaliumcyanid und 10 mg 18-Crown-6 in 2 ml Acetonitril wurde über Nacht am Rückfluß erhitzt. Das Lösungsmittel wurde im Vakuum abgedampft und der Rückstand zwischen 20 ml Ethylacetat und 20 ml Wasser verteilt. Die organische Phase wurde abgetrennt, getrocknet (MgSO&sub4;) und im Vakuum eingedampft, dann wurde der Rückstand auf Silicagel chromatographiert, wobei mit einer 1:1 Mischung von Ethylacetat und Pentan eluiert wurde. Durch Zerreiben des Rohprodukts mit Ether wurden 38 mg (19 %) eines gelben Feststoffs erhalten, Fp. 48-50ºC.
- Berechnet für C&sub7;H&sub8;N&sub4;O&sub3;: C, 42,86; H, 4,11; N1 28,56 %
- Gefunden: C, 42,89; H, 4,15; N, 28,78 %.
- Die Titelverbindung wurde aus 5-Cyano-3-methoxymethyl-1- methyl-4-nitropyrazol gemäß dem Verfahren von Beispiel 5 hergestellt und als cremefarbener Feststoff (68 %) erhalten, Fp. 82- 84ºC.
- Berechnet für C&sub7;H&sub1;&sub0;N&sub4;O: C, 50,59; H, 6,07; N, 33,72 %
- Gefunden: C, 50,81; H, 6,13; N, 33,94 %.
- Die Titelverbindung wurde aus 4-Amino-5-cyano-3-methoxymethyl-1-methylpyrazol gemäß dem Verfahren von Beispiel 6 hergestellt und als cremefarbener Feststoff (61 %) erhalten, Fp. 103- 105ºC.
- Berechnet für C&sub1;&sub6;H&sub1;&sub8;N&sub4;O&sub3;: C, 61,13; H, 5,77; N, 17,83 %
- Gefunden: C, 61,21; H 5,98; N, 17,80 %.
- Die Titelverbindung wurde aus 5-Cyano-4-(2-ethoxybenzamido)-3-methoxymethyl-1-methylpyrazol gemäß dem Verfahren von Beispiel 7 über die in situ Erzeugung des 5-primären Amid- Derivats hergestellt und als weißer Feststoff (98 %) erhalten, Fp. 160-161ºC.
- Berechnet für C&sub1;&sub6;H&sub1;&sub8;N&sub4;O&sub3;: C, 61,13; H, 5,77; N, 17,83 %
- Gefunden: C, 61,35; H, 5,75; N, 17,98 %.
- 470 mg (1,50 mmol) 5-(2-Ethoxyphenyl)-3-methoxymethyl-1- methyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on wurden in 3 ml Chlorsulfonsäure bei 0ºC gelöst. Die Lösung wurde 2 h lang bei Raumtemperatur gerührt, dann vorsichtig zu 50 ml Eiswasser zugesetzt. Die erhaltene Lösung wurde mit gesättigter Natriumcarbonatlösung neutralisiert, dann 2 x mit je 50 ml einer 20:1 Mischung von Dichlormethan und Methanol extrahiert. Die kombinierten organischen Extrakte wurden in Vakuum eingedampft, und der Rückstand wurde in 5 ml Ethanol gelöst und die Lösung mit 450 mg (4,5 mmol) N-Methypiperazin behandelt. Nach 1 h bei Raumtemperatur wurde das Lösungsmittel im Vakuum abgedampft und der Rückstand auf Silicagel chromatographiert, wobei mit einer Mischung von Dichlormethan, Methanol und wässeriger Ammoniumhydroxyd-Lösung (90:10:1 V) eluiert wurde. Durch Zerreiben des Rohprodukts mit Ethylacetat wurden 49 mg (7 %) der Titelverbindung als weißer Feststoff erhalten, Fp. 198-199ºC.
- Berechnet für C&sub2;&sub1;H&sub2;&sub8;N&sub6;O&sub5;S: C, 52,93; H, 5,92; N, 17,64 %
- Gefunden: C, 52,94; H, 6,04; N, 17,67 %.
- Nach Chromatographie und Kristallisation wurden auch aus einer Mischung von Ethylacetat und Methanol 51 mg (7 %) 3-Hydroxymethyl-1-methyl-5-[5-(4-methylpiperazinylsulfonyl)-2- ethoxyphenyl]-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7 on als weißer Feststoff isoliert, Fp. 209-210ºC.
- Berechnet für C&sub2;&sub0;H&sub2;&sub6;N&sub6;O&sub5;S: C, 51,94; H, 5,57; N, 18,17 %
- Gefunden: C, 51,94; H 5,77; N, 18,05 %.
- Dieses Pyrazol wurde aus 3-n-Propylpyrazol-5-carbonsäureethylester und Diethylsulfat gemäß dem in Beispiel 1 beschriebenen Verfahren hergestellt und als farbloses Öl (72 %) erhalten. Rt 0,5 (Kieselerde; Ethylacetat, Hexan; 1:1).
- Diese Carbonsäure wurde aus 1-Ethyl-3-n-propylpyrazol-5- carbonsäureethylester gemäß dem in Beispiel 2 beschriebenen Verfahren hergestellt und als blaßbrauner Feststoff (89 %) erhalten, Fp. 73º-77ºC.
- Berechnet für C&sub9;H&sub1;&sub4;N&sub2;O&sub2;: C, 59,32; H, 7,74; N, 15,37 %
- Gefunden: C, 58,62; H, 7,69; N, 15,23 %.
- Die Titelverbindung wurde aus 1-Ethyl-3-n-propylpyrazol-5- carbonsäure gemäß dem in Beispiel 3 beschriebenen Verfahren hergestellt und als farbloser Feststoff (96 %) erhalten, Fp. 120- 123ºC.
- Berechnet für C&sub9;H&sub1;&sub3;N&sub3;O&sub4;: C, 47,57; H, 5,77; N, 18,49 %
- Gefunden: C, 47,61; H 5,81; N, 18,54 %.
- Das Titel-Amid wurde aus 1-Ethyl-4-nitro-3-n-propylpyrazol- 5-carbonsäure gemäß dem in Beispiel 4 beschriebenen Verfahren hergestellt und als cremefarbener Feststoff (86 %) erhalten, Fp. 119-120ºC.
- Berechnet für C&sub9;H&sub1;&sub4;N&sub4;O&sub3;: C, 47,78; H, 6,24; N, 24,77 %
- Gefunden: C, 47,38; H, 6,18; N, 24,34 %.
- Die Titelverbindung wurde aus 1-Ethyl-4-nitro-3-n-propylpyrazol-5-carboxamid durch das Beispiel 5 beschriebene Verfahren hergestellt und als cremefarbener Feststoff (100 %) erhalten, Fp. 93-97ºC.
- Berechnet für C&sub9;H&sub1;&sub6;N&sub4;O: C, 55,08; H, 8,22; N, 28,55 %
- Gefunden: C, 55,17; H 8,34; N, 28,93 %.
- Das Titel-Amid wurde aus 4-Amino-1-ethyl-3-n-propylpyrazol- 5-carboxamid und 2-Ethoxybenzoylchlorid gemäß dem in Beispiel 6 beschriebenen Verfahren hergestellt und farbloser Feststoff (73 %) erhalten, Fp. 139-141ºC.
- Berechnet für C&sub1;&sub8;H&sub2;&sub4;N&sub4;O&sub3;: C, 62,77; H, 7,02 N, 16,27 %
- Gefunden: C, 63,03; H, 7,15; N, 16,50 %.
- Die Titelverbindung wurde aus 4-(2-Ethoxybenzamido)-1- ethyl-3-n-propylpyrazolo-5-carboxamid gemäß dem Verfahren von Beispiel 7 hergestellt und als farbloser Feststoff (46 %) erhalten, Fp. 112-114ºC.
- Berechnet für C&sub1;&sub8;H&sub2;&sub2;N&sub4;O&sub2;: C, 66,23; H, 6,79; N, 17,17 %
- Gefunden: C, 66,59; H, 6,85; N, 17,26 %.
- Die Titelverbindung wurde aus 5-(2-Ethoxyphenyl)-1-ethyl-3- n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on gemäß dem Verfahren von Beispiel 8 hergestellt und als Methylenchlorid- Solvat (86 %) erhalten, Fp. 170-172ºC.
- Berechnet für C&sub1;&sub8;H&sub2;&sub1;ClN&sub4;O&sub4;S; 1/6 CH&sub2;Cl&sub2;: C, 49,70; H, 4,90; N, 12,77 %
- Gefunden: C, 49,82; H, 4,84; N, 12,77 %.
- Das Titel-Sulfonamid wurde aus 5-(5-Chlorsulfonyl-2- ethoxyphenyl)-1-ethyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on und N-Methylpiperazin gemäß dem Verfahren von Beispiel 9 hergestellt und als farbloser Feststoff (43 %) erhalten, Fp. 160-162ºC. Rt 0,35 (Kieselerde; Dichlormethan, Methanol; 9:1).
- Berechnet für C&sub2;&sub3;H&sub3;&sub2;N&sub6;O&sub4;S: C, 56,54; H, 6,60; N, 17,20 %
- Gefunden: C, 57,24; H, 6,17; N, 16,83 %.
- Das Titel-Sulfonamid wurde aus 5-(5-Chlorsulfonyl-2- ethoxyphenyl)-1-ethyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on und N-(2-Hydroxyethyl)-piperazin gemäß dem Verfahren von Beispiel 9 hergestellt und als farbloser Feststoff (88 %) erhalten, Fp. 191-193ºC.
- Berechnet für C&sub2;&sub4;H&sub3;&sub4;N&sub6;O&sub5;S: C, 55,58; H, 6,61; N, 16,20 %
- Gefunden: C, 55,74; H, 6,55; N, 15,78 %.
Claims (8)
1. Verbindung der Formel:
worin R¹ H, C&sub1;-C&sub3;-Alkyl, C&sub3;-C&sub5;-Cycloalkyl oder
C&sub1;-C&sub3;-Perfluoralkyl bedeutet; R² H, C&sub1;-C&sub6;-Alkyl, gegebenenfalls substituiert
durch OH, C&sub1;-C&sub3;-Alkoxy oder C&sub3;-C&sub6;-Cycloalkyl oder
C&sub1;-C&sub3;-Perfluoralkyl darstellt; R³ C&sub1;-C&sub6;-Alkyl, C&sub3;-C&sub6;-Alkenyl,
C&sub3;-C&sub6;-Alkinyl, C&sub3;-C&sub7;-Cycloalkyl, C&sub1;-C&sub6;-Perfluoralkyl oder
(C&sub3;-C&sub6;-Cycloalkyl)-C&sub1;-C&sub6;-alkyl ist; R&sup4; zusammen mit dem
Stickstoffatom, an das es gebunden ist, eine Pyrrolidinyl-,
Piperidino-, Morpholino- oder 4-N-(R&sup6;)-Piperazinyl-Gruppe
bildet; R&sup5; H, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub3;-Alkoxy, NR&sup7;R&sup8; oder CONR&sup7;R&sup8;
bedeutet; R&sup6; H, C&sub1;-C&sub6;-Alkyl, (C&sub1;-C&sub3;-Alkoxy)-C&sub2;-C&sub6;-alkyl, Hydroxy-
C&sub2;-C&sub6;-alkyl, (R&sup7;R&sup8;N)-C&sub2;-C&sub6;-Alkyl, (R&sup7;R&sup8;NCO)-C&sub1;-C&sub6;-Alkyl,
CONR&sup7;R&sup8;, CSNR&sup7;R&sup8; oder C(NH)NR&sup7;R&sup8; darstellt; und R&sup7; und R&sup8;
jeweils unabhängig H, C&sub1;-C&sub4;-Alkyl, (C&sub1;-C&sub3;-Alkoxy)-C&sub2;-C&sub4;-alkyl
oder Hydroxy-C&sub2;-C&sub4;-alkyl sind; und pharmazeutisch annehmbare
Salze hievon.
2. Verbindung nach Anspruch 1, worin R¹ H, Methyl oder Ethyl
bedeutet; R² C&sub1;-C&sub3;-Alkyl, gegebenenfalls substituiert durch OH
oder Methoxy, darstellt; R³ C&sub2;-C&sub3;-Alkyl oder Allyl ist; R&sup4;
zusammen mit dem Stickstoffatom, an das es gebunden ist, eine
Piperidino- oder 4-N-(R&sup6;)-Piperazinyl-Gruppe bildet; R&sup5; H,
NR&sup7;R&sup8; oder CONR&sup7;R&sup8; bedeutet; R&sup6; H, C&sub1;-C&sub3;-Alkyl, Hydroxy-
C&sub2;-C&sub3;-alkyl, CONR&sup7;R&sup8;, CSNR&sup7;R&sup8; oder C(NH)NR&sup7;R&sup8; darstellt; und R&sup7;
und R&sup8; jeweils unabhängig H oder Methyl sind.
3. Verbindung nach Anspruch 2, worin R¹ Methyl bedeutet; R²
n-Propyl darstellt; R³ Ethyl, n-Propyl oder Allyl ist; R&sup4;
zusammen mit dem Stickstoffatom, an das es gebunden ist, eine
4-N-(R&sup6;)-Piperazinyl-Gruppe bildet; R&sup5; H bedeutet; und R&sup6; H,
C&sub1;-C&sub3;-Alkyl oder 2-Hydroxyethyl darstellt.
4. Verbindung nach Anspruch 3, wobei die Verbindung ausgewählt
ist aus:
5-[2-Allyloxy-5-(4-methylpiperazinylsulfonyl)-phenyl]-1-
methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
5-[2-Ethoxy-5-(piperazinylsulfonyl)-phenyl]-1-methyl-3-
n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
5-[2-Ethoxy-5-(4-methylpiperazinylsulfonyl)-phenyl]-1-
methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
5-[2-Ethoxy-5-[4-(2-propyl)-piperazinylsulfonyl]-phenyl]-1-
methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
5-[2-Ethoxy-5-[4-(2-hydroxyethyl)-piperazinylsulfonyl]phenyl]-1-methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on;
1-Methyl-5-[5-(piperazinylsulfonyl]-2-n-propoxyphenyl]-3-
n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on; und
5-[5-[4-(2-Hydroxyethyl)-piperazinylsulfonyl]-2-n-propoxyphenyl]-1-methyl-3-n-propyl-1,6-dihydro-7H-pyrazolo[4,3-d]pyrimidin-7-on,
und pharmazeutisch annehmbaren Salzen hievon.
5. Pharmazeutische Zusammensetzung, welche eine Verbindung der
Formel (I) oder ein pharmazeutisch annehmbares Salz hievon nach
einem der Ansprüche 1 bis 4 zusammen mit einem pharmazeutisch
annehmbaren Verdünnungsmittel oder Träger umfaßt.
6. Verbindung der Formel (I) oder ein pharmazeutisch
annehmbares Salz hievon nach einem der Ansprüche 1 bis 4 zur
Verwendung in der Medizin, insbesondere zur Behandlung von Angina,
Hypertonie, Herzversagen oder Arteriosklerose.
7. Verwendung einer Verbindung der Formel (I) oder eines
pharmazeutisch annehmbarern Salzes hievon nach einem der Ansprüche 1
bis 4 bei der Herstellung eines Medikaments, insbesondere zur
Behandlung von Angina, Hypertonie, Herzversagen,
Arteriosklerose, Schlaganfall, peripheren Gefäßerkrankungen, Zuständen
reduzierter Durchgängigkeit der Blutgefäße, chronischem Asthma,
Bronchitis, allergischem Asthma, Heuschnupfen, Glaukom oder
durch Störungen der Darmmotilität charakterisierten
Erkrankungen.
8. Verbindung der Formel:
worin R¹ und R² wie in Anspruch 1 definiert sind, R³ wie in
Anspruch 1 definiert ist, und auch H bedeutet, und Y Chlor, Brom
oder Fluor darstellt.
