Die Erfindung betrifft N-Oxide von Pyridylcarbonsäureestern,
ein Verfahren zur Herstellung dieser Verbindungen und pharmazeutische Mittel, die diese Verbindungen
enthalten.
Bekanntlich ist die Atherosklerose durch Anreicherung
von Lipiden in der Aorta, den coronaren, cerebralen und
]O peripheren Arterien bedingt, woraus ein erhöhtes Risiko
für Thrombosen oder Arterien verstopfung resultiert.
Je nach Art des erhöhten Plasma1ipoproteinspiegels
steht dabei die Erhöhung des Cholesterin- oder des Triglyceridspiegels im Vordergrund. Dabei gelten b e reits
Ch öl esterinspiegel von 200 - 300 mg/10 0 ml Serum
und Triglyceridspiegel von 150 - 200 mg/100 ml Serum
als überhöht.
Die beiden bekanntesten bisher zur Behandlung von Hyperlipidämien
verwendeten Wirkstoffe sind der Ethylester . von 2-(p-Chlorphenoxy)-isobuttersäure , bekannt als
Clofibrat, und deren Salze, und Nikotinsäure. Diese
Verbindungen wirken in verschiedener Weise auf die Serumlipide ein. Während bei Versuchstieren Dosen von
100" 500 mg/kg KG neben einer geringen Senkung des Gehalts an freien Fettsäuren vor allem eine Senkung des
Cholesterinspiegels bewirken, führt die Verabreichung von
3-Pyridylcarbinol oder Nikotinsäure und deren Salze bereits
bei niedriger Dosierung zwischen 10 und 1ü0mg/kg
2U einer starken Senkung des Fettsäurespiegels. Keine
der beiden Substanzen übt jedoch einen signifikanten
Einfluß auf die Senkung des Triglyceridspiegels aus.
Daneben ist die Nikotinsäure aufgrund ihrer unangenehmen bekannten Nebeneffekte (Blutandrang zum Kopf, Kopf-
3^ schmerzen, Übelkeit, Erbrechen) nur bedingt anwendbar,
so daß die Therapie oftmals vorzeitig abgesetzt werden muß.

Außerdem ist bekannt, daß zwar durch Clofibrat die
Triglycerid- und die Prae-ß-Lipoprote inwerte bis zu 50 %
unter den Ausgangswert gesenkt werden, dies jedoch für die Chol esterinwerte in dem Ausmaß nicht zutrifft. Nikotinsäure
und ihre Derivate hingegen wirken vorwiegend bei erhöhten Cholesterinwerten und erhöhten Fettsaurewerten,
senken hingegen erst sekundär über eine Hemmung der Gewebslipolyse die endogene Resynthese der Triglyceride
(vgl. Verhandlungen der Dtsch. Gesellschaft für Inn.
Medizin, 82. Kongreß, gehalten zu Wiesbaden vom 25. - 29.4.1976, Teil I, J.F. Bergmann Verlag, München).
Die genannten Wirkstoffe haben also die Eigenschaft, die Bl utspiegelwerte von nur einer der Lipidkomponenten,
beispielsweise die Triglyceride, signifikant zu senken,
während die anderen Lipidkomponenten nicht oder nur
zum geringen Teil therapeutisch beeinflußt werden. Dies
gelingt nur durch eine höhere Dosierung.
.^der Lipidforschunq Nach neuesten Ergebnissen auf dem GebietlToll ein brauenbarer
Wirkstoff nicht nur die Triglycerid- und ChI öl esterinwerte
reduzieren, sondern auch einen Anstieg der HDL-Lipoproteine(atherogener
Schutzfaktor) und einen entsprechenden Abfall der LDL- und VLDL-Lipoproteine
während der Behandlung bewirken. Außerdem sollte keine Peroxisomenproliferation auftreten und demzufolge
das Lebergewicht konstant bleiben.
Die DE-OS 30 09 099 beschreibt Nicotinsäureester, die
zwar einen Teil obiger Anforderungen erfüllen, nämlich ein-e 1 ipi dsenkende Wirkung auf beide Li pi dkomponenten
ausüben, die aber hinsichtlich der übrigen Anforderungen zu wünschen übrig lassen.
Aufgabe der Erfindung wares deshalb, weitere Verbindun-

düngen aufzufinden, die bei gleicher oder geringerer
Dosierung außer der reinen Lipidsenkung auch die anderen Faktoren, mit denen der Atherosklerose entgegengewirkt
werden kann, günstig beeinflussen.
überraschenderweise wurde nun gefunden, daß N-Oxide
von Pyridylcarbonsäureestern eine starke Wirkung sowohl
be"i erhöhten Triglycerid- als auch Cholesterin werten
im Serum aufweisen und gleichzeitig einen starken Anstieg
der va soprotekti ven F.DI- und eine Senkung der atherogenen LDL- und VLDL-Lipoproteinwerte bewirken.
Die erfindungsgemäßen Verbindungen verursachen auch keine Peroxisomenproliferation,so daß die Lebergewichte
konstant bleiben
Gegenstand der Erfindung sind deshalb Pyridylcarbon·
säureester-N-oxide der Formel:

C1-C5-AUyI, C5-C12-Cycloalkyl , 1-Adamantyl
oder Cyclohexylacetyl bedeutet und y ein Wasserstoffatom,
geradkettiges oder verzweigtes C.-C5-Al kyl , 2-Ethyl
benzofuranoyl-3 , oder C5-C12-CyCloalkyl bedeutet,
oder für 2-I2-(4-Chlorphenoxy)-2-methyl-propanoyloxy}-ethyl
steht.

Der -C-OR Rest kann in 2,3 oder ^-Stellung stehen, vorzugsweise
steht er in 3-Stellung. Der Phenylkern des
]O Restes R kann mono- oder disubstituiert sein, wobei
der Substituent X oder Y vorzugsweise ein Cyclohexyl- t
Cyclopentyl- oder 2-Ethylbenzofuranoyl-3- Rest ist. Beispiele für geeignete
Alkylsubstituenten X oder Y sind der Methyl-, Ethyl-, Propyl-, Jsopropyl-, Butyl-, tert-Butyl-, sek-Butyl- oder Pentylrest.
Das Verfahren zur Herstellung der erfindungsgemäßen
Verbindungen ist dadurch gekennzeichnet, daß man in
an sich bekannter Weise einen Pyridyl carbonsäureester, der nicht am Stickstoff oxidiert ist, mit
Peroxysäuren oder mit Eisessig/Wesserstoffperoxyd
N - ο χ i d i e r t.
Die Verbindungen gemäß der Erfindung können zu pharmazeutisehen
Mitteln verarbeitet werden, die neben dem
Wirkstoff auch einen Träger oder ein Verdünnungsmittel
enthalten. Sie können sowohl oral als auch parenteral verabreicht werden.

festepräparate zur oralen Verabreichung sind Kapseln,
Tabletten, Pillen, Pulver, Granulate. Jn solchen festen Zubereitungen ist der Wirkstoff mit wenigstens einem
inerten Verbindungsmittel wie Rohrzucker, Lactose oder
Stärke vermischt. Auch weitere Substanzen wie Gleit-35
mittel oder Puffer können anwesend sein. Die Tabletten
und Pillen können mit einem darmlöslichen überzug
versehen werden.

Verbindungen in Γ or ir. von Emulsionen, Lüsunaen , Suspc-nsio-
^1 η en, die üblicherweise verwendete; inerte Verdünnungsmittel
wie Wasser enthalten, zur Anwendung kommen. Außerdem können solche flüssigen Mittel auch Netz-, Ernulgier-
und Suspendiermittel, sowie Süß-, Geschmacks- und Geruchsstoffe
enthalten.

Die uosierung der Wirkstoffe in den Mitteln kann je nach Art
der Verabreichung und Behandlungsdauer variieren. Im allgemeinen
enthält ein zur Behandlung der Hyperlipidämie geeignetes
Mittel eine hyperlipidämisch wirksame Dosis min-
destens einer erfindungsgemaDen Verbindung in Kombination
mit einem pharmazeutisch akzeptablen Träger oder Verdünnungsmittel.

Methode
Weibliche CFY-Ratten (5o-6o g) wurden von Hacking und Churchill
Ltd. bezogen. Nach ihrer Ankunft wurden die Tiere gewogen und ir. Maschendraht-Einzelkäfigen einer 4-tägigen Akklimatisierung
unterzogen.
Die Raumtemperatur betrug 230C - 20C. Die relative Luftfeuchtigkeit und die Raumtemperatur wurden täglich gemessen. Die Beleuchtungsdauer betrug 12 Stunden (von 7.oo bis 19.oo Uhr) pro
24 Stunden; die restlichen 12 Stunden verbrachten die Tiere in Dunkelheit.
Während der gesamten Studie"erhielten die Ratten Wasser ad libitum,
Während der 4-tägigen Akklimatisierungzeit erhielten die Tiere
Spratt's pulverisierte Labortierdiät Nr. 2 mit niedrigem Fettgehalt ad libitum. Während der folgenden 14 Tage erhielten alle
Tiere Spratt's pulverisierte Labortierdiät Nr. 2 ad libitum, der
2Gewichtsprozent Cholesterin and" 1 Gewichtsprozent Cholsäure hinzugefügt worden waren. Während dieser Zeit wurde der Futterverbrauch pro Tier dreimal wöchentl i c-h bestimmt.
Zwischen 0.6.00 und 07.30 am Morgen des 14. Tages wurde jeder Ratte unter leichter Ätheranästhesie eine Blutprobe aus dem
Retro-orbita!-Plexus entnommen. Bei allen Blutproben wurden
Ge&amtserumchol-esteri-nf ti-BL-Chol esteri n, B-Li pop rote i ne sowie
die Triglyceridkonzentrationen bestimmt.
Basierend auf den Gesamtcholesterinspiegeln wurden die Tiere den
Behandlungsgruppen zugeordnet, wobei die Verteilung der Gesamtcholesterinspiegel
in allen Gruppen ähnlich und der initiale durchschnittliche Cholesterinspiegel in allen Gruppen annähernd
'gleichwar.
Danach schloß sich eine 14tätige Behandlungsperiode an, in der alle Tiere
die hypercholesterinämische Diät erhielten und mit Ausnahme der Kontrollen den Tieren mit dem Futter die angemessene Konzentration der Testsubstanz
verabreicht wurde (0,1 %).
Während der 14-tägigen Verabreichung der Substanz wurde
der Futterverbrauch pro Tier täglich, die Körpergewichte der
Tiere jed-och wöchentlich' bestinir.it.
Am letzten Tag der Studie wurde den Ratten zur gleichen Zeit wie am 14. Tag vor der Behandlung eine Blutprobe aus
dem Retro-orbita!-Plexus entnommen.
Die Blutproben wurden ebenfalls auf Gesamtserumcholesterin,
HDL-Cholesterin, ß-Lipoproteine und Triglyceridspiegel untersucht.
Die Tiere wurden anschließend getötet, die Lebern entnommen und gewogen.
Im Vergleich zu den Kontrolltieren konnten hochsignifikante
positive Veränderungen bei allen Parametern mit Ausnahme der Lebergewichte festgestellt werden (vgl. Tabelle 1).
Diese veränderten sich nicht.

Allgemeine Arbeitsvorschrift zur Herstellung von
Pyridylcarbonsäureester-N-oxiden _
Zu einer eisgekühlten, magnetisch gerührten Lösung des
betreffenden Nikotinates (20 mMol) in 50-100 ml Dichlormethan
gibt man portionsweise innerhalb von 15 Minuten 4,22 g (22 mMol) 90 %-ige 3-Chlorperbenzoesäure und rührt
30 Min. bei 0 C nach. Die weiteren Reaktionsbedingungen sind der Tabelle 2 zu entnehmen (DC-Kontrolle: SiO2;
CHCl3/MeOH 9:1).
Zur Aufarbeitung verdünnt man das Reaktionsgemisch mit Dichlormethan auf ca. 250 ml, wäscht die organische
Phase mit gesättigter Na2SO,-Lösung (2 χ 100 ml) ,
gesättigter NaHCC^-Lösung (3x100 ml) und zuletzt mit gesättigter
NaCl-Lösung (2x100 ml). Nach Trocknen über Na3SO4 wird das Solvens im Vakuum abgezogen und der
verbleibende Rückstand, gegebenenfalls unter Zuhilfenahme von Aktivkohle, umkristallisiert (s. Tabelle 2).
Auf diese Weise wurden folgende Verbindungen hergestellt:
