Abstract
Description
糖尿病性腎症は、糖尿病に起因する腎の細小血管障害であり、糸球体基底膜肥厚とメサンギウム領域拡大が基本的病理変化である。近年、糖尿病患者の増加とともに糖尿病性腎症で末期腎不全となり、透析治療に至る患者が増加している。高血糖状態の持続によりできるAGEは、1)血管透過性の亢進、2)タンパクやリポタンパクの沈着促進、3)一酸化窒素(NO)不活性化、4)細胞外基質産生促進、などを介して腎症発症に関与すると考えられている(例えば、非特許文献2参照)。
例えば、腎症、網膜症などの糖尿病合併症などを引き起こす成因となるAGEの生成を合成医薬(合成化合物)により阻害するものが開示されているが(特許文献1および2参照)、AGE産生抑制剤のような合成医薬はまだ開発段階であり、また、このような合成医薬には予期せぬ副作用が伴うことが多い。
しかしながら、このような植物抽出物中に存在するAGE阻害活性成分の阻害活性は、一般的に合成医薬(合成化合物)に比べ極めて低く、また活性成分の抽出物中での存在量も極めて低い。
さらに、桜抽出物の新規な生理活性として、AGEの1種であるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して効果的な抑制作用を有することを知見し、本発明を完成するに至ったものである。
他の目的は、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)によるヒト線維芽細胞のアポトーシスを抑制するアポトーシス抑制剤を提供することにある。
他の目的は、線維芽細胞におけるコラーゲン格子形成促進剤を提供することにある。
他の目的は、線維芽細胞内でAGE生成を効果的に阻害しうるAGE産生抑制剤を提供することにある。
1.本発明のAGE産生抑制剤は、桜の抽出物および/またはその処理物を有効成分として含有する。
2.本発明のAGE産生抑制剤は、桜の花の抽出物および/またはその処理物を有効成分として含有する。
3.本発明のAGE産生抑制剤は、桜の葉の抽出物および/またはその処理物を有効成分として含有する。

6.本発明のAGE産生抑制剤は、ケルセチン 3-O-β-D-グルコピラノシド(Quercetin 3-O-β-D-glucopyranoside)を有効成分として含有する。
8.本発明のアポトーシス抑制剤は、桜の花の抽出物および/またはその処理物を有効成分として含有するアポトーシス抑制剤であって、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して抑制作用を有する。
9.本発明のアポトーシス抑制剤は、桜の葉の抽出物および/またはその処理物を有効成分として含有するアポトーシス抑制剤であって、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して抑制作用を有する。

11.本発明のアポトーシス抑制剤は、1-O-(E)-カフェオイル-β-D-グルコピラノシド(1-O-(E)-Caffeoyl-β-D-glucopyranoside)を有効成分として含有するアポトーシス抑制剤であって、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して抑制作用を有する。
12.本発明のアポトーシス抑制剤は、ケルセチン 3-O-β-D-グルコピラノシド(Quercetin 3-O-β-D-glucopyranoside)および/またはケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(Quercetin 3-O-(6''-malony)-β-D-glucopyranoside)を有効成分として含有するアポトーシス抑制剤であって、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して抑制作用を有する。
14.本発明のヒト線維芽細胞のコラーゲン格子形成促進剤は、桜の花の抽出物および/またはその処理物を有効成分として含有する。

また、上記25~33に係る本発明によれば、CML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスを効果的に抑制し、ヒト線維芽細胞のコラーゲン格子形成を効果的に促進し、線維芽細胞内でAGEの生成を効果的に抑制することができる。これにより、皮膚老化の防止・改善に役立つ食品、医薬品、化粧品等の素材を提供することができる。
本発明のAGE産生抑制剤、線維芽細胞のアポトーシス抑制剤、ヒト線維芽細胞のコラーゲン格子形成促進剤、線維芽細胞内AGE産生抑制剤、および桜抽出物(以下、これらを纏めて示すときは、「AGE産生抑制剤等」という。)は、その生理活性の有効成分として、1-O-(E)-カフェオイル-β-D-グルコピラノシド(1-O-(E)-Caffeoyl-β-D-glucopyranoside)、1-O-(E)-クマロイル-β-D-グルコピラノシド(1-O-(E)-Coumaroyl-β-D-glucopyranoside)、1-O-(E)-シンナモイル-β-D-グルコピラノシド(1-O-(E)-Cinnamoyl-β-D-glucopyranoside)、ケンフェロール 3-O-β-D-グルコピラノシド(Kaempferol 3-O-β-D-glucopyranoside)、ケルセチン 3-O-β-D-グルコピラノシド(Quercetin 3-O-β-D-glucopyranoside)、ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド(Kaempferol 3-O-(6''-malony)-β-D-glucopyranoside)、ケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(Quercetin 3-O-(6''-malony)-β-D-glucopyranoside)から選ばれる少なくとも1種を含有する。
また、アルコール濃度が30wt%以上の場合、抽出温度は、0~95℃、好ましくは0~60℃程度で行うとよい。なお、含水エタノール抽出は、有効成分の含有率を向上させるため、種々の濃度で繰り返すとよい。
なお、精製方法としては、例えば、活性炭処理、樹脂吸着処理、シリカゲル処理、イオン交換樹脂、液-液向流分配、膜分離等の方法が挙げられる。
(a)桜の花および/または葉を20~50wt%の含水エタノールで抽出する工程
(b)前記工程(a)で得られた抽出物を吸着剤に吸着させた後、水で溶出して水溶出部を取り除く工程
(c)前記工程(b)に続いて前記抽出物を水溶性溶媒で溶出し、この溶出液を濃縮する工程を含む製造方法を採用するとよい。
このような製法によれば、桜の花および葉から上記化学式(1)~(7)のポリフェノールを高濃度に含む抽出物を効率よく得ることができる。
前記工程(c)水溶性溶媒としては、炭素数1~5の低級アルコールを用いることができ、特にメタノールおよび/またはエタノールを用いることが好ましい。
また、更にAGEの産生を抑制することにより特に線維芽細胞内のAGEの産生を抑制することにより、皮膚内のコラーゲン部分同士の架橋化を改善し、皮膚の硬化を防止することができる。
更に、AGEが生成した後における線維芽細胞のアポトーシスを抑制するため、皮膚の弾力性を保持することができ、更にシワやくすみの発生を予防することができる。
また、AGEが生成した後におけるコラーゲン架橋化を改善するため、皮膚の硬化を防止することができる。
桜抽出物を、例えば、油脂、エタノール、グリセリンあるいはこれらの混合物に溶解して液状にし、飲料に添加するか、固形食品に添加することが可能である。必要に応じてアラビアガム、デキストリン等のバインダーと混合して粉末状あるいは顆粒状にし、飲料に添加するか固形食品に添加することも可能である。

これらの粉体の形状(球状、棒状、針状、板状、不定形状、燐片状、紡錘状等)および粒子径に特に制限はない。なおこれらの粉体は、従来公知の表面処理、例えばフッ素化合物処理、シリコーン処理、シリコーン樹脂処理、ペンダント処理、シランカップリング剤処理、チタンカップリング剤処理、油剤処理、N-アシル化リジン処理、ポリアクリル酸処理、金属セッケン処理、アミノ酸処理、レシチン処理、無機化合物処理、プラズマ処理、メカノケミカル処理等によって事前に表面処理されていてもいなくても構わない。
アニオン性界向活性剤:脂肪酸セッケン、α-アシルスルホン酸塩、アルキルスルホン酸塩、アルキルアリルスルホン酸塩、アルキルナフタレンスルホン酸塩、アルキル硫酸塩、POEアルキルエーテル硫酸塩、アルキルアミド硫酸塩、アルキルリン酸塩、POEアルキルリン酸塩、アルキルアミドリン酸塩、アルキロイルアルキルタウリン塩、N-アシルアミノ酸塩、POEアルキルエーテルカルボン酸塩、アルキルスルホコハク酸塩、アルキルスルホ酢酸ナトリウム、アシル化加水分解コラーゲンペプチド塩、パーフルオロアルキルリン酸エステル等が挙げられる。

また、セルロースまたはその誘導体、ケラチン及びコラーゲンまたはその誘導体、アルギン酸カルシウム、プルラン、寒天、ゼラチン、タマリンド種子多糖類、キサンタンガム、カラギーナン、ハイメトキシルペクチン、ローメトキシルペクチン、グアーガム、アラビアゴム、結晶セルロース、アラビノガラクタン、カラヤガム、トラガカントガム、アルギン酸、アルブミン、カゼイン、カードラン、ジェランガム、デキストラン等の天然由来高分子化合物も好適に用いることができる。

亜硫酸水素ナトリウム、亜硫酸ナトリウム、エリソルビン酸、エリソルビン酸ナトリウム、チオジプロピオン酸ジラウリル、トコフェロール、トリルビグアナイド、ノルジヒドログアヤレチン酸、パラヒドロキシアニソール、ブチルヒドロキシアニソール、ジブチルヒドロキシトルエン、ステアリン酸アスコルビル、パルミチン酸アスコルビル、没食子酸オクチル、没食子酸プロピル、カロチノイド、フラボノイド、タンニン、リグナン、サポニン、リンゴエキスやチョウジエキスなとの酸化防止効果の認められる植物エキス等が挙げられる。
精製水、エタノール、低級アルコール、エーテル類、LPG(液化石油ガス)、フルオロカーボン、N-メチルピロリドン、フルオロアルコール、揮発性直鎮状シリコーン、次世代フロン等が挙げられる。
桜[ヤエザクラ (カンザン), Prunus lannesiana Wils. cv. Sekiyama]の花部を30%(W/W)含水エタノールで1時間抽出(60℃)を行った。その後、ろ過、濃縮を行い、桜の花抽出物(以下、桜花エキスともいう。)を得た(収率5wt%)。同様にして、桜の葉抽出物(以下、桜葉エキスともいう。)を得た(収率4wt%)。これらの桜花エキスおよび桜葉エキスをぞれぞれ実施例1および実施例2とした。
桜花の含有成分の探索の方法を図1に示す。即ち、実施例1の桜花エキス (90.10 g)をHP-20(三菱化学株製) カラムクロマトグラフィー(column chromatography)に付し, 水(H2O)溶出部(62.16 g, 68.99%)、メタノール(MeOH)溶出部(28.21g, 31.31%), およびアセトン(Acetone)溶出部(0.60g,0.67%)を得た。
得られたメタノール(MeOH)溶出部 (26.00 g)を順相シリカゲルクロマトグラフィー(silica-gel column chromatography)、 逆相ODSカラムクロマトグラフィー(column chromatography)およびHPLCを用いて繰り返し分離精製し、3種の既知フェニルプロパノイド配糖体;1-O-(E)-カフェオイル-β-D-グルコピラノシド(8.73g,9.6%)、1-O-(E)-クマロイル-β-D-グルコピラノシド(22.1mg,0.0245%)、1-O-(E)-シンナモイル-β-D-グルコピラノシド(21.0mg,0.0233%)、および4種の既知フラボノイド配糖体;ケンフェロール 3-O-β-D-グルコピラノシド (32.6 mg, 0.0361%)、ケルセチン 3-O-β-D-グルコピラノシド(86.0 mg, 0.0954%)、ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド(55.3mg, 0.0613%)、ケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(66.2 mg, 0.0734%)を単離した(実施例3~5および実施例6~9)。
これらの既知化合物については, 文献値の1H-NMR, 13C-NMR, MSスペクトルデータおよび旋光度の比較により同定した。 なお、図1に示されるこれら単離成分の収率(w/w)は実施例1の桜花エキスからの単離収率である。
[試験例1:AGE産生抑制作用]
桜花エキスおよびその含有成分並びに桜葉エキスのAGE形成に及ぼす作用について下記の方法で試験を行った。
D-グルコース(10%)とウシ血清アルブミン(fraction 5, 1%) を含有するリン酸緩衝液(pH: 7.4, 900 mL)に,桜花エキス(実施例1)およびその含有成分、並びに桜葉エキス(実施例2)を下記表1に示された濃度で含有するサンプル溶液(100 mL)を添加し,60℃で2日間静置した。反応液を蛍光光度が500付近になるように精製水で希釈し,蛍光光度(測定波長:370 nm, 励起波長:440 nm)を測定した。なお,エキスは精製水で希釈,成分はDMSOに溶解後,最終DMSO濃度が1%になるようにリン酸緩衝液で希釈して使用した。その結果を下記表1に示す。表1において各値は3例の平均値と標準誤差で示した。アスタリスクはサンプル未処理群とのDunnettの多重比較検定による有意差*:p<0.05、**:p<0.01を表す。
表1によれば、桜花エキス(実施例1)および桜葉エキス(実施例2)のいずれも濃度100~300(μg/mL)でAGEの生成を抑制していることが確認された。
また、表1に示すように、桜の花の主ポリフェノールである1-O-(E)-カフェオイル-β-D-グルコピラノシド(実施例3)は、高濃度(300μg/mL)で30%の阻害活性を示した。類縁化合物で水酸基数の少ない1-O-(E)-クマロイル-β-D-グルコピラノシド(実施例4)および1-O-(E)-シンナモイル-β-D-グルコピラノシド(実施例5)の阻害活性は減弱した。これに対し、フラボノイド配糖体の活性は総じて強く、ケルセチン 3-O-β-D-グルコピラノシド(実施例7)およびケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(実施例9)の活性は、対応するケンフェロール(kaempferol)をアグリコンとする化合物の2倍以上(IC50で)強い活性を示した。
皮膚に生じる 主AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)は皮膚細胞に障害を与えることが知られている。試験例2-1では、CML化コラーゲンよるヒト線維芽細胞のアポトーシスに対する桜花エキス(実施例1)の作用を下記の方法により検討した。
ウシ皮膚由来コラーゲン(50 mg)に1mM HCl(25 mL)を加え、37℃で必要に応じて揺り動かしながら2時間インキュベートして溶解した。これにPBS(pH7.4, 25 mL)、シアノ水素化ほう素ナトリウム(sodium cyanoborohydride)1.42 gおよびグリオキシル酸(glyoxylic acid)0.9 gを加えて混合し,37℃で24時間反応させた。反応溶液を透析用セル(透析分子量10000)に入れ,水で1日,PBSで6日間透析した。透析物をフリーズドライすることで、CML化コラーゲン21mgを得た。
正常二倍体線維芽細胞(40歳女性,正常皮膚由来,PDL20)を10%ウシ胎仔血清(FCS),ペニシリン(100 units/mL)およびストレプトマイシン(100 mg/mL)含有培地中で増殖・前培養を行い実験に使用した。
線維芽細胞(7×104 cells)を0.5%FCS、ペニシリン(100 units/mL)およびストレプトマイシン(100 mg/mL)含有D-MEM培地にサスペンドし、96-wellプレートに播種(100 mL/well)した。24時間培養後、培地に溶解したCML化コラーゲン溶液(2 mg/mL)およびサンプル溶液を添加(13 mL)し、24時間培養を行った。Normal群には、CML化コラーゲンの代わりにコラーゲンを添加した。
細胞生存率はMTTアッセイにより測定した。その結果を表2に示す。また、その際、鏡検による観察も行った。その結果として線維芽細胞(100倍鏡検)のホルマザン生成の様子を図2に示す。アポトーシスはカスパーゼ(caspase)3/7活性を蛍光法で測定することで調べた。その結果を下記表3に示す。なお、表2および3において各値は6例の平均値と標準誤差で示した。アスタリスクはDunnettの多重比較検定による有意差*:p<0.05、**:p<0.01、fはFisher's PLSD検定によるp<0.05を表す。
(1)細胞生存率
上記表2によれば、Control群の吸光度はnormal群より低く、CML化コラーゲンによる生存率の低下が観察された。鏡検像においても図2に示されるように、細胞内へのホルマザン蓄積の抑制が認められた。この様子からCML化コラーゲンによる線維芽細胞のアポトーシスが惹起されているものと推測された。これに対し、桜花エキス(実施例1)1000 mg/mLを添加した群では,ホルマザン生成の有意な増加が認められた。
上記表3は、表3に示されるようにControl群ではnormal群と比較して,カスパーゼ3/7活性の増加が見られ,CML化コラーゲンによるアポトーシスが惹起されていることが確認された。これに対し,桜花エキス(100 mg/mL以上)を添加した群ではカスパーゼ3/7活性の低下が見られた。300 mg/mL以上ではnormal群より活性が低かったことから桜花エキスはカスパーゼ 3/7の酵素活性や酵素発現を抑制している可能性が示唆された。
試験例2-2では、桜花エキス(実施例1)、桜葉エキス(実施例2)、および桜花エキス(実施例1)に含まれる7種の含有成分:1-O-(E)-カフェオイル-β-D-グルコピラノシド、1-O-(E)-クマロイル-β-D-グルコピラノシド、1-O-(E)-シンナモイル-β-D-グルコピラノシド、ケンフェロール 3-O-β-D-グルコピラノシド、ケルセチン 3-O-β-D-グルコピラノシド、ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド、ケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(実施例3~9)について、試験例2-1と同様の方法でCML化コラーゲンによるヒト線維芽細胞のアポトーシスに対する作用を検証した。なお、この試験例2-2では、正常二倍体線維芽細胞としてPDL22-32の細胞を使用した。比較対照として糖化抑制の医薬品である塩酸アミノグアニジンについても同様な方法で試験を行った。結果を下記表4に示す。表4において各値は5例の平均値と標準誤差で示した。アスタリスクはCML化コラーゲンを添加したサンプル未処理群とのDunnettの多重比較検定による有意差*:p<0.05、**:p<0.01を表す。
表4に示されるように、桜花エキス(実施例1)10~100 mg/mLでカスパーゼ3/7活性の低下が見られた。この結果より,桜花エキス(実施例1)がCML化コラーゲンによるアポトーシスを抑制していることが示唆された。含有成分では試験例1に示したAGE産生抑制試験で強い活性を示したケルセチン 3-O-β-D-グルコピラノシド(実施例7)の抑制作用が最も強く、類似化合物のケンフェロール 3-O-β-D-グルコピラノシド(実施例6)やケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(実施例9)にも強い活性が認められた。しかしながら,ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド(実施例8)は比較的活性が弱いものであった。一方,ケイヒ酸誘導体では含有量が最も高い1-O-(E)-カフェオイル-β-D-グルコピラノシド(実施例3)が特に強い抑制作用を示した。
このように桜花エキス(実施例1)、桜葉エキス(実施例2)及びその含有成分がCML化コラーゲンによる線維芽細胞のアポトーシスを抑制する理由については、AGE産生抑制作用が関与するものと考えられるが、上記の結果から推察すると、これらの有効成分がAGEであるCML化コラーゲンを分解するブレーカーの機能を果たすと見ることもできる。この点で、本発明のAGE産生抑制剤は、AGE分解・除去剤としても適用しうる。
線維芽細胞をコラーゲン溶液存在下で培養すると、コラーゲン格子の形成が認められる。一方で、この系に糖化刺激作用のある中間糖化物(グリオキサール)で糖化した線維芽細胞を添加すると、コラーゲン格子の形成が抑制されることが知られている。試験例3では、糖化したヒト線維芽細胞のコラーゲン格子形成に対する桜花エキスの作用を下記の方法により検討した。
新生児由来NB1RGB(継代数22)をD-MEM完全培地にサスペンドし(1.86×104 cells/mL),14 cmディッシュに15 mLずつ播種した。24時間培養後に,各種濃度のサンプルおよびグリオキサール(Glyoxal)を200 mMになるように添加し,5日間培養を行った。
ウシ皮膚由来コラーゲン(25 mg)を0.1%酢酸(8.3 mL)に溶解し,10倍濃縮Hanks液(1.66 L)を添加した。これに0.1%酢酸(3.5 mL)をさらに添加し,1M NaOHを滴下して溶液を中和した。この溶液をろ過滅菌して,コラーゲン液とした。
回収した線維芽細胞(5.4×104 cell)をFCS(50μm)にサスペンドした液に対して,コラーゲン450 mLを添加した混液を24穴プレートに播種した。24および48時間培養後,肉眼および顕微鏡による観察を行った。これらのうち、肉眼で観察し、撮影したものを図3、24時間場培養後の顕微鏡写真(100倍)を図4、48時間培養後の顕微鏡写真(200倍)を図5にそれぞれ示した。なお、比較対照として抗糖化剤である塩酸アミノグアニジンについても同様な方法で試験を行った。
線維芽細胞とコラーゲン溶液を培養した結果,図3に示すようにコラーゲン格子の形成が見られた。図3において白いもやが線維芽細胞の伸展突起とコラーゲン格子である。グリオキサールで処理したcontrolではnormalと比較して、コラーゲン格子の形成量は明らかに減少した。これに対し,桜花エキス(実施例1)100および1000 mg/mLをグリオキサールとともに処理した線維芽細胞を用いたウェルでは、格子形成量の増加が認められた。一方、塩酸グアニジン100 mg/mLで処理した線維芽細胞を用いたウェルでは、controlと比較して格子の形成量の増加は見られなかった。また、24時間培養後の鏡検像(図4)を見ると,normal,controlおよび塩酸グアニジンでは線維芽細胞が球状であるのに対し,桜花エキス(実施例1)100および1000 mg/mLのウェルでは明らかな線維芽細胞の伸展が認められた。また、48時間培養後の200倍に拡大した鏡検像(図5)では、controlで若干の線維芽細胞の伸展が見られたが、桜花エキス(実施例1)100および1000 mg/mLのウェルと比較すると弱いものであった。
試験例4では、桜花エキス(実施例1)および1-O-(E)-カフェオイル-β-D-グルコピラノシド(実施例3)がグリオキサールによる線維芽細胞(ヒト正常二倍体線維芽細胞)内AGE(carboxy methyl lysinタンパク)産生に及ぼす作用を下記の方法により検討した。
図6に示されるように、抗AGE抗体を用いたAGEの検出の結果,グリオキサール刺激によりAGE量は増加した(ControlとNonの比較)。これに対し、桜花エキス(実施例1)は10 mg/mLでAGEの生成を強く抑制した。100 mg/mLでは効果は減弱したが抑制作用が見られた。含有成分の1-O-(E)-カフェオイル-β-D-グルコピラノシド(CaGlu:実施例3) も1および10 mg/mLでAGEの生成を抑制した。以上の結果から,桜花エキスとその主成分はAGEの生成を抑制することが、細胞レベルで明らかになった。
桜花エキス(実施例1)を各種濃度で継続投与した後、AGEの前駆体であるグリオキサールを皮内投与し、AGE生成に及ぼす作用を調査した。また、この結果をもとに桜花エキス(実施例1)の推奨摂取量を設定した。試験方法は下記の通りである。
解凍した上清のタンパク含量をBCA法で測定後、グルコース由来AGEs測定用ELISAキット(Trans Genic Inc)を用いてAGE含量を測定した。また併せてウェスタンブロッティングによるAGEの検出も行った。結果を表5および図7に示す。
表5および図7に示すように、桜花エキス(実施例1)は用量依存性は見られなかったが、10~100mg/kgの経口投与でAGEの生成を抑制することが確認された。また、この結果より、桜花エキス(実施例1)の推奨摂取量は10~100mg/日となる。
桜花エキス(実施例1)を各種濃度で継続投与した後、CML化コラーゲンを皮内投与し、線維芽細胞をはじめとする真皮細胞のアポトーシスに及ぼすを調査した。試験方法は下記の通りである。
CML化コラーゲンの投与により,真皮のTUNNEL染色陽性細胞数は増加した。これに対し,桜花エキス(実施例1)は用量依存的にTUNNEL染色陽性細胞数を減少させた。この結果より,桜花エキス(実施例1)はin vivoにおいてもCML化コラーゲンによる真皮の線維芽細胞のアポトーシスを抑制することが判明した。

- 下記の化合物からなる群、1-O-(E)-カフェオイル-β-D-グルコピラノシド(1-O-(E)-Caffeoyl-β-D-glucopyranoside)、1-O-(E)-クマロイル-β-D-グルコピラノシド(1-O-(E)-Coumaroyl-β-D-glucopyranoside)、1-O-(E)-シンナモイル-β-D-グルコピラノシド(1-O-(E)-Cinnamoyl-β-D-glucopyranoside)、ケンフェロール 3-O-β-D-グルコピラノシド(Kaempferol 3-O-β-D-glucopyranoside)、ケルセチン 3-O-β-D-グルコピラノシド(Quercetin 3-O-β-D-glucopyranoside)、ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド(Kaempferol 3-O-(6''-malony)-β-D-glucopyranoside)、ケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(Quercetin 3-O-(6''-malony)-β-D-glucopyranoside)の全種を含有してなる桜抽出物。
- 下記の化合物からなる群、1-O-(E)-カフェオイル-β-D-グルコピラノシド(1-O-(E)-Caffeoyl-β-D-glucopyranoside)、1-O-(E)-クマロイル-β-D-グルコピラノシド(1-O-(E)-Coumaroyl-β-D-glucopyranoside)、1-O-(E)-シンナモイル-β-D-グルコピラノシド(1-O-(E)-Cinnamoyl-β-D-glucopyranoside)、ケンフェロール 3-O-β-D-グルコピラノシド(Kaempferol 3-O-β-D-glucopyranoside)、ケルセチン 3-O-β-D-グルコピラノシド(Quercetin 3-O-β-D-glucopyranoside)、ケンフェロール3-O-(6''-マロニル)-β-D-グルコピラノシド(Kaempferol 3-O-(6''-malony)-β-D-glucopyranoside)、ケルセチン 3-O-(6''-マロニル)-β-D-グルコピラノシド(Quercetin 3-O-(6''-malony)-β-D-glucopyranoside)の全種を含有する桜抽出物であって、
下記工程(a)~(c)、
(a)桜の花および/または葉を20~50wt%の含水エタノールで抽出する工程
(b)前記工程(a)で得られた抽出物を吸着剤に吸着させた後、水で溶出して水溶出部を取り除く工程
(c)前記工程(b)に続いて前記抽出物を水溶性溶媒で溶出し、この溶出液を濃縮する工程
を含む製造方法により製造される、前記桜抽出物。 - 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有するAGE産生抑制剤。
- 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有するアポトーシス抑制剤であって、AGEであるCML化コラーゲン(Carboxymethyl lysine-collagen)による線維芽細胞のアポトーシスに対して抑制作用を有するアポトーシス抑制剤。
- 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有するヒト線維芽細胞のコラーゲン格子形成促進剤。
- 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有する線維芽細胞内AGE産生抑制剤。
- 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有する糖尿病合併症の予防・治療剤。
- 請求項27記載の桜抽出物および/またはその処理物を有効成分として含有する皮膚老化防止・改善剤。
