又、近年、ヒトに対する抗原性、血中半減期などの観点から、抗体中のアミノ酸配列に対して置換などの何らかの改変が行われた改変抗体が医薬品として開発されている。例えば、ヒトに対する抗原性を減少させる為にヒト化やキメラ化等が行われたキメラ抗体やヒト化抗体が多数報告されている。これらのヒト化抗体やキメラ化抗体は医薬品として優れた性質を有していると考えられている。さらに、抗体の血中半減期などを改善する為に、ヒト由来の抗体定常領域にアミノ酸の改変が行われた抗体定常領域も報告されている(WO2009/041613:特許文献1)。
しかしながら、抗体のアゴニスト活性を増強する為に抗体の定常領域のアミノ酸を改変した報告はない。

抗体の重鎖定常領域にはIgG1、IgG2、IgG3、IgG4タイプの定常領域が存在する。本発明は、特に限定されないが、アミノ酸配列が改変されたIgG1定常領域に関する。より具体的には本発明は、アミノ酸配列が改変されたヒトIgG1定常領域に関する。ヒトIgG1定常領域のアミノ酸配列は公知である(配列番号:32)。
本発明のヒトIgG1定常領域としては、遺伝子多型による複数のアロタイプ配列がSequences of proteins of immunological interest, NIH Publication No.91-3242に記載されているが、本発明においてはそのいずれであっても良い。
また本発明の抗体重鎖定常領域は、本発明に基づいて導入されたアミノ酸配列の改変に加え、更に付加的な改変を含むことができる。付加的な改変は、たとえば、アミノ酸の置換、欠損、あるいは修飾のいずれか、あるいはそれらの組み合わせから選択することができる。具体的には、そのアミノ酸配列に次のような改変を含む定常領域は、いずれも本発明に含まれる。
*配列番号:32(ヒトIgG1定常領域)のアミノ酸配列に対して本発明に基づく改変を導入する。
*改変された配列番号:32(ヒトIgG1定常領域)のアミノ酸配列に対して本発明に基づく改変を導入する。

本発明のヒンジ領域にはUpperヒンジ領域、Middleヒンジ領域、Lowerヒンジ領域が含まれる。本発明の抗体重鎖定常領域は、アッパーヒンジ領域において少なくとも1つのアミノ酸の欠損を含む。通常、アッパーヒンジ領域はEUナンバリング216-225の領域である。

本発明は、アミノ酸配列が改変された抗体重鎖定常領域を提供する。好ましくは、本発明は抗体に増強したアゴニスト活性を付与することが可能な抗体定常領域を提供する。
より具体的には、配列番号:32(IgG1定常領域)のアミノ酸配列において、アッパーヒンジ領域の少なくとも1つのアミノ酸の欠損を含むアミノ酸配列を有する抗体定常領域を提供する。
アミノ酸が欠損する部位は特に限定されず、アッパーヒンジ領域の如何なるアミノ酸が欠損してもよい。

本発明により提供される抗体重鎖定常領域は、少なくとも上述のアミノ酸置換が導入されている限り、同時に他のアミノ酸の改変(置換、欠損、付加および/または挿入など)や修飾が導入されていてもよい。
さらに、本発明は本発明の抗体定常領域をコードする遺伝子、該遺伝子を含むベクター、該ベクターを含む細胞(宿主細胞など)、該細胞を培養して本発明の抗体定常領域を製造する方法を提供する。遺伝子、ベクター、細胞、製造方法については後述の通りである。
さらに、本発明は上述のいずれかに記載のアミノ酸配列が改変された重鎖定常領域を含む抗体を提供する。
さらに、本発明は、ヒトIgG1の重鎖アッパーヒンジ領域の少なくとも1つのアミノ酸が欠損され又は他のアミノ酸に置換された抗体定常領域を含むアゴニスト抗体であって、かつ当該欠損又は置換前と比較してアゴニスト活性が増強している抗体を提供する。
本発明の抗体を構成する可変領域は、任意の抗原を認識する可変領域であることが出来る。本発明における好ましい可変領域の例として、アゴニスト活性を有する抗体の可変領域を示すことが出来る。たとえば、アゴニスト活性を有する抗HLA-A抗体の可変領域を、本発明の抗体を構成する可変領域とすることが出来る。重鎖可変領域を構成するアミノ酸配列は、その抗原結合活性が維持される限り、1または複数のアミノ酸残基の改変が許容される。
例えば、可変領域のN末端のグルタミンのピログルタミル化によるピログルタミン酸への修飾は当業者によく知られた修飾である。したがって、本発明の抗体は、その重鎖のN末端がグルタミンの場合には、それがピログルタミン酸に修飾された可変領域を含む。
なお、抗体の軽鎖定常領域にはκ鎖とλ鎖タイプの定常領域が存在しているが、いずれの軽鎖定常領域であってもよい。さらに、本発明において軽鎖定常領域は、アミノ酸の置換、欠損、付加および/または挿入などの改変が行われた軽鎖定常領域であってもよい。
また本発明の抗体分子には、本発明の抗体定常領域を含む限り、抗体の修飾物も含まれる。
抗体の修飾物の例としては、例えば、ポリエチレングリコール(PEG)や細胞障害性物質等の各種分子と結合させた抗体を挙げることができる。このような抗体修飾物は、本発明の抗体に化学的な修飾を施すことによって得ることができる。抗体の修飾方法はこの分野においてすでに確立されている。
本発明の抗体定常領域は任意の抗原に対する抗体の定常領域として使用することが可能であり、抗原は特に限定されない。

無細胞系の指標としては、酵素反応やタンパク質、DNA、RNAの量的及び/又は質的な変化を用いることができる。酵素反応としては、例えば、アミノ酸転移反応、糖転移反応、脱水反応、脱水素反応、基質切断反応等を用いることができる。また、タンパク質のリン酸化、脱リン酸化、二量化、多量化、分解、乖離等や、DNA、RNAの増幅、切断、伸長を用いることができる。例えばシグナル伝達経路の下流に存在するタンパク質のリン酸化を検出指標とすることができる。
細胞系の指標としては、細胞の表現型の変化、例えば、産生物質の量的及び/又は質的変化、増殖活性の変化、細胞数の変化、形態の変化、特性の変化等を用いることができる。産生物質としては、分泌タンパク質、表面抗原、細胞内タンパク質、mRNA等を用いることができる。形態の変化としては、突起形成及び/又は突起の数の変化、偏平度の変化、伸長度/縦横比の変化、細胞の大きさの変化、内部構造の変化、細胞集団としての異形性/均一性、細胞密度の変化等を用いることができる。これらの形態の変化は検鏡下での観察で確認することができる。特性の変化としては、足場依存性、サイトカイン依存応答性、ホルモン依存性、薬剤耐性、細胞運動性、細胞遊走活性、拍動性、細胞内物質の変化等を用いることができる。細胞運動性としては、細胞浸潤活性、細胞遊走活性がある。また、細胞内物質の変化としては例えば、酵素活性、mRNA量、Ca2+やcAMP等の細胞内情報伝達物質量、細胞内タンパク質量等を用いることができる。また、細胞膜受容体の場合には、受容体の刺激によって誘導される細胞の増殖活性の変化を指標とすることができる。
組織系の指標としては、使用する組織に応じた機能変化を検出指標とすることができる。生体系の指標としては組織重量変化、血液系の変化、例えば血球細胞数の変化、タンパク質量や、酵素活性、電解質量の変化、また、循環器系の変化、例えば、血圧、心拍数の変化等を用いることができる。
例えば、吸収スペクトルは一般的に用いられるフォトメータやプレートリーダ等、発光はルミノメータ等、蛍光はフルオロメータ等で測定することができる。質量は質量分析計を用いて測定することができる。放射活性は、放射線の種類に応じてガンマカウンターなどの測定機器を用いて、蛍光偏光度はBEACON(宝酒造)、表面プラズモン共鳴シグナルはBIACORE、時間分解蛍光、蛍光共鳴エネルギー移動などはARVOなどにより測定できる。さらに、フローサイトメータなども測定に用いることができる。これらの測定方法は、一つの測定方法で2種以上の検出指標を測定しても良く、簡便であれば、2種以上の測定を同時及び/又は連続して測定することによりさらに多数の検出指標を測定することも可能である。例えば、蛍光と蛍光共鳴エネルギー移動を同時にフルオロメータで測定することができる。
アゴニスト依存性増殖を示す細胞も当業者に公知の方法により作製することが可能であり、例えば、細胞膜上のタンパク質が細胞増殖シグナルを発する受容体である場合には、該受容体を発現している細胞を用いればよい。又、細胞膜上のタンパク質が細胞増殖シグナルを出さない受容体である場合には、細胞増殖シグナルを発する受容体の細胞内領域と、細胞増殖シグナルを出さない受容体の細胞外領域からなるキメラ受容体を作製し、該キメラ受容体を細胞で発現させればよい。細胞増殖シグナルを発する受容体の例としては、例えば、G-CSF受容体、mpl、neu、GM-CSF受容体、EPO受容体、c-kit、FLT-3等を挙げることができる。受容体を発現させる細胞としては、例えば、BaF3、NFS60、FDCP-1、FDCP-2、CTLL-2、DA-1、KT-3等を挙げることができる。
本発明は、本発明の抗体を含む、医薬組成物を提供する。
本発明の医薬組成物は、抗体に加えて医薬的に許容し得る担体を導入し、公知の方法で製剤化することが可能である。例えば、水もしくはそれ以外の薬学的に許容し得る液との無菌性溶液、又は懸濁液剤の注射剤の形で非経口的に使用できる。例えば、薬理学上許容される担体もしくは媒体、具体的には、滅菌水や生理食塩水、植物油、乳化剤、懸濁剤、界面活性剤、安定剤、香味剤、賦形剤、ベヒクル、防腐剤、結合剤などと適宜組み合わせて、一般に認められた製薬実施に要求される単位用量形態で混和することによって製剤化することが考えられる。これら製剤における有効成分量は指示された範囲の適当な容量が得られるようにするものである。
注射のための無菌組成物は注射用蒸留水のようなベヒクルを用いて通常の製剤実施に従って処方することができる。
油性液としてはゴマ油、大豆油が挙げられ、溶解補助剤として安息香酸ベンジル、ベンジルアルコールと併用してもよい。また、緩衝剤、例えばリン酸塩緩衝液、酢酸ナトリウム緩衝液、無痛化剤、例えば、塩酸プロカイン、安定剤、例えばベンジルアルコール、フェノール、酸化防止剤と配合してもよい。調製された注射液は通常、適当なアンプルに充填させる。
また、患者の年齢、症状により適宜投与方法を選択することができる。抗体または抗体をコードするポリヌクレオチドを含有する医薬組成物の投与量としては、例えば、一回につき体重1kgあたり0.0001mgから1000mgの範囲で選ぶことが可能である。あるいは、例えば、患者あたり0.001から100000mg/bodyの範囲で投与量を選ぶことができるが、これらの数値に必ずしも制限されるものではない。投与量、投与方法は、患者の体重や年齢、症状などにより変動するが、当業者であれば適宜選択することが可能である。

[実施例1] マウス抗HLA-A抗体の取得
ハイブリドーマ作製
1.1. 免疫用可溶型HLA-A-flag(sHLA-A-flag)の調製
1.1.1. sHLA-A発現ベクターの構築とHLA産生CHO株の作製
HLA-A6802(GenBank No. AM235885)の細胞外ドメインのC末に、flagタグを付与したHLA-A-flag(配列番号:1、307~314はflagタグ)を動物細胞発現用のベクターに導入した。塩基配列の確認は、BigDye Terminator Cycle Sequencing Kit(Applied Biosystems)を用い、DNAシークエンサーABI PRISM 3700 DNA Sequencer(Applied Biosystems)にて、添付説明書記載の方法に従い決定した。作製した発現ベクターをCHO細胞に導入し、HLA産生CHO株を構築した。
1.1.2. sHLA-A-flagの精製
1.1.1で作製したCHO細胞の培養上清から、Q-Sepharose FF (GE Healthcare)、ANTI-FLAG M2 Affinity Gel (SIGMA)およびHiLoad 16/60 Superdex 200 pg (GE Healthcare) を用いて、可溶型HLA-Flagタンパク質を精製した。ゲルろ過溶出画分は、Centriprep YM-10 (Millipore) にて濃縮後、Dc Protein Assay Kit (BIO-RAD) にてウシガンマグロブリンに換算し、タンパク質濃度を算出した。
1.2. マウス抗HLA-A抗体産生ハイブリドーマの作製
1.2.1. HLA-A免疫マウスを用いたハイブリドーマの作製
MRL/lpr、雄、4週齢マウス(日本チャールズリバー)を免疫に用いた。
初回免疫では、精製した可溶型HLA-A-FLAG (sHLA-flag)蛋白をcompleteアジュバンドH37Ra(DIFCO #231131)で100μg / headになるようにマウス匹数分調整し、エマルジョンを作成した後、100μlずつを皮下に免疫した。二回目以降の免疫では、incomplete アジュバンド (DIFCO #263910)を用いてエマルジョンを作成し、50μg / headの免疫を行った。免疫は1週間おきに行い、血清抗体価の上昇を確認後、最終免疫(免疫6回目)で、sHLA-flagを50μg/200μlになるようにPBSで調整し、200μl/headでマウスにi.v.投与した。
最終免疫の4日後、マウスの脾臓細胞とマウスミエローマ細胞P3X63Ag8U.1(P3U1と称す、ATCC CRL-1597)をPEG1500(Roche Diagnostics)を用いた常法に従い細胞融合した。融合細胞、すなわちハイブリドーマは、HAT培地 [RPMI1640 + PS, 10% FCS, HAT (Sigma, H0262), 5% BM condimed H1 (Roche: #1088947)] にて培養した。
1.2.2. ハイブリドーマ細胞の選抜
一次スクリーニングは、フュージョンから約1週間後に、細胞凝集誘導活性を指標に行った。細胞凝集誘導活性は、ARH77細胞を2 x 104 cells/well・40μlで96 well plateに撒き、各ハイブリドーマの培養上清80μlを加え37℃で4時間培養した。各ウェルを顕微鏡で1つ1つ観察し、細胞凝集を起こしているウェルを目視で判断した。細胞凝集を起こしているウェルのハイブリドーマ細胞を陽性クローンとして選択した。細胞凝集誘導活性が陽性であったウェルのハイブリドーマ細胞を2.5 cells/wellとなるように96 well plateにまきなおし、約10日間培養した後に、再び細胞凝集誘導活性を調べた。二次スクリーニングは、抗体の細胞死誘導活性を指標に実施した。ARH77細胞を、1~8 x 105 cells /well で24 well plateに撒き、これに各抗体を加え、200μl培地中で37℃4~5時間以上培養した。これをエッペンに回収後、100~200μl propidium iodide (PI) 溶液 (5μg/ml in FACS buffer)に懸濁し、室温で15分インキュベートした。その後、遠心により細胞を沈殿させた後、500μl FACS bufferに懸濁し、FACS(Beckman Coulter, ELITE)を行ってPI陽性細胞(死細胞)の割合を測定し、各クローンの細胞死誘導活性を比較した。この結果、最も細胞死誘導活性の強かったクローンF17B1を選抜した。
ハイブリドーマ細胞から、RNeasy Mini Kits(QIAGEN)を用いてトータルRNAを抽出し、SMART RACE cDNA Amplification Kit(BD Biosciences)によりcDNAを合成した。作製したcDNAを用いて、PCRにより、抗体の可変領域遺伝子をクローニングベクターに挿入した。各DNA断片の塩基配列は、BigDye Terminator Cycle Sequencing Kit(Applied Biosystems)を用い、DNAシークエンサーABI PRISM 3700 DNA Sequencer(Applied Biosystems)にて、添付説明書記載の方法に従い決定した。決定したF17マウス抗体のH鎖可変領域を配列番号:2に、L鎖可変領域を配列番号:3に示した。CDR,FRの決定はKabat numberingに従って行った。
マウスF17B1抗体の可変領域をヒトのgermline配列、またはヒト抗体配列と比較し、抗体のヒト化に用いるFR配列を決定した。使用したFR配列を表1にまとめた。
ヒト化H鎖可変領域H2(配列番号:4)は表1に記載されるFR1、FR2、FR3、FR4から構成される。作製したH2と定常領域G1d(配列番号:6)を結合したH2_G1d(配列番号:8)を動物細胞発現ベクターに導入した。
ヒト化L鎖可変領域(配列番号:5)は表1に記載されるFR1、FR2、FR3、FR4から構成される。FR3は2種類のgermline配列 hVk2_28; GVPDRFSGSGSGTDFTLKISRVEAEDVGVYYC(配列番号:18)とVk6_21; GVPSRFSGSGSGTDFTLTINSLEAEDAATYYC(配列番号:19)から構成される。作製したL1と定常領域k0(配列番号:7)を結合したL1_k0(配列番号:9)を動物細胞発現ベクターに導入した。
作製した発現ベクターを用いて、H鎖としてH2_G1d(配列番号:8)、L鎖としてL1_k0(配列番号:9)からなるH2_G1d/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。
ヒトMHC classIを発現しているヒトリンパ芽球腫細胞株IM-9(ATCC,CCL-159)を用いて、以下に示す通り抗ヒトMHC classI抗体H2-G1d_L1-k0による細胞死誘導活性を測定した。
精製したH2-G1d_L1-k0を培地にて20 μg/mLに調整し、96well U bottom plate(BM Equipment)を用いて希釈公比3、合計9系列の希釈液を調製した(final conc.;10-0.0015 μg/mL)。IM-9細胞を10% FBS (BOVOGEN)、2 mM L-glutamine、1.5 g/l sodium bicarbonate、4.5 g/L glucose、10 mM HEPES、1 mM sodium pyruvateを含むRPMI1640倍地 (SIGMA) で1.2x105 cells/mLとなるように調製した。H2-G1d_L1-k0希釈溶液50μLに細胞懸濁液50μLを播種し、37℃、5% CO2インキュベーターにて3日間培養した。培養終了後、各wellにCell Counting Kit-8(Dojindo)を10μL添加し、37℃、5% CO2インキュベーターにて4時間反応させた。その後、吸光度(450 nm)を測定した(Molecular Devices, Emax)。H2-G1d_L1-k0の細胞死誘導活性は、培地のみ(細胞及び抗体非添加)のwellの値を0%, 細胞のみ(抗体非添加)のwellの値を100%として、阻害率(%)を算出した。測定はn=5で行い、その平均値を用いた。その結果、図1に示すように、H2-G1d_L1-k0はIM9細胞株において、濃度依存的に細胞増殖を抑制することが明らかとなった。
HLA class I に結合し、細胞死活性を誘導する抗体は幾つか知られている(Rolf et al, The journal of immunology, 1996, 156:1415-1424)。これらの抗体はHLA classIを活性化させることによって、細胞死を誘導する。マウスF17B1抗体もこれらと同様にHLAを活性化させることにより細胞死を誘導するAgonist抗体だと予想される。多くのAgonist抗体がFab断片のみでは活性を示さないことから、2価で結合し2量体を形成することが重要であると考えられる。また、全長抗体では十分な活性を示さない抗体でもsc(Fv)2等の形態に低分子化することでAgonist活性が上昇することが報告されている。これらのことより、本発明者らはAgonist活性を示すには2つのFabの距離が重要であると予想した。2つのFabの距離や動きやすさは抗体のサブクラス間によって異なる。2つのFabの距離あるいは動きやすさに大きな影響を与えるのがHinge領域である(EMBO J. 1988 Jul;7(7):1989-94, Schneider WP et al. Proc Natl Acad Sci U S A. 1988 Apr;85(8):2509-13)。そこで、Hinge領域を改変することによって、Agonist活性を増強することを考えた。
IgG1においてEUナンバリング(Sequences of proteins of immunological interest, NIH Publication No.91-3242 を参照)221-225のHinge領域はopen-turn helixを形成している (Marquart M et al. J Mol Biol. 1980 Aug 25;141(4):369-91, Ito W et al ,Biochemistry. 1985 Nov 5;24(23):6467-74.)。IgG1の221-225のアミノ酸配列のうちThr-His-Thrの配列はIgG2,4には存在しない(表2)。そこで、この配列に注目し、G1d(配列番号:6)に対して、223-225のThr-His-Thrを欠損させた新規定常領域M97(配列番号:20)を作製することを考えた。M97の細胞死活性に対する影響を検討するために、H2_M97(配列番号:21)の発現ベクターを参考例1の方法に従って行った。H鎖としてH2_M97(配列番号:21)、L鎖としてL1_k0(配列番号:9)からなるH2_M97/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。作製した各抗体の細胞死活性を実施例4に記載した方法で実施した。
図2に示すように、Thr-His-Thrを欠損させることによって、細胞死活性が向上することが確認された。
天然型のIgG1において、226と229番目に存在するCysはもう一方のH鎖とジスルフィド結合を形成する。また、H鎖EUナンバリング220番目のCysとL鎖EUナンバリング214番目のCysがジスルフィド結合をしている。ジスルフィド結合をしている220番目と226番目のCysの間にAsp-Lys-Thr-His-Thrの5アミノ酸が存在する。その5残基のうち3残基のThr-His-Thr配列を欠損させることにより細胞死活性が向上したことから、残りの2残基のAsp-Lys配列を欠損させることにより、さらに細胞死活性が向上することを考えた。また、Hinge領域を短くすることにより細胞死活性が向上することから、218番目のLys、219番目のSerを欠損させることにより、さらに細胞死活性が向上することを考えた。
M97(配列番号:20)に対して、H鎖222番目のLysを欠損させた定常領域をM167(配列番号:39)、H鎖221番目のAspと222番目のLysを欠損させた定常領域M168(配列番号:40)、H鎖218番目のLysと219番目のSerと221番目のAspと222番目のLysを欠損させた定常領域M169(配列番号:37)を作製することを考えた。作製した各定常領域の配列を表2にまとめた。
H2_M167(配列番号:43)、H2_M168(配列番号:44)、 H2_M169(配列番号:41)、H2_M154(配列番号:42)の発現ベクターを参考例1の方法に従って行った。H鎖としてH2_M167(配列番号:43)、L鎖としてL1_k0(配列番号:9)からなるH2_M167/L1_k0、H鎖としてH2_M168(配列番号:44)、L鎖としてL1_k0(配列番号:9)からなるH2_M168/L1_k0、H鎖としてH2_M169(配列番号:41)、L鎖としてL1_k0(配列番号:9)からなるH2_M169/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。作製した各抗体との細胞死活性を実施例4に記載した方法で実施した。

図3に示すように、Hinge regionの配列を短くすることにより、細胞死活性が向上する傾向が見られた。
天然型のIgG1においては、L鎖EUナンバリング214番目のCysとH鎖EUナンバリング220番目のCysとがジスルフィド結合をしている(Saphire EO, et al. Science. 293, 1155-9 (2001).)。Agonist活性において、2つのFabの距離や構造が重要であると考え、L鎖の214番目のCysとH鎖とのジスルフィド結合を変えることによって、活性が向上する可能性を検討した。
そこで、実施例5で作製したM97(配列番号:20)に対して、H鎖EUナンバリング220番目のCysをSerに、219番目のSerをCysに置換した、新規定常領域M98(配列番号:22)を作製することを考えた。M98の細胞死活性に対する影響を検討するために、H2_M98(配列番号:23)の発現ベクターを参考例1の方法に従って行った。H鎖としてH2_M98(配列番号:23)、L鎖としてL1_k0(配列番号:9)からなるH2_M98/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。作製した各抗体の細胞死活性を実施例4に記載した方法で実施した。
図4に示すように、Cysの位置を変えることによって細胞死活性が向上したことが確認された。
実施例5,6,7よりHinge領域を改変することにより、細胞死活性が上昇することが明らかとなった。各サブクラスのHinge領域の配列を比較すると、サブクラスにより配列が異なっていることがわかる。そこで、Hinge領域の配列、およびその周辺の配列を改変することによって細胞死活性を上昇させることを考えた。
Hinge領域のUpper Hingeのうち、実施例5,6,7で改変したアミノ酸以外の配列では、217、221、221のPro,Asp,Lysのアミノ酸配列が異なる。また、IgG1のCH1 domainに存在するアミノ酸のうち、192,193,199,214のSer,Leu,IIe,Lysは、他のサブクラスのアミノ酸配列と異なり、かつHinge領域周辺に存在しているため、Hingeの構造に影響を与える可能性が考えられた。
そこで、IgG1のアミノ酸配列を置換したM101(配列番号:24)、M132(配列番号:26) 、M146(配列番号:45)、M148(配列番号:33) 、M152(配列番号:34)を作製することを考えた。作製した各定常領域のHinge領域周辺の配列を表3にまとめた。H2_M101(配列番号:25)、H2_M132(配列番号:27)、H2_M146(配列番号:46)、H2_M148(配列番号:35)、 H2_M152(配列番号:36)の発現ベクターを参考例1の方法に従って行った。H鎖としてH2_M101(配列番号:25)、L鎖としてL1_k0(配列番号:9)からなるH2_M101/L1_k0、H鎖としてH2_M132(配列番号:27)、L鎖としてL1_k0(配列番号:9)からなるH2_M132/L1_k0、H鎖としてH2_M146(配列番号:46)、L鎖としてL1_k0(配列番号:9)からなるH2_M146/L1_k0、H鎖としてH2_M148(配列番号:35)、L鎖としてL1_k0(配列番号:9)からなるH2_M148/L1_k0、H鎖としてH2_M152(配列番号:36)、L鎖としてL1_k0(配列番号:9)からなるH2_M152/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。作製した各抗体と実施例5で作製したH2_M97/L1_k0の細胞死活性を実施例4に記載した方法で実施した。

EU numbering 216-225から構成されるUpper Hingeだけでなく231-238で構成されるLower Hingeも定常領域の構造に影響を与えることがマウス定常領域を用いて明らかになっている(Kim H et al. J Mol Biol. 1994 Feb 11;236(1):300-9)。そこで、M101(配列番号:24)、M132(配列番号:26)のLower HingeをIgG2のそれと置換したM133(配列番号:28)、M134(配列番号:30)を作製することを考えた。
H2_M133(配列番号:29)、H2_M134(配列番号:31)の発現ベクターを参考例1の方法に従って行った。H鎖としてH2_M133(配列番号:29)、L鎖としてL1_k0(配列番号:9)からなるH2_M133/L1_k0、H鎖としてH2_M134(配列番号:31)、L鎖としてL1_k0(配列番号:9)からなるH2_M134/L1_k0を作製した。抗体の発現と精製は参考例1に記した方法で実施した。作製した各抗体と実施例5で作製したH2_M97/L1_k0の細胞死活性を実施例4に記載した方法で実施した。

図6に示すように、IgG1のLower HingeをIgG2のそれと置換すると、細胞死活性が減少する傾向が見られた。細胞死活性にはLower Hingeではなく、Upper Hingeが重要であることが示された。
実施例8で作製したH2-M132/L1-k0におけるHingeの欠損による、活性の増強効果を確認検証するために、M132(配列番号:26)に対して、H鎖221番目のAspと222番目のLysを欠損させた定常領域をM154(配列番号:38)を作製することを考えた。H鎖としてH2_M154(配列番号:42)、L鎖としてL1_k0(配列番号:9)からなるH2_M154/L1_k0を作製した。

図7に示すように、Hinge regionの配列を短くすることにより、細胞死活性が向上する傾向が見られた。このことから、Hinge部位の欠損は、IgG1だけでなく、IgG1を改変した定常領域においても、活性を向上させることが明らかとなった。
目的の抗体のH鎖およびL鎖の塩基配列をコードする遺伝子は、Assemble PCR等を用いて当業者公知の方法で行った。アミノ酸置換の導入はQuikChange Site-Directed Mutagenesis Kit(Stratagene)あるいはPCR等を用いて当業者公知の方法で行った。得られたプラスミド断片を動物細胞発現ベクターに挿入し、目的のH鎖発現ベクターおよびL鎖発現ベクターを作製した。得られた発現ベクターの塩基配列は当業者公知の方法で決定した。抗体の発現は以下の方法を用いて行った。ヒト胎児腎癌細胞由来HEK293H株(Invitrogen)を10 % Fetal Bovine Serum (Invitrogen)を含むDMEM培地(Invitrogen)へ懸濁し、5~6 × 105個/mLの細胞密度で接着細胞用ディッシュ(直径10 cm, CORNING)の各ディッシュへ10 mLずつ蒔きこみCO2インキュベーター(37℃、5% CO2)内で一昼夜培養した後に、培地を吸引除去し、CHO-S-SFM-II(Invitrogen)培地6.9 mLを添加した。調製したプラスミドをlipofection法により細胞へ導入した。得られた培養上清を回収した後、遠心分離(約2000 g、5分間、室温)して細胞を除去し、さらに0.22μmフィルターMILLEX(R)-GV(Millipore)を通して滅菌して培養上清を得た。得られた培養上清からrProtein A SepharoseTM Fast Flow(Amersham Biosciences)を用いて当業者公知の方法で抗体を精製した。精製抗体濃度は、分光光度計を用いて280 nmでの吸光度を測定した。得られた値からPACE法により算出された吸光係数を用いて抗体濃度を算出した(Protein Science 1995 ; 4 : 2411-2423)

- 配列番号:32(IgG1定常領域)のアミノ酸配列において、アッパーヒンジ領域の少なくとも1つのアミノ酸の欠損を含むアミノ酸配列を有する抗体定常領域。
- EUナンバリング218番目のLys、219番目のSer、221番目のAsp、222番目のLys、223番目のThr、224番目のHis、225番目のThrから選択される少なくとも1つのアミノ酸の欠損を含む請求項1に記載の抗体定常領域。
- EUナンバリング223番目のThr、224番目のHis、225番目のThrから選択される少なくとも1つのアミノ酸の欠損を含む請求項1に記載の抗体定常領域。
- EUナンバリング223番目のThr、224番目のHisおよび225番目のThrの欠損を含むことを特徴とする請求項1に記載の抗体定常領域。
- EUナンバリング222番目のLysの欠損をさらに含むことを特徴とする請求項4に記載の抗体定常領域。
- EUナンバリング221番目のAspおよび222番目のLysの欠損をさらに含むことを特徴とする請求項4に記載の抗体定常領域。
- EUナンバリング218番目のLys、219番目のSer、221番目のAspおよび222番目のLysの欠損をさらに含むことを特徴とする請求項4に記載の抗体定常領域。
- EUナンバリング192番目のSer、193番目のLeu、199番目のIle、214番目のLys、217番目のPro、219番目のSer、220番目のCys、221番目のAsp、222番目のLysから選択される少なくとも1つのアミノ酸の他のアミノ酸への置換をさらに含むことを特徴とする請求項1~7いずれかに記載の抗体定常領域。
- EUナンバリング219番目のSerおよび220番目のCysの他のアミノ酸への置換を含むことを特徴とする請求項8に記載の抗体定常領域。
- 219番目のSer、220番目のCys、221番目のAspおよび222番目のLysの他のアミノ酸への置換を含むことを特徴とする請求項8に記載の抗体定常領域。
- EUナンバリング214番目のLys、217番目のPro、219番目のSer、220番目のCys、221番目のAspおよび222番目のLysの他のアミノ酸への置換を含むことを特徴とする請求項8に記載の抗体定常領域。
