Abstract
1. Способ увеличения экспрессии NGN3 и NKX6.1 в популяции клеток, экспрессирующих маркеры, характерные для эндокринной линии поджелудочной железы, включающий следующие стадии:a) культивирования плюрипотентных стволовых клеток,b) дифференцировки плюрипотентных стволовых клеток в клетки, экспрессирующие маркеры, характерные для линии дефинитивной эндодермы,c) дифференцировки клеток, экспрессирующих маркеры, характерные для линии дефинитивной эндодермы, в клетки, экспрессирующие маркеры, характерные для линии панкреатической эндодермы, путем добавления в среду, используемую для дифференцировки линии дефинитивной эндодермы, соединений, выбираемых из группы, которая состоит из H-9, H-89, GF 109203X, HA-1004, PP2, PP1, LY 294002, Вортманнина, SB-203580, SB-202190, Тирфостина 25, Тирфостина, AG1478, Тирфостина 46, GW 5074, Кенпауллона, HNMPA, AG490, Y27632 и ML-7, иd) дифференцировки клеток, экспрессирующих маркеры, характерные для линии панкреатической эндодермы, в клетки, экспресмирующие маркеры, характерные для эндокринной линии поджелудочной железы.2. Claims (2)
1. Способ увеличения экспрессии NGN3 и NKX6.1 в популяции клеток, экспрессирующих маркеры, характерные для эндокринной линии поджелудочной железы, включающий следующие стадии:
a) культивирования плюрипотентных стволовых клеток,
b) дифференцировки плюрипотентных стволовых клеток в клетки, экспрессирующие маркеры, характерные для линии дефинитивной эндодермы,
c) дифференцировки клеток, экспрессирующих маркеры, характерные для линии дефинитивной эндодермы, в клетки, экспрессирующие маркеры, характерные для линии панкреатической эндодермы, путем добавления в среду, используемую для дифференцировки линии дефинитивной эндодермы, соединений, выбираемых из группы, которая состоит из H-9, H-89, GF 109203X, HA-1004, PP2, PP1, LY 294002, Вортманнина, SB-203580, SB-202190, Тирфостина 25, Тирфостина, AG1478, Тирфостина 46, GW 5074, Кенпауллона, HNMPA, AG490, Y27632 и ML-7, и
