Abstract
Foreliggende oppfinnelse gjelder en fremgangsmåte for oppkonsentrering av epotiloner i dyrkningsmedier, en fremgangsmåte for fremstilling av epotiloner, en fremgangsmåte for å skille epotilon A og B fra hverandre og en stamme erholdt ved mutagenese for fremstilling av epotiloner, såvel som beslektede utførelser. Krystallformer av epotilon B beskrives også.
Description
Foreliggende oppfinnelse gjelder en ny fremgangsmåte for separasjon av epotiloner, fortrinnsvis epotilon A og B.
Av de eksisterende cytotoksiske aktive bestanddeler for behandling av tumorer spiller Taxol® (Paclitaxel; Bristol-Myers Squibb), et mikrotubulistabiliserende middel, en viktig rolle og har en bemerkelsesverdig kommersiell suksess. Imidlertid har Taxol<®> en rekke ulemper. Særlig er den svært lave løselighet i vann et problem. Det ble derfor nødvendig å tilføre 'Taxol" i et preparat sammen med "Cremophor EL"
(polyoksyetylert lakserolje, BASF, Ludwigshafen, Tyskland). "Cremophor EL" har alvorlige bivirkninger og forårsaker for eksempel allergier som i minst ett tilfelle har ført til en pasients død.
Selv om Taxan-klassen av mikrotubulistabiliserende anti-cancermidler har blitt anbefalt som "kanskje det viktigste tilskudd til det farmasøytiske arsenal mot cancer i det siste tiår "(se Rowinsky E.K., Ann. Rev. Med. 48,353-374 (1997)), og trass i den kommersielle suksess til "Taxol" synes disse forbindelser fortsatt ikke å representere et virkelig stort gjennombrudd innen cancerkjemoterapi. Behandling med "Taxol" er forbundet med en serie av betydelige bivirkninger, og noen få primærklasser av kompakte tumorer, nemlig kolontumorer og prostatatumorer, responderer på denne forbindelse kun i lite omfang (se Rowinsky E.K., inter alia). I tillegg kan virkningen av Taxol svekkes og til og med fullstendig nøytraliseres ved ervervede resistens-mekanismer, særlig slike basert på overekspresjon av fosfoproteiner som fungerer som utstrømningspumper for aktive bestanddeler, for eksempel "flermedikamentsresistens" grunnet overekspresjon av flermedikaments-transportglykoprotein "P-glykoprotein".
Epotilon A og B representerer en ny klasse av mikrotubulistabiliserende, cytotoksiske aktive bestanddeler (se Gerth, K. Et al., J. Antibiot. 49,560-3 (1966)) med formelen:
hvori R står for hydrogen (epotilon A) eller metyl (epotilon B).
Disse forbindelser har følgende fordeler framfor "Taxol":
a) De har bedre vannløselighet og er således lettere tilgjengelig for preparatdannelse. b) Det har blitt rapportert at de i cellekultureksperimenter også er aktive mot proliferasjon av celler som, grunnet aktiviteten av P-glycoproteinpumpen er "flermedikamentresistente", viser resistens overfor behandling med andre kjemoterapeutiske midler, innbefattet Taxol (se Bolag, D. M., et al., "Epothilones, a new class of microtubule-stabilizing agents with a Taxol-like mechanism of action", Cancer Research 55,2325-33 (1995)), og c) Det kunne vises at de fortsatt er svært effektive in vitro mot en 'Taxol"-resistent ovariekarsinomcellelinje med modifisert P-tubulin (se Kowalski, R.J., et al., J.
Biol. Chem. 272(4)M 2534-2541 (1997)).
Farmasøytisk anvendelse av epotilonene, for eksempel for tumorbehandlmg, er mulig på analog måte til den beskrevet for Taxol, se for eksempel US 5.641.802; US 5.496.804; US 5.565.478.
Oppfinnelsen gjelder en fremgangsmåte for separasjon av epotiloner, nærmere bestemt epotilon A og B, fra hverandre, som særpreges ved kromatografi på en reversfase-kolonne med et elueringsmiddel som omfatter et lavere alkylcyanid.
De generelle begreper som benyttes heri ovenfor og nedenfor har fortrinnsvis betydningene gitt nedenfor: Når det heri ovenfor eller nedenfor vises til dokumenter, inkorporeres disse heri i den grad det er nødvendig.
Forstavelsen "lavere" angir alltid at det således betegnede radikal inneholdes fortrinnsvis opp til maksimum 7 karbonatomer, mer foretrukket opp til 4 karbonatomer, og er forgrenet eller uforgrenet. Lavere alkyl kan for eksempel være uforgrenet eller forgrenet en eller flere ganger og er f. eks. metyl, etyl, propyl som isopropyl eller n-propyl, butyl som isobutyl, sec.-butyl, tert.-butyl eller n-butyl, eller også pentyl, for eksempel amyl eller n-pentyl.
Et dyrkningsmedium for bioteknologisk fremstilling av epotiloner som inneholder mikroorganismer som produserer disse forbindelser, særlig myksobakterier, som produsenter av naturlige forbindelser er primært er medium som omfatter en tilsvarende (naturlig forekommende, eller også er holdbar ved dyrking eller, fortrinnsvis, ved genetisk modifisering) mikroorganisme, nærmere bestemt en myksobakteriestamme som kan produsere disse forbindelser, fortrinnsvis en myksobakterie fra slekten Sorangium, ytterligere foretrukket (særlig for epotilonproduksjon) en mikroorganisme av typen Sorangium Cellulosum Soce90 (se WO 93/10121) eller et biomateriale avledet fra denne (for eksempel ved mutagenese eller rekombinant genteknologi) eller fra deler av denne myksobakterie, fortrinnsvis en tilsvarende avledet stamme, spesielt foretrukket stammen med referansebetegnelsen BCE33/10 eller mutanter av denne, og i tillegg, sammen med vann, fortrinnsvis andre konvensjonelle og nødvendige bestanddeler av dyrkningsmedier som biopolymerer, sukker, aminosyrer, salter, nukleinsyrer, vitaminer, antibiotika, semiokjemikalier, dyrkningsmedier, ekstrakter fra biomaterialer som gjær eller andre celleekstrakter, soyamel, stivelse, for eksempel potetstivelse, og/eller spormetaller, for eksempel jemjoner i kompleksbundet form, eller egnede kombinasjoner av alle eller noen av disse bestanddeler og/eller også analoge tilsetninger. Det tilsvarende dyrkningsmedium er kjent blant fagfolk eller kan fremstilles ved kjente prosesser (se for eksempel dyrkningsmediene i eksemplene i den foreliggende beskrivelse eller i WO 93/10121).
En foretrukket myksobakterie er en stamme selektert ved UV-mutagenese og seleksjon for økt dannelse av epotilon A og/eller B sammenlignet med Sorangium cellulosum Soce90 som er deponert hos DSM under aksesjonsnummeret 6773, særlig mutanten BCE33/10, som ble deponert under aksesjonsbetegnelsen DSM 11999 den 9. Februar 1998 hos Den Tyske Samling av Mikroorganismer og Celleculturer (DSMZ, Braunschweig, Tyskland).
Dyrking og morfologisk beskrivelse av stamme BCE 33/10: Stammen kan dyrke med cellulose som eneste karbon- og energikilde med kaliumnitrat som eneste nitrogenkilde f.eks. på filterpapir (ST21 mineralsaltagar (0.1% KN03; 0.1% MgS04 x 7 H20; 0.1% CaCl2 x 2 H20; 0.1% K2HP04; 0.01% MnS04 x 7 H20; 0.02% FeCl3; 0.002% gjærekstrakt, standard spormetalløsmng, 1% agar). På dette medium dannes mørkt rødbrune til svartbrune fruktlegemer. Disse består av små sporangioler (tilnærmet 15 til 30 um diameter) og foreligger i tette hoper og klumper av varierende størrelse.
Den vegetative basillus har form som er typisk for Sorangium (relativt kompakt, under fasekontrastmikroskop en mørk, sylindrisk basillus med brede, avrundede ender, gjennomsnittlig 3 til 6 um lang og 1 um tykk).
Epotiloner er hovedsakelig epotilon A og/eller 6, men også andre epotiloner, for eksempel epotilon C og D, navngitt i internasjonal pantentsøknad WO 97/19086 og WO 98/22461, epotilon E og F navngitt i WO 98/22461, og ytterligere epotiloner tilgjengelige fra tilsvarende mikroorganismer.
En vannløselig, kompleksdannende bestanddel er hovedsakelig et vannløselig oligo-eller polypeptidderivat eller, fortrinnsvis, et oligo- eller polysakkairdderivat med cyklisk eller helisk struktur som danner et intramolekylært hulrom, som grunnet sine tilstrekkelig hydrofobe egenskaper er i stand til å binde epotiloner, særlig epotilon A og/eller epotilon B. En vannløselig, kompleksdannende bestanddel som foretrekkes spesielt er en som er utvalgt blant cyklodekstriner eller (fortrinnsvis) cyklodekstrin-derivater eller blandinger av disse.
I den påfølgende beskrivelse av opparbeidelsen og rensingen er epotilon ment å være et epotilon som kan erholdes fra den tilsvarende mikroorganisme, fortrinnsvis epotilon C, D, E, F, eller mer foretrukket A, eller spesielt foretrukket epotilon B. Dersom ikke annet er angitt når "epotiloner" nevnes er dette ment å være et generelt begrep som omfatter enkeltepotiloner eller blandinger av disse.
Opparbeidelse av epotilonene oppnås ved konvensjonelle fremgangsmåter, først ved å skille en kultur i en væskefase (et sentrifugat eller filtrat) og en fastfase (celler) ved hjelp av filtrering eller sentrifugering (rørsentrifuge eller separator). Den del eller hoveddel av epotilonene som foreligger i sentrifugatet eller filtratet blandes så direkte med et syntetisk resin, for eksempel et resin basert på polystyren som støttemedium (i det påfølgende også for enkelthets skyld betegnet et polysytrenresin), for eksempel Amberlite XAD-16 [Rohm & Haas Germany GmbH, Frankfurt] eller Diaion HP-20 [Resindion S.R.L., Mitsubishi Chemical Co., Milano] (fortrinnsvis med et forhold mellom sentrifugat og resin på tilnærmet 10:1 til 100:1 (vol/vol), fortrinnsvis tilnærmet 50:1 (vol/vol)). Etter et kontakttidsrom på fortrinnsvis 0.25 til 50 timer, mer foretrukket 0.8 til 22 timer, separeres resinet, for eksempel ved filtrering eller sentrifugering. Etter adsorpsjonen vaskes resinet om nødvendig med et kraftig polart løsemiddel, fortrinnsvis med vann. Desorpsjon av epotilonene oppnås så med et egnet løsemiddel, fortrinnsvis med en alkohol, mest foretrukket isopropanol. Væskefasen, fortrinnsvis isopropanolfasen, fjernes så fira løsemiddelet, fortrinnsvis ved hjelp av forutgående, samtidig eller påfølgende tilsetning av vann, fortrinnsvis i en sirkulasjonsevaporator, hvorved væskefasen konsentreres om nødvendig, og den resulterende vannfase ekstraheres med et løsemiddel som er egnet for dannelse av en ny fase, for eksempel en ester, for eksempel et lavere alkanol-lavere alkanoat, typisk etylacetat eller isopropylacetat. Epotilonene overføres ved dette til den organiske fase. Deretter konsentreres den organiske fase etter behov, fortrinnsvis til tørrhet, for eksempel ved benyttelse av en rotasjonsevaporator.
Etter dette foregår den videre prosessering ved benyttelse av følgende trinn, hvori rensetrinnet ved hjelp av revers-fasekromatografi med evaluering med et nitril er det oppfinneriske trinn og således obligatorisk, mens de andre trinn er valgfrie: molekylifltrering (gelkromatografi), f.eks. på en kolonne med materiale som
Sephadex LH-20 (Pharmacia, Uppsala, Sverige) med en alkohol, for eksempel metanol, som elueringsmiddel,
separasjon av epotilonene ved revers-fasekromatografi etter at de er løst i et egnet løsemiddel, og eluering med en blanding av nitril/vann (obligatorisk), fortrinnsvis karakterisert ved at kromatografien utføres på en kolonne med materiale, fortrinnsvis et RP-18-materiale, som er ladet med hydrokarbonkjeder, for eksempel hydrokarbonskjeder med 18 karbonatomer, og et elueringsmiddel som omfatter et nitril, fortrinnsvis et lavere alkylnitril, mest foretrukket acetonitril, benyttes, fortrinnsvis benyttes en blanding av nitril og vann, mest foretrukket en blanding av acetonitril og vann, fortrinnsvis med et forhold mellom nitril og vann på tilnærmet 1:99 til 99:1, mer foretrukket mellom 1:9 og 9:1, f.eks. mellom 2:8 og 7:3, f.eks. 3:7 eller 4:6.

adsorpsjonskromatografi, fortrinnsvis ved tilsetning til en kieselgelkolonne og elusjon med et egnet løsemiddel eller en løsemiddelblanding, fortrinnsvis en blanding av ester og hydrokarbon, for eksempel lavere alkylalkanoat/C4-Cio-alkan, fortrinnsvis etyl- eller isopropylacetat/n-heksan, i hvilken forholdet mellom esteren
og hydrokarbonet fortrinnsvis ligger i området 99:1 til 1:99, fortrinnsvis 10:1 til 1:10, for eksempel 4:1,
oppløsning av restmaterialet som kan erholdes etter konsentrering i et egnet
løsemiddel, for eksempel en alkohol som metanol,
sammenblanding med aktivt karbon og fjerning av dette,,
rekrystallisering, f.eks. fra egnede løsemidler eller løsemiddelblandinger som for eksempel består av estere, ester/hydrokarbonblandinger eller alkoholer, fortrinnsvis etyl- eller isopropylacetat: toluen 1:10 til 10:1, fortrinnsvis 2:3 (epotilon A) eller metanol eller etylacetat (epotilon 8),
hvorved de resulterende løsninger eller suspensjoner konsentreres mellom hvert benyttede trinn om nødvendig og/eller væskekomponenter og faste komponenter skilles fra hverandre, fortrinnsvis ved filtrering eller sentrifugering av løsninger/suspensjoner. De mer presise definisjoner som nevnes nedenfor kan fortrinnsvis benyttes i enkelt-trinnene ovenfor.
Opparbeidelse og rensing utføres fortrinnsvis som følger: Etter høsting separeres en kultur i væskefase (sentrifugat) og fastfase (celler) ved hjelp av sentrifugering (rørsentrifuge eller separator). Hovedmengden av epotilonene foreligger i sentrifugatet, som så direkte blandes med et polystyrenresin, for eksempel Amberlite XAD-16 [Rohm & Haas Germany GmbH, Frankfurt] eller Diaion HP-20 [Resindion S.R.L., Mitsubishi Chemical Co., Milano] (fortrinnsvis med et forhold mellom sentrifugat og resin på tilnærmet 10:1 til 100:1 (vol/vol), fortrinnsvis tilnærmet 50: (vol/vol)) og omrøres i en rystemaskin. I dette trinn overføres epotilonene fra cyklodekstrinet til resinet. Etter et kontakttidsrom på tilnærmet 1 time separeres resinet ved sentrifugering eller filtrering. Adsorpsjon av epotilonene til resinet kan også oppnås i en kromatografisøyle ved å plassere resinet i søylen og la sentrifugatet renne gjennom resinet. Etter adsorpsjon vaskes resinet med vann. Desorpsjon av epotilonene fra resinet oppnås med isopropanol. Isopropanol fjernes så fra isopropanolfasen, fortrinnsvis ved tilsetning av vann, fortrinnsvis i en sirkulasjonevaporator, og den resulterende vannfase ekstraheres med etylacetat. Epotilonene overføres fira vannfasen til etylacetatfasen. Så konsentreres etylacetatekstraktet til tørrhet ved benyttelse av en rotasjonsevaporator. En utgangs-konsentrasjon av epotilonene oppnås så ved hjelp av molekylfiltrering (f.eks. Sephadex LH-20 [Pharmacia, Uppsala, Sverige] med metanol som elueringsmiddel). Toppfraksjonene fra molekylfiltreringen inneholder epotilon A og B og har et totalinnhold av epotiloner på > 10%. Separasjon av disse topp fraksjoner, som inneholder epotilon A og B i blanding, i enkeltbestanddelene følger så ved hjelp av kromatografi på en "reversfase", f.eks. en RP-18-fase (en fase som er modifisert med alkylradikaler med 18 karbonatomer pr. kjede) med et egnet elueringsmiddel, fortrinnsvis et som inneholder et nitril som for eksempel acetonitril (dette gir bedre separasjon enn for eksempel alkoholer som metanol). Epotilon A elueres før epotilon B. Toppfraksjonene med epotilon B kan fortsatt inneholde små mengder epotilon A, som kan fjernes ved ytterligere separasjon på RP-18. Endelig krystalliseres epotilon A-fraksjonen direkte fra etylacetat: Toluen~2:3, og epotilon B-fraksjonen fra metanol eller etylacetat.
Oppfinnelsen gjelder en fremgangsmåte for separasjon av epotiloner, fortrinnsvis epotilon A og B, fra hverandre, som særpreges ved kromatografi på en revers-fasekolonne med et elueringsmiddel som inneholder et lavere alkylcyanid, hvor kromatografien utføres på et kolonnemateriale, fortrinnsvis et RP-18-materiale, som er ladet med hydrokarbonkjeder, for eksempel kjeder med 18 karbonatomer, og benyttelse av et elueringsmiddel som inneholder et nitril, fortrinnsvis et lavere alkylnitril, mer foretrukket acetonitril, fortrinnsvis en blanding av nitril og vann, mer foretrukket en blanding av acetonitril og vann, fortrinnsvis med et forhold mellom nitril og vann på tilnærmet 1:99 til 99:1, mer foretrukket 1:9 til 9:1, f.eks. mellom 2:8 og 7:3, f.eks. 3:7 eller 4:6. Denne separasjon kan følge en filtrering med en molekylsikt eller utføres fortrinnsvis direkte ved benyttelse av restmaterialet etter adsorpsjon av epotilonene fra dyrkningsmediet som inneholder en kompleksdannende bestanddel til resinet. En fordel ved separasjon med et elueringsmiddel som inneholder et lavere alkylcyanid fremfor benyttelse av alkoholer, for eksempel metanol, er bedre separasjon av epotilon A og B.
Det påfølgende eksempel skal bidra til å illustrere oppfinnelsen uten å begrense dens område.
Forsiktig: Ved håndtering av epotiloner må egnede beskyttelsestiltak iverksettes etter behov, deres høye giftighet tas i betraktning.
Formentoren på 750 liter som ble benyttet i det påfølgende er en fermentor av raffinert stål fra firmaet Alpha AG, Nidau,Sveits.
Eksempel: Opparbeidelse av epotilonene: Isolering fra en hovedkultur på
500 liter:
Hovedkultur: 500 liter fermentering med 1% 2-(hydroksypropyl)-p-
cyklodekstrin): Fermenteringen utføres i en 750 liter fermentor. Mediet inneholder 10 g/l 2-(Hydroksypropyl)-(J-cyklodekstrin. Forløpet av fermenteringen er vist i tabell. Kulturen høstes etter 7 dager og opparbeides.
Høstningsvolumet fra hovedkulturen på 500 liter over er på 450 liter og det separeres ved benyttelse en Westfalia klarningsseparator Type S A-20-06 (rpm = 6500) i væskefase (sentrifugat + vaskevann = 650 liter) og fastfase (celler = tilnærmet 15 kg). Hovedmengden av epotiloner foreligger i sentrifugatet. Den nedsentrifugerte cellemasse inneholder <15% av det målte epotiloninnhold og prosesseres ikke videre. Sentrifugatet på 650 liter plasseres så i rørekar på 4000 liter, blandes med 10 liter Amberlite XAD-16 (sentrifugatrresinvolum = 65:1) og omrøres. Etter et kontakttidsrom på tilnærmet 2 timer frasentrifugeres resinet i en Heine overstrømssentrifuge (beholdervolum 40 liter,
rpm = 2800). Resinet fjernes fra sentrifugen og vaskes med 10-15 liter deionisert vann. Desorpsjon oppnås ved å omrøre resinet to ganger, hver gang med 30 liter isopropanol i rørekar på 30 liter av glass i 30 minutter. Separasjon av isopropanolfasen fra resinet finner sted ved benyttelse av et sugefilter. Isopropanol fjernes så fra de sammenslåtte isopropanolekstrakter ved tilsetning av 15-20 liter vann i en sirkulasjonsevaporator under vakuum (Schmid-Verdampfer), og den resulterende vannfase på tilnærmet 10 liter ekstraheres tre ganger, hver gang med 10 liter etylacetat. Ekstraksjonen utføres i rørekar
av glass på 30 liter. Etylacetatekstraktet konsentreres til 3-5 liter i en sirkulasjonsevaporator under vakuum (Schmid-Verdampfer), og konsentreres deretter til tørrhet i rotasjonsevaporator (Btichi-type) under vakuum. Resultatet er et etylacetatekstrakt på 50-2 g. Etylacetatekstraktet løses i 500 ml metanol, den uløselige del frafiltreres ved benyttelse av et foldefilter, og løsningen påsettes en 10 kg Sephadex LH 20-kolonne (Pharmacia, Uppsala, Sverige) (kolonnediameter 20 cm, høyde tilnærmet 1.2 m). Elueringen utføres med metanol som elueringsmiddel. Epotilon A og B foreligger hovedsakelig i fraksjonene 21-23 (med en fraksjonsstørrelse på 1 liter). Disse fraksjoner konsentreres til tørrhet under vakuum i en rotasjonsevaporator (totalvekt 9.0 g). Disse toppfraksjoner fra Sephadex (9.0 g) oppløses så i 92 ml acetonitril:-vann:-metylenklorid - 50:40:2, løsningen filtreres gjennom et foldefilter og påsettes en RP-kolonne (utstyr: Prepbar 200, Merck, 2.0 kg LiChrospher RP-18 Merck, partikkelstørrelse 12 um kolonnediameter 10 cm, høyde 42 cm, Merck, Darmstadt, Tyskland). Elueringen utføres med acetonitril: vann = 3:7 (gjennomstrømningshastighet ~ 500 ml/min., retensjonstid for epotilon A = tilnærmet 51-59 minutter, retensjonstid for epotilon B = ca. 60-69 minutter). Fraksjoneringen følges med en UV-detektor ved 250 nm. Fraksjonene konsentreres til tørrhet under vakuum i en Buchi-Rotavapor rotasjonsevaporator. Vekten av toppfraksjonen av epotilon A er 700 mg, og ifølge HPLC (extern standard) er innholdet 75.1%. Toppfraksjonen for epotilon B veier 1980 mg, og innholdet ifølge HPLC (ekstern standard) er 86.6%.

Fremgangsmåte for separasjon av epotiloner fira hverandre, fortrinnsvis epotilon A og B, karakterisert ved at den omfatter kromatografi på en reversefasekolonne med et elueringsmiddel som inneholder et lavere alkylcyanid.
2.
Fremgangsmåte ifølge krav 1, karakterisert ved at det benyttes et kolonnemateriale som er ladet med hydrokarbonkjeder som inneholder IS karbonatomer og at elueringsmiddelet som benyttes er en blanding av vann og acetonitril.
