Denne oppfinnelse vedrører forbindelsen N-[4-(3-klor-4-fluor-fenylamino)-7-(3-morfolin-4-yl-propoksy)-kinazonlin-6-yl]-akrylamid som er en irreversibel inhibitor for tyrosin-kinaser. Denne oppfinnelsen vedrører også anvendelse av denne forbindelsen til fremstilling av et farmasøytisk preparat for behandling av kreft, aterosklerose, restenose, endometriose og psoriasis og en farmasøytisk blanding som inneholder forbindelsen N-[4-(3-klor-4-fluor-fenylamino)-7-(3-morfolin-4-yl-propoksy)-kinazonlin-6-yl]-akrylamid.
Bakgrunn for oppfinnelsen
Kreft har vært ansett som en sykdom i det intracellulære signalsystem eller signaltransduksjonsmekanisme. Celler får instruksjoner fra mange ekstracellulæ-re kilder som instruerer dem til enten å formere seg eller ikke formere seg. Hen-sikten med signaltransduksjonssystemet er å motta disse og andre signaler på celleoverflaten, få dem inn i cellen og deretter føre signalene videre til kjernen, cytoskjellettet, og transport og proteinsyntese maskineriet.
Den vanligste årsak til kreft er en rekke defekter, enten i disse proteiner når de muteres, eller i reguleringen av mengden av protein i cellen slik at det produse-res for mye eller for lite. Noen ganger kan cellen starte å produsere en autentisk vekstfaktor for sine egne reseptorer når den ikke skulle, den såkalte autokrine sløyfemekanisme. Mutasjoner på celleoverflatereseptorene, som normalt signaliserer inn i cellen ved hjelp av tyrosin-kinaser, kan føre til aktivering av kinasen i fravær av ligande, og føre et signal videre som i virkeligheten ikke er der. Alternativt kan mange overfla-te kinaser overuttrykkes på celleoverflaten, som fører til utilsiktet sterk respons på et svakt signal. Det er mange nivåer inne i cellen ved hvilke mutasjon eller overekspresjon kan føre til de samme falske signal i cellen, og det er mange andre typer signaldefekter som er involvert i kreft. Denne oppfinnelsen berører kreft som drives av de tre nettopp beskrevne mekanismer, og som involverer celleoverflate-reseptorer av den epidermale vektsfaktorreseptor tyrosin-kinasefamilie (EGFR). Denne familie består av EGF reseptoren (også kjent som erbB1), erbB2 reseptoren, og dens konstitutive aktive onkoprotein mutant Neu, erbB3 reseptoren, og erbB4 reseptoren. I tillegg kan andre biologiske prosesser som drives gjennom medlemmer av EGF familien av reseptorer også behandles med forbindelser iføl-ge oppfinnelsen beskrevet nedenunder.
EGFR har sine to viktigste ligander epidermal vekstfaktor (EGF) og Transformeringsvekstfaktor alpha (TGFalpha). Reseptorene synes bare å ha små funk-sjoner hos voksne mennesker, men er åpenbart involvert i sykdomsprosessen ved en stor del av alle kreftformer, spesielt tykktarm- og brystkreft. De nær beslektede erbB2 (HER2), erbB3 (HER3), og erbB4 (HER4) har en familie av Hereguliner som sine hovedligander, og reseptor-overekspresjon og mutanter er entydig blitt påvist som hovedrisikofaktoren hos brystkreft med dårlig prognose. I tillegg har det blitt demonstrert at alle fire medlemmer av denne familien av reseptor kan danne heterodimere signalkomplekser med andre medlemmer av familien, og at dette kan føre til synergistisk transformeringskapasitet hvis mer enn ett medlem av familien overuttrykkes i en ondartet kreft. Overekspresjon av flere enn ett familie-medlem har vært vist å være relativt vanlig i ondartet kreft hos mennesker.
I tillegg til kreft er restenose også en sykdom hvori uønsket cellulær formering opptrer. Restenose medfører formering av vaskulære, glatte muskelceller. Restenose er et klinisk hovedproblem i forbindelse med koronær angioplasti og andre medisinske fremgangsmåter. Restenose opptrer generelt innenfor 0 til 6 måneder hos ca. 30% til 50% av pasienter som gjennomgår ballong-angioplasti for å klare tilstoppede koronære arterier i et forsøk på å behandle hjertesykdom som skyldes tilstoppede arterier. Den resulterende restenose forårsaker betydelig pasient-morbiditet og helsevern-omkostninger.
Prosessen restenose igangsettes ved skade på blodkaret, innbefattende arterier og vener, med den etterfølgende frigjøring av trombogene, vasoaktive og mitogene faktorer. Endothelial og dyp-kar skade fører til blodplate-aggregering, trombedannelse, inflammasjon og aktivering av makrofage og glattmuskelceller. Disse hendelser induserer produksjon av og frigjøring av vekstfaktorer og cytoki-ner, som igjen kan bevirke sin egen syntese og frigjøring fra målceller. Således igangsettes en selvdrivende prosess som involvever vekstfaktorer så som EGF, blodplateavledet vekstfaktor (PDGF) eller fibroblastvekstfaktor (FGFer). Således vil det være nyttig å ha irreversible inhibitorer for signaltransduksjons-veier, spesielt av tyrosin-kinaser som EGF, PDGF, FGF, eller src tyronin kinaser.
Den proliferatiye hudsykdom psoriasis har for tiden ingen god kur. Den behandles ofte med antikreftmidlet så som methotrexat, som har meget alvorlige bivirkninger, og som ikke er meget effektiv, da toksisiteten begrenser doser som skal brukes. Det antas at TGF alpha er hovedvekstfaktoren som overproduseres i psoriasis, da 50% av transgene mus som overuttrykker TGF alpha utviklet psoriasis. Dette tyder på at en god inhibitor for EGFR signalisering kunne anvendes som antipsoriasis-middel, fortrinnsvis men ikke nødvendigvis, ved topisk dosering.
Det er spesielt fordelaktig å ha irreversible tyrosin-kinaseinhibitorer sammenlignet med reversible inhibitorer, fordi irreversible inhibitorer kan anvendes i forlenget undertrykkelse av tyrosin-kinasen, bare begrenset av den normale has-tighet av reseptor resyntese, også kalt turnover.

Detaljert beskrivelse av oppfinnelsen
Den foreliggende oppfinnelse tilveiebringer forbindelsen N-[4-(3-klor-4-fluor-fenylamino)-7-(3-morfolin-4-yl-propoksy)-kinazonlin-6-yl]-akrylamid (Forbindelse 1), eller de farmasøytisk akseptable salter derav.
Forbindelse 1 er en irreversibel inhibitor av tyrosin-kinaser, spesielt EGFR tyrosin-kinase. Andre tyrosin-kinaser som kan inhiberes med Forbindelse 1 omfatter FGFR, PDGFR, c-src, erbB2 og erbB4. En terapeutisk effektiv mengde av Forbindelse 1 kan administreres til en pasient med kreft, eller en pasient med restenose eller med risiko for å få restenose, eller en pasient med psoriasis, aterosklerose eller med risiko for å få aterosklerose eller endometriose. Pasienter med behov for inhibering av tyrosin fosforylering av erbB3 er pasienter som har eller risikerer å få sykdommer nevnt heri i forbindelse med inhiberingen av EGFR og inhiberingen av VEGF-sekresjon.
Forbindelsel kan administreres til mennesker og dyr enten oralt, rektalt, parenteralt (intravenøst, intramuskulært eller subkutant), intracistemalt, intravaginalt, intraperitonealt, intravesikalt, lokalt, (pulvere, salver eller dråper) eller som en bukal eller nesespray. Forbindelsene kan administreres alene eller som en del av en farmasøytisk akseptabel blanding som omfatter farmasøytisk akseptable eksipienter.
Blandinger egnet for parenteral injeksjon kan innbefatte fysiologisk akseptable, sterile, vandige eller ikke-vandige løsninger, dispersjoner, suspensjoner eller emulsjoner og sterile pulvere for rekonstrtusjon i sterile injiserbare løsninger eller dispersjoner. Eksempler på egnede vandige og ikke-vandige bærere og fortynningsmidler, løsningsmidler eller bærere omfatter vann, etanol, polyoler (propy-lenglykol, polyetylenglykol, glycerol og lignende), egnede blandinger derav, vege-tabilske oljer (så som olivenolje) og injiserbare organiske estere så som etyloleat. Riktig flyteevne kan opprettholdes, for eksempel ved bruk av et belegg så som lecithin, eller ved opprettholdelse av den nødvendige partikkelstørrelse i dette tilfellet dispersjoner og ved anvendelse av overflateaktive midler.
Disse blandinger kan også inneholde hjelpemidler så som preserverings-, fukte-, emulgerings- og dispergeringsmidler. Forhindring av aktivitet av mikro-organismer kan sikres ved forskjellige antibakterielle og antisopp-midler, for eksempel parabener, klorbutanol, fenol, sorbinsyre og lignende. Det kan også være ønskelig å innta isotone midler, for eksempel sukkere, natriumklorid og lignende. Forlenget absorbsjon av den injiserbare farmasøytiske form kan tilveiebringes ved bruk av midler som forsinker absorbsjon, for eksempel aluminium-monostearat of gelatin.
Faste doseringsformer for oral administrasjon omfatter kapsler, tabletter, piller, pulvere og granuler. I tilfellet kapsler, tabletter og piller, kan doseringsformene også omfatte buffermidler.
Faste blandinger av en lignende type kan også anvendes som fyllmidler i fyllte myk- og hard-gelatinkapsler ved bruk av slike eksipienter som laktose eller melkesukker, så vel som høymolekylære polyetylenglykoler og lignende.
Faste doseringsformer slik som tabletter, drageer, kapsler, piller og granuler kan fremstilles med belegg og skall, så som enteriske belegg og andre som er velkjente på området. De kan inneholde opasiterende midler, og kan også være av slik sammensetning at de frigjør den aktive forbindelse i en viss del av fordøy-elseskanalen på en forsinket måte. Eksempler på innstøpte blandinger som kan brukes er polymersubstanser og vokser. Den aktive forbindelse kan også foreligge i mikroinnkapslet form om nødvendig med én eller flere av de ovenfor nevnte eksipienter.
Flytende doseringsformer for oral administrering omfatter farmasøytisk akseptable emulsjoner, løsninger, suspensjoner, siruper og eliksirer. I tillegg til den aktive forbindelse kan de flytende doseringsformer inneholde inerte fortynningsmidler som er vanlige å bruke på området så som vann eller andre løsningsmid-ler, solubiliseringsmidler og emulgeringsmidler, som for eksempel etylalkohol, isopropylalkohol, etylkarbonat.etylacetat, benzylalkohol, benzylbenzoat, propylen-glykol, 1,3-butylenglykol, dimetylformamid, oljer, spesielt bomullsfrøolje, jordnøtt-olje, maisfrøolje, olivenolje, kastorolje og sesamolje, glycerol, tetrahydrofurfuryl-alkohol, polyetylenglykoler og fettsyreestere av sorbitan eller blandinger av disse substanser eller lignende.
Ved siden av slike inerte fortynningsmidler kan blandingen også inneholde hjelpestoffer så som f uktemidler, emulgerings- og oppslemmingsmidler, søtnings-midler, smaksmidler og parfymeringsmidler.
Suspensjoner kan i tillegg til den aktive forbindelse inneholde oppslemmingsmidler som for eksempel etoksylerte isostearylalkoholer, polyoksyetylen-sorbitol og sorbitan-estere, mikrokrystallinsk cellulose, aluminium-metahydroksyd, bentonitt, agar-agar og tragakant, eller blandinger av slike substanser og lignende.
Blandinger for rektal administrering er fortrinnsvis suppositorer som kan fremstilles ved å blande forbindelsene ifølge foreliggende oppfinnelse med egnede, ikke-irriterende eksipienter eller bærere så som kokossmør, polyetylenglykol eller en suppositorievoks, som er faste ved vanlig temperatur, men flytende ved legemstemperatur og derfor smelter i rektum eller vaginalrommet og frigjør den aktive forbindelse.
Doseringsformer for topisk administrering omfatter salver, pulvere, sprayer og inhalasjoner. Den aktive forbindelse blandes under sterile betingelser med en fysiologisk akseptabel bærer og andre preserveringsmidler, buffere eller driv-midler etter behov. Oftalmiske formuleringer, øyesalver, pulvere og løsninger ligger også innenfor omfanget av denne oppfinnelsen.
Uttrykket "farmasøytisk akseptable salter, estere, amider og prodrugs" slik de heri brukes, viser til slike karboksylatsalter, aminosyreaddisjonssalter, estere, amider og prodrugs av forbindelsene ifølge foreliggende oppfinnelse som er innenfor omfanget av sunn medisinsk bedømmelse egnet for bruk i kontakt med vevene til pasienten uten uakseptabel toksisitet, irritasjon, allergisk reaksjon og lignende, i overensstemmelse med et rimelig gevinst/risk-forhold, og effektiv for den tilsiktede bruk, samt Zwitterionformene, hvor mulig, av forbindelsene ifølge oppfinnelsen. Uttrykket "salter" viser til de relativt ikke-toksiske, uorganiske og organiske syreaddisjonssalter av forbindelsene ifølge foreliggende oppfinnelse. Disse salter kan fremstilles in situ under den endelige isolering og rensing av forbindelsen eller ved separat å omsette den rensede forbindelse i sin fri baseform med en egnet organisk eller uorganisk syre og isolere det således dannede salt. Representative salter omfatter hydrobromidet, hydrokloridet, sulfatet, bisulfatet, nitratet, avcetatet, oksalatet, valeratet, oleatet, palmitatet, stearatet, lauratet, bora-tet, benzoatet, laktatet, fosfatet, tosylatet, citratet, maleatet, fumaratet, suksinatet, tartratet, naftylatet, mesylatet, glukoheptonatet, laktobionatet og laurylsulfonatsal-tene, og lignende. Disse kan omfatte kationer basert på alkali- og jordalkali-metallene, så som natrium, litium, kalium, kalsium, magnesium og lignende, samt ikke-toksiske ammonium, kvatemære ammonium og aminkationer, omfattende, men ikke begrenset til ammonium, tetrametylammonium, tetraetyl-ammonium, metylamin, dimetylamin, trimetylamin, trietylamin, etylamin og lignende (se for eksempel S.M. Eksempler på farmasøytisk akseptable, ikke-toksiske estere av forbindelsene ifølge denne oppfinnelsen omfatter Ci-C6-alkylestere hvori alkylgruppen er en rett eller forgrenet kjede. Akseptable estere innebefatter også C5-C7-cyklo-alkylestere samt arylalkylestere, så som, men ikke begrenset til, benzyl, C1-C4-alkylestere er foretrukket. Estere av forbindelsen ifølge foreliggende oppfinnelse kan fremstilles i henhold til vanlige metoder.
Eksempler på farmasøytisk akseptable, ikke-toksiske amider av forbindelsen ifølge oppfinnelsen omfatter amider som stammer fra ammoniakk, primære CrCe-alkylaminer og sekundære Ci-C6-dialkylaminer, hvori alkylgruppene er rette eller forgrenede. I tilfellet sekundære aminer, kan aminet også foreligge i form av en 5- eller 6-leddet heterocyklus inneholdende ett nitrogenatom. Amider avledet fra ammoniakk, primære Ci-C3-alkylaminer og sekundære Ci-C2-dialkylaminer er foretrukket. Amider av forbindelsen ifølge oppfinnelsen kan fremstilles i henhold til vanlige metoder.
Uttrykket "prodrug" viser til forbindelser som raskt overføres in vitro og gir stamforbindelsen med formelen ovenfor, for eksempel ved hydrolyse i blod. En grundig diskusjon er gitt i T. For et normalt, vok-sent menneske med en legemsvekt på ca. 70 kg er en dosering i området ca. 0,01 til ca. 100 mg pr. kg kroppsvekt tilstrekkelig. Den spesifikke anvendte dose kan imidlertid variere. Pulverisert 4-klor-7-fluor-6-nitrokinazonltn ( J- Med. Chem., 1996; 39:918 (82,77 g, 363,7 mmol) ble satsvis tilsatt gjennom 10 minutter til en meka-nisk rørt løsning av 3-klor-4-fluoranilin (53,183 g, 365,3 mmol) og N,N-dimetylanilin (88,5 g, 730 mmol) i ispopropanol (1,09 I) holdt under nitrogen på et isbad. Isobutylklorformiat (27,2 g, 0,20 mol) ble tilsatt dråpevis over 10 min. til en løsning av akrylsyre (14,40 g, 0,20 mol) og trietylamin (40,48 g, 0,40 mol) i THF (800 ml) rørt under nitrogen ved 0°C. Etter 10 min. til ble den hvite oppslemming overført til et -25°C kjølebad og rørt under nitrogen i 20 min. 6-amino-[(3-klor-4-fluorfenyl)amino]-7-[3-(4-morfolino)propoksy]kinazonlin (43,19 g, 0,100 mol) i THF (500 ml) ble tilsatt dråpevis over 1,33 timer. A431 humane epidermoide karcinomaceller og MDA-MB-453-celler erholdtes fra the American Type Culture Collection, Rockville, MD og ble holdt som mo-nosjikt i dMEM (Dulbecco's modified eagle medium)/F12, 50:50 (Gibco(BRL, Bethesda, MD) inneholdende 10% føtalt bovinserum. Cellene ble dyrket ved 37°C i en fuktet atmosfære inneholdende 5% C02 i luft.
Rensing av e<p>idermal vekstfaktorreseptortvrosin- kinase
Human EGF reseptortyrosin-kinase (EGFR) ble isolert fra A431 humane epidermoide karcinomaceller ved den følgende metode. Celler ble dyrket i fulle flasker i dMEM/F12 media (Gibco/BRL, Bethesda, MD) inneholdende 10% føtalt kalveserum. Ca. 10<9> celler ble lyset i 2 volumer buffer inneholdende 20 mM Hepes, pH 7,4, 5 mM EGTA, 1% Triton X-100, 10% glyserol, 0,1 mM natriumortova-nadat, 5 mM natriumfluorid, 4 mM pyrofosfat, 4 mM benzaid, 1 mM DTT, 80 ug/ml aprotinin, 40 ug/ml leupeptin, og 1 mM fenylmetylsulfonylfluorid (PMSF). Enzymmålinger for IC5o-bestemmelser ble utført i 96 brønns filterplater (Millipore MADVN6550, Millipore, Bedford, MA). Totalvolumet var 0,1 ml inneholdende 20 mM Hepes, pH 7,4, 50 uM natriumvanadat, 40 mM magnesiumklorid, 10 uM ATP inneholdende 0,5 uCi [<32>0]ATP, 20 ug poly Glutaminsyre/tyrosin (Sigma Chemical Co., St. Louis, MO), 10 ng EGF reseptortyrosin-kinase, og tilsvarende fortynninger av inhibitor Alle komponenter unntatt ATP settes til brønnen og platen inkuberes under risting i 10 min. ved 25°C. Reaksjonen startes ved tilsetning av [^PjATP, og platen inkuberes ved 25°C i 10 min.. Reaksjonen stoppes ved tilsetning av 0,1 ml 20% trikloreddiksyre (TCA). Platen holdes på 4°C i minst 15 min. for å la substratet utfelles. Brønnene vaskes så 5 ganger med 0,2 ml 10% TCA og <32>P innbygging bestemmes med en Wallac beta platefeller. Målinger som anven-der intracellulære kinaseområder av PDGF, FGF og insulinreseptorer, samt slike for c-src ble utført som beskrevet for EGF-reseptoren, unntatt at 10 mM mangan-klorid ble tatt med i reaksjonen.
EGF- oa Hereaulin- avhenqiqe tvrosinfosforvlerinasmålinaer
A431 humane epidermoide karcinomaceller eller MDA-MB-453-celler ble dyrket i 6-brønnsplater til ca. 80% sammenflytning og deretter inkubert i serumfrie media i 18 timer. Cellene ble utsatt for forskjellige konsentrasjoner av enten forbindelse 1 eller eksempel 21 i 2 timer og deretter stimulert med enten 100 ng/ml EGF (A431) eller 10 ng/ml heregulin (MDA-MB-453) i 5 min.. Celleekstrakter ble foretatt, og reduksjonen i fosfotyrosin ble bestemt ved western blot.
Western blot prosedyre
Ekstrakter ble foretatt ved å lyse monosjiktene i 0,2 ml av kokende Laemlli buffer (2% natriumdodecylsulfat, 5% beta-merkaptoetanol, 10% glyserol og 50 mM Tris, pH 6,8), og lysatene ble oppvarmet til 100°C i 5 min.. Proteiner i lysatet ble skilt ved polyakrylamidgelelektroforese og elektroforetisk overført til nitrocellu-lose. Membran ble vasket én gang i [10 mM Tris, pH 7,2,150 mM NaCI, 0,01% natriumazid] (TNA) og blokket natten over i TNA inneholdende 5% bovin serumal-bumin og 1% ovalbumin. Membranen ble blottet i 2 timer med antifosfotyrosin-antistoff (UBl, Lake Placid, NY, 1 ug/ml i blokkeringsbuffer) og deretter vasket to ganger i TNA, én gang i TNA inneholdende 0,05% Tween-20 og 0,05% nonidet P-40 og to ganger i TNA. Membranene ble deretter inkubert i 2 timer i blokkingsbuf-fer inneholdende 0,1 |iCi/ml [<125>l] protein A og deretter vasket igjen som ovenfor. Etter blottene var tørre, ble de satt i en filmkassett og eksponert mot X-AR x-røntgenfilm i 1 til 7 dager. Båndintensiteter ble bestemt med et laserdensitometer.
Dataen i tabell 1 viser at eksempel 21 og forbindelse 1 var omtrent like aktive mot renset EGF-reseptortyrosin-kinase, EGF-bevirket reseptor-autofosforylering, og heregulin-bevirket tyrosinfosforylering in vitro.
IN VIVO TUMOR INHIBERINGSMÅLING
Det humane epidermoide karcinoma, A431, ble propagert in vivo ved seriell transplantasjon. Denne tumormodell ble valgt for disse studier pga. sin kjente av-hengighet av EGF-reseptoren for vekst og i tidlig stadium respons på monoklonal antistoffterapi in vivo. I dette eksperimentet ble hårløse mus som veide 18-22 gram implantert subkutant med 30 mg A431 tumorfragmenter i området for høyre aksille på dag 0. Tumorer fikk vokse til en masse på 100 til 150 mg, på hvilket tidspunkt de tumorbærende dyr ble statistisk oppdelt og fordelt i behandlingsbur. Dyr ble behandlet peroralt i 15 påfølgende dager med isetionatsaltene fra eksempel 21 og forbindelse 1 i vann. Isetionatsaltene kan fremstilles ved titrering av forbindelsene i løsning med 2 ekvivalenter isetionsyre. Behandling var basert på gjennomsnittsgruppevekt Kontrolldyrene fikk vann.
Evaluering av eksperimentet ble basert på tumorvekstforsinkelse, T-C, definert som forskjellen i dager for de behandlede og kontrolltumorer skulle nå en evalueringsstørrelse på 750 mg. Data kan også representeres som prosentdel av tumorvekstinhibering, definert som tumorvekstforsinkelsen delt på antallet av be-handlingstider 100%. Større tumorvekstforsinkelser og tumorvekstinhiberingsver-dier er indikative for mer aktive forbindelser. Statistisk analyse ble utført ved å bruke JMP for Macintosh (SAS Institute, Inc., Cary, NC).
Verdiene for forbindelse 1 er signifikant forskjellig fra dem for eksempel 21 basert på den nødvendige tid for individuelle tumorer skulle nå 750 mg. p<0,05, studentens t-test.
VEGF-SEKRESJONSMÅLING
Vaskulære endotelial vekstfaktor (VEGF) som også er kjent som vaskulær permeabilitetsfaktor (VPF) antas å være angiogenesehovedstimulatoren for de fleste typer kreft. Det finnes mange indusere for tumor VEGF-sekresjon, og to av de sterkeste er epidermal vekstfaktor (EGF) og transformeringsvekstfaktor alfa, som begge er ligander for EGF-reseptor (EGFR).
Inhiberingen av VEGF-sekresjon fører til en antiangiogen effekt og kan føre til tumorregresjon, da selv delvis inhibering av VEGF-sekresjon kan føre til dest-ruksjon av nydannede uferdige blodkar i tumorer. (Benjamin L.E. and Keshel E., Conditional switching of VEGF expression in tumors: induction of endothelial cell shedding and regression of hemangioblastoma-like vessels by VEGF withdrawal. Proe. Nati. Acad. Sei. USA, 1997; 94:8761-8766).
Vi har sammenlignet to EGFR-tyrosin-kinaseinhibitorer, forbindelse 1 og eksempel 21, for deres effekt på A431-tumorceller. Mot A431 tumorer in vivo ga forbindelse 1 uventet en større antitumorrespons (T-C = 53 dager) enn eksempel 21 (T-C - 26 dager). Se målingen som er angitt ovenfor. Så undersøkte vi effek-ten av begge forbindelser på A431 sekresjon av VEGF. Til vår overraskelse var forbindelse 1 bedre enn eksempel 21 ved inhibering av VEGF-sekresjon fra A431 celler in vitro. Ved 0,5 uM blokkerte forbindelse 1 EGF- eller TGF-alfastimulert VEGF-sekresjon. Derimot inhiberte 0,5 jiM eksempel 21 VEGF-sekresjon i nesten samme grad som observert med 0,1 uM av forbindelse 1. 0,1 |iM av forbindelse 1 inhiberte EGF-stimulert VEGF-sekresjon med 63% og 0,5 |iM inhiberte med 86%. Derimot inhiberte 0,1 \ iM av eksempel 21 EGF-stimulert VEGF-sekresjon med 48% og 0,5 jiM av eksempel 21 inhiberte EGF-stimulert VEGF-sekresjon med 57%. Lignende resultater ble oppnådd ved å bruke TGF-alfa til å stimulere VEGF-sekresjon i stedet for EGF. 0,5 u.M av forbindelse 1 inhiberte TGF-alfa-stimulert VEGF-sekresjon med mer enn 80%, mens den samme konsentrasjon av eksempel 21 inhiberte VEGF-sekresjon med 57%. Forbindelse 1 viste seg også å inhibere grunn-nivået av VEGF-sekrsjon fra A431-celler, mens eksempel 21 ikke hadde noen effekt.
Metoder
A431 humane epidermoide karcinomaceller oppnådd fra the American Type Culture Collection (Rockville, MD) ble dyrket i Dulbecco's MEM/F12 media (DMEM/F12) tilsatt 10% føtalt bovinserum. Før medikament og vekstbehandling ble cellene vasket tre ganger i serumfritt DMEM/F12. Celler ble behandlet i 2 timer med den angitte konsentrasjon av forbindelse 1 eller eksempel 21 før tilsetningen av enten 10 ng/ml EGF eller TGF-alfa. Etter 48 timer inkubasjon ved 37°C ble media fjernet og lagret frosset ved -70°C. VEGF ELISA-målinger (Intergren, Co., Purchase, NY) utføres på mediaprøver etter produsentens instruksjoner. Resultater av denne måling er vist nedenunder i tabellform.
Claims (15)

6. Anvendelse av en forbindelse ifølge krav 1 til fremstilling av et farmasøytisk preparat for behandling eller forebygging av restenose.
