Description
Foreliggende oppfinnelse vedrører en fremgangsmåte for fremstilling av et plasmid som i en mikrobiell organisme kan utrykke et spesielt polypeptid med kjent aminosyresekvens, hvor et gen som koder for polypeptidet er innført i en et plasmid og satt under kontroll av en ekspresjonspromoter.
Genetisk ekspresjon:
DNA (deoksyribonukleinsyre) hvorfra gener fremstilles, omfatter både proteinkodende eller "strukturelle" gener og kontrollområder som formidler ekspresjonen av deres informasjon gjennom tilveiebringelse av seter for RNA-polymerase-binding, informasjon for ribosomale bindingsseter, osv. Innkodet protein blir "uttrykt" fra dets tilsvarende DNA ved en flertrinnsprosess i en organisme hvorved: 1. Enzymet RNA-polymerase aktiveres i kontrollområdet (i det følgende betegnet "promoter") og beveger seg langs det strukturelle gen og transkriberer dets innkodende informasjon til mRNA inntil transkripsjon er avsluttet ved en eller flere "stopp"-kodoner. 2. mRNA oversettes ved ribosomene til et protein hvis aminosyresekvens genet koder, idet dette begynner ved et translasjonsstartsignal, vanligvis ATG (som er translatert "f-metionin").
Ifølge den genetiske kode spesifiserer DNA hver aminosyre ved en triplet eller "kodon" av tre tilgrensende nukleotider individuelt valgt fra adenosin, tymidin, cytidin og guanin eller, slik som her benyttet, A, T, C eller G. Disse opptrer i den kodende kjede eller kodende sekvens av dobbeltkjedet ("dupleks") DNA, hvis gjenværende eller "komplementære" kjede er dannet av nukleotider ("baser") som hydrogenbindes til deres komplementer i den kodende kjede. A kompiementerer T, og C komplementerer G. Disse og andre trekk som vedrører bakgrunnen for foreliggende oppfinnelse, er inngående omtalt i Benjamin Lewin, Gene Expression, 1, 2 (1974) og 3 (1977), John Wiley and Sons, N.Y.
DNA- spalting og llgering:
En rekke forskjellige teknikker er tilgjengelige for DNA-kombinasjon, hvorved tilgrensende ender av separate DNA-fragmenter tilpasses på en eller annen måte for å lette llgering. Det sistnevnte begrep angår dannelse av fosfor-diester-bindinger mellom tilgrensende nukleotider, oftest ved hjelp av enzymet T4 DNA-ligase. Butte ender kan således direkte ligeres. Fragmentene inneholdende komplementære enkeltkjeder ved deres tilgrensende ender blir alternativt begunstiget ved hydrogenbInding, hvilket posisjonerer de respektive ender for etterfølgende llgering. Slike enkeltkjeder som betegnes kohesive termini, kan dannes ved addisjon av nukleotider til butte ender under anvendelse av terminal transferase, og 1 visse tilfeller ved å fjerne en av kjedene ved en butt ende ved hjelp av et enzym, slik som"Y-ekso-nuklease. Den mest vanlige teknikk er å bruke restriksjonsendonukleaser (i det følgende betegnet "restriksjonsenzymer"), som spalter fosfodiesterbindinger i og omkring enestående sekvenser av nukleotider med en lengde på ca. 4-6 basepar ("restriksjonsseter"). Mange restriksjonsenzymer og deres gjenkjennende seter er kjente, kfr.f.eks. R.J. Roberts, CRC Critical Reviews in Biochemistry, 123 (Nov. 1976). Mange foretar kutt i. sikksakkmønster hvilket gir korte komplementære enkeltkjedede sekvenser ved endene på dupleks-fragmentene. Som komplementære sekvenser kan de fremskytende eller "kohesive" ender rekombineres ved basepardannelse. Når to forskjellige molekyler spaltes med dette enzym, vil tverr-pardannelse av de komplementære enkeltkjeder utvikle et nytt DNA-molekyl som kan gis kovalent integritet ved anvendelse av ligase for å tette enkeltkjede-bruddene som gjenstår ved sammenføyningspunktet. Restriksjonsenzymer som gir ko- terminale eller "butte" ender på dupleks DNA som har blitt spaltet, tillater rekomblnasjon via f.eks.T4-ligase med andre sekvenser som har butte ender.
Klonlngsvektorer og rekombinant DNA:
For foreliggende formål menes det med en "kloningsvektor" en lkke-kromosomal lengde av dupleks DNA omfattende en intakt replikon, slik at vektoren kan replikeres når den anbringes inne i en encellet organisme ("mikrobe") ved transformasjon. En således transformert organisme kalles en "transformant". De klonlngsvektorer som i dag anvendes, er avledet fra virus og bakterier og er vanligvis ringer av bakterie-DNA betegnet "plasmider".
Biokjemisk forskning i de senere år har ført til konstruksjon av "rekombinant"-klonlngsvektorer hvori f.eks. plasmider dannes slik at de inneholder eksogent DNA. I visse tilfeller kan rekomblnanten inneholde "heterologt" DNA, dvs. DNA som koder for polypeptider som vanligvis ikke dannes av den organisme som er mottagelig for transformasjon ved hjelp av rekombinant-vektoren. Plasmidene blir således spaltet med restriksjonsenzymer slik at det oppnås lineært DNA som har ligerbåre termini. Disse bindes til et eksogent gen med ligerbare termini, slik at det oppnås en biologisk funksjo-nell del med en intakt replikon og en fenotyp egenskap som er nyttig ved valg av transformanter. Rekombinantvektoren innføres i en mikroorganisme ved transformasjon, og transfor-manten isoleres og klones med det formål å oppnå store populasjoner som inneholder kopier av det eksogene gen, og i visse tilfeller, med det formål å uttrykke det protein for hvilket genet koder. Rekomblnant- DNA ekspresjon
Foruten bruken av klonlngsvektorer for å øke mengden av gener ved replikasjon har det med hell vært foretatt noen forsøk på å uttrykke proteiner som genene koder for. Her ble først et gen for hjernehormonet somatostatlon under innvirkning av lac-promoteren uttrykt i E. coli bakterier, kfr. K. Itakura et al., Science 198, 1056, (1977). Senere ble A- og B-kjedene i humaninsulin uttrykt på samme måte og kombinert for dannelse av hormonet, kfr. D.V. Goeddel et al., Proe. Nat'l. Acad. Sei, USA, 106 (1979). I hvert tilfelle ble genene konstruert i sin helhet ved syntese. I hvert tilfelle ville proteolytiske enzymer i cellen antageligvis nedbryte det ønskede produkt, hvilket nødvendiggjør dets dannelse i konjugert form, dvs. i tandem med et annet protein som beskyttet det ved oppdeling i seksjoner uten innbyrdes forbindelse, og som kunne avspaltes ekstracellulært for dannelse av det ønskede produkt. Dette arbeid er beskrevet i GB nr. 2007.675.A, 2007.670.A, 2007.676.A, og 2008.123.A.
Mens forsøk med det syntetiske gen har vist seg å være nyttig i de ovenfor omtalte tilfeller, oppstår virkelige vanskelig-heter med langt; større proteinprodukter, f.eks. veksthormon, interferon, osv/., hvis gener er tilsvarende mer komplekse og mindre mottagelige for enkel syntese. Samtidig ville det være ønskelig å uttrykke slike produkter uledsaget av konjugat protein, idet nødvendigheten av dettes ekspresjon krever bruk av ressurser i organismen som heller kan benyttes bedre til konstruksjon av det ønskede produkt.
Andre har forsøkt å uttrykke gener avledet ikke ved organisk syntese, men ved revers transkripsjon fra den tilsvarende mRNA renset fra. vev. Denne utførelse har vært forbundet med to problemer. For det første kan revers transkriptase stoppe transkripsjonen fra mRNA like før fullendelse av cDNA for hele den ønskede aminosyresekvens. Villa-Komaroff et al. oppnådde således f.eks. cDNA for rotte-proinsulin som manglet kodoner for de tre første aminosyrer i insulin-forløperen, kfr. Proe. Nat'l. Acad. Sei., USA 75 3727 (1978). Videre har revers transkripsjon av mRNA for polypeptider som er uttrykt i forløper-form, gitt cDNA for forløper-formen 1 stedet for det bioaktlve protein som resulterer når ledersekvenser i en eukaryot celle, fjernes enzymatisk. Hittil har det ikke vært vist at en bakteriell celle har en slik egenskap, slik at mRNA-transkripter har gitt ekspresjonsprodukter inneholdende ledersekvensene i forløper-formen i stedet for det bioaktlve protein, kfr. Villa-Komaroff, supra (rotte proinsulin); P.H. Seeburg et al., Nature 276, 795 (1978) (rotte-prevekst-hormon).
Forsøk foretatt av andre for bakterielt å uttrykke human-hormoner (eller deres forløpere) fra mRNA-transkripter, har av og til bare ledet til dannelse av konjugerte proteiner som tydeligvis ikke er mottagelige for ekstracellulær spalting; Seeburg, supra, (p-laktamase-preveksthormon).

Humant veksthormon ("HGH") utskilles i hypofysen. Det består av 191 aminosyrer og er med sin molekylvekt på ca. 21.500 mer enn tre ganger så stort som insulin. Tidligere kunne humant veksthormon bare oppnås ved tungvint ekstraksjon fra en begrenset kilde, nemlig hypofysen fra døde mennesker. Den følgelige knapphet av stoffet har begrenset dets anvendelse til behandling av dvergvekst ved nedsatt hypofyseaktivitet, og selv her antyder pålitelige beregninger at humant avledet HGH er tilgjengelig i tilstrekkelig mengde til å tjene høyst ca. 50# av aktuelle pasienter.
Ifølge foreliggende oppfinnelse er det tilveiebragt en fremgangsmåte for fremstilling av et plasmid som i en bakteriell organisme kan uttrykke et spesielt polypeptid med kjent aminosyresekvens, hvorved et gen som koder for polypeptidet innføres 1 et plasmid og settes under kontroll av en ekspresjonspromoter, og denne fremgangsmåten er kjennetegnet ved at man: a) tilveiebringer et cDNA-fragment som omfatter en vesentlig del av, men mindre enn hele den kodende sekvensen for nevnte polypeptid; b) gjennom organisk syntese tilveiebringer ett eller flere genfragmenter som koder for resten av aminosyresekvensen i polypeptidet, idet minst ett av nevnte fragmenter koder for amino-terminaldelen 1 polypeptidet; og c) innordner cDNA'et fra trinn a) og DNA'et oppnådd ved organisk syntese i trinn b) i et plasmid i riktig leseramme 1
forhold til hverandre og under kontroll av en ekspresjonspromoter ;
hvorved det dannes et plasmid som kan uttrykke aminosyresekvensen til nevnte polypeptid.
Det er foretrukket at nevnte humant veksthormon uttrykkes uledsaget av fremmed protein.
Dette representerer den første anledning hvorved et medisinsk betydelig polypeptid-hormon (humant veksthormon) har blitt bakterielt uttrykt med samtidig unngåelse av både intra-cellulær proteo.lyse og nødvendighet for oppdeling i seksjoner ("compartmentalizing") av den bioaktlve form i fremmed protein før ekstracellulær spalting. Mikrobielle kilder for human-veksthormon tilgjengeliggjort ved foreliggende oppfinnelse gir for første gang tilstrekkelig mengder av hormonet for behandling av dvergvekst ved nedsatt hypofyse-aktlvitet sammen med andre anvendelser som hittil har ligget utenfor kapasiteten hos vevsavledede hormonkilder, inkludert diffus gastrisk blødning, pseudoarthrosis, brannsårterapi, sårleging, dystrofi og ben-sammenvoksing.
Foreliggende oppfinnelse vil fremgå mer detaljert fra nedenstående beskrivelse og fra de medfølgende tegninger som angår en foretrukken utførelse av foreliggende oppfinnelse, idet
figur 1 viser det syntetiske skjema for konstruksjon av et genfragment som koder for de første 24 aminosyrer i humant veksthormon sammen med startsignal-ATG og bindingselementer benyttet ved kloning. Pilene i den kodende eller den øvre kjede ("U") og i den komplementære eller lavere kjede ("L") Indikerer oligonukleotidene som er sammeføyet til dannelse av det viste fragment;
figur 2 viser sammenføyning av "U"- og "L"-oligonukleotidene til dannelse av genfragmentet i figur 1, og dets innføring i en plasmid-klonlngsvektor;
figur 3 illustrerer DNA-sekvenser (bare kodende kjede) hos Haelll-restriksjonsenzymfragmentet i et mRNA-transkript fra hypofysen, med de nummererte aminosyrer i humant veksthormon for hvilket de koder. De viktigste restriksjonsseter er indikert, slik som tilfellet er for DNA (etter stopp) for utranslatert mRNA;
figur 4 illustrerer konstruksjonen av en kloningsvektor for et genfragment som koder for aminosyre i humant veksthormon som ikke er fremstilt syntetisk, og konstruksjonen av nevnte genfragment som komplementær DNA ved revers transkripsjon fra mRNA isolert fra en kilde for human hypofyse; og
figur 5 illustrerer konstruksjonen av et plasmid som i bakterier kan uttrykke humant veksthormon begynnende med plasmidene 1 figurene 2 og 3.
Den generelle metode ifølge oppfinnelsen innebærer kombina-sjonen i et enkelt plasmid av flere genfragmenter som i kombinasjon koder for ekspresjon av det ønskede produkt. Av disse er minst ett cDNA-fragment oppnådd ved revers transkripsjon fra mRNA isolert fra vev, som ved fremgangsmåten ifølge A. Ullrich et al., Science 196, 1313 (1977). cDNA tilveiebringer en vesentlig del og fortrinnsvis i det minste en større del, av kodonene for det ønskede produkt, mens gjenværende deler av genet tilveiebringes syntetisk. De syntetiske og mRNA-transkriptfragmentene klones separat for tilveiebringelse av tilstrekkelige mengder for bruk i det senere kombinas;jonstrinn.
En rekke hensyn innvirker på fordelingen av kodoner for endeproduktet, slik som mellom syntetisk og cDNA, særlig DNA-sekvensen hos komplementært DNA bestemt ved metoden ifølge Maxam og Gilbert, Proe. Nat'1. Acad. Sei, UA, 74, 560 (1977). Komplementært DNA oppnådd ved revers transkripsjon vil uunngåelig Inneholde kodoner for i det minste en karboksy-terminaldel av det ønskede produkt, samt andre kodoner for utranslatert mRNA i nedstrømsretningen fra translasjons-stoppsignalet (slngalene) tilgrensende karboksyterminusen. Tilstedeværelsen av cDNA for utranslatert RNA er stort sett irrelevant, skjønt urimelig lange sekvenser av denne type kan fjernes slik som ved spalting med restriksjonsenzym, for å bevare cellulære forråd benyttet ved replikering og ekspresjon av DNA for det ønskede produkt. I spesielle tilfeller vil cDNA inneholde kodoner for hele den ønskede aminosyresekvens samt fremmede kodoner i oppstrømsretningen fra amino-terminalen i det ønskede produkt. F.eks. er mange, om ikke alle, polypeptid-hormoner uttrykt i forløper-form med leder-eller slgnalsekvenser av protein involvert, f.eks. i transport i cellemembranen. Ved ekspresjon fra eukaryotiske celler blir disse sekvenser fjernet enzymatisk slik at hormonet kommer inn i de periplasmatiske rom i sin frie, bioaktlve form. Man kan imidlertid ikke stole på mikrobielle celler når det gjelder å utføre denne funksjon, og det er følgelig ønskelig å fjerne sekvenser som koder for silke signaler eller ledersekvenser fra mRNA-transkriptet. I løpet av denne fjerningsprosess går også translasjons-startsignalet tapt, og noen kodoner for det ønskede produkt vil nesten uvegerlig også bli fjernet. Den syntetiske komponent i det kvasi-syntetiske genprodukt returnerer disse sistnevnte kodoner samtidig som de på nytt tilfører et translasjonsstartsignal hvor vektoren, i hvilken det hybride gen til slutt vil innordne seg, mangler en riktig beliggende start.
Eliminasjon av ledersekvensen fra prevekst-hormon cDNA fremmes ved tilgjengeligheten av et restriksjonssete innen den veksthormon-kodende del av genet. Oppfinnelsen kan Ikke desto mindre utføres uten hensyn til tilgjengeligheten av et slikt sete, eller ihvertfall uten hensyn til tilgjengeligheten av et restriksjonssete som er tilstrekkelig nær amino-terminalen i det ønskede polypeptid for å unngå behovet for utstrakt syntese av genkomponenten som ikke er avledet fra mRNA. I enhver cDNA som koder for det ønskede polypeptid og en ledersekvens eller en annen bloinaktiverende sekvens, vil således grensen mellom den sistnevntes kodoner og de i det ferdige polypeptid fremgå fra aminosyresekvensen i det ferdige polypeptid. Man kan ganske enkelt foreta spalting til det kodende gen fra det valgte peptid og dermed fjerne den uønskede ledersekvens eller en annen sekvens. F.eks., gitt cDNA slik som
hvor endepunktet for spalting er indikert med en pil , kan således bet Inge 1 sene for eks onuklease-spal ting velges slik at man fjerner de øvre sekvenser "a" og "b", hvoretter Sl-nuklease-spalting automat! sk vil eliminere de nedre sekvenser
"c" og "d". Alternativt og mer nøyaktig kan man benytte DNA-polymerase-spalting i nærvær av deoksymikleotid-trifosfater ( "d(A,T,C,G)TP" )i. I det foregående eksempel vil således DNA-polymerase i nærvær av dGTP fjerne sekvens "c" (deretter stopp ved "g"), Sl-nuklease vil deretter spalte "a"; DNA-polymerase i nærvær av dTTP vil fjerne "d", deretter stopp ved "T", og Sl-nuklease vil deretter fjerne "b", osv. Se generelt A. Kornberg, DNA Synthesis, pp. 87-88, W.H. Freeman og Co., San Francisco (1974).
Mer spesielt kan man på enkel måte konstruere et restriksjonssete ved et passende punkt i den del av det cDNA som koder for det ønskede produkt, ved anvendelse av "mismatch"-reparasjonssynteseteknikken ifølge A. Razin et al., Proe. Nat* 1. Acad. Sei. USA 75, 4268 (1978). Ved hjelp av denne teknikk kan en eller flere baser substitueres i en eksisterende DNA-sekvens under anvendelse av primere inneholdende den mispassede substituent. Minst syv palindrome 4-basepar sekvenser gjenkjennes entydig av kjente restriksjonsenzymer, dvs. AGCT (Alul), CCG (Hpall), CGCG (Thai), GATC (Sau3A), GCGT (Hha), GGCC (Haelll) og TC GA (Taql). Når cDNA-sekvensen inneholder en sekvens som adskiller seg fra en slik posisjon i en enkelt bane, hvilket statistisk er meget sannsynlig, vil reparasjonssyntese gi repllkat cDNA inneholdende den riktige substituentbase og således det ønskede restriksjonssete. Spalting vil fjerne DNA fra den uønskede leder hvoretter syntese vil gi kodoner som er nødvendig for ekspresjon av det fullstendige polypeptid. F.eks.: ;
Det vil naturligvis forstås at lengre restriksjonsseter likeledes kan innføres når det er ønsket, eller at suksessive reparasjoner kan skape 4-basepar-restriksjonsseter når bare to baser som er felles for setet, forekommer ved det ønskede punkt, etc. ;Det vil forekomme anvendelser hvorved det er ønskelig ikke bare å uttrykke aminosekvensen for det ønskede produkt, men også et mål for fremmed, men spesifikt konstruert protein. Fire slike anvendelser kan eksempelvis nevnes. Først, det kvasi-syntetiske gen kan representere et hapten eller en annen immunologisk determinant hvorpå immunogenlsitet gis ved konjugering til ytterligere protein, slik at vaksiner fremstilles, se generelt britisk patent nr. 2008.123.A. Det kan videre være ønskelig av bio-sikkerhetsmessige grunner å uttrykke det ønskede produkt som et konjugat med et annet, bio-inaktiverende protein som er slik konstruert at det tillater ekstracellulær spalting for dannelse av den aktive form. For det tredje vil det være anvendelser hvor transport-signal-polypeptider vil være forut for det ønskede produkt og dermed tillate produksjon av dette slik at det kan utskilles gjennom cellemembranen, men dette forutsetter at signal-peptidet deretter kan spaltes. Sluttlig, fremmed konjugat som er slik konstruert at det tillater spesifikk spalting ekstracellulært, kan benyttes for å seksjonsoppdele ønskede produkter som ellers er mottagelige for nedbrytning av proteaser endogent i forhold til den mikrobielle vert. I det minste i de sist angitte tre anvendelser kan det syntetiske adaptor-molekyl benyttes for å fullende kodingssekvensen i mRNA-transkripteit, i tillegg inkorporere kodoner for aminosyresekvenser som er spesifikt spaltbare, slik som ved enzymatisk behandling. Trypsin vil f.eks. spaltes spesifikt ved arg-arg eller lys-lys, etc., se britisk patent nr. 2008.123.A, Supra. ;Fra det ovenstående fremkommer mange anvendelser som hver har følgende trekk felles: det anvendes et mRNA-transkript som koder for en vesentlig del av den ønskede aminosyresekvens i polypeptidet, men som hvis det uttrykkes alene, ville gi et forskjellig polypeptid enten mindre eller større enn det ønskede produkt; ;proteinkodende kodoner for aminosyresekvenser andre enn de som inneholdes i det ønskede produkt, hvis slike er til stede, fjernes; ;organiske synteser gir fragment(er) som koder for resten av den ønskede sekvens; og ;mRNA-transkrlptet og det syntetiske fragment(er) kombineres og plasseres i en promoterholdig kloningsvektor for replikasjon og ekspresjon av enten det ønskede produkt uten eksternt konjugert protein, eller det ønskede produkt konjugert til, men spesifikt spaltbart fra eksternt protein. ;Ekspresjonsproduktet vil naturligvis i hvert tilfelle begynne med aminosyren som er kodet for av translasjons-startsignalet (i tilfellet for ATG, f-metionin). Man kan forvente at denne fjernes intracellulært eller lhvertfall gjør at bio- aktiviteten til det sluttlige produkt forblir vesentlig upåvirket. ;En foretrukken utførelse som illustrerer oppfinnelsen, er omtalt i det nedenstående, hvorved det konstrueres, klones og uttrykkes mikrobielt et kvasi-syntetisk gen som koder for humant veksthormon. ;Konstruksjon og ekspresjon av en kloningsvektor for humant veksthormon 1. Kloning av HaeIII- fragmentet i mRNA- transkrlptet ( figurene ;3 og 4) ;Polyadenylert mRNA for humant veksthormon (HGH) ble fremstilt fra hypofyseveksthormon-produserende tumorer ved hjelp av metoden ifølge A. Ullrich et al., Science 196, 1313 ;(1977). 1,5 pg dobbeltkjedet (ds") cDNA ble fremstilt fra 5 jjg av det RNA slik som vesentlig beskrevet av Wickens et al., J. Biol.Chem. 253 2483 (1978), med unntagelse av at RNA-polymerase "Klenow fragment", H. Klenow, Proe. Nat'l. Aci. USA. 65, 168 (1970), ble utbyttet med DNA-polymerase I i syntesen av den andre kjede. Restriksjonsmønsteret for HGH er slik at Haelll-restriksjonsseter er til stede i det 3' ikke-kodende område og i den sekvens som koder for aminosyrer 23 og 24 i HGH, som vist på figur 3. Behandling av ds HGH cDNA med Haelll gir et DNA-fragment med 551 basepar ("bp") som koder for aminosyrer 24-191 i HGH. Således ble 90 ng cDNA behandlet med Haelll, elektroforesebehandlet på en 8% polyakrylamidgel, hvoretter området ved 550 bp ble eluert. Omtrent 1 ng cDNA ble oppnådd. ;pBR322 fremstilt ifølge F. Bolivar et al., Gene 2 (1977) 95-113 ble valgt som kloningsbærer for cDNA. pBR322 er blitt fullstendigkarakterisertav J.G. Sutcliffe, Cold Spring Harbor Symposium, 43, 70 (1978) og er et multikopi-replike- ;rende plasmid som utviser både amplcillin- og tetracyklin-resistens p.g.a. dets innhold av de tilsvarende gener ("Ap<K>" og "Tc<*>", resp. figur 4), og som inneholder gjenkjennings-posisjoner for restriksjonsenzymene Pstl, EcoRI og Hindlll som vist på figuren.
Terminal-deoksynukleotidyl-transferase (TdT) ble således benyttet for å addere omtrent 20 dC-rester pr. 3'-terminal som beskrevet av Chang, A.Y.C. supra. 60 ng av Pstl-behandling pBR322 ble likeledes endeavslutningsbehandlet med ca. 10 dG-rester pr. 3'-terminal. Forsterkning av dC-ende-avsluttet ds cDNA med den dG-endeavsluttede vektor DNA ble foretatt i 130 pl 10 mM tris-HCl (pH 7,5), 100 mM NaCl, 0,25 mM EDTA. Blandingen ble oppvarmet til 70°C, fikk avkjøles langsomt til 37°C (12 timer), og deretter til 20°C (6 timer) før den ble benyttet for å transformere E. coil xl776. DNA-sekvensanalyse av plasmidet pHGH31 klonet i xl776 ved hjelp av metoden ifølge Maxam og Gilbert, Proe. Nat'l. Acad. Sei. USA 74, 560 (1977), resulterte i bekreftelse av kodonene for aminosyrer 24-191 i HGH som vist på figur 3.

xl776 har et obligatkrav for diaminopimelinsyre (DAP) og kan ikke syntetisere mukopolysakkaridcholansyren. Det gjennomgår således DAP-fri død i alle miljøer hvor DAP er begrensende, men hvor tilstrekkelige næringsmidler eksisterer til å underholde cellulær metabolisme og vekst. Det kreves tymin eller tymidin og gjennomgår tyminfri død med nedbrytning av DNA når tymin og tymidin er fraværende i miljøet, men når tilstrekkelige næringsmidler er til stede til å underholde metabolisk aktivitet. xl776 er meget følsom overfor galle og er således ikke 1 stand til å overleve og således ikke i
stand til å overleve passasje gjennom mavetarmkanalen hos rotter. xl776 er meget følsom overfor vaskemidler, anti-biotika, legemidler og kjemikalier. xl776 kan ikke utføre hverken mørke- eller fotoreparasjon av UV-lndusert skade og er således flere størrelsesordener mer følsom overfor sollys enn udyrkede stammer av E. coil. xl776 er resistent overfor mange transduserende farger og er konjugasjonsmangelfull med hensyn til arv av mange forskjellige typer konjugative plasmider p.g.a. tilstedeværelsen av forskjellige mutasjoner. xl776 er resistent overfor nalidiksinsyre, cykloserin og trimetoprim. Disse legemidler kan derfor tilsettes til media for å tillate kontroll av stammen og utelukke transformasjon av kontaminerende elementer under transformasjon.
xl776 vokser med en generasjonstid på ca. 50 min. i enten L-buljong eller Penassay-bul jong ved tilsetning av 100 jjg DAP/ml og 4 yg tymidin/ml, og når sluttlige tettheter på 8-10x 10<8>celler/ml ved stasjonær fase. Forsiktig omrøring ved dreiing og rysting frem og tilbake i 1-2 minutter suspenderer cellene tilstrekkelig under opprettholdelse av 100% leve-dyktighet. 2. Konstruksjon og kloning av det syntetiske genfragment
( figurene 1 og 2)
Metoden for konstruksjon av det HGH-kvasi-syntetiske gen omfattet konstruksjon av et syntetisk fragment omfattende et buttendet restriksjonsspaltingssete tilgrensende det punkt ved hvilket fragmentet ville bli sammenføyet med mRNA-transkriptet. Som vist på figur 1 inneholdt således det syntetiske gen for de første 24 aminosyrer i HGH et Haelll-spaltingssete etter aminosyre 23. Den distale enden 1 det syntetiske fragment ble forsynt med et "bindende element" som ga adgang til sammenføyning med en enkeltkjedet terminal resulterende fra restriksjonsspaltinger i plasmidet hvori mRNA-transkriptet og det syntetiske fragment til slutt ville bli tilføyet.
Som vist på figur 1 har 5'-endene i dupleksfragmentet enkeltkjedede, kohesive termini for EcoRI- og Hindlll-restriksjonsendonukleaser for å lette plasmidkonstruksjon. Metionin-kodonet ved den venstre ende gir et sete for initiering av translasjon. Tolv forskjellige oligonukleotider som varierer i størrelse fra undekaner til heksadekaner ble syntetisert ved den forbedrede fosfotriester-metoden ifølge Crea, R. Proe. Nat'1. Acad. Sei. USA 75, 5765(1978). Disse oligonukleotider, U^- U^, og - , er vist med piler.
Mengder på lOpg av U2til U5og L2til L5ble fosforylert under anvendelse av T4polynukleotid-kinase og ("y<32->P)ATP ved hjelp av en kjent metode, C. Goeddel, D.V. et al., Proe. Nat'1. Acad. Sei. USA 76, 106 (1979).
Tre separate T^ligasekatalyserte reaksjoner ble foretatt: 10 pg av 5'-0H fragment U^ble kombinert med det fosforylerte U2, L5og Lfcj fosforylert U3, U4, L3og L4ble kombinert; og 10 >jg av 5'-fragment Lj ble kombinert med fosforylert L2, U5og Ufc. Disse ligeringer ble utført ved 4°C i 6 timer i 300 jjI av 20 mM tris-HCl (pH 7,5), mM MgCl2, 10 mM ditiotreitol, 0,5 mM ATP under anvendelse av 100 enheter med T4-ligase. De tre 1igerlngsblandinger ble deretter kombinert, 100 enheter T4-ligase ble tilsatt, og reaksjonen fikk forløpe i 12 timer ved 20°C. Blandingen ble etanolutfelt og elektroforesebehandlet på en 10% polyakrylamidgel. Båndet som migrerte ved 84 basepar, ble skilt fra gelen og eluert. pBR322 (ljjg) ble behandlet med EcoRI og Hindlll, det største fragment ble isolert ved gel-elektroforese og ligert med det syntetiske DNA. Denne blanding ble benyttet til å transformere E. coli K-12 stamme 294 (ende A, thi~, hsr~, hsmjj"1"). Stamme 294 ble deponert den 30. oktober 1978 ved American Type Culture Collection (ATCC Nr. 31446), uten begrensning. Sekvensanalyse Ifølge Maxam og Gilbert-teknikken, Supra, på den EcoRI-Hindlll-tllføyende del fra et plasmid pHGH av en transformant bekreftet det som er vist på flg. 1.
3. Konstruksjon av<p>lasmid for bakteriell ekspresjon av HGH
( figur 5)
Med det syntetiske fragment i pHGH3 og mRNA-transkriptet pHGH31 ble det konstruert et replikerbart plasmid inneholdende begge fragmenter, under anvendelse av ekspresjonsplasmidet pHG6 som vist på figur 5. Ekspresjonsplasmidet som inneholder tandem-lac-promotere, ble først konstruert som følger. Et 285 basepar EcoRI-fragment inneholdende to 95 basepar UV5 lac-promoterfragmenter adskilt ved et 95 baseparheterologt DNA-fragment, ble Isolert fra plasmid pKB268, K. Backman, et al., Cell. Vol. 13, 65-71 (1978). 285 bp-fragmentet ble innført i EcoRI-setet i pBR322 og en klon pGHl Isolert med promoterene orientert mot og i riktig leseramme i forhold til genet for tetracyklin-resistens. EcoRI-posisjonen beliggende distalt i forhold til sistnevnte gen ble ødelagt ved partiell EcoRI-nedbrytning, reparasjon av de resulterende enkeltkjedede EcoRI-ender med DNA-polymerase I og sirkulasjon av plasmidet ved buttende llgering. Det resulterende plasmid, pGH6, inneholder et enkelt EcoRI-sete som er riktig plassert med hensyn til promote-systemet hvori det kompletterte gen for HGH kunne innføres.
For å klargjøre det syntetiske fragment for kombinasjon med RNA-transkriptet ble 10 pg av pHGH spaltet med EcoRI- og Haelll-restriksjonsendonukleaser, og 77 basepar-fragmenter inneholdende kodende sekvenser for HGH aminosyrer 1-23 ble isolert fra en 8% polyakrylamidgel.
Plasmidet pHGH31 (5>jg) ble deretter spaltet med Haelll. 551 bp HGH-sekvensen, og et komigrerende 540 bp HaeIII-fragment av pBR322 ble renset ved gelelektroforese. Etterfølgende behandling med Xmal spaltet bare HGH-sekvensen og fjernet dermed 39-baseparene fra det 3'-ikke-kodende området. Det resulterende 512 bp-fragment ble separert fra 540 bp pBR322 Haelll-delen ved elektroforese på en 6$ polyakrylamidgel. 0,3 g av 77 bp EcoRI-HaeIII-fragmentet ble polymerisert med T4-ligase i en 6 >j1 reaksjonsbeholder i 14 timer ved 4°C. Blandingen ble oppvarmet til 70°C i 5 minutter for å inaktivere ligasen, og deretter behandlet med EcoRI (for å spalte fragmenter som var dimerisert gjennom deres EcoRI-seter) og med Smal (for å spalte Xmal-dimerer) hvilket ga et 591 bp-fragment med en EcoRI "kohesiv" ende og en Smal "butt" ende. Etter rensing på en b% polyakrylamidgel ble omtremt 30 hg av dette fragment oppnådd. Det skal bemerkes at ekspresjonsplasmidet pGH6 ikke inneholder noen Xmal gjenkjennelsess-ete. Smal gjenkjenner imidlertid det samme sete som Xmal, men skjærer gjennom dens midtre del og gir dermed butte ender. Den Sma-avspaltede terminal i fragmentet avledet fra gHGH31 kan følgelig være buttende-ligert Inn i pGH6.
Ekspresjonsplasmidet pGH6 inneholdende lac UV5-promotere ble behandlet suksessivt med Hindlll nuklease Sl og EcoRI og renset ved gelelektroforese. 50 ng av den resulterende vektor som hadde en EcoRI-kohesiv ende og en butt ende, ble ligert til 10 ng av 591 bp. HGH DNA. Llgerlngsblandingen ble benyttet for å transformere E. coli xl776. Kolonier ble valgt for vekst på tetracyklln (12,5 jjg/ml). Det skal påpekes at innføring av hybrid-HGH-genet i pGH6 ødelegger promoteren med hensyn til det tetracykllnresistente gen, men at tandem-lac-promoteren tillater gjennomlesing av det strukturelle gen for tet-resistens under bibeholdelse av denne utvelgelses-egenskap. Omtrent 400 transformanter ble oppnådd. Filter-hybridisering ifølge Grunstein/Hogness-metoden, Proe. Nat'1. Acad. Sei. USA, 72, 3961 (1975), identifiserte 12 kolonier inneholdende HGH-sekvenser. Plasmidet isolert fra tre av disse kolonier ga de forventede restriksjonsmønstre ved spalting med Haelll, PvuII og Pstl. DNA-.sekvensen for en klon, pHGH107, ble bestemt.
Fjerning av EcoRI-setet i pGH107 vil etterlate ATG-startsignalet i samme avstand fra ribosombindingssete-kodonene i lac-promoteren som tilfellet er i naturen mellom slike kodoner og startsignalet for B-galaktosidase. For å bestemme om ekspresjon ville forøkes ved å etterligne dette naturlige mellomrom ble pGH107 omdannet til pGH107-l ved åpning av den førstnevnte med EcoRI, spalting av de resulterende enkeltkjedede ender med Sl-endenuklease, resirkularisering ved buttende-ligering med T4-ligase. Selvom det resulterende plasmid likeledes viste seg å kunne uttrykke HGH, gjorde det dette i mindre grad enn tilfellet var for pGH107, som vist ved direkte radioimmunobestemmelse.
Det er klart at variasjoner av det ovenfor angitte kan foretas avhengig blant annet av promotersystem, parenteralt plasmid, ønsket polypeptidprodukt, osv. Andre promotersystemer som kan anvendes 1 foreliggende oppfinnelse, inkluderer f.eks. X-promoteren, arabinose-operon (phi 80 d ara) eller kolisin El, galaktose, alkalisk fosfatase eller tryptofan-promotersystemer.
E. coil xl776 kan, selvom det er foretrukket brukt i forbindelse med foreliggende oppfinnelse utført i laboratoriemåle-stokk, vise seg å ha begrenset nyttevirkning i industriell stor målestokk p.g.a. svekkelser som med vilje er inkorporert deri av bio-sikkerhetsmessige grunner. Med passende nivåer av fysisk, snarere enn biologisk innhold, kunne slike organismer som E. coil K-12 stamme 294 supra og E. coli stamme RR1, genotype: Pro~Leu~Thi~R3-recA-Str<r>Lac y~, benyttes i operasjon i større målestokk. E. coli RR1 er avledet fra E. coli HB101 (E.W. Boyer et al., J. Mol. Bio (1969) 41 459-472) ved krysning med E. coli K12 stamme KL16 som Hfr-donor- Se J. E. Miller, Experlments in Molecular Genetlcs (Cold Spring Harbor, New York, 1972). En kultur av E. coli RR1 ble deponert den 30. oktober 1978 hos the American Type Culture Collection, uten begrensning med hensyn til adgang (ATCC No. 31343). En kultur av xl776 ble likeledes deponert den 3. juli 1979 hos the American Type Collection (ATCC No. 31537). Deponeringer av de følgende ble foretatt hos the American Type Culture Collection den 3. juli 1979: plasmid pHGH107 (ATCC No. 40011); plasmid pGH6 (ATCC No. 40012); stamme xl776 transformert med pHGH107 (ATCC No. 31538) og E. coli Kli stamme 294 transformert med pGH6 (ATCC No. 31539).
Claims (2)
1. Fremgangsmåte for fremstilling av et plasmid som i en bakteriell organisme kan uttrykke et spesielt polypeptid med kjent aminosyresekvens, hvorved et gen som koder for polypeptidet innføres i et plasmid og settes under kontroll av en ekspresjonspromoter,
karakterisert vedat man: a) tilveiebringer et cDNA-fragment som omfatter en vesentlig del av, men mindre enn hele den kodende sekvensen for nevnte polypeptid; b) gjennom organisk syntese tilveiebringer ett eller flere genfragmenter som koder for resten av aminosyresekvensen i polypeptidet, idet minst ett av nevnte fragmenter koder for amino-terminaldelen i polypeptidet; og c) innordner cDNA'et fra trinn a) og DNA<*>et oppnådd ved organisk syntese i trinn b) i et plasmid i riktig leseramme i forhold til hverandre og under kontroll av en ekspresjonspromoter ;
hvorved det dannes et plasmid som kan uttrykke aminosyresekvensen til nevnte polypeptid.
2. Fremgangsmåte ifølge krav 1,karakterisert vedat ved at humant veksthormon uttrykkes uledsaget av fremmed protein.
