Description

Liposomer er ganske bredt beskrevet i litteraturen, og deres struktur er velkjent. De har vanligvis en løk-lignende multilamellær struktur som omfatter en flerhet av fosfolipide biskikt som er skilt fra hverandre med vandig materiale. En annen liposom-type som er kjent, består av et enkelt fosfolipid biskikt som inneslutter vandig materiale, og disse unilamellære former blir noen ganger referert til som "blærer". Innkapslingen av en biologisk aktiv forbindelse i liposomer kan oppnås ved en rekke forskjellige fremgangsmåter. Dessuten kan liposomene aggregere og utfelles som et sediment. Aggregering og sediment-ering kan reduseres ved inkorporering av ladede lipider i liposomene, men det er ikke dermed garantert noen tilfreds-stillende lagringstid. Tapet av den biologisk aktive forbindelse fra liposomene og inn i det ytre vandige medium er en annen faktor som begrenser potensialet av disse preparater som praktiske dosisformer. Dette er spesielt tungtveiende for lav-molekylære, vann-løselige forbindelser, men lipid-løselige forbindelser kan også fordeles inn i det ytre vandige medium inntil likevekt er nådd. Dersom innholdet av forbindelse er lite, og/ eller volumet av det ytre vandige medium er stort, kan dette tap representere en betydelig andel av det totale innhold av den biologisk aktive forbindelse i liposomene.
Alle disse faktorer begrenser anvendelsen av liposomer som praktiske bærere for biologisk aktive forbindelser, spesielt ved medisinsk terapi. En løsning kan være å fremstille og lagre filmen av den lipid/biologisk aktive forbindelse, og så dispergere filmen for å danne liposomer efter behov like før anvendelse. Disse enhetsdosis-filmpreparater og lagringen medfører imidler-tid alvorlige praktiske vanskeligheter, og denne idé tilveiebringer derfor ikke noer. praktisk løsning på de problemer som er omtalt ovenfor. Når et frysetørket, potensialt liposom blanding fremstilt i henhold til denne oppfinnelse blir redispergert i et egnet vandig medium, for eksempel isotonisk saltløsning, blir det uventet dannet liposomer som er lik dem som fremstilles ved den kjente filmdispergeringsmetode. Eksempler på egnede amfipatiske lipider er naturlige lecitiner, f.eks. egglecitin eller soyabønne-lecitin, eller syntetiske lecitiner, for eksempel mettede syntetiske lecitiner, f.eks. dimyristoyl-fosfatidylcholin, dipalmitoy1-fosfatidylcholin eller distearoyl-fosfatidylcholin, eller umettede syntetiske lecitiner, f.eks. dioleyl-fosfatidylcholin eller dilinoleyl-fosfatidylcholin. Det kan anvendes enten et enkelt amfipatisk lipid eller en blanding av amfipatiske lipider.
Som angitt ovenfor kan den biologisk aktive forbindelse være hvilken som helst lipid-løselig eller lipid-bundet forbindelse som har en egenskap av biologisk interesse. De eventuelle adj uvant i a innbefatter (1) substanser

Løsnings-midlene blir værende faste under frysetørke-prosessen uten at det er nødvendig med utvendig avkjøling (utvendig avkjøling anvendes selvsagt ved prosessens start). Spesielt egnede løsningsmidler er t-butanol, dioksan, eddiksyre, pyridin og -piperidin. Det kan eventuelt også være til stede en eller flere andre væsker som hjelper tii med oppløsning av ingrediensene, for eksempel vann eller kloroform.
Liposomforløperen fremstilt ifølge oppfinnelsen kan hensiktsmessig anvendes til fremstilling av vandige liposom-preparater .

Eksempel 1
Dipalmitoyl-fosfatidylcholin (herefter "DPPC", 59,6 mg),
3 3 M-cortisol-21-palmitat (herefter " H-CP", 6,54 mg) og stearyl-aminacetat (3,81 mg) ble oppløst i redestillert t-butanol (3 ml) ved 60°C. Løsningsmidlet ble så fjernet under vakuum ved anvendelse av en kommersiell fryse-tørker. Det ble således oppnådd en fryse-tørket, potensialt liposom blanding som et pulver som kunne lagres i lukkede beholdere inntil det var behov for det.
Destillert vann (10 ml) ble satt til det fryse-tørkede pulver, og den resulterende blanding ble oppvarmet til ca. 70 C på et vannbad. Forsiktig risting av kolben forårsaket at pulveret dispergerte, og dette ga en melkeaktig suspensjon som viste seg å inneholde liposomer når den ble undersøkt mikroskopisk. Det ble tatt duplikat-prøver (50^ul) av suspensjonen for å foreta gnist-telling, for å bestemme steroid-innholdet før vasking. Resten av suspensjonen ble fortynnet til et volum på 25 ml med destillert vann, og-ble ultrasentrifugert ved 120.000 g i 30 minutter. Liposom-proppen ble dispergert til et volum på 10 ml med destillert vann, og det ble tatt duplikat-prøver (50^ul) av den vaskede suspensjon for å foreta gnist-telling. Sammenligning av telling-ene før og efter vasking viste at•90% av de gjenværende steroider i det fryse-tørkede pulver var inkorporert i de vaskede liposomer.
Eksempel 2
DPPC (49 mg) og <3>H-CP (7 mg) ble oppløst i redestillert t-butanol (5,05 ml) ved 60°C. Det ble tatt duplikat-prøver (5^ul) for å foreta gnist-telling, og den gjenværende løsning ble umiddelbart frosset på veggen .av en 250 ml's kolbe med rund bunn ved nedsenking i en fryseblanding (metanol og fast karbondioksyd). Så ble t-butanolen fjernet ved hjelp av en kommersiell fryse-tørker. Det ble således oppnådd en fryse-tørket, potensialt liposom blanding. Denne blanding ble skrapet bort fra kolbens vegg, og tre prøver (10, 11 og 12,7 mg) ble innveid i prøveglass. Det ble satt destillert vann (2,5 ml) til hvert prøveglass, og blandingen ble oppvarmet til 50°C på et vannbad. Blandingen ble så dispergert for å danne liposomer ved kraftig risting. Duplikat-prøver (50^ul) av hvert liposom-preparat ble tatt for å foreta gnist-telling. Liposom-preparatene ble innført i tørre, veide ultrasentrifuge-rør, fortynnet til 10 ml med destillert vann og ultrasentrifugert ved 120.000 g i 40 minutter ved 4°C.
De overflytende væsker ble fjernet fra lipid-proppene, og to av de tre propper ble tørket i en vakuumovn til en vekt som represen-terte 50 vekt% vanninnhold. Den tredje propp ble resuspendert i vann (2,5 ml) og det ble tatt duplikat-prøver (50^ul) for å foreta gnist-telling.
De radioaktive tellinger viste at efter dispersjon av de tre fryse-tørkede prøver i vann, var 86+4 % av det originale steroid til stede i dispersjonene. Tapet av radioaktivitet skyldtes tapet av en liten fraksjon av den potensialt liposome blanding under fryse-tørkingen. sfter at den tredje prøve var vasket, var det tilbake 73% av det totale steroid forbundet med liposomene.
Duplikat-prøver (6 mg) av begge de tørkede liposom-propper ble så veid inn i beholdere for å foreta differensial-avsøknings-kalorimetri (herefter "DSC"). DSC-spektrene av blandingene mellom 0 og 50°C ble nedskrevet på et Perkin Eimer differensial-avsøknings-kalorimeter. Det ble også dannet sammen-ligningsprøver for DSC ved å blande de samme vektmengder av DPPC og "^H-CP som i den opprinnelige blanding, og så blande dem med 50 vekt% vann. Disse tjente som "ikke-liposome" sammenligningsblandinger. DSC-spektrene for disse sammenligningsblandinger ble målt som beskrevet ovenfor.
DSC-spektret for DPPC alene består av en hoved-overgangs-endoterm ved 41°C og en pre-overgangs-endoterm ved 35°C. "Halv-spiss " -lin j ebredden av hoved-endotermen er tilnærmet 3°C. For-søkene beskrevet ovenfor viste at for de "ikke-liposome" sammenligningsblandinger ble begge spisser observert i DSC-spektrene i blandingene, og linjebredden forble ved ca. 3°C. Dette er antatt å vise at i enkle blandinger (dvs. ikke liposomer) forandrer ikke steroidet DSC-spektret til lipidet. - Spektrene av duplikat-liposom-preparatene.viste bare én overgang (hoved-endotermen), og den gjennomsnittlige linjebredde for disse preparater var 5,8°C, en betraktelig øket bredde sammenlignet med sammenligningsblandingene. Denne økede bredde kommer av molekylær innvirkning mellom lipidet og steroidet i liposomene fremstilt ved den ovenfor angitte fremgangsmåte. Det kan derfor ikke være noen tvil om at liposomer fremstilt ved fremgangsmåten ovenfor, inneholdt steroidet i liposomene.
Eksempel 3
Alle fremgangsmåter beskrevet i dette eksempel ble utført 1 et sterilt rom (sterilisert med formaldehyd, og så spylt med steril luft) ved anvendelse av konvensjonelle aseptiske forholds-regler. Fryse-tørkeren (Edwards Speedivac Centrifugal fryse-tørker, modell 5PS) var sterilisert med formaldehyd og så spylt med steril luft. De andre apparater som ble anvendt var enten sterilisert med tørr varme eller ved behandling i autoklav.
DPPC (697,2 mg), dipalmitoyl-fos fatidsyre (99,6 mg) og cortisol-21-palmitat (99,6 mg) ble oppløst ved 60°C i redestillert t-butanol (60 ml). Den varme løsning ble umiddelbart sterilisert ved å føres gjennom et 0,22^u 'MF-millipore' filter (Millipore Corporation, Bedford, MA, USA) holdt ved en temperatur på 50°C. Denne gjennomføring ble foretatt to ganger. 2 ml's aliquoter av den sterile løsning ble anbrakt i 5 ml's sterile multi-dose prøveglass. Innholdet i glassene ble fryse-tørket ved anvendelse av ovennevnte fryse-tørker, og hvert prøve-glass ble så lukket ved anvendelse av steril gummi-lukking og et multi-dose metalldeksel. Det ble således oppnådd innelukkede prøver av en steril, fryse-tørket, potensialt liposom blanding.
En steril 0,9% vekt/volum vandig løsning av natriumklorid ble innført i ett av de ovennevnte lukkede prøveglass, og dette ble så oppvarmet til 50°C på et vannbad, og ristet kraftig. Det ble således oppnådd et sterilt liposom-preparat som var egnet til administrasjon ved injeksjon.
Claims (3)
1. Fremgangsmåte for fremstilling av en liposom-forløper som kan dispergeres i et egnet vandig medium for fremstilling av et vandig liposom-preparat og som omfatter at minst ett liposom-dannende amfipatisk lipid, minst én lipid-løselig eller lipid-bundet biologisk aktiv forbindelse som kan være et legemiddel og eventuelt minst ett adjuvans oppløses i et løsningsmiddel for å danne en løsning, karakterisert ved at løsningen frysetørkes for å danne en frysetørket, potensial liposom blanding, idet det anvendes et løsningsmiddel som holder seg fast under frysetørkingen.
2. Fremgangsmåte ifølge krav 1, karakterisert ved at løsningsmidlet har et smeltepunkt som ligger nær rom-temperatur .
3. Fremgangsmåte ifølge krav 2, karakterisert ved at løsningsmidlet er t-butanol, n-butanol, dioksan, eddiksyre, pyridin eller piperidin.
