Claims (1)
- 【特許請求の範囲】 1. 細胞を解析するための下記のステップを含む方法: (a) 細胞が1種以上の蛍光レポーター分子を含有するときに複数の細胞を 含有する場所の配列を提供するステップ、 (b) 細胞内の蛍光レポーター分子からの蛍光シグナルを入手するために細 胞を含有する場所各々に含まれる多数の細胞をスキャンするステップ、 (c) 蛍光シグナルをデジタルデータに変換するステップ、及び (d) 細胞内の蛍光レポーター分子の分布、環境又は活性を測定するために デジタルデータを利用するステップ。 2. 場所の配列がマイクロタイタープレート内のウエルである請求項1に記 載の方法。 3. 場所の配列がマイクロプレート上のマイクロウエルである請求項1に記 載の方法。 4. 蛍光レポーターが細胞に添加される請求項1に記載の方法。 5. 蛍光レポーターが細胞によって産生される請求項1に記載の方法。 6. コンピューター手段がデジタルデータを平均細胞質レポーター分子蛍光 強度と平均核蛍光レポーター分子強度との差分に変換する請求項1に記載の方法 。 7. コンピューター手段がデジタルデータを核領域内の平均細胞質蛍光レポ ーター分子強度に変換する請求項1に記載の方法。 8. コンピューター手段がデジタルデータを細胞質マスク内の平均蛍光レポ ーター分子強度に変換する請求項1に記載の方法。 9. 複数の様々な蛍光レポーター分子が細胞内に存在する請求項1に記載の 方法。 10. 下記を含む細胞スクリーニングシステム: (a) 対物レンズ、細胞を保持するための場所の配列を備えたプレートを保 持するために適しているXYステージを有しており、さらにその場所と対物レン ズをアライメントするためにプレートを移動させるための手段及びフォーカシン グを実行するための方向にプレートを移動させるための手段を有する高倍率蛍光 光学系; (b) デジタルカメラ; (c) 励起光を場所の配列内の細胞に方向付けるための光源及び細胞から放 射された蛍光をデジタルカメラに方向付けるための手段; (d) デジタルカメラからのデジタルデータを受信して処理するための下記 を含むコンピューター手段:及び i) カメラから画像を受信するためのデジタルフレームグラバー、 ii) ユーザーインタラクション(交互作用)及 びアッセイ結果表示のためのディスプレイ、 iii) データの記憶及び保存を行うためのデジタル記憶媒体、及び iv) 結果の制御、収集、処理及び表示のための手段。 11. 蛍光レポーター分子を含む細胞のデータのグラフ及び画像を表示する ために、コンピューター手段と操作可能に接続されているPCスクリーンを有す る、請求項10に記載の細胞スクリーニングシステム。 12. コンピューター手段がバイオインフォーマティクス・データベースに データを記憶する請求項10に記載の細胞スクリーニングシステム。 13. さらに細胞を含有する場所の配列の各々において高スループットモー ドで複数の細胞をスキャンするステップ、及び細胞を含有するサブセットの場所 だけを高含量モードで選択的にスキャンするステップを含む請求項1に記載の方 法。 14. さらに複数の細胞を含有する場所の配列からシグナルを測定する取付 型リーダーと、複数の細胞を含む場所の配列と測定値の両方を細胞スクリーニン グシステムへ移すための方法を含む請求項10に記載の細胞スクリーニングシス テム。 15. 光学手段がシステムの倍率を変更するための機械的−光学手段を含む 請求項10に記載の細胞スクリーニングシステム。 16. さらに複数の細胞を含む場所の配列の周囲の温度、CO2濃度及び湿 度を維持するためのチャンバー及び制御システムを含む請求項10に記載の細胞 スクリーニングシステム。 17. 光学手段が同焦点走査型照明及び検出システムを含む請求項10に記 載の細胞スクリーニングシステム。 18. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図9に記載の手順を実 行することを惹起するための1セットの指示を含有するプログラムを含む機械可 読記憶媒体。 19. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図11に記載の手順を 実行することを惹起するための1セットの指示を含有するプログラムを含む機械 可読記憶媒体。 20. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図12に記載の手順を 実行することを惹起するための1セットの指示を含有するプログラムを含む機械 可読記憶媒体。 21. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図13に記載の手順を 実行することを惹起するための1セットの指示を含有するプログラムを含む機械 可読記憶媒体。 22. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図14に記 載の手順を実行することを惹起するための1セットの指示を含有するプログラム を含む機械可読記憶媒体。 23. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが図15に記載の手順を 実行することを惹起するための1セットの指示を含有するプログラムを含む機械 可読記憶媒体。 24. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが特異的な細胞の成分及 びプロセスの分布及び活性を検出するための手順を実行することを惹起するため の1セットの指示を含有するプログラムを含む機械可読記憶媒体。 25. 特異的な細胞プロセスがタンパク質の核トランスロケーションを含む 請求項24に記載の機械可読記憶媒体。 26. 前記タンパク質が膜タンパク質を含む請求項 25に記載の機械可読記憶媒体。 27. 特異的な細胞プロセスが細胞肥大を含む請求項24に記載の機械可読 記憶媒体。 28. 特異的な細胞プロセスがアポトーシスを含む請求項24に記載の機械 可読記憶媒体。 29. 特異的な細胞プロセスがタンパク質のプロテアーゼ誘発性トランスロ ケーションを含む請求項24に記載の機械可読記憶媒体。 30. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、細胞スクリーニングシステムが新規のレポーターアゴ ニスト及びアンタゴニストを識別するための手順を実行することを惹起するため の1セットの指示を含有するプログラムを含む機械可読記憶媒体。 31. 請求項10に記載の細胞スクリーニングシステムが図9に記載の手順 を実行することを惹起するための1セットの指示を含有するプログラムを含む機 械可読記憶媒体。 32. 請求項10に記載の細胞スクリーニングシステムが図11に記載の手 順を実行することを惹起するための1セットの指示を含有するプログラムを含む 機械可 読記憶媒体。 33. 請求項10に記載の細胞スクリーニングシステムが図12に記載の手 順を実行することを惹起するための1セットの指示を含有するプログラムを含む 機械可読記憶媒体。 34. 請求項10に記載の細胞スクリーニングシステムが図13に記載の手 順を実行することを惹起するための1セットの指示を含有するプログラムを含む 機械可読記憶媒体。 35. 請求項10に記載の細胞スクリーニングシステムが図14に記載の手 順を実行することを惹起するための1セットの指示を含有するプログラムを含む 機械可読記憶媒体。 36. 請求項10に記載の細胞スクリーニングシステムが図15に記載の手 順を実行することを惹起するための1セットの指示を含有するプログラムを含む 機械可読記憶媒体。 37. 請求項10に記載の細胞スクリーニングシステムが特異的な細胞の成 分及びプロセスの分布及び活性を検出するための手順を実行することを惹起する ための1セットの指示を含有するプログラムを含む機械可読記憶媒体。 38. 特異的な細胞プロセスがタンパク質の核トランスロケーションを含む 請求項37に記載の機械可読記憶媒体。 39. 前記タンパク質が膜タンパク質を含む請求項37に記載の機械可読記 憶媒体。 40. 特異的な細胞プロセスが細胞肥大を含む請求項37に記載の機械可読 記憶媒体。 41. 特異的な細胞プロセスがアポトーシスを含む請求項37に記載の機械 可読記憶媒体。 42. 特異的な細胞プロセスがタンパク質のプロテアーゼ誘発性トランスロ ケーションを含む請求項37に記載の機械可読記憶媒体。 43. 細胞スクリーニングシステムが細胞を保持するために適合したステー ジ及びそのステージを移動させるための手段、デジタルカメラ、デジタルカメラ からのデジタルデータを受信及び処理するための光源、及びデジタルカメラから のデジタルデータを受信及び処理するためのコンピューター手段を備えた高倍率 蛍光光学系を含むときに、請求項10に記載の細胞スクリーニングシステムが新 規のレポーターアゴニスト及びアンタゴニストを識別するための手順を実行する ことを惹起するための1セットの指示を含有するプログラムを含む機械可読記憶 媒体。
