図9を参照し、実行されるべきアッセイに基づいて選択されるべきオペレータ−指向性パラメータ、試料内の蛍光シグナルの分布についての細胞スクリーニングシステムによるデータ取得、および対話形式のデータレビューおよび解析を含む細胞を分析するための好ましい実施形態が提供される。自動走査の開始において、オペレータは、試料を説明する情報100を入力し、フィルタ設定および蛍光チャネルを測定して、使用される生物学的標識および求められる情報を適合させ、カメラ設定を調整して、試料の明るさを適合させる。ある範囲の試料を取り扱う柔軟性のためには、ソウトウェアは、核および細胞質を同定するのに使用される種々のパラメータ設定の選択および異なる蛍光試薬の選択、形態または明るさに基づく注目する細胞の同定、およびウェル当たりの分析すべき細胞の数の同定を可能とする。これらのパラメータは、各自動実行につき容易な検索のために、システムに保存される。このシステムの対話形式細胞同定モードは、分析すべき細胞のサイズ、形状および同一性の範囲等の形態学的パラメータの限界の選択を単純化する。使用者は、プレートのいずれのウェルをシステムが走査するか、およびいくつの視野およびいくつの細胞を各ウェルで分析すべきかを特定する。

細胞サイズの変化は、肥大、細胞付着および展延、分化、成長および分裂、壊死およびプログラムされた細胞死滅、細胞移動度、形態形成、チューブ形成、およびコロニー形成等の多数の細胞条件に関連する。
これらの種々の剤での細胞染色のためのプロトコルは当業者によく知られている。細胞を生きたまま、または固定後に染色し、細胞面積を測定することができる。例えば、DiIC16で染色した生細胞は均一に標識された原形質膜を有し、細胞の突出した断面積は色素の蛍光強度によってバックグラウンドから均一に区別される。CMFDA等のサイトゾル染料で染色した生細胞は、細胞の厚みに比例する蛍光強度を生じる。細胞標識は細胞の薄い領域では薄暗いが、全細胞面積はバックグラウンドから区別することができる。固定細胞は、重合されたアクチンを標識するALEXATM488ファロイジン等の細胞骨格マーカで染色することができる。ファロイジンは細胞質を均一に染色しないが、依然として、全細胞面積をバックグラウンドから区別する。
以下の戦略を用い、細胞肥大を分析するスクリーニングを実行する。初代ラット筋細胞を96ウェルプレート中で培養し、種々の化合物で処理し、次いで、固定し、ヘキスト等のDNA標識と組み合わせて、細胞表面マーカまたはローダミン−ファロイジン等の細胞骨格のための蛍光マーカに対する抗体等の、細胞膜または細胞質、または細胞骨格のための蛍光マーカで標識する。
細胞展延は、基質付着リガンドに対する細胞表面受容体の応答の尺度である。
以下は受容体の内部化の動力学を測定するためのスクリーニングの例である。
HCSの1つのクラスは、細胞内構成要素の薬物−誘導動的再分布を含む。ヒトグルココルチコイド受容体(hGR(細胞の複雑な環境応答機構における単一「センサー」)は、細胞に拡散したステロイド分子に結合する。リガンド−受容体複合体は核に移動し、そこで転写活性化が起こる(フトン(Htun)ら,Proc.Natl.Acad.Sci.93:4845,1996)。
アポトーシスは、膨大な分子事象および経路を含む複雑な細胞プログラムである。このプロセスに対する薬物作用のメカニズムを理解するには、時間および空間分解にて可能な限り多くのこれらの細胞内事象を測定するのが必須である。従って、細胞試料調製をほとんど要求しないアポトーシススクリーニングは、いくつかのアポトーシス−関連パラメータの自動読み出しを提供し、理想的であろう。細胞スクリーニングシステムのためにデザインされた細胞ベースアッセイは、パクリタキセル−誘導アポトーシスの形態学的、小器官およびマクロ分子ホールマークのいくつかを同時に定量するのに用いられてきた。
プラスミド作成物。緑蛍光タンパク質−カスパーゼ(コ−エン(Cohen)(1997)、Biochemical J.326:1〜16;リアング(Liang)ら(1997)J.of Molec.Biol.274:291〜302)キメラのコード配列を含む真核生物発現プラスミドを、GFP変異体を使用して調製する。この作成物を使用して、真核細胞をトランスフェクトする。
2つの材料および/または情報源が、この実施形態の活用に必要であり、これにより、特徴づけられていない遺伝子の機能の評価が可能となる。第1に、哺乳動物細胞へのトランスフェクションに適したcDNA配列を含む、疾病関連配列バンク(群)を使用できる。各RADEまたは差次的発現実験により、数100個までの配列が作成されるので、豊富にDASを供給することが可能である。第2に、一次配列データベース探索からの情報を使用して、DNAを、シグナル配列、7回膜貫通モチーフ、保存プロテアーゼ活性部位ドメイン、または他の同定可能なモチーフを含むカテゴリーを含むがこれに限定されない、広いカテゴリーに入れることができる。これらの源から獲得した情報に基づいて、トランスフェクトする方法の種類および指標細胞系を選択する。大量のモチーフがすでに十分に特徴づけられており、既存のゲノムデータベースの多くの遺伝子に含まれる線形配列にコードされている。

画像データは、固定または生指標細胞から得ることができる。得られた各画像から形態計測データを抽出するために、以下の解析法を使用する:
1.各核および細胞質画像を閾値処理し、核または細胞境界外の各ピクセルについて値=0を有するマスクを作成する
2.最初の画像上にマスクを重層し、フィールド(すなわち核または細胞)中の各物体を検出し、そのサイズ、形状、および積分強度を計算する
3.上で得られた全細胞マスクを、対応する発光微小管画像に重層し、以下の分類因子のセットの1つ以上を適用して、微小管形態および微小管形態に対する薬物の作用を決定する。

3.画像テクスチャーの測定値を使用した、微小管脱重合を定量するための分類因子
4.微小管の見かけの相互連結または分岐(または両方)を定量するための分類因子
5.試験化合物で処理した細胞の画像の時間シリーズに関する、上記の分類因子を使用した、微小管再構成の動力学の測定。
薬物発見についての主な興味は、ニューロンからの神経突起成長に影響を及ぼす化合物の同定である。神経成長を促進する薬物は、脊髄損傷、糖尿病および卒中などの疾病から生じる神経障害、パーキンソン病、および、アルツハイマー病を含む他の形態の痴呆を含むがこれに限定されない、神経障害および神経損傷をもたらす、多種多様の疾病および外傷の処置に有用である。
−細胞を含む位置のアレイを提供し、ここでの細胞は、細胞数を報告する少なくとも1つの第1の発光標識したレポータ分子、および、神経突起成長を報告する少なくとも1つの第2の発光標識したレポータ分子を有し、
−複数の細胞を含む各位置における複数の細胞を画像化または走査して、第1および第2の発光標識したレポータ分子から発光シグナルを得、
−発光シグナルを、デジタルデータに変換し、そして
−デジタルデータを使用して、自動的に測定を行ない、ここでの測定を使用して、細胞上または細胞内の、第1および第2の発光標識レポータ分子の分布、環境または活性の変化を自動的に計算し、ここでの計算した変化は、神経突起成長の測定値を提供することを含む。
1つの実施形態において、試料中の全細胞を、発光レポータ分子マーカで標識して、その位置を同定する。一旦細胞の位置が同定されると、細胞を計測できる。典型的には、核酸色素ヘキスト33342を、発光レポータ分子として使用して、全細胞の核を同定する。しかし、他の核標識も使用できる。核酸蛍光染色は2種類ある:生細胞の形質膜を通過できるもの、および、膜に不透過性であるもの。膜透過性核酸染色の例は、DAPI、ジヒドロエチジウム、ヨウ化ヘキシジウム、ヘキスト33258、およびSYTO(登録商標)色素シリーズ(モレキュラー・プローブ社)を含む。膜不透過性色素で核を標識するために、形質膜を透過化処理しなければならない。膜不透過性核酸色素の例は、シアニン核酸標識、例えば、TOTO(登録商標)、YOYO(登録商標)、BOBOTM、POPOTM、TO−PRO(登録商標)、YO−PRO(登録商標)、BO−PROTM、およびPO−PROTM(モレキュラー・プローブ社)、エチジウム類似体、例えばエチジウム−アクリジンヘテロダイマー、臭化エチジウム、エチジウムジアジド、およびエチジウムホモダイマー1および2、ヨウ化プロピジウム、および緑核酸染色SYTOX(登録商標)(モレキュラー・プローブ)を含む。さらに、ニューロンを低密度で蒔いた場合、細胞質などの細胞の他の成分を標識して、培養液中の全ての細胞を同定できる。好ましい実施形態において、核標識を使用する。いくつかの細胞質染色の例を以下に示す。
細胞を、慣用的な光学顕微鏡カバーガラスなどの、ガラス、プラスチック、またはシリコンウエハースを含むがこれに限定されない、任意の光学的に透明な材料から製造され得る、基質上に蒔く。特定の状況において、プラスチック基質は、細胞の良好な付着に十分である。しかし、いくつかの細胞型については、基質は、良好な付着および成長のために、特定の細胞外マトリックスでコーティングする必要がある。例えば、PC12細胞は、コラーゲン基質上で増殖する必要がある。次いで、試験する化合物(群)を細胞に加える。適切な時間の経過後、神経細胞を発光標識し(それらが以前に標識されていない場合)、次いで、画像を獲得し、自動的に解析して、下記のように神経突起成長を定量する。神経成長因子(NGF)で処理したPC12細胞を用いて実施したいくつかの実験では、細胞を発光標識し、画像化し、NGF処理の2から7日後に解析した。
神経細胞を最初に、上記のような基質上に蒔く。細胞を、試験すべき化合物、および、神経突起成長を刺激することが知られる対照化合物(例えば神経成長因子(NGF))で処理し、ここで、対照化合物での処理は、試験化合物処理の前、後、または同時に実施する。適切な時間の経過後、画像を獲得し、自動的に解析して、上記のように神経突起成長を定量する。
神経細胞を、最初に、上記のような基質上に蒔き、神経突起成長が可能となるように処理する。例えば、細胞を、上記のように、NGFと接触できる。神経突起成長が生じた後、細胞を、ニューロンおよび神経突起に対する毒性について試験する条件で処理する。かかる条件の例は、毒性の可能性のある濃度範囲の化合物の添加、細胞の増殖に重要な物理的パラメータ、またはいくつかの場合では、NGFなどの神経突起成長を刺激する因子の中止であり得る。適切な時間の経過後、画像を獲得し、下記したように自動的に解析し、神経突起成長を定量する。
神経細胞が低密度であるか、または高度の神経突起成長を有さない場合、個々の細胞を容易に同定できる。しかし、神経突起が成長し始め、細胞が数多くのプロセスを生じ始めると、これらのプロセスは交差し得、神経細胞は、大きな細胞のクラスターの一部となる。従って、全細胞クラスターが、1つの連結された発光実体となる。神経細胞体から成長する、異なるプロセスおよび構造は、軸索、樹状突起、神経突起、中間セグメント、末端セグメント、糸状仮足および成長錘状体を含む。画像獲得および解析のために、全てのプロセスおよび構造は、2つの群に分類される:(1)細胞体(神経細胞体としても知られる)および(2)神経突起。細胞体は、核を含み、粗く緻密で丸い形態を有する、ニューロンの中心部分である。細胞体から出現する全ての成長およびプロセスが、神経突起と分類される。神経突起は分岐し得るか、他の神経突起と交差し得るか、または、それから成長しているより小さなプロセスを有し得、その全てを、画像解析の目的のその親神経突起の一部と考える。従って、神経突起は、細胞体中にある1つの起源を有するが、それが分岐する場合には複数の終点を有し得る。本発明の方法の適用から得られた結果により、使用者は、その分類ガイドラインに従って神経突起を規定および分類できる。例えば、1つの刊行物において、軸索は、細胞体からの最も長い連続的な神経突起と定義され、0.7μmないし5.1μm長の間の神経突起セグメントは、糸状仮足と定義され、5.1μmより長いものは、細胞体から出現している場合には神経突起、または、末端が神経突起に付着している場合、末端セグメント突起と呼ばれる(ラマーカス(Ramakers)ら、1998、Developmental Brain Research、108:205〜216)。

a.好ましい実施形態(図25A―B)
方法への入力: 2つの画像を、方法への入力画像として提供する。:核チャネル画像および神経突起チャネル画像。核チャネル画像では、細胞の核を、ヘキスト33342またはいくつかの他の蛍光または発光核染色で標識する。神経突起チャネル画像では、神経細胞体およびその付着した神経突起を蛍光または発光標識する。
初期化相は、核およびニューロン画像の両方に対する、任意選択のバックグラウンド補正を用いて始める。バックグラウンド補正段階を適用して、不均一な照明の効果を低減し、検出を向上する。
バイナリ画像を、自動閾値の適用により、核チャネル画像から作成する。1つのニューロン核または核凝集塊あたり、1つの連結要素が核小体画像に存在する。各々の核の位置座標は、最初に、バックグラウンド補正した核画像にマスクとしてバイナリ核小体画像を適用し、次いで、ピーク検出慣行を使用して、ピーク最大強度を有するピクセルを選択することにより決定する。このピクセルは、各々の核の位置座標としてタグ化する。
バイナリ蓄積画像を、自動閾値処理の適用により、神経突起チャネル画像から作成する。この蓄積画像中のピクセルは、一般に、核小体画像のピクセルの上位集合である。
ニューロンは、細胞体から伸びている神経突起を有する細胞体からなる。各細胞体が1つの核を含み、細胞体は、核よりも広い面積を覆う。神経突起成長の定量を開始する前に、神経突起の源である、細胞体を同定する必要がある。従って、核小体画像(これはニューロンの核に対応する)の連結要素が拡張して、関連する神経細胞体が膨らむまで、各々の核のピークのピクセルの、一連の膨張を実施する。実施した膨張は、条件付膨張であり;膨張を適用する毎に、1つのピクセルの層を、その層中のピクセルがニューロン蓄積画像に存在する条件で、核小体に加える。これは、膨張に因る面積の増加は、依然として、細胞体の境界を越えて伸びていないことを意味する。各膨張中に、nfrontおよびnaddedの数を、核小体画像中の各連結要素について測定する。Nfrontは、単に追加のピクセルによる膨張である、単純な(条件のない)膨張により連結要素に加えられる、ピクセルの数である。Nfrontは、最新の膨張に因る物体の、ピクセル数で測定した、新しい円周と考えることができる。Naddedは、条件付膨張により実際に加えられる、ピクセルの数である。1ピクセルの膨張後に、蓄積画像で正である新しい円周のピクセルのみを計測するので条件付である。
種々の異なる量を、この方法により測定できる。第1に、細胞数およびニューロン数を測定および報告できる。各神経突起について、全長(全てのその分岐の長さの合計として測定)および分岐数を測定できる。各々の核について、それから出現する神経突起の数を測定できる。各クラスターについて、フォームファクタ(円周2÷(4π×面積))を測定し、神経突起成長度として報告する。さらに、各々の核からの神経突起長を合計し、閾値長よりも長い場合、核を、神経突起成長について正と同定する。これらの測定は、種々の方法で合わせて、表2に報告した出力特徴を出すことがでる。
1.PC6−3細胞における神経突起成長の測定
PC6−3細胞(PC12細胞のサブクローン)を、コラーゲンでコーティングしておいた、96ウェルマイクロプレートで増殖した。ウェルは、神経突起成長を刺激するための種々の濃度のNGF(神経成長因子)(0〜1000ng/ml)を含んだ。対照個体群は、NGFを含まなかった。2日後、細胞を固定し、間接的な免疫蛍光を、ウサギ抗βIII−チューブリン一次抗体およびALEXAFLUOR(商標)488コンジュゲートヤギ抗ウサギ二次抗体(モレキュラー・プローブ)を使用して、βIII−チューブリンに対して実施した。次いで、細胞を3.7%ホルムアルデヒド中で20分間固定し、固定溶液はまた、その核を標識するための10μg/mlのヘキスト33342を含んだ。細胞を、本発明の細胞スクリーニングシステムで画像化し、次いで、上記したプロトタイプ法で解析した。以下に示した結果は、NGF濃度の関数である、フォームファクタおよび平均神経突起長についてである。
2.PC12細胞の神経突起に対するドーパミン毒性の測定
PC12細胞を、1μg/mlのNGFの存在下で、コラーゲンIVでコーティングしたウェルを有する、96ウェルマイクロプレート上で7日間増殖した。
b.別の画像獲得実施形態(図26および27A―B参照)
別の実施形態において、この方法は、特定の状態、この場合、細胞型の混合した個体群を含む培養液中のニューロンである、フィールド中の細胞の比率を測定する。陽性状態は、細胞が明るく蛍光であることを意味し、陰性状態は、細胞が蛍光をほとんどまたは全く有さないことを意味する。ニューロン特異的レポータ分子を使用する場合、陽性状態は、どの神経細胞についても起こり、陰性状態は全ての他の細胞について起こる。ニューロンを同定するために、2つの画像を、上記に考察したように、1フィールドあたりで捉える。陽性状態の細胞数を、全細胞の総数と比較して、フィールド中の陽性状態の細胞の比率を得る。
検出閾値のコンピュータ計算
2つの対照ウェルを使用する:全細胞が陽性状態にある試料、および、全細胞が陰性状態にある試料。陽性状態の細胞を明るく標識し、陰性状態の細胞はそうしない。細胞セグメンテーションを向上するために、全ての画像を、ユーザーの決定した面積におよぶ局所平均強度を差し引くことにより、バックグラウンド補正できる。陰性対照では、細胞を含まないバックグラウンドおよび細胞の強度分布の変動を最小限にするように設定する。陽性対照では、細胞を含まないバックグラウンドおよび細胞の強度分布の変動を最小限にするように設定する。陽性状態検出閾値は、対照画像からコンピュータ計算した閾値の秤量合計として設定する。
核画像を、局所平均の差し引きにより、バックグラウンド補正する。閾値を画像に適用する。画像は、細胞を含まないバックグラウンドの薄いピクセルおよび細胞に関連したより明るいピクセルに因る、二項強度分布を有する。閾値は、これらの2つの分布の変動を最小限にするように設定する。8連結要素を標識および計測する。これにより、各々の核により覆われる面積が同定され、各々の個々の核のマスクが設定される。
核マスク画像の8連結要素を標識する。陽性状態の細胞を発光で標識した画像を、局所平均の差し引きにより、バックグラウンド補正する。次いで、陽性状態の細胞を、固定または自動閾値(陽性状態検出閾値より高いピクセルの選択)により選択する。
a.形態学的方法(図28):形態学的膨張(例えば5つのピクセルの)を選択した面積に適用する。選択した面積は、論理的に核マスクで「AND」し、次いで、得られた面積の8連結要素を標識および計測する。
各々のウェルについて、検出された核の数および陽性状態(すなわちニューロンである)の核の数を、記録および報告する。1ピクセルあたりの全積分強度および平均強度も報告できる。次に、神経突起成長法を、陽性状態の神経細胞に適用して、その神経突起成長を定量および特徴づける。陽性状態の核を使用して、陽性状態の細胞を係数化し、追跡する。
神経突起成長度を測定するために、神経細胞または細胞クラスターの円周および面積の両方を測定する。神経突起成長度を定量するために、我々は、上記で考察したような、フォームファクタ(FF)を使用する。関与する一連の段階の要約は以下の通りである:
1.バックグラウンド補正:好ましい実施形態と同じ
2.画像バイナリ化:その後、画像をバイナリ化して、全ての選択した細胞を用いてマスク画像を作成する。
1.PC12細胞における神経突起成長の測定

実施形態11 追加のスクリーン 形質膜と細胞質の間の転位置
プロフィラクチン複合体の解離およびプロフィリンの形質膜への結合
1つの実施形態において、プロフィリン膜結合の蛍光タンパク質バイオセンサーを、精製プロフィリン(フェデロブ(Federov)ら(1994)J.Molec.Biol.241:480〜482;ランブレッツ(Lanbretchts)ら(1995)Eur.J.Biochem.230:281〜286)を、2〜300n秒の範囲の蛍光寿命を有するプローブで標識することにより調製する。標識プロフィリンを、バルク添加方法を使用して、指標生細胞に導入し、指標細胞を、試験化合物で処理する。蛍光異方性画像化顕微鏡(ゴー(Gough)およびテーラー(1993)J.Cell.Biol.121:1095〜1107)を使用して、0.1秒から10時間までの範囲の処理後のある期間、細胞質と膜の間の、プロフィリンの蛍光誘導体の、試験化合物依存的な移動を測定する。
別の実施形態において、指標細胞を試験化合物で処理し、次いで、固定、洗浄および透過化処理した。指標細胞形質膜、細胞質、および核を全て、別個の色のマーカで標識し、次いで、Rhoタンパク質(セルフ(Self)ら(1995)Methods in Enzymology 256:3〜10;タナカ(Tanaka)ら(1995)、Methods in Enzymology 256:41〜49)を、第4の色で標識した抗体で抗原局在化する。4つの各標識を、細胞スクリーニングシステムを使用して別々に画像化し、この画像を使用して、試験化合物により奏効される転位置の阻害または活性化の量を計算する。この計算を行なうために、形質膜および細胞質に印しを付すのに使用したプローブの画像を使用して、免疫学的プローブの画像をマスクし、細胞内Rhoタンパク質の位置に印しを付す。各マスク下での単位面積あたりの積分輝度を使用して、形質膜積分輝度/面積を、細胞質積分輝度/面積で割ることにより、転位置指数を形成する。対照および実験ウェルの転位置指数値を比較することにより、転位置の比率を、各々の可能性あるリード化合物について計算する。

RNAをベースとした蛍光バイオセンサー
細胞骨格タンパク質の転写およびメッセージ局在化。細胞−基質粘着、細胞−細胞粘着、シグナル伝達、細胞周期事象、中間およびシグナル伝達分子の代謝、細胞歩行運動、細胞−細胞伝達、および細胞死を含む、一般的なクラスの細胞生理的応答の調節は、遺伝子発現の変化を含み得る。高含量スクリーンはまた、このクラスの生理的応答を測定するように設計できる。
生細胞におけるその細胞表面受容体への標識インスリンの結合。形質膜ドメインを特定の色の標識試薬で標識しておいた細胞を、適切な条件下で、適切な時間、異なる色の発光プローブで標識した、インスリン分子を含む溶液と共にインキュベートする(リーら(1997)Biochemistry 36:2701〜2708;マルチネス-ザグイラン(Martinez-Zaguilan)ら(1996)Am.J.Physiol.270:C1438〜C1446)。インキュベート後、非結合インスリン分子を洗浄して除去し、細胞を固定し、インスリンの形質膜上での分布および濃度を測定する。これを行なうために、細胞膜画像を、インスリン画像のマスクとして使用する。マスクしたインスリン画像からの積分強度を、既知量の標識インスリンを含む1セットの画像と比較する。細胞に結合したインスリンの量を、標準から決定し、細胞と共にインキュベートしたインスリンの総濃度と共に使用して、解離定数またはその細胞表面受容体へのインスリンを計算する。
全細胞標識
全細胞標識は、細胞の形状の動力学および細胞の移動を、細胞の蛍光画像を解析することにより、時間をかけて測定できるように、細胞成分を標識することにより行なわれる。
1つの実施形態において、全形質膜の標識は、全細胞の標識について上記したのと同じ方法のいくつかを使用する。全細胞表面を標識する発光分子は、形質膜を描写するように作用する。
1つの実施形態において、受容体により媒介されるエンドサイトーシスにより細胞に輸送されるリガンドを使用して、エンドソーム細胞小器官の動力学を追跡する。標識リガンドの例は、Bodipy FL標識低密度リポタンパク質複合体、テトラメチルローダミントランスフェリン類似体、および蛍光標識した表皮増殖因子(モレキュラー・プローブ社)を含む。
1つの実施形態において、膜透過リソソーム特異的発光試薬を使用して、生および固定細胞のリソソーム区分を標識する。これらの試薬は、発光分子ニュートラルレッド、N−(3−((2,4−ジニトロフェニル)アミノ)プロピル)−N−(3−アミノプロピル)メチルアミン、および、リソソーム内のpHを報告するLyso Trackerプローブ、並びに、リソソーム(モレキュラー・プローブ社)の動力学的分布を含む。
1つの実施形態において、反応性基を有する細胞透過蛍光色素(モレキュラー・プローブ社)を生細胞と反応させる。モノブロモビマン、5−クロロメチルフルオレセインジアセテート、カルボキシフルオレセインジアセテートスクシンイミジルエステル、およびクロロメチルテトラメチルローダミンを含む、反応性色素は、細胞の細胞質の長期標識に使用する、細胞透過性蛍光色素の例である。
1つの実施形態において、膜透過性核酸特異的発光試薬(モレキュラー・プローブ社)を使用して、生および固定細胞の核を標識する。これらの試薬は、シアニンをベースとした色素(例えば、TOTO(登録商標)、YOYO(登録商標)、およびBOBOTM)、フェナチジンおよびアクリジン(例えば、臭化エチジウム、ヨウ化プロピジウム、およびアクリジンオレンジ)、インドールおよびイミダゾール(例えば、ヘキスト33258、ヘキスト33342、および4’,6−ジアミジノ−2−フェニルインドール)および他の類似試薬(例えば、7−アミノアクチノマイシンD、ヒドロキシスチルバミジンおよびプソラレン)を含む。
1つの実施形態において、膜透過性ミトコンドリア特異的発光試薬(モレキュラー・プローブ社)を使用して、生および固定細胞のミトコンドリアを標識する。これらの試薬は、ローダミン123、テトラメチルロサミン、JC−1、およびMito Tracker反応性色素を含む。
1つの実施形態において、膜透過性小胞体特異的発光試薬(モレキュラー・プローブ社)を使用して、生および固定細胞の小胞体を標識する。これらの試薬は、短鎖カルボシアニン色素(例えばDiOC6およびDiOC3)、長鎖カルボシアニン色素(例えばDiIC16およびDiIC18)および発光標識したレクチン、例えばコンカナバリンAを含む。
1つの実施形態において、膜透過性ゴルジ体発光試薬(モレキュラー・プローブ社)を使用して、生および固定細胞のゴルジ体を標識する。これらの試薬は、発光標識した巨大分子、例えば、小麦凝集素およびブレフェルディンA、並びに、発光標識したセラミドを含む。
Claims (15)
- 神経突起成長を解析する自動化された方法であって、
a)細胞位置を報告する少なくとも1つの第1の発光標識したレポータ分子と、前記神経突起成長を報告する少なくとも1つの第2の発光標識したレポータ分子とを有する細胞を含む位置のアレイを与えるステップと、
b)前記少なくとも1つの第1の発光標識したレポータ分子から核画像を取得し、前記少なくとも1つの第2の発光標識したレポータ分子から神経突起画像を取得するステップと、
c)前記核画像から細胞体を自動的に特定するステップと、
d)前記細胞体から伸長する神経突起を自動的に特定するステップであって、
I)前記神経突起画像から蓄積画像を作成するステップと、
II)前記蓄積画像中の前記細胞体に存在しない正のピクセルであって、前記細胞体から伸長する神経突起に属する正のピクセルを特定するステップと
を含むステップと、
e)以下からなる群から選択された1つ以上の神経突起の特徴を自動的に決定するステップであって、
i)全細胞からの全神経突起長、
ii)全細胞からの神経突起分岐の総数、
iii)1細胞あたりの神経突起の数、
iv)1つの正のニューロンあたりの神経突起の数、
v)各細胞の神経突起長、
vi)1つの正のニューロンあたりの神経突起長、
vii)1つの神経突起あたりの神経突起長、
viii)神経突起成長について正である細胞の数、
ix)神経突起成長について正である細胞の比率、
x)1つのニューロンあたりの分岐の数、及び
xi)1つの神経突起あたりの分岐の数
前記特徴は前記細胞体からの神経突起成長の尺度を与えるステップと
を含む方法。 - 前記細胞体を自動的に特定するステップは、
A)前記核画像から核小体画像を作成するステップと、
B)前記核小体画像の条件付膨張を実施して前記細胞体を特定するステップと
を含む、請求項1に記載の方法。 - (a)前記核小体画像の1回の条件付膨張を実施して膨張画像を取得するステップと、
(b)前記膨張画像から1セットの節を決定するステップと、
(c)連結された節を繋ぐステップと、
(d)全神経突起長を追跡するまでステップ(a)から(c)を繰返すステップと
をさらに含む、請求項2に記載の方法。 - 複数の時点においてステップ(a)から(d)を繰り返すことをさらに含む、請求項3に記載の方法。
- 前記細胞を試験化合物と接触させるステップと、前記細胞体からの神経突起成長に対する前記試験化合物の影響を決定するステップとをさらに含む、請求項1に記載の方法。
- 前記細胞を、前記試験化合物との接触の前、後、又は同時に、神経毒と接触させるステップをさらに含む、請求項5に記載の方法。
- 前記細胞を、前記神経突起成長を刺激することが知られている対照化合物と接触させるステップと、前記対照化合物が前記細胞体の前記神経突起成長を誘導するのを前記試験化合物が阻止するかどうかを決定するステップとをさらに含む、請求項5に記載の方法。
- 複数の時点においてステップb)からe)を繰り返すことをさらに含む、請求項1に記載の方法。
- 前記第1の発光標識したレポータ分子は、DNA結合化合物を含む、請求項1に記載の方法。
- 前記第2の発光標識したレポータ分子は、ニューロン特異的である、請求項1に記載の方法。
- 前記ニューロン特異的な発光レポータ分子が、神経フィラメントタンパク質と、βIII−チューブリンと、毛様体神経栄養因子と、神経フィラメントタンパク質に特異的な抗体とからなる群から選択した分子を含む、請求項10に記載の方法。
- 請求項1から11のいずれか1つに記載の方法を細胞スクリーニングコンピュータシステムに実行させるための1セットの命令を含むプログラムを備えるコンピュータ解読可能な記録媒体。
- 前記膨張画像に対して複数回の条件付膨張が前記核小体画像のピクセルと連続する前記蓄積画像の残りのピクセルがなくなるまで連続して行われ、
1つの節と1つの既存の神経突起物体との会合が形成されるのは1つの節の1つ以上のピクセルが前記神経突起物体のピクセルに隣接する場合であり、
1つの節が2つ以上の神経突起物体と会合する場合に前記神経突起物体は接続点を表し、
複数の節が1つの神経突起物体と会合する場合に前記神経突起物体は分岐点を表し、
1つの節が1つのみの神経突起物体と会合し、かつ、前記神経突起物体が前記節とのみ会合する場合に前記節は前記神経突起物体の伸長である、請求項4に記載の方法。 - (i)前記蓄積画像を解析してトリプル点を特定するステップと、
(ii)前記蓄積画像から前記トリプル点を取り出して前記トリプル点における分岐と亜分岐とを分離するステップと、
(iii)連結要素標識を行って前記分岐及び前記亜分岐の数を計測するステップと
をさらに含む、請求項1から3のいずれか1つに記載の方法。 - 前記細胞のフォームファクタを自動的に決定するステップをさらに含む、請求項1から3のいずれか1つに記載の方法。
