Description
Claims (18)
- 対象物を含む蛍光画像を取得する蛍光画像取得部と、
前記対象物を含む干渉画像を取得する干渉画像取得部と、
前記干渉画像取得部により取得された干渉画像に基づいて光学的厚さ画像を求め、前記蛍光画像取得部により取得された蛍光画像および前記光学的厚さ画像の双方に共通に設定した関心領域において、前記蛍光画像の蛍光強度の積分値と前記光学的厚さ画像の光学的厚さの積分値とに基づいて前記対象物の蛍光発現率を求める演算部と、
を備える蛍光測定装置。 - 前記演算部は、前記関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値を、前記関心領域内の前記対象物の蛍光発現率の指標として求める、
請求項1に記載の蛍光測定装置。 - 前記演算部は、第1閾値以下である蛍光発現率を有する第1対象物のみを含む第1関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値、第2閾値以上である蛍光発現率を有する第2対象物のみを含む第2関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値、および、測定対象の関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値に基づいて、前記測定対象の関心領域内における前記第1対象物および前記第2対象物の混合割合を求める、
請求項2に記載の蛍光測定装置。 - 前記演算部は、前記光学的厚さ画像に基づいて関心領域を設定する、
請求項1〜3の何れか1項に記載の蛍光測定装置。 - 前記演算部は、前記光学的厚さ画像の情報を用いて前記蛍光画像に対して背景補正およびシェーディング補正を行い、当該補正後の蛍光画像の蛍光強度の積分値を求める、
請求項1〜4の何れか1項に記載の蛍光測定装置。 - 前記干渉画像取得部は、インコヒーレント光を用いて干渉画像を取得する、
請求項1〜5の何れか1項に記載の蛍光測定装置。 - 前記干渉画像取得部および前記蛍光画像取得部それぞれの光学系の少なくとも一部が共通である、
請求項1〜6の何れか1項に記載の蛍光測定装置。 - 前記蛍光画像取得部は、対象物の集合体を含む蛍光画像を取得し、
前記干渉画像取得部は、前記集合体を含む干渉画像を取得し、
前記演算部は、前記蛍光画像取得部により取得された蛍光画像のうちの前記集合体の蛍光の積分値と、前記干渉画像取得部により取得された干渉画像から求められた光学的厚さ画像のうちの前記集合体の光学的厚さの積分値とに基づいて、前記集合体の蛍光発現率を求める、
請求項1〜7の何れか1項に記載の蛍光測定装置。 - 対象物を含む蛍光画像を蛍光画像取得部により取得する蛍光画像取得ステップと、
前記対象物を含む光学的厚さ画像を干渉画像取得部により取得する干渉画像取得ステップと、
前記干渉画像取得部により取得された干渉画像に基づいて光学的厚さ画像を求め、前記蛍光画像取得部により取得された蛍光画像および前記光学的厚さ画像の双方に共通に設定した関心領域において、前記蛍光画像の蛍光強度の積分値と前記光学的厚さ画像の光学的厚さの積分値とに基づいて前記対象物の蛍光発現率を求める演算ステップと、
を備える蛍光測定方法。 - 前記演算ステップにおいて、前記関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値を、前記関心領域内の前記対象物の蛍光発現率の指標として求める、
請求項9に記載の蛍光測定方法。 - 前記演算ステップにおいて、第1閾値以下である蛍光発現率を有する第1対象物のみを含む第1関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値、第2閾値以上である蛍光発現率を有する第2対象物のみを含む第2関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値、および、測定対象の関心領域内の蛍光強度の積分値を光学的厚さの積分値で除算した値に基づいて、前記測定対象の関心領域内における前記第1対象物および前記第2対象物の混合割合を求める、
請求項10に記載の蛍光測定方法。 - 前記演算ステップにおいて、前記光学的厚さ画像に基づいて関心領域を設定する、
請求項9〜11の何れか1項に記載の蛍光測定方法。 - 前記演算ステップにおいて、前記光学的厚さ画像の情報を用いて前記蛍光画像に対して背景補正およびシェーディング補正を行い、当該補正後の蛍光画像の蛍光強度の積分値を求める、
請求項9〜12の何れか1項に記載の蛍光測定方法。 - 前記干渉画像取得ステップにおいて、前記干渉画像取得部がインコヒーレント光を用いて干渉画像を取得する、
請求項9〜13の何れか1項に記載の蛍光測定方法。 - 前記干渉画像取得部および前記蛍光画像取得部それぞれの光学系の少なくとも一部が共通である、
請求項9〜14の何れか1項に記載の蛍光測定方法。 - 前記蛍光画像取得ステップにおいて、対象物の集合体を含む蛍光画像を前記蛍光画像取得部により取得し、
前記干渉画像取得ステップにおいて、前記集合体を含む干渉画像を前記干渉画像取得部により取得し、
前記演算ステップにおいて、前記蛍光画像取得部により取得された蛍光画像のうちの前記集合体の蛍光の積分値と、前記干渉画像取得部により取得された干渉画像から求められた光学的厚さ画像のうちの前記集合体の光学的厚さの積分値とに基づいて、前記集合体の蛍光発現率を求める、
請求項9〜15の何れか1項に記載の蛍光測定方法。 - 前記対象物が細胞である、
請求項9〜16の何れか1項に記載の蛍光測定方法。 - 前記蛍光画像取得ステップにおいて、前記対象物としての細胞を各ウェルに入れたマルチウェルプレートを前記蛍光画像取得部によりタイリング撮影して蛍光画像を取得し、
前記干渉画像取得ステップにおいて、前記マルチウェルプレートを前記干渉画像取得部によりタイリング撮影して干渉画像を取得し、
前記演算ステップにおいて、前記蛍光画像および前記光学的厚さ画像それぞれにおいて前記マルチウェルプレートの各ウェルに関心領域を設定する、
