Claims (14)
1. Polipeptid koji sadrži molekule bazirane na građevnim elementima koje sadrže domene petlji koje su topološki slične domenama trećeg fibronektin domena, polipeptid ima sekvencu aminokiseline baziranu na konsenzus sekvenci trećeg fibronektin domena koja ima:

2. 4. Izolirana proteinska građa prema patentnom zahtjevu 3, naznačena time da navedene građa veže metu s najmanje jednim afinitetom izabranim iz KD za manje ili jednako 10-9M, manje ili jednako 10-10M, manje ili jednako 10-11M, manje ili jednako 10-12M, manje ili jednako 10-13M, manje ili jednako 10-14M, i manje ili jednako 10-15M, kao što je određeno rezonancijom površinskih plazmona ili KinExA metodom, kao i gdje petlje formiraju mjesta za vezivanje za IgG.

6. Metoda iz patentnog zahtjeva 5, pri čemu su nasumični kodoni izabrani iz grupe NNS i NNK, tako da su nasumični kodoni uvršteni na pozicijama kodiranja najmanje jedne regije petlje izabranih iz grupe koja se sastoji od ostataka u/ ili oko pozicija 13-16, 22-28, 38-43, 51-54, 60-64, i 75-81 od SEQ ID BR: 142, 143 i 147-151.
7. Biblioteka proizvedena metodom iz patentnog zahtjeva 5 ili patentnog zahtjeva 6, opcionalno, pri čemu su:
(i) polinukleotidi biblioteke operativno vezani za prikazanu sekvencu kodiranja proteina i uvedeni su u ekspresijski vektor za prikaz, i izraženi kao fuzioni protein; ili
(ii) prikazani protein je izabran iz proteina usidrenog u eukariotsku ćelijsku membranu, proteina prokariotske ćelije i fage omotača proteina.
8. Metoda generiranja vezanja platformskog proteina za određenu metu sa unaprijed definiranim afinitetom vezivanja, koja sadrži kontaktiranje biblioteke iz patentnog zahtjeva 7, sa određenom metom, i izoliranje platformskog proteina koji se veže za određenu metu sa unaprijed definiranim afinitetom, opcionalno, pri čemu je:
(i) korak izoliranja sastavljen od izoliranja platformskih molekula, vezanja za određenu metu i testiranja afiniteta vezanja izoliranih platformskih molekula za specifičnu metu, kao i pri čemu je korak izoliranja sastavljen od pretraživanja biblioteke sa određenom metom, hvatanja platformskih molekula, koja se vezuje za određenu metu i izoliranja vezujućih platformskih molekula;
(ii) afinitet, mjeren sa KD je manji ili jednak približno 10-7M.
9. Izolirana molekula nukleinske kiseline kodiranog platformskog proteina prema bilo kojem od patentnih zahtjeva od 1-4.
