Description
PATENTKRAV 1. Fremgangsmåde til fremstilling af erythropoietin ved følgende trin: (a) dyrkning i et egnet medium af eukaryote celler, som indeholder en DNA-sekvens, der koder for erythropoietin, og (b) isolering og separering af det erythropoietin, der produceres fra cellerne og mediet, med det forbehold, at en fremgangsmåde, som ikke resulterer i erythropoietin, som inkorporerer fucose og N-acetylgalac-tosamin, er udelukket. 2. Fremgangsmåde til fremstilling af rekombinant erythropoietin ved følgende trin: (a) dyrkning i et egnet medium af eukaryote celler, som -operativt bundet til en ekspressionskontrolsekvens - indeholder en DNA-sekvens, der koder for erythropoietin, og (b) isolering og separering af det således producerede erythropoietin fra cellerne og mediet, kendetegnet ved, at der anvendes eukaryote celler, som har evne til at producere N- og O-bundet glycosylering med inkorporering af fucose og N-acetylgalactosamin, og at ery-thropoietinet med N- og O-bundet glycosylering isoleres og adskilles fra cellerne og mediet. 3. Fremgangsmåde til fremstilling af rekombinant erythropoietin ved følgende trin: (a) dyrkning i et egnet medium af CHO-celler, som - operativt bundet til en ekspressionskontrolsekvens - indeholder en DNA-sekvens, der koder for erythropoietin, og (b) isolering og separering af det således producerede erythropoietin fra cellerne og mediet, kendetegnet ved, at der anvendes CHO-celler, som har evne til at producere N- og O-bundet glycosylering med inkorporering af fucose og N-acetylgalactosamin, og at erythropoieti-net med N- og O-bundet glycosylering isoleres og adskilles fra cellerne og mediet. 4. Fremgangsmåde ifølge krav 2 eller 3, hvor det rekombinan-te erythropoietin har et glycosyleringsmønster, der omfatter relative molære niveauer af hexoser i forhold til N-acetylgalactosamin (Nacglc) på 1,4:1, specifikt galactose:Nacglc = 0,9:1 og mannose:Nacglc = 0,5:1.
