Description
- Die vorliegende Erfindung betrifft die Verwendung substituierter 4-Aminopyrimidine zur antimikrobiellen Behandlung von Oberflächen, als antimikrobielle Wirksubstanz gegen grampositive und gramnegative Bakterien, Hefen und Pilze sowie zur Konservierung von Kosmetika, Haushaltsprodukten, Textilien und Kunststoffen und zur Verwendung in Desinfektionsmitteln.
- Aminopyrimidinderivate und deren Verwendung werden in
EP 0 323 757 ,EP 0 407 899 ,EP 0 519 211 ,WO 95/07278 US 6207668 ,EP 0 424 125 undUS 4 435 402 beschrieben. Keines dieser Dokumente offenbart die Verwendung der speziellen Verbindungen der vorliegenden Erfindung. - Die vorliegende Erfindung bezieht sich auf die Verwendung von 4-Aminopyrimidinen der Formel R1 Wasserstoff; C1-C5-Alkyl, das nicht substituiert oder durch ein oder mehrere Halogenatome substituiert ist; Biphenyl oder C6-C10-Aryl, das nicht substituiert oder durch Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder durch Amino substituiert ist; einen Pyrrol-, Furan-, Thiophen-, Imidazol-, Pyrazol-, 1,2,3-Triazol-, 1,2,4-Triazol-, Pyridazin-, Pyrimidin-, Pyrazin-, Pyran-, Thiopyran-, 1,4-Dioxan-, 1,2-Oxazin-, 1,3-Oxazin-, 1,4-Oxazin-, Indol-, Benzothiophen-, Benzofuran-, Pyrrolidin-, Piperidin-, Piperazin-, Morpholin- oder Thiomorpholinrest; oder Cyclo-C3-C7-alkyl;

zur antimikrobiellen Behandlung von Oberflächen. - C1-C20-Alkylreste sind geradkettige oder verzweigte Alkylreste wie z.B. Methyl, Ethyl, n-Propyl, Isopropyl, n-Butyl, sec-Butyl, tert-Butyl, Amyl, Isoamyl oder tert-Amyl, Heptyl, Octyl, Isooctyl, Nonyl, Decyl, Undecyl, Dodecyl, Tetradecyl, Pentadecyl, Hexadecyl, Heptadecyl, Octadecyl oder Eicosyl.
- C3-C7-Cycloalkyl bedeutet z. B. Cyclopropyl, Cyclobutyl, Cyclopentyl, Cyclohexyl oder Cycloheptyl. Diese Reste können substituiert sein, z. B. durch einen oder mehrere gleiche oder verschiedene C1-C4-Alkylreste, insbesondere durch Methyl, und/oder durch Hydroxy. Sind Cycloalkylreste durch einen oder mehrere Substituenten substituiert, so sind sie bevorzugt durch einen, zwei oder vier, insbesondere durch einen oder zwei, gleiche oder verschiedene Substituenten substituiert.
- C1-C5-Alkoxyreste sind geradkettige oder verzweigte Reste, wie z. B. Methoxy, Ethoxy, Propoxy, Butoxy oder Pentyloxy.
- C6-C10-Aryl- und Heteroarylreste können nicht substituiert sein oder einen oder mehrere, z. B. ein, zwei, drei oder vier, gleiche oder verschiedene Substituenten tragen, die sich in beliebigen Positionen befinden können. Beispiele für solche Substituenten sind z. B. C1-C4-Alkyl, Halogen, Hydroxy, C1-C4-Alkoxy, Trifluormethyl, Cyano, Hydroxycarbonyl, C1-C4-Alkoxycarbonyl, Aminocarbonyl, Amino, C1-C4-Alkylamino, Di-C1-C4-alkylamino und C1-C4-Alkylcarbonylamino.
- Heteroarylreste leiten sich von Heterocyclen ab, die ein zwei, drei oder vier gleiche oder verschiedene Ringheteroatome enthalten, besonders bevorzugt von Heterocyclen, die ein, zwei oder drei, insbesondere ein oder zwei, gleiche oder verschiedene Heteroatome enthalten. Die Heterocyclen können mono- oder polycyclisch sein, z. B. mono-, bi- oder tricyclisch. Bevorzugt sind sie mono- oder bicyclisch, insbesondere monocyclisch. Die Ringe enthalten bevorzugt 5, 6 oder 7 Ringglieder. Beispiele für monocyclische und bicyclische heterocyclische Systeme, von denen sich in den Verbindungen der Formel (1) vorkommende Reste ableiten können, sind z. B. Pyrrol, Furan, Thiophen, Imidazol, Pyrazol, 1,2,3-Triazol, 1,2,4-Triazol, Pyridin, Pyridazin, Pyrimidin, Pyrazin, Pyran, Thiopyran, 1,4-Dioxan, 1,2-Oxazin, 1,3-Oxazin, 1,4-Oxazin, Indol, Benzothiophen, Benzofuran, Pyrrolidin, Piperidin, Piperazin, Morpholin und Thiomorpholin.
- Ungesättigte Heterocyclen können z. B. eine, zwei oder drei ungesättigte Doppelbindungen im Ringsystem enthalten. 5-gliedrig Ringe und 6-gliedrige Ringe in monoyclischen und polycyclischen Heterocyclen können insbesondere auch aromatisch sein.
- Halogen bedeutet Fluor, Chlor, Brom oder Iod, vorzugsweise Fluor oder Chlor.
- Vorzugsweise werden erfindungsgemäß Verbindungen der Formel (1) verwendet, wobei R5 gegebenenfalls durch eine oder mehrere -O- oder-Gruppen oder einen bivalenten heterocyclischen Rest unterbrochenes R''R'''N-C1-C20-Alkyl;
R' Wasserstoff oder C1-C5-Alkyl;
R'' und R''' unabhängig voneinander Wasserstoff oder Methyl
bedeuten; und
R1, R2, R3 und R4 wie für Formel (1) definiert sind. -

- Unter diesen Verbindungen sind solche bevorzugt, wobei
R5 R''R'''N-C5-C20-Alkyl; und
R'' und R''' unabhängig voneinander Wasserstoff oder Methyl
bedeuten. - Bevorzugt werden auch Verbindungen der Formel (1) verwendet, wobei
R4 und R5 zusammen mit dem Stickstoffatom einen Rest der Formel (1a) bilden, worin
R'''' Wasserstoff oder Pyridyl-C1-C3-alkyl; und
n1 und n2 in jedem Fall 2
bedeuten. - Bevorzugt verwendet werden auch Verbindungen der Formel (1), wobei
R1 und R2 unabhängig voneinander Wasserstoff; C1-C5-Alkyl; nicht substituiertes oder mit Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder Amino substituiertes Phenyl; Biphenyl; Cyclo-C3-C7-alkyl; 3-Pyridyl; 4-Pyridyl; 2-Thiophenyl; 3-Thiophenyl; oder Thiazolyl; bedeuten, oder Verbindungen der Formel (1), wobei
R3 Wasserstoff; oder Phenyl bedeutet, oder Verbindungen der Formel (1), wobei
R4 Wasserstoff bedeutet. - Insbesondere werden Verbindungen der Formel

- Die Herstellung der erfindungsgemäß verwendeten 4-Aminopyrimidine erfolgt nach an sich bekannten Verfahren (J. Org. Chem.; 1967, 32, 1591). Hierzu wird eine Cyanoverbindung (R1-C≡N) in einem geeigneten Lösungsmittel wie z. B. Methanol, Ethanol, Isopropanol, DMF, Tetrahydrofuran, usw. mit Ammoniumacetat oder Ammoniumchlorid bei einer Temperatur von –10 °C bis 100 °C innerhalb 1 h bis 24 h zur entsprechenden Amidinverbindung umgesetzt.
- Anschließend erfolgt eine Kondensation der Amidinverbindung mit einem entsprechenden β-Ketoesterunter Verwendung einer Hilfsbase, wie z. B. Natriumcarbonat, Kaliumhydroxid, Natriumethylat, Natriummethylat, Kalium-tert-butylat, usw. in einem geeignetem Lösungsmittel wie z. B. Methanol, Ethanol, Butanol, tert-Butanol, THF, DMF, Acetonitril, Toluol, Xylol, usw. innerhalb 1-24 h bei einer Temperatur zwischen 40 und 120 °C.

- Die substituierten 4-Aminopyrimidinewerden durch Reaktion der 4-Chlor-2-pyrimidinverbindung mit einem primären oder sekundären Amin (R4R5NH) in einem geeigneten Lösungsmittel wie z. B. DMF, Dioxan, Toluol, Xylol, Ethanol, Butanol, und einer Hilfsbase wie z. B. Triethylamin, DIEA, Natriumcarbonat, Kaliumhydroxid, usw. oder unter Verwendung eines Überschusses an Amin innerhalb 1-24 h bei 40-130 °C erhalten.

- Die Herstellung der Verbindungen der Formel (2) erfolgt durch Reaktion eines Überschusses von 2-10 Äquivalenten der Diaminverbindung H2N(CH2)nX(CH2)mNH2 in z. B. DMF, Dichlormethan, THF oder Dioxan mit Tritylchlorid-Polystyrolharz bei einer Temperatur zwischen 10 und 50 °C innerhalb 0,5-24 h. Danach werden 2-10 Äquivalente der entsprechend substituierten 4,6-Dichlorpyrimidinein einem geeigneten Lösungsmittel wie z. B. Dichlormethan, DMF, THF, Toluol mit den polymer gebundenen Diaminen innerhalb 2 bis 48 h bei 10-120 °C umgesetzt. Die 4-Chlorpyrimidine werden mit 2 bis 10 Äquivalenten verschiedener Boronsäuren, 1 bis 10 % Palladium-Katalysator und 2 bis 10 Äquivalenten Hilfsbase, wie z. B. CaCO3, NaCO3 in z. B. THF, DMF oder Dioxan umgesetzt. Nach Waschen des Harzes zur Entfernung des Überschusses werden die Zielverbindungen mit 1-30 % Trifluoressigsäure (TFA) in Dichlormethan (DCM) innerhalb 1-5 h bei 25 °C abgespalten. Zur weiteren Aufreinigung werden die Substanzen aus tBuOH/Wasser 4:1 mit 1-10 % HOAc und einmal aus tBuOH/Wasser 4:1 gefriergetrocknet.

- Einige der erfindungsgemäß verwendeten 4-Aminopyrimidine sind aus der Literatur bekannt, und einige sind neue Verbindungen.
- Die erfindungsgemäß eingesetzten 4-Aminopyrimidine zeigen ausgeprägte antimikrobielle Wirkung, insbesondere gegen pathogene grampositive und gramnegative Bakterien sowie gegen Bakterien der Hautflora, und außerdem gegen Hefen und Schimmelpilze. Sie eignen sich daher insbesondere zur Desinfektion, Desodorierung, sowie der allgemeinen und antimikrobiellen Behandlung der Haut und Schleimhäute sowie Hautanhangsgebilde (Haare), ganz besonders zur Hände- und Wunddesinfektion.
- Sie sind daher geeignet als antimikrobielle Wirksubstanzen und Konservierungsmittel in Körperpflegemitteln, wie z. B. Shampoos, Badezusätzen, Haarpflegemitteln, flüssigen und festen Seifen (auf Basis synthetischer Tenside und Salze von gesättigten und/oder ungesättigten Fettsäuren), Lotionen und Cremes, Deodorantien, anderen wässerigen oder alkoholischen Lösungen, z. B. Reinigungslösungen für die Haut, feuchten Reinigungstüchern, Ölen oder Pudern.
- Einen weiteren Erfindungsgegenstand bildet daher ein Körperpflegemittel, enthaltend mindestens eine Verbindung der Formel (1) sowie kosmetisch verträgliche Träger- oder Hilfsstoffe.
- Das erfindungsgemäße Körperpflegemittel enthält 0,01 bis 15 Gew.-%, vorzugsweise 0,1 bis 10 Gew.-%, basierend auf dem Gesamtgewicht der Zusammensetzung, einer Verbindung der Formel (1) und kosmetisch verträgliche Hilfsstoffe.
- Je nachdem, in welcher Form das Körperpflegemittel vorliegt, weist es neben dem 4-Aminopyrimidin der Formel (1) noch weitere Bestandteile auf, wie z. B. Sequestriermittel, Farbstoffe, Parfümöle, Verdickungs- bzw. Festigungsmittel (Konsistenzregler), Emmollients, UV-Absorber, Hautschutzmittel, Antioxidantien, die mechanischen Eigenschaften verbessernde Additive wie Dicarbonsäuren und/oder Aluminium-, Zink-, Calcium-, Magnesium-Salze von C14-C22-Fettsäuren und gegebenenfalls Konservierungsmittel.
- Das erfindungsgemäße Körperpflegemittel kann als Wasser-in-Öl- oder Öl-in-Wasser-Emulsion, als alkoholische oder alkoholhaltige Formulierung, als vesikulare Dispersion eines ionischen oder nichtionischen amphiphilen Lipids, als Gel, fester Stift oder als Aerosol-Formulierung formuliert werden.
- Als Wasser-in-Öl- oder Öl-in-Wasser-Emulsion enthält der kosmetisch verträgliche Hilfsstoff vorzugsweise 5 bis 50 % einer Ölphase, 5 bis 20 % eines Emulgators und 30 bis 90 % Wasser. Die Ölphase kann dabei irgendein für kosmetische Formulierungen geeignetes Öl enthalten, wie z. B. ein oder mehrere Kohlenwasserstofföle, ein Wachs, ein natürliches Öl, ein Silikon-Öl, einen Fettsäureester oder einen Fettalkohol. Bevorzugte Mono- oder Polyole sind Ethanol, Isopropanol, Propylenglycol, Hexylenglycol, Glycerin und Sorbitol.
- Erfindungsgemäße kosmetische Formulierungen werden in verschiedenen Bereichen eingesetzt. Insbesondere kommen z. B. die folgenden Mittel in Betracht:
- – Mittel zur Hautpflege, wie z. B. Hautwasch- und Reinigungsmittel in Form von stückförmigen oder flüssigen Seifen, Syndets oder Waschpasten,
- – Badepräparate, wie z. B. flüssige (Schaumbäder, Milche, Duschpräparate) oder feste Badepräparate, wie z. B. Badetabletten und Badesalze;
- – Hautpflegemittel, wie z. B. Hautemulsionen, Mehrfachemulsionen oder Hautöle;
- – Dekorative Körperpflegemittel, wie z. B. Gesichts-Make-Ups in Form von Tages- oder Pudercremes, Gesichtspuder (lose oder gepreßt), Rouge oder Creme-Make-Ups, Augenpflegemittel, wie z. B. Lidschattenpräparate, Wimperntusche, Eyeliner, Augencremes oder Eye-Fix-Cremes; Lippenpflegemittel, wie z. B. Lippenstift, Lip Gloss, Lippenkonturstift, Nagelpflegemittel, wie Nagellack, Nagellackentferner, Nagelhärter, oder Nagelhautentferner;
- – Intimpflegemittel, wie z. B. Intim-Waschlotionen oder Intimsprays;
- – Fußpflegemittel, wie z. B. Fußbäder, Fußpuder, Fußcremes bzw. Fußbalsame, spezielle Deomittel und Antitranspirantien oder hornhautbeseitigende Mittel;
- – Lichtschutzmittel, wie Sonnenmilche, -lotionen, -cremes, -öle, Sun-blocker oder Tropicals, Vorbräunungspräparate oder After-sun-Präparate;
- – Hautbräunungsmittel, wie z. B. Selbstbräunungscremes;
- – Depigmentierungsmittel, wie z. B. Präparate zur Hautbleichung oder Mittel zur Hautaufhellung;
- – Insektenabweisende Mittel („Repellents"), wie z. B. Insektenöle, -lotionen, -sprays oder -stifte;
- – Deodorantien, wie Deosprays, Pumpsprays, Deogele, -stifte oder -roller;
- – Antitranspirantien, wie z. B. Antitranspirantstifte, -cremes oder -roller;
- – Mittel zur Reinigung und Pflege von unreiner Haut, wie z. B. Syndets (fest oder flüssig), Peeling- oder Scrubb-Präparate oder Peeling-Masken;
- – Haarentfernungsmittel in chemischer Form (Depilation), wie z. B. Haarentfernungspulver, flüssige Enthaarungsmittel, cremige oder pastöse Enthaarungsmittel, Enthaarungsmittel in Gelform oder Aerosolschäume;
- – Rasiermittel, wie z. B. Rasierseife, schäumende Rasiercremes, nichtschäumende Rasiercremes, -schäume, -gele, Preshave-Präparate für die Trockenrasur, Aftershaves oder Aftershave-Lotionen;
- – Duftmittel, wie z. B. Duftwässer (Eau de Cologne, Eau de Toilette, Eau de Parfum, Parfum de Toilette, Parfüm), Parfümöle oder Parfümcremes;
- – Mittel zur Zahn-, Zahnersatz- und Mundpflege, wie z. B. Zahncremes, Gel-Zahncremes, Zahnpulver, Mundwasserkonzentrate, Anti-Plaque-Mundspülungen, Prothesenreiniger oder Prothesenhaftmittel;
- – Kosmetische Mittel zur Haarbehandlung, wie z. B. Haarwaschmittel in Form von Shampoos, Haarkonditioniermittel, Haarpflegemittel, wie z. B. Vorbehandlungsmittel, Haarwasser, Frisiercremes, Frisiergele, Pomaden, Haarspülungen, Kurpackungen, Intensivhaarkuren, Mittel zur Haarverformung, wie z. B. Wellmittel zur Herstellung von Dauerwellen (Heißwelle, Mildwelle, Kaltwelle), Haarglättungspräparate, flüssige Haarfestiger, Haarschäume, Haarsprays, Blondiermittel, wie z. B. Wasserstoffperoxidlösungen, aufhellende Shampoos, Blondiercremes, Blondierpulver, Blondierbreie oder -öle, temporäre, semitemporäre oder permanente Haarfärbemittel, Präparate mit selbstoxidierenden Farbstoffen, oder natürliche Haarfärbemittel, wie Henna oder Kamille.
- Eine antimikrobielle Seife hat z. B. folgende Zusammensetzung:
0,01 bis 5 Gew.-% einer Verbindung der Formel (1)
0,3 bis 1 Gew.-% Titandioxid,
1 bis 10 Gew.-% Stearinsäure,
ad 100 % Seifengrundlage, wie z.B. die Natriumsalze der Talgfett- und Kokosfettsäure oder Glycerin. - Ein Shampoo hat z. B. die folgende Zusammensetzung:
0,01 bis 5 Gew.-% einer Verbindung der Formel (1),
12,0 Gew.-% Natrium-Laureth-2-sulfat,
4,0 Gew.-% Cocamidopropylbetain,
3,0 Gew.-% NaCl und
Wasser ad 100 %. - Ein Deodorant hat z. B. die folgende Zusammensetzung:
0,01 bis 5 Gew.-% einer Verbindung der Formel (1),
60 Gew.-% Ethanol,
0,3 Gew.-% Parfümöl und
Wasser ad 100 %. - Einen weiteren Erfindungsgegenstand bildet eine orale Zusammensetzung, enthaltend 0,01 bis 15 Gew.-%, basierend auf dem Gesamtgewicht der Zusammensetzung, einer Verbindung der Formel (1) und oral verträgliche Hilfsstoffe.
- Beispiel für eine orale Zusammensetzung:
10 Gew.-% Sorbitol,
10 Gew.-% Glycerin,
15 Gew.-% Ethanol,
15 Gew.-% Propylenglycol,
0,5 Gew.-% Natriumlaurylsulfat,
0,25 Gew.-% Natriummethylcocyltaurat,
0,25 Gew.-% Polyoxypropylen/Polyoxyethylen-Blockcopolymer,
0,10 Gew.-% Pfefferminzgeschmacksstoff,
0,1 bis 0,5 Gew.-% einer Verbindung der Formel (1) und
48,6 Gew.-% Wasser. - Die erfindungsgemäße orale Zusammensetzung kann z. B. in Form eines Gels, einer Paste, einer Creme oder einer wässerigen Zubereitung (Mundwasser) vorliegen.
- Weiterhin kann die erfindungsgemäße orale Zusammensetzung Verbindungen enthalten, die Fluoridionen freisetzen, die gegen die Bildung von Karies wirksam sind, z. B. anorganische Fluoridsalze, wie z. B. Natrium-, Kalium-, Ammonium- oder Calciumfluorid oder organische Fluoridsalze, wie z. B. Aminfluoride, die unter dem Handelsnamen Olafluor bekannt sind.
- Weiterhin eignen sich die erfindungsgemäß verwendeten 4-Aminopyrimidine der Formel (1) für die Behandlung, insbesondere Konservierung von textilen Fasermaterialien. Es handelt sich dabei um ungefärbte und gefärbte oder bedruckte Fasermaterialien z. B. aus Seide, Wolle, Polyamid oder Polyurethanen, und insbesondere cellulosehaltige Fasermaterialien aller Art. Solche Fasermaterialien sind beispielsweise natürliche Cellulosefasern, wie Baumwolle, Leinen, Jute und Hanf, sowie Zellstoff und regenerierte Cellulose. Bevorzugte geeignete textile Fasermaterialien sind aus Baumwolle.
- Die erfindungsgemäßen 4-Aminopyrimidine eignen sich auch zur Behandlung, insbesondere zur antimikrobiellen Ausrüstung oder Konservierung von Kunststoffen, wie z. B. Polyethylen, Polypropylen, Polyurethan, Polyester, Polyamid, Polycarbonat, Latex usw. Einsatzbereiche dafür sind z. B. Fußbodenbeläge, Kunststoffbeschichtungen, Kunststoffbehälter- und Verpackungsmaterialien; Küchen- und Badezimmer-Utensilien (z. B. Bürsten, Duschvorhänge; Schwämme, Badezimmermatten), Latex, Filtermaterialien (Luft- und Wasserfilter), Kunststoffartikel, die im medizinischen Bereich eingesetzt werden, wie z. B. Verbandmaterialien, Spritzen, Katheter usw., sog. „medizinische Vorrichtungen", Handschuhe und Matratzen.
- Auch Papier, wie z. B. Hygienepapiere können mit den erfindungsgemäßen 4-Aminopyrimidinen antimikrobiell ausgerüstet werden.
- Weiterhin können Nonwovens, wie z. B. Windeln, Damenbinden, Dameneinlagen, Tücher für den Hygiene- und Haushaltsbereich erfindungsgemäß antimikrobiell ausgerüstet werden.
- Weiterhin finden die 4-Aminopyrimidine der Formel (1) Verwendung in Wasch- und Reinigungsformulierungen, wie z. B. in Flüssig- oder Pulverwaschmitteln oder Weichspülern.
- Die 4-Aminopyrimidine der Formel (1) können insbesondere auch in Haushalts- und Allzweckreinigern zur Reinigung und Desinfektion von harten Oberflächen eingesetzt werden.
- Ein Reinigungsmittel hat z. B. folgende Zusammensetzung:
0,01 bis 5 % einer Verbindung der Formel (1),
3,0 % Octylalkohol 4EO,
1,3 % Fettalkohol-C8-C10-Polyglucosid,
3,0 % Isopropanol
ad 100 % Wasser. - Neben der Konservierung von Kosmetik- und Haushaltsprodukten ist auch die Konservierung und antimikrobielle Ausrüstung von technischen Produkten sowie der Einsatz als Biozid in technischen Prozessen möglich, wie z. B. bei der Papierbehandlung, insbesondere in Papierbehandlungsflotten, Druckverdickern aus Stärke oder Celluloseabkömmlingen, Lacken und Anstrichfarben.
- Die 4-Aminopyrimidine der Formel (1) eignen sich auch zur antimikrobiellen Holzbehandlung sowie zur antimikrobiellen Behandlung, Konservierung und Ausrüstung von Leder.
- Weiterhin eignen sich die erfindungsgemäßen Verbindungen zum Schutz von kosmetischen Produkten und Haushaltsprodukten vor mikrobieller Verderbnis.
- Die folgenden Beispiele veranschaulichen die vorliegende Erfindung, schränken diese aber nicht ein.
- Ausführungsbeispiele
- Allgemeine Arbeitsvorschrift zur parallelen Synthese von 4-Aminopyrimidinen
- Beispiel 1
- Herstellung von 4-Chlor-6-methyl-2-phenylpyrimidin
- 2,2 g Benzamidinhydrochlorid (14,05 mmol) werden in 10 ml EtOH absolut mit 5,43 ml (14,05 mmol) einer 20%igen Natriumethylat-Lösung umgesetzt und anschließend mit 1,66 g Acetessigsäuremethylester (14,29 mmol) 4 h bei 90 °C kondensiert.
- Das Rohprodukt wird eingedampft und in 30 ml Toluol aufgenommen.
- 4,31 g Phosphor(V)-oxidchlorid (28,1 mmol) werden dazugegeben und die Reaktionsmischung 3 h bei 120 °C erhitzt. Nach Abkühlen auf 20 °C wird der Überschuß mit Natronlauge neutralisiert, die Mischung mit Essigester verdünnt und anschließend mit Wasser und gesättigter Natriumchloridlösung gewaschen.
- Die Produktlösung wird über Natriumsulfat getrocknet und eingedampft. Es werden 2,2 g 4-Chlor-6-methyl-2-phenylpyrimidin (77,7 % d. Th.) erhalten.
- Beispiel 2: Umsetzung von 4-Chlor-6-methyl-2-phenylpyrimidin mit Monoaminen
- 20,5 mg 4-Chlor-6-methyl-2-phenylpyrimidin (0,1 mmol) werden mit 3 Äqu. Monoaminen (0,3 mmol) in 0,5 ml abs. Dioxan 20 h bei 100 °C erwärmt. Nach Abkühlung werden die Produkte im Vakuum eingeengt, in t-BuOH/Wasser 4/l aufgenommen und gefriergetrocknet. Die Endprodukte werden mittels LC-MS analysiert.
- Beispiel 3: Belegung von Tritylchlorid-Polystyrol-Harz mit N,N-Bis(3-aminopropyl)methylaminen und Umsetzung mit 4,6-Dichlor-2,5-diphenylpyrimidin
- Je 50 mg Harz (1,29 mmol/g) werden in 1 ml DMF mit 94 mg N,N-Bis(3-aminopropyl)methylamin (0,645 mmol) 3 h bei 25 °C geschüttelt. Das Harz wird abfiltriert und mit DCM, MeOH, THF, MeOH und DCM gewaschen und im Vakuum getrocknet.
- Das Harz wird in 1 ml DMF mit 0,194 g 4,6-Dichlor-2,5-diphenylpyrimidin (0,645 mmol) und 90 μl Triethylamin (0,645 mmol) 3 h bei 25 °C geschüttelt.
- Das Harz wird abfiltriert und mit DCM, MeOH, THF, MeOH, DCM und MeOH gewaschen und im Vakuum getrocknet.
- Beispiel 4: Parallele Umsetzung der 4-Amino-6-chlor-1,5-diphenylpyrimidin-TCP-Harze mit verschiedenen Boronsäuren und Abspaltung
- Das Harz wird mit 126,1 g Cäsiumcarbonat (6 Äqu., 0,387 mmol) und 300 μl einer Toluollösung von 0,1 Äqu. eines Palladium-Katalysators (
WO 01/16057 - Nach Abkühlen wird das Harz abfiltriert und mit DMF, MeOH, THF, MeOH und DCM gewaschen und im Vakuum getrocknet.
- Die Produkte werden mit 1,5 ml einer 5 % TFA/DCM-Lösung 3 h bei Raumtemperatur abgespalten. Das Harz wird anschließend mit 1 ml DCM und 1 ml MeOH gewaschen und die vereinigten Lösungen zur Trockne eingedampft. Die Endprodukte werden mittels LC-MS analysiert.
- Beispiel 5: Herstellung von 4-Chlor-6-methyl-2-tolylpyrimidin
- 2,5 g 4-Methyl-benzamidinhydrochlorid (14,65 mmol) werden in 10 ml EtOH absolut mit 5,66 ml einer 20-%-Lösung Natriumethylat (14,65 mmol) umgesetzt und anschließend mit 1,73 g Acetessigsäuremethylester (14,88 mmol) 4 h bei 90 °C kondensiert. Das Rohprodukt wird eingedampft und in 30 ml Toluol aufgenommen. 6,74 g Phosphor(V)-oxidchlorid (44,0 mmol) werden dazugegeben und die Reaktionsmischung 3 h bei 120 °C erhitzt. Nach Abkühlen auf 20 °C wird der Überschuß mit Natronlauge neutralisiert, die Mischung mit Essigester verdünnt, mit gesättigter Natriumhydrogencarbonat-Lösung und anschließend mit Wasser gewaschen. Die Produktlösung wird eingedampft und säulenchromatographisch getrennt (Hexan/EE: 5/1). Es werden 2,1 g 4-Chlor-6-methyl-2-tolylpyrimidin (66,5 % d. Th.) erhalten.
NMR: 1H (ppm in DMSO): 2,4, s, 3H; 2,55, s, 3H; 7,3, d, 2H; 7,5, s, 1H; 8,25, d, 2H - Beispiel 6: Umsetzung von 4-Chlor-6-methyl-2-(4-methyl)-phenylpyrimidin mit Monoaminen
- 21,9 mg 4-Chlor-6-methyl-2-(4-methyl)-phenylpyrimidin (0,1 mmol) werden mit 3 Äqu. Monoaminen (0,3 mmol) in 0,5 ml abs. Dioxan 20 h bei 100 °C erwärmt. Nach Abkühlung werden die Produkte im Vakuum eingeengt, in t-BuOH/Wasser 4/l aufgenommen und gefriergetrocknet. Die Endprodukte werden mittels LC-MS analysiert.
- Beispiel 8: Herstellung von 4-Chlor-2-isopropyl-6-methylpyrimidin
- 76,1 g 2-Isopropyl-6-methyl-4-pyrimidinol [2814-20-2] (500 mmol) werden in 300 ml Toluol bei 90 °C gelöst. 80,5 g Phosphor(V)-oxidchlorid (525 mmol) werden bei 90 bis 103 °C dazugetropft und die Reaktionsmischung 2 h bei 110 °C erhitzt. Nach Abkühlen auf 20 °C wird die Reaktionsmischung mit Natronlauge 4M unter Kühlen auf pH 8 gestellt. Die wässerige Phase wird abgetrennt und mit 100 ml Toluol extrahiert. Die vereinigten organischen Phasen werden 3 Mal mit je 100 ml Wasser gewaschen und bei RT unter 2 mbar getrocknet. Man erhält 89,7 g (105 %, enthält Toluol).
- Beispiel 10: Bestimmung der minimalen Hemmkonzentration (MHK-Wert in Mikrotiterplatten

- Beispiele für Testorganismen:
Bakterien: Pseudomonas aeruginosa CIP A-22 (= PA) Escherichia coli NCTC 8196 (= EC) Staphylococcus aureus ATCC 9144 (= SA) Candida albicans ATCC 10231 (= CA) - Verfahren:
- Die Testsubstanzen werden in Dimethylsulfoxid (DMSO) vorgelöst und in einer Verdünnungsreihe von 1:2 getestet.
- Bakterien und Hefe werden über Nacht in CASO-Bouillon angezüchtet.
- Alle Testkeim-Suspensionen werden mit 0,85%iger Kochsalzlösung auf eine Keimzahl von 1 – 5 × 106 KBE/ml eingestellt.
- Die Testsubstanzen werden à 8 μl pro Loch in Mikrotiterplatten vorpipettiert.
- Die voreingestellten Keimsuspensionen werden 1:100 in CASO-Bouillon verdünnt und à 192 μl pro Loch den Testsubstanzen zugegeben.
- Die Testansätze werden 48 Stunden bei 37 °C inkubiert.
- Nach Inkubation wird das Wachstum anhand der Trübung der Testansätze (optische Dichte) bei 620 nm in einem Mikroplate-Reader bestimmt.
- Als minimale Hemmkonzentration (MHK-Wert) wird diejenige Substanzkonzentration angegeben, bei der eine deutliche Wachstumshemmung (≤ 20 % Wachstum verglichen mit der Wachstumskontrolle) der Testkeime festzustellen ist.
- Pro Testkeim und Substanzkonzentration werden drei Mikrotiterplatten angesetzt. Alle Substanzen werden im Doppel geprüft.
- In der Tabelle 2 sind die mikrobiologischen Testergebnisse zusammengestellt:

- Verwendung eines 4-Aminopyrimidins der Formel R1 Wasserstoff; C1-C5-Alkyl, das nicht substituiert oder durch ein oder mehrere Halogenatome substituiert ist; Biphenyl oder C6-C10-Aryl, das nicht substituiert oder durch Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder durch Amino substituiert ist; einen Pyrrol-, Furan-, Thiophen-, Imidazol-, Pyrazol-, 1,2,3-Triazol-, 1,2,4-Triazol-, Pyridazin-, Pyrimidin-, Pyrazin-, Pyran-, Thiopyran-, 1,4-Dioxan-, 1,2-Oxazin-, 1,3-Oxazin-, 1,4-Oxazin-, Indol-, Benzothiophen-, Benzofuran-, Pyrrolidin-, Piperidin-, Piperazin-, Morpholin- oder Thiomorpholinrest; oder Cyclo-C3-C7-alkyl; R2 Wasserstoff; C1-C5-Alkyl, das nicht substituiert oder durch ein oder mehrere Halogenatome substituiert ist; Biphenyl oder C6-C10-Aryl, das nicht substituiert oder durch Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder durch Amino substituiert ist, einen 5- bis 7-gliedrigen Heteroarylrest; oder Cyclo-C3-C7-alkyl; R3 Wasserstoff; Phenyl oder C1-C5-Alkyl, das nicht substituiert oder durch ein oder mehrere Halogenatome substituiert ist; R4 Wasserstoff; C1-C10-Alkyl; Phenyl; oder einen 5- bis 7-gliedrigen Heteroarylrest; R5 NR''R'''-C1-C20-Alkyl, das nicht substituiert oder durch einen heterocyclischen Rest substituiert oder nicht unterbrochen oder durch eine oder mehrere -O- oder -Gruppen oder durch einen bivalenten heterocyclischen Rest unterbrochen ist, oder R4 und R5 zusammen mit dem sie verbindenden Stickstoffatom einen Rest der Formel (1a) R' Wasserstoff; oder C1-C3-Alkyl; R'' und R''' unabhängig voneinander Wasserstoff; C1-C5-Alkyl; oder Hydroxy-C1-C5-alkyl; R'''' Wasserstoff; C1-C4-Alkyl; oder Heteroaryl-C1-C4-alkyl; und n1 und n2 unabhängig voneinander 1 bis 8 bedeuten; zur antimikrobiellen Behandlung von Oberflächen.
- Verwendung nach Anspruch 1, wobei R5 gegebenenfalls durch eine oder mehrere -O- oder-Gruppen oder einen bivalenten heterocyclischen Rest unterbrochenes R''R'''N-C1-C20-Alkyl; R' Wasserstoff oder C1-C5-Alkyl; R'' und R''' unabhängig voneinander Wasserstoff oder Methyl bedeuten; und R1, R2, R3 und R4 wie in Anspruch 1 definiert sind.
- Verwendung nach Anspruch 4, wobei R5 R''R'''N-C5-C20-Alkyl; und R'' und R''' unabhängig voneinander Wasserstoff oder Methyl bedeuten.
- Verwendung nach Anspruch 1, wobei R4 und R5 zusammen mit dem sie verbindenden Stickstoffatom einen Rest der Formel (1a) bilden, worin R'''' Wasserstoff oder Pyridyl-C1-C3-alkyl; und n1 und n2 in jedem Fall 2 bedeuten.
- Verwendung nach einem der Ansprüche 1 bis 6, wobei R1 Wasserstoff; C1-C5-Alkyl; nicht substituiertes oder mit Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder Amino substituiertes Phenyl; Biphenyl, Cyclo-C3-C7-alkyl, 2-Thiophenyl; 3-Thiophenyl oder Thiazolyl; und R2 Wasserstoff, C1-C5-Alkyl; nicht substituiertes oder mit Halogen, C1-C5-Alkyl, C1-C5-Alkoxy oder Amino substituiertes Phenyl; Biphenyl, Cyclo-C3-C7-alkyl, 3-Pyridyl; 4-Pyridyl; 2-Thiophenyl; 3-Thiophenyl oder Thiazolyl bedeuten.
- Verwendung nach Anspruch 1, wobei R3 Wasserstoff oder Phenyl bedeutet.
- Verwendung nach einem der Ansprüche 1 bis 7, wobei R4 Wasserstoff bedeutet.
- Verwendung nach Anspruch 1, wobei die Verbindung der Formel (1) zur antimikrobiellen Behandlung, Desodorierung und Desinfektion der Haut, Schleimhäute und Haare verwendet wird.
- Verwendung nach Anspruch 1, wobei die Verbindung der Formel (1) zur Behandlung von textilen Fasermaterialien verwendet wird.
- Verwendung nach Anspruch 1, wobei die Verbindung der Formel (1) zur Konservierung verwendet wird.
- Verwendung nach Anspruch 1, wobei die Verbindung der Formel (1) in Wasch- und Reinigungsformulierungen verwendet wird.
- Verwendung nach Anspruch 1, wobei die Verbindung der Formel (1) zur antimikrobiellen Ausrüstung und Konservierung von Kunststoffen, Papier, Nonwovens, Holz oder Leder verwendet wird.
- Verwendung einer Verbindung der Formel (1) zur antimikrobiellen Ausrüstung und Konservierung von technischen Produkten, insbesondere Druckverdickern aus Stärke oder Celluloseabkömmlingen, Lacken und Anstrichfarben.
- Verwendung einer Verbindung der Formel (1) als Biozid in technischen Prozessen.
- Körperpflegemittel, umfassend 0,01 bis 15 Gew.-%, bezogen auf das Gesamtgewicht der Zusammensetzung, einer Verbindung der Formel (1) und kosmetisch verträgliche Hilfsstoffe.
- Orale Zusammensetzung, umfassend 0,01 bis 15 Gew.-%, bezogen auf das Gesamtgewicht der Zusammensetzung, einer Verbindung der Formel (1) und oral verträgliche Hilfsstoffe.
