in der A und B je ein Wasscrsloffatom oder zusammen
die Methylengruppe R1 ein Wasserstoff-, Chlor- oder
Bromatom. R2 ein !-'luoratom, die Cyan- oder Trifluormclhylgruppc
und R, cine Alkyl- oder Cycloalkylgruppc, welche jeweils 3 bis 5 Kohlcnstoffatomc
enthalten kann,die 2-Mclhyl-4-hydroxybutyl-(2)-gruppe,
die Hydroxy-tcrt.-bulylgruppc. die 3-Äthinyl-pcnlyl-(3)-gruppe
oder die l-(3,4-MclhyIendioxyphcnyl)-propyI-(2)-gruppc bedeuten, und dere«* physiologisch
verträgliche Säurcaddilionssalzc.
Gegenstand der Erfindung sind ferner Verfahren zur Herstellung der erfindungsgemäßen Verbindungen ^o
sowie Arzneimittel, die diese Verbindungen enthalten, entsprechend den verstehenden Patentansprüchen.
Die Verbindungen der obige;', allgemeinen Formel I
besitzen wertvolle pharmakoloi:ischc Eigenschaften, neben einer analgclischcn, ulcriisspasrm ytischen und .v>
einer antispastischen Wirkung auf die quergestreifte Muskulatur insbesondere /lymimetisehc und/oder
//,-blockierende Wirkungen, wobei je nach ihrer Substitution
die eine oder andere Wirkung im Vordergrund steht.
Die neuen Verbindungen lassen sich in an sich bekannter Weise nach folgenden Verfahren herstellen:
a) Reduktion eines Acctophcnons der allgemeinen Formel Il

in der R1 bis R, wie eingangs definiert sind.
Die Reduktion wird in einem Lösungsmittel wie Methanol. Melhanol/Wasscr, Äthanol. Isopropanol,
Äther, Tetrahydrofuran oder Dioxan zweckmäßigerweise mit einem komplexen Metallhyrid wie Lithiumaluminiumhydrid
oder Natriumborhydrid, mit AIuminiumisopropylat in Gegenwart eines primären oder
sekundären Alkohols oder mit katalytisch angeregtem Wasserstoff und bei Temperaturen zwischen -201C
und der Siedetemperatur des verwendeten Lösungsmittels durchgerührt.
Die Reduktion mit komplexen Metallhydridcn wird jedoch vorzugsweise mit Natriumborhydrid und bei
Raumtemperatur durchgeführt. Bei Verwendung eines hs
reaktionsfähigeren komplexen Metallhydrids wie Liihiumaliiminiumhydrid
und gegebenenfalls bei erhöhten Temperaturen kann die bei der Dclinilion
des Restes R2 erwähnte Cyangruppc gleichzeitig milreduzicrl
werden.
Führt man die Reduktion mit Wasserstoff in Gegenwart eines Katalysators wie Raney-Nickel, Platin
oder Palladium/Tierkohlc durch, so kann die bei der Definition des Restes R3 erwähnte Alkinylgruppe in
den entsprechenden Alkylrest übergeführt werden.
b) Zur Herstellung von Verbindungen der allgemeinen Formel I, in der Rj mit Ausnahme des eingangs
erwähnten Alkinylrestes wie eingangs definiert ist und R1 ein Chlor- oder Bromatom darstellt:
Halogenierung einer Verbindung der allgemeinen Formel III in einem Lösungsmittel

in der R2 und R3 mit Ausnahme des eingangs erwähnten
Alkinylrestes wie eingangs definiert sind.
Die Umsetzung wird mit einem Halogenierungsmittel,
z. B. mit Chlor, Brom oder Phenyljoddichlorid, vorzugsweise in einem Lösungsmittel, z. B. in 50- bis
IOO%iger Essigsäure oder in Tetrahydrofuran in Gegenwart einer tertiären organischen "Base, und
zweckmäßigerweise bei Temperaturen zwischen 0 und 50"C durchgeführt. Pro Mol einer Verbindung der
allgemeinen Formel III, welche als Base oder auch als Salz, z. B. als Morrohydrochlorid oder Dihydrochlorid,
eingesetzt werden kann, werden zweckmäßigerweise I Mol an Halogenierungsmittel oder
ein geringer Überschuß verwendet. Falls bei der Umsetzung ein halogenwasserstoffsaures Salz entsteht,
kann dieses als solches direkt isoliert, oder es kann gcwünschtcnfalls über die Base weiter gereinigt werden.
c) Abspaltung eines Schutzrestes in einem Lösungsmittel von einer Verbindung der allgemeinen Formel

einen beliebigen Sehutzresl für eine Aminogruppc
darstellt.
Bedeutet Y beispielsweise einen beliebigen Acylrcst,
z. B. den Acetyl-, Benzoyl- oder p-Toluolsulfonylrest.
so erfolgt die Abspaltung dieses Restes hydrolytisch, z. B. mit äthanolischer Salzsäure oder Natronlauge
bei Temperaturen bis ziim Siedepunkt des verwendeten
Lösungsmittels. Bedeutet hierbei in einer Verbindung der allgemeinen Formel IV, IVa oder IVb R, eine
Cyangruppe, so kann diese gleichzeitig zur Carboxylgruppe verseift werden.
Bedeutet Y beispielsweise einen Benzylrcst, so erfolgt die Abspaltung dieses Restes hydrogenolytisch,
z. B. mit Wasserstoff in Gegenwart eines Katalysators, vorzugsweise in einem Lösungsmittel wie Methanol,
Methanol/Salzsäure und bei Raumtemperatur. Bei der Umsetzung kann die bei der Definition von R,
erwähnte Alkinylgruppc gleichzeitig in den entsprechenden Alkylrest übergeführt werden.
d) Zur Herstellung von Verbindungen der allgemeinen Formel I, in der R1 ein WasserslolTaloii darstellt:
Enlhalogenierung einer Verbindung dci allgemeinen
Formel V
in der R2 und R3 wie eingangs definiert sind und Hai
ein Chlor-, Brom- oder Jodalom darstellt.
Die Enthalogenierung erfolgt in einem Lösungsmittel und zweckmäßigerweisc entweder mit Triphenylphosphin
in Benzol oder mit Wasserstoff in Methanol und in Gegenwart eines Hydrierungskatalysators.
Je nach dem verwendeten Katalysator wird die Reaktion bei Raumtemperatur oder erhöhter
Temperatur, beispielsweise bei Temperaturen zwischen 100 und 150"C, und bei Normaldruck oder mäßigem
überdruck durchgeführt, bei Anwendung von Rancy-Nickel oder Palladium/Kohle beispielsweise erfolgt
die Enthalogenierung bei Raumtemperatur und Normaldruck. Bedeutet hierbei in einer Verbindung der
allgemeinen Formel V R3 die eingangs erwähnte Alkinylgruppe, so Vann diese bei der hydrogcnolytischen
Enthalogenierung zu dem entsprechenden Alkylrest hydriert werden.
Eine gemäß den Verfahren a) bis d) erhaltene Verbindung der allgemeinen Formel I kann gcwünschlcnfalls
mit Formaldehyd in ein entsprechendes Oxazolidin der allgemeinen Formel I übergeführt werden.
Diese Umsetzung erfolgt in einem Lösungsmittel wie Äthanol, Benzol, Toluol oder Dioxan unter wasserabspaltenden
Bedingungen, z. B. in Gegenwart von wasserfreiem Kupfcr(II (-sulfat, bei Temperaturen bis
zur Siedetemperatur des verwendeten Lösungsmittels.
z.B. bei Temperaturen zwischen 20 und KK) C. Besonders vorteilhaft wird die Umsetzung jedoch am
Wasserabscheider in Gegenwart eines Lösungsmittels wie Benzol oder Toluol durchgeführt.
Die erhaltenen Verbindungen der allgemeinen
Formel I können grwiinsehtenfalls mit anorganischen
oder organischen Siiurcii in ihre physiologisch verträglichcn
Säurcaddilionssalzc mit I ndcr 2 Äquivalenten der betreffenden Säure uhergcfuhrt werden.
Als Säuren haben sich beispielsweise Salzsäure. Btornwasscrstoffsäure,
Schwefelsäure, Phosphorsäure,
s Milchsäure, Zitronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen.
Die als Ausgangsstoffe verwendeten Verbindungen der allgemeinen Formel 11 lassen sich nach üblichen
Verfahren herstellen, z. B. durch Umsetzung der cni-
lü sprechenden 2-Halogcn-acelophcnonc mit den entsprechenden
Aminen.
Die Verbindungen der allgemeinen Formeln 111,1V,
IVa, IVb und V erhält man durch Umsetzung der entsprechenden 2-Halogcn-acctophenonc mit den enl-
is sprechenden Aminen und anschließende Reduktion der erhaltenen Ketone, beispielsweise mit Nalriumborhydrid.
Die bei den Verfahren a) bis d) verwendeten Ausgangsproduklc
der Formeln II V brauchen nicht in
κ, allen Fallen rein dargestellt werden, sie können auch
zweckmäßigerweisc als Rohprodukte verwendet werden.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen
der allgemeinen Formel I wertvolle phar-
2s makologischc Eigenschaften auf. insbesondere eine
/(2-r.':imctischc und/oder /(,-blockierende Wirkung,
wobei je nach ihrer Substitution die eine oder andere Wirkung im Vordergrund steht.
Beispielsweise wurden die Substanzen

Die neuen Verbindungen der allgemeinen Forme) I
lassen sich gegebenenfalls in Kombination mit anderen Wirksubstanzen in die üblichen pharmazeutischen
Zubereitungsformen einarbeiten. Hierbei beträgt die Einzeldosis 1 bis l00>', vorzugsweise jedoch 5 bis 5Oy. ^
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern:
I -(4-Amino-3-chlor-5-trifluormethyl-phenyl)-2-tert.-butylaminoäthanol-hydrochlorid
80g4'-Amino-2-tert.-butylamino-3'-chIor-5-trifluormethylacetophenon
-hydrochlorid (Zersetzung zwischen 223 und 231 C) werden in 500 ml Methanol
gelöst und auf - 15 C abgekühlt. Unter Rühren trägt man im Laufe einer Stunde portionsweise 9,5 g Natriumborhydrid
ein. wobei man die Temperatur auf -5 bis - 1511C hält. Nach einer weiteren Stunde bei
— 15r C säuert man mit 2 η-Salzsäure an und entfernt
das Methanol im Vakuum. Die zurückbleibende wäßrige Lösung wird mit 2n-Ammoniak alkalisch
gestellt und mit Essigester extrahiert. Man wäscht die organische Phase mit Wasser, trocknet und versetzt
mit 50 ml 4,5 η-Salzsäure in Isopropanol. Das ausgefallene
Hydrochlorid der obengenannten Substanz wird abgesaugt und mit Essigester und Äther gewaschen.
Schmelzpunkt: 205-2070C (Zers.).
Durch Konzentrieren der Mutterlauge läßt sich weitere Substanz gewinnen.
l-(4-Amino-3-chIor-5-trifluormethyI-phenyI)-2-tert.-perity!aminoäthano!-hydroch!ond
Λ
0,37 g I-(4-Amino-3-trifluormethyI-phenyl)-2-tert.-pentyiaminoäthanol-hydrobromid
und 0,2 ml Pyridin werden in 30 ml Tetrahydrofuran gelösl und auf (I C
abgekühlt. Man fügt 0.3 g l'henyljoddichlorid hinzu, hält 2 Stunden bei der genannten Temperatur und versetzt
nochmals mit 0.1 g l'henyljoddichlorid. Nach 20 Stunden bei etwa 4 C dampft man die Lösung ein.
verteilt zwischen Essigester und Wasser, stellt den wäßrigen Auszug mil 2n-Ammoniak alkalisch und extrahiert
erneut mit I ssigester. Die organische Phase wird mil Wasser gewaschen, getrocknet und mit einigen
Tropfen 4n-Salzsäure in Isopropanol versetzt. Das ausgefallene
Hydrochlorid der obengenannten Verbindung wird abgesaugt und mit Äther gewaschen

3 Stunden unter Rückfluß gekocht Man vertreibt das Äthanol im Vakuum und saugt das ausgefallene
Kristallisat ab. Nach Umkristallisieren aus Äthanol/Wasser beträgt der Schmelzpunkt 145 bis
147 C.
Zur tiberführung in das Monohydroehlorid löst
man in der berechneten Menge I η-Salzsäure, bringi
im Vakuum zur Trockne und kristallisiert den festen Rückstand aus Isopropanol/Äther um.

l-(4-Amino-3-trifluormethyl-phen>l)-2-tcrl.-buty
la mino-äthanol
0,45 g I - (4 - Amino - 3 - trifluormcthyl - phcnyl)-2-(N- benzyl - N - lert. - butyl (-amino - äthanol werden
in einer Hydrierbirne in IO ml Methanol und 1,4 ml I η-Salzsäure gelöst und in Gegenwart von 50 mg
Palladium/Kohle-Katalysator (10%) bis zur Aufnahme von I Mol Wasserstoff hydriert. Man trennt vom Katalysator
durch Filtrieren, dampft die Lösung im Vakuum ein und verteilt den Rückstand zwischen Essigester
und 2 η-Ammoniak. Die mit Wasser gewaschene organische Phase wird getrocknet und erneut im Vakuum
eingedampft. Der Rückstand kristallisiert aus Äthanol/Wasser.
Schmelzpunkt: 145 147 C.
l-(4-Amino-3-chlor-5-trifluormethyl-phenyl)-2-tert.-butylamino-äthanoI-hydrochlorid
0,76g I-(4-Amino-3-chlor-5-trifiuormethyl-phenyl)
- 2 - (N - benzyl - N - tert. - butyl) - amino - äthanol werden in einer Hydrierbirne in 20 ml Methanol und
1,95 ml I η-Salzsäure gelöst und in Gegenwart von 80 mg Palladium/Kohle-Katalysator (10%) bis zur
Aufnahme von I Mol Wasserstoff hydriert. Man bricht die Hydrierung ab, entfernt den Katalysator
durch Filtrieren und bringt das Filtrat im Vakuum zur Trockne. Den öligen Eindampfrückstand reinigt man
über eine Säule mit Kieselgel unter Verwendung von Chloroform : Methanol: konz. Ammoniak = 80 zu
20: I als Laufmittel. Die substanzhaltigen Fraktionen
werden vereinigt und im Vakuum vom Lösungsmittel
befreit. Die zurückbleibende, kristallisierte Base der
gewünschten Verbindung wird mit der berechneten Menge 1.07 n-Salzsäurc in Isopropanol in das Hydrochlorid
überführt und aus Essigester/Äther iimkristallisicrt.
Schmelzpunkt: 205 207 C (Zers).

werden in einer Hydrierbirne in 200 ml Methanol gelöst und mit 2 g
I'a Had iumox id/Bariumsulfat- Katalysator (5%) versetzt.
Man hydriert bis I Mol Wasserstoff aufgenommen worden ist. filtriert vom Katalysator ab und

alkalisch gestellt, und die wäürige Phase >o
wird mit Essigester extrahiert. Die organischen Extrakte wäscht man mit Wasser, trocknet und dampft
erneut ein. Den festen Rückstand löst man in Isopropanol und versetzt die Lösung mit 2 Äquivalenten
4 n-Salzsäurc in Isopropanol. Das auskristallisiertc z*.
Dihydrochlorid der genannten Verbindung wird abgesaugt und mit Isopropanol und Äther gewaschen.
Schmelzpunkt: 141.5 142 C (Zers).
Beispiel 7 ,0
l-(4-Amino-3-tril1uormcthyl-phenyl)-2-tert.-pentyI-amino-äthanol-hydrobromid
5 g I -(4-Amino-3-brom-5-trifluormcthyl-phenyl)-2- tert.-pentylamino-äthanol-hydrochlorid werden
zwischen Essigester und 2n-Ammoniak verteilt. Die organische Phase wird getrocknet und im Vakuum
eingedampft. Die zurückbleibende Base löst man in K)OmI Methanol in einer Hydrierbirne, versetzt
mit 2.5 g Palladiumoxid/Bariumsulfat-Katalysator (5%) und hydriert bis zur Aufnahme von I Mol
Wasserstoff. Nach dem Entfj;nen des Katalysators
durch Filtrieren dampft man das Filtrat im Vakuum ein und kristallisiert den festen Rückstand aus Isopropanol
um. Das erhaltene Hydrobromid der ge- 4·, nannten Verbindung schmilzt bei 174 175 C (Zers.).
5-(4-Amino-3-chlor-5-trifluormethyl-phenyl)-3-tert.-buty
1-1,3-oxazolidin
1,35 g I -H-AminoO-chlor-S-trifluormethyl-phenyI)-2-tert.-butyIamino-äthanol
werden in 30 ml Benzol gelöst und mit 1,2 ml 40%iger wäßriger Formaldehydlösung versetzt. Man engt bei Normaldruck
auf das halbe Volumen ein, versetzt mit etwa 35 ml Benzol und kocht 5 Stunden unter Rückfluß. Dann
gibt man weitere 1,5 ml Formaldehydlösung hinzu und wiederholt das beschriebene Verfahren noch
drei Mal. Man bringt im Vakuum zur Trockne. löst in Äther und filtriert von einigen unlöslichen Flocken
ab. Das Filtrat wird mit ätherischer Salzsäure schwach sauer gestellt und das ausgefallene Produkt durch
Reiben kristallisiert. Man saugt ab und kristallisiert aus Aceton/Äther um. Das erhaltene Hydrochlerid
der oben genannten Verbindung schmilzt bei 163 bis 165° C (Zers.).

Verfahren zur Herstellung von neuen Aminophenyl-älhanolaminen der allgemeinen Formel IOACH CW, N(I)H,Nin der A und B je ein WasscrslolTatom oder zu-.summen die Methylengruppe, R, ein Wasserstoff-, Chlor- oder Bromatom, R2 ein Fluoratom, die Cyan- oder Trifluormcthylgruppe und R, eine Alkyl- oder Cycloalkylgruppe, welche jeweils 3 bis 5 Kohlenstoffatome enthalten kann, die 2-Mclhyl-4-hydroxybutyl-(2)-gruppc, die Hydroxy-tert.-butylgruppe, die 3-Äthinylpcntyl-(3)-gruppc oder die I - (3,4 - Methy lendioxyphenyl) - propyl - (2) - gruppe bedeuten, sowie von deren physiologisch verträglichen Säurcadditionssalzcn, dadurch gekenn- so zeichnet, daß in an sich bekannter Weise ein Acetophenon der allgemeinen Formel Ileine Verbindung der allgemeinen Formel IIIOHCHCH1 NH1NR.,R1in der R1 und R, mit Ausnahme des eingangs erwähnten Alkinylrestes wie eingangs definiert sind, in einem I.ösiingsmiltel halogeniert wird oder von einer Verbinduni; der allgemeinen Formel IVOHCU2 N(IV)H -NR1oder von einer Vcrbindunu der allgemeinen Formel IVaOH-CH CH7-N(IVa)oder von einer Vcrbindunu der alliiemeincn Formel IVbH-NOHCH CH1 N(IVb)in denen R1 bis R, wie eingangs definiert sind und Y einen Schutzresl für eine Aminogruppc darstellt, der Schutzrest Y in einem Lösungsmittel abgespalten wird oder, zur Herstellung von Verbindungen der allgemeinen Formel I. i.i der R, ein Wasscrstoffatom darstellt, eine Verbindung der allgjmcincn Formel VCH, NH2NR,(II)fiOin der R, bis R, wie eingangs definiert sind, in einem Lösungsmittel reduziert wird oder, /ur Herstellung von Verbindungen der allgemeinen Formel I. in der R, mit Ausnahme des eingangs erwähnten Alkinylrestes wie eingangs definiert isl und R, ein Chlor- oder Bromalom darstellt.OHHai f ], CH CH, N7V " X RH2N f KlR1(V)in der R2 und R, wie eingangs deliniert sind und Hai ein ChIiT-. Brom- oder lodatom darstellt, in einem Lösungsmittel cnlhalogenicrl wird und gewünschlenfalls eine erhaltene Verbindung der allgemeinen Fo.mel I durch Umsetzung mit Formaldehyd in ein Oxa/.olidin tier allgemeinen Formel I und/oder gewiin•ichlenfalls in ein physiologischverträgliches Säureaddilionssalz übergeführt wird.
