Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Oxazolidinen der Formel I

sowie von deren physiologisch verträglichen Säureadditionssalzen mit anorganischen oder organischen Säuren.
In Formel I bedeuten: Rl ein Wasserstoff-, Fluor-, Chlor-, Brom-, Jodatom oder eine Cyanogruppe; R2 ein Fluoratom, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 5 Kohlenstoffatomen, eine Hydroxyalkyl-, Aminoalkyl-, Dialkyla minoalkyl-, Trifluormethyl-, Alkoxy-, Nitro-, Cyano-, Carboxy-, Carbalkoxy- oder Carbamoylgruppe; R3 ein Wasserstoffatom, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 6 Kohlenstoffatomen, eine Hydroxyalkyl-, Cycloalkyl-, Cycloalkylalkyl-, Alkenyl-, Alkinyl- oder gegebenenfalls substituierte Aralkylgruppe; und R4 ein Wasserstoffatom oder eine geradkettige oder verzweigte Alkylgruppe.
Die Verbindungen der Formel I besitzen wertvolle pharmakologische Eigenschaften, neben einer analgetischen, uterusspasmolytischen und einer antispastischen Wirkung auf die quergestreifte Muskulatur, insbesondere ss2-mimetische und/oder ss1-blockierende Wirkungen, wobei je nach ihrer Substitution die eine oder andere Wirkung im Vordergrund steht. Die d(+)-Verbindungen weisen insbesondere eine selektive Wirkung auf die ss1-Rezeptoren und die 1(-)-Verbindungen eine bevorzugte Wirkung auf die ss2-Rezeptoren auf.

Die Umsetzung mit einem Aldehyd der Formel III erfolgt zweckmässigerweise in einem Lösungsmittel, wie Äthanol, Benzol, Toluol oder Dioxan unter wasserabspaltenden Bedingungen, z. B. in Gegenwart von wasserfreiem Kupfer(ll)- sulfat, bei Temperaturen bis zur Siedetemperatur des verwendeten Lösungsmittels, z. B. bei Temperaturen zwischen 20 und 100 "C, sie kann jedoch auch ohne Lösungsmittel durchgeführt werden.
Besonders vorteilhaft wird die Umsetzung jedoch mit Hilfe eines Wasserabscheiders in Gegenwart eines Lösungsmittels wie Benzol oder Toluol durchgeführt.
Die erhaltenen Verbindungen der Formel I können gewünschtenfalls mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Säureadditionssalze mit 1, 2 oder 3 Äquivalenten der betreffenden Säure übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Zitronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen.
Die als Ausgangsstoffe zu verwendenden Verbindungen können nach literaturbekannten Verfahren, z. B. gemäss belgischem Patent Nr. (Patentanmeldung Nr. 25 144) erhalten werden.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen der Formel I wertvolle pharmakologische Eigenschaften auf, insbesondere eine ss2-mimetische und/oder ss1-blockie- rende Wirkung, wobei je nach ihrer Substitution die eine oder andere Wirkung im Vordergrund steht. Die d(+ > Verbin- dungen weisen insbesondere eine selektive Wirkung auf die ssl-Rezeptoren und die l(-)-Verbindungen eine bevorzugte Wirkung auf die ss2-Rezeptoren auf.
Beispielsweise wurden die Substanz A = 544-Amino-3-brom-5fluor-phenyl)-3-tert.-butyl- oxazolidindihydrochlorid auf ihre Wirkung auf die ss-Rezeptoren untersucht:
Die P-mimetische Wirkung wurde als Antagonismus gegenüber dem durch i.v. Gabe von 20 v/kg Acetylcholin ausgelösten Bronchospasmus des narkotisierten Meerschweinchens in der Versuchsanordnung nach Konzett-Rössler nach i.v. Applikation geprüft. Aus der mit den verschiedenen Dosen erzielten prozentualen Abschwächung des Bronchospasmus wurde durch graphische Extrapolation eine ED50 bestimmt (siehe Tabelle 1).
Die akute Toxizität der Substanzen wurden an Gruppen von je 10 Mäusen bestimmt. Es wurde die LDso, die Dosis bei deren intravenöser Verabreichung 50% der Tiere innerhalb von 14 Tagen verstarben, nach der Methode von Litchfield und Wilcoxon berechnet (siehe Tabellen II).

Die neuen Verbindungen der Formel I lassen sich gegebenenfalls in Kombination mit anderen Wirksubstanzen in die üblichen pharmazeutischen Zubereitungsformen einarbeiten.
Hierbei beträgt die Einzeldosis 1 bis 100 y, vorzugsweise jedoch 5 bis 50 y.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern: Beispiel 1 5-(4-Amino-3-chlor-5-trifluormethyl-phenyl)-3-tert.- butyl-1,3-oxazolidin
1,35 g (1 -(4-Amino-3-chlor-5-trifluormethyl-phenyl)-2-tert.
-butylamino-äthanol werden in 30 ml Benzol gelöst und mit 1,2 ml 400/obiger wässriger Formaldehydlösung versetzt. Man engt bei Normaldruck auf das halbe Volumen ein, versetzt mit etwa 35 ml Benzol und kocht 5 Stunden unter Rückfluss.
Dann gibt man weitere 1,5 ml Formaldehydlösung hinzu und wiederholt das beschriebene Verfahren noch drei Mal. Man bringt im Vakuum zur Trockne, löst in Äther und filtriert von einigen unlöslichen Flocken ab. Das Filtrat wird mit äthe rischer Salzsäure schwach sauer gestellt und das ausgefallene Produkt durch Reiben kristallisiert. Man saugt ab und kristallisiert aus Aceton/Äther um. Das erhaltene Hydrochlorid der oben genannten Verbindung schmilzt bei 163-165 "C (Zers.).
Beispiel 2 5-(4-Amino-3-brom-5-fluor-phenyl)-3-tert.-butyl-oxazolidin
Schmelzpunkt des Dihydrochlorids: 164-178 "C (Zers.).
Hergestellt aus t-(4-Amino-3-brom-5-fluor-phenyl)-2-tert.-bu- tyl- amino-äthanol und 400/obiger Formaldehydlösung analog Beispiel 1.
Beispiel 3 2-Äthyl-5-(4-amino-3-brom-5-fluor.phenyl)-3-tert.- butyl-oxazolidin
5 g 1 -(4-Amino-3-brom-5-fluor-phenyl)-2-tert.-butylamino.ät- hanol werden in 100 ml Benzol gelöst. Hierzu gibt man 5 g Propionaldehyd und erhitzt während 6 Stunden am Wasserabscheider auf Rückflusstemperatur. Danach gibt man erneut 5 g Propionaldehyd hinzu und erhitzt weitere 3 Stunden. Nach dem Abkühlen wird im Vakuum zur Trockne eingeengt und der Rückstand über eine Chromatographie Säule, gefüllt mit 50 g Kieselgel, chromatographiert, wobei Benzol als Elutionsmittel verwendet wird. Die Substanz enthaltenden Eluate werden vereinigt und im Vakuum zur Trockne eingedampft, wobei die genannte Verbindung als Schaum anfällt.

The acute toxicity of the substances was determined in groups of 10 mice each. The LD 50, the dose of which 50% of the animals died within 14 days when administered intravenously, was calculated by the method of Litchfield and Wilcoxon (see Table II).

The new compounds of the formula I can, if appropriate, be incorporated into the customary pharmaceutical preparation forms in combination with other active substances.
The single dose here is 1 to 100 y, but preferably 5 to 50 y.
The following examples are intended to explain the invention in more detail: Example 1 5- (4-Amino-3-chloro-5-trifluoromethyl-phenyl) -3-tert-butyl-1,3-oxazolidine
1.35 g (1 - (4-Amino-3-chloro-5-trifluoromethyl-phenyl) -2-tert.
-butylamino-ethanol are dissolved in 30 ml of benzene and treated with 1.2 ml of 400 / above aqueous formaldehyde solution. It is concentrated to half its volume at normal pressure, mixed with about 35 ml of benzene and refluxed for 5 hours.
A further 1.5 ml of formaldehyde solution is then added and the procedure described is repeated three more times. It is brought to dryness in a vacuum, dissolved in ether and some insoluble flakes are filtered off. The filtrate is made slightly acidic with ethereal hydrochloric acid and the precipitated product is crystallized by rubbing. It is suctioned off and recrystallized from acetone / ether. The hydrochloride of the above compound obtained melts at 163-165 "C (decomp.).
Example 2 5- (4-Amino-3-bromo-5-fluoro-phenyl) -3-tert-butyl-oxazolidine

Example 3 2-Ethyl-5- (4-amino-3-bromo-5-fluoro-phenyl) -3-tert-butyl-oxazolidine
5 g of 1- (4-amino-3-bromo-5-fluoro-phenyl) -2-tert-butylamino-ethanol are dissolved in 100 ml of benzene. 5 g of propionaldehyde are added and the mixture is heated to reflux temperature for 6 hours on a water separator. Then another 5 g of propionaldehyde are added and the mixture is heated for another 3 hours. After cooling, the mixture is concentrated to dryness in vacuo and the residue is chromatographed on a chromatography column filled with 50 g of silica gel, benzene being used as the eluent. The eluates containing the substance are combined and evaporated to dryness in vacuo, the compound mentioned being obtained as a foam.
