Gegenstand der vorliegenden Erfindung ist ein Verfahren zur Herstellung neuer Derivate des 6,11-Dihydro-dibenz[b,e]oxepins der Formel I

in welcher bedeuten: Rt Wasserstoff, Halogen, oder eine Alkyl-, Alkoxybzw. Trifluormethylgruppe, R2 die Hydroxygruppe, Ra Wasserstoff, R4 Wasserstoff oder eine gegebenenfalls substituierte Alkyl- bzw. Aralkylgruppe, R und R6 Wasserstoff oder niedere Alkylgruppen, die auch zusammen eine Alkylenbrücke bilden können.
Es wurde gefunden, dass die Verbindungen I wertvolle pharmakologische Wirkungen aufweisen, insbesondere anticholinergische, antikonvulsive, muskelrelaxierende, sedierende, diuretische und/oder kreislaufaktive Eigenschaften.
Das erfindungsgemässe Verfahren zur Herstellung der neuen Dibenzoxepin-Derivate list dadurch gekennzeichnet, dass man ein Halogenderivat der Formel II

in welcher Hal ein Halogenatom vorstellt mit einem Alkalimetall oder Magnesium zur entsprechenden metallorganischen Verbindung umsetzt und diese mit einer Dibenzoxepin-Verbindung der Formel III

reagieren lässt. Die erhaltenen Verbindungen I kann man anschliessend dehydratisieren, wobei man eine Verbindung erhält, in der R2 und R3 zusammen eine doppelte Bindung darstellen.
Man kann die erhaltenen Verbindungen zur Herstellung von Verbindungen der Formel I verwenden, worin R2 ein Wasserstoffatom darstellt, indem man die Hydroxylgruppe R, im Wege der Hydrierung durch Wasserstoff ersetzt.
Zur Durchführung des erfindungsgemässen Verfahrens werden die Komponenten II und III unter den üblichen Bedingungen der metallorganischen Synthese miteinander umgesetzt. Vorzugsweise verwendet man als Metall Magnesium und arbeitet in einem inerten organischen Lösungsmittel (z. B. Tetrahydrofuran).
Es entstehen im allgemeinen Verbindungen der Formel I, in der R2 eine Hydroxygruppe bedeutet.
Durch Behandlung mit den üblichen dehydratisierenden Mitteln, z. B. alkoholischer Salzsäure, lassen sich hieraus Verbindungen der Formel I herstellen, in der R2 und R3 eine Doppelbindung bedeuten. Diese können anschliessend gewünschtenfalls in üblicher Weise zu Verbindungen der Formel I, in der R2 und R3 Wasserstoffatome bedeuten, hydriert werden, wobei man vorzugsweise die katalytische Hydrierung (z. B.
mittels Raney-Nickel) anwendet. Es ist aber auch möglich, die Verbindungen der Formel I, in der R2 eine Hydroxygruppe bedeutet, in schonender Weise direkt in Verbindungen mit R2 = Wasserstoff zu überführen, wobei man vorteilhaft zunächst die entsprechenden Halogenderivate herstellt (R2 = Hal) und diese z. B.
durch Behandlung mit komplexen Metallhydriden oder durch katalytische Hydrierung in die gesättigten Verbindungen 1 (R2 = Wasserstoff) überführt.
Die basischen Verfahrensprodukte lassen sich mit Hilfe anorganischer oder organischer Säuren in die entsprechenden Salze (z. B. die Hydrohalogenide) überführen.
Beispiel 1
Allgemeine Arbeitsvorschrift zur Herstellung von Verbindungen der Formel I mit R2 = OH
13,3 g 1-Methyl-4-chlor-piperidin (0,1 Mol) werden in 50 ml Tetrahydrofuran mit 2,4 g Magnesium zur entsprechenden metallorganischen Verbindung umgesetzt. Anschliessend lässt man bei Raumtemperatur eine Lösung von 0,05 Mol eines 6,11-Dihydrodibenz[b,e]oxepinons-1 1 in 50 ml Tetrahydrofuran zutropfen, rührt eine Stunde nach und versetzt dann mit gesättigter, wässriger Ammonchloridlösung. Nun wird mit Essigester ausgeschüttelt und die organische Schicht abgetrennt, getrocknet und eingedampft. Die auf diese Weise isolierte Base kristallisiert nach Anreiben mit Petroläther; sie kann durch Behandlung mit einer organischen oder anorganischen Säure in das entsprechende Salz übergeführt werden.
Die auf diese Weise hergestellten Verbindungen sind in der folgenden Tabelle I zusammengefasst.

Beispiel 2
Ailgemeine Arbeitsvorschrift zur Herstellung von Verbindungen der Formel I, in der R2 und R3 eine Doppelbindung bilden
0,05 Mol der nach Beispiel 1 hergestellten 11 -Hydroxy-11-(1 -methyl-piperidinyl-4)-Derivate der 6,11-Dihydro-dibenz[b,e]-oxepine werden in 150 ml gesättigter alkoholischer Salzsäure eine Stunde zum Sieden erhitzt. Anschliessend dampft man die Lösung ein und kristallisiert das entstandene Hydrochlorid aus Essigester oder Aceton um.
Die nach dieser Methode hergestellten Verbindungen sind in der folgenden Tabelle II zusammengefasst.

To carry out the process according to the invention, components II and III are reacted with one another under the usual conditions of organometallic synthesis. Magnesium is preferably used as the metal and the reaction is carried out in an inert organic solvent (e.g. tetrahydrofuran).

13.3 g of 1-methyl-4-chloropiperidine (0.1 mol) are reacted in 50 ml of tetrahydrofuran with 2.4 g of magnesium to give the corresponding organometallic compound. A solution of 0.05 mol of a 6,11-dihydrodibenz [b, e] oxepinone-11 in 50 ml of tetrahydrofuran is then added dropwise at room temperature, the mixture is stirred for one hour and then saturated aqueous ammonium chloride solution is added. It is then extracted with ethyl acetate and the organic layer is separated off, dried and evaporated. The base isolated in this way crystallizes after trituration with petroleum ether; it can be converted into the corresponding salt by treatment with an organic or inorganic acid.
The compounds prepared in this way are summarized in Table I below.

Example 2
General working procedure for the preparation of compounds of the formula I in which R2 and R3 form a double bond
0.05 mol of the 11-hydroxy-11- (1-methyl-piperidinyl-4) derivatives of 6,11-dihydro-dibenz [b, e] -oxepine prepared according to Example 1 are in 150 ml of saturated alcoholic hydrochloric acid for one hour heated to boiling. The solution is then evaporated and the resulting hydrochloride is recrystallized from ethyl acetate or acetone.
The compounds prepared by this method are summarized in Table II below.
