Die Erfindung betrifft ein Verfahren zur Herstellung von neuen Aminophenyläthanolaminen der Formel I

deren optisch aktiven Antipoden und deren physiologisch verträglichen Säureadditionssalzen mit anorganischen oder organischen Säuren.
In Formel I bedeuten:
R1 ein Chlor-, Brom- oder Jodatom;
R2 ein Fluoratom, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 5 Kohlenstoffatomen, eine Hydroxyalkyl-, Aminoalkyl-, Dialkylaminoalkyl-, Trifluormethyl-, Alkoxy-, Nitro-, Cyano-, Carboxy-, Carbalkoxy- oder Carbamoylgruppe; und R3 und R4, die gleich oder verschieden sein können, Wasserstoffatome, geradkettige oder verzweigte Alkylgruppen mit 1 bis 6 Kohlenstoffatomen, Hydroxyalkyl-, Cycloalkyl-, Cycloalkylalkyl- oder gegebenenfalls substituierte Aralkylgruppen.
Die Verbindungen der Formel I besitzen wertvolle pharmakologische Eigenschaften, neben einer analgetischen, uterusspasmolytischen und einer antispastischenWirkung auf die quargestreifte Muskulatur, insbesonderess2-mirnetische und/ oderss1-blockierendeWirkungen, wobei je nach ihrerSubstitution die eine oder andere Wirkung im Vordergrund steht. Die d(+)-Verbindungen weisen insbesondere eine selektive Wirkung auf die ssl-Rezeptoren und die l(-)-Verbindungen eine bevorzugte Wirkung auf die ss2.Rezeptoren auf.
Die Verbindungen der Formel I werden erfindungsgemäss hergestellt, indem man eine Verbindung der Formel II

chloriert, bromiert oder jodiert.
Dabei verwendet man als Chlorierungs-, Bromierungs- oder Jodierungsmittel beispielsweise Chlor, Brom, Jod, Tribromphenolbrom oder Phenyljodidchlorid und arbeitet vorzugsweise in einem Lösungsmittel, z. B. in 50- bis 100%iger Essigsäure, oder in Tetrahydrofuran in Gegenwart einer tertiären organischen Base, gegebenenfalls in Gegenwart eines Schwermetallsalzes wie Quecksilber(II)-oxid und zweckmässigerweise bei Temperaturen zwischen 0 und 50 C. Pro Mol einer Verbindung der Formel II, welche als Base oder auch als Salz, z. B. als Mono-, Di- oder Trihydrochlorid, eingesetzt werden kann, werden zweckmässigerweise 1 Mol an Chlorierungs-, Bromierungs- oder Jodierungsmittel oder ein geringer Überschuss verwendet.
Falls bei der Umsetzung ein halogenwasserstoffsaures Salze entsteht, kann dieses als solches direkt isoliert oder es kann gewünschtenfalls über die Base weiter gereinigt werden.
Erhält man gemäss dem Verfahren der eingangs erwähnten Art eine Verbindung der Formel I, in der R2 eine Cyanogruppe darstellt, so kann diese in die entsprechende Carbamoylverbindung übergeführt werden; erhaltene Carbamoyl- oder Carbalk oxyyerbindungen können in die entsprechende Carboxylverbindung der Formel I mittels Hydrolyse übergeführt werden.
Die erhaltenen Verbindungen der Formel I können gewünschtenfalls mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Säureadditionssalze mit 1, 2 oder 3 Aequivalenten der betreffenden Säure übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Zitronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen.
Fallen die Verbindungen der Formel I als Racemate an, so können sie eine Racematspaltung unterworfen werden. Die Racematspaltung der d,1-Form einer Verbindung der Formeln erfolgt vorzugsweise durch fraktionierte Kristallisation eines Gemisches ihrer diastereomeren Salze mit einer optisch aktiven Säure, z.B. der D(-)-Weinsäure, L(+)-Weinsäure, Dibenzoyl-D-weinsäure, Dibenzoyl-L-weinsäure, (+)-Kampfer- 1 0-sulfonsäure, L( - )-Äpfelsäure, L(+)-Mandelsäure, d-a-Brom- Kampfer-:T-sulfonsäure oder 1-Chinasäure. Die Racematspaltung kann jedoch auch durch Säulenchromatographie an einem optisch aktiven Trägermaterial, z. B. an Acetylcellulose, erfolgen.
Die als Ausgangsstoffe zu verwendenden Verbindungen können nach literaturbekannten Verfahren erhalten werden.
Die Verbindungen der Formel II erhält man z. B. durch Umsetzung der entsprechenden 2-Halogen-acetophenone mit den entsprechenden Aminen und anschliessende Reduktion der erhaltenen Ketone, beispielsweise mit Natriumborhydrid.
Die verwendeten Ausgangsprodukte brauchen nicht in allen Fällen rein dargestellt zu werden, sie können auch zweckmässigerweise als Rohprodukte verwendet werden.
Wie bereits erwähnt, weisen die neuen Verbindung der Formel I wertvolle pharmakologische Eigenschaften auf, insbesondere eine ss2-mimetische und/oder ss,-blockierende Wirkung, wobei je nach ihrer Substitution die eine oder andere Wirkung im Vordergrund steht. Die d(+)-Verbindungen weisen insbesondere eine selektive Wirkung auf die ss1-Rezeptoren und die 1()-Verbindungen eine bevorzugte Wirkung auf die ss2-Rezeptoren auf.

Die ss1-blockierende Wirkung wurde als Antagonismus gegenüber der durch eine Standarddosis von 1,0 v/kg iv. N Isopropyl- noradrenalin-Sulfat ausgelösten Tachykardie an narkotisierten Katzen geprüft. Aus der mit den verschiedenen Dosen erzielten gemittelten prozentualen Abschwächung der durch die N-Isopropyl-noradrenalin-sulfat-bedingten Herzfrequenzzunahme wurde durch graphische Extrapolation eine EDSo bestimmt (siehe Tabellen II und III).
Die ss2-mimetische Wirkung wurde als Antagonismus gegen über dem durch iv. Gabe von 20 v/kg Acetylcholin ausgelösten Bronchospasmus des narkotisierten Meerschweinchens in der Versuchsanordnung nach Konzett-Rössler nach iv. Applikation geprüft. Aus der mit den verschiedenen Dosen erzielten prozentualen Abschwächung des Bronchospasmus wurde durch graphische Extrapolation eine EDso bestimmt (siehe Tabelle I).
Die ss2-blockierende Wirkung wurde als Antagonismus gegenüber der broncholytischen Wirkung geprüft, die mit 5 v/kg i.v. N-Isopropyl- noradrenalin-Sulfat in der Versuchsanordnung nach Konzett-Rössler an narkotisierten Meerschweinchen beobachtet wird, wenn bei dieser der Bronchospasmus mit einer Standardmenge von 20 v/kg iv. Acetylcholin ausgelöst wird (siehe Tabelle III).
Die akute Toxizität der Substanzen wurden an Gruppen von je 10 Mäusen bestimmt. Es wurde die LD50, die Dosis bei deren intravenöser Verabreichung 50% der Tiere innerhalb von 14 Tagen verstarben, nach der Methode von Litchfield und Wilcoxon berechnet (siehe Tabellen II und III).

Die neuen Verbindungen der Formel I lassen sich gege benenfalls in Kombination mit anderen Wirksubstanzen in die üblichen pharmazeutischen Zubereitungsformen einarbeiten. Hierbei beträgt die Einzeldosis 1 bis 100 y, vorzugsweise jedoch 5 bis 50 y.
Die nachfolgenden Beispiele sollen die Erfindung näher erläutern:
Beispiel I 1 - (4-Amino-3-chlor-5-trtfluormethyl-phenyl) -2- tert. -pentylamino--äthanol-hydrochlorid
0,37 g 1-(4-Amino-3-trifluormethyl-phenyl)-2- tert.-pentylamino- äthanol-hydrobromid und 0,2 ml Pyridin werden in 30 ml Tetrahydrofuran gelöst und auf 0 C abgekühlt. Man fügt 0,3 g Phenyljoddichlorid hinzu, hält 2 Stunden bei der genannten Temperatur und versetzt nochmals mit 0,1 g Phenyljoddichlorid. Nach 20 Stunden bei etwa 4" C dampft man die Lösung ein, verteilt zwischen Essigester und Wasser, stellt den wässrigen Auszug mit 2n Ammoniak alkalisch und extrahiert erneut mit Essigester.
Die organische Phase wird mit Wasser gewaschen, getrocknet und mit einigen Tropfen 4n Salzsäure in Isopropanol versetzt. Das ausgefallene Hydrochlorid der obengenannten Verbindung wird abgesaugt und mit Äther gewaschen.
Schmelzpunkt: 176-178 C (Zers.).
Beispiel 2 1-(4-Amino-3-brom-S-methyl-phenyl)-2- tert.-butylamino äthanol-dihydrochlorid
2,6 g 1-(4-Amino-3-methyl-phenyl)-2- tert.-butylamino äthanol-hydrochlorid werden in 90 ml 50%iger Essigsäure gelöst und bei 0-5" mit 1,6 g Brom, gelöst in 5 ml Eisessig und
1 ml Wasser, versetzt. Nach 15 Minuten wird nicht verbrauchtes Brom mit Natriumhydrogensulfit zerstört. Man verdünnt mit Wasser und entfernt unlösliche Nebenprodukte durch Filtration über Kohle. Das Filtrat wird unter Kühlung mit Natronlauge alkalisch gestellt und mit Chloroform extrahiert. Nach dem Trocknen über Natriumsulfat wird die Chloroformlösung über Kohle filtriert und im Vakuum zur Trockne eingeengt. Die so erhaltene ölige Base wird in Isopropanol gelöst und mit isopropanolischer Salzsäure deutlich angesäuert.
Man engt im Vakuum etwas ein, kühlt, saugt das 1-(4-Amino-3-brom-5-methyl-phenyl)-2- tert.-butylamino- äthanol-dihydrochlorid ab und kristallisiert aus Isopropanol/Äther um.
Schmelzpunkt: ab 1480 C Zersetzung.
Beispiel 3 1 - (4-Amino-5-brom-3-hydroxymethyl-phenyl)-2- tert.-butylamino- äthanol
2,0 g 1-(4-Amino-3-hydroxymethyl-phenyl)-2- tert.-butylamino- äthanol werden in 18 ml Eisessig und 2 ml Wasser gelöst und tropfenweise mit 1,3 g Brom versetzt. Anschliessend wird das Reaktionsgemisch eingeengt, der Rückstand in Wasser gelöst, mit 2 n Ammoniak alkalisch gestellt und viermal mit Chloroform ausgeschüttelt. Die organische Phase engt man nach dem Trocknen mit Natriumsulfat ein und reinigt den Rückstand über eine Kieselgelsäule mit Äthanol als Eluens. Die substanzhaltigen Fraktionen werden vereinigt und im Vakuum eingeengt. Der Eindampfrückstand kristallisiert nach längerem Stehen unter Äther.
Schmelzpunkt: 121-124 C.
Beispiel 4 1 - (4-Amino-5-brom-3-hydroxymethyl-phenyl) -2- dimethylamino- äthanol
Schmelzpunkt: 87-92" C.
Hergestellt durch Bromierung von 1-(4-Amino-3-hydroxymethyl-phenyl)-2- dimethylamino- äthanol analog Beispiel 3.
Beispiel 5 1 -(4-Amino-5-brom-3-dimethylaminomethyl-phenyl)-2tert. -butylamino- äthanol
Schmelzpunkt: 127-129 C.
Hergestellt durch Bromierung von 1-(4-Amino-3-dime thylaminomethyl-phenyl)-2-tert.-butylamino-äthanol analog Beispiel 3.
Beispiel 6 1 -(4-Amlno-5-brnm-3-dimethylaminomethyl-phenyl)-2- dimethylamino-äthanol
Hergestellt durch Bromierung von 1-(4-Amino-3 -dimethyl- aminomethyl-phenyl)-2- dimethylamino-äthanol analog Beispiel 3.
Strukturbeweis durch NMR-Spektrum (CDCl3/CD3OD):
2,2 ppm Singulett [6 Protonen, N(CH3)2], 2,35 ppm Singulett [6 Protonen, N(CH3)2j, 1,8-3,0 ppm Multiplett (2 Protonen, CH2), 3,45 ppm Singulett (2 Protonen, CH2), 4,44,8 ppm Multiplett (1 Proton, CH); 6,96 ppm Doublett und 7,4 ppm Doublett (2 aromatische Protonen).
Beispiel 7 1 -(4-Amino-3-fluor-5-iod-phenyl)-2- isopropylamino- äthanolhydrochlorid
5,15 g 1-(4-Amino-3 -fluor-phenyl)-2- isopropylamino äthanol werden in 300 ml Essigsäure gelöst, mit 80 g Jod und 4,0 g Quecksilber-(II)-oxid versetzt und 2+ Stunden lang bei Raumtemperatur kräftig gerührt. Anschliessend wird vom Festkörper abfiltriert, das dunkelbraune Filtrat mit gesättigter Natriumhydrogensulfit-Lösung entfärbt und mit Wasser auf etwa 11 verdünnt Unter Kühlung wird mit 10 n Natronlauge alkalisch gestellt und mit Chloroform extrahiert. Die Chloroform-Phase wird über Natriumsulfat getrocknet und im Vakuum zur Trockne eingeengt. Der Rückstand wird in Methanol gelöst und mit ätherischer Salzsäure auf pH 4,5 angesäuert.
Die Lösung wird im Vakuum zur Trockne eingeengt und der feste Rückstand aus absolutem Aethanol kristallisiert.
Schmelzpunkt: 203-205 C (Zers.).
Beispiel 8 d-l -(4-Amino-3-chlor-5-fluor-phenyl)-2- tert.-butylamino äthanol
0,26 g d- 1-(4-Amino-3-fluor-phenyl)-2- tert.-butylamino äthanol-hydrochlorid und 0,2 ml Pyridin werden in 30 ml Tetrahydrofuran gelöst und auf 0 C abekühlt. Man fügt 0,3 g Phenyljoddichlorid hinzu, hält 2 Stunden bei der genannten Temperatur und versetzt nochmals mit 0,1 g Phenyljoddichlorid. Nach 20 Stunden bei etwa 4" C dampft man die Lösung ein, verteilt zwischen Essigester und Wasser, stellt den wässrigen Auszug mit 2 n Ammoniak alkalisch und extrahiert erneut mit Essigester. Die organische Phase wird mit Wasser gewaschen, getrocknet und im Vakuum zur Trockne eingeengt.
Man löst den Rückstand in absolutem Äthanol, neutralisiert mit äthanolischer Salzsäure und bringt das Hydrochlorid der genannten Verbindung durch Zugabe von Äther zur Kristallisation.
Schmelzpunkt: 210-211"C (Zersetzung).

Beispiel 9 d-1- (4-Amino-3-brom-5-fluor-phenyl)-2- tert. -butylamino äthanol
0,26 g d-1-(4-Amino-3-fluor-phenyl)-2- tert.-butylamino äthanol-hydrochlorid werden in 30 ml 50%iger Essigsäure gelöst und bei 0-5" C mit 0,16 g Brom, gelöst in 5 ml Eisessig und 1 ml Wasser, tropfenweise versetzt. Nach 15 Minuten wird das Reaktionsgemisch eingeengt, der Rückstand in Wasser gelöst, mit 2 n Ammoniak alkalisch gestellt und mit Chloroform extrahiert. Die Chloroformlösung wird mit Natriumsulfat getrocknet und im Vakuum zurTrockne eingeengt. Man überführt den Rückstand durch Lösen in Äthanol, Neutralisation mit äthanolischer Salzsäure und Zugabe von Äther in das Hydrochlorid der genannten Verbindung.
Schmelzpunkt: 2342350 C (Zersetzung).

205 g d,l-1- (4-Amino-3-brom-5-fluor-phenyl)-2- tert.butylamino-äthanol und 118 g Dibenzoyl-D-weinsäure werden in 2,5 1 heissem Äthanol gelöst, filtriert und einen Tag bei Raumtemperatur zur Kristallisation stehen gelassen. Das erhaltene Produkt wird sechsmal aus Methanol-Äther umkristallisiert, wobei man das reine d-[1-(4-Amino-3-brom-5fluor-phenyl)-2- tert.-butylamino- äthanol]- dibenzoyl-D-tartrat vom Schmelzpunkt 206-208" C (Zersetzung) erhält.

Das Salz wird in Methanol und konzentriertem Ammoniak unter Erwärmen gelöst und die Base durch Zusatz von Wasser zur Kristallisation gebracht. Die erhaltene Base löst man in absolutem Äthanol, neutralisiert mit absoluter äthanolischer Salzsäure und vervollständigt die Kristallisation des d-1-(4 Amino-3-brom-5- fluor-phenyl)-2- tert.-butylamino- äthanolhydrochlorids durch Zusatz von Äther.

Die Mutterlaugen der Fällung des d-[1-(4-Amino-3-brom
5- fluor-phenyl)-2- tertl-butylamino-äthanolj- dibenzoyl-D tartrats und die der ersten Umkristallisation werden vereinigt, auf ein kleineres Volumen eingeengt und durch Zugabe von konzentriertem Ammoniak und Wasser die Base gefällt. 140 g so erhaltenes 1-(4-Amino-3-brom-5- flur-phenyl)-2- tert. butylamino-äthanol (1-Form angereichert) werden in 1,8 1 absolutem Äthanol gelöst und mit einer Lösung von 82 g
Dibenzoyl-L-weinsäure in 500 ml absolutem Äthanol versetzt, auf ein Volumen von 11 eingeengt und drei Tage bei Raumtemperatur zur Kristallisation stehen gelassen. Das erhaltene Produkt wird sechsmal aus Methanol/Äther umkristallisiert.
Dabei erhält man das 1 -[ l-(4-Amino-3-brom-5-fluor- phenyl)-2- tert.-butylamino-äthanol]- dibenzoyl-L-tartrat in reiner Form.

Das Salz löst man in Methanol und konzentriertem Ammoniak unter Erwärmen und fällt die Base durch Zusatz von Wasser aus. Beispiel 16 1-(4-Amino-3-brom-S-carboxy-phenyl)-2- dimethylamino äthanol
2 g 1-(4-Amino-3-brom-5-cyan-phenyl)-2- dimethylamino äthanol werden 4 Stunden mit einer Lösung von 5 g Natriumhydroxyd in 120 ml 50%igem Äthanol gekocht. Man destilliert das Äthanol ab, verdünnt die verbleibende wässrige Lösung mit 100 ml Wasser und extrahiert dreimal mit Chloroform. Die wässrige Phase wird neutralisiert, zur Trockne eingeengt, der Rückstand wird mit Äthanol behandelt, abfiltriert, das Filtrat eingeengt, der Rückstand wieder mit Äthanol behandelt und abfiltriert.
Beim Eindampfen des letzten Filtrats verbleibt ein Rückstand von 1-(4-kmino-3-brom-5- carboxy-phenyl)-2- dimethylamino-äthanol, derbei 240-250 C (Zers.) schmilzt und dessen Struktur durch sein IR- und UV-Spektrum sichergestellt wurde: IR (KBr): CCC bei 1620 cm-t, NH+ bei 2000-3500 cm-1. UV (Äthanol): Maxima bei 220 und 330-340 nm, Schulter bei 250 nm.

Example 15 1 - (4-Amino-3-bromo-5-carbamoyl-phenyl) -2 - dimethylamino ethanol
2 g of 1- (4-amino-3-bromo-5-cyano-phenyl) -2-dimethylamino ethanol are boiled for 4 hours with a solution of 5 g of sodium hydroxide in 120 ml of 50% ethanol. The ethanol is distilled off and the remaining aqueous solution is diluted with
100 ml of water and extracted 3 times with chloroform. The chloroform solution is dried over sodium sulfate and evaporated in vacuo. The residue becomes solid and then recrystallized from chloroform.
Melting point: from 93 "C (decomp.).
Example 16 1- (4-Amino-3-bromo-S-carboxy-phenyl) -2-dimethylamino ethanol
2 g of 1- (4-amino-3-bromo-5-cyano-phenyl) -2-dimethylamino ethanol are boiled for 4 hours with a solution of 5 g of sodium hydroxide in 120 ml of 50% ethanol. The ethanol is distilled off, the remaining aqueous solution is diluted with 100 ml of water and extracted three times with chloroform. The aqueous phase is neutralized, concentrated to dryness, the residue is treated with ethanol and filtered off, the filtrate is concentrated, the residue is treated again with ethanol and filtered off.
When the last filtrate is evaporated, a residue of 1- (4-kmino-3-bromo-5-carboxy-phenyl) -2-dimethylamino-ethanol, which melts at 240-250 C (decomp.)
