Gegenstand der Erfindung ist ein Verfahren zur Herstellung neuer racemischer und optisch aktiver Aminophenyl-äthanolamine der allgemeinen Formel

und deren physiologisch verträglichen Additionssalze mit anorganischen und organischen Säuren.
In der obigen allgemeinen Formel (t) bedeutet
R Wasserstoff, Fluor, Chlor, Brom, Jod oder die Cyangruppe,
R Fluor, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 5 Kohlenstoffatomen, eine Hydro- xyalkyl-, Aminoalkyl-, Dialkylaminoalkyl-, Trifluormethyl-, Alkoxy-oder Cyangruppe und R Wasserstoff, eine geradkettige oder verzweigte Alkylgruppe mit 1 bis 6 Kohlenstoffatomen, eineHy- droxyalkyl-, Cycloalkyl-oder Cycloalkylalkylgruppe.
Die Verbindungen der obigen allgemeinen Formel (1) besitzen wertvolle pharmakologische Eigenschaften, neben einer analgetischen, uterusspasmolytischen und einer antispastischen Wirkung auf die quergestreifte Muskulatur insbesondere ss2 -mimetische und/oder ss 1-blockierende Wirkungen, wobei je nach ihrer Substitution die eine oder andere Wirkung im Vordergrund steht. Die d (+)-Verbindungen weisen insbesondere eine selektive Wirkung auf die ss1-Rezeptoren und die 1(-)-Verbindungen eine bevorzugte Wirkung auf die -Rezeptoren auf.
Die neuen Aminophenyl-äthanolamine lassen sich durch Hydrogenolyse einer Verbindungder allgemeinen Formel

in der R1'R 2 und R wie eingangs definiert sind und Y eine Benzylgruppe darstellt, herstellen.
Die Hydrogenolyse erfolgt mit Wasserstoff in Gegenwart eines Katalysators, wie Palladium auf Kohle, Palladiumoxydhydrat auf Bariumsulfat oder Platin, vorzugsweise in einem Lösungsmittel wie Methanol, Methanol/Salzsäure oder Äthanol, bei Raumtemperatur oder leicht erhöhten Temperaturen und bei Normaldruck oder leichtem Überdruck.
Die erfindungsgemäss erhaltenen Verbindungen der allgemeinen Formel (t) lassen sich anschliessend gewunschtenfalls in ihre optisch aktiven Antipoden mittels Racematspaltung zerlegen.
Die Racematspaltung der d, l-Form einer Verbindung der obigen allgemeinen Formel (I) erfolgt durch fraktionierte Kristallisation eines Gemisches ihrer diastereomeren Salze mit einer optisch aktiven Säure, z.B. der D(-)-Weinsäure, L(+)-Weinsäure, Dibenzoyl-D-weinsäure, Dibenzoyl-L-weinsäure, (+)-Kampfer- - 10-sulfonsäure, L (-)-Apfelsäure, L (+)-Mandelsäure, d-α-Brom-kampfer-#-sulfonsäure oder l-Chinasä re.
Die erhaltenen Verbindungen der allgemeinen Formel (I) können gewünschtenfalls mit anorganischen oder organischen Säuren in ihre physiologisch verträglichen Säureadditionssalze mit 1, 2 oder 3 Äquivalenten der betreffenden Säure übergeführt werden. Als Säuren haben sich beispielsweise Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Milchsäure, Citronensäure, Weinsäure, Maleinsäure oder Fumarsäure als geeignet erwiesen.
Die als Ausgangsstoffe verwendeten Verbindungen erhält man nach literaturbekannten Verfahren, So erhält manbeispielsweise die alsAusgangsstoffe verwendeten Verbindungen der allgemeinen Formel (H) durch

Umsetzung der entsprechenden 2-Halogen-acetophenone mit den entsprechenden Aminen und anschliessende Reduktion der erhaltenen Ketone, beispielsweise mit Natriumborhydrid, oder auch durch Halogenierung einer entsprechenden Verbindung oder durch katalytische Reduktion einer entsprechenden 4 - Nitrophenylverbindung.
Die verwendetenAusgangsprodukte brauchen nicht In allen Fällen rein dargestellt zu werden, sie können auch als Rohprodukte verwendet werden.
Wie bereits eingangs erwähnt, weisen die neuen Verbindungen der allgemeinen Formel (t) wertvolle pharmakologische Eigenschaften auf, Insbesondere eine -mimetische und/oder ss1-blockierende Wirkung, wobei je nach ihrer Substitution die eine oder andere Wirkung Im Vordergrund steht. Die d (+) -Verbindungen weisen insbesondere eine selektive Wirkung auf die ss-Rezeptoren und die l (-)-Verbindungen eine bevorzugte Wirkung auf die -Rezeptoren auf.

Die -blockierende Wirkung wurde als Antagonismus gegenüber der durch eine Standarddosis von 1,0 γ/kg i.v. N-Isoproyl-noradrenalin-Sulfat ausgelösten Tachykardie an narkotisierten Katzen geprüft.
Aus der mit den verschiedenen Dosen erzielten gemittelten prozentualenAbschwächung der durch die N-Isopropyl-noradrenalin-sulfatbedingten Herzfrequenzzunahme wurde durch graphische Extrapolation eine ED bestimmt (s. Tabellen II und III),
Die ss-mimetische Wirkung wurde als Antagonismus gegenüber dem durch i. v. Gabe von 20 7/kg Acetylcholin ausgelösten Bronchospasmus des narkotisierten Meerschweinchens In der Versuchsanordnung nach Konzett-Rössler nach i. v. Applikation geprüft. Aus der mit den verschiedenen Dosen erzielten prozentualen Abschwächung des Bronchospasmus wurde durch graphische Extrapolation eine ED bestimmt (s. Tabelle I).
Die ss-blockierende Wirkung wurde als Antagonismus gegenüber der broncholytischen Wirkung geprüft, die mit 5 γ/kg i.v. N-Isopropyl-noradrenalin-Sulfat in der Versuchsanordnung nach Konzett-Rössler an narkotisierten Meerschweinchen beobachtet wird, wenn bei dieser der Bronchospasmus mit einer Standardmenge von 20 y/kg i. v. Acetyleholin ausgelöst wird (s. Tabelle III).
Die akute Toxizität der Substanzen wurde an Gruppen von je 10 Mäusen bestimmt. Es wurde die LD, die Dosis bei deren intravenöser Verabreichung 50% der Tiere innerhalb von 14 Tagen verstarben, nach der Methode von Litchfield und Wilcoxon berechnet (s.

Die neuen Verbindungen der allgemeinen Formel g) lassen sich gegebenenfalls in Kombination mit andern Wirksubstanzen in die üblichen pharmazeutischen Zubereitungsformen einarbeiten. Hiebei beträgt die Einzeldosis 1 bis 100 γ, vorzugsweise jedoch 5 bis 50 y.

Die zurückbleibende, kristallisierte Base der gewünschten Verbindung wird mit der berechneten Menge 1, 07n Salzsäure in Isopropanol in das Hydrochlorid überführt und aus Essigester/Äther umkristallisiert.

Die Hydrierung wird nach Aufnahme von 41 ml Wasserstoff unterbrochen.
Man filtriert vom Katalysator ab und engt das Filtrat im Vakuum zur Trockne ein. Der Rückstand wird in Äthanol gelöst und durch Zugabe von Äther als Dihydrochlorid gefällt.

B e i s p i e l 4: 1-(4'-Amino-3'-methoxyphenyl)-2-tert. butylaminoäthanol
