まず、本発明の1つの態様に係る多孔性セルロース粒子の製造方法について説明する。本発明の多孔性セルロースの製造方法は、以下の(a)~(e)の工程を、この順で含む:
(a)二酢酸セルロースを溶媒に溶解して、二酢酸セルロース溶液を調製することと、
(b)前記二酢酸セルロース溶液を、該二酢酸セルロース溶液と混和しない媒質中に分散させて、分散系を得ることと、
(c)前記分散系を冷却することと、
(d)冷却された前記分散系に貧溶媒を添加することにより、酢酸セルロース粒子を析出させることと
(e)前記酢酸セルロース粒子を鹸化すること。
以下、上記各工程について順に説明する。
(a)においては、原料の二酢酸セルロースを溶媒に溶解して、二酢酸セルロース溶液を調製する。酢酸セルロースは、天然の高分子であるセルロースを酢酸エステル化することにより得られる半合成高分子である。工業的に広く使用されている酢酸セルロースは、二酢酸セルロースと三酢酸セルロースの2種類に大きく分けられ、その一般的な酢化度は、それぞれ約50~57%および約60~62%である。本発明の方法では、原料として、二酢酸セルロースを使用する。本発明において使用される二酢酸セルロースは、一般的に二酢酸セルロースと定義され得るものであれば特に限定されないが、酢化度が45~57%であることが好ましく、53~56%であることがより好ましい。酢化度が45~57%である二酢酸セルロースを使用することにより、より多くの種類の溶剤に溶解させることができる。
(b)においては、(a)で得られたニ酢酸セルロース溶液を、該二酢酸セルロース溶液と混和しない媒質(以下、分散媒とも称する)中に加え、撹拌する。それにより、分散媒中に二酢酸セルロース溶液の液滴が分散した分散系が得られる。
(c)においては、上記(b)で得られた分散系を冷却する。冷却することにより、得られるセルロース粒子の細孔径および粒径を制御することができる。
(d)においては、上記(c)で冷却した分散系に貧溶媒を添加する。それにより、ニ酢酸セルロース粒子を析出させることができる。
析出したニ酢酸セルロース粒子は、当業者に周知のいずれかの方法により分離され、次の(e)に供される。分離は、例えば、ろ過等により行うことができる。
(e)においては、上記(d)で析出させたニ酢酸セルロース粒子を鹸化する。それにより、ニ酢酸セルロースのエステル部分が加水分解して、セルロース粒子が得られる。ここで得られるセルロース粒子は、多孔性を有する。
また、二酢酸セルロースを溶解する溶媒として酢酸水溶液等の水性溶媒を使用した場合、得られる多孔性セルロース粒子の粒径は大きくなる傾向にある。一方、二酢酸セルロースを溶解する溶媒として有機溶媒を使用した場合、得られる多孔性セルロース粒子の粒径および細孔径は、小さくなる傾向にある。
また、使用する界面活性剤の添加量を多くすると、得られる多孔性セルロース粒子の粒径は小さくなり、逆に界面活性剤の量を少なくすれば、得られる多孔性セルロース粒子の粒径は大きくなる傾向にある。また、使用する界面活性剤の種類を変更することで、得られる多孔性セルロース粒子の粒径分布を変えることが可能である。ただし、界面活性剤の量や種類を変えても、多孔性セルロース粒子の細孔径には影響をさほど及ぼさない。
本発明の1つの実施形態によると、上記製造方法により製造され得る多孔性セルロース粒子が提供される。この多孔性セルロース粒子は、各種物質の分離精製において使用することができる。例えば、サイズ排除クロマトグラフィー等のゲル濾過法において、分子サイズの異なる物質を分別するために使用することができる。この際、本発明により得られる多孔性セルロース粒子をそのまま適用してもよいし、さらに置換基により修飾したり、架橋反応させた形態で適用してもよい。
まず、反応容器に硫酸化剤を準備する。本発明に用いる硫酸化剤は、セルロース粒子中の水酸基と反応して、多孔性セルロース粒子に硫酸基を導入できるものであれば特に限定されなく、そのような硫酸化剤としては、例えば、クロルスルホン酸-ピリジン錯体、ピペリジン-N-硫酸、無水硫酸-ジメチルホルムアミド錯体、三酸化硫黄-ピリジン錯体、三酸化硫黄-トリメチルアミン錯体、硫酸-トリメチルアミン複合体等を挙げることができる。硫酸化剤の使用量は、目的とする硫酸基の導入率及び反応条件によって任意に選択すればよく、例えば、多孔性セルロース粒子中の水酸基に対し、0.001~1当量を用いるのが適当である。
反応終了後、反応混合物にアルカリ水溶液、例えば水酸化ナトリウム水溶液を加えて中和してもよい。
その後、得られた反応混合物を濾過または遠心分離することにより、生成物を回収し、水で中性になるまで洗浄して、目的の硫酸化多孔性セルロース粒子を得ることができる。硫酸化多孔性セルロース粒子中の硫酸基の導入量は、硫酸化剤の使用量を変更することなどによって調整することができ、クロマトグラフィー充填剤の用途などに応じて適宜決定すればよい。
多孔性セルロース粒子のスルホン化処理は、特開2001-302702号公報または特開平9-235301号公報などを参照して行うことができる。実験条件を適宜設計変更することにより、目的のスルホン酸基含有基を目的の量だけ導入することができる。
85重量%酢酸水溶液352.5gに、二酢酸セルロース48.1g(和光純薬試薬、酢化度:53~56%)を加えて攪拌した。さらに、昇温して60℃で1時間撹拌することにより二酢酸セルロースを溶解し、ニ酢酸セルロース濃度が12重量%である透明な溶液を得た。該溶液を、界面活性剤ソルビタンモノオレート1.97gを含む、100℃のo-ジクロロベンゼン1.5L中に素早く注ぎ、回転数400rpmにて10分間攪拌して分散系を得た。続いてこの分散系を冷却し、30℃になったところで、貧溶媒である純水620mlを滴下した。その結果、ニ酢酸セルロースが析出し、球状の二酢酸セルロース粒子が得られた。その後、得られたニ酢酸セルロース粒子を大量のメタノール、次いで水で十分に洗浄した。そして、洗浄後のニ酢酸セルロース球状粒子を、目開き600μmの篩を用いて分級した。
上記実施例1で得られた多孔性セルロース粒子について、粒度分布を測定し、平均粒径を求めた。測定に使用した装置は、以下の通りである。
装置:Laser Scattering Particle Size distribution Analyzer Partica LA-950(HORIBA製)
上記装置を使用してメジアン径を測定することにより、平均粒径を算出した。
上記実施例1で得られた多孔性セルロース粒子について、Kav値を測定した。測定方法は、以下の通りである。
(1)使用機器及び試薬
カラム:エンプティカラム1/4×4.0mm I.D×300mm、10F(東ソー)
リザーバー:パッカ・3/8(東ソー)
ポンプ:POMP P-500 (Pharmacia)
圧力計:AP-53A(KEYENCE)
カラムとリザーバーとを接続し、カラム下部にエンドフィッティングを接続した。Kavを測定するセルロース粒子を減圧濾過した湿ゲルの状態で15g計りとり、50mLビーカーへ入れた。そこへ超純水20mLを加えて軽く攪拌した。これを、セルロース粒子が超純水に分散した状態でリザーバーの壁を伝わるようにカラムにゆっくりと加えた。ビーカーに残ったセルロース粒子を少量の超純水ですすぎ、ゆっくりとカラムに加えた。その後、リザーバーの上部ぎりぎりまで超純水を加え、リザーバーの蓋をした。リザーバーの上部にアダプターを接続し、ポンプで超純水を送液した。送液ラインの途中には、圧力計を接続しておき、圧力をモニターした。圧力が0.3MPaになるまで流速を上げ、その後30分間超純水を流しながら充填した。充填が終わったらポンプを止め、アダプターとリザーバーの蓋を外した。次に、リザーバーの中の超純水をピペットで吸い出した。リザーバーを外し、カラムからはみ出したセルロース粒子を除いて、エンドフィッティングを接続した。

セルロース粒子を充填したカラムをKav測定装置にセットした。流量0.4mL/minの流速で60分間、超純水を通液した。Kav測定サンプル10μLをカラムにアプライして、45分間超純水を通液した。Kav測定サンプルの検出はRI検出器を用いて行い、測定チャートを記録した。これらの操作を各Kav測定サンプルごとに実施した。得られた測定チャート(縦軸:RI検出強度、横軸:時間)は、正規分布を示した。RI検出強度が極大になるような時間を記録した。
Kav測定で得られた、RI検出強度が極大になるような時間と、そのときの通液量から、Kav測定サンプルの保持容量(mL)を算出した。
Kavは、以下の式にて算出した。
Kav=(Ve-V0)/(Vt-V0)
[式中、Veはサンプルの保持容量(mL)、Vtは空カラム体積(mL)、V0はデキストランT2000保持容量(mL)である。]
縦軸をKav値、横軸を分子量としてプロットしたときに、Kav値がゼロになる分子量が排除限界分子量である。
二酢酸セルロース溶液における二酢酸セルロース濃度を10重量%(実施例2)および4重量%(実施例3)としたことを除き、実施例1と同様にして多孔性セルロース粒子を製造した。得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
ニ酢酸セルロース溶液におけるニ酢酸セルロース濃度を10重量%としたこと、および貧溶媒として50%メタノール水溶液620mLを使用したことを除き、実施例1と同様にして多孔性セルロース粒子を製造した。得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
ニ酢酸セルロース溶液におけるニ酢酸セルロース濃度を6重量%としたことを除き、実施例4と同様にして多孔性セルロース粒子を製造した。得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
85重量%酢酸水溶液352.5gに、二酢酸セルロース39.2g(和光純薬試薬、酢化度:53~56%)を加えて攪拌した。さらに、昇温して60℃で1時間撹拌することにより二酢酸セルロースを溶解し、ニ酢酸セルロース濃度が10重量%である透明な溶液を得た。該溶液を、界面活性剤ソルビタンモノオレート1.97gを含む、100℃のo-ジクロロベンゼン1.5L中に素早く注ぎ、回転数400rpmにて10分間攪拌して分散系を得た。続いてこの分散系を0℃まで冷却し、そのまま0℃で1時間撹拌した後、貧溶媒である5℃以下の純水620mlを滴下した。その結果、ニ酢酸セルロースが析出し、球状のニ酢酸セルロース粒子が得られた。その後の洗浄および鹸化の工程は、実施例1と同様に行った。
得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
ニ酢酸セルロース溶液におけるニ酢酸セルロース濃度を8重量%(実施例7)、6重量%(実施例8)および4重量%(実施例9)としたことを除き、実施例6と同様にして多孔性セルロース粒子を製造した。得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
シクロヘキサノン352.5gに、二酢酸セルロース39.2g(和光純薬試薬、酢化度:53~56%)を加えて攪拌した。さらに、昇温して90℃で1時間撹拌することにより二酢酸セルロースを溶解し、二酢酸セルロース濃度が10重量%である透明な溶液を得た。該溶液を、界面活性剤ドデシルベンゼンスルホン酸ナトリウム1.97gを含む、90℃の水1.5L中に素早く注ぎ、回転数400rpmにて10分間攪拌して分散系を得た。続いてこの分散系を冷却し、30℃になったところで、貧溶媒であるメタノール620mlを滴下した。その結果、ニ酢酸セルロースが析出し、球状のニ酢酸セルロース粒子が得られた。その後の洗浄および鹸化の工程は、実施例1と同様に行った。
得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
実施例10と同様に、ニ酢酸セルロース分散系を調製した。その後、この分散系を0℃まで冷却し、そのまま0℃で1時間撹拌した後、貧溶媒である冷メタノール620mlを滴下した。その結果、ニ酢酸セルロースが析出し、球状のニ酢酸セルロース粒子が得られた。その後の洗浄および鹸化の工程は、実施例1と同様に行った。
得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
三酢酸セルロースを原料に用いて製造されている市販セルファインサルフェート(JNC社製、S含量950μg/g)を比較例に用いた。該品の粒度分布およびKav値を、実施例1と同様に測定した。
60重量%チオシアン酸カルシウム水溶液に、結晶性セルロース(旭化成:セオラスPH-101)を加えて110℃で撹拌し、セルロース濃度6重量%の溶液を調製した。該溶液を、界面活性剤ソルビタンモノオレエートを含む130℃のo-ジクロロベンゼン1.5L中に撹拌しながら注ぎ、分散系を得た。続いて、この分散系を冷却し、40℃になったところで、貧溶媒であるメタノールを滴下した。その結果、セルロースが析出し、球状のセルロース粒子が得られた。その後、多量のメタノールおよび水で洗浄を行った。
得られた多孔性セルロース粒子の粒度分布およびKav値を、実施例1と同様に測定した。
以下に、本発明の多孔性セルロース粒子の好ましい使用形態の一つであるクロマトグラフィー用充填剤に関する実施例を示す。
実施例6で製造した多孔性セルロース粒子を用い、以下の試料1~3のそれぞれの溶出時間(インジェクションから溶出ピークの最大値までの時間)を測定した。この測定は、移動相及び試料の溶剤として0.05Mリン酸pH8.0+0.5MNaClを用いた以外は、前述のゲル分配係数Kavの測定と同様に行った。

以下に、本発明の多孔性セルロース粒子の好ましい使用形態の一つである吸着剤に関する実施例を示す。
実施例2、実施例6、および比較例2により得られた多孔性セルロース粒子を、目開き125μm、目開き53μmの篩に通し、粒径125~53μmのセルロース粒子を分取した。得られたセルロース粒子を1mol当量のクロロスルホン酸と65℃で反応させ、硫酸化セルロース粒子を得た(以下、実施例2A、実施例6A、比較例2Aとする)。各硫酸化セルロース粒子のS含量(すなわち、硫酸化セルロース粒子1gに対する硫黄含量)をイオンクロマトグラフィー法により求めたところ(詳細は以下の試験例3に示す)、実施例2A:14000μg/g、実施例6A:18000μg/gであった。比較例2Aについては、多孔性セルロース粒子を上記方法により硫酸化しようとしたところ粒子が崩壊してしまったため、使用できなかった。
S含量は、イオンクロマトグラフィー法により求めることが出来る。具体的には、以下に記載の方法によって求めた。60℃で16~20時間真空乾燥したサンプルを乳鉢ですりつぶし、更に、105℃にて2時間乾燥した。この乾燥試料0.05gに2Mの塩酸2.5mlを加え、110℃にて16時間加水分解した。氷冷後、上澄液を1ml採取し、2Mの水酸化ナトリウム水溶液で中和し、25mlにメスアップした。カラムに横河電機社製ICS-A-23を用い、オーブン温度40℃、溶離液に3mM Na2CO3溶液、除去液に15mM硫酸をそれぞれ1ml/minの流量の条件で使用し、横河電機社製IC7000イオンクロマトアナライザーを用いて分析し、さらに後述の標準溶液から作成した検量線をもとにSO4濃度を求めた。ブランク値は乾燥試料を加えずに同様に操作した時の値とした。SO4標準液(関東化学社製 陰イオン混合標準液IV)の2μg/ml液を本測定法の標準溶液とし、これを更に段階希釈したものを同様の条件でイオンクロマトアナライザーにて分析し、検量線を作成した。硫黄含有割合は下記式にて求めた。なお、X試料、XブランクはSO4標準溶液による検量線から求めた濃度(×10-4%)である。

上記式により算出した硫黄含有割合に基づいて、修飾されたセルロース粒子(例えば、硫酸化セルロース粒子またはスルホン酸化セルロース粒子)の乾燥重量1g当りの硫黄含量(重量)を算出した。
各セルロース粒子を、50mMTris-HCl緩衝液(pH9.5)で十分に洗浄した。測定対象であるタンパク質を650mg用意し、50mMTris-HCl緩衝液(pH9.5)130mLに溶解してタンパク質溶液を調製した。タンパク質としては、以下のものを使用した。
(1)ヒトγグロブリン、血清由来(和光純薬社製)
(2)リゾチーム、鶏卵白由来(和光純薬社製)
表6の結果の通り、本発明の実施形態に係る多孔性セルロース粒子に適切なリガンドを導入することで、市販品と同等以上の吸着量を有する吸着剤としての利用が可能であることが判った。
文献:Microbiol Immunol 2012;56: 490-495を参考に、以下の手順で、インフルエンザウイルス吸着試験を行った。
(不活化ウイルス含有液の調製)
インフルエンザウイルスA/duck/Hokkaido/Vac-2/2004(H7N7)株を、発育鶏卵を用いて増殖させた。しょう尿液を回収して遠心し、上清を回収した。これに終濃度0.1%となるようβ-プロピオラクトンを加え、ウイルスを不活性化した。この不活性化ウイルス液を孔径0.45μmセルロースアセテート製メンブレンフィルターでろ過し、不活化ウイルス含有液として試験に供した。
実施例2、6および10により得られた多孔性セルロース粒子を、目開き125μm、目開き53μmの篩に通し、粒径125~53μmのセルロース粒子を分取した。得られたセルロース粒子を1mol当量のクロロスルホン酸と65℃で反応させ、硫酸化セルロース粒子を得た(以下、実施例2A、実施例6A、実施例10Aとする)。また、実施例10により得られた多孔性セルロース粒子を1mol当量のクロロスルホン酸と75℃で反応させ硫酸化セルロース粒子を得た(以下、実施例10Bとする)。各硫酸化セルロース粒子のS含量(すなわち、硫酸化セルロース粒子1gに対する硫黄含量)をイオンクロマトグラフィー法により求めたところ、実施例2A:14000μg/g、実施例6A:18000μg/g、実施例10A:1400μg/g、実施例10B:4000μg/gであった。
丸底96ウェルプレートの各ウェルに生理食塩水50μLを加え、縦1列目に評価サンプル(すなわち、上記で得られた溶出画分および参照としての不活化ウイルス含有液)50μLを加えてよく混合した。この2倍希釈されたサンプルのうち50μLを横隣のウェルに加えよく混合した。同様の操作を繰り返し、12番目のウェルまで2~4096倍の希釈サンプルを調製した。各サンプルに0.5%鶏赤血球浮遊液50μmLを加えて混合した後、室温で30分間放置した。30分後、血球が凝集して底に沈殿していないものをウイルス活性有りとし、活性が認められなくなる一つ前までの希釈倍率をサンプル50μLあたりのHA価(HAU/50μL)とした。また、ネガティブコントロールには生理食塩水を用いた。
例えば、多孔性セルロース粒子が硫酸基またはスルホン酸基含有基によって修飾され、且つ修飾された多孔性セルロース粒子に対するS含量(すなわち、修飾された多孔性セルロース粒子1gに対する硫黄含量)が800~5000μg/gであることが好ましく、800~4000μg/gであることがより好ましく、800~2000μg/gであることが特に好ましい。多孔性セルロース粒子に対するS含量が700μg/g以下の場合には、十分なウイルス吸着量を得られないことが実証されている。従って、S含量が上記範囲となるように多孔性セルロース粒子を修飾することにより、十分なウイルス吸着量を得られる。
(ウイルス含有液の調製)
B型肝炎ウイルスHBsAg-XT(Beacle Inc.)を、150μg/mlとなるよう、以下に示すクロマトグラフィーカラムの平衡化に用いる緩衝液に懸濁した。
(ウイルス吸着能の評価)
実施例14で調製した硫酸化セルロース粒子(実施例10Aおよび10B)を使用した。実施例10Aの硫酸化セルロース粒子(S含量:1400μg/g)、および実施例10Bの硫酸化セルロース粒子(S含量:4000μg/g)の各々を水に分散させ、減圧下で攪拌しながら脱気した。脱気した各々の分散液を、ガラスカラム(Φ3×50mm)に充填した。このカラムをクロマトグラフィーシステムに接続し、それぞれ0.02Mリン酸緩衝液(pH7.0)および0.05Mクエン酸緩衝液(pH5.0)で平衡化した。平衡化したカラムに、上記で調製したウイルス含有液を2ml通液した。通液は0.25ml/minの流速で行い、流出液は1mLずつ回収した。ウイルス含有液の通液後は、平衡化で用いた緩衝液を通液して非吸着分を洗浄した。続いて塩化ナトリウム水溶液の濃度を1.5Mまで直線勾配で上げて通液し、溶出画分を回収した(吸着分)。各フラクションは1mLずつ回収し、BCAアッセイ法で各フラクションにおけるタンパク質濃度を測定することで、ウイルスの回収量(すなわち吸着量)を求めた。なおこのBCAアッセイは、Thermo社マニュアルに従い、BSAを標準として行った。
Claims (16)
- (a)二酢酸セルロースを溶媒に溶解して、二酢酸セルロース溶液を調製することと、
(b)前記二酢酸セルロース溶液を、該二酢酸セルロース溶液と混和しない媒質中に分散させて、分散系を得ることと、
(c)前記分散系を冷却することと、
(d)冷却された前記分散系に貧溶媒を添加することにより、二酢酸セルロース粒子を析出させることと、
(e)前記二酢酸セルロース粒子を鹸化することと
を含む、多孔性セルロース粒子の製造方法。 - (a)において、前記二酢酸セルロース溶液の溶媒は、酢酸水溶液またはシクロヘキサノンである請求項1に記載の多孔性セルロース粒子の製造方法。
- (a)において、前記二酢酸セルロース溶液の溶媒は酢酸水溶液であり、前記酢酸水溶液における酢酸の含量は、前記酢酸水溶液に対して80~95重量%である請求項2に記載の多孔性セルロース粒子の製造方法。
- (d)において、前記貧溶媒は、水、アルコール類、グリコール類またはこれらの混合液である、請求項1~3のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- (b)において、前記二酢酸セルロース溶液と混和しない媒質は、水または有機媒質である請求項1~4のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- (b)において、前記二酢酸セルロース溶液と混和しない媒質は有機媒質であり、前記有機媒質は、トルエンまたはo-ジクロロベンゼンである請求項5に記載の多孔性セルロース粒子の製造方法。
- (a)において、前記二酢酸セルロースは、酢化度が45~57%である請求項1~6のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- (a)において、前記二酢酸セルロース溶液における二酢酸セルロースの含量は、前記二酢酸セルロース溶液に対して3~20重量%である請求項1~7のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- (a)において、前記二酢酸セルロースは、25℃~100℃の温度で前記溶媒に溶解される、請求項1~8のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- (a)において、前記二酢酸セルロースは、40℃~100℃の温度で前記溶媒に溶解される、請求項9に記載の多孔性セルロース粒子の製造方法。
- (c)において、前記冷却は、0~40℃の温度に冷却することにより行う請求項1~10のいずれか1項に記載の多孔性セルロース粒子の製造方法。
- 請求項1~11のいずれか1項に記載の多孔性セルロース粒子の製造方法により製造され得る多孔性セルロース粒子。
- (a)における前記二酢酸セルロース溶液中の二酢酸セルロースの含量は、前記二酢酸セルロース溶液に対して6~12重量%であり、(c)における前記冷却は、0~30℃の温度に冷却することにより行われ、分子量8000のPEGを用いて測定したKav値が0.01以上0.52以下であり、かつ分子量12000のPEGを用いて測定したKav値が0.001以上0.45以下である請求項12に記載の多孔性セルロース粒子。
- 請求項12または13に記載の多孔性セルロース粒子または修飾された前記多孔性セルロース粒子を含む、クロマトグラフィー用充填剤。
- ウイルス粒子を分離精製するために使用される、請求項14に記載のクロマトグラフィー用充填剤。
- 硫酸基またはスルホン酸基含有基によって修飾され、且つ修飾された多孔性セルロース粒子に対するS含量が800~5000μg/gである修飾された多孔性セルロース粒子を含む、インフルエンザウイルス粒子またはB型肝炎ウイルス粒子の分離精製用の請求項15に記載のクロマトグラフィー用充填剤。
