Description
本発明は、多孔質粒子とその製造方法、およびこれを用いたクロマトグラフィー用充填剤に関する。
バイオ医薬品の分離・精製に用いるクロマトグラフィー用の材料として、三次元ネットワーク状の骨格とその空隙からなる連通孔を持つ一体成型体であるモノリスが注目されている。モノリスを用いることで、従来のクロマトグラフィーよりも高処理量、高処理速度の分離・精製が可能となることが知られている(特許文献1)。
近年では、モノリスを粒子にした、連通孔を有する粒子が提案されている。粒子にすることの利点として、従来のクロマトグラフィー用分離カラムの充填剤と同様に使用できて取り扱いやすいこと、スケールアップがしやすいことがある。また、連通孔を有する粒子は生体高分子などの分離・精製の際に、粒子内部の連通孔の空隙も吸着サイトとして利用でき、高処理量、高処理速度の分離・精製が可能となる材料となる。連通孔を有する粒子の製造方法においては、例えば、モノリスを粉砕することで粒子状にする方法(特許文献2)と、連通孔を有する粒子を直接的に造粒する方法(特許文献3、4、5)とがある。

モノリスを粉砕して粒子にする方法では、粉砕で得られる粒子がいびつな形となるため、クロマトグラフィー用分離カラムへの均一な充填ができず、期待される性能が発揮されないという問題がある。また、球状粒子を直接造粒する方法では、粒子表面の連通孔形成が困難で塞がれてしまったり、粒子毎の連通孔の孔径が不均一であったり、連通孔の孔径の制御が難しいという問題がある。
そこで、本発明の目的は、制御された三次元ネットワーク状の骨格とその空隙からなる連通孔を有しており、かつ前記連通孔が粒子表面から内部まで貫通している多孔質粒子とその製造方法、およびこれを用いたクロマトグラフィー用充填剤を提供することである。
本発明者らは、上記の課題を解決するために鋭意研究した結果、制御された三次元ネットワーク状の骨格とその空隙からなる連通孔を有しており、かつ前記連通孔が粒子表面から内部まで貫通している多孔質粒子とその製造方法、およびこれを用いたクロマトグラフィー用充填剤を見出し、本発明を完成させた。
すなわち、本発明は以下の通りである。
[1]球状で、三次元ネットワーク状の骨格とその空隙からなる連通孔構造を有しており、かつ前記連通孔が粒子表面から内部にかけて貫通しており、内径0.78cm×長さ30cmのカラムに充填し流量0.4ml/min のときの分子量200万のデキストランの保持時間が20分以上であることを特徴とする、主成分が酢酸セルロースまたは、セルロースである多孔質粒子。

本発明に係る多孔質粒子は制御された三次元ネットワーク状の骨格とその空隙からなる連通孔を有しており、かつ前記連通孔が粒子表面から内部まで貫通している。そのため、生体高分子などの分離・精製の際に、粒子内部の連通孔の空隙も吸着サイトとして利用でき、高処理量、高処理速度が達成できる有用な材料となる。
本発明の多孔質粒子の形状は球状である。ここでいう球状とは、例えば、長径(最も長い径)が、短径(最も短い径)に対し、2倍以下である。本発明の多孔質粒子は、真球形で、長径と短径とが同じ長さに近いことが好ましい。
本発明の多孔質粒子は、三次元ネットワーク状の骨格とその空隙からなる連通孔構造を有しており、かつ前記連通孔が粒子表面から内部にかけて貫通している。三次元ネットワーク状の骨格と連通孔を有しているかどうかは、例えば、前記粒子の断面の共焦点レーザー顕微鏡観察、SEM観察の測定結果から判断することができる。また、前記連通孔が粒子表面から内部にかけて貫通していることは、例えば、本発明の多孔質粒子の表面の写真により確認できる。本発明の多孔質粒子は、前記連通孔が粒子表面において塞がれることなく、粒子の表面において開口している。
本発明の多孔質粒子は、以下の特性を満たす。内径0.78cm×長さ30cmのカラムに充填し流量0.4ml/min のときの分子量200万のデキストランの保持時間が20分以上である。粒子内部の空隙を吸着サイトとして利用するという観点からは、分子量200万のデキストランの保持時間は、25分以上であることが好ましく、27分以上であることがより好ましい。カラムの充填の方法は、粒子を純水で分散させてスラリーにした後、カラムに充填し、さらに純水を流量0.4ml/minで1時間以上流して、圧密化させるものとする。また、ここでいう保持時間とは、試料をカラムに注入してから、溶出ピークまでにかかる時間である。溶出ピークは例えば、HPLC装置(Agilent Technologies製、1260 Infinity)を用いることにより、RI検出強度が最大になる値から求めることができる。本発明における保持時間の測定は、移動相には純水を用い、測定温度は25℃にて行うものとする。
クロマトグラフィーの分野においては、多孔質粒子の細孔を測定する場合、分子量200万のデキストランを、排除限界分子量として測定を行うことが一般的である(特許文献6)。このことは、分子量200万のデキストランが多孔質粒子内部に入り込むことができないことが前提になっている。本発明の多孔質粒子は、従来の粒子と比較して分子量200万のデキストランの保持時間が長い。このことは、本発明の多孔質粒子が、分子量200万のデキストランが粒子内部に入り込める連通孔を有していることを示す。
一般に球状の多孔質粒子をカラムに充填した場合、空隙率は30~40%となることが知られている(非特許文献1)。ここでいう空隙率とは、カラム内部における粒子間の隙間の全体積をカラム全体の体積で割ったものである。例えば、試料が内径0.78cm×長さ30cmのカラム内部の充填された粒子間の隙間のみを通ってくると仮定した場合、流量0.4ml/min のときの保持時間は、理論的には、10.7~14.3分間となる。本発明の多孔質粒子の分子量200万のデキストランの保持時間が20分間以上であることは、粒子間の隙間だけでなく、粒子内部に分子量200万のデキストランが入り込むことを示している。
本発明の多孔質粒子は、より好ましくは以下の特性を満たす。内径0.78cm×長さ30cmのカラムに充填し流量0.4ml/min のときの粒径30nmのシリカ製ナノ粒子の保持時間が16分以上である。粒子内部の空隙を吸着サイトとして利用するという観点からは、粒径30nmのシリカ製ナノ粒子の保持時間は、18分以上であることがより好ましい。カラムの充填の方法、保持時間、測定の温度に関しては、前記の通りである。本発明の粒子は、従来の粒子と比較して粒径30nmのシリカ製ナノ粒子の保持時間が長い。このことから、本発明の粒子は、粒径30nmのシリカ製ナノ粒子が粒子内部に入り込める連通孔を有している。
本発明の多孔質粒子の主成分は、酢酸セルロースまたはセルロースである。ここでいう主成分とは、多孔質粒子中の含有量が50質量%以上である成分を意味する。本発明において使用される酢酸セルロースは、一般的に酢酸セルロースと定義され得るものであれば特に限定されないが、酢化度が45~57%であることが好ましい。
本発明の多孔質粒子は、モード径が1~1000μmである。クロマトグラフィー用充填剤として利用できるという観点からは、多孔質粒子のモード径は、35μm~200μmであることが特に好ましい。しかしながら、これらに限定されるものではない。なお、ここでいうモード径は、粒度分布測定装置:HORIBA製 Laser Scattering Particle Size distribution Analyzer Partica LA-950を使用してモード径を測定することにより算出した。
本発明の多孔質粒子のBET多点法による比表面積は1~200m2/gである。BET多点法による比表面積は、例えば、高速比表面積/細孔分布測定装置(マイクロトラックベル製BELSORP MAXII)を用いることにより測定することができる。例えば、本発明の粒子は、一昼夜凍結乾燥したのち、100℃、2時間、真空脱気して前処理をして、測定することができる。比表面積は、測定温度が77.3K、相対圧力が0.05から0.30の3点以上の圧力範囲において、固体表面に吸着した窒素ガス量の測定から得た吸着等温線(直線)から求めることができる。
以下、本発明における多孔質粒子の製造方法は、(a)酢酸セルロースを酢酸セルロースが可溶な溶媒と酢酸セルロースが不溶な溶媒との混合溶媒に加熱溶解して、酢酸セルロース溶液を調製する工程と、(b)前記酢酸セルロース溶液を、乳化安定剤を含む水中に分散させて、分散系を得る工程と、(c)前記分散系を冷却し、酢酸セルロース粒子を析出させる工程を含み、前記混合溶媒が水と混和しない有機溶媒である、多孔質粒子の製造方法である。
本発明の製造方法によれば、球状で、三次元ネットワーク状の骨格とその空隙からなる連通孔構造を有しており、かつ前記連通孔が粒子表面から内部にかけて貫通している、主成分が酢酸セルロースの多孔質粒子を好ましく製造することができる。本発明の製造方法は、得られる多孔質粒子の粒径および連通孔の孔径を容易に制御することができる。例えば、使用する酢酸セルロース原料の量、使用する良溶媒および貧溶媒の種類、比率、第三成分である高分子の分子量や濃度、界面活性剤の種類および添加量、冷却速度等によって制御することができる。前述した本発明の多孔質粒子は、本発明の製造方法において前記制御を行うことにより、好ましく製造することができる。
以下、上記各工程について順に説明する。

[(a)の工程]
(a)においては、原料の酢酸セルロースを混合溶媒に溶解して、酢酸セルロース溶液を調製する。酢酸セルロースは、天然の高分子であるセルロースを酢酸エステル化することにより得られる半合成高分子である。本発明において使用される酢酸セルロースは、一般的に酢酸セルロースと定義され得るものであれば特に限定されないが、重合度50~300、かつ酢化度が45~57%であることが好ましい。重合度50~300、かつ酢化度が45~57%である酢酸セルロースを使用することにより、より多くの種類の溶剤に溶解させることができる。
(a)においては、原料の酢酸セルロースを混合溶媒に溶解して、酢酸セルロース溶液を調製する。酢酸セルロースは、天然の高分子であるセルロースを酢酸エステル化することにより得られる半合成高分子である。本発明において使用される酢酸セルロースは、一般的に酢酸セルロースと定義され得るものであれば特に限定されないが、重合度50~300、かつ酢化度が45~57%であることが好ましい。重合度50~300、かつ酢化度が45~57%である酢酸セルロースを使用することにより、より多くの種類の溶剤に溶解させることができる。
(a)において、前記酢酸セルロース溶液が可溶な溶媒、すなわち良溶媒は、酢酸セルロースを溶解できるものであれば特に限定されないが、水への溶解度の低い有機溶媒が好ましい。ここでいう良溶媒とは、溶質が溶媒単独に溶解し、固形物を含まない透明な溶液が得られるものをいう。特にここでは、溶媒の沸点以下の温度において好ましくは1重量%以上の溶液が得られる溶媒のことをいう。溶媒は、1種類を単独で使用しても、2種類以上の溶媒を混合して使用してもよい。具体的にはベンジルアルコール、酢酸エチル、シクロヘキサノン、イソホロン;およびこれらの混合物を挙げることができる。中でも、連通孔を空けるという点において、ベンジルアルコールが好ましい。
(a)において、前記酢酸セルロース溶液が不溶な溶媒、すなわち貧溶媒は、低級アルコール類、グリコール類、エーテル類、エステル類;およびこれらの混合物を挙げることができる。貧溶媒とは良溶媒と比較して溶質に対して溶解性がないか又は溶解性の低い溶媒のことをいい、具体的には溶媒の沸点以下の温度において溶媒単独で溶質を溶解できず、透明な溶液が得られないものをいう。特にここでは好ましくは溶媒の沸点以下の温度において溶質を1重量%以上溶解できない溶媒のことをいう。水への溶解度の低い溶媒が好ましい。特に、低級アルコール類であることが好ましく、さらに連通孔を空けるという点において、1-ヘキサノールであることが好ましい。
(a)においては、混合溶媒における酢酸セルロースが可溶な溶媒すなわち良溶媒と酢酸セルロースが不溶な溶媒、すなわち貧溶媒の混合割合は良溶媒が5~95v/v%、貧溶媒が95~5v/v%が好ましい。混合溶媒に含まれる貧溶媒の比率が高いほど、連通孔の孔径は大きくなる傾向にある。
(a)において、混合溶媒中の酢酸セルロースの割合は0.1~30重量%が好ましく、5~20重量%がさらに好ましい。この範囲であれば、均一な溶液が得られ、きれいな連通孔を有する多孔質粒子が得られやすい。また、冷却した時に相分離による析出が生じやすく強度が十分なものとなり、粒子形状を維持しやすくなる。また酢酸セルロース溶液における酢酸セルロース濃度が低いほど、得られる多孔質粒子の粒径は小さく、連通孔の孔径は大きくなる傾向にある。
(a)において、混合溶媒は水と混和しない有機溶媒である。水と混和してしまうと、(b)の工程において、分散系を得ることができない。
(a)においては、混合溶媒に高分子である第三成分を含有させることができる。第三成分の高分子としては、ポリエチレングリコール、またはポリプロピレングリコールが好ましく、ポリプロピレングリコールがより好ましい。
(a)において、第三の成分がポリプロピレングリコールである場合、その分子量は、特に限定されないが、製造工程中の取扱い性と多孔質粒子が有する連通孔の孔径の観点から、100~3000の平均分子量(Mw)であることが好ましい。また第三成分の高分子の分子量が高いほど、連通孔の孔径は大きくなる傾向にある。
(a)において、多孔質粒子が有する連通孔の孔径の観点から、混合溶媒中の第三の成分の濃度は0.1~10重量%であることが好ましく、第三成分の濃度が高いほど、連通孔の孔径は大きくなる傾向にある。
(a)においては、加熱溶解において、加熱温度は、例えば、50~130℃の範囲が好ましい。この温度範囲において、酢酸セルロースが混合溶媒に溶解することが好ましい。この加熱溶解の時間は特に限定されず、例えば、加熱時間は3~24時間とすることができる。
[(b)の工程]
(b)においては、前記酢酸セルロース溶液を乳化安定剤を溶解した高温水中に、分散させる。この工程における、酢酸セルロース溶液と高温水との体積比は、酢酸セルロース溶液を分散相、高温水を連続相とする分散系が得られる範囲であれば、特に限定はされない。安定な分散系を得る観点からは、酢酸セルロース溶液と高温水との体積比(分散相/連続相)は1.0以下であることが、好ましい。また高温水の温度は、50~100℃であることが好ましく、60~95℃であることがより好ましい。

(b)において、分散させる方法に関しては、公知の方法を任意に適用することができる。例えば、攪拌機などのミキサーを用いる方法、ホモジナイザーを用いる方法、超音波を用いる方法などがある。また、一般的にマイクロリアクターと称されるものを用いて、細いノズルから酢酸セルロース溶液を押し出して液滴を得る方法や、均一な細孔径の多孔質膜から酢酸セルロース溶液、もしくは酢酸セルロース溶液と水と乳化安定剤の混合物を押し出してせん断をかける方法などがある。本発明においては、最も簡便な攪拌機により分散させる方法を検討した。
(b)において、乳化安定剤としては、前記分散系の安定性を高め、粒子の凝集を防ぐ効果があるものであれば、特に限定されない。例えば、でんぷん、ペクチン、アルギン酸、アルギン酸塩、ゼラチンなどの天然高分子、メチルセルロース、カルボキシメチルセルロース、ヒドロキシエチルセルロースなどの天然高分子加工物、ポリビニルアルコール、ポリ酢酸ビニルの部分ケン化物などのアルコール性OH含有合成高分子、スルホン化スチロールなどのSO3H基含有高分子、アクリル酸エステルなどのCOOH基含有高分子、ポリビニルピロリドンなどの含窒素合成高分子、硫酸バリウムやタルク、ベントナイト、酸化チタンなどの無機物質粉末、直鎖アルキルベンゼンスルホン酸ナトリウムなどのアニオン界面活性剤、アルキルトリメチルアンモニウム塩などのカチオン界面活性剤、アルキルジメチルアミンオキシドなどの両性界面活性剤、脂肪酸ソルビタンエステルなどのノニオン界面活性剤、またはこれらの混合物などを挙げることができる。特に、分散系を安定させるという観点から、ドデシルベンゼンスルホン酸ナトリウム、ポリビニルアルコール、カルボキシメチルセルロース、ヒドロキシエチルセルロース、またはこれらの混合物であることが好ましい。
(b)において使用する乳化安定剤の添加量は、分散系を安定させるという観点から、高温水における濃度として0.01重量%以上が好ましい。かかる濃度範囲において、乳化安定剤の添加量を多くすると、得られる多孔質粒子の粒径は小さくなる傾向にある。反対に、乳化安定剤の添加量が少ないと、得られる多孔質粒子の粒径は大きくなる傾向にある。
[(c)の工程]
(c)においては、上記(b)で得られた分散系を冷却する。冷却することにより熱誘起相分離が起こり、連通孔を有する多孔質粒子を得ることができる。

(c)において、分散系の冷却速度は、0.1℃/min~10℃/minであることが好ましい。(c)における冷却速度が遅いほど、連通孔の孔径は大きくなる傾向にある。反対に、冷却速度が速い場合は、連通孔の孔径は小さくなる傾向にある。前述の本発明の多孔質粒子を製造する場合は、冷却速度が0.1℃/min~5℃/minであることが好ましく、さらに0.1℃/min~2℃/minであることがより好ましい。
(c)において、分散系を冷却する温度は、酢酸セルロースが析出する温度であれば特に限定されない。しかし、多孔質粒子が析出しきるという点で、冷却する温度は0℃~50℃であることが好ましい。
[多孔質粒子または架橋、修飾された多孔質粒子]
本発明の1つの実施形態によると、上記製造方法により製造され得る多孔質粒子が提供される。この多孔質粒子は、各種物質の分離精製において使用することができる。さらに、ケン化反応したり、架橋反応したり、置換基により修飾したりした形態で適用してもよい。

多孔質粒子をケン化、架橋することで、クロマトグラフィー用充填剤としての使用に耐えうる機械的強度、耐流速性を付与することができる。 多孔質粒子の架橋処理は、特開2009―242770号などを参照して行うことができる。実験条件を適宜設計変更することにより、目的の機械的強度、耐流速性を付与することができる。
また、本発明の多孔質粒子の反応性官能基の少なくとも一部にリガンドを付加することによって、種々の物質を吸着可能な吸着剤を容易に得ることができる。例えば、インフルエンザウイルスや肝炎ウイルス、狂犬病ウイルス、ヒトパピローマウイルス、アデノ随伴ウイルスなどに対するウイルス吸着体、抗体医薬品精製用吸着体、エクソソーム吸着体、核酸吸着体等において利用され得る。具体的には、本発明の多孔質粒子をケン化、架橋した後、硫酸化多糖を導入することにより、ウイルス粒子の分離または精製に好適なクロマトグラフィー用充填剤を提供することができる。硫酸化多糖の導入に関しては、特開2011-220992号公報などを参照して行うことができる。
その他のリガンドとしては、特に限定されないが、2-ジエチルアミノエチル(DEAE)、カルボキシメチル(CM)、スルホン、4級アンモニウム(Q)等のイオン交換基やフェニル、ブチルなどの疎水性基、イオン交換と疎水基を併せ持ついわゆるミックスモードの分離に使えるリガンド、プロテインA、抗体等のタンパク吸着用リガンドやポリリジン等のポリカチオン、ヘパリンやポリグルタミン酸等のポリアニオンなどの機能性高分子等が挙げられ、多孔質粒子の使用目的に応じて任意に選択でき、その導入は、公知の方法に従って任意に行うことができる。

本発明のクロマトグラフィー用充填剤のインフルエンザウイルスに対する吸着性能は、例えば、特開2011-220992号公報に記載の方法で評価できる。評価に用いるインフルエンザウイルスは、A、B型、発育鶏卵由来、MDCK細胞などの培養細胞由来のインフルエンザウイルスが利用できる。
以下、実施例により本発明をより具体的に説明するが、本発明はこれら実施例の態様に

[実施例1]
酢酸セルロース14g(L-20、ダイセル製)をベンジルアルコール52.5ml、1-ヘキサノール47.5ml、ポリプロピレングリコール (和光純薬試薬、ジオール型、平均分子量1,000)2mlの混合溶媒に加えて攪拌した。さらに、昇温して120℃で4時間撹拌することにより酢酸セルロースを溶解し、透明な酢酸セルロース溶液を得た。また、PVA(JP-18E、日本酢ビ・ポバール(株)社製)60mg、カルボキシメチルセルロースナトリウム(和光純薬試薬)4.5gを混合溶媒で飽和した純水300mlに加え、昇温して80℃で1時間以上攪拌、溶解することで、分散媒を得た。分散媒300mlに前記酢酸セルロース溶液100gを素早く注ぎ、80℃、回転数250rpmにて15分間攪拌して分散系を得た。続いてこの分散系を20℃室温中で放置し40℃まで冷却した。この際、冷却速度は、平均して0.5℃/minであった。40℃になったところで、酢酸セルロースが析出し、球状の酢酸セルロース粒子が得られた。その後、得られた酢酸セルロース粒子を大量の水、次いでメタノールで十分に洗浄した。水で再び洗浄後、得られた酢酸セルロース球状粒子を、目開き250μmと目開き45μmの篩に通し、粒径45~250μmの多孔質粒子を得た。

以下に、本発明の多孔質粒子の好ましい使用形態の一つである架橋した多孔質粒子の製造例を示す。
[製造例1]
実施例1により得られた多孔質粒子55g-wet(含水率6.31)を124gの純水に32gのNa2SO4を溶解した液に加え、温度を40℃にして30分攪拌した。次に、2.93mol当量のNaOHを加え、40℃で2時間攪拌して反応させることで、ケン化した。反応後、50℃に温度を上げて、攪拌を継続しながら、0.9gの48%NaOH溶液と、0.5gのNaBH4を加え、30分かけて溶解、反応させた。溶解後、22gの48%NaOH溶液と30gのエピクロロヒドリンをそれぞれ4等分した量を、60分おきにおよそ3時間かけて添加した。添加終了後、温度50℃で16時間反応させた。温度を40℃以下に冷却し、酢酸5.8gを加えて中和した。反応混合物をろ過してゲルを回収し、純水でろ過洗浄し、目的の架橋した多孔質粒子を得た。
[製造例1]
実施例1により得られた多孔質粒子55g-wet(含水率6.31)を124gの純水に32gのNa2SO4を溶解した液に加え、温度を40℃にして30分攪拌した。次に、2.93mol当量のNaOHを加え、40℃で2時間攪拌して反応させることで、ケン化した。反応後、50℃に温度を上げて、攪拌を継続しながら、0.9gの48%NaOH溶液と、0.5gのNaBH4を加え、30分かけて溶解、反応させた。溶解後、22gの48%NaOH溶液と30gのエピクロロヒドリンをそれぞれ4等分した量を、60分おきにおよそ3時間かけて添加した。添加終了後、温度50℃で16時間反応させた。温度を40℃以下に冷却し、酢酸5.8gを加えて中和した。反応混合物をろ過してゲルを回収し、純水でろ過洗浄し、目的の架橋した多孔質粒子を得た。

得られた実施例2~4の粒子を製造例1と同様にして架橋工程を行い、製造例2~4の架橋した多孔質粒子を得た。

[粒子の評価]
保持時間の測定
上記製造例1~4で得られた粒子について、内径0.78cm、長さ30cmのステンレス製カラム(東ソー製)に充填した後、各試料の保持時間を測定した。粒子の比較として、市販GCL-2000HF(JNC製)を用いた。充填方法は、粒子を純水で分散させてスラリーにした後、カラムに充填し、さらに純水を流量0.4ml/minで1時間以上流して、圧密化させた。試料としては、以下の表1のものを用いた。測定する際の移動相には、純水を用いた。RI検出強度が最大になる値を溶出ピークとし、その時間を保持時間とした。測定に使用した装置は、以下の通りである。

上記装置を使用して保持時間を測定した。
結果を図1および表2に示す。

測定の結果、製造例1と製造例2の粒子は分子量200万のデキストランの保持時間が20分以上であった。一方、製造例3と製造例4の粒子およびGCL-2000HFは20分未満であり、連通孔の孔径が小さいことが分かった(表2)。その他の高分子試料を用いた場合も、製造例1と製造例2の粒子は製造例3と製造例4の粒子およびGCL-2000HFよりも試料の保持時間が長かった(図1)。
製造例1の粒径30nmのシリカ製ナノ粒子の保持時間は、16分以上であった。一方、製造例2~4、およびGCL―2000HFの粒径30nmのシリカ製ナノ粒子の保持時間は、16分未満であった(表2)。
2.モード径の測定
上記製造例1~4で得られた粒子について、粒度分布を測定し、モード径を求めた。測定に使用した装置は、以下の通りである。

3.BET比表面積の測定
上記製造例1~4で得られた粒子について、BET比表面積を測定した。BET比表面積は、高速比表面積/細孔分布測定装置(マイクロトラックベル社製BELSORP MAXII)を用いた。凍結乾燥した製造例1~4の粒子を、さらに100℃、2時間(真空脱気)で脱気して前処理をして試料とした。測定温度が77.3K、相対圧力が0.05から0.30の5点以上の圧力範囲において固体表面に吸着した窒素ガス量の測定から得た吸着等温線(直線)から求めた。
評価結果を表3に示す。
上記製造例1~4で得られた粒子について、BET比表面積を測定した。BET比表面積は、高速比表面積/細孔分布測定装置(マイクロトラックベル社製BELSORP MAXII)を用いた。凍結乾燥した製造例1~4の粒子を、さらに100℃、2時間(真空脱気)で脱気して前処理をして試料とした。測定温度が77.3K、相対圧力が0.05から0.30の5点以上の圧力範囲において固体表面に吸着した窒素ガス量の測定から得た吸着等温線(直線)から求めた。

4.SEM観察
日立ハイテクノロジーズ製の超高分解能電界放出形走査電子顕微鏡「SU8020」を使用して、SEM観察を行った。前処理として凍結乾燥した製造例1~4の粒子に、撮影のため、Auコーティングを行った。電子顕微鏡写真では、得られた粒子がほぼ真球状であり、表面および断面に連通孔があることが認められる(図2~7)。
日立ハイテクノロジーズ製の超高分解能電界放出形走査電子顕微鏡「SU8020」を使用して、SEM観察を行った。前処理として凍結乾燥した製造例1~4の粒子に、撮影のため、Auコーティングを行った。電子顕微鏡写真では、得られた粒子がほぼ真球状であり、表面および断面に連通孔があることが認められる(図2~7)。
以下に、本発明の多孔質粒子の好ましい使用形態の一つであるクロマトグラフィー用充填剤に関する製造例を示す。

1)架橋した多孔質粒子のエポキシ化
製造例1より得られた架橋した多孔質粒子20g-wet(含水率6.25)を24mlの純水とともに撹拌機付き反応器に仕込んだ。よく撹拌を行いながら内部温度を30℃に調整し、エピクロロヒドリン15.6gを添加した。15分撹拌後、48%NaOH水溶液14.6gを添加して2時間反応を行った。反応終了後、反応液を濾過し、濾液が中性になるまで水で十分に洗浄した。30分間吸引乾燥を行い、エポキシ活性化湿潤ゲル20g-wetを得た。エポキシ活性化湿潤ゲル1.0gを1.3Mチオ硫酸ナトリウム溶液3.0mlとともに30℃に温度設定した振騰機で1時間振騰し、0.1mol・L塩酸で滴定することによりエポキシ量を定量した。このエポキシ基量は230μmol/g-dryであった。
[製造例5](多孔質粒子と硫酸化多糖の結合)
1)架橋した多孔質粒子のエポキシ化
製造例1より得られた架橋した多孔質粒子20g-wet(含水率6.25)を24mlの純水とともに撹拌機付き反応器に仕込んだ。よく撹拌を行いながら内部温度を30℃に調整し、エピクロロヒドリン15.6gを添加した。15分撹拌後、48%NaOH水溶液14.6gを添加して2時間反応を行った。反応終了後、反応液を濾過し、濾液が中性になるまで水で十分に洗浄した。30分間吸引乾燥を行い、エポキシ活性化湿潤ゲル20g-wetを得た。エポキシ活性化湿潤ゲル1.0gを1.3Mチオ硫酸ナトリウム溶液3.0mlとともに30℃に温度設定した振騰機で1時間振騰し、0.1mol・L塩酸で滴定することによりエポキシ量を定量した。このエポキシ基量は230μmol/g-dryであった。
2)硫酸化多糖の結合
デキストラン硫酸ナトリウムDS-500(名糖産業株式会社製)1.63gを、18mlの純水とともに撹拌機付き反応器に仕込み、デキストラン硫酸ナトリウムが完全に溶解するまで撹拌した後、1)で製造したエポキシ活性化湿潤ゲル15g(含水率5.78、エポキシ基量230μmol/g-dry)を、反応器内に加えた。良く撹拌しながら内部温度を30℃に調整し、15分撹拌後、Na2SO48.4g、Na2HPO413.5g、48%NaOH水溶液1.3gを添加した。40℃に温度を上げて、6時間反応を行った。反応終了後、純水9.7mlに48%NaOH水溶液0.9mlを添加した水溶液にNaBH40.2gを溶解させた溶液を加え、さらに16時間反応させた。反応後、反応液を濾過し、濾液が中性になるまで水で十分に洗浄した。30分間吸引乾燥を行い、デキストラン硫酸化された多孔質粒子15g-wetを得た。
デキストラン硫酸ナトリウムDS-500(名糖産業株式会社製)1.63gを、18mlの純水とともに撹拌機付き反応器に仕込み、デキストラン硫酸ナトリウムが完全に溶解するまで撹拌した後、1)で製造したエポキシ活性化湿潤ゲル15g(含水率5.78、エポキシ基量230μmol/g-dry)を、反応器内に加えた。良く撹拌しながら内部温度を30℃に調整し、15分撹拌後、Na2SO48.4g、Na2HPO413.5g、48%NaOH水溶液1.3gを添加した。40℃に温度を上げて、6時間反応を行った。反応終了後、純水9.7mlに48%NaOH水溶液0.9mlを添加した水溶液にNaBH40.2gを溶解させた溶液を加え、さらに16時間反応させた。反応後、反応液を濾過し、濾液が中性になるまで水で十分に洗浄した。30分間吸引乾燥を行い、デキストラン硫酸化された多孔質粒子15g-wetを得た。
[製造例6]
製造例4より得られた架橋した多孔質粒子を製造例5と同様に硫酸化多糖を結合させ、製造例6とした。
製造例4より得られた架橋した多孔質粒子を製造例5と同様に硫酸化多糖を結合させ、製造例6とした。
3)ウイルス評価試験
(1)市販セルファインサルフェイトと製造例5のクロマトグラフィー用充填剤との比較
充填剤として、市販セルファインサルフェイト(JNC製)と製造例5、製造例6のクロマトグラフィー用充填剤を用いた。比較のために用いた市販セルファインサルフェイトは表面にリガンドとして硫酸化多糖を持つ真密セルロース粒子である。
各充填剤を水に分散させ減圧下で攪拌しながら脱気した。脱気した後、φ5×30mm
のカラムに各クロマトグラフィー用充填剤を充填し、カラムをAKTA avant (Cytiva)に取り付け、カラム容量の10倍以上の0.01Mリン酸バッファー、0.12M NaCl、pH7.4を流し平衡化した。その後、セルロースアセテート製の0.45μmメンブレンフィルター(製品名 GD/X シリンジフィルター)でろ過したH1N1株の試験ウイルス液30ml(使用時の総HA価:76800)のカラムへの通液を開始すると共に、カラムからの流出液を1.5mlずつ回収した。上記試験ウイルス液の通液終了後、0.01Mリン酸バッファー、0.12M NaCl、pH7.4の通液を開始し、非吸着画分を洗い流した。非吸着画分の洗浄が終了した後、0.01Mリン酸バッファー、2M NaCl,pH7.4の通液を開始し、カラムに吸着しているウイルスが完全に溶出されるまで通液を実施した。溶出終了後、回収した流出液の1.5ml画分の50μLを使用し、HA価を測定した。さらに、試験ウイルス液50μLのHA価の1/10に到達する直前の画分までの総液量(μL)を求めた。本測定系に於ける10%DBCは以下のようにして求めた。
10%DBC(HA価/ml-gel)
=(試験ウイルス液50μLのHA価の1/10に到達する直前の画分までの総液量× 1/50×試験ウイルス液50μLのHA価)/カラム容量(ml-gel)
(1)市販セルファインサルフェイトと製造例5のクロマトグラフィー用充填剤との比較
充填剤として、市販セルファインサルフェイト(JNC製)と製造例5、製造例6のクロマトグラフィー用充填剤を用いた。比較のために用いた市販セルファインサルフェイトは表面にリガンドとして硫酸化多糖を持つ真密セルロース粒子である。
各充填剤を水に分散させ減圧下で攪拌しながら脱気した。脱気した後、φ5×30mm
のカラムに各クロマトグラフィー用充填剤を充填し、カラムをAKTA avant (Cytiva)に取り付け、カラム容量の10倍以上の0.01Mリン酸バッファー、0.12M NaCl、pH7.4を流し平衡化した。その後、セルロースアセテート製の0.45μmメンブレンフィルター(製品名 GD/X シリンジフィルター)でろ過したH1N1株の試験ウイルス液30ml(使用時の総HA価:76800)のカラムへの通液を開始すると共に、カラムからの流出液を1.5mlずつ回収した。上記試験ウイルス液の通液終了後、0.01Mリン酸バッファー、0.12M NaCl、pH7.4の通液を開始し、非吸着画分を洗い流した。非吸着画分の洗浄が終了した後、0.01Mリン酸バッファー、2M NaCl,pH7.4の通液を開始し、カラムに吸着しているウイルスが完全に溶出されるまで通液を実施した。溶出終了後、回収した流出液の1.5ml画分の50μLを使用し、HA価を測定した。さらに、試験ウイルス液50μLのHA価の1/10に到達する直前の画分までの総液量(μL)を求めた。本測定系に於ける10%DBCは以下のようにして求めた。
10%DBC(HA価/ml-gel)
=(試験ウイルス液50μLのHA価の1/10に到達する直前の画分までの総液量× 1/50×試験ウイルス液50μLのHA価)/カラム容量(ml-gel)
(2)ウイルス活性HA価の測定
丸底96ウェルプレートの各ウェルに生理食塩水50μLを加え、縦1列目に評価サン
プル(すなわち、上記で得られた溶出画分および参照としての不活化ウイルス含有液)5
0μLを加えてよく混合した。この2倍希釈されたサンプルのうち50μLを横隣のウェ
ルに加えよく混合した。同様の操作を繰り返し、12番目のウェルまで2~4096倍の
希釈サンプルを調製した。各サンプルに0.5%鶏赤血球浮遊液50μLを加えて混合
した後、室温で1時間放置した。1時間後、血球が凝集して底に沈殿していないものを
ウイルス活性有りとし、活性が認められなくなる一つ前までの希釈倍率をサンプル50μ
LあたりのHA価(HAU/50μL)とした。また、ネガティブコントロールには生理
食塩水を用いた。
丸底96ウェルプレートの各ウェルに生理食塩水50μLを加え、縦1列目に評価サン
プル(すなわち、上記で得られた溶出画分および参照としての不活化ウイルス含有液)5
0μLを加えてよく混合した。この2倍希釈されたサンプルのうち50μLを横隣のウェ
ルに加えよく混合した。同様の操作を繰り返し、12番目のウェルまで2~4096倍の
希釈サンプルを調製した。各サンプルに0.5%鶏赤血球浮遊液50μLを加えて混合
した後、室温で1時間放置した。1時間後、血球が凝集して底に沈殿していないものを
ウイルス活性有りとし、活性が認められなくなる一つ前までの希釈倍率をサンプル50μ
LあたりのHA価(HAU/50μL)とした。また、ネガティブコントロールには生理
食塩水を用いた。
その結果、ウイルス吸着量は、市販セルファインサルフェイトを用いた場合には、64000HA/ml-gelであり、製造例5のクロマトグラフィー用充填剤を用いた場合には217600HA/ml-gelであった。また、製造例6のクロマトグラフィー用充填剤を用いた場合には、112000HA/ml-gelであった。
親密粒子である市販セルファインサルフェイトと比較して、製造例5、製造例6のクロマトグラフィー用充填剤は、インフルエンザウイルスの10%DBCがそれぞれ3.4倍、1.75倍を示した。分子量200万のデキストランの保持時間が29.6分である多孔質粒子を利用した製造例5は高い10%DBCを示した。一方、分子量200万のデキストランの保持時間が15.0分である多孔質粒子を利用した製造例6は製造例5と比較して低い10%DBCであった。
親密粒子である市販セルファインサルフェイトと比較して、製造例5、製造例6のクロマトグラフィー用充填剤は、インフルエンザウイルスの10%DBCがそれぞれ3.4倍、1.75倍を示した。分子量200万のデキストランの保持時間が29.6分である多孔質粒子を利用した製造例5は高い10%DBCを示した。一方、分子量200万のデキストランの保持時間が15.0分である多孔質粒子を利用した製造例6は製造例5と比較して低い10%DBCであった。
[圧力損失と線速度の関係]
内径10mmのクロマトグラフィー用カラム(JNC製)に製造例5、製造例6で得られた粒子をそれぞれ10cm(±1cm)充填した。このカラムに20℃の純粋を通液しその際のカラム入口とカラム出口の圧力差である圧力損失を測定した。なお、通液は初期においては、0.5ml/minから始めて、その後流量を段階的に増加させ。1分以上通液を継続した後、圧力損失を測定した。ブランクとして、粒子を詰めずにカラムのみにかかる圧力損失を測定しておき、粒子にかかる圧力を以下の式で算出した。

また、線速度は以下の式で計算した。
線速度(cm/h)=測定時の流量(ml/h)/カラム断面積(cm2)
内径10mmのクロマトグラフィー用カラム(JNC製)に製造例5、製造例6で得られた粒子をそれぞれ10cm(±1cm)充填した。このカラムに20℃の純粋を通液しその際のカラム入口とカラム出口の圧力差である圧力損失を測定した。なお、通液は初期においては、0.5ml/minから始めて、その後流量を段階的に増加させ。1分以上通液を継続した後、圧力損失を測定した。ブランクとして、粒子を詰めずにカラムのみにかかる圧力損失を測定しておき、粒子にかかる圧力を以下の式で算出した。

また、線速度は以下の式で計算した。
線速度(cm/h)=測定時の流量(ml/h)/カラム断面積(cm2)
結果を図8に示した。図8から明らかであるとおり、製造例5、製造例6は市販セルファインサルフェイトに対し、同じ線速度において、圧力損失が少なく、高い流速特性を維持しているといえる。
本結果から、本発明の多孔質粒子をクロマトグラフィー用充填剤として用いた場合は、対照とする市販セルファインサルフェイトに対し、10%DBCが3.4倍で、さらに圧力損失特性も良好であった。
本発明では、制御された三次元ネットワーク状の骨格とその空隙からなる連通孔を有しており、かつ前記連通孔が粒子表面から内部まで貫通している多孔質粒子とその製造方法、およびこれを用いたクロマトグラフィー用充填剤としての利用を示した。本発明の製造方法で得られた多孔質粒子は、粒子内部の連通孔の空隙も吸着サイトとして利用できるクロマトグラフィー用充填剤へ応用できる。そのため、医薬品などの分離・精製プロセスにおいて、高処理量、高処理速度が達成でき、産業上非常に有用である。
Claims (14)
- 球状で、三次元ネットワーク状の骨格とその空隙からなる連通孔構造を有しており、かつ前記連通孔が粒子表面から内部にかけて貫通しており、 内径0.78cm×長さ30cmのカラムに充填し流量0.4ml/min のときの分子量200万のデキストランの保持時間が20分以上であることを特徴とする、主成分が酢酸セルロースまたは、セルロースである多孔質粒子。
- 内径0.78cm×長さ30cmのカラムに充填し流量0.4ml/min のときの粒径30nmのシリカ製ナノ粒子の保持時間が16分以上であることを特徴とする、請求項1に記載の多孔質粒子。
- モード径が1~1000μmの範囲である請求項1または2に記載の多孔質粒子。
- BET多点法による比表面積が1~200m2/gの範囲である請求項1~3のいずれか一項に記載の多孔質粒子。
- (a)酢酸セルロースを酢酸セルロースが可溶な溶媒と酢酸セルロースが不溶な溶媒との混合溶媒に加熱溶解して、酢酸セルロース溶液を調製する工程と、(b)前記酢酸セルロース溶液を、乳化安定剤を含む水中に分散させて、分散系を得る工程と、(c)前記分散系を冷却し、酢酸セルロース粒子を析出させる工程を含み、前記混合溶媒が水と混和しない有機溶媒である、多孔質粒子の製造方法。
- (a)において、前記酢酸セルロースが可溶な溶媒は、ベンジルアルコール、酢酸エチル、シクロヘキサノン、またはイソホロンである請求項5に記載の多孔質粒子の製造方法。
- (a)において、前記酢酸セルロースが不溶な溶媒は、アルコール類、グリコール類、エーテル類、またはエステル類である請求項5または6に記載の多孔質粒子の製造方法。
- (a)において、混合溶媒における酢酸セルロースが可溶な溶媒と酢酸セルロースが不溶な溶媒との体積比が5:95~95:5である請求項5~7のいずれか一項に記載の多孔質粒子の製造方法。
- (a)において、混合溶媒における酢酸セルロースの含量は、前記酢酸セルロース溶液に対して0.1~30重量%である請求項5~8のいずれか一項に記載の多孔質粒子の製造方法。
- (a)において、混合溶媒が高分子である第三成分を含有し、第三成分がポリエチレングリコール、またはポリプロピレングリコールである請求項5~9のいずれか一項に記載の多孔質粒子の製造方法。
- (b)において、前記乳化安定剤は、ドデシルベンゼンスルホン酸ナトリウム、ポリビニルアルコール、カルボキシメチルセルロース、ヒドロキシエチルセルロース、またはこれらの混合物である請求項5~10のいずれか一項に記載の多孔質粒子の製造方法。
- (c)において、分散系の冷却速度は、0.1℃/min~10℃/minである請求項5~11のいずれか一項に記載の多孔質粒子の製造方法。
- 請求項1~4のいずれか一項に記載の多孔質粒子、またはケン化または架橋または修飾された前記多孔質粒子を含む、クロマトグラフィー用充填剤。
- ウイルス粒子を分離精製するために使用される、請求項13に記載のクロマトグラフィー用充填剤。
