Claims (28)

1. Слитый белок IL-7, включающий цепочку иммуноглобулина и молекулу IL-7, которая является модифицированной по сравнению с IL-7 дикого типа, связанные вместе непосредственно или посредством молекулы линкера, где модификация в IL-7 представляет собой
(i) аминокислотные остатки в положениях 70 и 91 являются гликозилированными, а аминокислотный остаток в положении 116 является негликозилированным;
и/или (ii) в пределах остатка IL-7 существуют дисульфидные связи между Cys2 и Cys92, Cys34 и Cys129 и Cys47 и Cys141.
2. Слитый белок IL-7 по п.1, в котором аминокислотные остатки в положениях 70 и 91 IL-7 являются гликозилированными, а аминокислотный остаток в положении 116 IL-7 является негликозилированным, и существуют дисульфидные связи в пределах остатка IL-7 между Cys2 и Cys92, Cys34 и Cys129 и Cys47 и Cys141.
3. Слитый белок IL-7 по п.1, в котором аминокислотный остаток в положении 116 представляет собой аспарагин.
4. Слитый белок IL-7 по п.1, в котором аминокислотный остаток в положении 116 является негликозилированным.
5. Слитый белок IL-7 по п.1, в котором аминокислотный остаток в положении 116 является измененным так, что он не служит сайтом гликозилирования.
6. Слитый белок IL-7 по п.1, в котором молекула IL-7 включает делецию.
7. Слитый белок IL-7 по п.6, в котором укороченный IL-7 включает делецию восемнадцати аминокислот от аминокислоты 96 до аминокислоты 114 SEQ ID NO: 1.
8. Слитый белок IL-7 по п.1, в котором молекула IL-7 представляет собой IL-7 человека.
9. Слитый белок IL-7 по п.1, в котором IL-7 представляет собой остаток зрелого IL-7.
10. Слитый белок IL-7 по п.1, в котором цепочка иммуноглобулина включает, по крайней мере, часть константного домена.
11. Слитый белок IL-7 по п.10, в котором константный домен является выбранным из группы, состоящей из СН1, СН2 и СН3.
12. Слитый белок IL-7 по п.10, в котором константный домен представляет собой константный домен IgG1.

14. Слитый белок IL-7 по п.13, в котором модификация Asn297 представляет собой Asn297Gln.
15. Слитый белок IL-7 по п.13, который дополнительно включает модификацию в Тyr296.
16. Слитый белок IL-7 по п.15, в котором модификация Тyr296 представляет собой Тyr296Аlа.
17. Слитый белок IL-7 по п.10, в котором константный домен представляет собой константный домен IgG2.
18. Слитый белок IL-7 по п.17, в котором цепочка IgG2 включает шарнир IgG1.
19. Слитый белок IL-7 по п.17, в котором Asn297 константного домена IgG2 является модифицированным.
20. Слитый белок IL-7 по п.19, в котором модификация Asn297 представляет собой Asn297Gln.
21. Слитый белок IL-7 по п.19, дополнительно включающий модификацию в Phe296.
22. Слитый белок IL-7 по п.21, в котором модификация Phe296 представляет собой Phe296Ala.
23. Слитый белок IL-7 по п.1 или 2, в котором остаток IL-7 слитого белка обладает повышенной биологической активностью по сравнению с IL-7 дикого типа.
24. Изолированная молекула ДНК, кодирующая слитый белок IL-7 по любому из пп.1-23.
25. Культивируемая хозяйская клетка, включающая ДНК по п.24.
26. Способ получения слитого белка, включающий: трансформацию хозяйской клетки с помощью ДНК по п.24, культивирование хозяйской клетки, а также экспрессию и сбор слитого белка IL-7.
27. Фармацевтическая композиция, приемлемая для лечения иммунных расстройств или рака, включающая фармацевтически эффективное количество слитого белка IL-7 по любому из пп.1-23, необязательно вместе с фармацевтически приемлемым носителем, разбавителем или наполнителем.
28. Применение слитого белка IL-7 по любому из пп.1-23 для получения лекарственного средства для лечения иммунных дефицитов и рака.
