Abstract
本发明属于治疗性生物制剂领域,具体讲,涉及一种新型羧基末端肽和一种白介素,本发明还涉及该新型羧基末端肽的应用,以及采用本发明的羧基末端肽制备得到的长效人白介素7。本发明通过羧基末端肽的作用,延长与其融合的白介素蛋白的半衰期,并且对白介素的活性无明显影响或可增强其活性。

技术领域
本发明属于治疗性生物制剂领域,涉及一种新型羧基末端肽及长效白介素-7。
背景技术
人白介素7(IL-7)于1988年确认并定位于染色体8q12-13。人IL-7蛋白含有六个外显子,小鼠IL-7为5个外显子,分子量约为25kDa,人比小鼠多的为第5个外显子,并多了17个氨基酸。IL-7主要由非造血基质细胞和上皮细胞产生,包括成纤维网状细胞、淋巴器官的T细胞区、骨髓基质细胞、主要组织相容性复合体(MHC)、(MHC)Ⅱ类+胸腺上皮细胞、肝和肠上皮细胞、内皮细胞、成纤维细胞、角质形成细胞、滤泡树突状细胞及平滑肌细胞,DC细胞和巨噬细胞也可产生少量的IL-7。IL-7分布广泛,绝大多数器官包括大脑均表达。因为IL-7存在于大多数组织内,并且几乎所有的外周T细胞均表达IL-7受体,最近研究表明IL-7通过一个简单的IL-7R介导的反馈回路直接控制信号传导和激活。IL-7由153个氨基酸组成,含有三个二硫键,三个N糖基化,一个O糖基化位点。
IL-7是γc家族细胞因子,与细胞表面IL-7受体复合物结合,α链(IL-7R,CD127)和γ链(CD132)共同形成的受体复合物。其中CD127为IL-7和胸腺基质淋巴细胞生成素(TSLP)共同受体,而CD132为IL-7和IL-2,IL-4,IL-9,IL-15和IL-21共同使用。IL-7二聚体受体在效应T细胞和记忆T细胞上高表达,该受体也在单核细胞、DC细胞、发育中的B细胞和T细胞上表达,但是不在成熟的B细胞上表达。人IL-7能够结合鼠IL-7受体,并能够通过该受体传导信号,反之亦然。刺激的IL-7R激活Janus激酶/信号转导和转录激活(JAK/STAT主要是JAK1,JAK3和STAT5),转录因子(JAK/STAT主要JAK1,JAK3,STAT5),Src家族激酶和磷脂酰肌醇3-激酶信号通路(phosphatidylinositol 3-kinase(PI3K-AKT)signaling pathways)。
IL-7是免疫调节和免疫增强的重要细胞因子,在免疫系统的各个阶段均起作用,从骨髓基质细胞,到胸腺,再到外周血的各种效应细胞,都可见IL-7的作用。在B细胞分化的某些阶段、T细胞和NK细胞的存活、发育和稳态等多方面都发挥着重要作用。因此认为IL-7是记忆T细胞产生和维持的关键因素之一。在造血干细胞向淋巴细胞分化和T细胞的阴阳性选择中有重要作用,还可以使初始T细胞活化后经过多轮扩增变成效应T细胞。研究表明,IL-7能够增强CD4+效应T细胞作用,并促使具有IL-7受体的效应T细胞发展成为长期存活的记忆T细胞。在胸腺发育早期对IL-7的需求是因为IL-7有利于Rag-1基因的表达,该基因是TCR重组必须的。Il-7对T细胞发育至关重要,参与T细胞的阴阳性选择,并促进T细胞受体(TCR)β,γ和δ基因重排。对于γδT细胞的分化发育,IL-7是必不可少的。另外,IL-7也能促进记忆细胞反向分化成效应CD4+和CD8+细胞。另外,IL-7也能促进记忆细胞反向分化成效应CD4+和CD8+细胞。IL-7对于B细胞的发育也很重要,尤其是pre-pro-B细胞发育成B细胞。IL-7对B淋巴系细胞发育的作用首先是与分泌IL-7的骨髓基质细胞作用,在V、D、J重排过程中也发挥作用。IL-7使Igu链成功表达,并与替代轻链、Igα和Igβ的信号亚单位结合形成B细胞受体(pre-BCR),pre-BCR促使一群大的pre-B细胞(pre-BII cells)产生和扩增。随着年龄的增长,胸腺退化,对淋巴细胞从胸腺向外周血转移初始T细胞的能力显著降低。与初始T细胞相比,当记忆细胞针对回忆性抗原的免疫能力增强,但是会限制其针对新抗原的免疫能力。因此,IL-7作用于前T细胞的生物活性大小与年龄相反的,越小越强。
homeostatic peripheral expansion(HPE)是指在去除或部分去除T细胞的宿主上过继移植T细胞或对剩余的T细胞进行扩增的方式来重构T细胞数量和功能的手段。在HPE中,低亲和力的抗原作用更大,因为在淋巴细胞减少的情况下,T细胞针对广谱抗原的扩增便于维持足够T细胞受体(TCR)的丰度。IL-7水平提高介导T细胞针对自身抗原提供持续的刺激信号扩增,这种信号就是低亲和力抗原刺激T细胞受体的信号。对于透析所致的淋巴细胞减少,HIV所致的淋巴细胞CD4+减少,骨髓移植等淋巴细胞减少或者由于治疗自身免疫性疾病采用了单抗而导致T细胞减少的情况,IL-7可以增加TCR的多样性和T细胞数量。另外,抗凋亡效应是IL-7生物活性的重要贡献,IL-7通过上调Bcl-2和Mcl-1提供抗凋亡信号,抑制前凋亡信号,如BAX或BIM,便于维持长期记忆性T细胞。
IL-7的受体在体内广泛分布,为α链(IL-7R,CD127)和γ链(CD132)共同形成的受体复合物。IL-7R可以反馈调节IL-7的浓度。因为CD132的表达基本上是恒定的,所以它能够过调节CD127的表达量来调节IL-7的受体表达量,从而调节IL-7的功能。但T细胞数量正常时,IL-7信号降低CD127在成熟T细胞表面的表达,这种下调或许可以提高有限数量的IL-7作用于更多的T细胞。IL-7下调CD127的表达也可以限制T细胞的无限制分化。因此,动态调节CD127的表达在效应T细胞向记忆T细胞转化过程中至关重要。
IL-7注射于正常小鼠时,会导致外周淋巴器官的T细胞增加,当注射到环磷酰胺使淋巴细胞减少或在骨髓移植中全身照射小鼠,会加速T细胞的恢复而使脾脏和淋巴结的T细胞增加。这些研究为IL-7对胸腺依赖或非胸腺依赖的免疫重构提供了依据。IL-7治疗可以提高免疫反应,尤其可以大幅度提高对低亲和性或弱的免疫原的反应。由于自身抗原—肿瘤抗原没有免疫原性或免疫原性很弱,IL-7的上述特征使得其成为一种针对肿瘤免疫治疗的极具可能的潜在佐剂。目前,艾滋病、肝炎和结核病(HIV、HBV、HCV、TB)等疾病在我国甚至全世界都是严重的公共卫生问题;并且随着人口老龄化,肿瘤患病率越来越高。目前,IL-7作为治疗慢性病毒性感染疾病、结核感染和肿瘤的备选药物或佐剂,开展了多达27项相关的临床试验,应用前景光明。
接受IL-7治疗的病人显示CD4+和CD8+的数量都增加,有剂量效应关系,并伴随着T细胞循环加快。淋巴细胞的增加与年龄关系不大,然而幼稚T细胞尤其幼稚CD8+T细胞最先诱导进入循环,因此他们的数量比记忆性细胞的数量多。TCR谱测定显示;IL-7治疗后,T细胞受体丰度大大增加,选择性的增加了高表达CD127的群落。CD4+和CD8+T亚群的增加是对IL-7剂量依赖性的。在一些病人中,骨髓中的前B细胞增加,但是在外周循环并不出现这种增加。与对照组相比,没有显示针对恶性肿瘤的抗原增加了免疫反应性,但是相关数据比较少,还需进一步验证。
HIV病毒的细胞毒性结合对细胞的其他间接性的影响,如慢性活化免疫系统导致消耗T细胞库,不完全恢复失去的T细胞等使CD4+T细胞逐渐消失,导致针对特异性免疫反应的能力缺失。IL-7能提高HIV-1特异性CD8+的分化和细胞毒杀伤活性(CTL)。由于IL-7治疗能够提高CD4+,所以可作为治疗HIV的候选药物或佐剂。也能提高用HIV-1膜蛋白免疫后的CD4+T细胞的体液免疫,提高α干扰素抗HIV复制的作用,还可以提高CD4+和CD8+T细胞的抗凋亡的能力。HIV感染者PBMC体外培养证明IL-7能够增强HIV复制能力,并且提高CXCR4和CXCR5受体的表达,使HIV更容易进入细胞。
IL-7用于肿瘤治疗的机制在于是:1.在化疗后可以使用IL-7来重构免疫系统;2.通常在使用药物化疗后,淋巴细胞减少,T细胞数量也减少,尤其是CD4+细胞,免疫细胞低于正常值长达数月至数年。一种策略是利用过继性免疫治疗技术,将细胞在体外激活和扩增,然后回输给患者,这种策略也可用在放射性病人或化疗后的病人。重构肿瘤病人免疫系统,使用IL-7治疗年龄越小越好,因为小孩具有产生初始T细胞谱的能力更强,并且比成人更完善。
通过糖基化来延长半衰期:Ceaglio等通过定点突变的方法将一段N糖基化共有序列插入到干扰素分子的不同位置,构成4N和5N-干扰素的变异体,使得分子大小和电荷数均有所增加,后者的生物学活性与未突变的重组人干扰素-α2活性相当,但降低了肾脏对干扰素的清除作用,延长了半衰期。
人绒毛膜促性腺激素(HCG)-β亚单羧基端肽(CTP):HCG是由胎盘的滋养层细胞分泌的一种糖蛋白,它是由α和β二聚体的糖蛋白组成。HCG-β末端比人黄体生成素末端多37个氨基酸。常用其中的一部分融合其他蛋白,增加其半衰期(WO/2014/080401A2)。CTP蛋白具有延长融合蛋白在体内半衰期的作用,不影响被融合蛋白的生物活性,并且来源于人体,不会产生抗体。为了增加蛋白的分子量,可以在同一蛋白上融合一个或多个CTP蛋白,可在蛋白的N端融合,也可在蛋白的C端融合。有研究表明融合多个CTP也不影响蛋白的结构,且效果更好。CTP与促卵泡激素或生长激素融合的蛋白,在体内没发现免疫性,并且与促卵泡激素融合的蛋白已经在欧洲获得批号,融合CTP蛋白的生长激素也在进行二期实验。
常用的流式细胞仪筛选高表达细胞株的方法:直接针对分泌蛋白的:微胶滴法(Gelmicrodrops)、亲和捕获术(Affinity capture surface display,ACSD)、分泌捕获技术(Secretionand capture technology,SECANTTM)和冷冻捕获法等,但这些方法不是很准确,检测指标和是否是高表达细胞株相关性较低。流式细胞仪对于细胞分泌性蛋白检测效果不好,而针对膜蛋白检测效果较好。如目的蛋白协同表达某一膜蛋白,用流式细胞仪针对膜蛋白分选,则可获得高表达细胞。IRES原件采用5’端介导翻译机制,可以较低效率的翻译其后面的蛋白。人CD20不在CHO细胞上表达,通过IRES元件把CD20与目的蛋白核酸序列结合,基因表达盒如附图1所示。转录于同一mRNA上,所以转录水平是一致的,但可翻译成不同的蛋白,目的蛋白分泌在培养液中,CD20在细胞膜上,这样可以通过检测CD20来评估目的蛋白的表达情况,筛选出表达量高的细胞株。属于同一mRNA的CD20的表达量与目的蛋白表达量是线性相关的,可反应同一mRNA的翻译情况,所以其既可以作为流式细胞仪筛选标记,同时又能反映细胞目的蛋白表达水平。本发明的优点是不受特异性抗体的限制,可以筛选目前还没有抗体的蛋白质。
鉴于此,特提出本发明。
发明内容
本发明的首要发明目的在于提出一种新型的羧基末端肽。
本发明的第二发明目的在于提出一种长效白介素-7。
本发明的第三发明目的在于提出该长效白介素-7的应用。

本发明涉及一种羧基末端肽,该羧基末端肽上具有N-糖基化位点,该N-糖基化位点可设置于羧基末端肽的任意位置,并优选设置于羧基末端肽的末端;并且,本发明的N-糖基化位点可包含所有的可构成N-糖基化位点的氨基酸序列,进一步优选的,其氨基酸序列如SEQ IDNO:1所示。
本发明还涉及一种如SEQ ID NO:2所示的编码权利要求1所述羧基末端肽的核苷酸序列。
本发明还涉及一种重组表达载体,该重组表达载体含有SEQ ID NO:2所示的核苷酸序列。
本发明还涉及一种宿主细胞,该宿主细胞含有上述重组表达载体,宿主细胞为CHO细胞。
本发明还涉及一种长效干扰素,该长效干扰素由本发明的羧基末端肽与白介素-7连接而成,该长效白介素-7的核苷酸序列如SEQ ID NO:5所示,该白介素-7的核苷酸序列如SEQ IDNO:3所示。
该长效白介素-7的制备方法为:将编码N糖基化修饰的羧基末端肽的核苷酸序列与编码白介素-7的核苷酸序列连接,克隆到表达载体pcDNA5/FRT和pIRES,脂质体转染Flp-In-CHO和CHO-K1细胞,筛选高表达的细胞株。其中,筛选高表达细胞株的方法为Hygromycin B加压筛选和流式细胞仪针对pIRES的IRES元件后表达的人CD20筛选;浓缩和纯化方法为:细胞培养上清用(NH4)2SO4进行沉淀浓缩,等电点沉淀、疏水层析、离子交换、Blue Sepharose 6FF和分子筛s-200对样品进行纯化。
本发明还涉及该长效白介素-7在制备治疗肿瘤、慢性病毒性疾病、免疫性疾病或放射、化疗以及移植引发的免疫系统损伤的药物中的应用。
下面对本发明的内容做进一步的说明书。
人绒毛膜促性腺激素(HCG)-β亚单羧基端肽(CTP):HCG是由胎盘的滋养层细胞分泌的一种糖蛋白,它是由α和β二聚体的糖蛋白组成。HCG-β末端比人黄体生成素末端多37个氨基酸。CTP是人绒毛膜促性腺激素亚基(hCGβ)的羧基末端肽(C-terminal peptide,CTP),含有四个O-糖基化位点,其中的19-30个氨基酸常被用于融合蛋白表达的CTP蛋白。
本发明提出了一种全新的羧基末端肽,其氨基酸序列如SEQ ID NO:1所示。编码该多肽的核苷酸序列如SEQ ID NO:2所示。hCGβ亚基含有145个氨基酸,109-145位氨基酸是CTP,共37个氨基酸,本发明去掉前面13个氨基酸,利用后面的24个氨基酸作为长效的CTP分子。本发明CTP在N端加上4个连接肽,与前面要表达的蛋白质的核酸相连接,在C端加上4个氨基酸为N-糖基化位点。

两端下划线为实线的部分为酶切位点和酶切位点保护碱基,字体倾斜部分为IL-7序列,黑体部分为linker序列,而下划线为虚线的部分为CTP蛋白,双实现下划线部分为增加的N糖基化位点。
本发明还涉及该羧基末端肽在制备长效多肽、长效蛋白或长效细胞因子中的应用。通过O-糖基化位点和N-糖基化位点的协同作用,从而不仅可以延长与其融合的蛋白分子的半衰期,并且可增强其活性。同时在IL-7的末端融合了经过改造的CTPON蛋白,CTPON蛋白的主要功能就是延长预期融合蛋白的半衰期,并且不影响被融合蛋白的生物活性。所以本发明使用了两种方式延长蛋白在体内的半衰期,一是增加蛋白的糖基化,二是融合长效蛋—CTPON蛋白。
目前白介素-7广泛地被认为是慢性病毒性疾病如HIV/HBV/HCV等,一些肿瘤、免疫性疾病和放射、化疗以及移植等免疫系统损伤后免疫系统重构的潜在有效药物。但普通白介素-7半衰期短,在临床使用中给病人带来较多的不便,而长效IL-7既可以减轻病人的痛苦,同时可以使血药浓度保持比较平稳,有利于疾病的治疗。因此白介素-7长效研究成为热点。本发明为IL-7的长效化提供了一个全新的方式。
本发明的长效白介素-7的制备方法为:
1. IL-7-CTP和IL-7-CTPON载体构建及稳定细胞株筛选
首先对人CTP蛋白进行改造,在其末端增加了一个N糖基化氨基酸,接着对IL-7-CTPON进行密码子优化,参考密码子为仓鼠密码子表,优化方法为本专业常用手段。而后合成带有酶切位点的IL-7-CTPON,酶切位点为:Nhe Ⅰ/Not Ⅰ;而后应用PCR技术获得IL-7序列,引入酶切位点Nhe Ⅰ/Not Ⅰ,并与表达载体pcDNA5/FRT连接。分别构建了pcDNA5/FRT-IL-7和pcDNA5/FRT-IL-7-CTPON两个质粒,测序确定质粒构建成功。按照本专业的常规手段,提取质粒,并纯化质粒后使用脂质体转染技术将构建的质粒和辅助质粒pOG44按照说明书以1:9的比例转入Flp-In-CHO细胞,转染24小时后用600ug/ml Hygromycin B筛选,得到稳定表达IL-7和IL-7-CTPON的细胞株进行扩大培养。
使用PCR技术将IL-7-CTPON酶切位点换成Nhe I和ECOR I,插入pIRES载体的第一个克隆位点;将人CD20mRNA序列反向优化后合成,在其两端引入酶切位点:Xba Ⅰ/Not Ⅰ将其插入在IRES元件后面的第二个克隆位点。克隆在DH5α菌里后提取质粒,并测序验证后,按照上述手段,将质粒转染进CHO-K1细胞里,在G418 500ug/ml筛选作用后使用流式细胞仪筛选出表达量较高的细胞株,并继续测定和放大培养。而后再使用ELISA法测定细胞表达量。
2. IL-7-CTP和IL-7-CTPON蛋白纯化
由于IL-7-CTP和IL-7-CTPON均是糖蛋白,并且有数个糖基化位点,所以在对其进行真核表达时,可能会出现不同糖基化的蛋白,即分子量大小不一的数个蛋白,因此在纯化的各阶段会获得不同的蛋白。
针对本表达产物的纯化使用了硫酸铵沉淀、等电点沉淀、疏水层析、Blue Sepharose 6FastFlow和分子筛s-200等获得IL-7-CTP和IL-7-CTPON蛋白,获得较纯的表达产物,该产物为多种糖基化不同的蛋白构成的混合物,如附图2所示。
对IL-7-CTPON较纯的产品进行western blot检测后,发现至少有6中不同分子量大小的糖蛋白构成,IL-7-CTP也有4种分子量大小的糖蛋白构成,如附图6所示。
本发明的有益效果为:
本发明制备得到的长效白介素的半衰期得到大大延长,从而得到了一种生理活性高、体内半衰期显著增加的长效白介素。IL-7是免疫调节和免疫增强的重要细胞因子,在免疫系统的各个阶段均起作用,从骨髓基质细胞,到胸腺,再到外周血的各种效应细胞,都可见IL-7的作用。在B细胞分化的某些阶段、T细胞和NK细胞的存活、发育和稳态等多方面都发挥着重要作用。因此认为IL-7是记忆T细胞产生和维持的关键因素之一。
在体外IL-7可以刺激小鼠的T细胞增殖,也可以刺激人的外周血单核细胞增殖。本试验通过在体外使用本实验室表达的IL-7-CTP和IL-7-CTPON对小鼠CTLL细胞的增殖实验,以R&D公司的原核表达的IL-7为对照产品,采用了promega的G4000测定。
IL-7-RD的ED50在为0.635ng/ml,比活性为1.58×106U/mg;IL-7-CTP的ED50为0.675ng/ml,比活性为1.48×106U/mg,IL-7-CTPON的ED50为0.90ng/ml,比活性为1.11×106U/mg。
以R&D公司的原核IL-7为参照,小鼠分别注射三种白介素-7,在0.5,2,6,26.2和30小时分别测定血清中白介素-7的含量,IL-7-RD的半衰期为5.79小时,MRT为8.17小时,IL-7的半衰期为17.32小时,MRT为28.12小时,IL-7-CTPON的半衰期为25.68小时,MRT为36.27小时。
IL-7和IL-7-CTPON在体外促进人外周血单核细胞的增殖,经实验检测,本发明的IL-7-CTPON和IL-7-CTP均可以促使人外周血单个核细胞的增殖,并且在相同浓度下,IL-7-CTPON效果优于IL-7-CTP,IL-7-CTP效果又优于IL-7-RD;并且在一定浓度范围内(<10ng/ml)可见,随着浓度增加,效果越明显;在三种药物相同剂量组中可见,第五天效果比作用3天的效果明显。表明本发明的IL-7-CTP和IL-7-CTPON的在体外具有很强的生物活性。

具体实施方式
实施例1
1材料
1.1主要试剂与材料
IL-7-CTPON序列均由北京天一辉远生物科技有限公司合成。大肠杆菌E.Coli菌株DH5α感受态购自天根生物公司。克隆载体pMD18T购自Takara公司。T4DNA连接酶为Promega产品。Flp-In-CHO细胞、pOG44质粒和pcDNA5/FRT质粒购自Invitrogen公司(美国)IL-7-RD标准蛋白购自R&D公司;IL-7Human ELISAKit购自Abcam公司。Taq DNA聚合酶、限制性内切酶Nhe Ⅰ/Hind Ⅲ、Not Ⅰ等均购自Takara公司。蛋白纯化凝胶SP SepharoseFF,Blue Sepharose6,S-200均购自美国GE公司。TRIZOL、RT-PCR Kit、Ⅲ逆转录试剂盒,购自Invitrogen公司;Qiagen RNeasy protect min kit、RNase free DNase set均购自Qiagen公司;Non-Radioactive Cell Proliferation AssaypromegaG4000试剂盒购自Promega公司。
2.方法
2.1.序列优化:
由于CHO细胞来源于中国仓鼠的卵巢细胞,而IL-7和CTP均来源于人,二者之间存在物种间遗传背景的差异,因此所使用的密码子频率有差异,可能会因为转运RNA的数量等不一样,影响翻译和表达速度。为了更好的利用仓鼠体内的转录、翻译和翻译后修饰机制,需要对表达的IL-7-CTP进行密码子优化,优化成小鼠偏嗜的密码子,减少因为密码子的不一致而影响目标的表达量。密码子的优化通常是通过大量的测序确认需要用到的表达体系的密码子,然后根据其密码子偏好性,通常将需要表达的目标的密码子的第三位进行修改,使之蛋白没有改变,但是密码子已经改变。本发明对IL-7的核苷酸序列的密码子进行优化,优化后的核苷酸序列如SEQ ID NO:3所示。序列优化后,与原序列相似性为73%,但是密码子适应指数(codon adaptation index,CAI)由0.68升高到1.0,有效密码子数(effectivenumber of codons,ENC)由54降到21,与仓鼠的密码子更加符合,优化后GC%含量也得到提高。
2.2.载体构建和单克隆筛选
直接合成带有酶切位点的IL-7-CTPON(SEQ ID NO:5),酶切位点为:Nhe Ⅰ/Not Ⅰ。
IL-7-CTP的制备:
使用PCR从IL-7-CTPON上扩增IL-7-CTP序列(SEQ ID NO:4),并引入酶切位点Nhe
Ⅰ/Not Ⅰ;
PCR体系为:

PCR反应条件:95℃5min,(95℃30s,58℃30s,72℃60s)35循环,72℃7min。
产物纯化回收后,与pGEM-T-easy载体连接,转化DH5α大肠杆菌,经测序鉴定正确的质粒,用NheI和ECORI分别双酶切质粒和pIRES载体,回收酶切下来的IL-7-CTP和载体,以T4连接酶连接后转化DH5α大肠杆菌,经测序确认正确的菌落留存待用。
小提质粒pcDNA5/FRT、IL-7-CTP和IL-7-CTPON质粒。对三种质粒分别进行双酶切,酶切体系如表2所示:

37℃酶切过夜,胶回收酶切产物。
将IL-7-CTP和IL-7-CTPON质粒与载体pcDNA5/FRT在T4连接酶链接4℃连接过夜。转化DH5α大肠杆菌,测序确定质粒构建成功。
提取质粒,纯化质粒后使用脂质体转染技术将构建的质粒和辅助质粒pOG44按照说明书以1:9的比例转入Flp-In-CHO细胞,转染24小时后用600ug/ml Hygromycin B筛选,得到稳定表达IL-7和IL-7-CTPON的细胞株进行扩大培养。
使用引物SEQ ID NO:8、SEQ ID NO:9从pcDNA5/FRT-frt-IL-7-CTPON上扩增IL-7-CTPON片段,获得含有NheI和ECoRI酶切位点的IL-7-CTPON;
使用引物SEQ ID NO:8、SEQ ID NO:10从pcDNA5/FRT-frt-IL-7-CTP上扩增IL-7-CTP片段,获得含有NheI和ECoRI酶切位点的IL-7-CTP。

将人CD20mRNA序列反向优化后合成(CD20核苷酸序列如SEQ ID NO:11所示),在其两端引入酶切位点:Xba Ⅰ/Not Ⅰ将其插入在IRES元件后面的第二个克隆位点。克隆在DH5α菌里后提取质粒,并测序验证后,将质粒转染进CHO-K1细胞里,经G418 500ug/ml筛选作用,将G418筛选到的CHO-K1-IL-7-CTP-IRES-CD20或CHO-K1-IL-7-CTPON-IRES-CD20表达抗G418的细胞使用PBS清洗2遍,清洗后用BD公司FITC标记的anti-human CD20染色。每1×107细胞在250ul培养液中加入5ul抗体,在室温孵育15分钟,间断性的轻微晃动,再次使用PBS清洗两遍后,去除多余的CD20抗体及粘附在细胞表面的抗体,上流式细胞仪分选。
将选出的细胞在48孔板中培养,培养时前10小时不加G418,血清为15%,加入1%的PS防止污染。因为流式细胞仪筛选时对细胞是有一定的损伤的,所以部分细胞会死亡。当4~5天后细胞铺满48孔板,一部分继续转入6孔板,一部分按照有限稀释法使细胞在96孔板中单克隆化,10~14天后,对收到的单个细胞形成的克隆,转入24孔板中培养。在24孔水平使用ELISA鉴定表达量,方法按照筛选FLP-INTM-CHO-IL-7-CTP的方法,对高表达细胞株进行冻存和扩大培养。
2.3.纯化条件:
IL-7-CTP和IL-7-CTPON均通过以下方式纯化,通过(NH4)2SO4沉淀浓缩,等电点沉淀、疏水层析、离子交换、Blue Sepharose 6FF和分子筛s-200对样品进行纯化,得到90%纯度的IL-7和IL-7-CTPON,其SDS-PAGE电泳图如附图2所示。
2.3.1硫酸铵溶液沉淀:
缓慢加入30%硫酸铵溶液,充分搅拌,低温搅拌过夜,离心,收集离心上清后缓慢加入硫酸铵粉末,使硫酸铵浓度达到75~85%,充分搅拌过夜,离心,收集沉淀,将沉淀用20nMPB稀释。稀释后在20nM PB的透析,后经超滤管浓缩。
2.3.2等电点沉淀
调节pH值为4.4后离心去沉淀,调节PH值为5.0,同样离心去沉淀,调节PH值为6.0,同样离心去沉淀。
2.3.3疏水层析
将上清pH调节至7.0~7.2,在0.05M PB中平衡,以0.05nM PB和1.5M的硫酸铵线性梯度洗脱、0.05M PB和1.0M硫酸铵洗脱、0.05M PB和0.6M硫酸铵洗脱。
2.3.4离子交换
将目的峰的洗脱液在50nM PB(PH 7.0)充分平衡后再使用离子交换(Blue Sepharose6)进行纯化,以0.5M NaCl和50nM PB(PH 7.0)到2.0M NaCl和50nM PB(PH 7.0)线性洗脱。
2.3.5亲和层析:
将目的峰在0.02M PB中平衡后,使用0.15M NaCl(pH 7.2)的溶液中平衡,然后上样,使用Blue Sepharose 6FF纯化,起始缓冲液为(0.02M PB,0.15M NaCl,pH 7.2)洗脱,待回到基线后,用buffer A(0.02M PB,2M NaCl,pH 7.2)洗脱至基线。接着使用buffer B(0.02M PB,2M NaCl,50%ethylene glycol,pH 7.2)清洗至基线,重新平衡清洗纯化柱以备下次使用。
2.3.6分子筛s-200层析:
将目的峰在0.05nM PB和0.5M NaCl中平衡后,使用分子筛S-200进行进一步纯化。流速设定为20cm/h。对目的峰采用WB和ELISA等方法鉴定,最终获得纯化产物。
2.4纯化后的蛋白样品进行western blot检测后,IL-7-CTPON发现至少有6种不同分子量大小的糖蛋白构成,IL-7-CTP有4种大小不同的糖蛋白构成,如附图6所示;样品送清华大学生物医学测试中心检测蛋白序列,鉴定目的蛋白序列正确。
实施例2长效性测定
Balb/c小鼠分为三组,每组6只,分别给予IL-7-RD、IL-7-CTP和IL-7-CTPON腹腔注射(40μg/kg),在0.5h、2h、6h、26.2h,30h小时分别取血,离心后获得血清,应用ELISA试剂盒检测血清中的各样品含量。应用药物动力学软件DAS3.0计算小鼠体内的药代动力学参数如表4所示。

实施例3比活性的测定
每个细胞孔中加入50ul 10%FBS的1640培养基,向第一列孔中加入50ul 40ng/ml的IL-7-CTP或IL-7-RD培养液,充分混匀后,取50μl加入后一孔,按照上述步骤,将IL-7倍比稀释加入到第十一孔,从第十一孔中取出50μl抛弃。每孔中加入50ul 1×106细胞,孵育36小时后加入15ul MTT,4小时后加入100ul溶解/终止反应液,过夜等结晶紫完全溶解后在570nm波长检测各浓度吸光值,根据吸光值大小画出曲线,从曲线中计算ED50,1/ED50即为比活性,如附图3所示。
IL-7-RD的ED50在为0.635ng/ml,比活性为1.58×106U/mg;IL-7-CTP的ED50为0.90ng/ml,纯度为83.2%时的比活性为1.11×106U/mg,IL-7-CTP的ED50为0.70ng/ml,比活性为1.43×106U/mg。
实施例4 PHA-P活化后,IL-7刺激健康人PBMC增殖作用
按照常规方法分离人外周血单个核细胞,使用羧基荧光素二醋酸盐琥珀酰亚胺酯(CFSE)染色。将一份使用PHA-P 5μg/ml的量活化2天,使用含3%FBS的PBS清洗两遍,进行CFSE染色。向圆底96孔板中加入含有不同浓度的IL-7-RD、IL-7-CTP和IL-7-CTPON的培养液,浓度分为0、1.25、2.5、5、10、20ng/ml共六个梯度,每个梯度2个复孔。每孔加入1×105个PBMC细胞,细胞培养72小时后,使用流式细胞仪检测细胞增殖情况,如附图4所示(柱状图从上倒下依次为P9、P8、P7、P6、P5、P4)。
将另一份PBMC细胞使用PHA-P 5μg/ml的量活化2天,使用含3%FBS的PBS清洗两遍,进行CFSE染色。向圆底96孔板中加入含有不同浓度的IL-7-RD、IL-7-CTP和IL-7-CTPON的培养液,浓度分为0、1.25、2.5、5、10、20ng/ml共六个梯度,每个梯度2个复孔。每孔加入1×105个PBMC细胞,细胞培养5天后,使用流式细胞仪检测细胞增殖情况,如附图5所示(柱状图从上倒下依次为P9、P8、P7、P6、P5)。
Claims (10)
1.一种羧基末端肽,其特征在于,所述羧基末端肽上设置有N-糖基化位点,所述N-糖基化位点设置于羧基末端肽的任意位置,并优选设置于羧基末端肽的末端;更优选的,其氨基酸序列如SEQ ID NO:1所示。
2.一种如SEQ ID NO:2所示的编码权利要求1所述羧基末端肽的核苷酸序列。
3.一种重组表达载体,其特征在于,所述的重组表达载体含有SEQ ID NO:2所示的核苷酸序列。
4.一种宿主细胞,其特征在于,所述的宿主细胞含有权利要求3所述的重组表达载体。
5.根据权利要求4所述的宿主细胞,其特征在于,所述的宿主细胞为CHO细胞。
6.一种长效白介素-7,其特征在于,所述长效白介素-7由权利要求1所述的羧基末端肽与白介素-7通过连接蛋白连接而成,所述长效白介素-7的核苷酸序列如SEQ ID NO:5所示。
7.根据权利要求6所述的长效白介素-7,其特征在于,所述白介素-7的核苷酸序列如SEQ ID NO:3所示。
8.根据权利要求6所述的长效白介素-7,其特征在于,所述长效白介素-7的制备方法为:将编码N糖基化修饰的羧基末端肽的核苷酸序列与编码白介素-7的核苷酸序列连接,克隆到表达载体pcDNA5/FRT和pIRES,脂质体转染Flp-In-CHO和CHO-K1细胞,筛选高表达的细胞株。
9.根据权利要求8所述的长效白介素-7稳定表达株的获得,筛选高表达细胞株的方法为Hygromycin B加压筛选和流式细胞仪针对pIRES的IRES元件后表达的人CD20筛选;浓缩和纯化方法为:细胞培养上清用(NH4)2SO4进行沉淀浓缩,等电点沉淀、疏水层析、离子交换、Blue Sepharose 6FF和分子筛s-200对样品进行纯化。
10.权利要求6所述的长效白介素-7在制备治疗肿瘤、慢性病毒性疾病、免疫性疾病或放射、化疗以及移植引发的免疫系统损伤的药物中的应用。
