Abstract
Description

野生型(WT)G-CSFWT:G-CSFRWT複合体は、2:2ヘテロ二量体である。G-CSFは、G-CSFRの細胞外ドメイン(ECD)との2つの結合界面を有する。G-CSFとG-CSFRの細胞外サイトカイン受容体相同(CRH)ドメインとの間のより大きな界面は、サイトIIと呼ばれる。G-CSFとG-CSFRのN末端Ig様(Ig)細胞外ドメインとの間のより小さな界面は、サイトIIIと呼ばれる(図1を参照)。
共進化した排他的なG-CSFE:G-CSFRE変異ペア(デザイン)を作成するために、コンピュテーショナルデザインワークフローを採用した(図2を参照)。まず、サイトIIでの排他的デザインを作成した。
表2に記載のデザインは、インシリコでG-CSFE:G-CSFR(CRH)Eのペアリングに有利であるZymeCAD(商標)におけるパッキングメトリックを有し(表3を参照)、塩橋、水素結合の存在と、激しい衝突の不在など、目視検査上、インシリコにおいて満足のいく相互作用を示した(図5を参照)。また、これらのデザインは、WT G-CSFまたはG-CSFR(CRH)とのミスペアリングに対して高い選択性を示した(表3を参照)。
選択されたサイトIIデザインは、バキュロウイルスベースの昆虫細胞システムにおいて共発現させたときにサイトII複合体を形成する能力について、G-CSFE:G-CSFR(CRH)Eの形式でプルダウン実験でスクリーニングした。また、G-CSFE変異体の各デザインとG-CSFR(CRH)WTを共発現させることにより、その逆でG-CSFR(CRH)E変異体の各デザインをGCSFWTと共発現させることにより、デザインがWT受容体またはサイトカインとミスペアリング複合体を形成できるかどうかを評価した。G-CSFEのみの発現は、サイトカイン変異体の単一感染によって確認した。
簡単に言えば、サイトII G-CSFデザインと対応するペアのG-CSFR(CRH)変異体は、昆虫細胞トランスフェクションベクターに別々にクローニングした。G-CSF WT(残基1-173、表1)及び変異体を、N末端分泌シグナル及び配列
を有するC末端TEV切断可能Twin Strepタグがインフレームに存在する改変pAcGP67bトランスファーベクター(Pharmingen)にクローニングした。受容体細胞外ドメインの中では、サイトIIデザイン変異体を有するCRHドメイン(残基98-308、表1)のみが、N末端分泌シグナル及び配列
のTEV切断可能ヘキサヒスチジン(配列番号89)タグがインフレームに存在する改変pAcGP67bトランスファーベクターにクローニングされた。すべてのコンストラクトを合成し、昆虫細胞の発現のためにコドンを最適化した(Genscript)。トランスファーベクターDNAは、Midi-prep(ThermoScientific,cat.K0481)により調製し、エンドトキシンを含まず、A260/280吸光度比が1.8~2.0であった。組換えウイルス生成は、製造元(Expression Systems,California)の説明に従って付着法を用いて、組換え、線状化バキュロウイルスDNAとベクターDNAを、Spodoptera frugiperda 9(Sf9)細胞に、共トランスフェクションすることにより達成された。トランスフェクションの約1時間前に、0.46×106細胞ml-1で、健康な対数期Sf9細胞2mLを6ウェル組織培養プレート(Greiner,cat.657-160)のウェルごとに播種した。トランスフェクション混合物は以下のように調製した:100μlのトランスフェクション培地(Expression Systems,California,cat.95-020-100)を2本の滅菌1.5mlMicrofugeチューブA及びBのそれぞれに入れた。チューブAには、0.4μgの組換えBestBac 2.0Δ v-cath/chiA線状化DNA(Expression Systems,California,cat.91-002)及び2μgのベクターDNAを加えた。チューブBには、1.2μlの5XのExpress2TRトランスフェクション試薬(Expres2ION,cat.S2-55A-001)を添加した。溶液A及び溶液Bを約24℃で5分間インキュベートした後、それらを合わせて、30分間インキュベートした。インキュベーション後、800μlのトランスフェクション培地を各トランスフェクション反応物に加え、容量を1mlまで増加させた。古いESF921培地をウェルから取り除き、細胞の単層を乱さないように滴下して1mLのトランスフェクション混合液で置換した。プレート(複数可)を前後左右に穏やかに揺り動かして、トランスフェクション混合物を均一に分散させ、27℃で4時間インキュベートした。4時間後、トランスフェクション混合物を共トランスフェクションプレート(複数可)から取り出し、ゲンタマイシンを10μg/ml(cat.15750-060)で含有する2mLの新鮮なESF921昆虫細胞培養培地(Expression Systems,California、cat.96-001-01)を滴下して加えた。蒸発を防ぐため、プレートをサランラップで包み、滅菌プラスチックボックスに入れ、27℃で4~5日間インキュベートした。トランスフェクション後4日目または5日目に、P1上清を収集し、5000rpmで5分間遠心分離して清澄化し、新しい滅菌チューブに移し、光から保護して4℃で保存した。
サイトIIデザイン#6、7、8、9、15、17、30、34、35、36は、G-CSFE単独での発現を示し、G-CSFE上のTwin Strepタグを介してプルダウンされた十分に安定なペアリングしたG-CSFE:G-CSFRE複合体を形成した(図6を参照)。例えば、操作された複合体のプルダウン後のサイトIIデザイン#6は、SDS-PAGEで、そのG-CSFE変異体の約22kDaのバンドと、対応する共発現したG-CSFR(CRH)E変異体の約33kDaのバンドを示した(図6を参照)。
G-CSFに結合するサイトIII受容体Igドメインの結合活性効果は、2:2ヘテロ二量体化学量論のG-CSF:G-CSFR(Ig-CRH)複合体の形成に寄与する(図1を参照)。WTサイトIII界面を有するサイトIIにのみ存在する選択的デザインは、完全に排他的な2:2 G-CSFE:G-CSFR(Ig-CRH)E複合体を作成するには不十分であるように思われる。WTサイトカインまたは受容体へのミスペアリング結合よりも選択的であるペアリングした共進化デザインの結合を優先するために、実施例1で述べたインシリコデザインワークフローをサイトIII界面に適用し、選択的サイトIIIデザインを作成した(図2を参照)。
まず、サイトIII界面相互作用の構造解析を実施した。サイトIII界面は、55.64kcal/molのAMBERエネルギーをG-CSF:G-CSFR複合体にもたらし、692.6Å2の界面領域のサイトIIと比較して、571.5Å2の全体的に小さい界面領域を有する。サイトIII界面の詳細な検査により、サイトII界面と比較して、静電相互作用及び水素結合相互作用が少ないことが明らかである(図8を参照)。サイトIIIでの重要な相互作用は、例えば、G-CSFのE46と受容体IgドメインのR41間、さらにG-CSFのR147と受容体IgドメインのE93間の塩橋である(図9を参照)。両方の相互作用は、サイトIIIの引力AMBERエネルギー合計に対して、それぞれ15.9%と15.4%を占める。さらなる相互作用は、例えば、受容体IgドメインのQ87を介して作られ、G-CSFサイトIIIのバックボーンへの側鎖アミドとの水素結合相互作用を形成し、E46及びL49など、サイトカインの周囲の側鎖とのLennard Jones相互作用を有する。この相互作用は、サイトIIIの引力AMBERエネルギー合計の12.3%を占める。
表4に記載のデザインは、G-CSFE:G-CSFR(Ig)Eのペアリングに有利であるZymeCAD(商標)におけるインシリコパッキングメトリックを有し、WT G-CSFまたはG-CSFR(Ig)とのミスペアリングに対して高い選択性を持っている。したがって、表4に示す同じバリアントG-CSF及び受容体デザインのさまざまなサイトIII変異は、それぞれ野生型G-CSFR及びG-CSFよりも優先的な結合を示すと予測される。
2:2のヘテロ二量体の操作した複合体を介した結合及びシグナル伝達を可能にし、野生型サイトカインまたは受容体との交差反応性が低いか、または完全に消失した完全選択性デザインペアG-CSFE:G-CSF(Ig-CRH)Eを開発するために、実施例1及び3の選択したサイトII及びIIIデザインを組み合わせ(表6を参照)、インビトロで試験した。
サイトカインTSTタグを介したプルダウンを伴う共発現アッセイは、上記の実施例2で説明したように実施したが、受容体コンストラクトがG-CSFR(Ig-CRH)と呼ばれるIg及びCRHドメイン(残基2-308、表1)を含むという点で異なる。
組合せデザイン401及び402は、本デザインサイトカインが、それらの共進化した、操作された受容体をプルダウンするがWT受容体はプルダウンせず、その逆にWT G-CSFが、操作された受容体をプルダウンしなかったという点で、共発現アッセイにおいて完全に選択的であった(図10を参照)。
野生型及び操作されたサイトカイン及び受容体バリアントを作製し、それらの生物物理学的特性を比較するために、組換えタンパク質を発現させ、昆虫細胞から精製した。
G-CSFE及びG-CSFWTを、上記のようにクローン化した。組換えタンパク質の分取スケール生成は、2~4Lの健常な対数期Tni細胞において以下のように行った。2×106細胞ml-1での800mLのTniに、2ml細胞当たり20μlのサイトカインバリアントP2ウイルスを接種し、135rpmで振盪しながら27℃で70時間インキュベートした。インキュベーション後、5500rpmで15分間遠心分離することにより細胞をペレット化し、上清を2回、まず1μmのA/E型ガラス繊維フィルター(PALL,cat.61631)、続いて0.45μmのPVDFメンブレンフィルター(Sigma Aldrich,cat.HVLP04700)を通じてろ過した。プロテアーゼ阻害剤カクテルIII(Sigma Aldrich,cat.539134)を加え、上清緩衝液をHBS(20mMのHEPES pH8、150mMのNaCl)に交換し、タンジェンシャルフローで300mLに濃縮した。タンパク質を、3x3mLのb.v Streptactin-XT superflowによりバッチモードで精製し、4℃で2x1時間、1x一晩、撹拌しながらインキュベートした。溶出前に樹脂を10CVのHBS緩衝液で洗浄した。タンパク質を4x5mLのBXT溶出緩衝液で溶出した。溶出液をnanodrop A280及び還元型SDS-PAGEにより分析し、約2mLに濃縮し、TEV:タンパク質比1:80で18℃で一晩、TEVと転倒式に混合してインキュベートすることによりTST精製タグを切断した。切断されたタンパク質は、20mMのBisTris pH6.5、150mMのNaClで平衡化したSX75 16/600またはSX200 16/600サイズ排除カラム(GE Healthcare,cat.28-9893-33または28-9893-35)にロードする前に、SDS-PAGEによって確認した(図11を参照)。タンパク質含有フラクションを還元型SDS-PAGEにより分析し、プールして、nanodrop A280測定により濃度を測定した。
野生型、G-CSFE、及びG-CSFRE変異体は、還元型SDS-PAGEにより判断すると、SEC後の純度は90%を超えていた(図11及び12を参照)。デザイン401及び402のG-CSFEの1Lの培養物当たりのSEC後の収量は、それぞれ2.7mg及び1.6mgであった。デザイン401及び402のG-CSFREの1Lの生産当たりのSEC後の収量は、それぞれ1.7mg及び1.5mgであった。WT G-CSFは、1Lの培養物当たり2.1mgのSEC後の収量で精製され、WT G-CSFRは、1Lの培養物当たり3.1mgのSEC後の収量で精製された。
共進化した受容体変異体に対するデザインサイトカインの親和性を決定するために、G-CSFRWTに対するG-CSFEの親和性を測定した。また、G-CSFEとG-CSFRWTの親和性、及びG-CSFWTとG-CSFREの親和性を、SPRによりデザインのサブセット(ミスペアリングした)について測定した。
SPR結合アッセイは、Biacore T200装置(GE Healthcare,Mississauga,ON,Canada)で、PBS-T(PBS+0.05%(v/v)Tween 20)ランニング緩衝液を用いて、25℃の温度で実施された。CM5 Series Sセンサチップ、Biacoreアミンカップリングキット(NHS,EDC,及び1Mのエタノールアミン)、及び10mMの酢酸ナトリウム緩衝液は、すべてGE Healthcareから購入した.0.05% Tween20を含むPBSランニング緩衝液(PBS-T)はTeknova Inc.(Hollister,CA)から購入した。デザインは、3つの異なる固定化配向において評価した。
曲線の会合及び解離相を適合させることによって速度論的導出親和性定数(KD)を得た。WT G-CSFR(Ig-CRH)に対するWT G-CSFのKDは、1.8~2.5E-9の範囲であった。速度論的パラメータを適合させることができなかった場合、定常状態の親和性定数を導出することを試みた。これらの場合、KD変化倍率は、WT G-CSF:WT G-CSFR(Ig-CRH)ペアの定常状態から導出されたKDから算出され、表7に示される。

WTサイトカイン及び受容体と比較したG-CSFE及びG-CSFRE変異体の熱安定性を測定するために、示差走査熱量測定(DSC)を実施した。
バリアントの熱安定性は、示差走査熱量測定(DSC)により以下のように評価した:濃度1~2mg/mLの精製試料950mLを、Nano DSC(TA instruments,New Castle,DE)を用いたDSC分析に使用した。各分析の開始時に、ベースラインの安定化のために緩衝液ブランクの注入を行った。各試料を、60psiの窒素圧力下、60℃/時の速度で25~95℃で走査した。得られたサーモグラムを参照し、NanoAnalyzeソフトウェアを用いて解析し、熱安定性の指標となる融解温度(Tm)を測定した。
操作されたバリアントの熱安定性は、同じ条件及び実験設定下で測定された、操作された分子と同等の野生型分子との間の最も顕著な遷移(最高のエンタルピー)の差として報告される。測定されたWT GCSFのTmは、独立した実験間で52.2~55.4℃の間で変動し、WT G-CSFRのTmは50.5℃を示した。デザイン#15及び34を除いて試験したG-CSFE変異体は、WT G-CSFのTmとは5℃未満異なるTmを示した(表10及び図14を参照)。試験したすべての受容体変異体は、WT受容体と同じ熱安定性を示した(表10及び図14を参照)。
WT G-CSFと比較したG-CSFE変異体の単分散性を測定するために、UPLC-SECによって変異体を分析した。
SEC精製したタンパク質試料に対して、Acquity BEH125 SECカラム(4.6x150mm,ステンレススチール,1.7μm粒子)(Waters LTD,Mississauga,ON)を30℃に設定し、PDA検出器を備えたAgilent Technologies 1260 infinity IIシステムに取り付けてUPLC-SECを実施した。分析時間は、150mMのNaCl、20mMのHEPES pH8.0または150mMのNaCl、20mMのBisTris pH6.5のランニング緩衝液を用いて、流速0.4mL/分で7分間のランタイムで構成された。溶出は、210~500nmの範囲でUV吸光度によりモニターし、クロマトグラムは280nmで抽出した。ピーク積分は、OpenLAB(商標)CDS ChemStation(商標)ソフトウェアを使用して実施した。
WT G-CSF及びデザイン34、35、及び130のG-CSFE変異体は、100%単分散性であった(表11を参照)。変異体8、9、15、117、135は、65.3~79.5%のより低い単分散性を示した。デザイン#134サイトカインは、pH8.0で57.3%の単分散性を示したが、pH6.5では86.6%の単分散性に改善した。移動相のより低いpHでの単分散性の改善は、pIのシフト(例えば、WT G-CSFでの算出されたpI5.41からデザイン#134のG-CSFEでの算出されたpI8.35へ)に起因する可能性がある。
デザインG-CSFEサイトカイン変異体がデザインG-CSFR(Ig-CRH)E受容体変異体を介してシグナル伝達し、免疫細胞増殖を引き起こすことができるかどうかを調べるために、gp130膜貫通(TM)ドメイン及び細胞内シグナル伝達ドメイン(ICD)、ならびにIL-2Rβ細胞内シグナル伝達ドメインに融合したG-CSFR ECDを使用して、一本鎖キメラG-CSF受容体(G-CSFRWT-ICDgp130-IL-2Rβ)を構築した。また、ヘテロ二量体受容体として共発現するように設計された2つのサブユニットからなるキメラG-CSFRを利用した:1)G-CSFRWT-ICDIL-2RβサブユニットはIL-2RβTM及びICDに融合したG-CSFR ECDからなり、2)G-CSFRWT-ICDγcサブユニットは共通γ鎖(γc,IL-2Rγ2Rγ)TM及びICDに融合したG-CSFR ECDからなる。
一本鎖キメラ受容体構築物は、G-CSFRシグナルペプチド及びECD、次いでgp130 TM及び部分ICD、ならびにIL-2Rβ部分ICDを含むように設計した(表12)。ヘテロ二量体キメラ受容体構築物は、以下を含むように設計した:1)G-CSFRシグナルペプチド及びECD、次いでIL-2RβTM及びICD(表13);ならびに2)G-CSFRシグナルペプチド及びECD、次いでγc TM及びICD(表14)。キメラ受容体構築物をレンチウイルストランスファープラスミドにクローニングし、構築物配列をサンガーシークエンシングにより確認した。トランスファープラスミド及びレンチウイルスパッケージングプラスミド(psPAX2、pVSVG)を、以下のようにレンチウイルスパッケージング細胞株HEK293T/17細胞(ATCC)に共トランスフェクションさせた:細胞を、10%ウシ胎児血清及びペニシリン/ストレプトマイシンを含有するDMEM中で一晩プレーティングし、トランスフェクションの2~4時間前に培地交換した。プラスミドDNA及び水をポリプロピレンチューブ内で混合し、CaCl2(0.25M)を滴下して加えた。2~5分間インキュベートした後、2xのHEPES緩衝生理食塩水(0.28MのNaCl、1.5mMのNa2HPO4、0.1MのHEPES)と1:1で混合することにより、DNAを沈殿させた。沈殿したDNA混合物を細胞上に添加し、37℃、5%のCO2で一晩インキュベートした。翌日、HEK293T/17培地を交換し、細胞をさらに24時間インキュベートした。翌朝、プレートから細胞上清を回収し、軽く遠心分離して破片を除去し、0.45μmフィルターで濾過した。上清をBeckman Optima L-XP超遠心機のSW-32Tiローターを用いて25,000rpmで90分間回転させた。上清を除去し、ペレットを適当量のOpti-MEM培地に再懸濁させた。ウイルスの連続希釈液をBAF3細胞(10%ウシ胎児血清、ペニシリン、ストレプトマイシン、及び100IU/mlのhIL-2を含有する増殖用RPMI)に添加することにより、ウイルス力価を測定した。形質導入の48~72時間後、細胞を抗ヒトG-CSFR APCコンジュゲート抗体(1:50希釈)及びeBioscience(商標)Fixable Viability Dye eFluor(商標)450(1:1000希釈)とともに4℃で15分間インキュベートして洗浄し、Cytek AuroraまたはBD FACS Caliburフローサイトメーターで分析した。この方法により測定した推定力価を用いて、32D-IL-2Rβ細胞株(10%ウシ胎児血清、ペニシリン、ストレプトマイシン、及び300IU/mlのhIL-2を含有するRPMIにおいて増殖)に、MOI0.5でキメラ受容体構築物をコードするレンチウイルス上清を形質導入した。形質導入は、適切な量のウイルス上清を細胞に添加し、24時間インキュベートし、細胞培地を交換することにより行った。形質導入から3~4日後、上記のようにフローサイトメトリーによりヒトG-CSFRの発現を確認した。BrdUアッセイを行う前に、細胞をG-CSFWT中で約14~28日間増殖させた。
BrdUアッセイでは、一本鎖キメラ受容体構築物G-CSFRWT-ICDgp130-IL-2Rβまたはヘテロ二量体受容体構築物G-CSFRWT-ICDIL-2Rβ+G-CSFRWT-ICDγcでトランスフェクトした細胞は、G-CSFWT(30ng/ml)に反応してhIL-2(300IU/ml)と比較して同等または優れた増殖が示された。細胞は、サイトカインの非存在下では増殖しなかった(図15を参照)。
SPRまたは共発現アッセイによって判断されるように十分に選択的であったサイトII/IIIの組み合わせデザインを、32D-IL-2Rβ細胞の増殖を誘発する能力についてインビトロで試験した。
デザイン137の点変異を、表12~14に記載の構築物に導入した。G-CSFR137-ICDgp130-IL-2Rβ(ホモ二量体)またはG-CSFR137-ICDIL-2Rβ+G-CSFR137-ICDγc(ヘテロ二量体)構築物のクローニング及び発現は、上記と同じ手順に従った。BrdUアッセイを行う前に、細胞をG-CSF137中で約14~28日間増殖させた。
BrdUアッセイでは、一本鎖キメラ受容体構築物G-CSFR137-ICDgp130-IL-2Rβまたはヘテロ二量体受容体構築物G-CSFR137-ICDIL-2Rβ+G-CSFR137-ICDγcで形質導入された細胞は、G-CSF137(30ng/ml)においてhIL-2(300IU/ml)と比較して同等または優れた増殖が示された。細胞はサイトカインの非存在下では増殖せず、G-CSFWT(30ng/ml)において劣った増殖を示した(図16を参照)。
続いて、32D-IL-2R2Rβ細胞においてペアリングしたシグナル伝達を回復することができたサイトII/IIIの組み合わせデザインを、一本鎖キメラ受容体構築物G-CSFRWT-ICDgp130-IL-2Rβまたはヘテロ二量体受容体構築物G-CSFRWT-ICDIL-2Rβ+G-CSFRWT-ICDγcで形質導入された32D-IL-2R2Rβ細胞の増殖を誘発する能力について試験した。

32D-IL-2Rβ細胞の操作されたサイトカイン受容体複合体を介して増殖シグナル伝達を回復することができ、G-CSFWTまたはWT-GCSFR-ICD-IL2の結合を介して有意にシグナル伝達しなかったサイトII/IIIの組み合わせデザインを、G-CSFWTまたはG-CSF137に応答して下流のシグナル伝達分子を活性化させる能力についてウエスタンブロットによって評価した。
G-CSFRWT-ICDgp130-IL-2Rβ(ホモ二量体)またはG-CSFRWT-ICDIL-2Rβ+G-CSFRWT-ICDγc(ヘテロ二量体)構築物のクローニング及び発現は、上記と同じ手順に従った。ウエスタンブロットアッセイを実施する前に、細胞をG-CSF137またはG-CSFWTで約14~28日間増殖させた。非形質導入細胞はIL-2において維持した。
非形質転換32D-IL-2Rβ細胞において、IL-2のみでの刺激に応答した活性化IL-2R関連シグナル伝達分子を検出した。G-CSFRWT-ICDgp130-IL-2RβまたはG-CSFRWT-ICDIL-2Rβ+G-CSFRWT-ICDγcのいずれかを発現する32D-IL-2Rβ細胞において、IL-2またはG-CSFWTに応答した活性化シグナル伝達分子の同様のパターンが観察された。G-CSFRWT-ICDgp130-IL-2Rβを発現するまたはG-CSFRWT-ICDIL-2Rβ+G-CSFRWT-ICDγcを発現する32D-IL-2Rβ細胞のG-CSF137刺激に応答したIL-2R関連シグナル伝達分子の活性化は観察されなかった。G-CSFR137-ICDgp130-IL-2RβまたはG-CSFR137-ICDIL-2Rβ+G-CSFR137-ICDγcを発現する32D-IL-2Rβ細胞において、IL-2またはG-CSF137に応答した活性化シグナル伝達分子の同様のパターンが観察された。G-CSFR137-ICDgp130-IL-2Rβを発現するまたはG-CSFR137-ICDIL-2Rβ+G-CSFR137-ICDγcを発現する32D-IL-2Rβ細胞のG-CSFWT刺激に応答したIL-2R2R2R関連シグナル伝達分子の活性化は観察されなかった。
初代細胞及び細胞株:レンチウイルスパッケージング細胞株HEK293T/17(ATCC)は、10%ウシ胎児血清及びペニシリン/ストレプトマイシンを含有するDMEM中で培養した。BAF3-IL-2Rβ細胞を、BAF3細胞株へのヒトIL-2Rβサブユニットの安定的トランスフェクションによって事前に生成し、10%ウシ胎児血清、ペニシリン、ストレプトマイシン、及び100IU/mlのヒトIL-2(hIL-2)(PROLEUKIN(登録商標),Novartis Pharmaceuticals Canada)を含有するRPMI-1640において増殖させた。32D-IL-2Rβ細胞株を、32D細胞株へのヒトIL-2Rβサブユニットの安定的トランスフェクションによって事前に生成し、10%ウシ胎児血清、ペニシリン、ストレプトマイシン、及び300IU/mlのhIL-2、または示された他のサイトカインを含有するRPMI-1640において増殖させた。ヒトPBMC由来T細胞(Hemacare)を、3%ヒトAB血清(Sigma-Aldrich,H4522)、及び300IU/mlのhIL-2、または示された他のサイトカインを含有するTexMACS(商標)培地(Milenyi Biotec,130-097-196)において増殖させた。ヒト腫瘍関連リンパ球(TAL)を、T細胞培地(以下の50:50混合物)において、初代腹水試料を14日間培養することにより生成した:1)10%ウシ胎児血清、50uMβ-メルカプトエタノール、10mMのHEPES、2mMのL-グルタミン、ペニシリン、ストレプトマイシン含有RPMI-1640;及び2)最終濃度3000IU/mlのhIL-2を含有するAIM V(商標)培地(ThermoFisher,12055083)。この高用量のIL-2増殖の後、TALを300IU/mlのhIL-2または示された他のサイトカインを含有するT細胞培地において培養した。レトロウイルスパッケージング細胞株Platinum-E(Cell Biolabs,RV-101)を、10%FBS、ペニシリン/ストレプトマイシン、プロマイシン(1mcg/ml)、及びブラストサイジン(10mcg/ml)を含有するDMEMにおいて培養した。
PBMC由来T細胞または腫瘍関連リンパ球(TAL)に、図1に示すキメラ受容体構築物をコードするレンチウイルスで形質導入し、細胞を洗浄した後、示されたサイトカインで再プレーティングした。細胞を3~4日ごとに計数した。G/γcは、そのN末端にMycエピトープをタグ付けされ(Myc/G/γc)、G/IL-2Rβは、そのN末端にFlagエピトープをタグ付けされ(Flag/G/IL-2Rβ);これらのエピトープタグは、フローサイトメトリーによる検出を補助し、受容体の機能には影響を与えない。予想通り、すべてのT細胞培養物は陽性対照サイトカインであるIL-2(300IU/ml)に反応して増殖を示した。G-CSF(100ng/ml)で刺激した後、PMBC由来のT細胞及びG2R-1キメラサイトカイン受容体を発現するTALについてのみ、増殖が観察された(図22)。レンチウイルスの形質導入効率は、T細胞の100%未満が、示されたキメラサイトカイン受容体を発現するように100%未満であったことに留意されたい。これは、IL-2と比較してG2R-1によって媒介される増殖率が低いことの原因であると考えられる。同様に、増殖の増加は、G-CSFRキメラ受容体サブユニットG2R-1及びG2R-2を発現し、G-CSFで刺激された32D-IL-2Rβ細胞(ヒトIL-2Rβサブユニットを安定して発現する)において観察された(図21)。T細胞とは対照的に、G/IL-2Rβキメラ受容体サブユニットのみを発現する32D-IL-2Rβ細胞は、G-CSFに反応して増殖した(図2);G-CSF誘導性増殖は、G/γcキメラ受容体サブユニットのみを発現する32D-IL-2Rβ細胞では見られなかった(図2)。
G2R-2キメラサイトカイン受容体をコードするレンチウイルスベクター(図23及び25に概略的に示す)を形質導入した後の32D-IL-2Rβ細胞株、PBMC由来のヒトT細胞及びヒト腫瘍関連リンパ球に対してフローサイトメトリーを行い、細胞が細胞表面にG-CSFR ECDを発現するかどうかを測定した。G-CSFR陽性細胞は、すべての形質導入された細胞タイプにおいて検出された(図26)。別の実験では、G/IL-2Rβ、G2R-1、及びG2R-2キメラ受容体を発現する32D-IL-2Rβ細胞は、フローサイトメトリーによりG-CSFR ECDに対して陽性であった(図21B~Dの下段)。
ヒトPBMC由来のT細胞及びヒト腫瘍関連リンパ球に、G2R-2受容体構築物(図4及び図6)をレンチウイルスを形質導入し、洗浄し、示されたサイトカインで再プレーティングした。いくつかの実験では、T細胞もTransActで刺激することにより定期的に再活性化させた。生細胞は3~4日ごとに計数した。PMBC由来のT細胞(図27A)及び腫瘍関連リンパ球(図27B、Cの2つの独立した実験)の増殖は、G2R-2キメラ受容体を発現する細胞においてG-CSF(100ng/ml)で刺激した後に観察されたが、非形質導入細胞では観察されなかった。
CD4選択及びCD8選択ヒトT細胞に、G2R-2をコードするレンチウイルスベクター(図25)を形質導入するか、または示された場所では形質導入しないままにした。細胞を洗浄し、示されたサイトカインで再プレーティングして3~4日ごとに計数した。G2R-2を発現するCD4またはCD8選択TALの増殖は、G-CSF(100ng/ml)またはIL-2(300IU/ml)での刺激後に観察されたが、添加したサイトカインがない場合(培地のみ)では観察されなかった(図28及び図29)。図28では、各線は5つの患者試料のうちの1つからの結果を表す。
G-CSFによる刺激時の、G2R-2または一本鎖G/IL-2Rβ(G2R-1の構成要素)を発現する初代マウスT細胞とモック形質導入細胞の増殖を評価するために、BrdU取り込みアッセイを実施した。すべての細胞を、アッセイ前にIL-2で3日間増殖させた。G2R-2またはG/IL-2Rβの細胞表面発現は、フローサイトメトリーにより確認した(図32A)。示されたように、次に細胞をIL-2(300IU/ml)、野生型G-CSF(100ng/ml)、またはサイトカインなしでプレーティングした。G-CSFによる刺激後の細胞周期進行の促進は、非形質導入細胞または一本鎖G/IL-2Rβを発現する細胞よりもG2R-2を発現する細胞において観察された(図32B、C)。パネルB及びCは、それぞれ、すべての生細胞またはG-CSFR+細胞についての結果を示す。
キメラサイトカイン受容体が、実際にIL-2と同様のサイトカインシグナル伝達を活性化させることができることを確認するために、サイトカイン受容体がさまざまなシグナル伝達分子を活性化させる能力を評価した。G2R-2を発現する腫瘍関連リンパ球及びPBMC由来T細胞は、あらかじめG-CSFにおいて、非形質導入細胞は、あらかじめIL-2において増殖させた。細胞を洗浄し、次にIL-2(300IU/ml)、野生型G-CSF(100ng/ml)、またはサイトカインなしで刺激し、示されたシグナル伝達分子に対する抗体を用いて細胞溶解物にウエスタンブロットを行った(図33)。パネルA及びBはTALの結果を示し、パネルCはPBMC由来T細胞の結果を示す。G2R-2を発現するT細胞は、G-CSFにより刺激すると、非形質導入細胞または形質導入細胞のIL-2刺激後に見られるのと同程度に(G-CSFがJak2リン酸化を誘発したのに対し、IL-2はJak3リン酸化を誘発したという予想される例外を除いて)、IL-2関連シグナル伝達分子を活性化した。
キメラサイトカイン受容体G2R-2または一本鎖G/IL-2Rβ(G2R-1由来)が、サイトカインシグナル伝達を活性化させることができるかどうかを評価するために、これらのサイトカイン受容体のさまざまなシグナル伝達分子を活性化させる能力を、G2R-2またはG/IL-2Rβを発現するマウス初代T細胞とモック形質導入細胞の細胞溶解物のウエスタンブロットによって評価した。すべての細胞を、アッセイ前にIL-2で3日間増殖させた。その後、細胞を洗浄し、IL-2(300IU/ml)、野生型G-CSF(100ng/ml)、またはサイトカインなしで刺激した。G2R-2を発現するT細胞は、G-CSFにより刺激すると、非形質導入細胞または形質導入細胞のIL-2刺激後に見られるのと同程度に(G-CSFがJak2リン酸化を誘発したのに対し、IL-2はJak3リン酸化を誘発したという予想される例外を除いて)、IL-2関連シグナル伝達分子を活性化した(図34)。対照的に、G/IL-2Rβは、G-CSFへの曝露時にサイトカインシグナル伝達を活性化しなかった。
キメラサイトカイン受容体を発現する細胞が直交G-CSFに応答して選択的に活性化され得るかどうかを判定するために、32D-IL-2Rβ細胞または初代マウスT細胞に、野生型G-CSFR ECDを含むキメラ受容体G2R-1及びG2R-2(G2R-1 WT ECD、G2R-2 WT ECD)と、アミノ酸置換R41E、R141E、及びR167Dを有するG-CSFR ECDを含むキメラ受容体G2R-1及びG2R-2(G2R-1 134 ECD、G2R-2 134 ECD)を形質導入した。細胞を、IL-2、野生型G-CSF、またはG2R-1 134 ECD、及びG2R-2 134 ECDに結合できるが野生型G-CSFRへの結合が著しく低下した直交G-CSF(130 G-CSF)のいずれかで刺激した。BrdU取り込みアッセイを行い、サイトカイン刺激による細胞周期の進行の促進能力を評価した(図3)。G2R-2 134 ECDを発現する32D-IL-2Rβ細胞は、130 G-CSF(アミノ酸置換E46R、L108K、及びD112Rを保有;30ng/ml)で刺激すると細胞周期の進行を示したが、野生型G-CSF(30ng/ml)で刺激すると細胞周期の進行は起こらなかった。操作されたサイトカイン:受容体ECDペアの直交性は、「クリスクロス」増殖アッセイで初代マウスT細胞を刺激することによってさらに示され、このアッセイでは、WT、130、134、304、または307 ECDを有するG2R-3(図23)を発現する細胞がWT、130、304または307サイトカイン(100ng/ml)で刺激された(図36)。130 ECDは、アミノ酸置換:R41E及びR167Dを有する。304 ECDは、アミノ酸置換:R41E、E93K、及びR167Dを有し、304サイトカインは、アミノ酸置換:E46R、L108K、D112R、及びR147Eを有する。307 ECDは、アミノ酸置換:R41E、D197K、D200K、及びR288Eを有し、307サイトカインは、アミノ酸置換:S12E、K16D、E19K、及びE46Rを有する。図36のパネルA及びBは、反復実験を表す。
キメラサイトカイン受容体を発現する細胞が直交G-CSFに応答して細胞内サイトカインシグナル伝達イベントを選択的に活性化し得るかどうかを判定するために、32D-IL-2Rβ細胞に、野生型G-CSFR ECDを含むキメラ受容体G2R-1及びG2R-2(G2R-1 WT ECD及びG2R-2 WT ECD)と、アミノ酸置換R41E、R141E、及びR167Dを有するG-CSFR ECDを含むキメラ受容体G2R-1及びG2R-2(G2R-1 134 ECD、G2R-2 134 ECD)を形質導入した。細胞を、IL-2(300IU/ml)、野生型G-CSF(30ng/ml)、またはG2R-1 134 ECD、G2R-2 134 ECDに結合できるが野生型G-CSFRへの結合が著しく低下した直交G-CSF(130 G-CSF-E46R_L108K_D112R;30ng/ml)のいずれかで刺激した。サイトカインへの曝露時にサイトカインシグナル伝達を活性化させる細胞の能力を評価するために、細胞溶解物に対してウエスタンブロットを実施した(図37)。G2R-2 134 ECDを発現する細胞は、野生型G-CSFではなく130G-CSFで刺激時のサイトカインシグナル伝達の証拠を示した。さらに、G2R-2 WT ECDを発現する細胞は、130G-CSFで刺激するとサイトカインシグナル伝達を活性化させることができなかった。
G2R-3キメラ受容体が初代ヒトT細胞においてG-CSFで刺激するとサイトカインシグナル伝達関連イベントを促進できるかどうかを判定するために、TALにG2R-3をコードするレンチウイルスベクターを用いて形質導入した。T細胞増殖アッセイを実施して、IL-2(300IU/ml)、野生型G-CSF(100ng/ml)、またはサイトカインなしでの刺激時における細胞の増殖を試験した。生細胞は3~4日ごとに計数した。非形質導入対応物とは対照的に、G2R-3を発現する初代TALは、G-CSFに応答して培養液中で増殖させた(図40A)。G-CSFによる刺激時にサイトカインシグナル伝達イベントが活性化されたかどうかを判定するために、細胞溶解物にウエスタンブロットを行い、細胞内シグナル伝達を評価した。細胞を増殖アッセイから回収し、洗浄した後、IL-2(300IU/ml)または野生型G-CSF(100ng/ml)で刺激した。G2R-3を発現する初代TALは、G-CSFがJak2リン酸化を誘発したのに対し、IL-2がJak3リン酸化を誘発したという予想される例外を除いて、G-CSFに応答してIL-2関連シグナル伝達イベントを実証した(図40B)。
304(R41E_E93K_R167D)または307(R41E_D197K_D200K_R288E) ECDを有するキメラサイトカイン受容体G2R-3が直交リガンド130、304、または307 G-CSFによる刺激に応答して増殖及び拡大増殖を誘発できるかどうかを評価した。初代PBMC由来ヒトT細胞にGR-3 304 ECDまたはG2R-3 307(R41E_D197K_D200K_R288E)ECDをコードするレンチウイルスベクターを用いて形質導入した。IL-2(300IU/ml)、304 G-CSF(100ng/ml)、307 G-CSF(100ng/ml)、またはサイトカインなしで培養したときの細胞の増殖倍率を評価するためにT細胞増殖アッセイを実施した。生細胞は3~4日ごとに計数した。G2R-3 304 ECDを発現するT細胞を、IL-2または304 G-CSFに応答して培養液中で増殖させた(図43A)。G2R-3 307 ECDを発現するT細胞を、IL-2または307 G-CSFに応答して培養液中で増殖させた(図43B)。非形質導入T細胞のみを、IL-2に応答して増殖させた(図43C)。
キメラサイトカイン受容体構築物が初代ヒト腫瘍関連リンパ球(TAL)の表面上で発現できるかどうかを評価するために、TALにG21R-1、G21R-2、G12R-1、及びG2R-3キメラ受容体をコードするレンチウイルスベクターを用いて形質導入し、細胞をフローサイトメトリーによって細胞表面上のG-CSFR ECD発現について試験した(図39)。4つのキメラサイトカイン受容体デザインすべてについて、G-CSFR ECD陽性細胞が検出された。
G12R-1及びG21R-1キメラ受容体が初代T細胞の表面上で発現することができるかどうかを判定するために、初代マウスT細胞にG12R-1及びG21R-1キメラ受容体をコードするレトロウイルスベクターを用いて形質導入し、フローサイトメトリーにより分析した(図45)。
G21R-1及びG21R-2構築物が初代細胞においてサイトカインシグナル伝達イベントを誘発することができるかどうかを判定するために、初代PBMC由来ヒトT細胞に、G21R-1またはG21R-2キメラサイトカイン受容体をコードするレンチウイルスベクターを用いて形質導入した。細胞をphospho-STAT3(p-STAT3)特異的抗体で細胞内染色し、フローサイトメトリーにより評価し、STAT3活性化の尺度であるSTAT3リン酸化の程度を測定した(図46)。G-CSF(100ng/ml)で刺激すると、G21R-1またはG21R-2のいずれかで形質導入したG-CSFR陽性細胞のサブセットにおいて、リン酸化STAT3を発現する細胞数が増加した。対照的に、G-CSFR陰性(すなわち非発現)細胞は、G-CSFで刺激してもリン酸化STAT3の増加を示さなかったが、IL-21で刺激すると増加した。
キメラサイトカイン受容体G21R-1がサイトカインシグナル伝達イベントを活性化できるかどうかを判定するために、初代マウスT細胞にG21R-1をコードするレトロウイルスベクターを用いて形質導入し、フローサイトメトリーにより評価して、G-CSFによる刺激時にリン酸化STAT3を検出した。生細胞をCD8またはCD4でゲーティングし、サイトカインなし、IL-21(1ng/ml)、またはG-CSF(100ng/ml)での刺激後、phospho-STAT3陽性染色細胞の割合をCD8及びCD4細胞集団について測定した。G-CSFで刺激すると、G21R-1を発現する細胞(非形質導入細胞ではない)は、リン酸化STAT3の量の増加を示した(図47A及び47B)。G-CSFで刺激するとG21R-1またはG12R-1を発現する細胞の細胞内サイトカインシグナル伝達を評価するためにウエスタンブロットを実施した。予想通り、G21R-1を発現し、G-CSFで刺激された細胞は、STAT3のリン酸化の増加を示し、phospho-STAT4及びphospho-STAT5においてわずかに増加した(図47C)。また予想通り、G12R-1を発現する細胞では、G-CSFに応答して、STAT4の強いリン酸化が見られた。G-CSFは、モック形質導入細胞においてシグナル伝達イベントを誘発しなかった。(G12R-1群では、陽性対照(hIL-12 10ng/ml)がシグナル伝達イベントを誘発しなかったと思われることに留意されたい;これは、マウスIL-12RへのヒトIL-12の結合が不十分なためである可能性がある。)
キメラサイトカイン受容体によって媒介されるサイトカインシグナル伝達イベント及び細胞増殖を評価するために、初代マウスT細胞にG2R-2、G2R-3、G7R-1、G21/7R-1、G27/2R-1、G21/2R-1、G12/2R-1、またはG21/12/2R-1をコードするレトロウイルスベクターを用いて形質導入した。G-CSFによる刺激時の細胞周期の進行を評価するために、BrdU取り込みアッセイを実施した。細胞を回収し、洗浄した後、IL-2(300IU/ml)、野生型G-CSF(100ng/ml)、またはサイトカインなしにおいて再プレーティングした。G-CSF誘導性細胞周期の進行は、G2R-2、G2R-3、G7R-1、G21/7R-1、もしくはG27/2R-1(図48A、48B)、またはG21/2R-1、G12/2R-1、もしくはG21/12/2R-1を発現する初代マウスT細胞において見られた(図49A、49B)。G-CSFR ECDの発現は、フローサイトメトリーによっても検出可能であった(図48C、49C)。
134 ECDを有するキメラサイトカイン受容体G12/2R-1が、直交リガンド130 G-CSFでの刺激に応答して増殖及び拡大増殖を誘発できるかどうかを判定するために、初代PBMC由来ヒトT細胞にG12/2R-1 134 ECDをコードするレンチウイルスベクターを用いて形質導入した。IL-2(300IU/ml)、130 G-CSF(100ng/ml)、またはサイトカインなしで培養したときの細胞の増殖倍率を評価するためにT細胞増殖アッセイを実施した。生細胞は4~5日ごとに計数した。G12/2R-1 134 ECDを発現する初代ヒトT細胞は、IL-2または130 G-CSFに応答して培養液中で増殖したが(図50A)、培地のみでは限定的な一過性の増殖を示した。
細胞内シグナル伝達イベントを評価するために、初代PBMC由来ヒトT細胞に、G2R-3 304 ECDまたはG12/2R-1 304 ECDをコードするレンチウイルスベクターを用いて形質導入した。304 G-CSF(100ng/mL)、IL-2(300IU/mL)、IL-2、及びIL-12(10ng/mL)、または培地のみで刺激時の、G2R-3 304 ECDもしくはG12/2R-1 304 ECDを発現する細胞、または非導入細胞における細胞内サイトカインシグナル伝達を評価するためにウエスタンブロットを実施した。形質導入されたT細胞と非形質導入T細胞の両方において、STAT5の強いリン酸化が、IL-2+IL-12、またはIL-2のみでの刺激のいずれかに応答して検出された(図53)。STAT4の強いリン酸化は、IL-2+IL-12の両方による刺激に応答して検出されたが、STAT4の弱いリン酸化のみが、IL-2のみでの刺激に応答して検出された。G2R-3 304 ECDを発現する細胞では、STAT4の弱いリン酸化及びSTAT5の強いリン酸化が304 G-CSFでの刺激に応答して検出され、IL-2のみに応答して見られるパターンに類似していた。G12/2R-1 304 ECDを発現する細胞では、STAT4及びSTAT5の強いリン酸化が304 G-CSFの刺激に応答して検出され、IL-2+IL-12に対する応答に見られるパターンに類似していた。非形質導入T細胞は、304 G-CSFに対する応答を示さなかった。
