Przedmiotem wynalazku jest sposób wytwarzania nowego 4-(4-bifenylilo)-butanolu-1 o wzorze 1.Wedlug wynalazku nowy zwiazek wytwarza sie przez redukcje zwiazku o wzorze 2 za pomoca katalitycz¬ nie aktywowanego wodoru.Jako katalizatory stosuje sie, np. katalizatory z metali szlachetnych, zwlaszcza pallad na weglu kostnym.Redukcje prowadzi sie na ogól w temperaturze 0—100°C, zwlaszcza w temperaturze pokojowej, pod cisnieniem wodoru 1—100 atm, korzystnie 1—10 atm w obecnosci odpowiedniego rozpuszczalnika, np. alkoholu, estru kwasu karboksylowego lub nizszego kwasu karboksylowego. Jako substancje porównawcza stosunkowo fenylobutazon w bada¬ niach na dzialanie przeciwwysiekowe wobec obrzeku kaolinowego i karagenowego, wywolanego na tylnych lapach szczurów oraz na dzialanie wrzodotwórcze i ostra toksycznosc po dawkach badanej substancji, stosowanej doustnie.2 91409 a) Obrzek kaolinowy na tylnej lapie szczura.Wyzwolenie obrzeku nastapilo wedlug wskazówek Hillebrecht'a (Arzneimittel-Forsch. 4,607 (1954) przez podpodeszwowe wstrzykniecie 0,05 ml 10% zawiesiny kaolinu w 0,85% roztworze NaCI.Pomiar grubosci lapy przeprowadzono metoda Doepfner'a i Cerletti'ego (Int. Przez graficzna ekstrapolacje procentowych wartosci zahamowania obrzeku, okreslonych z róznych dawek, oznaczono dawke, któraprowadzi do 35% zmniejszenia obrzeku (ED35). b) Obrzek karagenowy na tylnej lapie szczura.Wyzwolenie obrzeku wywolanego karagenem nastepowalo wedlug wskazówek Winter'a i inn. (Proc. Soc. exp. itfoL lyied. tli, 544 (1962), pod podpodeszwowym wstrzyknieciu 0,05 ml 1% roztworu karagenu w 85% Roztworze NaCI. Badana substancje wprowadzono na 60 minut przed wywolaniem obrzeku. Dla oceny dzialania hamujacego obrzek oznaczono wartosci pomiarowe, otrzymane po 3 godzinach od wywolania obrzeku.Pozostalaszczególy postepowania byly analogiczne do opisanych przy obrzeku wywolanym kaolinem. i J.."'\ ,c) Dzialanie wrzodotwórcze.Badanie na dzialanie wrzodotwórcze prowadzono na szczurach FW 49 obu plci (1 :1), o ciezarze 130—150 g. Zwierzeta otrzymaly badana na dzialanie wrzodotwórcze substancje raz dziennie w ciagu 3 kolejno nastepujacych dni, w postaci roztartej z tyloza mieszaniny, za ppmoca sondy przelykowo-zoladkowej. W 4 godziny po ostatnim aplikowaniu zwierzeta zabito i badano sluzówka zoladka i dwunastnicy na owrzodzenia. Ze skladu procentowego zwierzat, które po róznych dawkach wykazaly co najmniej jeden wrzód, obliczono wartosc ED50 metoda LitchfiekTa i Wilcoxon'a (J. Pharmacol. exp. Therap. 96, 99 (1945)). d) ostra toksycznosc.Ostra toksycznosc LD50 oznaczono po podawaniu doustnym samcom i samicom (1:1) szczurów FW49 o przecietnym ciezarze 135 g. Podawano substancje roztarta w tylozie. Obliczenia LD50 prowadzono, o ile to bylo mozliwe wedlug LitchfiekTa i Wilcoxon'a,z procentowego skladu zwierzat, które po róznych dawkach padly wciagu 14dni. e) Wskaznik terapeutyczny.Wskaznik terapeutyczny jako miernik zakresu terapeutycznego oblicza sie przez otrzymanie ilorazu zwartosci ED50 oznaczonej dla wlasciwosci wrzódotwórczej wzglednie zwartosci LD50 (doustnie), oznaczonej na szczurach oraz zwartosci ED35 oznaczonej w badaniu dzialania przeciwwysiekowego (test na obrzek kaolinowy i karagenowy).Wyniki otrzymane w tych badaniach przedstawione sa w tablicy 1 i 2.Nowa substancja przewyzsza znany fenylobutazon pod wzgledem dzialania przeciwzapaleniowego.Toksycznosc i czynnosc wrzodotwórcza nowej substancji nie zwieksza sie w takim stopniu, jakiego nalezaloby sie spodziewac, sadzac ze wzrostu czynnosci przeciwzapelniowej. Dodaje sie najpierw nieco octanu etylu, a nastepnie wody lodowatej, zakwasza 50% kwasem siarkowym, alkalizuje dodatkiem lugu sodowego i ekstrahuje eterem. Roztwór eterowy przemywa sie woda, suszy nad siarczanem sodowym i uwalnia od rozpuszczalnika. Pozostalosc rozciera sie z eterem naftowym i przekrystalizowuje z ukladu cykloheksan/octan etylu. Otrzymuje sie 18,5 (76,5% wydajnosci teoretycznej) bezbarwnego 1 -(4-bifenylilo)-butanodiolu-1,4 o temperaturze topnienia 79—80°C. b) 4-(4-bifenylilo)-butanol-1. 9,68 g (0,04 mola) 1-(4-bifenylilo)-butanodiolu-1,4 (temperatura topnienia 79-80°C) rozpuszcza sie w 100 ml metanolu, dodaje 1 g palladu na wegiu jako katalizatora i uwodornia w temperaturze pokojowej przy 5 atm cisnienia, az zostanie pobrana wyliczona ilosc wodoru. Katalizator odciaga sie, z przesaczu oddestylowuje sie rozpuszczalnik, przy czym otrzymuje sie stala pozostalosc, która przekrystalizowuje sie z eteru naftowego i malej ilosci octanu etylu. Otrzymuje sie 8,32 g (93% wydajnosci teoretycznej) bezbarwnych krysztalów 4-/4-bifenylilo)-butanolu-1, o temperaturze topnienia 75—76°C. PL
