Die Erfindung betrifft das 4-(4-Biphenylyl)-1-butanol der Formel
CH;, CH- OH
(I)
und Verfahren zu seiner Herstellung.
Die neue Verbindung läßt sich nach folgenden Verfahren herstellen:
a. Durch Reduktion von Verbindungen der allgemeinen Formel II,
O~O~c"2"~cH~cH
(H)
in der B die Hydroxy-·, eine beliebige Alkoxy-, Aralkoxy-, Aryloxy-
oder Acyloxygruppe oder ein Halogenatom bedeutet.
Die Reduktion erfolgt mittelr; komplexer Hydride oder mit katalytisch
aktivierten Wasserstoff.
Als komplexe Hydride kommen bevorzugt Lithiumaluminiumhydrid",
Lithiumborhydrid, Natrium-bis-(2-methoxyäthoxy)-dihydroaluminat,
Natriumaluminiumhydrid oder Natriumborhydrid, letzteres in Anwesenheit von wasserfreiem Aluminiumchlorid oder von Bortrifluorid,
in Frage. Verbindungen der allgemeinen Formel Tif in der B ein
Halogenatom darstellt, können auch mit Natriumborhydrid allein reduziert werde.n.
Die Reduktion erfolgt in einem geeigneten Lösungsmittel wie zum Beispiel Tetrahydrofuran, Äther, Dioxan, Dimethoxyäthan,
Diäthylenglykoldimethylather, Benzol oder Gemischen derselben,
vorzugsweise bei Raumtemperatur oder bei Temperaturen bis zum Siedepunkt des verwendeten Lösungsmittels.
Die neue Verbindung der Formel I läßt sich, wie bereits erwähnt, auch durch Reduktion mittels katalytisch aktiviertem Wasserstoff
von Verbindungen der allgemeinen Formel II oder aber von Verbindungen der allgemeinen Formel III
CH-CH-Y Z
™ herstellen, in der A eine Hydroxyrnethylen·- oder Ketogruppe, B' die
Hydroxy- oder eine beliebige Alkoxy-, Aralkoxy-, oder Aryloxygruppe
und Y und Z Wasserstoffatorne oder zusammen eine Doppelbindung
bedeuten, wobei B' auch eine Acyloxygruppe sein kann, sofern A die Ketogruppe ist, oder ein Halogenatom, sofern A die
Ketogruppe und Y und Z zusammen eine Doppelbindung bedeuten.
Als Katalysatoren eignen sich insbesondere Kupferchromoxid oder Zinkchromoxid. Die Reduktion wird in einem inerten Lösungsmittel,
wie zum Beispiel Dioxan oder einem höher siedenden Kohlenwasserstoff, bei Temperaturen zwischen 150 und 300 C und einem V/asserstoffdruck
von 150 bis 300 at durchgeführt.

Die Verbindung der Formel I kann auch aus einer Verbindung der
Formel IV

mittels katalytisch aktiviertem Wasserstoff erhalten werden.
Als Katalysatoren dienen zum Beispiel Edelmetallkatalysatoren, hiervon vorzugsweise Palladium auf Tierkohle. Die Reduktion
erfolgt im allgemeinen'bei Temperaturen zwischen 0° und 1000C,
verzugsweise bei "Raumtemperatur, bei einem Wasserstoffdruck von
1 bis 100 at, bevorzugt bei 3 bis 10 at und in Gegenwart eines geeigneten Lösungsmittels, wie zum Beispiel eines Alkohols, Carbonsäureesters
oder niederer Carbonsäuren. Man kann die Verbindung der allgemeinen Formel IV jedoch auch in Gegenwart von Kupferchromoxid
oder Zinkchromoxid als Katalysatoren bei Temperaturen zwischen
150 und 300 C und einem Wasserstoffdruck vo:
der neuen Verbindung der Formel I hydrieren.

b. Durch Umsetzung einer Grignard-Verbindung der allgemeinen Formel

in der Hai ein Chlor-, Brom- oder Jodatom darstellt, mit ÄthylenoxJd.
Die Umsetzung wird in einem Lösungsmittel wie Äther,
Tetrahydrofuran, Benzol oder deren Gemischen und bei Temperaturen zwischen -1IO und +70 C, vorzugsweise bei Temperaturen
zwischen 0°C und 40 C, durchgeführt. Es kann von Vorteil sein,

wenn man die Umsetzung unter Verwendung eines Schutzgases, etwa von Stickstoff, durchführt.
Die Ausgangsverbindungen der allgemeinen Formeln II, III, IV und V sind bekannt oder nach gängigen Verfahren erhältlich.
Die Ausgangsver'bindungen der allgemeinen Formel II erhält man zum Beispiel durch katalytisch^ Hydrierung von ^-(H-Biphenylyl)-4-oxo-buttersäure.Diese
Reduktion erfolgt zweckmäßigerweise in einem polaren Lösungsmittel wie Eisessig, in Gegenwart
einer starken Säure (zum Beispiel Perchlorsäure), bei Temperaturen zwischen 0 und 50 C und bei einem Wasserstoffdruck von
1 bis 5 at. Als Katalysatoren eignen sich besonders Palladium auf r Bariumsulfat oder Palladium auf Tierkohle (vgl. auch deutsche
Patentanmeldung P 21 12 840.5). Die so erhaltenen Verbindungen der
• allgemeinen Formel II, in der B die Hydroxylgruppe bedeutet, lassen sich gegebenenfalls anschließend verestern oder in an sich üblicher
Weise in die entsprechenden Säurehalogenide oder -anhydride überführen.
Die als Ausgangsverbindung verwendete 4~(4-Biphenylyl)-4-oxobuttersäure
oder deren Ester kann durch Umsetzung von Biphenyl mit Bernsteinsäureanhydrid oder einem Bernsteinsäureenterhalogenid
in Gegenwart einer Lewis-Säure, zum Beispiel von wasserfreiem Aluminiumchlorid, in einem Lösungsmittel wie zum Beispiel
Nitrobenzol oder Methylenchlorid, bei Temperaturen zwischen 0 und 80 C, erhalten werden.
Die Ausgangsverbindungen der allgemeinen Formel III, in der A die
Hydroxymethylengruppe, Y und Z zusammen eine Doppelbindung und B' die Hydroxy- oder Alkoxygruppe bedeuten, werden durch Reduktion
mittels Alumminiumisopropylat in Gegenwart von Isopropanol aus solchen Verbindungen der Formel III erhalten, in der A die Ketogruppe,
Y und Z eine Doppelbindung und B'die Hydroxy- oder eine
Alkoxygruppe darstellen. Die letzteren Verbindungen lassen sich nach literaturbekannten Verfahren, zum Beispiel durch Friedel-
-Crafts-Acylierung von Biphenyl mit Maleinsäureanhydrid in Gegenwart
von Aluminiumchlorid (vgl. H. G. Oddy, J. Ame'r. Chem. Soc. Hß,,

Verbindungen der allgemeinen Formel III, in der A die Hydroxymethylengruppe,
Y und Z Wasserstoffatome und B1 eine Hydroxy-,
Alkoxy-, Aralkoxy- oder Aryloxygruppe darstellen, lassen sich durch Reduktion von 4-(il-Biphenylyl)-^-oxo-buttersäure oder von
deren Estern zum Beispiel mittels Aluminiumisopropylat in Anwesenheit von Isopropanol gewinnen (vgl. deutsche Patentanmeldung
P 21 12 715.1).
Die Verbindungen der allgemeinen Formel III, in der A eine Ketogruppe,
Y und Z eine Doppelbindung und B; die Hydroxygruppe bedeuten,
können in an sich üblicher Weise in ihre gemischten Säureanhydride, z.B. mit Pivalinsäure, Benzoesäure, Essigsäure, oder
in ihre Säurehalogenide, Verbindungen der allgemeinen Formel III, in der A die Ketogruppe, Y und Z Wasserstoffatome und B'die Hydroxygruppe
darstellen, in ihre gemischten Säureanhydride übergeführt werden.
Das Butandiol der Formel IV erhält man zum Beispiel durch Reduktion
von ^-(A-BiphenylyD-^-oxo-buttersäure mittels Lithiumaluminiumhydrid
oder mittels Natriumborhydrid in Anwesenheit von wasserfreiem Aluminiumchlorid bei Zimmertemperatur und in Anwesenheit
eines geeigneten Lösungsmittels wie zum Beispiel von Dimethoxyäthan.
Die neue Verbindung der Formel I weist wertvolle pharmakologische Eigenschaften auf, sie besitzt insbesondere eine gute antiphlogistische
Wirkung.
Die Prüfung auf die antiphlogistische Wirkung erfolgte beispielsweise
nach Hillebrecht (Arzneimittelforschung ^, 607 - 6l4
und nach Winter et al. (Proc. Soc. exp. Biol. Med. Ill, 51^
/1962/), wobei die Auswertung nach der von Doepfner und Cerletti (Int. Arch. Allergy a. appl. Immun. 12, 89 - 97 /1958/) beschrie
benen Methode durchgeführt wurde.
Die nachstehenden Beispiele sollen die Erfindung näher erläutern

Man versetzt eine Lösung von 7,21 g (0,03 Mol) 4-(4-Biphenyly])-,buttersäure_(F.:
118 - 119°C) in 100 ml absolutem Tetrahydrofuran portionsweise unter Rühren mit 1,13 g (0,03 Mol) Lithiumaluminiumhydrid
und erhitzt anschließend H Stunden unter Rückfluß. Nach dem
Abkühlen versetzt man mit Eiswasser, säuert mit 50 #iger Schwefelsäure
an und extrahiert mit Äther. Die Ätherlösung wird mit Natriumhydrogencarbonatlösung
und Wasser gewaschen, getrocknet und fc vom Lösungsmittel befreit. Den verbleibenden Rückstand destilliert
man im Vakuum und erhält 6,1 g (90 % der Theorie) 4-(Jl-Biphenylyl)-1-butanol
vom Kp Q ^ : I117 - 1480O

Man trägt in eine Aufschlämmung von 1,8 g Lithiumaluminiuinhydrid
in 50 ml wasserfreiem Äther unter Rühren eine Suspension von 12 g
4-(4-Biphenylyl)-buttersäure (F. 117 - 119°C) in 50 ml wasserfreiem
Äther portionsweise ein, erwärmt anschließend 5 Stunden unter Rückluß, gibt etwas Essigester zu, trägt in Eiswasser ein,
säuert dann mit Schwefelsäure an, macht mit Natronlauge alkalisch und trennt die Ätherlösung ab. Der wässerige Ablauf wird noch zweimal
mit je 50 ml Äther ausgeschüttelt. Die vereinigten Ätherlösungen
wäscht man neutral und dampft dann das Lösungsmittel ab.
Den Rückstand (10 g) kristallisiert man aus Petroläther um. Das farblose 4-(4-Biphenylyl)-l-butanol hat den P. 75 - 76°C und
0,1 mm'

Man löst 7,1 E 4-(4-Biphenylyl)-buttersäureäthylester in 50 ml
Dioxan, gibt 3,0 g Kupferchromoxid-Katalysator zu und reduziert
bei 250 C und bei einem Wasserstoffdruck von 250 Atm. bis die
berechnete Menge Wasserstoff aufgenommen ist. Dann wird abfiltriert,
das Lösungsmittel im Vakuum entfernt und der Rückstand eine Stunde mit 10 ml 20 #iger Natronlauge erwärmt. Nach dem Abkühlen
wird mit Äther extrahiert und eingedampft. Der erhaltene Rückstand
wird im Vakuum destilliert und man erhält so 3,2 g 4-(4-Biphenylyl)·
1-butanol vom KpQ lm): 118 - 14 90C .
Schmelzpunkt: 75-76 C (aus Petroläther).

15>3 G (0,05^1 Mol) 4-(4~Biphenylyl)~4~oxo-buttersäureäthylester
(F.: 8O-81 C) werden in 100 ml Dioxan mit 15 g Kupfer-Chromoxid-Katalysator
versetzt und bei 23O°C und 300 Atm. hydriert. Die Wasserstoffaufnahme
ist nach etwa 1 1/2 Stunden beendet. Danach wird der Katalysator abgesaugt, das Piltrat vom Lösungsmittel befreit
und der Rückstand in Äther aufgenommen. Man schüttelt die organische
Phase mit 10 Siiger Natronlauge, dann mit Wasser durch und befreit sie vom Lösungsmittel. Bei der Destillation des Rückstandes
erhält man 7,4 g (60 % der Theorie) des oben genannten Carbinols vom KP0 159°C
Schmelzpunkt: 75~76°C (aus Petroläther).

13,8 g (0,049 Mol) 4-(4-Biphenylyl)-4-oxo-crotonsäureäthylester .(F.: 78 - 8O0C) werden in 100 ml wasserfreiem Dioxan gelöst, mit
14 g Kupferchromoxid-Katalysator versetzt und bei 230 C und 300Atir..
Wasserstoffdruck reduziert bis die berechnete Menge Wasserstoff nach etwa einer Stunde aufgenommen ist. Die weitere Aufarbeitung
erfolgt analog Beispiel 4.
Ausbeute: 7,8 g (70 % der Theorie) vom KpQ ±mm'. 148 - l49°C

Zu einer Lösung von 5,37 g (0,02 Mol) 4-(4-Biphenylyl)-buttersäureäthylester
in 50 ml Dimethoxyäthan gibt man unter Rühren und Kühlen auf 10 - 15°C nacheinander 0,48 g (0,0125 Mol) Natriumborhydrid
und portionsweise 0,56 g (0,0042 Mol) Aluminiumchlorid.
Anschließend rührt man 1 1/2 Stunden bei Raumtemperatur weiter, trägt den Reaktionsansatz in eine Mischung aus 350 ml Eiswasser
und 25 ml konz. Salzsäure ein und nimmt das abgeschiedene Carbinol in 250 ml Äther auf. Die organische Phase \iird abgetrennt, mit Wasser
gewaschen, über Natriumsulfat getrocknet und vom Lösungsmittel befreit. Der Rückstand erstarrt und wird aus Petroläther unter Zusatz
von wenig Essigester umkristallisiert. Man erhält 2,51g (55$
der Theorie) farblose Kristalle des 4-(4-Biphenylyl)-l-butanols
vom F. 75 - 76°C.

Zu einer Suspension von 1,9 g (0,05 Mol) Natriumborhydrid in 50 ml
.1,2-Dimethoxyäthan tropft man unter Rühren bei 15 - 200C 12,93 g
(0,05 Mol) 4-(4-Biphenylyl)-buttersäurechlorid in 30 ml 1,2-Dimethoxyäthan.
Nach beendeter Zugabe rührt man noch 2 Stunden bei Raumtemperatur und gießt dann den Reaktionsansatz auf eine Mischung
aus 400 g Eis und 50 ml konz. Salzsäure. Die dabei abgeschiedene
Substanz nimmt man in 300 ml Äther auf. Man wäscht den Ätherextrakt neutral, trocknet.ihn über Natriumsulfat und
dampft das Lösungsmittel ab. Der Rückstand wird fest. Man kristallisiert ihn aus Petroläther unter Zusatz von Cyclohexan um und erhält
8,3 g (73 % der Theorie) farblose Kristalle des oben genannten Carbinols vom F. 75 - 76°C.
a) 4-(4-BiphenyIyI)-I,4-butandiol
7,6 g Lithiumaluminiumhydrid schlämmt man in 100 ml wasserfreiem Tetrahydrofuran auf, trägt unter Rühren und Kühlen auf etwa
10 C die Lösung von 25,4 g 4-(4-Biphenylyl)--!4-oxo-buttersäure
(B1. 187 C) in 100 ml wasserfreiem Tetrahydrofuran ein und erhitzt dann 3 Stunden unter Rückfluß. Man gibt zuerst etwas
Essigester zu, dann Eiswasser, säuert mit 50 #iger Schwefelsäure an, macht durch Zugabe von Natronlauge alkalisch und extrahiert
mit Äther. Die Ätherlösung wird mit Wasser gewaschen, mit Natriumsulfat getrocknet und vom Lösungsmittel befreit.
Den verbleibenden Rückstand verreibt man mit Petroläther und kristallisiert ihn aus Cyclohexan/Essigester um.
Man erhält 18,5 g (76,5 % der Theorie) farbloses <f-(4-Biphenylyl)
1,4-butandiol vom P. 79 - 800C.

b) 4-(4-Biphenylyl)-l-butanol
9,68 g (0,04 MoI) l-(4-Biphenylyl)-l,4-butandiol (F. 79 - 80 C)
löst man in 100 ml Methanol, fügt 1 g Palladium auf Kohle als Katalysator zu und hydriert bei Raumtemperatur und 5 Atm.
Druck, bis die berechnete Menge Viasserstoff aufgenommen ist. Dann saugt man vom Katalysator ab, destilliert das Lösungsmittel
aus dem Piltrat ab, wobei man einen festen Rückstand erhält, den man aus Petroläther und wenig Essigester umkristallisiert. Man
erhält 8,32 g (93 % der Theorie) farblose Kristalle des 4-(4-Biphenylyl)-l-butanols
vom P. 75-76 C.
4-(4-Biphenylyl)-l~butanol·
Zu einer Lösung von 5,37g (0,02 Mol) 4-(4-Biphenylyl)-buttersäureäthylester
in 100 ml wasserfreiem Tetrahydrofuran tropft man unter Rühren 10,6g (0,04 Mol) Natrium-bis-(2-methoxyäthoxy)-dihydroaluminat
(70 %ig in Benzol) und rührt noch zwei Stunden, Anschließend dampft man ein, versetzt den Rückstand mit
200 ml Wasser und säuert mit 15 ^iger Salzsäure an. Das abgeschiedene
öl wird in Äther aufgenommen und mit Wasser neutral gewaschen.
Der nach dem Verdampfen des Äthers verbleibende Rückstand wird aus Cyclohexan unter Zusatz von wenig Essigester umkristallisiert.
Man erhält so 4,18 g (92 % der Theorie) des oben genannten Carbinols als farblose Kristalle vom P. 75 - 760C.

4-(4-Biphenyly1)-1-butanol
Zu einer Lösung von 13,42 g (0,05 Mol) 4-(4-Biphenylyl)-buttersäureäthy!ester
in 125 ml absolutem Äther gibt man portionsweise 8,18g (0,0375 Mol) Lithiumborhydrid und kocht anschließend
24 Stunden unter Rückfluß, wobei sich eine zähe Mas-

se abscheidet. Anschließend versetzt man mit 2n Schwefelsäure und trennt die ätherische Schicht ab, extrahiert die wässerige Phase
mit Äther und wäscht die vereinigten Ätherauszüge mit Wasser, gesättigter
Natriumhydrogencarbonatlösung und Wasser. Die organische Phase trocknet man über Natriumsulfat und entfernt das Lösungsmittel
im Vakuum. Der erhaltene farblose, kristalline Rückstand wird aus Cyclohexan unter Verwendung von Aktivkohle umkristallisiert.
Ausbeute: 9,44g (83 % der Theorie), F.: 75 - 76°C.

4- (4-Biphenylyl)-l-butanol
12,02g (0,05 Mol) 4-(4-Biphenylyl)-buttersäure werden in einer
Mischung von 75 ml absolutem Tetrahydrofuran und 5,05 g (0,05 Mol)
absolutem Triethylamin gelöst und bei -20 bis -30 C langsam und
unter starkem Rühren mit 5,40 g (0,05 Mol) Chlorameisensäureäthylester versetzt. Nach 15 Minuten wird vom Triäthylammoniumchlorid
unter WasserausSchluß abgesaugt. In diese Lösung werden unter Ausschluß
von Feuchtigkeit 4,35 g (0,20 Mol) Lithiumborhydrid in
kleinen Portionen eingetragen. Man rührt zwei Stunden bei Zimmertemperatur,
kocht anschließend 48 Stunden unter Rückfluß und destilliert schließlich den größten Teil des Lösungsmittels ab. Nach
dem Zersetzen mit 2 η Schwefelsäure extrahiert man erschöpfend mit Äther. Die Ätherauszüge werden nacheinander mit Wasser, gesättigter
Natriumhydrogencarbonatlösung und nochmals mit V/asser gewaschen,
über Natriumsulfat getrocknet und eingeengt. Der Rückstand, ein fahlgelbes öl, wird im Feinvakuum (Kpn . : 150 151
C) destilliert, dann an insgesamt 700 g Kieselgel unter Verwendung von Benzol zum Eluieren chrornatographiert. Durch Aufarbeitung
der Eluate erhält man reines 4-(4-Biphenylyl)-lbutanol von: Kpn Λ : 148 - 149°C und F. 75 - 76°C (Cyclchexan-Essigsäureäthylester).
Ausbeute: 7,65 g (68 % der Theorie).

In die Lösung einer Grignardverbindung aus l6,0 g (0,06l MoI)
2-(4-Biphenylyl)-äthylbromid (F. 39 - 400C) und 1,5 g (0,062 Grammatom)
Magnesiumspänen in 150 ml absolutem Äther wird bei ca. O0C eine gekühlte Lösung von 4,0 g (0,091 Mol) Äthylenoxid in
80 ml absolutem Äther unter kräftigem Rühren eingetropft. Man rührt bei 0° bis 10°C weiter, läßt über Nacht stehen, kocht das
Gemisch noch 30 Minuten am Rückfluß und arbeitet nach dein Abkühlen
in üblicher Weise unter Zusatz von 2n-Salzsäure auf. Die neutral gewaschene Ätherlösung wird getrocknet und eingeengt. Man
destilliert den Rückstand im Feinvakuum (Kpn ± mm'. 148 - l49°C)
und kristallisiert aus Petroläther um. Dabei erhält man 4-(4-Biphenylyl)-l-butanol
vom F. 75 - 76°C (Ausbeute: 54 % der Theorie).

4-(4-Biphenylyl)-1-butanol
Eine Lösung von 20,0 g (0,078 Mol) 4-(4-Biphenylyl)-buttersäure~ methylester (Kpn Q7 mm: 145 - 148 C) in 100 ml absolutem Äther
tropft man bei ca. 100C unter Rühren zu einer Suspension von 5j9 g
(0,156 Mol) Lithiumaluminiumhydrid in 400 ml absolutem Äther, k Man rührt das Gemisch 15 Stunden bei Labortemperatur, kocht 2 Stunden
unter Rückfluß, zerlegt überschüssiges Hydrid durch Zugabe von etwas Essigester, versetzt unter Kühlung mit Wasser und säuert mit
5n-Schwefelsäure an. Die Ätherlösung wird abgetrennt, zusammen mit weiteren Ätherextrakten neutral gewaschen, getrocknet und eingeengt.
Der kritalline Rückstand ergibt beim Umkristallisieren aus Petroläther 15,7 g (89 % der Theorie) 4-(4-Biphenylyl)-1-butanol
vom Schmelzpunkt 7475 - 76°C.
Die neue Verbindung der Formel I läßt sich zur pharmazeutischen Anwendung, gegebenenfalls in Kombination mit anderen
Wirksubstanzen, in die üblichen pharmazeutischen Zubereitungsformen einarbeiten. Die Einzeldosis beträgt 10 bis 200 mg,
vorzugsweise 50 bis I50 mg, die Tagesdosis 50 bis 500 mg, vorzugsweise
70 bis 300 mg.

In dem auf 70 C erwärmten Wasser werden nacheinander, DOWSS, Benzoesäure, Natriumcylamat und Polyvinylpyrrolidon gelöst. Man
gibt Glycerin und Aerosil dazu, kühlt auf Raumtemperatur ab und suspendiert mit Hilfe eines Eintauchhomogenisators die feinpulverisierte
Wirksubstanz. Anschließend wird aromatisiert und mit Wasser auf das gegebene Volumen aufgefüllt.

Herstellungsverfahren
In etwa 15 % der benötigten Wassermenge werden nacheinander DOfIMS
gelöst und der feinpulverisierte Wirkstoff suspendiert. Das restliche Wasser wird auf 80 C erwärmt und Veegum sovrie Aerosil suspendiert.
Danach löst man nacheinander Benzoesäure, !latriuincyclamat
sowie Polyvinylpyrrolidon und fügt das Glycerin zu. Die Lösung wird auf Raumtemperatur abgekühlt, aromatisiert und die Wirkstoff suspension eingerührt. Das fertige Präparat wird homogenisiert.

- Patentansprüche1.) Als neue Verbindung 4-(4-Biphenylyl)-l-butanol der Formel2.) Verfahren zur Herstellung von 4-(4-Biphenylyl)-l-butanol der FormelCH—CHCHg(Ddadurch gekennzeichnet, daßa.) eine Verbindung der allgemeinen Formel_ I I I ■ ^\l ff Vk. >—(· )'CH2 CHg CH2 C^(II)in der B die Hydroxy-, eine beliebige Alkoxy-, Aralkoxy-, Aryloxy- oder Acyloxygruppe oder ein Halogenatom bedeutet, mittels komplexer Hydride in einem Lösungsmittel oder mit katalytisch aktiviertem Wasserstoff oder eine Verbindung der allgemeinen Formeln309816/1198oderGH- CH2- CK2- CK2OH (IY ) OHwobei in der Formel III A eine Hydroxymethylen- oder Ketogruppe, B1 die Hydroxy- oder eine beliebige Alkoxy-, Aralkoxy- oder Aryloxygruppe und Y und Z Wasserstoffatcme oder zusammen eine Doppelbindung bedeuten, und B1 auch eine Acyloxygruppe sein kann, sofern A die Ketogruppe ist, oder ein Halogenatom, sofern A die Ketogruppe und Y und Z zusammen eine Doppelbindung darstellen, mittels katalytisch aktiviertem Wasserstoff reduziert wird, oderb.) eine Grignard-Verbindung der allgemeinen FormelHal (Y)in der Hai ein Chlor-, Brom- oder Jodatom darstellt, mit Ä'thylenoxid in Gegenwart eines Lösungsmittels umgesetzt wird.3.) Verfahren gemäß Anspruch 2, dadurch gekennzeichnet, daß zur Reduktion von Verbindungen der allgemeinen Formel II als komplexe Hydride Lithiumaluminiumhydrid, Lithiumborhydrid, Natrium-bis-(2-methoxyäthoxy)-dihydroaluminat, Natriumaluminiumhydrid, Natriumborhydrid in Anwesenheit von wasserfreiem Aluminiumchlorid oder von Bortrifluorid oder, falls in einer Verbindung der allgemeinen Formel II B ein Halogenatom darstellt, auch Natriumborhydrid verwendet v/erden.309816/1198l\.) Verfahren gemäß Anspruch 2 a und. 3, gekennzeichnet durch die Verwendung von Tetrahydrofuran, A'ther, Dioxan, Dirnethoxyäthan, Diäthylenglykoldimethylather, Benzol oder Gemischen davon als Lösungsmittel.5.) Verfahren gemäß Anspruch 2, dadurch gekennzeichnet;, daß zur Reduktion von Verbindungen der allgemeinen Formeln II und III mit katalytisch aktiviertem Wasserstoff als Katalysatoren Kupferchromoxdd oder Zinkchromoxid, zur Reduktion einer Verbindung der Formel IV Palladiumkatalysatoren verwendet i^erden.6.) Verfahren gemäß Anspruch 5» dadurch gekennzeichnet, daß die Reduktion in Anwesenheit von Kupferchromoxid oder Zinkchromoxid bei 150 bis 3000C und einem Wasserstoffdruck von 150 bis 300 at durchgeführt wird.7.) Verfahren gemäß Anspruch 5, dadurch gekennzeichnet, daß die Reduktion der Verbindung von Formel IV mit Palladium auf Tierkohle und bei Zimmertemperatur bei einem v/afjserstoffdruck von 3 bis 10 at in Gegenwart eines Lösungsmittels durchgeführt wird.8.) Verfahren gemäß Anspruch 2 b, dadurch gekennzeichnet, daß als Lösungsmittel Diethylether, Tetrahydrofuran, Dirnethoxyäthan, Benzol oder Gemische davon verwendet v/erden.9.) Arzneimittelzubereitung, gekennzeichnet durch einen Gehalt an H-( Ji-Biphenylyl)-l-butanol in Kombination mit den gebräuchlichen Träger- und/oder Hilfsstoffen.10.) Verfahren zur Herstellung von Arzneimittelzubereitungen gemäß Anspruch 9, dadurch gekennzeichnet, daß 'l-(4-Biphenylyl)-lbutanol mit den üblichen Träger- und/oder Hilfsstoffen zusammen verarbeitet wird.309816/1T98
