Description
Foreliggende oppfinnelse vedrører en ny klasse av forbindelser som har strukturer som er beslektet med visse naturlig forekommende og syntetiske chalkoner, såvel som et farmasøytisk preparat samt farmasøytisk anvendelse derav.
Forbindelsen 1,3-difenyl-2-propen-l-on er kjent ved trival-navnet "chalkon". Hange naturlig forekommende flavonoider - deler strukturelle trekk med chalkon og er omtalt ved den generiske betegnelsen "chalkoner". Visse flavonoider som inkluderer dem som også er klassifisert som chalkoner, er i den senere tid også blitt vist å ha anticanceraktivitet (Cancer Research, 48, 5754, 1988) og kjemopreventiv aktivitet i enkelte tumorer (J. Nat. Prod., 53, 23, 1990).
Spesielt er quercetin, et ubikvitært flavanoid funnet i planter, blitt vist til å virke på proliferasjonen av humane levkemiceller (Br. J. av Haematology, 75, 489, 1990) og på andre cellelinjer (Br. J. Cancer, 62, 94, 942, 1990, Int. J. Cancer, 46, 112, 1990, Gynecologic Oncology, 45, 13, 1992) og til å ha en synergisk virkning sammen med vanlige antiblastiske legemidler.
I tillegg har enkelte naturlige eller syntetiske chalkoner, beskrevet i Internasjonal Patentpublikasjon nr. WO 91/1774 9 og i Internasjonal Patentpublikasjon nr. WO 96/192 09 (Baylor College of Medicine) vist seg å ha en signifikant antiproliferasjonsaktivitet på en rekke ulike cellelinjer.
Skjønt virkningsmekanismen i forbindelse med antiproliferasjonsaktiviteten til flavonoider og chalkoner fremdeles er ukjent, menes den å være forbundet med interaksjonen av disse forbindelser med type II østrogenreseptorer.
Virkningen av disse polyfenolsubstanser in vivo er ganske sikkert mye mere komplisert. Alle disse forbindelser er generelt karakterisert ved en nesten fullstendig uoppløselig-het i vann og, in vivo, ved en svært dårlig biotilgjengelig-het som er forbundet med en hurtig metabolisme av fenoler og en markert affinitet for lipider og proteiner.
Man har nå overraskende funnet at visse nye chalkoner, chalkonderivater og chalkonanaloger, særlig dem hvor fenyl-ringen i 3-stillingen er substituert eller erstattet med ringer inneholdende ett eller flere heteroatomer, har en større antiproliferasjonsaktivitet både på sensitive cancerceller og celler som er resistente overfor vanlige kjemoterapeutiske legemidler inkluderende den siste generasjon av anti-neoplastiske midler, paklitaxel og docetaxel.
Således ifølge et aspekt av den foreliggende oppfinnelse er der tilveiebragt forbindelser med generell formel (A)
hvor:
Ar representerer fenyl som kan være usubstituert eller
substituert med en, to eller tre substituenter som uavhengig er valgt blant Cl, Br, F, -OMe, N02, CF3, C^ lavere alkyl (foretrukket metyl), -NMe2, -NEt2, -SCH3, -NHCOCH3; 2-tienyl;
2-furyl; 3-pyridyl; 4-pyridyl eller 3 -indolyl; R representerer -OCH2R1, hvor Rx er valgt fra -CH=CMe2, -CMe=CH2, -C=CH, med den betingelse at når Ar representerer fenyl, C4-alkyl-fenyl, 4-metoksyfenyl eller 3,4-di-metoksyfenyl, er R annet enn 3-metyl-2-butenyloksy.
Innen denne klassen er Ar foretrukket valgt fra usubstituert fenyl, 3-pyridyl, 4-pyridyl og 3-indolyl. Særlig foretrukket er forbindelser hvor R representerer -OCH2-CH=CMe2,
-OCH2-CMe=CH2 eller -0CH2-C<=>CH.
Forbindelser med formel (A) som inneholder en basisk amino-funksjon kan omdannes til syreaddisjonssalter med farmasøy-tisk aksepterbare'syrer slik som saltsyre og fosforsyre og slike salter er inkludert i den foreliggende oppfinnelse.
Oppfinnelsen vedrøres også et farmasøytisk preparat som er kjennetegnet ved at det omfatter en forbindelse med formel

Oppfinnelsen vedrører videre anvendelse av en forbindelse med formel (A):

enn 3-metyl-2-butenyloksy,
for fremstilling av et medikament for behandling eller forebygging av neoplasmer, for behandling eller forebygging av menopausal lidelser eller for behandling eller forebygging av osteoporose.
In vitro resultater har vist at forbindelsene ifølge oppfinnelsen har en markert affinitet til type II østrogenreseptorer og er inhibitorer av tyrosin kinase. Videre kan de inhibere aktiviteten av P-170 proteinpumpen som medierer MDR i tumorceller, og de kan antagonisere proliferasjonen av både hormonavhengig og kjetnores ist ente tumorale celler i det samme omfang.
Virkningsmekanismene til forbindelsene med formel (A) er mest sannsynlig forskjellige fra dem hos strukturelt beslektede kjente forbindelser, (inkluderende naturlig forekommende chalkoner) ettersom de kan inhibere protein P-170 fullstendig. På grunn av dette viser forbindelsene ifølge oppfinnelsen en mere markert aktivitet både in vitro og in vivo enn andre produkter som er kjent innen teknikken.
Affiniteten til enkelte forbindelser for type 11 reseptorer og den antiproliferative aktivitet på ovarial-tumorceller er vist i tabell 1.
Bindingen til type II .østrogenreseptorer ble evaluert i ovarial-tumorceller og tumorceller i andre målorganer. Cellene ble dyrket i et enkeltlag i henhold til kjent teknikk rapportert i litteraturen. For å gjøre testen reproduserbar, ble cellene trypsinbehandlet hver uke og anbragt på plater i en tetthet på 8xl04 celler/cm2 og inkubert ved 37°C i luft som inneholdt 5% C02.
De forskjellige forbindelser som ble testet, oppløst i etylalkohol, ble tilsatt i seriefortynninger og de behandlede celler og kontrollceller ble inkubert med <3>H-østradiol eller i nærvær av dietylstilbestrol i henhold til metodene beskrevet i litteraturen. Antiproliferasjonsaktiviteten ble bekreftet på samme måte ved å tilsette forbindelsene, oppløst i DMSO, til mediet og utføre en celletelling etter 72 timer.
Forbindelsene i henhold til den foreliggende oppfinnelse inhiberer celleproliferasjonen in vivo som vist ved å under-søke størrelsen av tumorene som er implantert i atymiske "nakne" mus i henhold til teknikkene som er omfattende rapportert innen litteraturen.
Behandling av dyr med doser som strekker seg fra 1 mg/kg til 200 mg/kg ga en markert reduksjon i størrelsen av tumorene med total tilbakegang av tumoren i en rekke av de behandlede dyr.
Forbindelsene i henhold til oppfinnelsen kan passende administreres enteralt eller parenteralt (f.eks. oralt eller ved injeksjon) i farmasøytisk aksepterbare vehikler. Dosen for behandling og forebygging av neoplasmer med forbindelsene ifølge oppfinnelsen (som er hovedsakelig ikke-toksiske) kan variere fra 50 mg til 1.000 mg pr. døgn i en tid som strekker seg fra måned til flere år avhengig av sykdommens natur og alvorlighet.
Forbindelsene i henhold til oppfinnelsen er særlig anvendbare i kombinasjonsterapier sammen med andre anti-neoplastiske legemidler og/eller fysiske anticancerbehandlinger slik som radioterapi. De kan således f.eks. med fordel administreres i antitumorterapi før behandling med antiblastiske legemidler, slik at dosen .av de sistnevnte og følgelig deres uønskede bivirkninger kan reduseres til pasientens fordel.
Gitt deres klare antiproliferasjonsaktivitet på hormon-avhengige tumorceller og aktiviteten på proteinkinase, kan behandling av pasienter fortsettes etter den tradisjonelle kjemoterapi eller kirurgiske operasjon for fjerning av tumoren for å blokkere metastatisk diffusjon. De samme forbindelser i henhold til oppfinnelsen kan anvendes ved profylaktiske behandling for å forebygge tumorer i uterus, ovarier og bryst, såvel som for reduksjon av typiske menopausallidelser.
I slike tilfeller kan dosene strekke seg fra 5 mg/kg opp til 100 mg/kg pr. døgn og videre kan produktene fordelaktig anvendes oralt i formuleringer som inneholder fosfolipider som forenkler deres resorpsjon.
Forbindelsene i henhold til oppfinnelsen kan syntetiseres ved hjelp av to hovedmåter: a) ved reaksjonen mellom en ekvimolar oppløsning av acetofenon og et passende aldehyd i etylalkohol i nærvær
av KOH, og
b) ved å reagere en ekvimolar oppløsning av acetofenon og et passende aldehyd i etylalkohol i nærvær av piperidin og
eddiksyre under svak motstrøm.
På slutten av reaksjonen kan produktene krystalliseres fra den alkoholiske oppløsning eller renses ved hjelp av kromatografi. Reaksjonsproduktene har generelt en høyere renhets-grad, slik at krystallisering vanligvis er tilstrekkelig for å oppnå de ønskede produkter i en høy grad av renhet.
De etterfølgende eksempler illustrerer oppfinnelsen mer detaljert uten å begrense denne.
Eksempel I - Generelle betingelser for å oppnå chalkoner.
Metode A.
En oppløsning av 50% KOH tilsettes til en ekvimolar
oppløsning av acetofenon {0,075 mol) og aldehyd (0,075 mol) i 95% etanol, idet tilsetningen utføres under kraftig omrøring ved romtemperatur. Reaksjorisblåndingen får stå med omrøring over natten og fortynnes deretter med vann og surgjøres, presipitatet separeres ved filtrering og tørkes under vakuum. Forbindelsene krystalliseres med etanol og eller separeres først ved kromatografi og krystalliseres deretter med etanol.
Metode B.
En oppløsning av acetofenon (0,075 mol),- aldehyd (0,075 mol), piperidin (15 ml) og eddiksyre (75 ml) i 95% etylalkohol
(80 ml) motstrøms-oppvarmes i 5 timer. Molekylsiler tilsettes til oppløsningen for å eliminere vann og det hele får hvile over natten.- Presipitatet som generelt oppnås samles og krystalliseres. Dersom produktet ikke presipiterer under disse forhold, avdampes løsningsmidlet i vakuum og resten renses ved kromatografi på en silikagelkolonne.
Eksempel II - Fremstilling av 1-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-{pyridin-4-yl)propen-l-on, forbindelse I.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis under omrøring til en oppløsning som inneholder 2,2 g (10 mmol) 2-hydroksy-4-(3-metylbut-2-enyloksy)-acetofenon og 1,07 g
(10 mmol) pyridin-4-karboksyaldehyd i 25 ml 95% etanol. Oppløsningen får stå med omrøring over natten ved romtemperatur og helles deretter i 60 ml vann. Den surgjøres deretter med fortynnet HCl og filtreres, resten krystalliseres to ganger med 95% etanol. Dette gir 1,9 g av et produkt med følgende egenskaper: smp. 99-100°C, <1>H NMR 6 (CHC13) : 1.68 (S, 3H, CH3) , 1.7 (s, 3H, CH3), 4.53 (d, 2H, 0CH2), 5.45 (mt 1H, CH=), 6.5-6.7 (m, 2H, olefiner), 8.7-9.1 (m, 7H, Ar). Masse: M/z (%): 309 (M<+>, 7.57), 241 (59.12), 163 (62 .41) , 69 (100) .
Eksempel III - Fremstilling av 1-[2-hydroksy-4-<3-metylbut-2-enyloksy)-fenyl]-3-{pyridin-3-yl)propen-l-on, forbindelse II.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis og under omrøring til en oppløsning som inneholder 2,2 g (10 mmol) 2-hydroksy-4-(3-metylbut-2-enyloksy)-acetofenon og 1,07 g
(10 mmol) pyridin-3-karboksyaldehyd i 25 ml 95% etanol. Oppløsningen omrøres over hatten ved romtemperatur og helles deretter i 60 ml vann. Den surgjøres deretter med fortynnet HCl og filtreres, resten krystalliseres to ganger med 95% etanol. Dette gir 1,6 g av et produkt med følgende
egenskaper: smp. 177-179°C, <1>H NMR 6 (CHC13) : 1.65 (s, 3H, CH3), 1.72 {S, 3H, CH3), 4.64 (d, 2H, OCH7), 5.42 (ra, 1H, CH=), 6.5-6.65 (m, 2H, olefiner), 7.8-9.4 (m, 7H, Ar). Masse: M/z (%): 309 (M<+>, 10.76), 241 (71.17), 163 (43.17), 69 (100).
Eksempel IV - Fremstilling av 1-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-(4-acetamido)-fenyl-propen-l-on, forbindelse III.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis og under omrøring til en oppløsning som inneholder 2,2 g (10 mmol) 2-hydroksy-4-(3-metylbut.-2-enyloksy)-acetofenon og 1,63 g
(10 mmol) 4-acetamidbenzaldehyd i 25 ml 95% etanol. Oppløsningen omrøres over natten ved romtemperatur og helles deretter inn i 60 ml vann. Den surgjøres med fortynnet HCl og ekstraheres med metylenklorid. Den organiske oppløsning-vaskes med vann, tørkes og tørrevaporeres. Resten renses ved kromatografi på en silikakolonne med eluering med en blanding av toluen/etylacetat i et forhold på 9:1. Etter samling av utgangsacetofenonet (30%), elueres det ønskede produkt med følgende egenskaper: smp. 150-152°C, <1>H NMR 6 (CHC13):1.75 (s, 3H, CH3) , 1.8 (s, 3H, CH3) , 2.2 (s, 3H, COCH3) , 4.55 (d, 2H, 0CH2), 5.6 (m, 1H, CH=), 6.4-6.5 (rn, 2H, (olefiner), 7.3 (bred, 1H, NH), 7.4-7.9 (m, 7H, Ar). Masse: m/z (%): 365 (M+, 48.38), 297 (100), 148 (70.77), 69 (97.15).
Eksempel V - Fremstilling av 1-[2-hydroksy-4-(2-metyl-allyloksy)-fenyl]-3-(4-dimetylamin)-fenyl-propen-l-on, forbindelse IV.
En oppløsning som inneholder 2,06 g (10 mmol) 2-hydroksy-4-(2-metylallyloksy)-acetofenon og 1,79 g (10 mmol) 4-dimetylaminbenzaldehyd, 11 ml 95% etanol, 2 ml piperidin og 15 ml iseddik motstrøms-oppvarmes i 5 timer. Etter tilsetning av molekylsiler får oppløsningen stå over natten, den filtreres deretter og presipitatet krystalliseres med 95% etanol. Dette gir 0,75 g av et produkt med følgende egenskaper: smp. 85-87°C, <1>H NMR 6 (CHC13) : 1.85 (s, 3H, CH3) , 3.02 (S, 6H, NMe) , 4.48 (s, 2H, CH2 = ) , 5.05 (d, 2H, 0CH2) , 6.4-6.5 (m, 2H, olefiner), 6.6-7.9 (m, 7H, Ar). Masse: m/z (%): 337 (M<+>, 55.97), 147 (100), 134 (67.67), 55 (8.87).
Eksempel VI - Fremstilling av 1-[2-hydxoksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-(indol-3-yl)-propen-1-on, forbindelse V.
En oppløsning som inneholder 2,2 g (10 mmol) 2-hydr-oksy-4- (2-enyloksy)-acetofenon, 1,45 g (10 mmol) indol-3-karboksy-aldehyd, 11 ml 95% etanol, 2 ml piperidin og 15 ml iseddik motstrøms-oppvarmes i 5 timer. Etter tilsetning av molekylsiler får oppløsningen stå over natten, den tørrevaporeres og underkastes kromatografi på en silikagelkolonne under eluering med petroleumseter/etylacetat i forholdet 3:7. Etter eliminering av utgangsacetofenonet, samles 0,8 g av det ønskede produkt med følgende egenskaper: smp. 2 28-23 0°C (Toluen), <1>H NMR 6 (DMSO): 1.72 (s, 3H, CH3), 1.76 (s, 3H, CH3),4.64 (d, 2H, OCH2), 5.45 (t, 1H, CH=), 6.5-6.6 (m, 2H, olefiner), 7.2-8.2 (m, 8H, Ar), 12 (s, 1H, OH). Masse: m/z (%) : 347 (M<+>, 55.88), 143 (100), 130 (85.83), 69 (40.80).
Eksempel VII - Fremstilling av 1-[2-hydroksy-4-(2-metyl-allyloksy)-fenyl]-3-(indol-3-yl) -propen-l-on,
forbindelse VI.
En oppløsning som inneholder 2,06 g (10 mmol) 2-hydroksy-4-(2-metylallyloksy)-acetofenon, 1,45 g (10 mmol) indol-3-karboksyaldehyd, 11 ml 95% etanol, 2 ml piperidin og 15 ml iseddik motstrøms-oppvarmes i 5 timer. Etter tilsetning av molekylsiler får oppløsningen stå over natten, den tørr-evaporeres deretter og underkastes, kromatografi på en silikagelkolonne under eluering med petroleumseter/etylacetat i et forhold 3:1. Etter eliminering av utgangsacetofenonet, samles 0,7 g av det ønskede produkt med følgende egenskaper: smp. 29-30°C, <1>H NMR 5 (CHC13) : 1.8 (s, 3H, CH3) , 4.55 (s, 2H, CH2=), 5.05 (d, 2H, 0CH2), 6.4-6.5 (m, 2H, olefiner), 7.4-7.9 (m, 8H, Ar). Masse: m/z (%): 333 (M<+>, 44.03), 143 (100), 130 (48.23), 115 (17.10).
Eksempel VIII - Fremstilling av I-[2-hydroksy-4-(prop-2-ynyloksy)-fenyl]-3-(indol-3-yl)-propen-1-on, forbindelse VII.
En oppløsning som inneholder 1,9 g (10 mmol) 2-hydroksy-4-(prop-2-ynyloksy)-acetofenon, 1,45 g (10 mmol) indol-3-karboksyaldehyd, 11 ml 95% etanol, 2 ml piperidin og 15 ml iseddik motstrøms-oppvarmes i 5 timer. Etter tilsetning av molekylsiler får oppløsningen stå over natten, den tørr-evaporeres deretter og underkastes kromatografi på en silikagelkolonne under eluering med petroleumseter/etylacetat i et forhold 3:1. Etter eliminering av utgangsacetofenonet, samles 0,8 g av det ønskede produkt med følgende egenskaper: smp. 228-230°C, <X>H NMR 6{CHC13): 2.5 (t, I H, CH=), 4.65 (s, 2H, OCH2) , 6.4-6.5, (m, 2H, olefiner), 7.1-8.2 (m, 9H, Ar og NH) , 11 ( bred, 1 H, OH). Masse m/z (%) : 317 {M<1>, 27.02), 143 (50.70), 130 (23.40), 91 (41.09).
Eksempel IX - Fremstilling av 1-[2-hydroksy-4-(2-metyl-allyloksy) -fenyl]-3-(pyridin-3-yl)-propen-l-on, forbindelse VIII.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis og under omrøring til en oppløsning som inneholder 2,06 g (10 mmol) 2-hydroksy-4-(2-metylallyloksy)-acetofenon og 1,07 g
(10 mmol) pyridin-3-karboksyaldehyd i 25 ml 95% etanol. Oppløsningen holdes under omrøring over natten ved romtemperatur og helles deretter inn i 60 ml vann. Den sur-gjøres med fortynnet HCl og filtreres, resten krystalliseres med 95% etanol. Dette gir 1,4 g av et produkt med følgende egenskaper: smp. 164-166°C,
XH NMR 6 (CHC13) : 1.8 (&, 3H, CH3) , 4.5 (s, 2H, CH2 = ) , 5.8 (d, 2H, OCH2) , 6.4-6.6 (m,'2H, olefiner) , 7.3-8.9 (m, 7H, Ar). Masse: m/z (%): 295 (M<+>, 45.98), 240 (21.93), 217 (35.55), 132 (24.23), 55 (100).
Eksempel X - Fremstilling av 1-[2-hydroksy-4-(2-metyl-allyloksy) -fenyl]-3-(3-metoksyfenyl)-propen-l-on, forbindelse IX.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis og under omrøring til en oppløsning som inneholder 2,06 g (10 mmol) 2-hydroksy-4-(2-metylallyloksy)-acetofenon og 1,36 g
(10 mmol) 3-metoksybenzaldehyd i 25 ml 95% etanol. Oppløs-ningen holdes under omrøring over natten ved romtemperatur og helles deretter inn i 60 ml vann.- Den surgjøres deretter med
fortynnet HCl og filtreres, resten krystalliseres med 95% etanol. Dette gir 1,7 g av et produkt med følgende egenskaper: smp. 97-100°C, <1>H NMR 6 (CHC13): 2.54 {t, 1H, CH=), 3.82 (s, 3H, OCH3) , 4.70 (s, 2H, 0CH2) , 6.5-6.6 (m, 2H, olefiner), 6.9-7.9 (m, 7H, Ar). Masse m/z (%): 308 (M<+>, 100), 269 (19.75), 161 (56.42), 134 (65.79), 118 (32.73).
Eksempel XI - Fremstilling av l-[2-hydroksy-4-(prop-2-ynyloksy) -fenyl] -3- (pyridin-3-yl) -propen-l-on, forbindelse X.
En oppløsning av 50% KOH (3 ml) tilsettes dråpevis og under omrøring til en oppløsning som inneholder 1,9 g (10 mmol) 2-hydroksy-4-(prop-2-ynyloksy)-acetofenon og 1,07 g (10 mmol) pyridin-3-karboksyaldehyd i 25 ml 95% etanol. Oppløsningen holdes under omrøring over natten ved romtemperatur og helles deretter inn i 60 ml vann. Den surgjøres deretter med fortynnet HCl og filtreres, resten krystalliseres med 95% etanol. Dette gir 1,5 g av et produkt med følgende egenskaper: smp. 115-in°C, ^ NMR 6 (CHC13)2.5 (t, 1H, CH=) , 4.8 (s, 2H, OCH2) , 6.55-6.65 (m, 2H, olefiner), 7.3-8.9 (m, 7H, Ar). Masse: m/z (%): 279 (M+, 100), 240 (27.47), 201 (74.86), 147 (24.90), 104 (49.93).
Claims (6)
- l. Forbindelse, karakterisert ved at den har den generelle formel (A) hvor: ' Ar representerer fenyl, som kan være usubstituert eller ■substituert med en, to eller tre substituenter som uavhengig er valgt fra Cl, ■Br, F, -OMe, N02, CF3, cL.4 lavere alkyl, -NMe2, -NEt2, -SCH3, -NHCOCH3; 2-tienyl; 2- furyl,- 3-pyridyl; 4-pyridyl eller 3- indolyl; R representerer -OCH2R1, hvor Rx er valgt fra -CH=CMe2, -CMe=CH2, - C=CU; med den betingelse at når Ar representerer fenyl, C4-alkyl-fenyl, 4-metoksyfenyl eller 3,4-dime-toksyfenyl, er R annet et 3-metyl-2-butenyloksy.
- 2. Forbindelse som angitt i krav l, karakterisert ved at Ar er valgt fra usubstituert fenyl, 4-dimetylaminofenyl, 4-acetamidofenyl, 3-pyridyl, 4-pyridyl og 3-indolyl.
- 3. Forbindelse som angitt i krav 1, karakterisert ved at den er valgt blant: 1-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-(pyridin-4-yl)-propen-l-on, 1-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl] -3-(pyridin-3-yl)-propen-l-on, 1-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-(4-aceta-mido)-fenyl-propen-l-on,l-[2-hydroksy-4-(2-metylallyloksy)-fenyl]-3-(4-dimetylamin)-fenyl-propen-1-on,l-[2-hydroksy-4-(3-metylbut-2-enyloksy)-fenyl]-3-(indol-3-yl)-propen-l-on,l-[2-hydroksy-4-(2-metylallyloksy)-fenyl]-3-(indol-3-yl)-propen-l-on,l-[2-hydroksy-4-(prop-2-ynyloksy)-fenyl]-3-(indol-3-yl)-propen-l-on,l-[2-hydroksy-4-(2-metylallyloksy)-fenyl]-3-(pyridin-3-yl)-propen-l-on,l-[2-hydroksy-4-(2-metylallyloksy)-fenyl]-3-{3-metoksyfenyl)-propen-l-on,l-[2-hydroksy-2-(prop-2-ynyloksy)-fenyl]-3-(pyridin-3-yl)-propen-l-on.
- 4. Farmasøytisk preparat, karakterisert ved at det omfatter en forbindelse med formel (A): hvor Ar representerer: fenyl som kan være usbustituert eller substituert med en, to- eller tre substituenter som uavhengig er valgt .fra Cl, Br, F, -OMe, N02, CF3, Cx.4 lavere alkyl, -NMe2, -NEt2, -SCH3, -NHCOCH3; 2-tienyl; 2-furyl; 3-pyridyl; 4-pyridyl eller 3-indolyl; R representerer: -OCHjR-l, hvor Rx er valgt fra -CH=CMe2, - CMe2=CH2, -C=CH; med den betingelse at når Ar representerer fenyl eller C4-alkylfenyl, er R annet enn 3-metyl-2-butenyloksy, og en farmasøytisk aksepterbar eksipiens.
- 5. Forbindelse for anvendelse som et farmasøytika, karakterisert ved at den er som angitt i ett eller flere av kravene 1 til 3.
- 6. Anvendelse av en forbindelse med formel (A) : hvor Ar representerer: fenyl som kan være usubstituert eller substituert med en, to eller tre substituenter som uavhengig er valgt fra Cl, Br, F, -OMe, N02, CF3, C1.4 lavere alkyl, -NMe2, -NEt2, -SCH3, -NHCOCH3; 2-tienyl; 2-furyl; 3-pyridyl; 4-pyridyl eller 3-indolyl; R representerer: -OCH^, hvor R er valgt fra -CH=CMe2, - CMe2=CH2, -C^CH; med den betingelse at når Ar representerer fenyl, er R annet enn 3-metyl-2-butenyloksy, for fremstilling av et medikament for behandling eller forebygging av neoplasmer, for behandling eller forebygging av menopausal lidelser eller for behandling eller forebygging av osteoporose.
