a) ホルムアルデヒド、1,3−シクロヘキサンジオン又は1,3−シクロペンタンジオン、及びアニリン(II):
b) 中間体(III)を酸化して、N−オキシド(IV):
c) N−オキシド(IV)を塩素化剤と反応させて、中間体(V):
d) 中間体(V)をアニリンArNH2(VI)と反応させて、式(I)の化合物を得る工程;
e) 得られた式(I)の化合物を酸で処理して、上記式(I)の化合物の塩を得る任意の工程。
6,7−(メチレンジオキシ)−3,4−ジヒドロアクリジン−1(2H)−オン(4)
3,4−(メチレンジオキシ)アニリン1(6.17g)、パラホルムアルデヒド3(1.35g)、及びシクロヘキサン−1,3−ジオン2(5.04g)のEtOH(220mL)の等分子(45mmol)混合物を、1.5時間還流した。溶媒の半分を減圧下で除去し、得られた固体をろ去した。このジヒドロ及び芳香族化合物の混合物(7.56g)のジクロロメタン(1.60L)溶液に活性化MnO2(純度85%)(6.74g)を加えた。室温で1.5時間撹拌した後、反応混合物をセライトパッドでろ過し、溶媒を除去し、白色粉末として分析的に純粋な4(7.22g,66%)を得た。mp: 210 ℃; 1H NMR (300 MHz, DMSO-d6): δ = 2.12 (q, J = 6 Hz, 2H, 3-H), 2.69 (t, J = 6 Hz, 2H, 2-H), 3.13 (t, J = 6 Hz, 2H, 4-H), 6.25 (s, 2H, O-CH2-O), 7.33 (s, 1H, 5-H), 7.50 (s, 1H, 8-H), 8.63 (s, 1H, 9-H).

アクリジン4(1.91g,7.9mmol)のジクロロメタン(100mL)溶液に、m−クロロ過安息香酸(純度70%)(3.91g,15.8mmol)を加えた。室温で72時間攪拌した後、1M水酸化ナトリウム溶液(160mL)を加え、反応混合物を10分間激しく攪拌した。次いで、水層をジクロロメタン(2x50mL)で抽出した。まとめた有機相を中性のpHとなるまで水で洗浄し、Na2SO4で乾燥し、ろ過した。次いで、溶媒を減圧下で除去し、黄色粉末として5(1.40g,68%)を得、さらに精製することなく次工程で用いた。試料を、元素分析のためにシリカゲルのフラッシュクロマトグラフィー(ジクロロメタン/メタノール95/5)で精製した。mp: 262 ℃; 1H NMR (300 MHz, DMSO-d6): δ = 2.13 (q, J = 6.2 Hz, 2H, 3-H), 2.67 (t, J = 6.2 Hz, 2H, 2-H), 3.18 (t, J = 6.2 Hz, 2H, 4-H), 6.31 (s, 2H, O-CH2-O), 7.63 (s, 1H, 8-H), 7.86 (s, 1H, 5-H), 8.23 (s, 1H, 9-H).
9−クロロ−6,7−(メチレンジオキシ)−3,4−ジヒドロアクリジン−1(2H)−オン(6)
アクリジンN−オキシド5(1.36g,5.2mmol)のジクロロメタン(250mL)溶液にPOCl3(5.9mL,63.6mmol)を加えた。48時間の還流後、水(100mL)を加え、次いで、8のpHになるまで飽和NaHCO3水溶液を加えた。水層をジクロロメタン(2x100mL)で抽出した後、有機相を水で洗浄し、Na2SO4で乾燥し、ろ過した。次いで、溶媒を減圧下で除去し、薄茶色粉末として6(1.33g、93%)を得、さらに精製することなく次工程で用いた。試料をシリカゲルのフラッシュクロマトグラフィー(ジクロロメタン)で精製した。Mp: 214 ℃; 1H NMR (300 MHz, DMSO-d6): δ = 2.21 (q, J = 6.4 Hz, 2H, 3-H), 2.81 (t, J= 6.4 Hz, 2H, 2-H), 3.25 (t, J = 6.4 Hz, 2H, 4-H), 6.19 (s, 2H, O-CH2-O), 7.29 (s, 1H, 5-H), 7.70 (s, 1H, 8-H).

クロロアクリジン6(698mg,2.53mmol)及び3,4,5−トリメトキシアニリン(7)(510mg,2.78mmol)のEtOH(350mL)混合物を、5時間還流した。溶媒を減圧留去し、得られた析出物をろ去し、次いでEtOHから再結晶し、淡黄色粉末としてSGR307.HCl(848mg、73%)を得た。mp: 244-246 ℃; 1H NMR (300 MHz, DMSO-d6): δ = 2.16 (q, J = 6.1 Hz, 2H, 3-H), 2.79 (t, J = 6.1 Hz, 2H, 2-H), 3.30 (t, J = 6.1 Hz, 2H, 4-H), 3.70 (s, 3H, 4’-OMe), 3.72 (s, 6H, 2 x OMe), 6.26 (s, 2H, O-CH2-O), 6.72 (s, 1H, 8-H), 6.75 (s, 2H, 2’-H et 6’-H), 7.56 (s, 1H, 5-H), 12.73 (s, 1H, NH), 15.15 (s, 1H, 10-H); HRMS (ES+): m/z calculated for C23H23N2O6[M+]: 423.1556, found: 423,1508.

内皮細胞の形態に対する化合物の効果を評価するために、ヒト臍帯静脈内皮細胞(HUVEC)の永久ヒト細胞株A549との融合物に由来するEAhy926内皮細胞株を用いた(Edgell et al, Proc.Natl.Acad.Sci.U.S.A., 1983, vol 80, issue 12, 3734-3737)。EAhy926細胞株は、親のHUVECの大部分の生化学的マーカーを発現するため、ネイティブHUVECの良い代表として考えられる(Bouis et al., Angiogenesis., 2001, 4, 91-102)。EAhy926細胞を、加湿雰囲気下(37℃、5%CO2)で、2mM L−グルタミン、10%ウシ胎児血清、100U/mLペニシリン及び100μg/mLストレプトマイシンを含むDMEM中で培養させた。急激に培養したEAhy926細胞を、5000細胞/100μL/ウェルで96ウェルプレートに播種した。播種の24時間後、培地を吸引し、試験化合物を含む100μLの培地を、細胞を含むウェルに加え(3重)、2時間インキュベートした(37℃、5%CO2)。最も形態学的に活性な化合物のために必要な場合に、用いる最高濃度は、100μMとし、それに続いて、低いナノモル濃度までの二倍段階希釈液を用いた。2時間のインキュベーション期間後、100X及び200Xの倍率で各ウェルの代表的な中央領域のデジタル写真を撮影した。対照の1%DMSO処理細胞で15%未満である通常の有糸分裂細胞を排除するために、結果を、フィールド内の15%を超える細胞のラウンディングアップをもたらし得る最小濃度として示す。通常、同一プレート上で3重の濃度で行った。CA4を正の内部標準として実験に含めた。
Eahy926内皮細胞を、6ウェルプレートのマトリゲル層に播種し、48時間マイクロ血管構造(索)を形成させた。索を、0.005ないし100μMの様々な濃度のSGR307.HCl及び1μMのコンブレタスタチンA4(CA4)に2時間曝した。本実験で用いた対照溶媒は、1%DMSO最終濃度であった。これは、この溶媒が正の内部対照として使用するコンブレタスタチンA4を可溶化するために必要であったためである。同一プレート領域のデジタル写真を、薬剤使用前及び二時間後に撮影した。
微小管のチューブリンの集合を、記載されているような96ウェルプレート型(Bane et al in Zhou, J. (Ed.), Microtubule Protocols. Humana Press, Totowa, New Jersey, 2007, 281-288; Barron et al., Anal. Biochem., 2003, 315, 49-56)で、蛍光色素DAPI(4’,6−ジアミジノ−2−フェニルインドール)(Bonne et al., J. Biol. Chem., 1985, 260, 2819-2825)を用いて行った。標準アッセイを以下のように行った:ウェルを、10μM DAPI及び様々な濃度の試験化合物を伴うPMEバッファー(100mM PIPES(1,4−ピペラジンビス(エタンスルホン酸));1mM MgSO4;2mM EGTA)中のチューブリン(細胞骨格、純度97%、最終濃度1mg/ml)で充填した。正の内部標準対照として30μMのコルヒチンを用いた。試験化合物の用いた最終濃度は、30μMで開始し、阻害が観察されなくなるまで3倍ずつ希釈して低下させた。それぞれの濃度につき三つのウェルで行った。室温で45分間プレインキュベーションした後、1mM GTP(5μL)を各ウェルに加え、チューブリン重合を開始し、次いで、プレートを37℃でさらなる2時間恒温Victorプレートリーダーに移した。次いで、蛍光を360nmの励起波長及び450nmの発光で読み取った。阻害割合を以下のように測定した:1−(ΔF(サンプル/ΔF(対照)X100、ここで、ΔF対照=F(阻害なし)−F(完結阻害)、ΔFサンプル=F(サンプル)−F(コルヒチンでの完結阻害)。チューブリン重合の化合物誘導阻害のIC50は、重合阻害の程度が最大値の50%となる濃度であり、非線形回帰プログラムSigmaPlot(Jandel Scientific)を用いて可変勾配を伴うS字状モデルに適合した薬剤濃度の対数の関数としての阻害割合の片対数プロットで決定される。これらの条件において、チューブリン重合のコルヒチン阻害のIC50値は0.36μMであった。
マウスB16黒色腫細胞、ルイス肺癌細胞及び結腸26癌細胞を、2mM L−グルタミン、10%ウシ胎児血清、100U/mLペニシリン及び100μg/mLストレプトマイシンを含むDMEM培地で培養した(37℃、5%CO2)。SGR307.HCl及びその他の本発明の化合物を、2.5mg/mLのストック濃度で水に溶解し、細胞培地でさらに希釈した。急激に培養した細胞を、100μLの培地中1ウェルあたり5000細胞で96ウェルプレートに播種した。播種の24時間後、0.01ないし100μMの範囲の最終濃度の試験化合物を含む100μlの培地を、細胞を含むウェルに加え(3重)、5%CO2にて37℃で48時間インキュベートした。48時間の試験化合物への曝露の期間の後、細胞生存率を、MTT(メチルチアゾールテトラゾリウム)試験(Scudiero et al., Cancer Res. 1988, 48, 4827-4833)を用いて評価した。吸光度を、マイクロプレートリーダー(BioKinetics Reader, EL340)にて562nmで読み取った。DMEMのみ及びMTTを伴う適切な対照を、バックグラウンド吸光度を減算するために用いた。50%まで細胞生存率を阻害する化合物の濃度(細胞の50%の阻害濃度、又はIC50)を、GraphPad Prismソフトウエアを用いて測定した。
ヒト細胞を、5%CO2下、75cm2フラスコ内で、ペニシリン、ストレプトマイシン及びアンフォテリシンB(Fungizone)の存在下、10%ウシ胎児血清(インビトロゲン)を補充したDMEM培地で培養した。細胞を、200μl培地中650細胞/ウェルの密度で96ウェル組織培養マイクロプレートに播種し、自動ベンチステーション(Beckman Biomek 3000)を用いて適用される0.5〜10μMの範囲の濃度でDMSOに溶解した化合物で24時間後に処理した。対照は、試験ウェルに対して同じ体積のDMSOを与えた(1%最終濃度)。72時間の曝露の時間後、MTS試薬(プロメガ)を加え、37℃で3時間インキュベートし、吸光度を490 nmで読み取った。結果を、比[(OD490処置/OD490対照)×100]で計算される細胞増殖阻害として示す。IC50(細胞増殖の50%阻害)の決定のために、実験は、2つの独立した2回の実験で行われた。
メスのC57BL/6及びBALB/cマウスを、Janvier(Le Genest Saint Isle、フランス)から購入した。マウスは、治療開始時に18gを上回る重量であった。食物及び水に自由に出入りさせた。マウスの腫瘍を、適切な血統のマウス系統の連続継代によって維持した。即ち、結腸腫瘍26(CT26)をBalb/cマウスに継代した(Corbett et al., Cancer Res. 1975, 35, 2434-24391975; Corbett et al., Cancer 1977 40, 2660-2680)。ルイス肺癌(Mayo et al, Cancer Chemotherapy Reports Part 2 Supplement, 1972, vol 3, issue 1, 325-330)がC57BL/6マウスで保持された。
SGR307.HClを、滅菌水、通常の食塩水又は0.5%デキストロースで希釈することにより調製し、体積200μlで腹腔内(i.p.)投与した。薬剤用量を、治療開始時の体重に基づいて調整した。用量反応評価を、各試験で行い、最高非毒性用量(HNTD)を測定した。それは、薬剤関連死を伴わず、20%以上の体重減少をもたらさない最高薬剤用量として定義される。動物の体重は腫瘍重量を含む。
腫瘍増殖阻害(T/C値)。これは、早期疾患に関して、抗腫瘍活性の測定で幅広く用いられる基準である。腫瘍容積を、処置群(T)及び対照群(C)で同日に測定した。対照群(C)の腫瘍容積中央値は、750ないし1500mm3の範囲に達した。その一方で処置群(T)の腫瘍容積中央値は、ゼロを含めて測定された。パーセントT/C値は以下のように計算される:
アメリカ国立癌研究所の標準に従って、T/C≦42%を、活性の最小レベルとみなす。T/C<10%を、さらなる開発を正当化する高い抗腫瘍活性レベルとみなす。
抗腫瘍活性を、南部諸州研究学会(SRI)で科学者によりはじめに定義されたようにLog細胞死滅値≧0.7を用いて評価した(Schabel et al., 1977)。以下のSRIスコアを用いて、以下のlog細胞死滅値に基づいて抗腫瘍活性を評価した:<0.7=−(不活性);0.7〜1.2=+;1.3〜1.9=++;2.0〜2.8=+++;>2.8=++++(高活性)。完全腫瘍退縮(CR)は、触診限界以下への腫瘍退縮として定義された(〜62mm3)。研究終了時に触診可能な腫瘍を有さない動物を、腫瘍を有さない生存個体(TFS)としてみなし、T−C値の計算から除外した。
SGR307.HCl及びその他の本発明の化合物の内皮細胞形態に対する効果
まず、本発明の化合物を、形質転換HUVEC(ヒト臍帯静脈内皮細胞)であるEAhy926内皮細胞に対して試験した。これらの細胞は、血管破壊剤(例えば、コンブレタスタチンA4)に短い2時間曝露した際にラウンディングアップを受ける。図1は、短い2時間の曝露時間で1μMの濃度にてSGR307.HClに曝露した内皮細胞の典型的なラウンディングアップを示す。内皮細胞の有意なラインディングアップが、0.06μMという低い濃度で観察された。
抗血管剤のその他の典型的な効果は、マトリゲル層上で増殖した内皮細胞の予め形成した索の破壊である。SGR307.HClの予め形成した内皮細胞索に対する効果を図2に示す。SGR307.HClによる内皮索の破壊は、2時間の曝露時間にて0.5μMという低い濃度で観察することができる。実際、SGR307.HClは、内部対照として含めた1μMのコンブレタスタチンA4と同様に効果的に内皮細胞索を破壊し得る。
内皮細胞のラウンディングアップ並びに内皮細胞索の破壊が、共に、細胞骨格構造の急速な破壊を示唆するため、次に、我々は、in vitroでのチューブリン重合に対する本発明の化合物の効果を試験した。図3は、チューブリン重合に対するSGR307.HClの効果を示す。SGR307.HClが1.09μMのIC50でチューブリン重合を阻害することが観察できる。同じ実験の条件で、標準的なチューブリン重合阻害剤ポドフィルロトキシン、コルヒチン及びコンブレタスタチンA4は、それぞれ、0.22、0.36及び0.29μMのIC50を示した。
SGR307.HClの細胞毒性活性を、3種のマウス腫瘍細胞、即ち、B16黒色腫、ルイス肺癌及び結腸26癌腫に対して試験した。表1で示される結果は、以下のIC50値を示す:B16、ルイス肺、及び結腸26細胞それぞれで4.5、10.6、及び1.2μM。
b48時間の曝露時間で50%細胞死滅をもたらすSGR307.HClの濃度。生存率は、MTT試薬を用いてアッセイした。
さらに、SGR307.HClの細胞毒性活性を、最も重要な癌タイプを含むヒト癌細胞株、並びに多剤耐性株、ホルモン感受性及び耐性株の大パネルを用いて評価した。
結腸26癌腫を有するマウスにおけるSGR307.HClの抗腫瘍活性
図4は、腫瘍移植後4、5、11、12、14及び15日目に31、50及び80mg/kgの薬剤用量で腹腔内注射(i.p.)した結腸26を有するマウスにおけるSGR307.HClの抗腫瘍作用を示す。図4−Aで示すことができるように、80mg/kgの用量は、20%を上回る体重減少をもたらす毒性があった。その一方で、31及び50mg/kgは、毒性がなく、この研究の間に体重増加をもたらした。

次いで、抗腫瘍活性を、ルイス肺癌を有するマウスで試験した。図5に示されたデータは、2週連続で週に5日間腹腔内投与した場合に、50及び55mg/kgの用量での重大な増殖遅延を示す。

pKA、B16黒色腫に対する細胞毒性(細胞毒性B16)活性、修飾HUVEC(EA.hy926)(CmarrEA.hy926)に対する形態学的効果(ラウンディングアップ)、及び化合物SGR307.HClのチューブリン重合阻害(IPT)を、先行技術の化合物及びその他の密接に関連する化合物のものと比較した(下記表5参照)。B16黒色腫に対する細胞毒性、チューブリン重合、並びに内皮細胞形態を評価するために用いられる試験は、実施例2に記載されたものである。

6SGR305は窒素上でプロトン化することができない
− Xの−CH2−又は−CH2CH2−、好ましくは−CH2CH2−が、化合物の抗血管活性を向上させる。
− R2及びR3が、一緒になって、好ましくは、架橋基−O−CH2−O−又は−O−CH2−CH2−O−を形成し、より好ましくは架橋基−O−CH2−O−を形成することにより、チューブリン阻害特性及び腫瘍細胞に対する細胞毒性を向上させる。さらに、架橋基−O−CH2−O−及び−O−CH2−CH2−O−は、プロトン化の計算pKaで測定されたように血漿溶解度を増加させる。
結論として、合成が容易でもある水溶性化合物SGR307.HClの主な生物学的特性としては、以下のものが挙げられる:
1.in vitroにてナノモル濃度で独自の作用機構、即ち、内皮細胞のラウンディングアップ及び内皮索の破壊を示す;
2.臨床的に用いられるポドフィルロトキシン類似体(即ち、トポイソメラーゼII阻害剤(テニポシド又はVM26、エトポシド又はVP16、及びエトポシドのプロドラッグであるエトポホス))と比べて、強力なチューブリン重合阻害剤である;
3.マウス癌細胞に対してマイクロモル濃度で細胞毒性がある;
4.いくつかのヒト腫瘍細胞株に対してナノモル濃度でin vitroにて高活性である;
5.ヒト多剤耐性(MDR)癌細胞に対して活性である;
6.シスプラチン耐性ヒト癌細胞株に対して活性である;
7.共に薬剤非感受性腫瘍としてみなされる結腸26癌腫及びルイス肺癌を含む充実性腫瘍を有するマウスにおいて、優れたin vivo抗腫瘍活性を示す。
Claims (19)
- 式(I):
Xは、−CH2−又は−CH2CH2−を示し;
R1及びR4は、独立して、H、ハロゲン、C1−C4−アルキルからなる群から選ばれ;
R2及びR3は、独立して、H、OH、C1−C4アルキル、C1−C4−アルコキシ、C1−C4−アルキルチオ、ハロゲン、トリフルオロメチル、CNからなる群から選ばれるか;
或いは、R2及びR3が、一緒になって、−(CH2)n−及び−O−(CH2)m−O−からなる群から選ばれる架橋基を形成し、nは、3ないし4の整数であり、mは、1又は2であり;
Arは、独立して、C1−C4−アルキル、OH、C1−C4−アルコキシ、C1−C4−アルキルチオ、ハロゲン、トリフルオロメチル、CNからなる群から選ばれる1〜4個の置換基で置換されていてもよいフェニル又はナフチル基を示す。]

- R2及びR3が、一緒になって、架橋基−O−(CH2)m−O−を形成し、mが、1又は2であり、R1及びR4が、共に、Hを示す請求項1又は2に記載の化合物。
- Xが、−CH2CH2−を示し、R1及びR4が、共に、Hを示す前記請求項のいずれかに記載の化合物。
- Arが、2又は3個のC1−C4−アルコキシ基で置換されたフェニル基を示す前記請求項のいずれかに記載の化合物。
- Arが、3,4,5−トリメトキシフェニル基である前記請求項のいずれかに記載の化合物。
- 上記式(I)の化合物が:
- 請求項1ないし8の何れかに記載の化合物、及び医薬上許容される賦形剤を含む医薬組成物。
- 組成物の総重量に対して0.0.1%ないし10重量%の上記式(I)の化合物を含む請求項9に記載の組成物。
- 非経口又は経口経路で投与するための組成物として製剤化される請求項9又は10の何れかに記載の組成物。
- 医薬として使用するための、請求項1ないし8の何れかに記載の化合物、又は請求項9ないし11の何れかに記載の組成物。
- 医薬として、同時に、交互に又は連続的に使用するための、少なくとも:
− 請求項1ないし8の何れかに定義された上記式(I)の化合物(好ましくは、その医薬上許容される塩であり、溶媒和物の形態であってもよい。)、及び医薬上許容される賦形剤を含む第一組成物、及び
− シスプラチン、メトトレキサート、シクロホスファミド、ドキソルビシン、フルオロウラシルのようなその他の抗腫瘍剤を含む第二組成物
を含むキット。 - 細胞毒性薬又は抗血管剤として使用するための、請求項12に記載の化合物又は組成物、又は請求項13に記載のキット。
- 癌の治療に使用するための、請求項12又は14に記載の化合物又は組成物、又は請求項13又は14に記載のキット。
- 癌が、黒色腫、肺癌、結腸癌、乳癌、脳腫瘍、膵臓癌、白血病、前立腺癌、リンパ腫、及び肝癌からなる群から選ばれる請求項15に記載の使用のための化合物又は組成物又はキット。
- 癌が、多剤耐性癌である、請求項15又は16に記載の使用のための化合物又は組成物又はキット。
- 単独で、又は電離放射線又は非電離放射線又は温熱療法と同時に、別々に又は順次組み合わせて使用する請求項15ないし17の何れかに記載の使用のための化合物又は組成物又はキット。
- 以下の連続した工程を含む、請求項1ないし8の何れかに記載の化合物の調製方法:
a) ホルムアルデヒド、1,3−シクロヘキサンジオン又は1,3−シクロペンタンジオン、及びアニリン(II):
b) 中間体(III)を酸化して、N−オキシド(IV):
c) N−オキシド(IV)を塩素化剤と反応させて、中間体(V):
d) 中間体(V)をアニリンArNH2(VI)と反応させて、式(I)の化合物を得る工程;
e) 得られた式(I)の化合物を酸で処理して、上記式(I)の化合物の塩を得る任意の工程。
