Description
Introduksjon
I de siste år har selektive serotoninreopptaks-inhibitorer (SSRI) begynt å bli anvendt for behandling av depresjons- og andre seritralnervesystemsykdommer, og spesielt blant disse er fluoksetin, citalopram, sertralin og par-oksetin. De har alle forskjellig kjemisk struktur som hjelper til å forklare deres forskjellige metabolske og farmakokinet-iske profiler. Deres ytelse som antidepressive midler kan sammenlignes med den til de klassiske trisykliske forbindelser, men deres fordel er at de er sikrere og tolereres bedre.
Foreliggende oppfinnelse angår et utall nye 4-substituerte piperidiner som har en aryloksyfunksjonalitet og som kraftig inhiberer serotonin- og/eller noradrenalinreopp-tak som et resultat av deres høye affinitet for deres neuron-ale transportører. Dette karakteristiske trekk gir dem et forøkt antidepressivt potensial i human terapi. Andre potensielle terapeutiske anvendelser av disse forbindelser er behandling av nervøs bulimi, alkoholavhengighet, angst, obsessiv-kompulsiv sykdom, panikk, smerte, premenstruelt syndrom og sosial fobi, så vel som migreneprofylakse. Biblio-grafi beskriver også andre piperidinderivater med aryloksy-funks jonalitet som potensielle antidepressive midler, dog med en kjemisk natur som avviker vesentlig fra de her krevede, da piperidinet er substituert ved 3-stillingen. Dette er f.eks. tilfellet for slike forbindelser som 3-[{2-metoksyfenoksy)-fenyllmetylpiperidin 1 (Melloni, P., Carniel, G., Deila Torre, A., Bonsignalari, A., Buonamici, M.( Pozzi, 0., Ricciardi, S., Rossi, A.C., Eur. J. Med. Chem. Chim. Ther. 1984, 3, 235-242; Melloni, P., Deila Torre, A., De Munari,
S., Meroni, M., Tonani, R-, Gazetta Chimica Italiana 1985, 11S, 159-163) og 3-[(fenoksy)fenyl]metylpiperidin 2 (FR

Fremstilling og/eller isolering av rene enantiomerer er ikke beskrevet i noe tilfelle. Forbindelsene krevet i foreliggende beskrivelse, utviser imidlertid et enkelt kiralt senter da de har piperidinringen substituert ved 4-stilling. De er blitt fremstilt som racemiske blandinger og som rene enantiomerer under anvendelse av syntesemetoder som avviker fra de som ble anvendt ved fremstilling av 1 og 2.
Enn videre er andre piperidinderivater som har aryloksyfunksjonalitet og piperidinringen substituert ved 4-stilling, blitt beskrevet som potensielle antidepressive midler {formel 3. og 4) . Når det gjelder type 3.-forbindelser (JP 96 40.999 CA124: 343333n),
er aryloksygruppen direkte forbundet til piperidinringen, mens i type 4-forbindelser (JP 96 40.999 CA124: 343333n) er angitte gruppe forbundet til piperidinringen via en metylen-gruppe som ikke har noen ytterligere substitusjoner. Forbindelsene beskrevet her, avviker sterkt fra disse, da de har aryloksygruppen forbundet til piperidinringen via en metylen-gruppe hvori, i alle tilfeller, én av metylengruppehydro-genene er substituert med en arylgruppe som er substituert eller ikke-substituert som definert i det etterfølgende.
Disse forbindelser er derfor strukturelt forskjellige fra type 3_ og 4, og syntesemetodikken anvendt ved fremstilling av disse, er også fullstendig forskjellig.
Beskrivelse
De nye 4-substituerte piperidiner beskrevet ifølge foreliggende oppfinnelse, er kjennetegnet ved at de har generell formel (I)
hvori Ri og R2 er ikke-substituerte arylradikaler eller arylradikaler som er mono- eller polysubstituert med halogen (fluor, klor, brom, jod) , alkyl, alkoksy, cyan, tri f luonret-oksy, trifluormetyl, benzoyl, fenyl, nitro, amino, aminoalkyl, aminoaryl og karbonylamino, og deres farmasøytisk akseptable salter med uorganiske og organiske syrer.
Forbindelsene av generell formel (I) har et asym-metrisk senter og er blitt fremstilt som racemiske blandinger og som rene enantiomerer. Foreliggende oppfinnelse innbefatter alle optiske isomerer av forbindelsene av generell formel (I) og racemiske blandinger derav. Foreliggende oppfinnelse omfatter også de farmasøytisk akseptable salter av disse forbindelser med uorganiske syrer (slik som saltsyre, hydro-bromsyre, salpetersyre, svovelsyre og fosforsyre) og med organiske syrer {slik som eddiksyre, fumarsyre, vinsyre, oksalsyre, sitronsyre, p-toluensulfonsyre og metansulfonsyre).
De racemiske forbindelser av generell formel (I) fremstilles under anvendelse av velkjente syntesemetoder som starter med forbindelsene av generell formel (II).
Dannelsen av alkylaryletergruppen utføres under anvendelse av Mitsunobu-reaksjonen (Mitsunobu, 0., Synthesis 1981, 1; Hughes, D.L., Organic Reactions, 42, 335) med fenoler R2-0H, hvori R2 er et arylradikal som er substituert eller ikke, som beskrevet for generell formel (I), og forbindelsene av generell formel (II) hvori Ri er et arylradikal som er substituert eller ikke, som beskrevet for generell formel (I), og R3 er hydrogen eller R4, som er et alkoksykarbonylradikal, fortrinnsvis etoksykarbonyl og t-butoksykarbonyl.
Alkylaryletergruppen fremstilles også under anvendelse av en aromatisk nukleofil substitusjonsreaksjon (Berg-lund, R.A., Org. Proe. Res. Dev. 1997, I, 328-330) med forbindelsene av generell formel (II) definert ovenfor, og de fluorerte derivater R2-F, hvori R2 er et arylradikal som er mono- eller polysubstituert med halogen (fluor, klor, brom, jod), alkyl, alkoksy, cyan, trifluormetoksy, trifluormetyl, benzoyl, fenyl, nitro, amino, aminoalkyl, aminoaryl og karbonylamino. Forbindelsene av generell formel (II) fremstilles under anvendelse av konvensjonelle syntesemetoder som starter med forbindelsene av generell formel (III) (Duncan, R.L., Helsley, G.C., Welstead, W.J., DaVanzo, J.P., Funderburk, W.H., Lunsford, C.D., J. Med. Chem. 1970, 13 (1), 1), hvori R5 er et acetyl radi kal, etoksykarbonyl og R6 er cyan eller karboksy.
Forbindelsene av generell formel (III) definert ovenfor, overføres til forbindelsene av generell formel (IV), hvori Ri er et arylradikal som er substituert eller ikke, som-beskrevet for forbindelsene av generell formel (I), og R? er hydrogen, acetyl eller R4, som er et alkoksykarbonylradikal, fortrinnsvis etoksykarbonyl og t-butoksykarbonyl. Slik omdannelse foretas under anvendelse av to reaksjonstyper: a) en Friedel-Crafts-reaksjon av syrekloridene avledet fra forbindelsene av generell formel (III), hvori Rs er acetyl eller etoksykarbonyl og R6 er karboksy (Duncan, R.L., Helsley, G.C., Welstead, W.J., DaVanzo, J.P., Funderburk, W.H., Lunsford, C.D., J. Med. Chem. 1970, 13 (1), 1) med benzen eller hen-siktsmessig funksjonaliserte derivater derav; eller b) en Grignard reaktiv addisjonsreaksjon, fremstilt fra hensikts-messig funksjonaliserte arylhalogenider, til forbindelser av generell formel (III) hvori R5 er acetyl, etoksykarbonyl eller t-butoksykarbonyl og R6 er cyan (Duncan, R.L., Helsley, G.C., Welstead, W.J., DaVanzo, J.P., Funderburk, W.H., Lunsford, C.D., J. Med. Chem. 1970, 13 (1), 1). Reduksjon av forbindelsene av generell formel (IV) gir alkoholene av generell formel (II) definert ovenfor.
Enantiomerene som utgjør de racemiske blandinger av generell formel (I), erholdes under anvendelse av to forskjellige veier: a) oppløsning av den tilsvarende racemiske blanding ved splittkrystallisering av de diastereomere salter fremstilt med kirale syrer (D- eller L-dibenzoylvinsyre, D- eller L-vinsyre, D- eller L-di-p-toluylvinsyre og D- eller L-mandel-syre) og b) enantioselektiv syntese. I det sistnevnte tilfelle erholdes enantiomerer av generell formel (I) ved omsetning av fenoler Ra-0H eller de fluorerte, aromatiske derivater R2-F definert ovenfor, med enantiomerene av alkoholene av generell formel (II) som beskrevet for de racemiske blandinger av generell formel (I). I enantiomerene av alkoholene av generell formel (II) er Ri et arylradikal som er substituert eller ikke, som definert for forbindelsene av generell formel (I), og R3 er hydrogen eller R4, som er et alkoksykarbonylradikal, fortrinnsvis etoksykarbonyl og t-butoksykarbonyl. Enantiomerene av alkoholene av generell formel (II) definert ovenfor, erholdes ved enantioselektiv reduksjon (Ramachandran, P.V., Teodorovic, A.V., Rangaishenvi, M.V., Brown, H.C., J. Org. Chem. 1992, 57, 2379-2386) av forbindelsene av generell formel (IV) (Duncan, R.L., Helsley, G.C., Welstead, W.J., DaVanzo, J.P., Funderburk, W.H., Lunsford, C.D., J. Med. Chem. 1970, 13 (1), 1), hvori Ri er et arylradikal som er substituert eller ikke, som definert for forbindelsene av generell formel (I), og R? er hydrogen eller R4, definert ovenfor.
Den farmakologiske aktivitet av forbindelsene av generell formel (I) ble bestemt under anvendelse av vel etab-lerte in vitro og in vivo farmakologiske tester. Affiniteten-av forbindelsene for serotoninreopptaksreseptorene (5HT) ble evaluert i full rottecerebral cortex under anvendelse av [3H] - paroksetin som radioligand (Habert, E., Graham, D., Tahraoui, L., Claustre, Y., Langer, S.Z., Eur. J. Pharmacol. 1985, 118, 107-114), og som ga Ki-verdier varierende mellom 0,5 og 500 nmol/1. Affiniteten av forbindelsene for noradrenalin-(NA) reopptaksreseptorer ble evaluert i full rottecerebral cortex under anvendelse av [<3>H]-nisoksetin som radioligand (Tejani-Butt, S.M., J. Pharmacol. Exp. Ther. 1992, 260, 1, 427-436) som ga Ki-verdier mellom 1 og 500 nmol/1. De etter-følgende ble anvendt som utprøvninger som forutsier antidepressiv aktivitet: musehalesuspensjon (Stéru, L., Chermat, R., Thierry, B., Mico, J.A., Lenégre, A., Stéru, M., Simon, P., Porsolt, R.D., Prog. Neuropsychopharmacol. Biol. Psychiat. 1987, 11, 659-671), desperat atferd hos rotter eller mus (Porsolt, R.D., Anton, G., Blavet, N., Jalfre, M., Eur. J. Pharmacol. 1978, 48, 379-394) og økning av rotteyohimbin-fremkalt letalitet (Quinton, R.M., Brit. J. Pharmacol. 1963, 21, 51-66) . Forbindelsene med Ki-verdier som varierte mellom 0,5 og 40 nmol/1 for én av transportørene eller for begge, utviste en glimrende antidepressiv aktivitet i de modeller når de ble administrert innen 1 til 30 mg/kg-området oralt, intraperitonealt eller subkutant.
I en artikkel av Melloni et al. (Eur. J. Med. Chem. 19( 3), 235-242 (1984) er følgende typer av forbindelser beskrevet:
a) Forbindelser med en åpen struktur (I) som overhodet ikke er lik forbindelsene ifølge foreliggende søknad. b) Forbindelser med en syklisk struktur (II) avledet fra morfolin, og således fullstendig forskjellig fra piperidin-derivatene ifølge foreliggende søknad. Morfolingruppen er også til stede i forbindelser av formel (IV) som er trisykliske forbindelser og må også anses å være fullstendig forskjellig fra foreliggende forbindelser. c) Fire forbindelser med struktur III er beskrevet (se tabell III, side 238), men bare én av disse, forbindelse 62 (X =
CH2), har en piperidingruppe. Denne piperidingruppen er imidlertid substituert i 3-stilling av ringen og har således to kirale sentre som gir opphav til to par diastereoisomerer. Foreliggende 4-piperidinderivater kan ikke anses å være stillingsisomerer av 3-(aryloksyalkyl)piperidinene ifølge den kjente teknikk på grunn av de spesielle karakteristika av piperidinringen, uten noen plankonformasjon og med et nitrogenatom med det eneste elektronpar som er i stand til å interagere med atomer av kjemiske grupper bundet til piperidinringen. Substitusjoner på 3- eller 4-stillingene gir forbindelser med klart forskjellige steriske konformasjoner og også forskjellige egenskaper med hensyn til de farmakologiske reseptorer på grunn av vesentlige forskjeller i romlig kobling og elektronisk ladningsfordeling.
Med hensyn til den farmakologiske aktivitet er bare et morfolinderivat beskrevet av Melloni et al., med en klar forskjellig kjemisk struktur i forhold til foreliggende piperidiner, angitt å utvise glimrende aktivitet i reserpin-testen som ble utført. Resten av disse "gave less active or inactive compounds" (se side 23 9).
I Gazzetta Chimica Italiana, 115, 159-163 (1985) er enn videre samme type av 3-(aryloksyalkyl)piperidiner (blant andre forbindelser) beskrevet, og den farmakologiske aktivitet ble studert av Melloni et al. Forfatterne angir at "The compounds described in this paper were found to be inactive aa potential antidepressant agents" (se side 161). Så vidt søkeren kjenner til, har ingen forbindelse beskrevet i de ovennevnte publikasjoner noen gang blitt markedsført som en antidepressiv aktiv farmasøytisk bestanddel.
Når den farmakologiske aktivitet av de 4-substituerte piperidiner ifølge foreliggende oppfinnelse ble undersøkt, utviste på den annen side forbindelsene over-raskende og uventet glimrende dobbelt serotonin- og nor-adrenalinreopptaksinhiberende aktivitet, hvilket gir dem en unik dobbelt virkningsmåte som antidepressive midler i denne familie av molekyler. Den etterfølgende tabell viser de erholdte uventede resultater for flere av forbindelsene.
De etterfølgende eksempler illustrerer oppfinnelsens ramme.
Eksempel 1
{+/-)- 4-[( 4- trifluormetoksyfenoksy)- 2-( 4- fluorfenyl)] metylpiperidin- fumarat
En blanding av (+/-)-4-[(4-trifluormetoksyfenyl)-hydroksy]metyl-l-piperidinkarboksylsyre-1,1-dimetyletylester (2,25 g, 7,27 mmol), 2-pyridyldifenylfosfin (1,90 g,
7,27 mmol) og 1,3 g (7,4 mmol) 4-trifluormetoksyfenol i 40 ml tetrahydrofuran (THF) ble behandlet med en løsning av 1,15 ml dietyl-aza-dikarboksylat (DEAD) i 10 ml THF. Reaksjonsblandingen ble omrørt ved 20 °C i 4-6 timer og ble konsentrert. Residuet ble oppløst i etyleter, ble vasket med 10% vandig HCl-løsning og 5% vandig NaOH-løsning, ble tørket (vannfritt Na2S04) , filtrert og konsentrert. 2,4 g (71%) ble erholdt som en olje som ble oppløst i 50 ml diklormetan og behandlet med en løsning av 2,1 ml trifluoreddiksyre i 10 ml diklormetan. Etter 20 timer ved 20 °C ble denne vasket med en 5% vandig NaOH-løsning og mettet, vandig NaCl-løsning. Tørking (vannfritt Na2S04) , filtrering og konsentrering ga 1,3 g (71%) av produktet, som ble suspendert i 60 ml vannfri eter og behandlet med 0,42 g fumarsyre under dannelse av 1,0 g av fumaratet (60% utbytte) med et smp. = 130-134 °C. RMN^H (DMSO-ds) utviste et karakteristisk signal ved 4,31 ppm (d, J = 5,9 Hz, 1H, CHOAr) og RMN-<13>C (DMSO-d6) utviste ved 74,9 ppm et signal svarende til CHOAr-karbon.
Eksempel 2
(+/-)- 4- t( 4- fluorfenoksy)( 4- fluorfenyl)] metylpiperidin- hydro-klorid
En blanding av {+/-)-4-[(4-fluorfenyl)hydroksy]-metyl-1-piperidinkarboksylsyre-1,1-dimetyletylester
(16,33 mmol) og 1,9 g 4 fluorfenol i 50 ml THF ble behandlet med 5,0 g trifenylfosfin, og 3,45 ml DEAD-løsning i 10 ml THF ble deretter tilsatt. Etter 3 timer ble løsningsmidlet des-tillert, og den resulterende olje ble behandlet med heksan under dannelse av et bunnfall som ble filtrert. Filtratet ble konsentrert, og residuet ble oppløst i 100 ml diklormetan og
behandlet med 8 ml av en trifluoreddiksyreløsning i 30 ml diklormetan. Etter 15 timer ble reaksjonsblandingen opp-arbeidet som vanlig, og hydrokloridet ble fremstilt i THF under dannelse av 3,6 g derav som et amorft og svakt hygroskopisk, rosefarget fast materiale (utbytte: 70%) og med et smp. = 90 °C (d) . RMN-^-H (CDC13) av hydrokloridet utviste et karakteristisk signal ved 4,72 ppm (d, J = 5,8 Hz, CHOAr) og RMN-<13>C (CDC13) et signal ved 83,1 ppm svarende til CHOAr-karbon.
Følgende forbindelser ble analogt fremstilt: (+/-)-4-[(4-fluorfenoksy)(4-klorfenyl)]metylpiperidin-hydroklorid (54% utbytte, hygroskopisk),
(+/-)-4-[(4-metoksyfenoksy)(4-fluorfenyl)]metylpiperidin-fumarat (60% utbytte, smp. = 139-142 °C) ,
(+/-)-4-[(4-trifluormetylfenoksy)fenyl]metylpiperidin-hydroklorid (36% utbytte, hygroskopisk),
(+/-)-4-[(fenoksy)(4-klorfenyl)]metylpiperidin-hydroklorid (72% utbytte, smp. = 80 °C (d) ) ,
(+/-)-4-[(4-benzoylfenoksy)fenyl]metylpiperidin-hydroklorid (74% utbytte, smp. 70 °C (d)), og
(+/-)-4-[(4-trifluormetoksy)fenyl]metylpiperidin-fumarat (58% utbytte, smp. = 76 °C (d) ) .
Eksempel 3
(+/-)- 4 -[( 4- fluorfenoksy) fenyl] metylpiperidin- sulfat
En NaH-suspensjon (1,95 g, 60% mineralvann) i 20 ml dimetylsulfoksid (DMSO) ble behandlet med en løsning av (+/-)-4-(fenylhydroksy)metyl-1-piperidinkarboksylsyre-1,1-dimetyletylester (13,8 g, 47 mmol) i 36 ml DMSO. Kaliumbenzoat (7,5 g, 47 mmol) og 1,4-difluorbenzen (6,1 ml, 56 mmol) ble tilsatt, og reaksjonsblandingen ble oppvarmet til 85 °C inntil utgangssubstansen forsvant. Denne ble deretter behandlet med mettet, vandig NaCl og vannløsning og ble ekstrahert med etylester. Fordampningsresten av den organiske fase ble behandlet med 200 ml metanol og 200 ml 10% vandig HCl-løsning og ble kokt under tilbakeløpskjøling i 1 time. Produktet ble isolert under anvendelse av vanlig metodikk, under dannelse av en olje (9,6 g, 72% utbytte). RMN^H(CDC13) utviste et signal ved 4,70 ppm (d, J = 7,1 Hz, CHOAr) og RMN-<13>C (CDC13) et signal ved 85,0 ppm svarende til CHOAr-karbon. Oljen ble behandlet med 1,85 ml konsentrert H2S04-løsning i 90 ml vann under dannelse av sulfatet som et fast materiale med et smp. = 118-120 °C (75% utbytte).
Eksempel 4
(+/-)- 4-[( 3- fluorfenoksy) fenyl] metylpiperidin- sulfat
En NaH-suspensjon (0,40 g, 60% mineralvann) i 6 ml DMSO ble behandlet med en løsning av (+/-)-4-(fenylhydroksy)-metyl-1-piperidinkarboksylsyre-l,1-dimetyletylester (2,55 g, 8,75 mmol) i 6 ml DMSO. Kaliumbenzoat (1,35 g, 8,43 mmol) og 1,3-difluorbenzen (1,05 ml, 10,6 mmol) ble tilsatt, og reaksjonsblandingen ble oppvarmet til 85 °C inntil utgangsforbind-elsen forsvant. Den ble deretter behandlet med mettet, vandig NaCl og en vannløsning og ble ekstrahert med etylester. Fordampningsresten fra den organiske fase ble behandlet med 30 ml metanol og 30 ml av en 10% vandig HCl-løsning og ble kokt under tilbakeløpskjøling i 1 time. Vanlig opparbeidelse ga 2,16 g av en ravfarget olje (88% utbytte). RMN-<1>H (CDC13) utviste et signal ved 4,78 ppm (d, J = 6,4 Hz, 1 H, CHOAr) og RMN-<13>C (CDC13) et signal ved 84,6 ppm svarende til CHOAr-karbon. Oljen ble behandlet med 0,20 ml konsentrert H2S04-løsning i 10 ml vann under dannelse av sulfatet som et fast materiale med et smp. = 72-76 °C.

Oppløsning av (+/-)- 4-[( 3- fluorfenoksy) fenyl] metylpiperidin 4,45 g L-(-)-dibenzoylvinsyre ble tilsatt til 7,1 g (25 mmol) (+/-)-4-[(3-fluorfenoksy)fenyl]metylpiperidin opp-løst i 175 ml 96% etanol. Et hvitt, fast materiale ble erholdt (smp. = 212 °C (d)) som ble behandlet med 5% vandig NaOH-løs-ning og ekstrahert med kloroform, under dannelse av den venstredreiende isomer (96% ee, smp. = 59-62 °C [a] S46 - 11,4, c = 0,576, CHC13) .
De erholdte filtratvæsker ble konsentrert, og den frie base ble ekstrahert ved behandling med 5% vandig NaOH-løsning og kloroform. Det erholdte produkt oppløst i etanol, ble behandlet med D-{+)-dibenzoylvinsyre under anvendelse av den foregående prosedyre. Et hvitt, fast materiale ble erholdt (smp. = 208 °C (d)) som ble behandlet med 5% vandig NaOH-løsning og ekstrahert med kloroform under dannelse av den høyredreiende isomer (98% ee, smp. = 59-62 °C, [ct] 546 + 11, 4 , c = 0, 618, CHCI3) .

Eksempel 6
(+)- 4-[( 4- fluorfenoksy) fenyl] metylpiperidin
4-benzoylpiperidin (2,0 g, 10,6 mmol) ble tilsatt til en løsning av 6,8 g (+)-B-klordiisopinocanfeilboran ((+)-DIP-C1) (21,25 mmol) i 20 ml tørr diklormetan avkjølt til 3-4 °C. Etter omsetning i 72 timer ble 2,0 ml acetaldehyd (35,46 mmol) tilsatt, og blandingen ble omrørt ved romtemperatur i 3 timer. 24 ml av en vandig 6 N NaOH-løsning, diklormetan og mettet, vandig NaCl-løsning ble tilsatt. Fasene ble separert, og behandling av den organiske fase på vanlig måte ga (+)-a-fenyl-4-piperidinmetanol som et hvitt, fast materiale med et smp. = 64- 66 °C i 90% utbytte (84% ee).
1,8 g av aminoalkoholen (+)-a-fenyl-4-piperidin-metanol (9,6 mmol) ble oppløst i 10 ml metanol. Løsningen ble avkjølt til 0 °C, og en ditert.-butyldikarbonat- ((Boc)20)
(2,5 g, 11,27 mmol) løsning ble dråpevis tilsatt til 10 ml metanol. Blandingen ble omrørt i 24 timer ved romtemperatur,
metanolen ble konsentrert, vann ble tilsatt, og blandingen ble ekstrahert med diklormetan. Vanlig behandling av den organiske fase ga den ønskede alkohol som en svakt farget olje i 93% utbytte.
Alkoholen fremstilt ovenfor (2,7 g, 9,3 mmol) opp-løst i 25 ml DMSO, ble tilsatt til en NaH-suspensjon (60%, "0,6 g) i 5 ml DMSO. Kaliumbenzoat (1,53 g, 9,63 mmol) og 1,4-difluorbenzen (1,3 ml, 11,9 mmol) ble tilsatt, og blandingen ble oppvarmet (70-75 °C) inntil utgangssubstansen forsvant. Reaksjonsblandingen ble helt over i vann og mettet, vandig NaCl-løsning og ble ekstrahert med eter. Den erholdte olje ble kokt under tilbakeløpskjøling med en blanding av 40 ml metanol og 40 ml vandig saltsyreløsning i 1 time. Isolering av produktet under anvendelse av vanlige metoder ga (+)-4-[(4-fluorfenoksy)fenyl]metylpiperidin som en olje i 54% utbytte. Behandling av 0,5 g (1,75 mmol) av denne olje med D-dibenzoylvinsyre i etanol (96%, 30 ml) ga et bunnfall som ble filtrert (smp. = 198-199 °C) . Aminoeteren ble frigjort under dannelse av et hvitt, fast materiale med 96% ee, smp. = 102-104 °C, og [a] 546 + 15, c = 0,105, CHCI3.
Følgende forbindelser ble analogt fremstilt: (+)-4-[(4-nitrofenoksy)fenyl]metylpiperidin-hydro-klorid (96% ee, smp. = 55 °C (d) , [a] 436 + 36 , c = 0,045, etanol),
(-)-4-[(1-naftyloksy)fenyl]metylpiperidin-hydro-klorid (98% ee, smp. = 65 °C (d) , [a] S46 - 180, c = 0,080, CHC13) og
(+)-4-[(2-fluorfenoksy)fenyl]metylpiperidin-sulfat (97,6% ee, smp. = 105 °C (d) , [a] 546 + 31, c = 0,081, CHC13) .
Eksempel 7
{-)- 4-[( 4- fluorfenoksy) fenyl] metylpiperidin
4-benzoylpiperidin (7,35 g, 39,05 mmol) ble tilsatt til en løsning av 25 g (-)-DIP-Cl (78,125 mmol) i diklormetan (75 ml, tørr) avkjølt til 0-2 °C. Etter omsetning i 72 timer ble 5,2 ml acetaldehyd (92,2 mmol) tilsatt, og blandingen ble omrørt ved romtemperatur i 3 timer. 71 ml av en vandig 6 N NaOH-løsning, diklormetan og mettet, vandig NaCl-løsning ble tilsatt. Fasene ble separert, og vanlig behandling av den
organiske fase ga (-)-a-fenyl-4-piperidinmetanol som et hvitt, fast materiale med et smp. = 48-50 °C i 85% utbytte (86% ee). 2 g av aminoalkoholen (-)-a-fenyl-4-piperidin-metanol (10,7 mmol) ble oppløst i 10 ml metanol. Løsningen ble avkjølt til 0 °C, og en (Boc)20- (2,6 g, 11,73 mmol) løsning ble dråpevis tilsatt til 7 ml metanol. Blandingen ble omrørt i 20 timer ved romtemperatur, metanolen ble konsentrert, vann ble tilsatt, og blandingen ble ekstrahert med diklormetan. Vanlig behandling av den organiske fase ga den ønskede alkohol som en svakt farget olje i 90% utbytte.
Alkoholen fremstilt ovenfor (1,3 g, 4,5 mmol) opp-løst i 10 ml DMSO, ble tilsatt til en NaH-suspensjon (60%, 2,10 g) i 5 ml DMSO. Kaliumbenzoat (7,15 g, 4,5 mmol) og 1,4-difluorbenzen (0,75 ml, 6,86 mmol) ble tilsatt, og blandingen ble oppvarmet (70-75 °C) inntil utgangssubstansen forsvant. Reaksjonsblandingen ble helt over i vann og mettet, vandig NaCl-løsning og ble ekstrahert med eter. Den erholdte olje ble kokt under tilbakeløpskjøling med en blanding av 17 ml metanol og 17 ml vandig saltsyreløsning i 1 time. Vanlig opparbeidelse av reaksjonsblandingen ga (-)-4-[(4-fluorfenoksy)fenyl]metylpiperidin som en olje i 64% utbytte. Behandling av denne olje med L-dibenzoylvinsyre i etanol (96%, 35 ml) ga et bunnfall som ble filtrert (smp. = 193-194 °C) . Aminoeteren ble frigjort under dannelse av et hvitt, fast materiale med 98% ee, smp. = 100-102 °C, og [a] 546 -14, c = 0,2, CHC13.
Følgende forbindelser ble analogt fremstilt: (-)-4-[(4-nitrofenoksy)fenyl]metylpiperidin-hydro-klorid (98,7% ee, smp. = 59 °C (d) , [a] 436 - 31, c = 0,042, etanol),
(+)-4-[(1-naftyloksy)fenyl]metylpiperidin-hydro-klorid (94% ee, smp. = 115 °C (d) , [a] 546 + 156, c = 0,128, CHCI3) , og (-)-4-[(2-fluorfenoksy)fenyl]metylpiperidin-sulfat (97,6% ee, smp. = 90 °C (d) , [a] 546 - 31, c = 0,14 0, CHC13) .
Eksempel 8
(+/-)- 4 -[( 3- fluorfenoksy) ( 3- fluorfenyl)] metylpiperidin- sulfat
En blanding av 4-cyanpiperidin (5 g, 40,92 mmol), (Boc)20 (11,7 g, 53,7 mmol), natriumbikarbonat (11,7 g,
13 9,3 mmol) og 117 ml vann ble omrørt ved romtemperatur i
17 timer. Dette ble ekstrahert med diklormetan, og den organiske fase ble tørket (vannfritt Na2S04) , filtrert og konsentrert. Den resulterende olje ble renset ved flashkromatografi (Still, w.C. Kahn, M., Mitra, A., J. Org. Chem. 1978, 43, 2923) under dannelse av 4-cyan-l-piperidinkarboksylsyre-l,1-dimetyletylester som en gul olje i 43% utbytte.
En Mg (0,5 g) suspensjon i eter (tørr, 22 ml) ble behandlet med noen milliliter (ca. 1/4 av totalen) av en 1-brom-3-fluorbenzen- (2,15 ml, 19,4 mmol) løsning i 16 ml tørr eter og en jodkrystall. Blandingen ble oppvarmet inntil jevn tilbakeløpskokning ble observert og fargen forsvant. Resten av løsningen ble deretter dråpevis tilsatt under opprettholdelse av en mild tilbakeløpskokning. Etter endt tilsetning ble løs-ningen kokt under tilbakeløpskjøling i 1 time og 3 0 minutter og fikk avkjøles til romtemperatur. En 4-cyan-l-piperidin-karboksylsyre-1,1-dimetyletylester- (2,7 g, 12,84 mmol) løsning ble dråpevis tilsatt til 27 ml tørr eter, og den resulterende blanding ble kokt under tilbakeløpskjøling i 3 timer. 50 ml av en mettet, vandig NH4C1-løsning ble tilsatt og ekstrahert med eter. Vanlig behandling av den organiske fase ga en olje som ble renset ved flashkromatografi (Still, W.C, Kahn, M., Mitra, A., J. Org. Chem. 1978, 43, 2923) ga 2,4 g (61% utbytte) av 4-(3-fluorbenzoyl)-1-piperidinkarboks-ylsyre-l , 1-dimetyletylester som en gulaktig olje.
Det ovenfor erholdte produkt (2,4 g, 7,8 mmol) ble oppløst i 30 ml metanol, og NaBH4 (0,2 g) oppløst i 3,5 ml vann, ble tilsatt. Blandingen ble oppvarmet i 2 timer i et oljebad (50-60 °C) , og produktet ble isolert på vanlig måte under dannelse av (+/-)-4-[ (3-fluorfenyl)hydroksy]metyl-1-piperidinkarboksylsyre-1,1-dimetyletylester som en meget tett, gulaktig olje i kvantitativt utbytte.
En løsning av den racemiske alkohol fremstilt ovenfor (2,4 g, 7,8 rnrnol) i 25 ml DMSO ble dråpevis tilsatt til en NaH-suspensjon (60%) (0,62 g) i 15 ml DMSO. Kaliumbenzoat (1,53 g, 9,55 mmol) og 1,3-difluorbenzen (1,2 ml, 11,9 mmol) ble tilsatt, og blandingen ble oppvarmet i et oljebad (65-70 °C) inntil utgangssubstansen forsvant. Blandingen ble deretter helt over i en blanding av 50 ml mettet NaCl-løsning og 39 ml vann. Dette ble ekstrahert med eter, og vanlig behandling av eterfasen ga en olje som ble kokt under tilbakeløps-kjøling i en blanding av 40 ml metanol og 40 ml 10% vandig HC1-løsning i 1 time og 30 minutter. Det ønskede produkt (+/-)-4-[(3-fluorfenoksy)(3-fluorfenyl)]metylpiperidin ble erholdt som en ravfarget olje i 50% utbytte. RMN^H (CDC13) av dette produkt utviste et signal ved 4,55 ppm (d, J = 6,1 Hz, CHOAr) og RMN-<13>C (CDC13) et signal ved 83,9 ppm svarende til CHOAr-karbon. Den ovenfor fremstilte olje ble behandlet med 0,22 ml konsentrert H2S04-løsning i 16,5 ml vann under dannelse av sulfatet som et svakt farget, fast materiale (smp. = 158 °C (d)).
Følgende forbindelser ble analogt fremstilt: (+/-)-4-[(2-fluorfenoksy)(3-fluorfenyl)]metylpiperidin-hydroklorid (62% utbytte, smp. = 90 °C) og
(+/-)-4-[(4-fluorfenoksy)(3-fluorfenyl)]metylpiperidin-hydroklorid (30% utbytte, smp. = 65 °C) .
Claims (5)
1. 4-substituerte piperidiner, karakterisert ved at de har generell formel CI)
hvori Ri og R2 er ikke-substituerte arylradikaler eller arylradikaler som er mono- eller polysubstituert med halogen (fluor, klor, brom, jod), alkyl, alkoksy, cyan, trifluormetoksy, trifluormetyl, benzoyl, fenyl, nitro, amino, aminoalkyl, aminoaryl og karbonylamino, og deres farmasøytisk akseptable salter med uorganiske og organiske syrer.
2. 4-substituerte piperidiner av generell formel (I) ifølge krav 1,
karakterisert ved at de er erholdt som racemiske blandinger og som rene enantiomerer.
3. 4-substituerte piperidiner av generell formel (I) ifølge krav 1 og 2,
