Description
Foreliggende oppfinnelse vedrører analogifremgangsmåte ved fremstilling av et terapeutisk aktivt aminosyrederivat med den generelle formel I
hvor R betyr benzyloksykarbonyl eller 2-kinolylkarbonyl, eller et farmasøytisk akseptabelt syreaddisjonssalt derav.
Forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter er nye og har verdifulle farmako-logiske egenskaper. Særlig hemmer de proteaser av virusopprinnelse og kan anvendes ved profylakse eller behandling av virusinfeksjoner, spesielt infeksjoner som forår-sakes av HIV og andre retrovirus.
Gjenstand for foreliggende oppfinnelse er dessuten mel-lomprodukter som anvendes i fremgangsmåten.
De farmasøytisk akseptable syreaddisjonssalter av forbindelsene med formel I er salter dannet med uorganiske syrer, f.eks. hydrohalogensyrer, såsom saltsyre eller hydrobromsyre, svovelsyre, salpetersyre, fosforsyre etc, eller med organiske syrer, f.eks. eddiksyre, sitronsyre, maleinsyre, fumarsyre, vinsyre, metansulfonsyre, p-toluensulfonsyre etc.
I henhold til fremgangsmåten som fremskaffes ved foreliggende oppfinnelse, fremstilles forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter ved
(a) å omsette 2-[(3(S)-amino-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid med formel II
med en syre med den generelle formel III
hvor R har den betydning som er angitt ovenfor, eller et reaktivt derivat derav, eller
(b) å redusere en forbindelse med den generelle formel
IV
hvor R har den betydning som er angitt ovenfor, og isole-re den ønskede 2(R)-hydroksyisomer fra den erholdte blanding, eller (c) å omsette 2-[3(S)-[(L-asparaginyl)amino]-2(R)-hydr-oksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-iso-kinolin-3(S)-karboksamid med formel V med et middel som gir benzyloksykarbonyl- eller 2-kinol-ylkarbonylgruppen, og (d) om ønsket, å omdanne en erholdt forbindelse med formel I til et framasøytisk akseptabelt syreaddisjonssalt.
Omsettingen av en forbindelse med formel II med en syre med formel III ifølge utførelsesform (a) av fremgangsmåten kan utføres i henhold til kjente fremgangsmåter in-nenfor peptidkjemien. Således, når en syre med formel III anvendes, utføres reaksjonen fortrinnsvis i nærvær av et kondenseringsmiddel, såsom hydroksybenzotriazol og dicykloheksylkarbodiimid. Denne reaksjonen utføres med fordel i et inert organisk løsningsmiddel, såsom en eter (f.eks. dietyleter, tetrahydrofuran osv.) eller dimetylformamid ved en lav temperatur, fortrinnsvis omkring - 10"C til +5°C og spesielt omkring 0°C. Egnede reaktive syrederivater med formel III som kan anvendes er f.eks. de tilsvarende syrehalogenider (f.eks. syreklorider), syreanhydrider, blandede anhydrider, aktiverte estere etc. Når et reaktivt derivat anvendes, utføres reaksjonen med fordel i et inert organisk løsningsmiddel, såsom et halogenert alifatisk hydrokarbon (f.eks. diklormetan etc.) eller en eter (f.eks. dietyleter, tetrahydrofuran etc.) og, om ønsket, i nærvær av en organisk base (f.eks. N-etylmorfolin, diisopropyletylamin etc.) ved en lav temperatur, fortrinnsvis omkring -10°C til +5°C og særlig omkring 0°C.
Reduksjonen av en forbindelse med formel IV ifølge ut-førelsesform (b) av fremgangsmåten kan utføres i henhold til kjente metoder vedrørende reduksjon av en karbonyl-gruppe til en hydroksygruppe. Således kan f.eks. reduksjonen utføres under anvendelse av et kompleks metall-hydrid, såsom et alkali-metall-borhydrid, spesielt natri-umborhydrid, i et egnet organisk løsningsmiddel såsom, en alkanol (f.eks. metanol, etanol, propanol, isopropanol etc). Med fordel utføres reduksjonen ved ca. værelsestemperatur. Isoleringen av den ønskede 2(R)-hydroksy-isomer fra den erholdte blanding kan utføres i henhold til konvensjonelle metoder, f.eks. ved kromatografi og lignende.
I henhold til utførelsesform (c) av fremgangsmåten er det egnede middel som gir benzyloksykarbonylgruppen, benzylklorformiat. Egnede midler som gir 2-kinolylkarbonylgrup-pen er den tilsvarende syre eller det reaktive derivat derav, såsom de tilsvarende syrehalogenider (f.eks. syre-klorid), syreanhydrid, blandede anhydrider, aktiverte estere etc. Omsetningen av en forbindelse med formel V med de ovenfor nevnte midler utføres på samme måte som beskrevet tidligere i forbindelse med utførelsesform (a) av fremgangsmåten.
Omdannelsen av en forbindelse med formel I til et farma-søytisk akseptabelt syreaddisjonssalt i forbindelse med utførelsesform (d) av fremgangsmåten kan utføres ved å behandle en slik forbindelse på konvensjonell måte med en uorganisk syre, f.eks. en hydrohalogensyre, såsom saltsyre eller hydrobromsyre, svovelsyre, salpetersyre, fosforsyre etc., eller med en organisk syre, såsom eddiksyre, sitronsyre, maleinsyre, fumarsyre, vinsyre, metansulfonsyre, p-toluensulfonsyre etc.
Forbindelsen med formel II som anvendes som utgangsmateriale i utførelsesform (a) av fremgangsmåten er ny og er også gjenstand for foreliggende oppfinnelse.
Forbindelsen med formel II kan fremstilles f.eks. ved å omsette en forbindelse med den generelle formel VI
hvor R<1> betyr en aminobeskyttende gruppe (f.eks. tert.bu-toksykarbonyl eller benzyloksykarbonyl), og X betyr et
klor-eller bromatom, med N-tert.butyl-decahydro-(4aS,8aS)isokinolin-3(S)-karboksamid med formel VII
og redusere den resulterende forbindelse med den generel- hvor R<1> har den betydning som er angitt ovenfor, og iso-lere den ønskede 2(R)-hydroksyisomer fra den erholdte blanding og avspalte gruppen R<1> fra den resulterende forbindelse med den generelle formel IX
hvor R<1> har den betydning som er angitt ovenfor, for å få en forbindelse med formel II.
Omsetningen av en forbindelse med formel VI, fortrinnsvis én hvor R<1> betyr benzyloksykarbonyl, med en forbindelse med formel VII kan utføres på kjent måte; f.eks. i et inert organisk løsningsmiddel, såsom et halogenert alifatisk hydrokarbon (f.eks. diklormetan etc.) og i nærvær av en base (f.eks. et trialkylamin, såsom trietylamin etc.) med fordel ved ca. værelsestemperatur.
Reduksjonen av en forbindelse med formel VIII for å få en forbindelse med formel IX og den påfølgende isolering av den ønskede 2(R)-hydroksyisomer kan utføres som beskrevet tidligere i forbindelse med utførelsesform (b) av fremgangsmåten ifølge oppfinnelse, dvs. reduksjon av en forbindelse med formel IV og isolering av den ønskede 2(R)-hydroksyisomer fra den erholdte blanding.
Avspaltingen av gruppen R<1> fra en forbindelse med formel IX kan også utføres på kjent måte; f.eks. ved å anvende en sterk uorganisk syre, såsom en hydrohalogensyre eller en sterk organisk syre (f.eks. trifluoreddiksyre etc), med fordel ved ca. 0°C til ca. værelsestemperatur. Alter-nativt kan en hydrogenolytisk spaltbar aminobeskyttende gruppe R<1> avspaltes ved hjelp av hydrogen i nærvær av en edelmetallkatalysator (f.eks. en palladiumkatalysator, såsom palladium på trekull) i et organisk løsningsmiddel eller en løsningsmiddelblanding som er inert under reak-sjonsbetingelsene (f.eks. en alkanol, såsom etanol, isopropanol etc, en alkankarboksylsyreester, såsom etylacetat etc.) og med fordel ved ca. værelsestemperatur.
En ytterligere fremgangsmåte ved fremstilling av forbindelsen med formel II omfatter å først omsette en forbindelse med den generelle formel X hvor R<1> har den betydning som er angitt tidligere, med forbindelsen med formel VII som er nevnt tidligere, med fordel i et inert organisk løsningsmiddel, såsom en alkanol (f.eks. metanol etc), dimetylformamid eller lignende ved forhøyet temperatur, fortrinnsvis ved ca. 60°C til ca. 120°C, og deretter avspalte gruppen R<1> i reaksjons-produktet (en forbindelse med formel IX ovenfor) som beskrevet tidligere.
Forbindelsene med formel IV som anvendes som utgangsmateriale i utførelsesform (b) av fremgangsmåten kan fremstilles f.eks. ved å avspalte den aminobeskyttende gruppe R<1> fra en forbindelse med formel VIII og omsette produk-tet med en syre med formel III eller et reaktivt derivat derav. Denne omsetning kan utføres analogt med hva som er beskrevet tidligere i forbindelse med utførelsesform (a) av fremgangsmåten.
Forbindelsen med formel V som anvendes som utgangsmateriale i utførelsesform (c) av fremgangsmåten er ny og utgjør en ytterligere gjenstand for foreliggende oppfinnelse.
Forbindelsen med formel V kan fremstilles f.eks. ved
avspaltning av benzyloksykarbonylgruppen R fra forbindelsen med formel i, hvor R betyr benzyloksykarbonyl, eller den tertiære butoksykarbonylgruppe danner en forbindelse som tilsvarer formel I, men hvor R betyr tertiært but-oksykarbonyl. Denne sistnevnte forbindelse kan fremstil-
les f.eks. ved å omsette forbindelsen med formel II med N-(tert.butoksykarbonyl)-L-asparagin i henhold til utfør-elsesform (a) av fremgangsmåten. Den ovennevnte avspaltning utføres analogt med hva som er beskrevet tidligere i forbindelse med avspaltningen av gruppen R<1> fra en forbindelse med formel VIII.
Utgangsmaterialene med formel III og deres reaktive derivater samt forbindelsene med formlene VI, VII og X ovenfor, hvis de ikke er kjente forbindelser eller analoge med kjente forbindelser, kan fremstilles på lignende måte som de kjente forbindelser eller som beskrevet i de et-terfølgende eksempler eller analogt med dem. Videre er midlene som anvendes i utførelsesform (c) av fremgangsmåten vanlig kjente forbindelser.
Som nevnt tidligere, hemmer forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter proteaser av virusopprinnelse, og de er nyttige ved behandling eller profylakse av virusinfeksjoner, spesielt av infeksjoner som skyldes HIV eller andre retrovirus.
In-vitro-hemmingen av HIV-protease av forbindelsene som frembringes ved foreliggende oppfinnelse, kan vises ved hjelp av følgende forsøk: HIV-protease ble uttrykt i E. coli og delvis renset for løselige ekstrakter av bakterien ved ammoniumsulfatfrak-sjonering (0-30%). Proteaseaktivitet ble bestemt ved å anvende det beskyttede heksapeptid succinyl-Ser-Leu-Asn-Tyr-Pro-Ile-isobutylamid (S<1>) eller det beskyttede hepta-peptid succinyl-Val-Ser-Gln-Asn-Phe-Pro-Ile-isobutylamid (S<2>) som substrat. Avspaltning av substratet ble kvanti-fisert ved å måle produksjonen av H-Pro-Ile-isobutylamid ved spektrofotometrisk bestemmelse av N-terminalt prolin.
1,25 mM substrat ble oppløst i 125 mM citratbuffer (pH
5,5) som inneholdt 0,125 mg/ml Tween 20. 10 \ il av en løsning med forskjellige konsentrasjoner av forsøks-forbindelsen (oppløst i metanol eller dimetylsulfoksyd og fortynnet med vann som inneholdt 0,1% Tween 20) og 10 ul protease, ble tilsatt til 80 ul av det ovenfor nevnte buffrede substrat. Omsetningen ble utført ved 37°C i en bestemt tidsperiode og ble avsluttet ved å tilsette 1 ml fargereagens [30 iig/ml isatin og 1,5 mg/ml 2-(4-klor-benzoyl)benzosyre i 10% aceton i etanol (vol./vol.)]. Løsningen ble oppvarmet i et vannbad og deretter ble de fargede residuer gjenoppløst i 1 ml 1% pyrogallol i 33% vann i aceton (vekt/vol./vol.). Den optiske densitet i løsningen ble målt spektrofotometrisk ved ved 599 nm. Dannelsen av H-Pro-Ile-isobutylamid i nærvær av forsøks-forbindelsen ble sammenlignet med kontroller, og konsentrasjonen av forsøksforbindelsen som var nødvendig for å gi 50% hemming (I50) ble bestemt ved hjelp av en kurve som var fremstilt av de forskjellige konsentrasjoner av anvendt forsøksforbindelse.
Den in vitro antivirale aktivitet av forbindelsene med formel I kan vises ved undersøkelsen som er beskrevet nedenfor:
Aktivitet mot HIV:
Denne undersøkelse anvender HTLV-III (stamme RF) dyrket i C8166-celler (en human CD4+ T-lymfoblastoid linje) med RPM1 1640-medium med bikarbonatbuffer, antibiotika og 10% bovint fosterserum.
En suspensjon av celler infiseres med ti ganger TCD50-verdien med virus og adsorpsjonen får pågå i 90 minutter ved 37°C. Cellene vaskes tre ganger med mediet. Forsøket utføres i 6 mis vevkulturrør, og hvert rør inneholder 2 x IO<5> infiserte celler i 1,5 ml medium. Forsøksforbindel-sene oppløses enten i vandig medium eller i dimetylsulfoksyd, alt etter deres oppløselighet, og en 50 uls løs-ning av substansen tilsettes. Kulturene inkuberes ved 37"C i 72 timer i en fuktig atmosfære som inneholder 5% karbondioksyd i luft. Kulturene sentrifugeres deretter og en aliquot av supernatanten solubiliseres med Nonidet P40 og underkastes en antigen innfangningsassay som anvender et primært antiserum med spesiell rektivitet mot det virale protein 24 og et pepperotperoksidase deteksjons-system. Fargeutviklingen måles spekrofotometrisk og fremstilles grafisk mot konsentrasjonen av forsøkssubstansen. Konsentrasjonen som frembringer 50% beskyttelse bestemmes
En cytotoksisitetsundersøkelse som er basert på farge-opptagelse og metabolisme av isotopmerket aminosyre-innlemmelse kjøres parallelt med ovennevnte undersøkelse for å bestemme den antivirale selektivitet.
De erholdte resultater i ovennevnte forsøk med forbindelsene med formnel I som forsøksforbindelse er angitt i følgende tabell.
Forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter kan anvendes som medikamenter
(f.eks. i form av farmasøytiske preparater). De farmasøy-tiske preparater kan administreres enteralt, såsom oralt (f.eks. i form av tabletter, belagte tabletter, dragéer, harde og bløte gelatinkapsler, oppløsninger, emulsjoner eller suspensjoner), nasalt (f.eks. i form av nesespray-er) eller rektalt (f.eks, i form av suppositorier). Men adminsistrasjonen kan også skje parenteralt, såsom intra-muskulært eller intravenøst (f.eks. i form av injeksjons-løsninger).
For fremstilling av tabletter, belagte tabletter, drageer og hardgelatinkapsler kan forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter fremstilles med farmasøytisk inerte, uorganiske eller organiske eksipienser. Laktose, maisstivelse eller derivater derav, talkum, stearinsyre eller dens salter kan anvendes, f.eks. som eksipienser for tabletter, drageer og hardgelatinkapsler.
Egnede eksipienser for bløtgelatinkapsler er for eksempel vegetabilske oljer, vokser, fetter, halvfaste og flytende polyoler etc.
Egnede eksipienser for fremstilling av løsninger og siru-per er for eksempel vann, polyoler, sakkarose, invert sukker, glukose etc.
Egnede eksipienser for injeksjonsløsninger er for eksempel vann, alkoholer, polyoler, glyserol, vegetabilske oljer etc.
Egnede eksipienser for suppositorier er for eksempel naturlige eller herdede oljer, vokser, fetter, halvfaste eller flytende polyoler etc.
Videre kan de farmasølytiske preparater inneholde konser-veringsmidler, oppløsningsmidler, viskositetsøkende substanser, stabiliseringsmidler, fuktemidler, emulgerings-midler, søtningsstoffer, fargestoffer, aromastoffer, salter for å variere det osmotiske trykk, buffere, beleg-ningsmidler eller antioksydanter. De kan også ytterligere inneholde andre terapeutisk verdifulle substanser.
Forbindelsene med formel I og deres farmasøytisk akseptable syreaddisjonssalter kan anvendes ved behandling eller profylakse av virusinfeksjoner, spesielt retrovirusinfek-sjoner. Doseringen kan variere innen vide grenser og vil naturligvis anpasses til de individuelle krav i hvert enkelt tilfelle. I tilfelle oral administrasjon vil det vanligvis være nok med en daglig dose på ca. 3 mg til ca. 3 g, fortrinnsvis ca. 10 mg til ca. 1 g (f.eks. ca. 300 mg pr. person), fordelt på fortrinnsvis 1-3 enhetsdoser som f.eks. kan være av samme mengde. Det er imidlertid underforstått at den øvre grense som er angitt ovenfor kan overskrides hvis dette blir funnet å være nødvendig.
Følgende eksempler illustrerer foreliggende oppfinnelse.
EKSEMPEL 1
En oppløsning av 561 mg 2-[3(S)-amino-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid og 372 mg N-(benzyloksykarbonyl)-L-asparagin i 20 ml tørt tetrahydrofuran ble avkjølt i en is/saltblanding. 189 mg hydroksybenzotriazol, 161 mg N-etylmorfolin og 317 mg dicykloheksylkarbodiimid ble tilsatt, og blandingen ble omrørt i 16 timer. Blandingen ble deretter fortynnet med etylacetat og filtrert. Filtratet ble vasket med vandig natriumbikarbonatløsning og natrium-kloridløsning. Løsningsmiddelet ble fjernet ved inndamp-ning, og residuet ble kromatografert på kisel-gel med diklormetan/metanol (9:1) for elueringen, hvilket ga 434 mg 2-[3(S)-[[N-(benzyloksykarbonyl)-L-asparaginyl]amino]-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-

2-[3(S)-amino-2(R)-hydroksy-4-fenylbutyl-N-tert.butyl-decahydro- (4aS,8aS)-isokinolin-3(S)-karboksamidet som ble anvendt som utgangsmateriale, ble fremstilt som følger: (i) En suspensjon av 12,676 g (71,6 mmol) av 1,2,3,4-tetra-hydro-3(S)-isokinolinkarboksylsyre (Chem. Pharm. Bull. 1983, 31, 312) i 200 ml 90% eddiksyre ble hydrogenert ved 80°C og under 140 atmosfærers trykk over 5% rho-dium på trekull i 24 timer. Blandingen fikk avkjøles til værelsestemperatur og katalysatoren ble deretter filtrert av. Filtratet ble inndampet, hvilket ga en gummi som ble oppløst i 10 ml etylacetat og tilsatt sakte til 100 ml kraftig omrørt diisopropyleter. En harpikslignende felling oppsto. Supernatantvæskene ble fjernet ved dekante-ring, og fellingen ble ekstrahert med varmt etylacetat. Denne varme løsning ble helt i en kraftig omrørt blanding av 150 ml dietyleter/diisopropyleter (1:1) hvilket ga et lyst grått fast stoff som ble oppsamlet ved filtrtering, vasket med dietyleter og tørket. Det ble erholdt 5,209 g av en blanding av decahydroisokinolin-3(S)-karboksylsyrer som hovedsakelig besto av (ca. 65%) 4aS,8aS-isomeren sammen med 4aR,8aR-isomeren (ca.25%) og ca 10% av trans-isomeren; MS: m/e 184 [M+H]<+>. (ii) 9,036 g (49,9 mmol) av den foregående blanding av decahydroisokinolin-3(S)-karboksylsyrer ble oppløst i 50 ml (50 mmol) IM natriumhydroksydløsning, og den resulterende løsning ble avkjølt til 0°C. 7,40 ml (51,87 mmol) benzylklorformiat og 58,7 ml (58,7 mmol) IM natriumhydr-oksydløsning ble tilsatt dråpevis i løpet av én time mens temperaturen ble holdt ved 0-5°C ved avkjøling. Blandingen ble deretter omrørt i ytterligere to timer, og i løpet av denne tid fikk blandingen anta værelsestemperatur. 100 ml dietyleter ble tilsatt og blandingen ble filtrert, hvorved den uløselige R,R-isomer ble fjernet. Det vandige sjikt av filtratet ble skilt vekk og justert til pH 1,5-2 ved å tilsette konsentrert saltsyre, hvorved en olje falt ut. Blandingen ble ekstrahert to ganger med 100 ml etylacetat hver gang. De kombinerte organiske ekstrakter ble vasket med vann, tørket over vannfritt natriumsulfat og inndampet, hvilket ga en olje. Denne olje ble oppløst i 35 ml etylacetat, og 2,85 ml (25 mmol) cykloheksylamin ble tilsatt, den hvite felling ble oppsamlet ved filtrering hvilket ga, etter flere frak-sjonene omkrystalliseringer fra metanol/etylacetat, 2,38 g av cykloheksaminsaltet av 2-(benzyloksy-karbonyl)-deca-hydro-(4aS,8aS9-isokinolin-3(S)-karboksylsyre; MS: m/e 318 [M+H]<+. >(iii) 2,334 g av cykloheksylaminsaltet av 2-(benzyloksykarbonyl) -decahydro- (4aS ,8aS)-isokinolin-3(S)-karboksylsyre ble fordelt mellom 50 ml etylacetat og 50 ml 10% sitronsyreløsning. Den organiske fase ble adskilt, vasket med vann, filtrert og inndampet, hvilket ga 1,87 g 2-(benzyloksykarbonyl)-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksylsyre i form av en fargeløs gummi; MS: m/e 318
[M+H]<+.>
(iv) En oppløsning av 0,634 g (2,0 mmol) 2-(benzyloksykarbonyl) -decahydro-(4aS,8aS)-isokinolin-3(S)-karboksylsyre i 6 ml dimetoksyetan ble behandlet med 0,23 g (2,0 mmol) N-hydroksysuccinimid og 0,412 g (2,0 mmol) dicykloheksylkarbodiimid. Blandingen ble omrørt ved værelsestemperatur i 18 timer. Blandingen ble filtrert og filtratet ble inndampet, hvilket ga 0,879 g av hydroksysuccin-imidesteren av den foregående syre i form av en lys gul olje. En oppløsning av 0,828 g (2,0 mmol) av den foregående N-hydroksysuccinimidester i 5 ml diklormetan ble omrørt, avkjølt til 0°C og behandlet med 0,219 g (3,0 mmol) tert.butylamin. Blandingen ble omrørt ved 0°C i 2 timer og deretter ved værelsestemperatur i 4,5 timer. Blandingen ble deretter vasket med 2M saltsyre, natrium-karbonatløsning og natriumkloridløsning, tørket over vannfritt magnesiumsulfat og inndampet. Residuet ble oppløst i 20 ml dietyleter og filtrert. Filtratet ble inndampet, hvilket ga 0,712 g 2-(benzyloksykarbonyl)-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid i form av et hvitt fast stoff; MS: m/e 373 [M+H]<+>. (v) En oppløsning av 0,689 g (1,85 mmol) 2-(benzyloksykarbonyl) -N-tert .butyl-decahydro- (4aS ,8aS)-isokinolin-3(S)-karboksymid i 20 ml etanol ble hydrogenert i nærvær av 0,01 g 10% palladium på trekull ved værelsestemperatur og under atmosfæretrykk i 18 timer. Katalysatoren ble fjernet ved filtrering og løsningsmiddelet ble fjernet ved inndamping, hvilket ga i kavntitativt utbytte N-ter.-butyl-decahydro-(4aS,8aS9-isokinolin-3(S)-karboksamid som en klar olje; MS: m/e 239 [M+H]<+> som ble brukt i neste trinn uten ytterligere rensing. (vi) En oppløsning av 440 mg N-tert.buty1-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid og 549 mg 3(S)-(benzyloksyformamid)-1,2(S)-epoksy-4-fenylbutan i 6 ml etanol ble omrørt ved 60°C i 7 timer. Ytterligere 54 mg 3(s)-(benzylformamid)-1,2(S)-epoksy-4-fenylbutan ble tilsatt, og løsningen ble omrørt ved 20"C i 16 timer. Løsningsmiddelet ble fjernet ved inndamping og residuet ble kromatografert på kisel-gel med dietyleter/n-heksan/- metanol (47,5:47,5:5) for elueringen, hvilket ga 771 mg 2-[3(S)-(benzyloksyformamid)-2(R)-hydroksy-4-fenylbutyl-N-tert. butyl-decahydro-(4aS,8aS9-isokinolin-3(S)-karboksamid som et hvitt fast stoff; MS: m/e 53 6 [M+H]<+>. (vii) En oppløsning 747 mg 2-[3(S)-(benzyloksyformamid)-2(R)-hydroksy-4-fenylbutyl-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid i 40 ml etanol ble hydrogenert over 10% palladium på trekull ved 20°C og under atmosfæretrykk i 5 timer. Katalysatoren ble fjernet ved filtering og filtratet ble inndampet, hvilket ga 561 mg 2-[3(S)-amino-2(R)-hydroksy-4-fenylbutyl].N-tert.-butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid som et brungult fast stoff som ble anvendt i neste trinn uten ytterligere rensing.
EKSEMPEL 2
En oppløsning av 154 mg 2-[3(S)-[(L-asparaginyl)-amino]-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,
8aS)-isokinolin-3(S)-karboksamid og 52 mg kinaldinsyre i 6 ml tørt tetrahydrofuran ble avkjølt i en is/saltbland-ing. 41 mg hydroksybenzotriazol, 35 mg N-etylmorfolin og
68 mg dicykloheksylkarbodiimid ble tilsatt og blandingen ble omrørt i 64 timer. Blandingen ble fortynnet med etylacetat og filtrert. Filtratet ble vasket med vandig nat-riumbikarbonatløsning og med natriumkloridløsning og deretter inndampet. Residuet ble kromatografert på kisel-gel med diklormetan/metanol (9:1) for eluering, hvilket ga 50 mg N-tert.butyl-decahydro-2-[2-(R)-hydroksy-4-

2-[3(s)-[(L-asparaginyl)amino]-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS, 8aS)-isokinolin-3(S)-karboksamidet som ble anvendt som utgangsmateriale, ble fremstilt som følger: En oppløsning av 195 mg 2-[3(S)-[[N-(benzyloksykarbonyl)-L-asparaginyl]amino]-2-(R)-hydroksy-4-fenylbutyl]-N-tert-.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid i 20 ml etanol ble hydrogenert ved værelsestemperatur og atmosfæretrykk i 18 timer over 10 mg 10% palladium på trekull. Katalysatoren ble filtrert av og filtratet ble inndampet under redusert trykk, hvilket ga 154 mg 2-[3(S)-[(L-asparaginyl)amino]-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid som ble anvendt i neste trinn uten ytterligere rensing.

En oppløsning av 287 mg N-(2-kinolylkarbonyl)-L-asparagin og 401 mg 2-[ 3 (S) -amino-2 (R) -hydroksy-4-f enylbutyl ] -Il-tert .butyl-decahydro- (4aS ,8aS)-isokinolin-3(S)-karboksamid [fremstilt som beskrevet i eksempel 1 (i)-(vii)] i 3 ml tetrahydrofuran ble avkjølt til -10°C og 163 mg 3-hydroksy-1,2,3-benzotriazin-4(3H)-on og 220 mg dicykloheksylkarbodiimid ble tilsatt. Blandingen ble omrørt ved -10°C i 2 timer og ved 20°C i 16 timer, deretter fortynnet med etylacetat og filtret. Filtratet ble vasket med mettet natriumbikarbonatløsning og mettet natrium-kloridløsning og deretter inndampet. Residuet ble kromatografert på kisel-gel med 4% (volumprosent) metanol i diklormetan for elueringen, hvilket ga 537 mg N-tert.-butyl-decahydro-2-[2(R)-hydroksy-4-fenyl-3(S)-[[N-(2-kinolylkarbonyl)-L-asparaginyl]amino]-butyl]-(4aS,8aS)-isokinolin-3(S)-karboksamid som var identisk med produk-tet erholdt i første avsnitt av eksempel 2.
N-(2-kinolylkarbonyl)-L-asparaginet som ble anvendt som utgangsmateriale, ble fremstilt som følger: En blanding av 540 mg kinaldinsyre-succinamidester og 300 mg L-asparagin monohydrat i 2 ml dimetylformamid ble omrørt ved 20°C i 96 timer. Løsningsmiddelet ble fjernet ved inndamping, hvilket ga et hvitt fast stoff som ble omrørt kraftig i 10 ml diklormetan, filtrert av og vasket med diklormetan. Det ble således erholdt 431 mg N-(2-kinolylkarbonyl)-L-asparagin som et hvitt fast stoff; MS: m/e 288 [M+H]<+>.
Følgende eksempel illustrerer fremstillingen av et farma-søytisk preparat som inneholder en forbindelse med formel I eller et farmasøytisk akseptabelt syreaddisjonssalt derav som den aktive bestanddel:
EKSEMPEL A
En vandig oppløsning av den aktive bestanddel filtreres sterilt og blandes under oppvarming med en steril gela-tinløsning som inneholder fenol som konserveringsmiddel, og man anvender mengder således at 1,00 ml av den resulterende blanding inneholder 3,0 mg aktiv bestanddel, 150,0 mg gelatin, 4,7 mg fenol og destillert vann opp til 1,0 ml. Blandingen fylles inn i små ampuller med en kapa-sitet på 1,0 ml under aseptiske betingelser.
Claims (4)
1. Analogifremgangsmåte ved fremstilling av et terapeutisk aktivt aminosyrederivat med den generelle formel I
hvor R betyr benzyloksykarbonyl eller 2-kinolylkarbonyl, eller et farmasøytisk akseptabelt syreaddisjonssalt derav,
karakterisert ved(a) å omsette 2-[(3(S)-amino-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboks-
amid med formel II
med en syre med den generelle formel III hvor R har den betydning som er angitt ovenfor, eller et reaktivt derivat derav, eller (b) å redusere en forbindelse med den generelle formel IV
hvor R har den betydning som er angitt ovenfor, og isole-re den ønskede 2(R)-hydroksyisomer fra den erholdte blanding, eller (c) å omsette 2-[3 (S)-[ (L-asparaginyl) amino]-2.(R)-hydr-oksy-4-fenylbutyl]-N-tert.butyl-decahydro-(4aS,8aS)-iso-kinolin-3 (S) -karboksamid med formel V
med et middel som gir benzyloksykarbonyl- eller 2-kinol-ylkarbonylgruppen, og (d) om ønsket, å omdanne en erholdt forbindelse med formel I til et framasøytisk akseptabelt syreaddisjonssalt.
2. Fremgangsmåte ifølge krav 1 ved fremstilling av N-tert.butyl-decahydro-2-[2(R)-hydroksy-4-fenyl-3(S)-[[N-(2-kinolylkarbonyl)-asparaginyl]amino]butyl]-(4aS,8aS)-isokinolin-3(S)-karboksamid, karakterisert ved å anvende tilsvarende substituerte utgangsmateria-ler.
3. Forbindelse, karakterisert ved at den er 2-[3(S)-amino-2(R)-hydroksy-4-fenylbutyl]-N-tert.butyl-decahydro- (4 aS , 8aS)-isokinolin-3(S)-karboksamid.
4. Forbindelse, karakterisert ved at den er 2-[3(S)-[(L-asparaginyl)amino]-2(R)-hydroksy-4-fenyl-butyl]-N-tert.butyl-decahydro-(4aS,8aS)-isokinolin-3(S)-karboksamid.
